1. Identification of putative orthologs of clinically relevant antimicrobial peptides in the equine ocular surface and amniotic membrane

Abstract

ObjectivesThis study aimed to define the antimicrobial peptide (AMP) expression pattern of the equine ocular surface and amniotic membrane using a targeted qPCR approach and 3'Tag-sequencing. It will serve as a reference for future studies of ocular surface innate immunity and amniotic membrane therapies.ProceduresA targeted qPCR approach was used to investigate the presence of orthologs for three of the most highly expressed beta-defensins (DEFB1, DEFB4B, and DEFB103A) of the human ocular surface and amniotic membrane in equine corneal epithelium, conjunctiva, and amniotic membrane. 3'Tag-sequencing was performed on RNA from one sample of corneal epithelium, conjunctiva, and amniotic membrane to further characterize their AMP expression.ResultsEquine corneal epithelium, conjunctiva, and amniotic membrane expressed DEFB1, DEFB4B, and DEFB103A. DEFB103A was expressed at the highest amounts in corneal epithelium, while DEFB4B was most highly expressed in conjunctiva and amniotic membrane. 3'Tag-sequencing from all three tissues confirmed these findings and identified expression of five additional beta-defensins, 11 alpha-defensins and two cathelicidins, with the alpha-defensins showing higher normalized read counts than the beta-defensins.ConclusionsThis study identified AMP expression in the equine cornea and conjunctiva, suggesting that they play a key role in the protection of the equine eye, similar to the human ocular surface. We also determined that equine amniotic membrane expresses a substantial number of AMPs suggesting it could potentiate an antimicrobial effect as a corneal graft material. Future studies will focus on defining the antimicrobial activity of these AMPs and determining their role in microbial keratitis.

Abstract

Drawing provides opportunities for children to communicate their thoughts even when they do not have the vocabulary or the English proficiency to fully explain their ideas. This practical guide presents foundational information on the role of drawing in vocabulary development. The authors describe a research-based intervention designed to support and expand young multilingual learners' experiences with content area vocabulary. They provide teaching examples from several content area investigations carried out in Head Start contexts serving multilingual students. These vignettes, accompanied by student work samples and excerpts of dialogue, will help early childhood educators effectively integrate this pedagogical approach into their classrooms. The user-friendly text includes curriculum support materials such as lesson-planning templates and lists of recommended children's literature and media. "Guided Drawing With Multilingual Preschoolers" shows teachers how to use guided drawing in conjunction with established practices to help all young students develop language and content knowledge, particularly in science. This book features: (1) An innovative pedagogical intervention that was created by the authors to use in Head Start classrooms; (2) An actionable approach to teaching content area vocabulary in the classroom that works with young multilingual learners; (3) Tables with quick summaries of developmental milestones and teaching points; (4) Guidance for early educators who understand the importance of building word and world knowledge in authentic ways while children are learning English; (5) Teaching examples that highlight language-rich interactions and strategies for supporting multilingual learners; and (6) Curriculum connections to culturally relevant children's literature, media, and high-quality informational texts.

10. Exploring Assessment Demands and Task Supports in Early Childhood Phonological Awareness Assessments

Abstract

Phonological awareness is assessed in various ways in both research studies and early childhood classrooms. The measures used to assess phonological awareness are related closely, although they differ in the linguistic unit used (e.g., word, syllable, onset-rime, or phoneme), the position of the linguistic unit (e.g., initial, medial, final), the task operation (e.g., detection, segmentation, deletion, production), the response mode (e.g., verbal, nonverbal), the task support or demands (e.g., picture prompts or memory demands), and in words used (low vs. high age of acquisition). To understand phonological awareness assessments better, phonological awareness tasks used with young children were examined based on these variations. The results indicate that considerable variability exists in the cognitive demands on phonological awareness assessments used in early childhood settings. These results raise some important questions regarding how phonological awareness is assessed in early childhood. Moreover, variations in response format and task support/demand are likely to increase or decrease the complexity of the tasks, which could have implications for classroom instruction and intervention.

12. An Examination of the Relations between Oral Vocabulary and Phonological Awareness in Early Childhood

Abstract

This article reports a post hoc analysis conducted as part of a larger study in which 61 typically developing, 3-, 4-, and 5-year-olds were assessed in phonological awareness (PA), vocabulary (i.e., receptive, expressive, and definitional), and grammatical skill at baseline and 3, 6, and 9 months later. The larger study's purpose was to shed light on the theory of lexical reorganization claim that increases in receptive vocabulary provide the basis for PA development. Its results showed that expressive vocabulary accounted for additional PA growth in 4- and 5-year-olds but not in 3-year-olds. The post hoc study involved a closer analysis of PA growth in relation to expressive vocabulary. Its results suggest that increases in receptive vocabulary might be necessary as a foundation for initial PA development but that expressive-level vocabulary might be needed to hold words in memory to perform complex manipulations required in higher level PA tasks.

14. Examining Differential Effects of a Family Literacy Program on Language and Literacy Growth of English Language Learners with Varying Vocabularies

Abstract

Many English language learners (ELLs) and children living in poverty begin school with substantially less English vocabulary knowledge than their monolingual, economically advantaged peers. Without effective intervention, these vocabulary gaps are likely to contribute to long-term reading failure. This quasi-experimental study examined the extent to which a family literacy program (FLP) moderated vocabulary development of 158 ELLs (prekindergarten through third grade) from low-income families in relation to children's level of vocabulary knowledge (at, moderately below, or substantially below national norms) on program entry. The FLP activities focused on supporting parents' development of English literacy and on teaching them effective ways to engage their children in authentic, home-based literacy events and practices that could be expected to prepare their children for success in school. Findings indicated that although all children demonstrated substantial language and literacy growth, children with the lowest pretest vocabulary knowledge achieved the greatest vocabulary gains and these gains differed significantly from their demographically matched peers who did not participate in the FLP. Gains for treatment and control children with middle and high pretest vocabulary knowledge (relative to this sample) did not differ significantly. Findings suggest that an FLP emphasizing authentic literacy practices holds particular promise in closing vocabulary gaps among children who enter early childhood classrooms with especially limited English vocabulary knowledge.

Abstract

Mesenchymal stem cells (MSCs) are frequently delivered via needle injection for treatment of musculoskeletal injuries. The purpose of this study was to evaluate the effect of needle diameter on the viability of MSCs.Equine bone marrow-derived MSCs from 5 horses were suspended in PBS, and held at room temperature for 7 hours to mimic shipping conditions. Two replicate samples for each needle size (20, 22, 23, or 25-gauge [ga]) were aspirated into a 3 mL syringe and re-injected into the holding vial 3 times, to reproduce the resuspension of cells prior to injection in clinical cases. Cells were stained with fluorescein diacetate and propidium iodide to measure viability. Flow cytometry (FC) was performed to compare cell debris and intact cells between groups.MSC viability was higher when cells were passed through a 20-ga rather than a 25-ga needle. Cell suspensions passed through a 20-ga needle contained a larger percentage of intact cells, compared to 25-ga samples. The percentage of debris present in cell suspensions tended to increase with decreasing needle diameter. Neither horse nor passage had a significant effect on viability.Cell damage is more likely when MSCs are passed through 25-ga rather than 20-ga needles.Use of needles larger than 25-ga is recommended to maintain the viability of MSCs injected in horses.

Abstract

Purpose: Bone marrow concentrate (BMC) and platelet-rich plasma (PRP) are used extensively in regenerative medicine. The aim of this study was to determine differences in the cellular composition and cytokine concentrations of BMC and PRP and to compare two commercial BMC systems in the same patient cohort.Methods: Patients (29) undergoing orthopaedic surgery were enrolled. Bone marrow aspirate (BMA) was processed to generate BMC from two commercial systems (BMC-A and BMC-B). Blood was obtained to make PRP utilizing the same system as BMC-A. Bone marrow-derived samples were cultured to measure colony-forming units, and flow cytometry was performed to assess mesenchymal stem cell (MSC) markers. Cellular concentrations were assessed for all samples. Catabolic cytokines and growth factors important for cartilage repair were measured using multiplex ELISA.Results: Colony-forming units were increased in both BMCs compared to BMA (p < 0.0001). Surface markers were consistent with MSCs. Platelet counts were not significantly different between BMC-A and PRP, but there were differences in leucocyte concentrations. TGF-β1 and PDGF were not different between BMC-A and PRP. IL-1ra concentrations were greater (p = 0.0018) in BMC-A samples (13,432 pg/mL) than in PRP (588 pg/mL). The IL-1ra/IL-1β ratio in all BMC samples was above the value reported to inhibit IL-1β.Conclusions: The bioactive factors examined in this study have differing clinical effects on musculoskeletal tissue. Differences in the cellular and cytokine composition between PRP and BMC and between BMC systems should be taken into consideration by the clinician when choosing a biologic for therapeutic application.Level Of Evidence: Clinical, Level II. 38. Fetal ultrasonography of the distal limb: a new tool to assess equine fetal age and bone development.

Abstract

In Quarter Horses (QH), fetal age can be predicted within 2 weeks prior to 200 days of gestation via ultrasonography, but as pregnancy advances, accuracy decreases (Renaudin.et.al. Reprod Dom Anim. 2022;57: 743-753). Recently, fetal ultrasonographic measurement of the first phalanx (P1) length was introduced as a novel biometric parameter during late gestation, and the time of apparition and closure of P1's epiphyses were found to be potential markers of bone maturation in utero (Renaudin.et.al. AAEP.2021;67:264-265). The aim of this study was to determine if bone characteristics of the second phalanx (P2), distal metacarpal 3 (MC3), proximal and distal sesamoid can predict fetal age. Ten healthy pregnant mares (9 QH and 1 Thoroughbred carrying a QH fetus) with known ovulation dates were used. Fetuses were examined by transrectal ultrasound every 2 weeks from 9 months of gestation until parturition. At each examination, the length of the ossified portion of P2's diaphysis was measured, the presence or absence of P2's and distal MC3's epiphyses and the time of appearance of the proximal and distal sesamoid bones were documented. P2 length correlated strongly with days of gestation (P < 0.0001; r 2 = 0.75) and a correlation equation was established: y = 0.1413x -24.89 (y = predicted value of P2 length; x = fetal age). The P2 proximal epiphysis and MC3 distal epiphysis appeared on ultrasound between 285 and 306 days of gestation (mean: 294 days),and 260 and 294 days of gestation (mean: 268 days) respectively and did not close before parturition. P2 distal epiphysis was never imaged. Proximal and distal sesamoids were first observed between 286 and 306 days of gestation (mean: 295 days) and 300 and 329 days of gestation (mean: 317 days) respectively except for the smallest foal where they appeared at 331 and 335 days respectively. In conclusion, P2 length can be used in addition to P1 length as a biometric parameter in late gestation. Distal MC3's, P1's and P2's epiphyses, and sesamoid bones diaphyses appear in a chronological order with MC3 distal epiphysis appearing first, P1's second, P2's third, proximal sesamoids fourth and distal sesamoid last. Distal limb ossification can be considered complete in QHs when the MC3 distal epiphysis, P1 and P2 proximal epiphysis and all sesamoid bones are present. Clinically, fetal ultrasonography of the distal limb may be used to assess fetal age and in the decision-making process of not inducing parturition if in utero bone development is not complete. Abstract

Breast cancer is the most common cancer in women living in Australia, accounting for 27% of all diagnoses in 2007. It was also the leading cause of burden of disease for females in 2010. Previous research has highlighted the utility of cancer rehabilitation programs for a range of tumour streams including breast cancer. The aim of cancer rehabilitation is to improve physical, social and psychological outcomes during and after treatment. Based on the end of treatment program for early stage breast cancer 'Yesterday, Today and Tomorrow' conducted at Cabrini Health, a private hospital in Melbourne, a pilot breast cancer rehabilitation program has been developed to address the rehabilitation needs of women with early stage breast cancer. The program will function as an adjunct to the existing rehabilitation service and has a physical reconditioning and psycho-education component. In this paper the establishment of the program will be described and data on the outcomes of the first three months presented.

44. Subcutaneous dextrose for rehydration of elderly patients - An evidence-based review.

Abstract

Background: In the Rehabilitation and Aged Care Services Program at Southern Health in Victoria, saline hypodermoclysis is a relatively common method of rehydration. However, there were questions about the safety and effectiveness of subcutaneous infusion of other fluids and, in particular, dextrose solutions. This review aimed to assess the safety and effectiveness of rehydration of elderly patients with subcutaneous 5% dextrose solutions compared with intravenous 5% dextrose solutions. Method(s): The Cochrane Library, Medline, IDIS, CINAHL, Current Contents, Premedline, Australasian Medical Index, the Joanna Briggs Institute, the US National Guideline Clearinghouse and bibliographies of retrieved articles. Searching was undertaken in July 2003. Studies selected were primary studies (or systematic reviews of primary studies) providing evidence as to the effectiveness and safety of subcutaneous infusion of dextrose solutions for rehydration of elderly patients. We included articles published in English in the last 10 years. Data were extracted by a single researcher. Result(s): From our search we identified 15 potentially relevant articles. We obtained the full text of these articles to determine their relevance. After application of the inclusion criteria, four articles remained for appraisal including one systematic review, two randomised controlled trials and one cohort study. Conclusion(s): The four studies appraised all provide evidence that appropriate volumes of subcutaneous dextrose infusions (in the form of half-normal saline-glucose 5%, 40 g/L dextrose and 30 mmol/L NaCl, or 5% dextrose solution and 4 g/L NaCl, or two-thirds 5% glucose and one-third normal saline) can be used effectively for the treatment of dehydration, with similar rates of adverse effects to intravenous infusion. The evidence in this area is limited, and larger randomised controlled trials using validated outcome measures would be useful to confirm these results.

Abstract

We have undertaken a study on 200 autopsy lung samples collected from subjects who lived in the Rome urban area and were not occupationally exposed to mineral dusts. The samples belonged to subjects who died aged between 15 and 65, both male and female. Subjects suffering from diseases and drug addicts were excluded. The purpose of this investigation was to determine whether any correlation existed between subjects' life-style, in particular smoking habits, and the presence of mineral fibres and dusts in their lungs. The data obtained were compared with those on airborne mineral dusts in the environment of the subjects themselves, particularly data on the concentration and types of mineral fibres present in that environment.

Abstract

The ultrastructural expression of laminin and fibronectin was studied in human colonic mucosa with a pre-embedding immunoperoxidase technique. Both antigens were present in epithelial basement membrane. The lamina densa was uniformly and homogeneously immunostained by both fibronectin and laminin, while the lamina rara showed discrete, positive sites occasionally connecting the lamina densa with the epithelial plasma membrane. In the lamina reticularis, fibronectin was diffusely positive, while laminin showed a discontinuous positivity only in the uppermost region in vicinity of the lamina densa. Collagen fibrils were regularly decorated by fibronectin-positive material. Basement membranes in the capillary wall showed a similar pattern of reactivity both with laminin and fibronectin. These results point out that laminin as well as fibronectin are integral components of intestinal mucosa basement membranes.
