The objectives of this study were to determine the social behavior response to repeated social regrouping (SR), and if social rank and the changes of social rank affect the growth rate and some metabolites and hormones linked to energy status, in “resident” replacement dairy heifers subjected to SR for 205 d. Fourteen dairy heifers (153.3 ± 16.1 kg; 9 to 11 mo old), maintained under grazing conditions, were regrouped every 21 d with 5 new animals (total = 10 SR). Two days after each SR, heifers were weighed and their withers height was measured, and blood samples for nonesterified fatty acids, glucose, and IGF-1 were taken. Concentrations of nonesterified fatty acids, glucose, and IGF-1 were taken. Social behavior was registered by continuous sampling and social status was calculated by the ETlog software, considering the total number of agonistic interactions on 2 d before and 7 d after each SR. A dominance scale was constructed in each SR, and heifers were categorized as high-ranked (HRA), medium-ranked (MRA), and low-ranked (LRA). In addition, the social rank in the first and the last 5 SR were compared, resulting in 3 categories: heifers that remained in the same social rank (Mai), and heifers that lowered (Low) and raised (Rai) their social ranks. Variables were analyzed by repeated measures using a generalized lineal mixed model, considering the social rank in each SR or the change in social rank, the number of SR and their interaction as main effects, and the farm of origin and animal as random effects. Heifers performed more agonistic (2.6 vs. 1.8 ± 0.6, day of SR and 7 d later, respectively) and total (3.8 ± 1.2 vs. 2.5 ± 0.7, day of SR and 7 d later, respectively) interactions on the day of each SR than 7 d after, whereas the number of interactions decreased after the first 2 SR. Medium-ranked heifers presented greater IGF-1 concentrations than HRA heifers at SR1, SR4, and SR9, and than LRA heifers at SR3 and SR4, whereas IGF-1 concentrations were greater in HRA heifers than MRA heifers at SR3 and SR7. In addition, LRA heifers had greater IGF-1 concentrations than MRA ones at SR3 and compared with HRA heifers at SR9. Heifers that lowered their social had greater IGF-1 concentrations than Mai heifers along the SR (132.5 ± 17.1 vs. 97.8 ± 11.2 ng/mL, Low and Mai heifers, respectively) and at SR1, SR3, and SR9. In conclusion, although regrouping animals according to their characteristics can facilitate farm handling, it should be considered that endocrine profile may be affected according to heifers' position in the social hierarchy.

Abstract Markov chain Monte Carlo (MCMC) methods have been proposed to overcome computational problems in linkage and segregation analyses. This approach involves sampling genotypes at the marker and trait loci. Among MCMC methods, scalar-Gibbs is the easiest to implement, and it is used in genetics. However, the Markov chain that corresponds to scalar-Gibbs may not be irreducible when the marker locus has more than two alleles, and even when the chain is irreducible, mixing has been observed to be slow. Joint sampling of genotypes has been proposed as a strategy to overcome these problems. An algorithm that combines the Elston-Stewart algorithm and iterative peeling (ESIP sampler) to sample genotypes jointly from the entire pedigree is used in this study. Here, it is shown that the ESIP sampler yields an irreducible Markov chain, regardless of the number of alleles at a locus. Further, results obtained by ESIP sampler are compared with other methods in the literature. Of the methods that are guaranteed to be irreducible, ESIP was the most efficient.

These problems do not arise if the genotypes are sampled jointly from the entire pedigree. This paper proposes a method to jointly sample genotypes. The method combines the Elston-Stewart algorithm and iterative peeling, and is called the ESIP sampler. For a hypothetical pedigree, genotype probabilities are estimated from samples obtained using ESIP and also scalar-Gibbs. Approximate probabilities were also obtained by iterative peeling. Comparisons of these with exact genotypic probabilities obtained by the Elston-Stewart algorithm showed that ESIP and iterative peeling yielded genotypic probabilities that were very close to the exact values. Nevertheless, estimated probabilities from scalar-Gibbs with a chain of length 235 000, including a burn-in of 200 000 steps, were less accurate than probabilities estimated using ESIP with a chain of length 10 000, with a burn-in of 5 000 steps. The effective chain size (ECS) was estimated from the last 25 000 elements of the chain of length 125 000. For one of the ESIP samplers, the ECS ranged from 21 579 to 22 741, while for the scalar-Gibbs sampler, the ECS ranged from 64 to 671. Genotype probabilities were also estimated for a large real pedigree consisting of 3 223 individuals. For this pedigree, it is not feasible to obtain exact genotype probabilities by the Elston-Stewart algorithm. ESIP and iterative peeling yielded very similar results. However, results from scalar-Gibbs were less accurate.

AbstractForensic science plays a critical role in the United States criminal legal system. For decades, many feature-based fields of forensic science, such as firearm and toolmark identification, developed outside the scientific community’s purview. The results of these studies are widely relied on by judges nationwide. However, this reliance is misplaced. Black-box studies to date suffer from inappropriate sampling methods and high rates of missingness. Current black-box studies ignore both problems in arriving at the error rate estimates presented to courts. We explore the impact of each type of limitation using available data from black-box studies and court materials. We show that black-box studies rely on unrepresentative samples of examiners. Using a case study of a popular ballistics study, we find evidence that these nonrepresentative samples may commit fewer errors than the wider population from which they came. We also find evidence that the missingness in black-box studies is non-ignorable. Using data from a recent latent print study, we show that ignoring this missingness likely results in systematic underestimates of error rates. Finally, we offer concrete steps to overcome these limitations. Supplementary materials for this article areavailable online.

ABSTRACT: A linear relationship between heart rate (HR) and oxygen consumption (VO2) has been reported in homeothermic animals, indicating that is possible to estimate heat production through HR measurements. This relationship may depend on the animal activity and environmental conditions. The main objective of the present study was to evaluate the effect of the air temperature and animal posture and activity on heat production and VO2 in relation to HR. In addition, as a secondary objective, the energy cost of eating and ruminating versus idling and standing versus lying down was determined. Twelve Holstein lactating cows were housed inside climate-controlled respiration chambers for 8 d, where the air temperature was gradually increased from 7 to 21°C during the night and from 16 to 30°C during the day with daily increments of 2°C for both daytime and nighttime. During the 8-d data collection period, HR and gaseous exchange measurements were performed, and animal posture and activity were recorded continuously. The oxygen pulse (O2P), which represents the amount of oxygen that is consumed by the cow per heartbeat, was calculated as the ratio between VO2 and HR. Results showed that heat production and VO2 were linearly and positively associated with HR, but this relationship largely varied between individual cows. Within the range tested, O2P was unaffected by temperature, but we detected a tendency for an interaction of O2P with the temperature range tested during the night versus during the day. This indicates that the effect of air temperature on O2P is nonlinear. Standing and eating slightly increased O2P (1.0 and 2.5%) compared with lying down and idling, respectively, whereas rumination increased O2P by 5.1% compared with idling. It was concluded that the potential bias introduced by these effects on the O2P for the application of the technique is limited. The energy cost of eating and ruminating over idling was 223 ± 11 and 45 ± 6 kJ/kg0.75 per day, respectively, whereas the energy cost of standing over lying down was 53 ± 6 kJ/kg0.75 per day. We concluded that O2P in dairy cows was slightly affected by both animal posture and activity, but remained unaffected by air temperature within 8 to 32°C. Nonlinearity of the relationship between the O2P and air temperature suggests that caution is required extrapolating O2P beyond the temperature range evaluated in our experiment.

This project's main objective is to create an open-source database containing a sizeable number of high-quality images of shoe impressions. The Center for Statistics and Applications in Forensic Evidence (CSAFE) team collected images that represented those found at crime scenes and constructed a database that is publicly available to forensic science and research communities. The database includes images obtained from mixed impression types: full blood impression, partial blood impression, and dust impression. The impressions are made on different flooring (vinyl and tile), and captured via various lift techniques: gel lifts and handiprints (exemplar prints with high definition using graphite powder and clear sticky vinyl, backed by a white sheet), and saved in multiple digital file types (TIF, XMP, CR3, and JPG). Our data were collected to ensure reproducibility, using simple but well-described protocols and easily accessible materials.The complete dataset includes 936 unique shoeprint images saved in 3,275 digital files.Data were collected by trained volunteers making shoe impressions on flooring with the two mediums: spatter blood and graphite powder. To make an impression, volunteers wearing a shoe stepped into the material and then walked on the flooring. A separate “lighter” step was taken to create a partial print for the blood prints. The blood prints were brought to the photography station, where researchers labeled and photographed them. Graphite prints were covered with a gel lifter before being moved to the photography room. There, the researchers removed the lift, labeled, and then photographed them.Our data will be of significant use to researchers, examiners, and anyone who could benefit from using a large dataset like this. Footwear datasets are often difficult to find, especially ones that resemble crime scenes, so our data can help fill that gap.

The competitive exclusion principle establishes that the coexistence of closely related species requires a certain degree of resource partitioning. However, populations have individuals with different morphological or behavioral traits (e.g., maturity stages, sexes, temporal or spatial segregation). This interaction often results in a multi-level differentiation in food preferences and habits. We explored such resource partitioning between and within three batoid species: Hypanus dipterurus, Narcine entemedor, and Rhinoptera steindachneri in the southern Gulf of California, Mexico, using a combination of stomach content (excluding R. steindachneri) and stable isotope analyses. We found a clear differentiation between H. dipterurus and N. entemedor, where the latter exhibited more benthic habitats, supported by a greater association to infaunal prey and higher δ13C values. Though the degree and patterns of intra-specific segregation varied among species, there was a notable differentiation in both sex and stage of maturity, corresponding to changes in specialization (i.e., isotopic niche breadth) or trophic spectrum (varying prey importance and isotopic values per group). This work is a promising step towards understanding the dietary niche dynamics of these species in a potentially important feeding area within the southern Gulf of California, as well as the biological and ecological mechanisms that facilitate their coexistence.

ABSTRACT: The objective of this study was to assess hepatic ATP synthesis in Holstein cows of North American and New Zealand origins and the gluconeogenic pathway, one of the pathways with the highest ATP demands in the ruminant liver. Autumn-calving Holstein cows of New Zealand and North American origins were managed in a pasture-based system with supplementation of concentrate that represented approximately 33% of the predicted dry matter intake during 2017, 2018, and 2019, and hepatic biopsies were taken during mid-lactation at 174 ± 23 days in milk. Cows of both strains produced similar levels of solids-corrected milk, and no differences in body condition score were found. Plasma glucose concentrations were higher for cows of New Zealand versus North American origin. Hepatic mitochondrial function evaluated measuring oxygen consumption rates showed that mitochondrial parameters related to ATP synthesis and maximum respiratory rate were increased for cows of New Zealand compared with North American origin. However, hepatic gene expression of pyruvate carboxylase, phosphoenolpyruvate carboxykinase, and pyruvate dehydrogenase kinase was increased in North American compared with New Zealand cows. These results altogether suggest an increased activity of the tricarboxylic cycle in New Zealand cows, leading to increased ATP synthesis, whereas North American cows pull tricarboxylic cycle intermediates toward gluconeogenesis. The fact that this occurs during mid-lactation could account for the increased persistency of North American cows, especially in a pasture-based system. In addition, we observed an augmented mitochondrial density in New Zealand cows, which could be related to feed efficiency mechanisms. In sum, our results contribute to the elucidation of hepatic molecular mechanisms in dairy cows in production systems with higher inclusion of pastures.

Objetivo. Estimar el efecto que representa en la calidad de la dieta de la primera infancia uruguaya niños de 2 a 4 años el consumo de productos que contienen cantidades excesivas de nutrientes críticos asociados a las enfermedades no transmisibles (ENT) (azúcares libres, grasas totales, grasas saturadas y sodio), según el modelo de perfil de nutrientes de la Organización Panamericana de la Salud (OPS). Métodos. Se utilizó un recordatorio de ingesta de alimentos durante 24 horas en una muestra representativa de 401 participantes de la Encuesta de Nutrición, Desarrollo Infantil y Salud del año 2018. Se usó la clasificación de alimentos propuesta por el sistema NOVA para categorizarlos según su naturaleza y los procesos industriales a los que son sometidos. A partir de esto se lo analizó con base en el modelo de perfil de nutrientes de la OPS, lo cual permitió identificar los productos con contenido excesivo de estos nutrientes. Resultados. El 50 % de los niños consumieron tres o más productos con exceso de alguno de los nutrientes vinculados con las ENT. Aproximadamente 9 de cada 10 niños consumen productos con exceso de al menos uno de los nutrientes críticos estudiados. Conclusión. Las dietas que no contienen productos ultraprocesados y procesados con exceso de azúcares libres, grasas totales, grasas saturadas y sodio fueron la mejor opción para los niños de 2 a 4 años. La ingesta de productos con exceso de nutrientes críticos según la OPS (y cada gramo adicional consumido de tales productos) empeora la calidad de la dieta de manera significativa, e impide que se cumpla con las recomendaciones de la Organización Mundial de la Salud.

The connection of the Southern Ocean and Eastern Equatorial Pacific (EEP) has been used to explain the presence of an old, respired water mass into the Eastern Tropical North Pacific (ETNP) and Eastern North Pacific (ENP) during deglaciation. This water mass and its geochemical properties are transported along the thermocline via subsurface and intermediate circulation. Some questions remain regarding an intermediate water mass with old and less ventilated characteristics in the eastern Pacific because certain sedimentary records of δ13C, δ18O, and Δ14C in benthic foraminifera have failed to prove its presence. The top-down hypothesis has been proposed to explain the absence of this carbon isotopic signal in benthic and its presence in planktonic foraminifera in the EEP. To prove this hypothesis in the ETNP, oxygen and carbon isotopic compositions were determined for Neogloboquadrina dutertrei in a core collected at 700 m depth over the Magdalena margin, Mexico. From these new data, a reconstruction of the water column structure and the change in ocean circulation at the thermocline level during deglaciation was inferred. N. dutertrei δ18O values in the ETNP and those from Globigerina bulloides in the ENP, similar to those of N. dutertrei in the EEP, suggest that the tropical water mass extended as far as 32 °N. Specifically, N. dutertrei δ18O values were more positive in the Heinrich Stadial-1 (HS-1) and Younger Dryas (YD) than the Bolling Allerod-Antarctic Cold Reversal (BA-ACR) and Early Holocene (EH) suggesting the presence of a saline water mass with origin in the EEP. N. dutertrei δ13C depleted values during the HS-1 and YD suggested the presence of a water mass with old and respired carbon. This is consistent with the salinification or "spicy" inferred from δ18O for the ETNP. These isotopic findings in N. dutertrei corroborate the existence of a tropical water mass that probably originated in the Southern Ocean. Ankle Foot Orthoses (AFOs) are frequently prescribed to manage gait impairments in children with physical disability, and it is important that AFOs are prescribed and fitted appropriately to maximize potential benefits. AFO tuning, manipulation of the AFO footwear combination (AFO-FC) by means of video vector analysis, is routinely used to optimize AFO use. However, the incidence or types of changes that are implemented after this type of orthotic review are unknown. To investigate the impact of a multi-disciplinary video vector clinic on AFO provision in children with physical disability. All children who attended a video vector clinic over a period of 10-years from the establishment of the clinic were included in the study. Outcomes of the clinic were grouped into 5 categories: (1) No change to AFO-FC; (2) Altered/tuned AFO-FC; (3) Discontinued AFO-FC; (4) Recast AFO; (5) Change in prescription. Data were summarised narratively. 141 independently ambulant children were included. The diagnoses were bilateral cerebral palsy (39 %, n=55), unilateral cerebral palsy (38 %, n=54), spina bifida (9 %, n=13), hereditary spastic paraparesis (2 %, n=3) and other (11 %, n=16). No changes were made in 52 % of cases (n=74), tuning in 22 % of cases (n=31), the AFO was recast in 13 % of cases (n=19) and discontinued in 10 % of cases (n=14). A prescription change was recommended in 3 % of cases (n=4). Our findings suggest that the video vector clinic is a time efficient and effective means of assessing gait function in children with AFOs. Without assessment at the clinic, most of the children assessed would likely have been referred for a full and more time consuming 3-dimensional gait analysis. Video vector analysis at the initial AFO fitting may improve alignment and possibly reduce non-compliance at an earlier stage. • Video Vector analysis resulted in changes in 48 % of orthoses. • Children with CP accounted for 77 % of all those assessed at the clinic. • Video vector clinic is a time efficient assessment of gait in children with AFOs. The objective of the study was to characterize adaptations of hepatic metabolism of dairy cows of two Holstein strains with varying proportions of grazing in the feeding strategy. Multiparous autumn calving Holstein cows of New Zealand (NZH) and North American (NAH) strains were assigned to a randomized complete block design with a 2 x 2 factorial arrangement with two feeding strategies that varied in the proportions of pasture and supplementation: maximum pasture and supplementation with a pelleted concentrate (MaxP) or fixed pasture and supplementation with a total mixed ration (FixP) from May through November of 2018. Hepatic biopsies were taken at - 45 ± 17, 21 ± 7, 100 ± 23 and 180 ± 23 days in milk (DIM), representing prepartum, early lactation, early mid-lactation and late mid-lactation. The effects of DIM, feeding strategy (FS), strain and their interactions were analyzed with mixed models using repeated measures. Cows of both strains had similar triglyceride levels, mitochondrial function and carnitine palmitoyltransferase activity in liver during lactation. However, there was an effect of DIM and FS as liver triglyceride was higher for the MaxP strategy at 21 DIM and both mitochondrial function and carnitine palmitoyltransferase activity in liver were lower for the MaxP strategy at 21 DIM. Hepatic mitochondrial function and acetylation levels were affected by the interaction between strain and feeding strategy as both variables were higher for NAH cows in the MaxP strategy. Mid-lactation hepatic gene expression of enzymes related to fatty acid metabolism and nuclear receptors was higher for NZH than NAH cows. This work confirms the association between liver triglyceride, decreased hepatic mitochondrial function and greater mitochondrial acetylation levels in cows with a higher inclusion of pasture and suggests differential adaptative mechanisms between NAH and NZH cows to strategies with varying proportions of grazing in the feeding strategy.

Abstract In pasture-based systems, there are nutritional and climatic challenges exacerbated across lactation; thus, dairy cows require an enhanced adaptive capacity compared with cows in confined systems. We aimed to evaluate the effect of lactation stage (21 vs. 180 days in milk, DIM) and Holstein genetic strain (North American Holstein, NAH, n = 8; New Zealand Holstein, NZH, n = 8) on metabolic adaptations of grazing dairy cows through plasma metabolomic profiling and its association with classical metabolites. Although 67 metabolites were affected (FDR < 0.05) by DIM, no metabolite was observed to differ between genetic strains while only alanine was affected (FDR = 0.02) by the interaction between genetic strain and DIM. However, complementary tools for time-series analysis (ASCA analysis, MEBA ranking) indicated that alanine and the branched-chain amino acids (BCAA) differed between genetic strains in a lactation-stage dependent manner. Indeed, NZH cows had lower (P-Tukey < 0.05) plasma concentrations of leucine, isoleucine and valine than NAH cows at 21 DIM, probably signaling for greater insulin sensitivity. Metabolic pathway analysis also revealed that, independently of genetic strains, AA metabolism might be structurally involved in homeorhetic changes as 40% (19/46) of metabolic pathways differentially expressed (FDR < 0.05) between 21 and 180 DIM belonged to AA metabolism.
