Schistosomiasis mansoni is a debilitating neglected tropical disease caused by infection with the blood fluke, Schistosoma mansoni . Development of the larval stage of the parasite in an intermediate host snail of the genus Biomphalaria is an obligatory component of the life cycle. Enhanced understanding of the mechanism(s) of host defense in the intermediate host snail may hasten the development of new tools that block transmission of schistosomiasis. The embryonic cell line from B. glabrata , termed the Bge line, is a versatile resource in the investigation of snail-schistosome interactions. A key attribute of the Bge cell is its hemocyte-like phenotype, given the central role of the snail hemocyte in innate and cellular immunity. The allograft inflammatory factor 1 (AIF, aif ) is a conserved antigen typically expressed in phagocytic and granular leukocytes and is a marker of macrophage activation. AIF is highly expressed in isolates of B. glabrata that are resistant to infection with S. mansoni . We targeted the aif gene of B. glabrata ( Bg AIF) for programmed gene knockout using the CRISPR/Cas9 approach, to investigate the activity of CRISPR/Cas9 gene editing in this gastropod species and, in turn, to investigate the loss of AIF on Bge cell-mediated adherence to schistosome sporocysts. Bge cells were transformed with gene editing plasmid encoding the Cas9 nuclease driven by the CMV promoter and a guide RNA specific for exon 4 of the Bg AIF by square wave electroporation. Twelve independent transformations were carried out. Transcript levels of Bg AIF were significantly reduced by up to 73% (range, 26 to 73%; mean 49.5±20.2% S.D; n = 12) from two to nine days following transformation. Sanger direct sequencing and bioinformatic analysis of the sequence reads revealed on-target programmed mutation on the Bg AIF gene, including insertions of 1-2 bp and deletion of 8-30 bp, (range, 9 to 17% of Bg AIF gene; n =12). In addition, the adherence of gene-edited Bge cells to sporocysts was significantly impeded in comparison to control cells, as ascertained using the cell adherence index (CAI); 2.66±0.10 (control) vs . 2.30±0.22 (Δ Bg AIF cells), P < 0.05. This is the first report of gene editing in this medically important taxon of freshwater gastropods that are necessary for the transmission of schistosomiasis.

Snails of the family Lymnaeidae act as intermediate hosts in the biological cycle of Fasciola hepatica, which is a biological agent of fasciolosis, a parasitic disease of medical importance for humans and animals. The present work aimed to update and map the spatial distribution of the intermediate host snails of F. hepatica in Brazil. Data on the distribution of lymnaeids species were compiled from the Collection of Medical Malacology (Fiocruz-CMM, CPqRR), Collection of Malacology (MZUSP), “SpeciesLink” (CRIA) network and through systematic surveys in the literature. Our maps of the distribution of lymnaeids show that Pseudosuccinea columella is the most common species and it is widespread in the South and Southeast with few records in the Midwest, North and Northeast regions. The distribution of the Galba viatrix, G. cubensis and G. truncatula showed a few records in the South and Southeast regions, they were not reported for the Midwest, North and Northeast. In addition, in the South region there are a few records for G. viatrix and one occurrence of Lymnaea rupestris. Our findings resulted in the first map of the spatial distribution of Lymnaeidae species in Brazil which might be useful to better understand the fasciolosis distribution and delineate priority areas for control interventions. Moluscos da família Lymnaeidae são hospedeiros intermediários no ciclo biológico de Fasciola hepatica, agente etiológico da fasciolose, doença parasitária de importância médica para humanos e animais. O presente trabalho teve como objetivo datar e mapear a distribuição espacial dos hospedeiros intermediários de F. hepatica no Brasil. Os dados de distribuição das espécies de limneídeos foram obtidos das Coleções de Malacologia Medica (Fiocruz-CMM, CPqRR) e de Malacologia do Museu de Zoologia da Universidade de São Paulo (MZUSP), rede “SpeciesLink” (CRIA) e através de pesquisas sistemáticas na literatura. Os mapas de distribuição dos limneideos mostram que Pseudosuccinea columella é a espécie mais comum e distribuída nas regiões Sul e Sudeste com poucos registros nas regiões Centro-Oeste, Norte e Nordeste. Além disso, na região Sul os registros para G. viatrix são escassos e existe apenas um único relato de ocorrência para L. rupestris. Os dados obtidos resultaram no primeiro mapa de distribuição espacial das espécies de Lymnaeidae no Brasil, informação básica e relevante para estruturação de políticas de controle da fasciolose humana e animal.

INTRODUCTION: The increasing practice of ecotourism and rural tourism in the State of Minas Gerais, Brazil, highlights the importance of studies concerning the occurrence of potential intermediate hosts of Schistosoma mansoni. This study aimed to identify species of Biomphalaria snails in municipalities along the Estrada Real, an important Brazilian tourism project. METHODS: The specimens were collected in different water collections of 36 municipalities along the Estrada Real in the southeast of the State of Minas Gerais. Biomphalaria species were characterized using both morphological and molecular approaches. The research was conducted between August 2005 and September 2009 and all the sites visited were georeferenced using GPS. RESULTS: Six Biomphalaria species were found in 30 of the 36 municipalities studied: glabrata, tenagophila, straminea, peregrina, occidentalis and schrammi. The first three species of Biomphalaria, recognized as intermediate hosts of S. mansoni, were present in 33.3%, 47.2% and 8.3% of the municipalities studied, respectively. The mollusks were found in different types of water collections and no infection by S. mansoni was detected. The highest occurrence of Biomphalaria concentration was verified in the area covered by the Caminho Novo route (Diamantina/MG to Rio de Janeiro/RJ). CONCLUSIONS: Considering the occurrence of schistosomiasis in the State of Minas Gerais and the socioeconomic repercussions involved in the Estrada Real Project, this work focuses on the vulnerability of water collections due to the presence of Biomphalaria mollusks and emphasizes the need for epidemiological surveillance and sanitary and educational measures integrated with the local community and tourism sectors. INTRODUÇÃO: O aumento das práticas de ecoturismo e turismo rural, em Minas Gerais, Brasil, evidencia a importância de se realizarem estudos sobre a ocorrência de hospedeiros intermediários do Schistosoma mansoni, no estado. O presente trabalho objetivou a busca e identificação das espécies de caramujos Biomphalaria encontrados em municípios mineiros pertencentes à Estrada Real, um importante projeto de turismo brasileiro. MÉTODOS: Os moluscos foram coletados em 36 municípios da Estrada Real, no sudeste de Minas Gerais. A pesquisa foi realizada de agosto de 2005 a setembro de 2009 e todos os locais visitados foram georreferenciados com o uso de GPS. RESULTADOS: Dos 36 municípios estudados, 30 apresentaram a ocorrência de pelo menos uma entre as seis espécies de Biomphalaria: glabrata, tenagophila, straminea, peregrina, occidentalis e schrammi. As três primeiras espécies citadas, reconhecidas como hospedeiras intermediárias do S. mansoni, estavam presentes em 33,3%, 47,2% e 8,3% dos municípios estudados, respectivamente. Os moluscos foram encontrados em diferentes tipos de coleções hídricas e em nenhum deles foi detectada infecção pelo S. mansoni. Houve maior ocorrência de Biomphalaria na área referente ao Caminho Novo (Diamantina/MG ao Rio de Janeiro/RJ). CONCLUSÕES: Considerando-se a ocorrência da esquistossomose, no Estado de Minas Gerais, e as repercussões socioeconômicas que envolvem o projeto Estrada Real, este trabalho aponta para a vulnerabilidade das coleções hídricas devido à presença de moluscos Biomphalaria e enfatiza a necessidade de vigilância epidemiológica e medidas educativas e sanitárias integradas com a comunidade local e setores de turismo.

We compared the effects of larval and adult worm excretory-secretory (ES) products from hookworm on the proliferative responses and cytokine secretion in peripheral blood mononuclear cells (PBMCs) from hookwormpatients and egg-negative, nonendemic controls. When compared with negative controls, mitogen-stimulated PBMC from hookworm-infected individuals showed a significantly reduced proliferative response when adult worm ES antigen was added to the cultures. Furthermore, in hookworm-infected individuals a significant downmodulation of inflammatory interleukin (IL)-6 and tumor necrosis factor (TNF)-α secretion resulted when PBMCs were stimulated with mitogen in combination with larval or adult worm ES. Both, interferon (IFN)-γ and IL-10 secretion were significantly lower in stimulated PBMC from infected individuals; however the IFN-γ/IL-10 ratio was much lower in hookworm-infected patients. Comparable effects, although at lower concentrations, were achieved when PBMCs from both groups were incubated with living hookworm third-stage larvae. We suggest that hookworm ES products downmodulate proliferative responses and inflammation during the chronic phase of the disease and facilitate early larval survival or adult worm persistence in the gut.

We sequenced the internal transcribed spacer 2 of the ribosomal DNA (ITS2-DNAr) from the three Schistosoma mansoni intermediate hosts in Brazil: Biomphalaria glabrata, Biomphalaria tenagophila and Biomphalaria straminea. Analysis of a restriction map from those sequences allowed us to select putative restriction enzymes able to identify the snail species under study. Four restriction enzymes were used and HpaII provided simple species-specific profiles easily visualized in polyacrylamide gels. The use of ITS2 is advantageous as it provides a small fragment of 460 bp which may be easily amplified by PCR. In the current work, we showed that the amplification of ITS2-DNAr together with HpaII enzyme restriction is an auxiliary molecular tool for the morphological identification of such snails as well as for taxonomic and phylogenetic studies of neotropical planorbids. O sequenciamento da região espaçadora transcrita interna 2 do DNA ribossomal (ITS2-DNAr) das espécies brasileiras gênero Biomphalaria (B. glabrata, B. tenagophila and B. straminea) hospedeiras intermediárias do Schistosoma mansoni no Brasil, permitiu a análise dos sítios de restrição presentes nestas seqüências. A análise do mapa de restrição obtido dessas seqüências nos permitiu selecionar enzimas mais promissoras que gerassem perfis de restrição capazes de identificar essas espécies. Foram testadas 4 enzimas e a enzima HpaII foi selecionada por produzir perfis espécie específicos de fácil visualização em gel de poliacrilamida. A utilização da região ITS2 tem como vantagens a obtenção de um fragmento pequeno de 460bp, o qual pode ser facilmente amplificado por PCR. Neste trabalho, nos demonstramos que a amplificação da região ITS2-DNAr e a restrição deste com a enzima HpaII é uma ferramenta molecular auxiliar a identificação morfológica desses moluscos, bem como para estudos taxonômicos e filogenéticos de planorbídeos neotropicais.

A infecção por Ascaris lumbricoides decorre da ingestão de ovos embrionados deste parasita, o que justifica a pesquisa de substâncias que tenham efeito deletério sobre estes ovos. Nosso objetivo foi estudar a ação de 16 produtos detergentes e desinfetantes, de uso doméstico e laboratorial, sobre a embriogênese deste helminto. Crianças portadoras desta infecção foram tratadas com levamisol e os vermes fêmeas expelidos foram recolhidos e dissecados, para obtenção dos ovos intra-uterinos. Os ovos foram postos em contato com os produtos em diversas diluições e tempos, lavados e incubados a 28ºC, por 20 dias, para teste da viabilidade e determinação da porcentagem de embrionamento. Apenas um produto inibiu completamente o embrionamento dos ovos, em todos os tempos e diluições testados. Cinco produtos inibiram o embrionamento dos ovos em mais de 50%, seis inibiram o embrionamento em menos de 50% e três não tiveram efeito sobre o embrionamento dos ovos. Por outro lado, com um produto observou-se aumento da porcentagem de embrionamento dos ovos em relação aos controles.

Ascaris lumbricoides infection is acquired via ingestion of embryonated eggs of the parasite, thus justifying the search for ovicidal compounds. We studied the effect of 16 household and laboratory detergents and disinfectants on the embryogenesis of this helminth. Children carrying this infection were treated with levamisole. Eliminated female worms were collected and dissected to obtain eggs from the uteri. The eggs were placed in contact with various products at different dilutions and for various periods of time. After washing, eggs were incubated at 28ºC for 20 days, for viability tests and to determine the embryonation rate. Only one product completely inhibited the embryonation of eggs at every dilution and for every time period tested. Five products inhibited more than 50% of embryonation, while six inhibited less than 50%. Three products showed no effect on embryonation, while for one the percentage of embryonated eggs was higher than in the control tubes.

Descreve-se foco de transmissão ativa de esquistossomose mansoni em Belo Horizonte, MG, Brasil, localizado no Parque "Julien Rien", uma área de lazer com 48.500 m², criada pela Prefeitura Municipal, em 1980, na zona sul da cidade. Em capturas feitas durante 10 anos (1983-1992) foram coletados 3.361 exemplares de Biomphalaria glabrata (diâmetro médio= 9,3 mm), dos quais 23 (0,7%) estavam infectados pelo Schistosoma mansoni. O encontro de moluscos hospedeiros intermediários do S. mansoni em áreas urbanizadas (pequenos lagos artificiais cimentados de parques, jardins, prédios públicos, e outros) pode estar relacionado com a introdução de peixes e plantas aquáticas. É sugerida, como medida profilática, a não introdução de peixes e plantas aquáticas nesses locais, a não ser após período de quarentena de, no mínimo, 30 dias. Urban focus of schistosomiasis in an urban area of South-eastern, Brazil. An active, urban focus of schistosomiasis in Belo Horizonte, MG, Brazil is described. It is located in the "Julien Rien" Park, a leisure area of 48,500 m² created by the local administration in 1980 on the south side of city. Snail captures were made over 10 years (1983-1992) resulting in the collection of 3,361 Biomphalaria glabrata, ranging from 6.4 to 12.8 mm in diameter (mean = 9.3); 23 (0.7%) of these were found to be infected wi th Schistosoma mansoni. The finding of S. mansoni intermediate hosts in urban areas (ponds, small artificial lakes, etc) may be related to the introduction of fish and ornamental aquatic plants coming from specialized shops or areas in which they occur. It is suggested that the introduction of fish aquatic plants into urban water collections without a quarentine period should be avoided, as a prophylactic measure.
