Atmospheric deposition of particulate matter is an important indicator of air pollution and a significant factor in material surface fouling. The elemental composition of this nutrient-containing dust depends largely on the exposure region and time as well as on climate. Therefore, in this paper we report an analysis of atmospheric pollutions with a self-made low-cost bulk deposition sampler directed at sampling deposition via air transport and rainfall. We used the device in diverse environments - thus comparing an urban region, an area surrounded by forest and an area mainly dominated by agriculture. The total organic carbon (TOC) and total nitrogen (TN) amounts were selected as indicator parameters and analyzed in a biweekly rhythm for three and a half and two years, respectively. The TOC value responded to particulate matter in the urban area, especially significant were the influences of the New Year's firework in urban and pollen in the rural forest area. In contrast, the TN value was more under the influence of the nitrogen emissions in the agriculture-dominated area. However, the TN value did not correlate with the NOx values in the urban area because the atmospheric nitrogen emissions in the city might originate from various emission sources. Summarizing, the TOC and TN values of the self-made low-cost bulk deposition sampler were in good agreement with environmental events of their immediate surrounding. Moreover, the selected containers and sampling procedures are universally applicable to monitor and analyze organic as well as inorganic parameters (e.g. metal ions) of atmospheric deposition.

Individual Layer Fabrication (ILF) is a novel additive manufacturing process that was developed to create objects with high wood content and high mechanical strength. Here, thin and individually contoured wood composite panels are created via Binder Jetting and subsequent mechanical pressing. Like in Sheet Lamination, these panels are then laminated onto each other to create a three-dimensional object. With wood contents (more than 85 mass percent) and mechanical properties (more than 30 MPa flexural strength) on par with other engineered wood products like particle boards and plywood, the produced objects are well suited for the construction and furniture industry. To gain a deeper understanding of the process, the influence of processing parameters on the geometric and mechanical properties of the finished objects were investigated. As process parameters the amounts of adhesive and the pressing forces for both panel production and lamination were selected. It was discovered that the interaction between the amount of adhesive and the pressure used to produce the panels is highly relevant for the geometric properties. The three core mechanisms that are responsible for the mechanical properties of produced parts were identified and can be ranked in the following order: 1) the amount of adhesive in the panels binding the particles, 2) the density of the panels, 3) the amount of adhesive for laminating the panels.

Es wird über die Nachweise und die Ausbreitung der zwei nichtwirtswechselnden Blattlausarten Crypturaphis grassii Silvestri, 1935 und Neotoxoptera formosana (Takahashi, 1921) in Deutschland berichtet. Auf hohe Besiedelungsdichte reagiert die monophage, nur Alnus cordata besiedelnde C. grassii mit der verstärkten Produktion von geflügelten Morphen, die eine schnelle Ausbreitung ermöglichen. Diese Reaktion ist stark reduziert bei der polyphagen, zahlreiche Taxa des Genus Allium L. besiedelnden N. formosana, deren Ausbreitung hauptsächlich vom anthropogenen Transport besiedelter Wirtspflanzen abhängig ist.

Especially in the energy supply of multi-family houses, a wide variety of stakeholders are involved, from owners, to users, to energy service providers and society. They usually have different requirements and understandings of optimality, but ultimately have to make joint decisions and thus sensible compromises. In Germany in particular, there are a large number of multi-family houses and, at the same time, many government restrictions and subsidies in terms of energy supply. This makes it difficult to make clear recommendations for the choice of an energy supply concept that takes all stakeholder interests into account. We first identify the relevant stakeholders and define their objectives. In order to relate these with one another, we present a methodology based on energy system simulation and TOPSIS to make energy concepts objectively evaluable. A generic multi-family house with 40 residential units is examined, combining different energy technologies and insulation standards. There is no energy concept that satisfies all stakeholders equally and it is difficult to build coalitions between them. The best results are achieved by air-source heat pumps in combination with photovoltaic.

Abstract This work evaluates glacial dust as a source of sediment, and associated iron (Fe), to the Fe‐limited Gulf of Alaska (GoA). A reanalysis of GoA sediment data, using rare earth elements and thorium as provenance tracers, suggests a flux to the ocean surface of Copper River (AK) glacial dust, and associated Fe, that is comparable to the flux of dust from Asia, at least 1,000 km from the narrow mountain valley glacial dust source area. This work suggests dust from Asia may not be the largest source of Fe to the GoA. Dust models fail to accurately simulate this glacial dust transport because their coarse resolution underestimates wind speeds, and the dust flux. This work suggests that glacial dust fluxes may have been important in the geologic past (e.g., the last glacial maximum) from locations where there was more extensive coverage by glaciers than at present.

Abstract Pesticides are transferred outside of cropland and can affect animals and plants. Here we investigated the distribution of 97 current use pesticides in soil and vegetation as central exposure matrices of insects. Sampling was conducted on 53 sites along eleven altitudinal transects in the Vinschgau valley (South Tyrol, Italy), in Europe’s largest apple growing area. A total of 27 pesticides (10 insecticides, 11 fungicides and 6 herbicides) were detected, originating mostly from apple orchards. Residue numbers and concentrations decreased with altitude and distance to orchards, but were even detected at the highest sites. Predictive, detection-based mapping indicates that pesticide mixtures can occur anywhere from the valley floor to mountain peaks. This study demonstrates widespread pesticide contamination of Alpine environments, creating contaminated landscapes. As residue mixtures have been detected in remote alpine ecosystems and conservation areas, we call for a reduction of pesticide use to prevent further contamination and loss of biodiversity.

The upcoming 3GPP global mobile communication standard 6G strives to push the technological limits of radio frequency (RF) communication even further than its predecessors: Sum data rates beyond 100Gbit/s, RF bandwidths above 1GHz per link, and sub-millisecond latency necessitate very high performance development tools. We propose a new SDR firmware and software architecture designed explicitly to meet these challenging requirements. It relies on Ethernet and commercial off-the-shelf network and server components to maximize flexibility and to reduce costs. We analyze state-of-the-art solutions (USRP X440 and other RFSoC-based systems), derive architectural design goals, explain resulting design decision in detail, and exemplify our architecture’s implementation on the XCZU48DR RFSoC. Finally, we validate its performance via measurements and outline how the architecture surpasses the state of the art with respect to sustained RF recording, while maintaining high Ethernet bandwidth efficiency. Building a 6G integrated sensing and communication (ISAC) example, we demonstrate its real-time and rapid application development capabilities.

Abstract The decline of insect abundance and richness has been documented for decades and has received increased attention in recent years. In 2017, a study by Hallmann and colleagues on insect biomasses in German nature protected areas received a great deal of attention and provided the impetus for the creation of the project Diversity of Insects in Nature protected Areas (DINA). The aim of DINA was to investigate possible causes for the decline of insects in nature protected areas throughout Germany and to develop strategies for managing the problem. A major issue for the protection of insects is the lack of insect-specific regulations for nature protected areas and the lack of a risk assessment and verification of the measures applied. Most nature protected areas border on or enclose agricultural land and are structured in a mosaic, resulting in an abundance of small and narrow areas. This leads to fragmentation or even loss of endangered habitats and thus threaten biodiversity. In addition, the impact of agricultural practices, especially pesticides and fertilisers, leads to the degradation of biodiversity at the boundaries of nature protected areas, reducing their effective size. All affected stakeholders need to be involved in solving these threats by working on joint solutions. Furthermore, agriculture in and around nature protected areas must act to promote biodiversity and utilise and develop methods that reverse the current trend. This also requires subsidies from the state to ensure economic sustainability and promote biodiversity-promoting practices.

Abstract Introduction The incidence of community-acquired acute bacterial meningitis has decreased during the last decades. However, outcome remains poor with a significant proportion of patients not surviving and up to 50% of survivors suffering from long-term sequelae. These guidelines were developed by the Deutsche Gesellschaft für Neurologie (DGN) under guidance of the Arbeitsgemeinschaft der Wissenschaftlichen Medizinischen Fachgesellschaften (AWMF) to guide physicians through diagnostics and treatment of adult patients with acute bacterial meningitis. Recommendations The most important recommendations are: (i) In patients with suspected acute bacterial meningitis, we recommend that lumbar cerebrospinal fluid (with simultaneous collection of serum to determine the cerebrospinal fluid-serum glucose index and blood cultures) is obtained immediately after the clinical examination (in the absence of severely impaired consciousness, focal neurological deficits, and/or new epileptic seizures). (ii) Next, we recommend application of dexamethasone and empiric antibiotics intravenously. (iii) The recommended initial empiric antibiotic regimen consists of ampicillin and a group 3a cephalosporin (e.g., ceftriaxone). (iv) In patients with severely impaired consciousness, new onset focal neurological deficits (e.g. hemiparesis) and/or patients with newly occurring epileptic seizures, we recommend that dexamethasone and antibiotics are started immediately after the collection of blood; we further recommend that —if the imaging findings do not indicate otherwise —a lumbar CSF sample is taken directly after imaging. (v) Due to the frequent occurrence of intracranial and systemic complications, we suggest that patients with acute bacterial meningitis are treated at an intensive care unit in the initial phase of the disease. In the case of impaired consciousness, we suggest that this is done at an intensive care unit with experience in the treatment of patients with severe CNS diseases. Conclusions The German S2k-guidelines give up to date recommendations for workup, diagnostics and treatment in adult patients with acute bacterial meningitis.

Abstract We describe how typical relative humidity (RH) profiles change systematically through the phases of the Madden–Julian Oscillation (MJO). We use 18 years of classified RH‐profiles from the Atmospheric Infrared Sounder, clustered without prior knowledge of large‐scale dynamics or occurrence‐region. Occurrence probability (OP) anomalies of RH‐profiles classes exhibit classical MJO patterns: Large positive OP anomalies of the RH‐profile class with most moisture in the free troposphere are co‐located with the largest positive precipitation anomalies. About 10 days prior to this deep convective core large OP anomalies of a RH‐profile class occur consistent with lower atmospheric moistening, supporting a ubiquitous role of pre‐moistening in MJO propagation. Since each RH‐profile class is associated with distinct dynamics (most of which are subgrid‐scale to climate models) our results can serve as process‐based diagnostics to investigate parameterization shortcomings of climate models preventing them to simulate the MJO accurately.

Defect‐rich black titanium dioxide (B‐TiO2) has been extensively studied over the past decade due to its enhanced photoelectrochemical efficiency compared to titanium dioxide (TiO2), which is known for its outstanding photocatalytic stability. So far, most of the B‐TiO2 material is obtained by hydrogenation of crystalline TiO2, resulting in a disordered outer layer of a few nanometers thickness. Recently, a new sputtering process has been introduced to produce B‐TiO2 thin films without the usage of hydrogen. Herein, the influence of the sputtering process on the creation of Ti3+ defect states within the films is discussed. Comprehensive optical, structural, and electronic studies of the thin film suggest that increasing the density of Ti3+ states enhances the conductivity of the films and results in increased and broadband light absorption. In addition, the new sputtering method can also be used to alter the density of the defect states in the film in a controlled manner, allowing the optical and electronical properties of the thin film to be changed in a precise and controllable way.

Abstract The problem of global amphibian declines has prompted extensive research over the last three decades. Initially, the focus was on identifying and characterizing the extent of the problem, but more recently efforts have shifted to evidence‐based research designed to identify best solutions and to improve conservation outcomes. Despite extensive accumulation of knowledge on amphibian declines, there remain knowledge gaps and disconnects between science and action that hamper our ability to advance conservation efforts. Using input from participants at the ninth World Congress of Herpetology, a U.S. Geological Survey Powell Center symposium, amphibian on‐line forums for discussion, the International Union for Conservation of Nature Assisted Reproductive Technologies and Gamete Biobanking group, and respondents to a survey, we developed a list of 25 priority research questions for amphibian conservation at this stage of the Anthropocene. We identified amphibian conservation research priorities while accounting for expected tradeoffs in geographic scope, costs, and the taxonomic breadth of research needs. We aimed to solicit views from individuals rather than organizations while acknowledging inequities in participation. Emerging research priorities (i.e., those under‐represented in recently published amphibian conservation literature) were identified, and included the effects of climate change, community‐level (rather than single species‐level) drivers of declines, methodological improvements for research and monitoring, genomics, and effects of land‐use change. Improved inclusion of under‐represented members of the amphibian conservation community was also identified as a priority. These research needs represent critical knowledge gaps for amphibian conservation although filling these gaps may not be necessary for many conservation actions.

In this paper, we describe a teaching scenario using a virtual environment (known also in the context of the 'metaverse'). This is motivated by the challenges that arise during the pandemic. More and more teaching scenarios are transferred to online learning settings, which allow learning at any time and at any time. One of the possibilities are virtual 3D environment. These allow more intensive immersion than for example video conferences. Furthermore, they offer new didactic concepts, for example, for group activities. The benefit of using virtual 3D environments we demonstrate by a concrete learning scenario: the simulation of robot programming. A further advantage when using virtual 3D environments are personal assistants (conversational/ pedagogical agents), for example, to the ease the work load borne by teachers; meanwhile, this works well also with natural language due to advantage stage of artificial intelligence implementations.

Abstract Background Genetic variants within nearly 1000 loci are known to contribute to modulation of blood lipid levels. However, the biological pathways underlying these associations are frequently unknown, limiting understanding of these findings and hindering downstream translational efforts such as drug target discovery. Results To expand our understanding of the underlying biological pathways and mechanisms controlling blood lipid levels, we leverage a large multi-ancestry meta-analysis (N = 1,654,960) of blood lipids to prioritize putative causal genes for 2286 lipid associations using six gene prediction approaches. Using phenome-wide association (PheWAS) scans, we identify relationships of genetically predicted lipid levels to other diseases and conditions. We confirm known pleiotropic associations with cardiovascular phenotypes and determine novel associations, notably with cholelithiasis risk. We perform sex-stratified GWAS meta-analysis of lipid levels and show that 3–5% of autosomal lipid-associated loci demonstrate sex-biased effects. Finally, we report 21 novel lipid loci identified on the X chromosome. Many of the sex-biased autosomal and X chromosome lipid loci show pleiotropic associations with sex hormones, emphasizing the role of hormone regulation in lipid metabolism. Conclusions Taken together, our findings provide insights into the biological mechanisms through which associated variants lead to altered lipid levels and potentially cardiovascular disease risk.

Abstract The high share of power generation based on fluctuating renewable energy sources, especially wind and solar, has increased the levels of variability and uncertainty in power systems. The aim of this study is to develop a method for quantifying the variability of photovoltaic (PV) systems. The developed method investigates the power measurements of a PV system and quantifies its power and energy fluctuations in three steps. The first includes a classification of days into three classes according to the variability and power output of the PV system. The second consists of computing the empirical cumulative distribution functions of PV power ramps for the given classes. The third calculates the PV daily energy fluctuations based on predicted PV power output. This method of PV variability quantification was then applied to seven rooftop PV systems in different locations that feature different installed capacities, years of installation, orientations and solar cell types. The testing of this method indicates that it can be used to quantify the power and energy fluctuations of different PV systems, independent of their locations and technical characteristics. The proposed method was developed as part of a general approach for quantifying the flexibility potential of buildings and city districts with PV systems.

Working memory contents are represented in neural activity patterns across multiple regions of the cortical hierarchy. A division of labor has been proposed where more anterior regions harbor increasingly abstract and categorical representations while the most detailed representations are held in primary sensory cortices. Here, using fMRI and multivariate encoding modeling, we demonstrate that for color stimuli categorical codes are already present at the level of extrastriate visual cortex (V4 and VO1), even when subjects are neither implicitly nor explicitly encouraged to categorize the stimuli. Importantly, this categorical coding was observed during working memory, but not during perception. Thus, visual working memory is likely to rely at least in part on categorical representations. Significance statement: Working memory is the representational basis for human cognition. Recent work has demonstrated that numerous regions across the human brain can represent the contents of working memory. We use fMRI brain scanning and machine learning methods to demonstrate that different regions can represent the same content differently during working memory. Reading out the neural codes used to store working memory contents, we show that already in sensory cortex, areas V4 and VO1 represent color in a categorical format rather than a purely sensory fashion. Thereby, we provide a better understanding of how different regions of the brain might serve working memory and cognition.

Randomness is a fundamental property of human behavior. It occurs both in the form of intrinsic random variability, say when repetitions of a task yield slightly different behavioral outcomes, or in the form of explicit randomness, say when a person tries to avoid being predicted in a game of rock, paper and scissors. Randomness has frequently been studied using random sequence generation tasks (RSG). A key finding has been that humans are poor at deliberately producing random behavior. At the same time, it has been shown that people might be better randomizers if randomness is only an implicit (rather than an explicit) requirement of the task. We therefore hypothesized that randomization performance might vary with the exact instructions with which randomness is elicited. To test this, we acquired data from a large online sample (n = 388), where every participant made 1,000 binary choices based on one of the following instructions: choose either randomly, freely, irregularly, according to an imaginary coin toss or perform a perceptual guessing task. Our results show significant differences in randomness between the conditions as quantified by conditional entropy and estimated Markov order. The randomization scores were highest in the conditions where people were asked to be irregular or mentally simulate a random event (coin toss) thus yielding recommendations for future studies on randomization behavior.

A large-scale GWAS provides insight on diabetes-dependent genetic effects on the glomerular filtration rate, a common metric to monitor kidney health in disease.

Biodiversity is declining on a global scale. Especially tropical ecosystems, containing most of the planetary biodiversity, are at risk. Agricultural monocrop systems contribute to this decline as they replace original habitats and depend on extensive use of synthetic pesticides that impact ecosystems. In this review we use large-scale banana production for export purposes in Costa Rica as an example for pesticide impacts, as it is in production for over a century and uses pesticides extensively for more than fifty years. We summarise the research on pesticide exposure, effects and risks for aquatic and terrestrial environment, as well as for human health. We show that exposure to pesticides is high and relatively well-studied for aquatic systems and humans, but hardly any data are available for the terrestrial compartment including adjacent non target ecosystems such as rainforest fragments. Ecological effects are demonstrated on an organismic level for various aquatic species and processes but are not available at the population and community level. For human health studies exposure evaluation is crucial and recognised effects include various types of cancer and neurobiological dysfunctions particularly in children. With the many synthetic pesticides involved in banana production, the focus on insecticides, revealing highest aquatic risks, and partly herbicides should be extended to fungicides, which are applied aerially over larger areas. The risk assessment and regulation of pesticides so far relies on temperate models and test species and is therefore likely underestimating the risk of pesticide use in tropical ecosystems, with crops such as banana. We highlight further research approaches to improve risk assessment and, in parallel, urge to follow other strategies to reduce pesticides use and especially hazardous substances.

Abstract Kidneys are key regulators of phosphate homeostasis. Biallelic mutations of the renal Na+/phosphate cotransporter SLC34A1/NaPi-IIa cause idiopathic infantile hypercalcemia, whereas monoallelic mutations were frequently noted in adults with kidney stones. Genome-wide-association studies identified SLC34A1 as a risk locus for chronic kidney disease. Pathogenic mutations in SLC34A1 are present in 4% of the general population. Here, we characterize a mouse model carrying the 91del7 in-frame deletion, a frequent mutation whose significance remains unclear. Under normal dietary conditions, 12 weeks old heterozygous and homozygous males have similar plasma and urinary levels of phosphate as their wild type (WT) littermates, and comparable concentrations of parathyroid hormone, fibroblast growth factor 23 (FGF-23) and 1,25(OH)2 vitamin D3. Renal phosphate transport, and expression of NaPi-IIa and NaPi-IIc cotransporters, was indistinguishable in the three genotypes. Challenging mice with low dietary phosphate did not result in differences between genotypes with regard to urinary and plasma phosphate. Urinary and plasma phosphate, plasma FGF-23 and expression of cotransporters were similar in all genotypes after weaning. Urinary phosphate and bone mineral density were also comparable in 300 days old WT and mutant mice. In conclusion, mice carrying the 91del7 truncation do not show signs of impaired phosphate homeostasis.

Abstract Background Variants in SLC34A2 encoding the sodium-dependent phosphate transport protein 2b (NaPi-IIb) cause the rare lung disease pulmonary alveolar microlithiasis (PAM). PAM is characterised by the deposition of calcium-phosphate concretions in the alveoli usually progressing over time. No effective treatment is available. So far, 30 allelic variants in patients have been reported but only a few have been functionally characterised. This study aimed to determine the impact of selected SLC34A2 variants on transporter expression and phosphate uptake in cellular studies. Methods Two nonsense variants (c.910A > T and c.1456C > T), one frameshift (c.1328delT), and one in-frame deletion (c.1402_1404delACC) previously reported in patients with PAM were selected for investigation. Wild-type and mutant c-Myc-tagged human NaPi-IIb constructs were expressed in Xenopus laevis oocytes. The transport function was investigated with a 32Pi uptake assay. NaPi-IIb protein expression and localisation were determined with immunoblotting and immunohistochemistry, respectively. Results Oocytes injected with the wild-type human NaPi-IIb construct had significant 32Pi transport compared to water-injected oocytes. In addition, the protein had a molecular weight as expected for the glycosylated form, and it was readily detectable in the oocyte membrane. Although the protein from the Thr468del construct was synthesised and expressed in the oocyte membrane, phosphate transport was similar to non-injected control oocytes. All other mutants were non-functional and not expressed in the membrane, consistent with the expected impact of the truncations caused by premature stop codons. Conclusions Of four analysed SLC34A2 variants, only the Thr468del showed similar protein expression as the wild-type cotransporter in the oocyte membrane. All mutant transporters were non-functional, supporting that dysfunction of NaPi-IIb underlies the pathology of PAM.

Abstract Renewable energy generation curtailment increases due to more frequently occurring congestions in power system operation. Post‐contingency curative congestion management actions can reduce the necessity of renewable energy curtailment by enabling a more efficient utilization of transmission capacities. In this research, the potential of curative actions to substitute renewable energy curtailment is studied considering technical and economic criteria. Therefore, a novel pricing methodology for the market‐based provision of curative actions is introduced. The method is based on the security constraint optimal power flow technique. Simulations are carried out on a modified version of the IEEE 14‐bus network and a real‐world 110 kV distribution network. Battery energy storage systems are implemented as an exemplary technology to provide curative actions. The developed method achieves a positive power system impact by reducing operational costs and maximizing renewable energy integration. Also, novel business models for merchant‐owned battery energy storage systems are unveiled. The provision of curative actions further proves to be competitive to established battery storage applications. Additionally, results of different grid expansion scenarios of the 110 kV network reveal the need to coordinate power system planning and operation more extensively in the future.

Decarbonization requires new energy systems components to mitigate fossil fuel dependency, for instance electric vehicles and heat pumps, forming a sector integrated energy system. Energy management is a promising approach to integrate these devices more efficiently by orchestrating the respective consumption and generation. This study investigates the advantage of an advanced energy management algorithm that is applied to varying energy system scenarios. The energy management algorithm is based on economic principles and the system topology is represented by a rooted tree. Grid elements form parents, which act as auctioneers and devices act according to type specific demand and supply functions. This algorithm is compared to an approach where devices are not coordinated, at a system scale of six households. In order to account for different characteristics of the energy system, the different scenarios are defined according to a morphological analysis and are analysed by means of Monte-Carlo simulation. These scenarios vary the PV generation, heating technology, and building insulation. It is shown that the algorithm reduces peak loads across all scenarios by around 15 kW. Other key performance indicators, such as own consumption and self-sufficiency show a dependency on the scenarios, although the algorithm outperforms the reference in each one, achieving an increase in own consumption of at least 13 p.p. and 22 p.p. in terms of self-sufficiency.

After a short introduction about fatty acids, the article refers to an unclear statement by Gibson et al. about the kind and number of isomeric products formed by addition of hypochloric acid to palmitoleic acid and linoleic acid. A theoretical approach with detailed mechanistic and stereochemical considerations will lead the reader to the solution of the problem.

Emerging aquatic insects serve as one link between aquatic and adjacent riparian food webs via the flux of energy and nutrients. These insects provide high-quality subsidy to terrestrial predators. Thus, any disturbance of emergence processes may cascade to higher trophic levels and lead to effects across ecosystem boundaries. One stressor with potential impact on non-target aquatic insects, especially on non-biting midges (Diptera: Chironomidae), is the widely used mosquito control agent Bacillus thuringiensis var. israelensis (Bti). In a field experiment, we investigated emerging insect communities from Bti-treated (three applications, maximum field rate) and control floodplain pond mesocosms (FPMs) over 3.5 months for changes in their composition, diversity as well as the emergence dynamics and the individual weight of emerged aquatic insects over time. Bti treatments altered community compositions over the entire study duration – an effect mainly attributed to an earlier (∼10 days) and reduced (∼26%) peak in the emergence of Chironomidae, the dominant family (88% of collected individuals). The most reasonable explanation for this significant alteration is less resource competition caused by a decrease in chironomid larval density due to lethal effects of Bti. This is supported by the higher individual weight of Chironomidae emerging from treated FPMs (∼21%) during Bti application (April – May). A temporal shift in the emergence dynamics can cause changes in the availability of prey in linked terrestrial ecosystems. Consequently, terrestrial predators may be affected by a lack of appropriate prey leading to bottom-up and top-down effects in terrestrial food webs. This study indicates the importance of a responsible and elaborated use of Bti and additionally, highlights the need to include a temporal perspective in evaluations of stressors in aquatic-terrestrial meta-ecosystems.

Abstract In Germany, the decline of insect biomass was observed in nature conservation areas in agricultural landscapes. One of the main causal factors discussed is the use of synthetic pesticides in conventional agriculture. In a Germany-wide field study, we collected flying insects using Malaise traps in nature conservation areas adjacent to agricultural land. We used a multi-component chemical trace element analysis to detect 92 common agricultural pesticides in ethanol from insect traps sampled in May and August 2020. In total, residues of 47 current use pesticides were detected, and insect samples were on average contaminated with 16.7 pesticides. Residues of the herbicides metolachlor-S, prosulfocarb and terbuthylazine, and the fungicides azoxystrobin and fluopyram were recorded at all sites. The neonicotinoid thiacloprid was detected in 16 of 21 nature conservation areas, most likely due to final use before an EU-wide ban. A change in residue mixture composition was noticeable due to higher herbicide use in spring and increasing fungicide applications in summer. The number of substances of recorded residues is related to the proportion of agricultural production area in a radius of 2000 m. Therefore, a drastic pesticide reduction in large buffers around nature conservation areas is necessary to avoid contamination of their insect fauna.

Introduction: Phosphate homeostasis is regulated by a complex network involving the parathyroid hormone (PTH), fibroblast growth factor 23 (FGF23), and calcitriol acting on several organs including the kidney, intestine, bone, and parathyroid gland. Previously, we showed that activation of the Janus kinase 1 (Jak1)-signal transducer and activator of transcription 3 (Stat3) signaling pathway leads to altered mineral metabolism with higher FGF23 levels, lower PTH, and higher calcitriol levels. Here, we investigated if there are sex differences in the role of Jak1/Stat3 signaling pathway on phosphate metabolism and if this pathway is sensitive to extracellular phosphate alterations. Methods: We used a mouse model (Jak1S645P+/−) that resembles a constitutive activating mutation of the Jak1/Stat3 signaling pathway in humans and analyzed the impact of sex on mineral metabolism parameters. Furthermore, we challenged Jak1S645P+/− male and female mice with a high (1.2% w/w) and low (0.1% w/w) phosphate diet and a diet with phosphate with organic origin with lower bioavailability. Results: Female mice, as male mice, showed higher intact FGF23 levels but no phosphaturia, and higher calcitriol and lower PTH levels in plasma. A phosphate challenge did not alter the effect of Jak1/Stat3 activation on phosphate metabolism for both genders. However, under a low phosphate diet or a diet with lower phosphate availability, the animals showed a tendency to develop hypophosphatemia. Moreover, male and female mice showed similar phosphate metabolism parameters. The only exception was higher PTH levels in male mice than those in females. Discussion/Conclusion: Sex and extracellular phosphate levels do not affect the impact of Jak1/Stat3 activation on phosphate metabolism.
