2. Circulating Micrornas Predict Survival of Patients with Tumors of Glial Origin

Abstract

The World Health Organization has recently introduced molecular prognostic-diagnostic biomarkers in the classification of Central Nervous System (CNS) tumors. In order to characterize subclasses of tumors that cannot find a precise location in the current classification, and, or cannot be tested because of scant material, it is important to find new molecular biomarkers in tissue and, or biological fluid samples. In this study, we identified serum microRNAs that could serve as biomarkers for the diagnosis and prognosis of patients with tumors of glial origin. We retrospectively analyzed microRNA expression in the serum extracellular vesicles of patients with tumors of glial origin. Extracellular vesicles RNA was analyzed by Nanostring. qRT-PCR confirmed 6 overexpressed microRNAs: hsa-miR-4443, hsa-miR-422a, hsa-miR-494-3p, hsa-miR-502-5p, hsa-miR-520f-3p, and hsa-miR-549a. Hsa-miR-4443 was the only microRNA that showed significant differences in most comparisons. In situ hybridization (ISH), confirmed that our signature was mostly expressed in cancer cells.Importantly, hsa-miR-549a and hsa-miR-502-5p expression predicted prognosis in patients with tumors of glial origin. Although more studies are needed, we demonstrated that serum vesicles microRNA profiles are promising diagnostic and prognostic molecular biomarkers that will find an actual application in the clinical practice of CNS tumors. Abstract

The immune escape mechanisms at the base of tumor progression in endometrial cancer mimic immune tolerance mechanisms occurring at the maternal–fetal interface. The biological and immunological processes behind the maternal–fetal interface are finely tuned in time and space during embryo implantation and subsequent pregnancy stages; conversely, those behind cancer progression are often aberrant. The environment composition at the maternal–fetal interface parallels the pro-tumor microenvironment identified in many cancers, pointing to the possibility for the use of the maternal–fetal interface as a model to depict immune therapeutic targets in cancer. The framework of cancer environment signatures involved in immune adaptations, precisely timed in cancer progression, could reveal a specific “immune clock” in endometrial cancer, which might guide clinicians in patient risk class assessment, diagnostic workup, management, surgical and therapeutic approach, and surveillance strategies. Here, we review studies approaching this hypothesis, focusing on what is known so far about oncofetal similarities in immunity with the idea to individualize personalized immunotherapy targets, through the downregulation of the immune escape stage or the reactivation of the pro-inflammatory processes suppressed by the tumor.

In primates, females can advertise ovulation by means of morphological, behavioral, and chemical cues which, increasing female attractiveness to males, may grant females, beyond mere conception, likely direct/indirect benefits. Conspicuous sexual swelling is a unique morphological signal shown by some Old World primates: sexual hormones cyclically drive the female sexual skin, tail and rump to "exaggeratedly" swell. However, given the conspicuousness, long duration and potential imprecision to pinpoint ovulation, its actual functional significance rose puzzling questions and conflicting hypotheses. Furthermore, although it has been suggested to be particularly costly if compared to other signals, yet little is still known about actual costs females have to face during this particular phase of the cycle. In fact, swelling may increase female body weight, water retention, risk of wounds and infections, resulting in a likely increase of energetic needs, unless behavioral strategies to gain energy balance them out. We investigated female energetic cost in wild Macaca maura, by comparing stress levels (i.e., fecal glucocorticoid metabolite, FGCM), activity budget and nutrient intake among fully swollen, cycling non swollen (as energetic state baseline), and lactating females (widely known as mammal most costly state). Results suggest fully swollen females facing high energetic costs. In fact, they devote the majority of their time towards proceptive and positive interactions with males, while feeding less, reducing nutrient intake and exhibiting high FGCM levels. Contrary to lactating females, fully swollen females do not show behavioral strategies to balance likely energetic expenditure.

Abstract

Selection pressures on copulatory systems rapidly shape the anatomy of external genitals, affecting both the occurrence and form (size+shape) of baculum (os penis) and baubellum ( os clitoridis ) in primates. These heterotopic bones are usually located within the distal end of the penis and clitoris. However, the pattern of occurrence within the primate phylogenetic tree and the multitude of forms shown cannot be easily merged into a univocal functional interpretation. This study aims to develop a methodological protocol to supply a suitable dataset of bone virtual volumes. These data are useful in morphological approaches exploiting both traditional and geometric morphometric techniques. We provide results on samples from both captive individuals (i.e. fresh samples from corpses coming from zoos or research laboratories) and museum samples (i.e. the primate collection of “La Specola” Natural History Museum, Florence, IT). We followed a three-step protocol which entails progressive sophistication: (1) manual palpation (efficient for fresh tissues, much less so for museum tissues made stiff by either alcohol or formalin); (2) X-rays (confirming or not the palpation); (3a) if absence is confirmed, histological analysis is performed focused on hypothetical traces of cartilage or osteocytes; (3b) if presence is confirmed, high resolution 3D images are obtained by using a non-invasive microtomography system which allows the exploration of both the external and internal structure of bones. The main objectives of the research were (1) completing the primate genital bone occurrence matrix at the specific level, and (2) providing morphological data to test evolutionary hypotheses by overlapping form variation to reproductive and ecological correlates.

15. Habitat complexity and its use correlate with soil-transmitted Helmiantiasis in two social groups of Macaca maura (H.R. Schinz 1825), Endangered Primates Primates Endemic to Sulawesi Island, Indonesia

Abstract

Sulawesi endemic Macaca maura is included in the IUCN Red List as Endangered due to anthropogenic disturbance and fragmentation of its habitat. Residual populations have a scattered distribution in the karst forests of south Sulawesi. Here the dissolution of limestone layers has created a multi-level landscape hardly accessible for ground predators and humans. In this study, we aimed to obtain better knowledge on the ecological flexibility of M. maura in the use of such a complex habitat, and its consequences on health status. Since all data published on M. maura were obtained from a single group (group B), an additional group (G) was habituated to human presence. We analysed 50 vegetation plots (10 × 20 metres) to discriminate structural features in terms of feeding options (e.g. key food species diversity, density and DBH) and anthropogenic disturbance (e.g. human trails and solid litter). We then correlated these data with habitat use and helminth infection. We collected 74 faecal samples from 18 different adult individuals belonging to both groups. Vegetation analysis suggested that there were 2 suitable habitats: a Ground Forest (e.g. higher abundance of key food species) and a Karst Tower Forest (e.g. lower presence of human trails and solid litter). Gastrointestinal investigation revealed a positive correlation between the prevalence of Trichuris sp. and time devoted to ground food-related activities in all individuals (Spearman correlation, rs = 0.665, p = 0.003). Moreover, behavioural data confirmed that group B, the larger study group of the area, spent most of its activity time in the Ground Forest (N group B = 33 ± 1; N group G = 18 ± 1; χ 2 = 134.30, d.f. = 1, p < 0.001). Since resource availability and predation risk can influence foraging decisions and, ultimately, space use, the “group size effect” might explain the significantly higher proportion of time spent by group B in the Ground Forest. Consequently, due to the Trichuris faecal-oral contamination life-cycle, the chance of infecting individuals based on their feeding habits might be described according to the “soil-transmitted helminthiasis hypothesis”.

28. Analysis of tufted capuchin (Cebus apella) courtship and sexual behavior repertoire: changes throughout the female cycle and female inter-individual differences

Abstract

Sexual solicitations and initiative (proceptivity, sensu Beach, 1976) are important components of the sexuality of females of many primate species. In the tufted capuchin (Cebus apella), female proceptivity characterizes the species’ mating system. In Study 1 we defined and discussed 20 behavioral patterns based on the observation of 6 females and 5 males living in two social groups. In Study 2 each behavior, including mounting activity, was quantitatively assessed during the periovulatory and non-periovulatory cycle phases of four females, detected on the basis of urinary progestin levels (N=20 ovulatory cycles, 5 for each female); moreover, we monitored changes in females’ social interactions (agonism, grooming activity and play). Nine of the behaviors typically used by the female during courtship and in sexual interactions showed a dramatic increase during the periovulatory phase. Though males mounted females at an apparently higher rate during the periovulatory than the non- periovulatory phase, the difference was not significant. However, when adult male mounting is separated into those which occur within play and non-play contexts, there is a significant periovulatory phase effect for mounts not associated with play. Females groomed adult males at the same rate throughout the cycle. Agonism and play did not show any phase effect; however, females’ avoidance of adult males significantly increased during the periovulatory phase. Finally, each female made a statistically different use of the behavioral repertoire by performing some behaviors more than others. This variability among females during courtship calls for further research into whether it affects mating success.
