The changes of cortisol, red blood cells (RBC), hemoglobin (Hb), hematocrit (Hct), heart rate (HR), respiratory rate (RR) and rectal temperature (RT) were evaluated after a reining training session in eight Quarter Horses. All parameters were measured before exercise (T0), immediately after exercise (T1), 1 h after exercise (T2), 2 h after exercise (T3) and 24 h after exercise (T4). One-way analysis of variance (ANOVA) for repeated measures, followed Bonferroni's post hoc test, showed a significant effect of the reining training session (P0.0001) on cortisol, RBC, Hb, Hct, HR, RR and RT. Simple linear regression analysis showed the positive correlation (P0.05) between cortisol changes and variations of studied parameters in T1, T3 and T4. Exercise-induced cortisol concentrations reflect the physiological response of reining training, suggesting that the changes observed are useful to assess the performance in reining horses and their reining training adaptability.

The purpose of this study was to determine haematological, haematochemical, and electrophoretic reference values for the Girgentana goat in order to form a basis for clinical interpretation. The study included 348 female Girgentana goats aged from 1 to 6 years. The animals were divided into 3 groups: goats aged 1-2 years (group A), goats aged 3-4 years (group B), and goats aged 5-6 years (group C). Blood samples were collected from each animal every morning at the same time (0800) by means of jugular venipuncture, and red blood cell (RBC), white blood cell (WBC), neutrophil (NEU), lymphocyte (LYM), monocyte (MONO), eosinophil (EOS), and basophil (BASO) counts, and haemoglobin (Hb), packed cell volume percent (PCV), mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH), mean corpuscular haemoglobin concentration (MCHC), and red cell distribution width (RDW) were recorded. Additionally, the concentration of some haematochemical parameters, including alanine-aminotransferase (ALT), aspartate-aminotransferase (AST), g-glutamine-transferase (g-GT), creatinine, b-hydroxybutyrate, urea, non-esterified fatty acids (NEFA), potassium, sodium, and chloride, and the electrophoretic profile (albumin, a1-globulins, a2-globulins, b–globulins, g–globulins, total proteins and albumin:globulins ratio) were determined. Statistical analysis (one-way analysis of variance [ANOVA]), followed by Bonferroni’s test, showed that there were significant differences only in RBC, MCV, EOS, ALT, AST, creatinine, g-GT, sodium, urea, NEFA, g–globulins, and the albumin:globulins ratio (P < 0.05 was considered statistically significant). The values determined in the present study contribute to the knowledge of the reference ranges for the Girgentana goat and can be used for monitoring health and disease diagnosis.

Adipose tissue as a stem cell source is ubiquitously available and has several advantages compared to other sources, for example it is easily accessible in large quantities with minimal invasive harvesting procedure, and isolation of adipose-derived mesenchymal stem cells (MSCs) yields a high amount of stem cells, essential for stem cell-based therapies and tissue engineering. We have explored the effect of donor age, and the anatomical origin of the adipose tissue on several aspects of MSCs in dogs, such as cell yield, proliferative ability, multi-differentiation potential, colony-forming capacity, stemness marker expression. We also assessed the effect of cell passaging on the MSCs stemness. We found that the anatomical origin of the adipose tissue and the age of donors have effects only on the proliferative capacity of the MSCs. Moreover, cells show a progressive loss of the stemness characteristics with passages. Cell therapies need a suitable number of cells to use in clinical applications. Characterization of MSCs at different passages, allowed us to demonstrate that, under our culture conditions, the best quantitative and qualitative characteristics are obtained at early passages. Adult MSCs are of particular interest for the therapeutic approach to musculoskeletal diseases, and the dog provides an excellent preclinical model for the development of new approaches in regenerative medicine that might be applied to humans.

The aim of the present study was to measure the serum total proteins and the serum protein fractions (albumin, α1-, α2-, β-, and γ- globulins) of ten newborn lambs (Comisana breed) during the first 30 days of life in order to obtain useful information for neonatal care. From each animal, blood samples were collected via jugular venipuncture at the same hour (9.00) every 3 days for 30 days starting from birth (day 0). The concentrations of serum total proteins and albumin, α1-, α2-, β-, γ- globulins, and Albumin/Globulins (A/G) ratio were determined using an automated electrophoresis system. The aim of this study was to evaluate the influence of different physiological phases on serum total proteins and their fractions of ten Comisana ewes housed in Mediterranean area. From each animal, blood samples were collected at different physiological phases: late pregnancy, post-partum, early, mid-, end lactation and dry period. On all samples serum total proteins were determined by the biuret method, and albumin, α-globulins, β(1) -globulins, β(2) -globulins and γ-globulins concentrations were assessed using an automated system. One-way repeated measures analysis of variance was applied to determine the significant effect of different physiological phases on the parameters studied. During the late pregnancy and post-partum, total proteins, β1- and β2-globulins and γ-globulins showed the highest values. Starting from post-partum, α-globulins increased to reach their peaks in mid-lactation. Early lactation was characterized by low γ-globulins values. The increase in serum albumin concentration and the drop in some globulin fractions determined the significant increase in albumin/globulin ratio. The obtained results contributed to improve the knowledge on electrophoretic profile during the different physiological phases in ewes, confirming that pregnancy and lactation periods affect the protein metabolism. Particularly, serum protein fractions pattern could give information about dehydration, plasma volume expansion and hepatic function, which occur during the different physiological phases. Dynamics of the protein profile - from pregnancy to dry period - which are provided by our results, could be considered as guidelines for the management strategies to guarantee the nutritional needs of these animals during the different physiological phases and to avoid a decline of productive performance and consequently an economic loss.

The aim of this study was to assess the effect of the different aquatic conditions on anion transport of fish erythrocytes through the measurement of the sulphate uptake operating from band 3, the determination of reduced glutathione (GSH) and oxidized glutathione (GSSG). To this purpose, blood samples of 30 Liza aurata and 30 Salmo irideus fishes were collected via caudal vein and washed with physiological buffer. Successively, erythrocytes suspended at 3% haematocrit were used to measure the SO(4) (=) influx by atomic absorption spectrophotometry at 425 nm wavelength and the GSH concentration using an immunodiagnostic assay intended for the quantitative determination of glutathione in ethylenediaminetetraacetic acid in blood. All results were analyzed using unpaired Student's t-test and a P0.05 was considered statistically significant. The results of this study showed statistically significant differences about the sulphate uptake and the glutathione levels in S. irideus with respect to L. aurata (P0.0001). In conclusion, the different aquatic conditions play an important role on anion transport in fish erythrocytes, showing that environmental differences induce perturbations in erythrocyte membrane and should be evaluated to prevent physiological damages in fishes.

In this study we investigated the influence of body mass on the daily rhythm of total motor activity in ponies and horses. Six clinically healthy Shetland pony breed with a body mass of 180 ± 30 kg and six healthy horses Sella Italiana breed with a body mass of 530 ± 20 kg were used. Animals were equipped with actigraphy-based data loggers, Actiwatch-Mini® to record total activity. The application of two-way ANOVA showed a highly significant effect of time of day and body mass on motor activity. Cosinor analysis identified the periodic parameters and their acrophases during the two days of monitoring. Mesor and amplitude were statistically higher in horses than in ponies and no statistically significant differences was found about acrophases and robustness. In conclusion we suppose that the difference in the amount of total motor activity, in animals that differ in breed, as pony and horse, may lie in physical differences. En este estudio hemos investigado la influencia de la masa corporal con respecto al ritmo circadiano de toda la actividad motora en los ponis y los caballos. Han sido utilizados seis sanos Shetland poni con una masa corporal de 180 ± 30 kg y seis sanos caballos biotipo Sella Italiana con una masa corporal de 530 ± 20 kg clínicamente sanos. Los animales han sido equipados con registrador de datos Actiwatch-Mini® para grabar toda la actividad motora. La aplicación de ANOVA dos vías mostró un efecto altamente significante de la hora del día y de la masa corporal sobre la actividad motora. El análisis del Cosinor ha identificado los parámetros periódicos y ellos acrofases en los dos días de seguimiento. Mesor y amplitud fueron estadísticamente más alto en los caballos que en los ponis y ninguna significativa diferencia estadística se encontró sobre las acrofases y robustez. En conclusión suponemos que las diferencias sobre la cantidad de toda la actividad motora en animales de diferente biotipo, como poni y caballo, podrían ser diferencias físicas.

The aim of the present study was to evaluate the effect of shearing on physiological and oxidative stress parameters in ewes. Twenty Comisana ewes were used and divided into two groups. Ten ewes were left unshorn as a control group (Group A) and 10 ewes were shorn (Group B). All measurements were taken before and after shearing, and repeated 8 h after shearing and 1, 2, 3, 4, 5, 10 and 15 days after shearing. Reactive oxygen species (dROMs), antioxidant barrier (oxy-adsorbent), thiol antioxidant barrier (SHp) and packed cell volume (PCV) were assessed in blood samples collected by means of jugular venipuncture. Rectal temperature (RT), respiratory rate (RR) and heart rate (HR) were also measured. Two-way repeated measures analysis of variance (ANOVA), followed by Bonferroni's test, was used for the assessment of significant effects due to shearing and time. The statistical analysis showed significant increases (P0.01) of dROMs, oxy-adsorbent, SHp, and a significant decrease (P0.01) of RT and RR associated with time and shearing. Our results indicate that shearing causes a change in the ewe's homeostatic balance that leads to oxidative stress.

To evaluate the metabolic profile of pregnant and non-pregnant ewes with high or low degree of behavioral lateralization, we used 57 clinically healthy Sardinian ewes. They were divided into three groups: Group A (nonpregnant ewes), Group B (pregnant ewes with one fetus) and Group C (pregnant ewes with twin fetuses). Using behavioral tests, in all groups lateralization was established and ewes were distinguished into ewes with high and low level of lateralization. Every 30 days for 4 months (at 35, 76, 103 and 145 days of gestation), blood samples were collected and total cholesterol, triglycerides, nonesterified fatty acids (NEFA), glucose, insulin, creatinine, urea, uric acid, total protein, total bilirubin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), γ-glutamine transferase (γ-GT), β-hydroxybutyrate (β-OHB), chloride (Cl), potassium (K), sodium (Na) and calcium (Ca) were assessed. The repeated measures multifactor analysis of variance (MANOVA) showed statistical differences of time, pregnancy and degree of lateralization on some studied parameters. These results confirm that pregnancy represents a very important metabolic moment and show the effect of the degree of behavioral lateralization on the assessment of metabolic parameters in non-pregnant and pregnant ewes. Further studies are necessary to value the influence of these factors on other parameters such as hematological parameters and hormones.
