2. Associations of Alcohol Consumption With Left Atrial Morphology and Function in a Population at High Cardiovascular Risk

- Abstract
Excessive alcohol consumption has been associated with increased risk of atrial fibrillation, although the underlying mechanisms remain unclear. An enlarged left atrium and impaired left atrial function may lead to atrial fibrillation. The association of alcohol consumption with structural and functional left atrial measures, however, has received limited attention.We studied 503 participants from the PREDIMED-Plus (Prevención con Dieta Mediterránea) trial, a randomized trial testing intensive weight loss intervention with an energy-reduced Mediterranean diet and physical activity promotion in preventing cardiovascular disease in adults with metabolic syndrome. Participants underwent transthoracic echocardiography at baseline, year 3, and year 5 of the study. Outcomes of interest included volume index and reservoir, conduit, and contractile strains of the left atrium. Alcohol consumption was calculated through food frequency questionnaires and presented as drinks consumed per day. Multiple linear regression and mixed models estimated the association of alcohol consumption with left atrial measurements at baseline and through follow-up. Cross-sectionally, higher alcohol consumption (per 1 drink/day increases) was associated with larger left atrial volume (0.65 mL/m2 [95% CI, 0.18-1.11]) and lower left atrial reservoir and contractile strain (-0.44% [95% CI, -0.87 to -0.01]; and -0.44% [95% CI, -0.75 to -0.14]). Baseline alcohol consumption was not associated with changes in left atrial measurements, but increases in alcohol consumption (per 1 drink/day increase) during follow-up were associated with left atrial enlargement (0.71 mL/m2 [95% CI, 0.17-1.26]).In a population at high cardiovascular risk, increased alcohol consumption was associated with left atrial enlargemen

Many ongoing clinical trials for DLBCL patients involve microtubule targeting agents (MTAs), however, their anticancer activity is limited by severe side effects. Therefore, we chose to improve the therapeutic window of the MTA monomethyl auristatin E developing a nanoconjugate, T22-AUR, that selectively targets the CXCR4 receptor, which is overexpressed in many DLBCL cells (CXCR4+) and associated with poor prognosis.Methods: The T22-AUR specificity towards CXCR4 receptor was performed by flow cytometry in different DLBCL cell lines and running biodistribution assays in a subcutaneous mouse model bearing CXCR4+ DLBCL cells. Moreover, we determined T22-AUR cytotoxicity using cell viability assays, cell cycle analysis, DAPI staining and immunohistochemistry. Finally, the T22-AUR antineoplastic effect was evaluated in vivo in an extranodal CXCR4+ DLBCL mouse model whereas the toxicity analysis was assessed by histopathology in non-infiltrated mouse organs and by in vitro cytotoxic assays in human PBMCs.Results: We demonstrate that the T22-AUR nanoconjugate displays CXCR4-dependent targeting and internalization in CXCR4+ DLBCL cells in vitro as well as in a subcutaneous DLBCL mouse model. Moreover, it shows high cytotoxic effect in CXCR4+ DLBCL cells, including induction of G2/M mitotic arrest, DNA damage, mitotic catastrophe and apoptosis. Furthermore, the nanoconjugate shows a potent reduction in lymphoma mouse dissemination without histopathological alterations in non-DLBCL infiltrated organs. Importantly, T22-AUR also exhibits lack of toxicity in human PBMCs.Conclusion: T22-AUR exerts in vitro and in vivo anticancer effect on CXCR4+ DLBCL cells without off-target toxicity. Field emission scanning electron microscopy (FE-SEM) and X-ray diffraction (XRD) have been used to study the microstructural changes and phase development that take place during the hydration of cubic (pure) and orthorhombic (Na-doped) tricalcium aluminate (C3A) and gypsum in the absence and presence of lime. The results demonstrate that important differences occur in the hydration of each C3A polymorph and gypsum when no lime is added; orthorhombic C3A reacts faster with gypsum than the cubic phase, forming longer ettringite needles; however, the presence of lime slows down the formation of ettringite in the orthorhombic sample. Additional rheometric tests showed the possible effects on the setting time in these cementitious mixes.

- Abstract
The Nature Futures Framework (NFF) is a heuristic tool for co-creating positive futures for nature and people. It seeks to open up a diversity of futures through mainly three value perspectives on nature – Nature for Nature, Nature for Society, and Nature as Culture. This paper describes how the NFF can be applied in modelling to support decision-making. First, we describe key considerations for the NFF in developing qualitative and quantitative scenarios: i) multiple value perspectives on nature as a state space where pathways improving nature toward a frontier can be represented, ii) mutually reinforcing key feedbacks of social-ecological systems that are important for nature conservation and human wellbeing, iii) indicators of multiple knowledge systems describing the evolution of complex social-ecological dynamics. We then present three approaches to modelling Nature Futures scenarios in the review, screening, and design phases of policy processes. This paper seeks to facilitate the integration of relational values of nature in models and strengthen modelled linkages across biodiversity, nature’s contributions to people, and quality of life.

10. Multicausal etiology of anemia among women of reproductive age in Vietnam

BACKGROUND/OBJECTIVES: Anemia is a significant public health concern, especially among women and young children. An improved understanding of the complex etiology of anemia is crucial for developing appropriate prevention strategies. This paper examines the determinants of anemia in a large sample of Vietnamese women of reproductive age (WRA). METHODS: We included baseline data from 4986 WRA participating in a randomized controlled trial (PRECONCEPT). Hemoglobin (Hb) concentrations were measured with Hemocue. Plasma ferritin (Fe), retinol binding protein (RBP) and markers of inflammation were assessed using the ELISA technique. We used multivariate logistic regression to describe associations with anemia and structural equation modeling (SEM) to characterize direct and indirect pathways influencing Hb concentrations. RESULTS: Prevalence of anemia, iron deficiency (Fe < 12 µg/l), insufficient iron stores (Fe < 30 µg/l) and iron deficiency anemia was 19.7, 3.5, 14.4 and 1.9%, respectively. Ferritin concentration (0.29 per log-mg/dl), being an ethnic minority (-0.24 compared with Kinh), number of children (-0.17) and socioeconomic status (0.09) were directly associated with Hb concentration (P < 0.05). Similarly, RBP was directly (0.27 per mg/dl) associated with Hb and also indirectly (0.09 mg/dl) with ferritin. Hookworm infection was indirectly associated with Hb (-0.11) through RBP and ferritin. CONCLUSION: These findings illustrate the complex etiology of anemia and provide a useful framework for designing, targeting and evaluating appropriate strategies for the prevention and control of anemia. Contrary to expectations, iron deficiency accounted for a very small proportion of anemia in Northern Vietnam. Loco-regional recurrences and distant metastases represent the main cause of head and neck squamous cell carcinoma (HNSCC) mortality. The overexpression of chemokine receptor 4 (CXCR4) in HNSCC primary tumors associates with higher risk of developing loco-regional recurrences and distant metastases, thus making CXCR4 an ideal entry pathway for targeted drug delivery. In this context, our group has generated the self-assembling protein nanocarrier T22-GFP-H6, displaying multiple T22 peptidic ligands that specifically target CXCR4. This study aimed to validate T22-GFP-H6 as a suitable nanocarrier to selectively deliver cytotoxic agents to CXCR4+ tumors in a HNSCC model. Here we demonstrate that T22-GFP-H6 selectively internalizes in CXCR4+ HNSCC cells, achieving a high accumulation in CXCR4+ tumors in vivo, while showing negligible nanocarrier distribution in non-tumor bearing organs. Moreover, this T22-empowered nanocarrier can incorporate bacterial toxin domains to generate therapeutic nanotoxins that induce cell death in CXCR4-overexpressing tumors in the absence of histological alterations in normal organs. Altogether, these results show the potential use of this T22-empowered nanocarrier platform to incorporate polypeptidic domains of choice to selectively eliminate CXCR4+ cells in HNSCC. Remarkably, to our knowledge, this is the first study testing targeted protein-only nanoparticles in this cancer type, which may represent a novel treatment approach for HNSCC patients. © 2022 Chinese Pharmaceutical Association and Institute of Materia Medica, Chinese Academy of Medical Sciences

Preventive and modelling approaches to address the COVID-19 pandemic have been primarily based on the age or occupation, and often disregard the importance of heterogeneity in population contact structure and individual connectivity. To address this gap, we developed models based on Erdős-Rényi and a power law degree distribution that first incorporate the role of heterogeneity and connectivity and then can be expanded to make assumptions about demographic characteristics. Results demonstrate that variations in the number of connections of individuals within a population modify the impact of public health interventions such as lockdown or vaccination approaches. We conclude that the most effective strategy will vary depending on the underlying contact structure of individuals within a population and on timing of the interventions.Author summaryThe best strategy for public health interventions, such as lockdown or vaccination, depends on the contact structure of the population and the timing of the intervention. In general, for heterogeneous contact structures that mimic the COVID-19 spread, which is characterized by the presence of super spreaders, vaccinating the most connected individuals first was the most effective strategy to prevent infections and deaths, especially when coupled to serological tests. Models considering heterogeneity in human interactions need be used to identify the best potential vaccine prioritization strategies.

CXC chemokine receptor 4 (CXCR4) is a cell surface marker associated with several severe human pathologies, including colorectal cancer, for which intracellular targeting agents are currently missing.Results: Four different peptides that bind CXCR4 were tested for their ability to internalize a green fluorescent protein-based reporter nanoparticle into CXCR4+ cells. Among them, only the 18 mer peptide T22, an engineered segment derivative of polyphemusin II from the horseshoe crab, efficiently penetrated target cells via a rapid, receptor-specific endosomal route. This resulted in accumulation of the reporter nanoparticle in a fully fluorescent and stable form in the perinuclear region of the target cells, without toxicity either in cell culture or in an in vivo model of metastatic colorectal cancer.Conclusion: Given the urgent demand for targeting agents in the research, diagnosis, and treatment of CXCR4-linked diseases, including colorectal cancer and human immunodeficiency virus infection, T22 appears to be a promising tag for the intracellular delivery of protein drugs, nanoparticles, and imaging agents.Keywords: peptide tag, CXCR4, intracellular targeting, self-assembling, nanoparticles, colorectal cancer

- Abstract
Background: Results from retrospective studies suggest that use of neuromuscular blocking agents during general anaesthesia might be linked to postoperative pulmonary complications. We therefore aimed to assess whether the use of neuromuscular blocking agents is associated with postoperative pulmonary complications. Methods: We did a multicentre, prospective observational cohort study. Patients were recruited from 211 hospitals in 28 European countries. We included patients (aged ≥18 years) who received general anaesthesia for any in-hospital procedure except cardiac surgery. Patient characteristics, surgical and anaesthetic details, and chart review at discharge were prospectively collected over 2 weeks. Additionally, each patient underwent postoperative physical examination within 3 days of surgery to check for adverse pulmonary events. The study outcome was the incidence of postoperative pulmonary complications from the end of surgery up to postoperative day 28. Logistic regression analyses were adjusted for surgical factors and patients' preoperative physical status, providing adjusted odds ratios (ORadj) and adjusted absolute risk reduction (ARRadj). This study is registered with ClinicalTrials.gov, number NCT01865513. Findings: Between June 16, 2014, and April 29, 2015, data from 22 803 patients were collected. The use of neuromuscular blocking agents was associated with an increased incidence of postoperative pulmonary complications in patients who had undergone general anaesthesia (1658 [7·6%] of 21 694); ORadj 1·86, 95% CI 1·53–2·26; ARRadj −4·4%, 95% CI −5·5 to −3·2). Only 2·3% of high-risk surgical patients and those with adverse respiratory profiles were anaesthetised without neuromuscular blocking agents. The use of neuromuscular monitoring (ORadj 1·31, 95% CI 1·15–1·49; ARRadj −2·6%, 95% CI −3·9 to −1·4) and the administration of reversal agents (1·23, 1·07–1·41; −1·9%, −3·2 to −0·7) were not associated with a decreased risk of postoperative pulmonary

Les Pyrénées ne comptent que quatre sites établis autour de 1 000 m qui peuvent témoigner de l’occupation de l’étage montagnard avant le Néolithique ancien. À partir des vallées d’Andorre et sur le flanc sud de la chaîne orientale, les abris de Guilanya et Margineda permettent d’analyser les mutations environnementales et les évolutions dans la culture matérielle et spirituelle des groupes de chasseurs-collecteurs de la fin du Tardiglaciaire à l’Holocène ancien. Au côté d’originalités culturelles (incursion d’Aziliens du versant nord pendant l’Alleröd, Sauveterriens sans microlithes cantonnés dans les Pyrénées sèches), les innovations techniques bien cernées en stratigraphie sont plus précocement attestées au cœur de la chaîne axiale qu’en bordure du littoral et dans les piémonts où certains décalages techno-typologiques, tel l’Épimagdalénien, ont été individualisés. In the Pyrenees, mountain settlements higher than 1000 m are scarce before the Early Neolithic. Balma Guilanyà and Balma Margineda, rock-shelters located in the south-eastern face of the Pyrenees, allow analyzing the ecological and the hunter-gatherer adaptations along the limit Late Glacial/Early Holocene. In this area, some cultural processes (like the sudden apparition of the Azilian from the north Pyrenees in the Allerød, or the development of a Sauveterrien stage without standardized microlithic artifacts in the south Pyrenees) suggest that these technical innovation are detected sooner in the core of the mountain chain than in peripheral areas as the piedmont or the coast, where some techno-typological endurances (Epimagdalenian) has been recognized.

Oligonucleotide-protein conjugates have important applications in biomedicine. Simple and efficient methods are described for the preparation of these conjugates. Specifically, we describe a new method in which a bifunctional linker is attached to thiol-oligonucleotide to generate a reactive intermediate that is used to link to the protein. Having similar conjugation efficacy compared with the classical method in which the bifunctional linker is attached first to the protein, this new approach produces significantly more active conjugates with higher batch to batch reproducibility. In a second approach, direct conjugation is proposed using oligonucleotides carrying carboxyl groups. These methodologies have been applied to prepare nanoconjugates of an engineered nanoparticle protein carrying a T22 peptide with affinity for the CXCR4 chemokine receptor and oligomers of the antiproliferative nucleotide 2'-deoxy-5-fluorouridine in a very efficient way. The protocols have potential uses for the functionalization of proteins, amino-containing polymers or amino-lipids in order to produce complex therapeutic nucleic acid delivery systems.

- Abstract
Aquest doctorat. El projecte cobreix les investigacions sobre aïllants topològics (TI) de la família Bi2Se3 amb diferents defectes i l'estudi d'efectes de proximitat de TI a la heteroestructura de grafè amb TI. La primera part d'aquest projecte se centra principalment en l'efecte del desordre en les propietats electròniques de TI amb gruix ultrafí ( 3 nm). S'ha trobat que la manca de coincidència de rotació entre capes quíntuples de TI pot augmentar el "gap" de volum dels TI però preservar la textura d'espín tipus Rashba en l'estat de la superfície; mentre que la hidrogenació en una superfície de TI pot ajudar a reduir l'efecte túnel quàntic i tancar el "gap" de superfície en el punt Γ amb la textura d'espín tipus Rashba per a la pel·lícula TI ultrafina. A més, aquest esquema també pot crear un altre punt Dirac (DP) en el punt M amb textura de spin tipus Dresselhaus. La segona part del projecte investiga els efectes de proximitat de TI dins de la heteroestructura de grafè / TI i el DP en el grafè es plega des del punt K / K 'a Γ punt de la zona Brillouin, a causa del plegament de la banda, trobant que l'alineació entre el substrat de TI i el grafè té un paper clau en la formació de l'estructura de la banda i la textura d'espín del grafè. La configuració d'apilament "hollow" podria induir la distorsió d'unió de Kekulé a la capa de grafè, donant com a resultat l'engrandiment del "gap" (3.2 meV) i el Rashba SOC, el que dóna com a resultat la precessió d'espín propera al Γ punt . A més, aquesta textura atípica de Rashba espín fa que la component d'espín fora del pla disminueixi gradualment a mesura que el punt k s'allunya del punt Γ, el que porta a la anisotropia de gir a la capa de grafè. D'altra banda, la configuració d'apilament "bridge" o "top" podria portar l'evident divisió de la banda en direcció lateral, que podria ser l'origen de l'efecte Edelstein en la capa de grafè; no obstant, no hi ha una anisotropia d'espín evident en aquesta configuració. The first part of this project mainly focuses on the effect of disorder on the electronic properties of TI with ultrathin thickness ( 3 nm). It was found that rotation mismatch between quintuple of TI can enlarge the bulk gap of TI but preserve the Rashba type spin texture on the surface state; while, the hydrogenation on one TI surface can help reduce the quantum tunneling effect and close the surface gap at Γ point with Rashba type spin texture for ultrathin TI film. Furthermore, this scheme can also create another Dirac point (DP) at M point with Dresselhaus type spin texture. The second part of the project investigates in the proximity effects of TI within the heterostructure of graphene/TI and the DP on graphene is folded from K/K' point to Γ point in Brillouin zone, due to the band folding, and it was found that the alignment between TI substrate and graphene played the key role in forming the band structure and the spin texture of graphene. Hollow configuration could induce the Kekulé bonding distortion to graphene layer, mainly resulting in the enlarged gap (3.2 meV), and the Rashba SOC, resulting in the spin precession close to the Γ point. Furthermore, this atypcial Rashba spin texture has the out-of-plane spin component decrease gradually as the k point moves away from the Γ point, leading to the spin anisotropy on graphene layer. While, the bridge or the top configuration could bring the evident band splitting in lateral direction, which could be the origin of the Edelstein effect in graphene layer; however, there is no evident spin anisotropy in such configuration. 41. Axtrell, a new CV3 chondrite find from Texas

We describe a previously unreported meteorite found in Axtell, Texas, in 1943. Based on the mineralogical composition and texture of its matrix and the sizes and abundance of chondrules, we classify it as a CV3 carbonaceous chondrite. The dominant opaque phase in the chondrules is magnetite, and that in refractory inclusions is Ni-rich metal (awaruite). Axtell, therefore, belongs to the oxidized subgroup of CV3 chondrites, although unlike Allende it escaped strong sulfidation. The meteorite bears a strong textural resemblance to Allende, and its chondrule population and matrix appear to be quite similar to those of Allende, but its refractory inclusions, thermoluminescence properties, and cosmogenic Co-60 abundances are not. Our data are consistent with a terrestrial age for Axtell of approximately 100 years and a metamorphic grade slightly lower than that of Allende.

42. Vulvar Basal Cell Carcinoma: four case reports with immunohistochemical study

Basal cell carcinomas (BCC) are the most frequent tumours in humans and normally appear in photoexposed areas of the skin. It is widely accepted that BCCs originate at follicular stem cells and consequently are very rare in nonhairy areas. Here, we report 4 cases of vulvar BCC, 3 of which were located in a vulvar semimucous area, a nonphotoexposed area, and a nonhairy area. We have determined the CK7 and CK19 profile of all cases; both are markers of simple epithelium with glandular differentiation. Interestingly, all cases were positively stained for CK7 and CK19. Considering that the vulvar region is rich in sebaceous and apocrine units, we hypothesise a glandular origin of BCCs situated in the vulvar region.

A petrographic and chemical study of compact Type A CAI's from the Efremovka CV3 chondrite strongly suggests that the opaque assemblages (OA's) that they contain were molten at temperatures below the CAI silicate solidus, and that the V-rich magnetite presently observed in association with OA's formed by in situ oxidation of their FeNi.

47. Metal-rich meteorites from the aubrite parent body

Three metal-rich meteorites - Mt. Egerton, Horse Creek, and LEW 88055 - were studied and it is suggested that they formed in the aubrite parent body. LEW 85369 and 88631 may also have a common origin, but these rocks have not yet been studied in detail. This body was probably heated to about 1600 C by a very strong heat source. While molten, metal agglomerated into sizeable nodules which never segregated efficiently to form a core, but were trapped in the silicate mantle. Different clasts and lithologies in aubrites solidified and cooled under local equilibrium conditions of oxygen fugacity, and with different thermal histories. Impacts mixed clasts from throughout the parent body, creating the typical aubrite breccias.

49. Origin and history of chondrite regolith, fragmental and impact-melt breccias from Spain

Six ordinary chondrite breccias from the Museo Nacional de Ciencias Naturales, Madrid (Spain), are described and classified as follows: the solar gas-rich regolith breccia Oviedo (H5); the premetamorphic fragmental breccias Cabezo de Mayo (type 6, L-LL), and Sevilla (LL4); the fragmental breccias Canellas (H4) and Gerona (H5); and the impact melt breccia, Madrid (L6). It is confirmed that chondrites with typical light-dark structures and petrographic properties typical of regolith breccias may (Oviedo) or may not (Canellas) be solar gas-rich. Cabezo de Mayo and Sevilla show convincing evidence that they were assembled prior to peak metamorphism and were equilibrated during subsequent reheating. Compositions of olivine and low-Ca pyroxene in host chondrite and breccia clasts in Cabezo de Mayo are transitional between groups L and LL. It is suggested, based on mineralogic and oxygen isotopic compositions of host and clasts, that the rock formed on the L parent body by mixing, prior to peak metamorphism. This was followed by partial equilibrium of two different materials: the indigenous L chondrite host and exotic LL melt rock clasts.

Based on optical microsocpy and electron microprobe analysis of mafic minerals, four previously poorly described ordinary chondrites from Spain are classified. The classifications of Guarena (H6), Olmedilla de Alarcon (H5) and Reliegos (L5) are confirmed. Molina is reclassified as H5, based on new data.
