Abstract

Description of the subject. Dietary fiber is largely used in pig production but some contradictions appear in the literature regarding the effects on performance and health. Objectives. This paper aims to contribute to the clarification of the effects of a diet rich in sugar beet pulp on animal behavior, growth performance, carcass quality and gut health of fattening pigs. Method. Two successive batches of 24 fattening pigs were each divided into two groups fed ad libitum either a standard diet based on cereals (STD, 19% NSP [non-starch-polysaccharides]) or a fibrous diet based on 23% sugar beet pulp (HFD, 31% NSP). Results. Pigs activity rate and feeder occupancy duration were increased by 57% and 165% for group fed HFD, respectively (P < 0.05). The fecal bacteria counts showed increases with HFD for Lactobacillus (10.21 vs 9.84 log10 of cfu·g-1 of feces dry matter, P < 0.05) and Bifidobacterium (9.49 vs 8.88, P < 0.01) but decreases for Enterobacteriaceae (4.85 vs 5.97, P < 0.001). Reductions of the average daily gain (788 vs 876 g per day, P < 0.001) and the dressing percentage (75.7 vs 78.9%, P < 0.001) were observed with HFD. Gastric lesion score was decreased with HFD (0.82 vs 1.55, P < 0.05). For pigs fed HFD, the proportion of surface area occupied by goblet cells was increased in the jejunum (10.06 vs 7.99%, P < 0.01) and the number of CD3 lymphocytes was increased in the colon (1.24 vs 0.90·mm-², P < 0.05). Conclusions. HFD contributes to strengthen the gut health of fattening pigs, but it impairs growth performance and carcass traits.

23. Fetal infection with Neospora caninum in dairy and beef cattle in Belgium

Abstract

Neospora caninum is a protozoan parasite, which causes fetal and neonatal mortality in livestock and companion animals. In 224 abortions in Belgian cattle, different diagnostic methods were used to demonstrate infection, and the presence of N. caninum. An indirect fluorescent antibody test (IFAT) was used to analyze fetal and maternal sera and immunohistochemistry (IHC) was performed when lesions consistent with neosporosis were observed in the brain, heart or liver. Twenty dairy cattle sera out of 70 (29%) and 13 beef cattle sera out of 93 (14%) were positive by IFAT. A positive titer to N. caninum was found in seven and three fetuses born to beef and dairy cows, respectively. Lesions consistent with N. caninum infection were observed in 17 fetuses. Of nine positive beef fetuses, five were confirmed by IHC while, all but one dairy fetus were confirmed using the same technique. Age had no influence on the serological status of the mother (P = 0.486) whereas husbandry system had a borderline influence (P = 0.082). However, a strong association (P = 0.004) between the level of antibodies in the darn and the occurrence of lesions in the fetus was observed and lesions were more prominent in dairy than in beef fetuses. Additionally, the distribution of intra-cerebral lesions was more extensive in dairy than in beef fetuses (P < 0.0001). Age and serological status of the fetus were found to influence the occurrence of lesions in beef fetuses (both P < 0.001) but no such significant relationships could be demonstrated in dairy fetuses. 32. Measurement of beta 2-microglobulin in serum by a particle-enhanced nephelometric immunoassay.

Abstract

A particle-enhanced immunoassay of beta 2-microglobulin in serum is described. It is based on the agglutination of complexes formed between the serum beta 2-microglobulin and latex particles coated with F(ab')2 fragments of polyclonal anti-beta 2-microglobulin antibodies. The analytical range of the method is 0.50 to 16 mg/l; it can be extended by appropriate dilution to 0.12 to 80 mg/l with good precision (CV less than 5% over the whole range). The accuracy and the precision are confirmed by a good correlation with radioimmunoassay (n = 123, r = 0.993). No error due to antigen excess was observed, even up to 292 mg/l. The main advantages of the method are its simplicity, its low cost per test and its high sensitivity (final dilution of the sample at 1/1200) with no known interference. The calibration curve is stable for at least 2 weeks.

Abstract

A latex particle immunoturbidimetric assay in microplates has been developed for the quantitation of beta 2 microglobulin. The assay which involves three pipetting steps and one incubation, has a clinically useful range of 0.4-16 mg/l. A comparison of the method with a commercial radioimmunoassay in the analysis of 125 sera revealed a high degree of correlation. Although the assay was performed manually, it showed considerable potential for full automation.

Abstract

We describe here two latex immunoassays for total thyroxine (T4) and total triiodothyronine (T3). These homogeneous 60 min assays are quantified by optically counting the monomeric particles remaining after agglutination. When precision is assessed, both methods display coefficients of variation of 3-7% for within-run assays and 4-10% for between-run assays. The accuracy of the methods, as tested by dilution and spike recovery experiments, was found to be satisfactory. Two correlation studies were carried out to compare the present method with leading commercial methods. The coefficients obtained were: r = 0.92 and r = 0.93 with 150 sera for T3, and r = 0.95 (150 sera) and r = 0.93 (108 sera) for T4.

35. Immunoassay by particle counting for coagulation testing: application to the determination of protein C.

Abstract

Latex particles coated with F(ab)'2 fragments of anti-protein C IgG antibodies are agglutinated by protein C, and the quantity of particles agglutinated is proportional to the concentration of protein C. The reaction can be quantitated by optical particle counting. Based on this system, we designed an immunoassay for protein C. Precision measured at low, medium and high levels of protein C varied from 3.3% to 13.7%. Specificity was evaluated by dilution recovery. A correlation coefficient of r = 0.959 was found when the new method was compared with a chromogenic technique on 131 plasmas.

Abstract

A latex particle immunoassay has been developed for the quantification of choriogonadotropin in human serum using two monoclonal antibodies specific for the beta-chain of the hormone. The assay, based on optical counting of monomeric particles, was achieved in 40 min and the calibration curve was linear between 10 and 200 IU/l. Intra- and interassay precisions at three different levels of the curve varied between 3.3 and 10.9%. The method was validated by comparison with two different radioimmunoassays and correlation coefficients of 0.97-0.99 were obtained.

Abstract

By means of a new technique (Particle Counting Immunoassay), we have determined the level of ferritin in 470 samples of cerebrospinal fluid of patients with various neurological disorders. The median value obtained in a control group was 2.3 ng/ml with an upper limit at 5.5 ng/ml. the concentrations in the serum and cerebrospinal fluid were independent, but that in cerebrospinal fluid correlated with its total protein content. High values of ferritin were found in infectious meningo-encephalitis, in vascular diseases of the central nervous system, and, unexpectedly, in several cases of dementia without obvious vascular pathology. 48. Pancreatic Injury in Equine Acute Abdomen Evaluated by Plasma Trypsin Activity and Histopathology of Pancreatic Tissue

Abstract

In cases of equine acute abdominal disease, where pancreatic damage is suspected, pancreatic damage can be assessed by measuring increased trypsin activity in the plasma of horses suffering intestinal obstruction and severe shock. The pancreas is particularly vulnerable to splanchnic hypoperfusion because it is a highly active tissue. In this study, 10 horses undergoing abdominal surgery for intestinal obstruction were assayed for trypsin activity on admission and, because of extensive intestinal lesions that were not amenable to surgery, euthanasia was selected; the pancreas was removed before euthanasia. Trypsin activity in the plasma of these horses was significantly higher than in healthy horses (196 ng/ml ± 128.2 versus 28.5 ng/ml ± 19.2; P = 0.0026). Light and transmission electron microscopy revealed slight to severe lesions of vacuolar degeneration, a few zymogen granules, dilation of the endoplasmic reticulum, and swelling of mitochondria in the exocrine pancreas. The activation of an inflammatory cascade occurring during strangulating intestinal obstruction could increase pancreatic anoxic lesions caused by severe shock and hypoperfusion in the horse. Further studies will show the significance of pancreatic lesions and the ensuing damage in equine acute intestinal obstruction and shock.

To determine prevalence of antibodies against Schmallenberg virus in adult cows and proportion of infection transmitted to fetuses, we tested serum samples from 519 cow/calf pairs in Belgium in spring 2012. Abstract

The article describes examples of clinical based applied research as a tool to approach animal diseases. It relates the occurrence of serial episodes of an unidentified gastrointestinal (GI) disease among puppies and dogs, which was subsequently identified as canine parovirus. Findings concerning unusual cases of systemic disease in heifers which was discovered as malignant catarrhal fever (MCF) are discussed. The article also describes cases of horses suffering from equine atypical myopathy and cases of bluetongue in lynx.
