FAPEMIG - Fundação de Amparo a Pesquisa do Estado de Minas Gerais Cassia bakeriana Craib é uma planta pertencente à família Leguminosae- Caesalpioideae, nativa da Tailândia e bem adaptada ao bioma errado. O Trabalho objetivou estudar a composição química das flores de C. bakeriana Craib, bem como explorar o potencial antifúngico, antibacteriano, antioxidante, antiglicação, citotóxico e identificar os constituintes químicos das amostras mais ativas. A prospecção química dos extratos e partições das flores indicou presença de terpenos, esteroides, açucares, fenóis, flavonoides e antraquinonas. A composição química da partição n-hexânica flores (P-HF) foi determinada por CG-EM, possibilitando identificar 11 compostos: cinco ésteres de cadeia longa (5; 45%), dois ácidos graxos de cadeia longa (2; 18%), dois alcanos de cadeia longa (2; 18%) e dois terpenos (2; 18%). Por meio análise da CLAE-EM-IES, foi possível identificar no extrato etanólico das flores (EEF) e nas partições hexânica das flores (P-HF), partição diclorometano das flores (P-DF), partição acetato de etila das flores (P-AEF), partição n-butanol das flores (PBF), partição água das flores (P-AF) os compostos fenólicos e derivados, flavonoides, antraquinonas e ácidos graxos. Na quantificação dos teores de compostos polifenólicos, foram encontrados os melhores teores para fenóis para o extrato etanólico (123,4±1,2 mg EAG/ gextrato) seguida da partição acetato de etila 95,4±0,6 mg EAG/ gextrato, para proantocianidinas com n-butanol (92,9±0,0 mg EC/ gextrato) e para teor de flavonoides, o extrato etanólico (109,6±0,4 mg EQ/ gextrato), seguidamente da partição acetato de etila com (128,6±0,6 mg EQ/ gextrato). A avaliação antifúngica e antibacteriana foi realizada pelo Método da Microdiluição em Caldo, contra as espécies de Candida e contra bactérias da cavidade bucal, respectivamente. As partições mais ativas contra C. tropicalis e C. glabrata foram P-HF, P-DF, P-BF e PAF, com valores de CIM variando entre 2,9 a 11,72 μg ml-1. Quanto à atividade antibacteriana, esta foi considerada moderada para as partições P-DF e P-BF, com CIM de 50 μg ml-1 para ambas as partições, contra a bactéria A. naeslundii. A avaliação da atividade antioxidante foi realizada pelos métodos ORAC, DPPH e FRAP. Dentre as partições, a P-AEF apresentou o melhor resultado (IC50 de 9,8 ±0,4 μg ml-1), demonstrando altíssima atividade antioxidante em todos os métodos empregados, com resultados superiores ao controle positivo ácido ascórbico (1317,2 ± 11,3 Trolox eq/gamostra). No que se refere a capacidade de inibição da glicação, novamente a P-AE apresentou maior percentual de inibição da glicação (IC50 38,6 ± 0,4 ml-1) igual ao controle positivo (quercetina). O índice de seletividade (IS) indicou que os extratos e partições das flores de C. bakeriana não apresentaram nenhuma citotoxicidade para células Vero nas concentrações testadas. Esses achados são auspiciosos, uma vez que o uso dessas partições ricas em polifenóis em outros ensaios "in vivo" pode ser viável. Além disso, os dados deste estudo ampliam o mapeamento do conhecimento desta espécie vegetal estudada por nosso grupo de pesquisa, contribuindo para a sua conservação e proteção no bioma Cerrado. Cassia bakeriana Craib is a plant belonging to the Leguminosae-Caesalpioideae family, native to Thailand and adapted to the Cerrado biome. The work aimed to study the chemical composition of C. bakeriana Craib flowers, as well as explore the antifungal, antibacterial, antioxidant, antiglycation, cytotoxic potential and identify the chemical constituents of the most active samples. Chemical prospection of extracts and flower partitions indicated the presence of terpenes, steroids, sugars, phenols, flavonoids and anthraquinones. The chemical composition of the flowers n-hexanic partition (P-HF) was determined by GC-MS, enabling the identification of 11 compounds: five long-chain esters (5; 45%), two long-chain fatty acids (2; 18%), two long-chain alkanes (2; 18%) and two terpenes (2; 18%). Through HPLC-EM-IES analysis, it was possible to identify in the ethanol extract of the flowers (EEF) and in the hexane partition of the flowers (P-HF), dichloromethane partition of the flowers (PDF), ethyl acetate partition of the flowers (P-AEF), n-butanol partition of flowers (PBF), water partition of flowers (P-AF) phenolic compounds and derivatives, flavonoids, anthraquinones and fatty acids. In the quantification of the contents of polyphenolic compounds, the best contents for phenols were found for the ethanol extract (123,4±1.2 mg EAG/gextrat) followed by the partition of ethyl acetate 95,4±0.6 mg EAG/gextract, for proanthocyanidins with n-butanol (92,9±0.0 mg EC/gextract) and for flavonoid content, the ethanol extract (109,6±0.4 mg EQ/gextract), followed by ethyl acetate partition with (128,6 ± 0.6 mg EQ / gextract). The antifungal and antibacterial evaluation was carried out using the Broth Microdilution Method, against Candida species and against bacteria from the oral cavity, respectively. The most active partitions against Candida tropicalis and Candida glabrata were P-HF, P-DF, P-BF, and P-AF with MIC values ranging from 2,9 to 11,72 μg ml-1. For the antibacterial activity, it was considered moderate for the P-DF and P-BF partitions, with a MIC of 50 μg ml-1 for both partitions, against the bacteria A. naeslundii. The evaluation of the antioxidant activity was carried out by the ORAC, DPPH and FRAP methods. Among the partitions, P-AEF had the best result (IC50 of 9,8 ± 0,4μg L-1), demonstrating remarkably high antioxidant activity in all tested methods, with results superior to the positive control ascorbic acid (1317,2 ± 11,3 μmol Trolox eq/gsample). Regarding the glycation inhibition capacity, again the P-AE presented a higher percentage of glycation inhibition (IC50 38,6 ± 0.4 μg mL–1) equal to the positive control (quercetin). The selectivity index (SI) indicated that the extracts and partitions of C. bakeriana Craib flowers did not show any cytotoxicity to Vero cells at the tested concentrations. These findings are auspicious since the use of this polyphenols-rich fractions in further “in vivo” assays could be feasible. Diversos metabólitos secundários (especializados) já foram isolados das espécies de Cassia, além de relatos do uso na medicina tradicional de várias partes das plantas para o tratamento da hanseníase, infecções por micose, doenças oftálmicas, de pele e hepáticas. Como a C. bakeriana possui poucos estudos publicados na literatura, neste trabalho foi avaliada a composição química e as atividades antioxidante e biológicas das cascas. Dessa forma, foram preparados os extratos hexânico (EH) e etanólico (EE) das cascas por maceração, seguido do fracionamento via extração líquido-líquido do EE com solventes de polaridade crescente, obtendo as frações diclorometano (FD), acetato de etila (FAE), n-butanol (FB) e aquosa (FA). Em relação aos ensaios, a capacidade antioxidante foi avaliada pelos métodos da capacidade de absorção do radical de oxigênio (ORAC) e do sequestro do radical 2,2-difenil-1-picril-hidrazil (DPPH). No método ORAC os valores obtidos para o EE e para as FD, FAE e FB foram bem acima dos valores obtidos para o controle ácido ascórbico (2498,50 ± 109,60 μmol troloxeq gamostra–1), sendo o valor mais expressivo para a FD com 4514,37 ± 176,03 μmol troloxeq gamostra–1. Em relação ao método DPPH, os valores de CI50 ficaram um pouco acima aos observados para o controle (4,07 ± 1,21 μg mL–1) e o melhor resultado foi para a FAE com inibição do radical em 93,30 ± 0,57% e CI50 de 25,90 ± 3,2 μg mL–1. Também foram avaliadas as atividades antidiabética através da capacidade dos extratos e frações de inibirem as enzimas α-amilase e α-glicosidase, com o melhor resultado obtido para a inibição da enzima α-amilase pela FAE com percentual de inibição de 94,00 ± 1,13% e CI50 de 1,05 ± 0,21 μg mL–1. Em relação a inibição da glicação, a FB apresentou valores promissores com inibição de 85,15 ± 2,47% e CI50 de 76,95 ± 5,87 μg mL–1. Através da espectrometria de massas (EM) foi possível identificar os compostos presentes no EE e nas FAE, FD e FB, que foram as mais ativas. As principais classes de metabólitos identificados foram flavonoides, proantocianidinas e antraquinonas, que podem ser responsáveis pelas atividades apresentadas. Foi a primeira vez que as atividades antioxidante e antidiabética foram avaliadas para as cascas e também a primeira avaliação da composição química completa do EE e frações. Estes resultados demonstram que os extratos e frações das cascas da C. bakeriana são promissores como fontes de compostos bioativos, além da contribuição para a quimiossistemática do gênero. Cassia bakeriana is a species found in Thailand and belongs to the Fabaceae family and the Cassia genus, which has great importance within this family. Several specialized metabolites have been isolated from Cassia species, in addition to the reports of the use of many parts of this plant in traditional medicine, for the treatment of hanseniasis, mycoses, eye infections, skin, and hepatic diseases. As C. bakeriana does not have many studies published in the literature, in this work, we evaluated the chemical composition, the antioxidant activities, and the biologic activities of the bark. Then, the hexane extract (EH) and ethanol extract (EE) were prepared by maceration, and the fractionation by liquid-liquid extraction of the EE with solvents of increasing polarity, obtaining the dichloromethane fractions (FD), ethyl acetate (FEA), n-butanol (FB) and water (FA). Regarding the tests, the antioxidant capacity was evaluated by ORAC and DPPH methods. In the ORAC method, the values obtained for the EE and for FD, FAE and FB were well above the values obtained for the control (ascorbic acid), and the best results were FD with 4514.37 ± 176.03 μmol troloxeq gsample–1. Regarding the DPPH method, the IC50 values were slightly above those observed for the control, the best result was FAE with 93,30% of radical inhibition and IC50 de 25.90 ± 3.2 μg mL–1. Also, the capacity inhibitory activities of α-amylase and α-glucosidase enzymes from extracts and fractions were evaluated. The most important result was the α-amylase inhibition by FEA with 94.00±1.13% of inhibition and IC50 of 1.05±0.21 μg mL–1. Another activity evaluated was the glycation inhibitory capacity, with FB showing the most promising value with inhibition of 85.15±2.47% and IC50 of 76.95±5.87 μg mL–1. By Mass Spectrometry (MS) it was possible to identify many of the compounds in the EE, FEA, FD, and FB, which were the most active ones. The main classes of the identified metabolites were flavonoids, proanthocyanidins and anthraquinones, which can be the responsible for the activities shown in the extracts and fractions. It was the first time that the antioxidant and antidiabetic activities for the bark were evaluated and the first complete chemical composition for the EE and other fractions. These studies showed that extracts and fractions on the C. bakeriana are promising sources of bioactive compounds, in addition to the contribution to the chemosystematics of the genre. O presente estudo teve como objetivo explorar o potencial antifúngico, antioxidante e antidiabético dos extratos e partições das folhas de C. bakeriana Craib bem como, identificar os constituintes químicos das amostras mais ativas. A atividade antifúngica foi realizada pelo método da microdiluição em caldo contra as espécies de Candida spp. As partições mais ativas para a C. glabrata foi a diclorometano (P-D) e acetato de etila (P-AE) (CIM 93,75 e 187,5 µg mL–1, respectivamente). A avaliação da atividade antioxidante foi realizada pelo método ORAC e do radical DPPH. O extrato etanólico (EE) e as partições apresentaram elevada atividade antioxidante no método ORAC com resultados superiores ao controle positivo ácido ascórbico (2498,5 ± 109,6 μmol Troloxeq/gamostra). No método do radical DPPH as partições n-butanol (P-B), P-AE e P-D apresentaram maior atividade antioxidante (IC50 21,7 ± 3,2; 21,9 ± 4,3 e 31,7 ± 10,9 μg mL–1, respectivamente). Em relação à inibição enzimática, o EE apresentou maior inibição da α-amilase (IC50 5,00 ± 0,85 µg mL–1). A P-D apresentou maior inibição da α-glicosidase (IC50 359,55 ± 2,90 µg mL–1). O EH apresentou elevada inibição da lipase pancreática (IC50 25,26 ± 8,77 µg mL–1). No que se refere a capacidade de inibição da glicação, as P-D e P-AE foram as mais ativas (IC50 37,85 ± 0,49 e 53,25 ± 11,24 µg mL–1, respectivamente). Através do fracionamento da P-D por CLAE-DAD semi-preparativo, os flavonóides canferol (XX) e canferol-3-O-ramnosídeo (XVII) foram isolados e caracterizados por RMN e EM-IES. Estes compostos apresentaram elevada atividade antioxidante no método ORAC (4198,0 ± 42,6 e 4153,6 ± 3,8 μmol Troloxeq/gamostra, respectivamente), mas apenas XX foi ativo para o radical DPPH (IC50 10,2 ± 0,1 μg mL–1). O composto XX apresentou elevada inibição da α-amilase (IC50 1,5 ± 0,14 μg mL–1). Os compostos XX e XVII apresentaram elevada inibição da glicação (IC50 64,16 ±1,53 e 84,27 ± 11,38 μg mL–1, respectivamente). Através da análise CLAE-EM-IES do EE, P-D e P-AE, alguns compostos como ácidos fenólicos, flavonoides, megastigmanos, ácidos graxos e derivados, esfingolipídios, proantocianidinas, cianidina e antraquinonas foram identificados. In relation to enzymatic inhibition, the EE showed higher α-amylase inhibition (IC50 5.00 ± 0.85 μg mL–1). The P-D showed greater α-glycosidase inhibition (IC50 359.55 ± 2.90 μg mL–1). The EH showed high inhibition of pancreatic lipase (IC50 25.26 ± 8.77 µg mL–1). In relation to glycation inhibition capacity, P-D and P-AE were also the most active (IC50 37.85 ± 0.49 and 53.25 ± 11.24 μg mL–1, respectively). By fractionation of P-D by semi-preparative CLAE-DAD, the flavonoids canferol (XX) and canferol-3-O-rhamnoside (XVII) were isolated and characterized by NMR and MS-IES. These compounds showed high antioxidant activity in the ORAC method (4198.0 ± 42.6 and 4153.6 ± 3.8 μmol Troloxeq/gsample, respectively), but only compound XX was active by DPPH radial method (IC50 10.2 ± 0.1 μg mL–1). The isolated compounds XX showed high inhibition of α-amylase (IC50 1.5 ± 0.14 μg mL–1). The compounds XX and XVII showed high inhibition of glycation (IC50 64.16 ± 1.53 and 84.27 ± 11.38 μg mL–1, respectively). Through the HPLC-ESI-MS analysis of EE, Fr-D and Fr-AE, some compounds such as phenolics acid, flavonoids, megastigmane, fatty acids and derivatives, sphingolipids, proanthocyanidins, cyanidin and anthraquinones were identified. The antimicrobial potential of extracts of bark and leaves of Cassia bakeriana Craib, Fabaceae, against aerobic and anaerobic oral bacteria was evaluated by the microdilution broth method. For crude ethanol extracts and organic fractions tested, the bark dichloromethane phase showed a significant antibacterial effect, with MIC values ranging from 12.5 to 100 μg/ml for most of the microorganisms tested. Thus, a bioassay-guided fractionation of this fraction was performed. This fractionation led to isolation of the 1,8-dihydroxy-anthraquinone-3-carboxylic acid, also known as cassic acid or rhein. It is the first time that this bioactive anthraquinone has been isolated from this plant. Rhein exhibited good selectivity and high activity against anaerobic microorganisms, with MIC values ranging between 3.12 μg/ml (11.0 μM) and 25 μg/ml (88.0 μM). These results were considered very promising since the most active samples and rhein showed greater selectivity against oral microorganisms than toxicity to Vero cells. The chemical composition of the essential oils from leaves, bark and wood of Cassia bakeriana Craib. was determined by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). Alcohols, aldehydes and fatty acids were the major components in leaf and bark oil, while wood essential oil was rich in fatty acids. Terpenes such as linalool, (E)-nerolidol and phytol were present in low concentrations. The antimicrobial activity against aerobic and anaerobic oral bacteria was evaluated using the microdilution method, as was the cell viability test carried out with Vero cells. The oils from leaves and bark showed high antimicrobial activity, with minimum inhibitory concentrations between 62.5 and 125 µg·mL⁻¹ for most of the tested bacteria, including Streptococcus mutans, the main etiological agent of dental caries. Leaves oil displayed the lowest cytotoxic effect (EC₅₀ of 153 µg·mL⁻¹), while wood oil exhibited the highest toxicity to Vero cells. C. bakeriana oils are thus a source of biologically active compounds against aerobic and anaerobic oral microorganisms. This study is the first report on the chemical composition, antimicrobial activity and cytotoxicity of C. bakeriana.

Fundação de Amparo a Pesquisa do Estado de Minas Gerais The species Cassia bakeriana Craib. is popularly known as pink cassia and belongs to the genus Cassia and family Leguminoseae. The present study had several goals: determine the macromolecular composition of the wood and bark of C. bakeriana; determine the content of phenolics and proanthocyanidins, as well as the antioxidant activity of extracts (wood, bark and leaves); evaluate the antimicrobial activity of various extracts of bark and leaves; characterize and determine the antimicrobial activity of essential oils (wood, bark and leaves); phytochemical study of the dichloromethane extract of the bark and the determination of the cytotoxic potential of the samples with the higher antioxidant and antimicrobial activities. The percentages of extractives, acid insoluble lignin (Klason), acid-soluble lignin, holocellulose, α-cellulose, hemicellulose and ashes, in wood were: 13.04, 21.0, 0.76, 67.4, 35.35, 32.06 and 2.08, respectively. The values found in bark: 53.88, 15.06, 0.6, 27.41, 20.32, 7.1 and 4.87. The contents of total phenols by Folin-Ciocalteau method and proanthocyanidins by vanillin in sulphuric acid indicated a positive correlation with the increasing polarity of the extracts of bark and leaves, with higher levels in the bark extracts. The extracts in ethanol, dichloromethane, ethyl acetate and methanol of leaves and bark of C. bakeriana showed good antioxidant activity by DPPH free radical scavenging method with CE50 values below 50 μg mL-1, and special attention to methanol extract of leaves with CE50 of 8.67 μg mL-1. The same extracts that showed good antioxidant activity, showed low toxicity to Vero cells (cytotoxic concentration ˃512 mg mL-1). The components of the essential oils from wood, bark and leaves of C. bakeriana were identified by GC-MS and tested for antimicrobial activity against aerobic and anaerobic oral microorganisms by broth microdilution method. Although these extracts did not showed high concentration of terpenes, the oils from leaves and bark showed significant antimicrobial activity with MIC\'s ranging between 62.5 and 125 μg mL-1, for most of the bacteria used in tests. According to the MIC\'s and selectivity index, the oil from the leaves showed promising antimicrobial activity and low cytotoxicity to Vero cells. The chloroform extract obtained from the bark in soxhlet extractor and the dichloromethane extract obtained by liquid-liquid partition also showed activity against oral microorganisms with MIC\'s below μg mL-1. Both extracts showed higher antimicrobial activity against anaerobes. The dichloromethane extract of the bark was subjected to biofractioning directed by antimicrobial activity, using phytochemicals methods, as well as HPLC coupled to mass spectrometry with electrospray ionization technique. The 1,8-dihydroxy-anthraquinone-3-carboxylic acid (rhein) was isolated from the dichloromethane fraction of the bark and identified spectroscopic methods as 1H-NMR, HSQC, COSY, FTIR and mass spectrometry. The compound rhein showed strong antimicrobial activity against oral anaerobic microorganisms. A espécie Cassia bakeriana Craib. é conhecida popularmente como cássia rósea e pertence ao gênero Cassia e á família Leguminoseae. O presente trabalho teve como objetivos: determinar a composição macromolecular da madeira e da casca de C. bakeriana; determinar o teor de fenóis e proantocianidinas, assim como a atividade antioxidante de extratos (madeira, casca e folhas); avaliar a atividade antimicrobiana de diversos extratos de casca e folhas; caracterizar e determinar a atividade antimicrobiana dos óleos essenciais (madeira, casca e folhas); realizar o estudo fitoquímico do extrato em diclorometano da casca e determinar o potencial citotóxico das amostras com maior atividade antioxidante e antimicrobiana. As porcentagens de extrativos, lignina insolúvel em ácido (Klason), lignina solúvel em ácido, holocelulose, α-celulose, hemiceluloses e cinzas para a madeira foram respectivamente 13,04; 21,0; 0,76; 67,4; 35,35; 32,06 e 2,08; e para a casca 53,88; 15,06; 0,6; 27,41; 20,32; 7,1 e 4,87. O conteúdo de fenóis totais pelo método Folin-Ciocalteau e proantocianidinas pelo método da vanilina sulfúrica indicaram uma correlação positiva com o aumento da polaridade dos extratos da casca e folhas, com maiores teores para a casca. Os extratos em etanol, diclorometano, acetato de etila e metanol das folhas e casca de C. bakeriana indicaram forte atividade antioxidante pelo método do sequestro do radical livre DPPH com valores de CE50 abaixo de 50 μg mL-1, com destaque para o extrato metanólico das folhas com CE50 de 8,67 μg mL-1. Os mesmos extratos que indicaram forte atividade antioxidante, apresentaram baixa toxicidade para células Vero (concentração citotóxica ˃512 μg mL-1). Os componentes dos óleos essenciais da madeira, casca e folhas de C. bakeriana foram identificados por CG-EM e submetidos aos testes de atividade antimicrobiana frente a microrganismos bucais aeróbios e anaeróbios pelo método da microdiluição em caldo. Embora não tenham apresentado elevada concentração de terpenos, os óleos das folhas e casca mostraram significativa atividade antimicrobiana com CIM s variando entre 62,5 e 125 μg mL-1, para a maioria das bactérias utilizadas nos ensaios. De acordo com as CIM s e índice de seletividade, o óleo das folhas apresentou promissora atividade antimicrobiana e baixa citotoxicidade para células Vero. O extrato clorofórmico da casca obtido em soxhlet e o extrato em diclorometanoda casca obtido por partição líquido-líquido também mostraram atividade frente aos microrganismos bucais com valores de CIM s abaixo de 100 μg mL-1. Ambos os extratos indicaram maior efeito antimicrobiano contra os microrganismos anaeróbios. O extrato em diclorometano da casca foi submetido ao fracionamento bioguiado por atividade antimicrobiana, utilizando métodos fitoquímicos, assim como pela técnica de CLAE acoplado à espectrometria de massas com ionização por eletrospray. O ácido 1,8-diidroxi-antraquinona-3-carboxílico (rhein) foi isolado da fração em diclorometano da casca e identificado por técnicas espectroscópicas de RMN-1H, HSQC, COSY, IVTF e espectrometria de massas.
