Introduction: Head and neck cancer is an umbrella term for tumor manifestations across the head and neck regions, including the oral cavity, pharynx (including the naso, oro, and hypopharynx), larynx, and sinuses. Treatment options for head and neck cancer include surgery, radiation therapy, chemotherapy, and immunotherapy, with specific treatment plans depending upon individual tumor location and staging, together with overall patient health status. Furthermore, definitive chemoradiotherapy (CRT) has emerged as a highly effective therapeutic option for locoregional advanced head and neck squamous cell cancer. However, such therapy has also been linked to the development of spondylodiscitis. Spondylodiscitis consists of an infection starting at the vertebral endplates and spreading into the intervertebral discs, typically manifesting in adults. Case Presentation and Conclusion: This case report describes our clinical team’s experience in managing three separate cases of spondylodiscitis following CRT for head and neck tumors that presented at our clinic for diagnosis and treatment in order to identify predisposing factors that underlie the link between CRT and spondylodiscitis.

3. Cluster classification of a Brazilian gastric cancer cohort reveals remarkable populational differences in normal p53 rate

ABSTRACT Objective: Gastric adenocarcinoma remains an aggressive disease with a poor prognosis, as evidenced by a 5-year survival rate of approximately 31%. The histological classifications already proposed do not accurately reflect the high biological heterogeneity of this neoplasm, particularly in diverse populations, and new classification systems using genetic markers have recently been proposed. Following these newly proposed models, we aimed to assess the cluster distribution in a Brazilian cohort. Furthermore, we evaluated whether the inclusion of other clinical and histological parameters could enhance the predictive value. Methods: We used a previously described four-immunohistochemistry/EBER-ISH marker to classify a cohort of 30 Brazilian patients with gastric adenocarcinoma into five different clusters and compared the distribution with other genetically diverse populations. Furthermore, we used artificial intelligence methods to evaluate whether other clinical and pathological parameters could improve the results of the methodology. Results: Disclosing the genetic variability between populations, we observed a more balanced distribution of the aberrant/normal p53 ratio (0.6) between patients negative for the other markers tested, unlike previous studies with Asian and North American populations. In addition, decision tree analysis reinforced the efficiency of these new classifications, as the stratification accuracy was not altered with or without additional data. Conclusion: Our study underscores the importance of local research in characterizing diverse populations and highlights the complementary role of molecular biomarkers in personalized medicine for gastric adenocarcinoma, enhancing diagnostic accuracy and potentially improving survival rates.

O presente artigo, por meio de uma pesquisa qualitativa e revisão bibliográfica, busca analisar como a prática do advocacy desempenhado pelas ONGs junto aos órgãos de proteção dos direitos humanos no sistema onusiano, principalmente através da leitura de relatórios alternativos, auxilia no combate do descumprimento dos tratados internacionais pactuados pelos Estados-parte através da atuação denominada “truque de ilusionista”. O “truque de ilusionista” refere-se a artimanhas usufruídas pelos Estados para dissimular a realidade dos fatos em seus relatórios enviados à ONU com o intuito de reafirmar os acordos convencionados através de uma ótica própria. Nesse sentido, esta pesquisa tem como objetivo principal analisar como a advocacy das ONGs efetiva os direitos humanos, especialmente com relação as questões de gênero, ao frustrar o “truque de ilusionista” exercido pelos Estados. Sobretudo, como a atividade das Organizações Não-Governamentais de realizarem relatórios alternativos para serem expedidos aos comitês da ONU podem influenciar na análise dos relatórios oficiais remetidos pelos Estados-parte. Para a execução de tal exame, recorreu-se a leitura de publicações que versassem sobre a referida temática sob um viés qualitativo. Concluir-se que a advocacy das ONGs desempenha um papel chave dentro do sistema onusiano de direitos humanos e um mecanismo eficaz no monitoramento de violações desses direitos.

5. Characterization of acquisition of physiological quality in soybean seeds from desiccated plants

ABSTRACT: Desiccation interrupts the acquisition of physiological quality in soybean seeds, as this quality peaks at R9. However, this interruption can be mitigated by the stage in which herbicide is applied and which herbicide is used. The aim of this study was to characterize the acquisition of physiological quality in soybean seeds from plants desiccated by herbicides at different stages. A completely randomized experimental design was used in a 3×6 factorial arrangement, consisting of three phenological stages (R6, R7.1, R7.3) and six different desiccant treatments (Diquat 1.5 L.ha-1; Glufosinate 1.5 L.ha-1 and 2.0 L.ha-1; Diquat 1.0 L.ha-1 + Glufosinate 1.0 L.ha-1; Saflufenacil 140 g.ha1 + Glufosinate 1.0 L.ha-1; and Saflufenacil 140 g.ha-1 + Diquat 1.0 L.ha-1), with four replications. The following evaluations were made: desiccation rate, germination, desiccation tolerance (DT), longevity (P50), viability after one year of storage, and thousand seed weight. Seeds are able to germinate and have DT when plant desiccation through application of Diquat, Diquat + Glufosinate, Saflufenacil + Diquat, and Saflufenacil + Glufosinate occurs at R6, and for Glufosinate, at R7.1. The use of Diquat + Glufosinate and their combination with Saflufenacil showed less impact on acquisition of soybean seed longevity, which was better when application was made at R7.3.

A História da Matemática e a Etnomatemática, são tendências da Educação Matemática que buscam por um ensino de matemática mais significativo. O objetivo deste artigo é mapear trabalhos do SNHM que aliam Etnomatemática e História da Matemática que possam contribuir para o ensino de conteúdos matemáticos na educação básica. Para alcançar este objetivo, utilizamos como proposta metodológica o Mapeamento Sistemático, analisando os trabalhos aprovados nas Comunicações Orais/Científicas dos anais da SNHM das edições de 1995 a 2023. Foi possível compreender que nos trabalhos encontrados há uma preocupação em valorizar, entender o saber/fazer de povos e comunidades e buscar, de alguma forma, trazer esse conhecimento para dentro das salas de aula, demonstrando as distintas formas de fazer matemática e desmistificando a ideia de que a matemática é uma disciplina de difícil aprendizado, a-histórica e aculturada. Ainda falta muito para percorrer, a Etnomatemática e a História da Matemática têm muito em comum e sua integração pode tornar o ensino da disciplina mais cativante, menos distante e mais relevante para a vida dos estudantes.

Cervical cancer (CC) is a global public health concern, primarily caused by persistent infection with oncogenic types of human papillomavirus (HPV). The World Health Organization (WHO) has established a plan to eliminate CC as a public health issue by the year 2100. However, the implementation of the HPV vaccine is impeded by vaccine restrictions and misinformation despite its demonstrated effectiveness. The CC treatment is influenced by the disease stage, with an unfavorable prognosis for those in advanced stages. This study aimed to investigate the potential of long non-coding RNAs (lncRNAs) in CC by identifying and characterizing related lncRNAs, elucidating their regulatory mechanisms and molecular interactions, and analyzing their expression patterns in patients with diverse responses to chemoradiotherapy. Non-stem cells from CC were isolated using flow cytometry sorting and used for total RNA extraction. The RNA was used to build libraries that were subsequently sequenced using the Illumina Nextseq 550.417 lncRNAs that showed differentially expressed between CC patients who responded or not to treatment. Further analysis demonstrated that these lncRNAs significantly interact with several molecules, which play crucial roles in CC progression and therapeutic resistance. Statistical analysis correlated the expression profile of these lncRNAs with treatment efficacy. Three lncRNAs, ENSG00000267838, ENSG00000266340, and FRMD6-AS1, were identified with positive expression related to non-response to chemoradiotherapy and worse progression-free survival in CC patients. Specifically, lncRNA ENSG00000267838 has its up-regulation related to non-response and down-regulation to response to chemoradiotherapy treatment.

The dry stacking of filtered tailings is an option to deal with safety-related issues involving traditional slurry disposition in impoundments. Filtered tailings can be compacted to pre-define design specifications, which minimizes structural instability problems, such as those related to liquefaction. Yet, comprehending the tailing’s response under various stress states is essential to designing any dry stacking facility properly. Thus, the present research evaluated the mechanical response of cemented and uncemented compacted filtered iron ore tailings, considering different molding characteristics related to compaction degree and molding moisture content. Therefore, a series of one-dimensional compression tests and consolidated isotropically drained triaxial tests (CID), using 300 kPa and 3000 kPa effective confining pressures, were carried out for different specimens compacted at various molding characteristics. In addition, changes in gradation owing to both compression and shearing were evaluated using sedimentation with scanning electron microscope tests. The overall results have indicated that the 3% Portland cement addition enhanced the strength and stiffness of the compacted iron ore tailings, considering the lower confining pressure. Nevertheless, the same was not evidenced for the higher confining stress. Moreover, the dry-side molded specimens were initially stiffer, and significant particle breakage did not occur owing to one-dimensional compression but only due to shearing (triaxial condition).

9. Citogenética de pequenos mamíferos a partir da extração da medula óssea: uma descrição mais detalhada da técnica in vivo com ajustes

A citogenética é uma ferramenta eficaz na identificação de espécies e resolução de problemas taxonômicos de pequenos mamíferos. Os protocolos in vivo são úteis devido ao seu baixo custo, mas apresentam divergência metodológica, precisando ser frequentemente ajustados. Neste estudo, apresentamos um método padronizado para a análise citogenética de pequenos mamíferos a partir da extração de medula óssea femoral, e demonstramos a importância dessa padronização em contraste com outras abordagens. Aplicamos a técnica de Ford e Hamerton (1956) com as seguintes atualizações: 1) adequação das quantidades e concentrações de soluções utilizadas (colchicina e solução hipotônica); 2) descrição detalhada das etapas; 3) utilização de centrífuga como método de separação; 4) adição de procedimento de montagem de lâminas, o qual melhora a visualização de metáfases. Seis protocolos citogenéticos que utilizam medula óssea foram comparados em etapas importantes da fase de preparação da amostra. Houve diferenças na concentração de soluções de colchicina entre todos os métodos. O tempo de ação da solução também variou (de 15 minutos a 6 horas). O tempo e a concentração das soluções de colchicina são etapas cruciais para a condensação dos cromossomos e geração de boas metáfases. Deste modo, destacamos que a metodologia apresentada em nosso estudo contribui com a padronização de um protocolo citogenético in vivo indicado para pequenos mamíferos, que possibilita o aproveitamento do material e redução no número de animais sacrificados para ajustes de metodologia, gerando amostras viáveis desde as primeiras tentativas.

Este artigo trata de uma pesquisa de orientação fenomenológica, que buscou empreender um movimento hermenêutico na análise de fotografias produzidas, com o auxílio de smartphones, por discentes dos Cursos de Licenciatura em Educação do Campo da Universidade Federal do Maranhão, durante a execução do componente curricular intitulado “Matemática Básica”, ao longo dos anos de 2016 a 2019. Na pesquisa, compreendemos essas fotografias como narrativas que, como tais, a partir das ideias de Paul Ricouer (2004; 2010a; 2010b; 2019), podem modificar as pessoas que as leem e carregam consigo uma identidade narrativa. Orientada pela pergunta O que se mostra a partir da leitura das as fotografias produzidas por licenciandas e licenciandos em Educação do Campo sobre as mulheres campesinas e a sua relação com a matemática?, defini como corpus de análise somente as fotografias que, de algum modo, traziam o lugar e os saberes matemáticos dessas mulheres. Com base na categoria final “Mulheres campesinas, saberes matemáticos e invisibilização”, foi possível compreender que essas mulheres se encontram em um espaço de transição, são seres inconclusos, assim como as licenciandas e licenciandos que atuaram como fotógrafas e fotógrafos. Das narrativas produzidas, é perceptível algo que fica – o culto, o respeito à natureza e a contribuição dessas mulheres no sustento familiar – e algo que se modifica, qual seja, o lugar delas e o lugar de seus saberes matemáticos e científicos.

14. Heatwaves in Mozambique 1983–2016: Characteristics, trends and city-level summaries using high-resolution CHIRTS-daily

The intensity, frequency, and duration of heatwaves are increasing worldwide. Still, heatwaves are systematically underreported and underresearched across the African continent. This study examines heatwaves across Mozambique, a country highly vulnerable to a variety of climate risks yet where heatwaves have received little to no attention. A spatio-temporal analysis was conducted for five heatwave characteristics (heatwave number, frequency, duration, amplitude, and magnitude) and corresponding trends from 1983 to 2016. This was done using the remotely sensed CHIRTS-daily, which presents one of the most accurate and highest resolution (5 × 5 km) daily temperature product currently available, especially for data-scarce regions. Three heatwave definitions were analyzed and compared, which are based on (1) the 90th percentile of daily maximum temperature (TX90), (2) the 90th percentile of daily minimum temperature (TN90), and (3) the Excess Heat Factor (EHF). Results were overlayed with high-resolution population data to obtain heatwave exposure and likely potential implications. Our findings show that Mozambique has experienced many heatwaves over the past decades. On average, 2–18.6 annual heatwave days (HWF) were recorded with the longest heatwaves (HWD) lasting X- 11.5 days. More and longer heatwaves were observed in the North and along the coast of Mozambique. Heatwave magnitude (HWM) ranged from 0.3 to 6.8 °C and amplitude (HWA) from 0.8 to 11.7 °C, with highest values in South and Central Mozambique. O presente relato narra a experiência de estágio obrigatório na Educação Infantil, realizado na 7ª fase do curso de Pedagogia, da Universidade Federal de Santa Catarina, no ano de 2021. O estágio foi realizado integralmente de forma remota, devido ao distanciamento social causado pela pandemia do novo Corona vírus. No texto narramos os desafios de estagiar sem o contato direto com as crianças, professores(as) e instituições, trazendo inquietudes, inseguranças e indagações no que se refere ao nosso processo formativo nesta conjuntura, articuladas a reflexões e considerações a respeito da possibilidade de resistência, na utilização deste distanciamento, como um tempo tempestivo para, em conjunto, (re)pensar, explorar e escrever sobre as especificidades da Educação Infantil e o tempo da infância como um direito, bem como advogar a importância das ferramentas da ação pedagógica como instrumentos imprescindíveis para uma permanente construção da docência na Educação Infantil.

Para que ocorra a extensão universitária é necessário elencar uma população-alvo para dirigirem-se as ações extensionistas. No entanto, estas ações tendem a se estender para além da comunidade, por ação de formação de multiplicadores de conhecimento na própria população atendida. O objetivo deste estudo é fazer um relato de experiência da contribuição de ações de extensão para além do público-alvo quando se trabalha temas relacionados à saúde como o uso de plantas e chás medicinais e a Hipertensão Arterial Sistêmica junto a cuidadores de Pessoas com Deficiência (PcD). Este projeto de extensão atuou junto a quinze cuidadoras de pessoas com deficiência a partir de temas sugeridos por elas. Na atividade sobre plantas e chás medicinais as metodologias utilizadas foram caixa de mitos e verdades, roda de conversa, jogo de bingo e distribuição de fôlderes. Para a atividade sobre hipertensão utilizou-se “tempestade de ideias”, aferição da pressão arterial e distribuição de fôlderes. Essas ações foram realizadas em 3 encontros entre março e maio de 2019. Como resultado, tivemos a participação de 11 a 16 cuidadoras (sendo uma participante ocasional) e 2 a 3 funcionárias da Instituição parceira nos encontros. Observou-se que as metodologias ativas empregadas foram importantes para o aprendizado e fixação dos conteúdos abordados segundo as falas das próprias cuidadoras. As ações possibilitaram que os saberes construídos fossem compartilhados com amigos e familiares das participantes, caracterizando a contribuição do projeto para formação de multiplicadores do conhecimento. Palavras-chave: Ensino; Disseminação de informação; Educação em saúde Knowledge multipliers: Role of extension actions with caregivers of people with disabilities Abstract: For the university extension to occur, it is necessary to establish a target population to which extension actions will be directed. However, these actions tend to spread beyond the community by forming knowledge multipliers in the served population. This study aimed to report the experience of the contribution of extension actions beyond the target audience when working on health-related topics such as the use of medicinal plants and teas and Systemic Arterial Hypertension with caregivers of People with Disabilities (PwD). This extension project worked with fifteen caregivers of PwD based on themes suggested by them. In the medicinal plants and teas activity, the methodologies used were the box of myths and truths, conversation wheel, bingo game, and distribution of folders. For the activity on hypertension, we used "brainstorm," blood pressure measurement, and distribution of folders. These actions were carried out in 3 meetings between March and May 2019. As a result, 11 to 16 caregivers (one casual) and 2 to 3 employees from the partner institution participate in the meetings. It was observed that the active methodologies used were important for learning and fixing the contents addressed, according to the words of the caregivers themselves. 17. Legal action for access to resources inefficiently made available in health care systems in Brazil: a case study on obstructive sleep apnea

ABSTRACT Objective: Obstructive sleep apnea (OSA) is a highly prevalent chronic disease, associated with morbidity and mortality. Although effective treatment for OSA is commercially available, their provision is not guaranteed by lines of care throughout Brazil, making legal action necessary. This study aimed at presenting data related to the volume of legal proceedings regarding the access to diagnosis and treatment of OSA in Brazil. Methods: This was a descriptive study of national scope, evaluating the period between January of 2016 and December of 2020. The number of lawsuits was analyzed according to the object of the demand (diagnosis or treatment). Projections of total expenses were carried out according to the number of lawsuits. Results: We identified 1,462 legal proceedings (17.6% and 82.4% related to diagnosis and treatment, respectively). The projection of expenditure for OSA diagnosis in the public and private spheres were R$575,227 and R$188,002, respectively. The projection of expenditure for OSA treatment in the public and private spheres were R$2,656,696 and R$253,050, respectively. There was a reduction in the number of lawsuits between 2017 and 2019. Conclusions: Legal action as a strategy for accessing diagnostic and therapeutic resources related to OSA is a recurrent practice, resulting in inefficiency and inequity. The reduction in the number of lawsuits between 2017 and 2019 might be explained by the expansion of local health care policies or by barriers in the journey of patients with OSA, such as difficulties in being referred to specialized health care and low availability of diagnostic resources.

Introduction. Knee osteoarthrosis, whether subtle or marked, appears to alter the stability and performance of the knee joint in activities of daily living that prevent the maintenance of bipedal posture. However, there is still a gap in the literature as to how knee osteoarthritis can affect static balance. Objective. To analyze the performance of postural control in elderly diagnosed with unilateral knee osteoarthrosis. Materials and Methods. 40 elderly people of both sexes participated in this study, divided into two groups containing 20 elderly each. Group 1 (G1) consists of elderly patients who have received a diagnosis of unilateral knee osteoarthritis. Despite undergoing conservative treatment, their condition has shown insufficient improvement, leading to a clinical recommendation for total knee arthroplasty (TKA). The G2 group was made up of 20 elderly with an average age of 71.09 years, considered active, who do not have a diagnosis of osteoarthritis in the knee joint and practice physical activity. With the aid of a Baroscan pressure platform, the center of pressure (COP) displacement in the anteroposterior (COPAP) direction and mediolateral direction (COPML) and the area of center of pressure displacement were evaluated during bipedal postural control with eyes open and eyes closed. Results. During postural control with eyes open and eyes closed, the G1 group showed greater displacement of the COP in the anteroposterior direction—COPAP (p=0.007)—and mediolateral direction—COPML (p=0.033)—when compared to the G2 group. As for the area of displacement of the COP, group G1 presented a larger area of displacement (p=0.002) than group G2 during bipedal postural control with open eyes. For the condition with eyes closed, both groups showed similar behaviors, which resulted in no present statistically significant differences. Conclusion. The results suggest that unilateral knee osteoarthritis influences bipedal postural control and activities of daily living that require this static balance, since information from the somatosensory system is reduced, resulting in stability of tasks that require body control and promoting the risk of falls. From a clinical perspective, the results suggest that the assessment of bipedal postural control can assist orthopedic physicians in assessing joint stability in patients with unilateral knee osteoarthrosis.

The Degree in Rural Education at Federal University of Maranhão (UFMA) began its activities in 2009 and was institutionalized in 2014. In order to encourage discussions, as well advance in the consolidation of this degree, we ask ourselves what the guiding principles of the curriculum are evident in the Pedagogical Political Project (PPP) of the Degree Course in Rural Education — Natural Sciences and Mathematics, at UFMA. Guided by this question, we bring a history of the creation of the course, we present the general aspects of its curricular organization and, finally, we weave reflections, in the hermeneutic perspective of Paul Ricoeur (1987), about the guiding principles of the curriculum that is evidenced in the PPP, namely: Education as a political act of social transformation; Teacher training in an emancipatory perspective and Alternation as a space for reflection-action-reflection. The interpretative exercise points to the need to broaden the debate regarding training by area of knowledge and interdisciplinarity in your curriculum.

IntroductionMethotrexate (MTX), a folic acid antagonist and nucleotide synthesis inhibitor, is a cornerstone drug used against acute lymphoblastic leukemia (ALL), but its mechanism of action and resistance continues to be unraveled even after decades of clinical use.MethodsTo better understand the mechanisms of this drug, we accessed the intracellular metabolic content of 13 ALL cell lines treated with MTX by 1H-NMR, and correlated metabolome data with cell proliferation and gene expression. Further, we validated these findings by inhibiting the cellular antioxidant system of the cells in vitro and in vivo in the presence of MTX.ResultsMTX altered the concentration of 31 out of 70 metabolites analyzed, suggesting inhibition of the glycine cleavage system, the pentose phosphate pathway, purine and pyrimidine synthesis, phospholipid metabolism, and bile acid uptake. We found that glutathione (GSH) levels were associated with MTX resistance in both treated and untreated cells, suggesting a new constitutive metabolic-based mechanism of resistance to the drug. Gene expression analyses showed that eight genes involved in GSH metabolism were correlated to GSH concentrations, 2 of which (gamma-glutamyltransferase 1 [GGT1] and thioredoxin reductase 3 [TXNRD3]) were also correlated to MTX resistance. Gene set enrichment analysis (GSEA) confirmed the association between GSH metabolism and MTX resistance. Pharmacological inhibition or stimulation of the main antioxidant systems of the cell, GSH and thioredoxin, confirmed their importance in MTX resistance. Arsenic trioxide (ATO), a thioredoxin inhibitor used against acute promyelocytic leukemia, potentiated MTX cytotoxicity in vitro in some of the ALL cell lines tested. Likewise, the ATO+MTX combination decreased tumor burden and extended the survival of NOD scid gamma (NSG) mice transplanted with patient-derived ALL xenograft, but only in one of four ALLs tested.ConclusionAltogether, our results show that the cellular antioxidant defense systems contribute to leukemia resistance to MTX, and targeting these pathways, especially the thioredoxin antioxidant system, may be a promising strategy for resensitizing ALL to MTX.

Adjuvant emulsions are widely used to enhance the antibody response in animals used as immunoglobulin source to produce snake antivenoms. We tested the performance of four commercial emulsion adjuvants (Montanide, Freund, Carbigen, and Emulsigen-D) and an experimental adjuvant (QH-769) in the antibody response of horses towards venoms of the African snakes Bitis arietans, Echis ocellatus, Dendroaspis polylepis and Naja nigricollis. Montanide, Freund and Carbigen adjuvants generated the highest immune response but induced moderate/severe local lesions at the site of injection. In contrast, Emulsigen-D and QH-769 adjuvants generated the lowest immune response and low incidence of local lesions. No evidence of systemic alterations was observed in the horses immunized with any of the adjuvants. It is suggested that the use of Montanide or Freund-based emulsions in the first immunization steps, followed by the use of Emulsigen-D, QH-769 or similar adjuvants in the following injections, could result in a satisfactory immune response against snake venoms, while not inducing serious local deleterious effects.

The focus of this study is to investigate the gene expression related to the formation of neutrophil extracellular traps (NETs) stimulated by Calloselasma rhodostoma L-amino acid oxidase (Cr-LAAO). LAAOs found in snake venom have been shown to activate human neutrophils, leading to the production of reactive oxygen species (ROS), chemotaxis, phagocytosis, and the release of pro-inflammatory cytokines and lipid mediators. Additionally, it has been found that Cr-LAAO activates NADPH oxidase, which is responsible for the release of ROS. Neutrophils are known to release NETs to combat pathogens, and this process involves the migration of DNA from the nucleus to the cytoplasm, where it merges with the contents of the granules to produce NETs. Initially, the formation of NETs was associated with cell death, and this process was known as NETosis. However, two forms of NETosis have now been identified: classical or suicidal NETosis, which results in cell death, and vital NETosis, in which the cell retains its viability and many of its effector functions. To evaluate the gene expression related to the formation of NETs, a microarray assay was performed on human neutrophils stimulated with Cr-LAAO. The results show that Cr-LAAO stimulates the expression of important genes for the formation of NETs, such as TXNIP, FOXO3, PPARA, ELANE, CXCL8, and PADI4. This is the first report that shows the transcriptome of neutrophils related to Cr-LAAO-stimulated NETosis, which may lead to the development of local inflammatory effects observed in snakebite victims.

Angelman Syndrome is a rare neurodevelopmental disorder caused by several (epi)genetic alterations. The patients present strong neurological impairment due to the absence of a functional maternal UBE3A gene in neurons. Here, we generated and characterized a new induced pluripotent stem cell (iPSC) line from a female child with Angelman syndrome harbouring a class II deletion. iPSCs were reprogrammed from fibroblasts using Sendai viruses. The new iPSCs express pluripotency markers, are capable of trilineage in vitro differentiation and have the expected imprinting status of Angelman syndrome. These iPSCs are a valuable tool to elucidate the pathophysiological mechanisms associated with this disease.

Este trabalho tem por objetivo refletir sobre aspectos da formação inicial de professores de Física a partir da perspectiva de estudantes do curso de Física Licenciatura da Universidade Federal do Maranhão. São apresentadas algumas problemáticas que interferem no processo de formação docente e contribuem para o elevado índice de evasão, assim como, se discute sobre o desenvolvimento da prática docente, nos primeiros anos do curso, analisando contribuições que algumas disciplinas e programas oferecem. Para embasar tais reflexões, além da análise do projeto pedagógico do curso, foi aplicado um questionário que coletou informações junto a estudantes, matriculados nas disciplinas Didática e Prática de Ensino I, ministradas no sexto e sétimo período, respectivamente. Os dados obtidos reafirmam prováveis motivos para a evasão do curso, entre eles, a falta de conhecimento de conceitos referentes a conteúdos da Educação Básica. Eles mostraram também, que estudantes que tiveram experiências na docência, se sentem estimulados a desenvolverem de forma mais consciente suas funções em sala de aula. As reflexões apresentadas aqui evidenciam a necessidade da realização de mais pesquisas referentes à formação inicial de professores de Física, bem como, a implantação de ações que promovam mudanças efetivas na organização de cursos de Licenciatura em Física.

A aplicação de nitrogênio (N) na dose correta e no momento adequado pode favorecer a produtividade e a qualidade das sementes de trigo. Neste sentido, objetivou-se avaliar o efeito do parcelamento da adubação nitrogenada sobre a produtividade e a qualidade fisiológica de sementes de cultivares de trigo. As sementes utilizadas no experimento foram produzidas no Centro Nacional de Pesquisa de Soja (Embrapa Soja), em Londrina-PR, em delineamento experimental de blocos casualizados em esquema fatorial 2×7, com quatro repetições. Os tratamentos constaram de duas cultivares e sete combinações de aplicação da adubação nitrogenada. Foram avaliados: produtividade, germinação, primeira contagem da germinação, comprimento e massa seca de plântulas, emergência de plântulas em areia e índice de velocidade de emergência. Os dados foram submetidos à análise de variância com comparação de médias pelo teste F, entre os genótipos, e Skott-Knott, entre as combinações de N. O efeito da fertilização nitrogenada sobre a produtividade de sementes varia com a cultivar utilizada. Para ambas as cultivares, a adubação nitrogenada aplicada em uma única dose eleva o potencial fisiológico de sementes. O suprimento inadequado de N prejudica a qualidade fisiológica sementes produzidas.

26. The Ototoxicity of Chloroquine and Hydroxychloroquine: A Systematic Review

Introduction Chloroquine and hydroxychloroquine are antimalarial drugs widely used in the treatment of rheumatic diseases. With the global pandemic caused by the new coronavirus, there was an increase in the prescription of these drugs, which led to a major concern regarding their ototoxic effects. Objectives The objective of the present study was to assess existing scientific evidence about the toxic effects of chloroquine and hydroxychloroquine on the peripheral and/or central auditory system. Data Synthesis A systematic literature review was performed by searching the PubMed (Medline), Scopus, Web of Science, LILACS, and SciELO electronic databases, in a search of articles that fullfiled the predefined inclusion and exclusion criteria. The review was conducted in three phases and, in all of them, analyses were performed by two independent researchers. Disagreements were discussed with a third researcher until a consensus was reached. A total of 437 articles were found and 8 were included in this review. Seven of the included studies reported hearing loss in their samples and presented a diagnostic hypothesis of ototoxicity induced by chloroquine or hydroxychloroquine. The most common type of hearing loss was sensorineural, with varying laterality and degrees of severity. The most frequently used audiological test was pure tone audiometry, and only two studies assessed brainstem evoked responses. Conclusion The scientific evidence compiled in this research showed that chloroquine and hydroxychloroquine have an ototoxic effect in the peripheral auditory system. These drugs can cause cochlear damage, including changes in the stria vascularis and lesions in sensory hair cells.

In March 2022, large parts over the north Indian plains including the breadbasket region, and southern Pakistan began experiencing prolonged heat, which continued into May. The event was exacerbated due to prevailing dry conditions in the region, resulting in devastating consequences for public health and agriculture. Using event attribution methods, we analyse the role of human-induced climate change in altering the chances of such an event. To capture the extent of the impacts, we choose March–April average of daily maximum temperature over the most affected region in India and Pakistan as the variable. In observations, the 2022 event has a return period of ∼1-in-100 years. For each of the climate models, we then calculate the change in probability and intensity of a 1-in-100 year event between the actual and counterfactual worlds for quantifying the role of climate change. We estimate that human-caused climate change made this heatwave about 1 °C hotter and 30 times more likely in the current, 2022 climate, as compared to the 1.2 °C cooler, pre-industrial climate. Under a future global warming of 2 °C above pre-industrial levels, heatwaves like this are expected to become even more common (2–20 times more likely) and hotter (by 0 °C–1.5 °C) compared to now. Stronger and frequent heat waves in the future will impact vulnerable groups as conditions in some regions exceed limits for human survivability. Therefore, mitigation is essential for avoiding loss of lives and livelihood. Heat Action Plans have proved effective to help reduce heat-related mortality in both countries.

Familiares cuidadores de pessoas com deficiência (PcD) abdicam muitas vezes de seu bem-estar em função dos cuidados necessários de seu ente cuidado. Assim, o objetivo deste estudo foi trabalhar a Fisiologia do sono e do sistema digestório com as cuidadoras de PcD que fazem acompanhamento em Centro de Reabilitação de Porto Alegre ligadas ao projeto de extensão universitária denominado “Apoiando e educando as famílias de pessoa com deficiência”, estimulando-se o bem-estar dessas mulheres. Como metodologias para o estudo da Fisiologia junto às cuidadoras foram utilizadas rodas de conversa, a Escala de Sonolência de EPWORTH, moldes anatômicos do sistema digestório e questionário sobre constipação intestinal em dois encontros com duração de 90 minutos cada um. Participaram das atividades relativas à Fisiologia do sono 9 cuidadoras e observou-se que 66,6% delas deveriam cuidar mais de seu sono ou apresentavam sono inadequado. Quanto a Fisiologia do sistema digestório, 11 cuidadoras participaram das atividades e 20% foram consideradas constipadas. O estudo sobre a Fisiologia do sono e do sistema digestório fez com que as cuidadoras pudessem refletir sobre o autocuidado em relação a estes aspectos e oportunizou que as extensionistas colocassem em prática o conhecimento da Universidade. Thus, this study aimed to work on sleep and digestive system physiology with PwD caregivers whom follow-up on the Rehabilitation Center of Porto Alegre (Brazil) linked to the university extension project called "Supporting and educating families of people with disabilities," stimulating the well-being of these women. As methodologies for studying physiology with caregivers, we used conversation wheels, the EPWORTH Sleepiness Scale, anatomical molds of the digestive system, and intestinal constipation questionnaire in two 90-minute meetings. Nine caregivers participated in sleep physiology activities, and it was observed that 66.6% of respondents should take better care of their sleep or had an inadequate sleep. Regarding the physiology of the digestive system, 11 caregivers participated in the activities, and 20% were considered constipated. The study of sleep and digestive system physiology allowed caregivers to think about self-care, improving their well-being, and helped the extensionists to put the University's knowledge into practice. A presente pesquisa busca analisar o efeito do índice porosidade/teor volumétrico de ligante no comportamento de um solo argiloso. Os fatores controláveis são as quantidades de cinza volante e de cal dolomítica e o peso específico aparente seco. Os resultados indicam aumento da resistência a compressão não confinada com a adição de cal e cinza, assim como com a redução da porosidade. Além disso, foi possível comprovar a eficácia do índice porosidade/teor volumétrico de ligante na análise do comportamento de solos argilosos.

This article’s aim is to introduce a research about the history of relations between Brazil and Vietnam since its establishment in 1989. A PWC’s report (2017) indicated that, in the next three decades, Vietnam will be the country with the highest economic growth (GDP per capita) in the world, registering an average of 5.1% annually. In a context where 4 of the world's 7 largest GDPs will be in Asia (expected for 2050), this growth is significant and deserves attention. Through a bibliographic review of Vietnam's recent history, as well as use of portals from both governments and research reports, it seeks, in a first step, to demonstrate the recent changes that have taken place in the country and then to demonstrate how Vietnamese foreign policy was designed at the end of the 20th century and at the beginning of the 21st century. Finally, the research includes data on official visits, support in multilateral institutions and bilateral trade between Brazil and Vietnam.

A determinação da densidade ótima de semeadura do trigo, adequada ao genótipo e ao ambiente, é essencial para evitar o acamamento e a competição entre as plantas, o que pode reduzir a produtividade e a qualidade das sementes. Neste sentido, objetivou-se avaliar o efeito da densidade de semeadura (nas cultivares BRS Sabiá e BRS Gralha-Azul) na produtividade e na qualidade fisiológica de sementes de duas cultivares de trigo. As sementes foram produzidas no campo experimental da Empresa Brasileira de Pesquisa Agropecuária, em Londrina-PR, no Centro Nacional de Pesquisa de Soja (Embrapa Soja) em delineamento experimental de blocos casualizados em esquema fatorial 2x4, com quatro repetições. Foram realizadas as seguintes avaliações: produtividade de sementes, massa de mil sementes, germinação, primeira contagem da germinação, comprimento e massa seca de plântulas, envelhecimento acelerado, emergência de plântulas em areia e índice de velocidade de emergência. De modo geral, observou-se que o aumento da densidade de semeadura aumenta a produtividade das sementes e não altera a geminação e a emergência de plântulas de ambas cultivares. A cultivar BRS Gralha-Azul mostrou-se mais produtiva que a BRS Sabiá. O aumento da densidade de semeadura aumenta o vigor das sementes para a cultivar BRS Sabiá e reduz para a cultivar BRS Gralha-Azul.

ABSTRACT Objective: To reflect on the repercussions of premature babies’ oral ability concerning breastfeeding, under the light of the theory of causation. Methods: Theoretical production of reflections based on Hobbes’s theory of causation. Results: The study addresses the understanding of oral abilities as the main accident regarding the capacity of the premature newborn, which, coupled with other accidents that make up the other domains, concerning breastfeeding, is an integral cause of the phenomenon. Final considerations: Although there are protocols, even if some of them are inconsistent or incomplete, the use of criteria such as weight and gestational age as standards to understand this readiness can still be observed. However, the effect manifests itself even in the absence of these accidents, showing them as partial causes of the phenomenon, while oral ability is, by itself, a necessary cause for this event to take place.

Abstract: Initial seed vigor and sowing density interact in establishment of plants in the field, and can thus affect expression of the plasticity of the wheat crop and the yield and physiological quality of the seeds produced. The aim of this study was to evaluate the effect of seed vigor levels in combination with sowing densities on the yield and physiological potential of wheat seeds. The cultivars BRS Gralha-Azul and BRS Sabiá were used in a randomized block experimental design in a 2 × 4 factorial arrangement, with four replications. The treatments consisted of seeds of high and low vigor and four sowing densities (150, 250, 350, and 450 seeds.m-2). The following evaluations were made: seed yield, germination, first germination count, seedling length, seedling dry matter, accelerated aging, emergence speed index, and seedling emergence in sand. An increase in sowing density favors the seed yield of both cultivars; however, it reduces the vigor of the seeds produced by the cultivar BRS Gralha-Azul, especially of the seeds produced by plants originating from high vigor seeds. For the cultivar BRS Sabiá, an increase in sowing density decreases the germination performance of seeds produced by plants originating from low vigor seeds.

Pluripotent stem cells (PSCs) are central to development as they are the precursors of all cell types in the embryo. Therefore, maintaining a stable karyotype is essential, both for their physiological role as well as for their use in regenerative medicine. Karyotype abnormalities in PSCs in culture are common but the underlying causes remain unknown. To gain insight, we explore the composition of the centromere and kinetochore in human embryonic and induced PSCs. Centromere function depends on CENP-A nucleosome-defined chromatin. We show that while PSCs maintain abundant pools of CENP-A, CENP-C and CENP-T, these essential centromere components are strongly reduced at stem cell centromeres. Outer kinetochore recruitment is also impaired to a lesser extent, indicating an overall weaker kinetochore while the inner centromere protein Aurora B remains unaffected. We further show that, similar to differentiated human cells, CENP-A chromatin assembly in PSCs requires transition into G1 phase. Finally, reprogramming experiments indicate that reduction of centromeric CENP-A levels is an early event during dedifferentiation, coinciding with global chromatin remodelling. Our characterization of centromeres in human stem cells suggests a possible link between impaired centromere function and stem cell aneuploidies.

Analisar a utilização dos cateteres centrais de inserção periférica em recém-nascidos internados na Unidade de Terapia Intensiva Neonatal. Pesquisa retrospectiva, descritiva e quantitativa, realizada em 80 prontuários de uma maternidade escola federal do Rio de Janeiro, no período de abril a julho de 2018. A utilização do dispositivo foi maior em recém-nascidos pré-termo (83%), o peso predominante foi abaixo de 1.500 gramas (60,9%), a indicação mais frequente para a inserção do cateter foi o baixo peso, associado ou não a terapia intravenosa prolongada (35,2%) e o tempo de permanência foi superior a cinco dias (81,8%). A ocorrência de eventos adversos estava presente em 31,8% dos casos. Ressalta-se a importância da correta indicação, manuseio, cuidado no uso do dispositivo, além de identificação dos eventos adversos, para que se estabeleçam medidas de prevenção e treinamento da equipe, a fim de diminuir os riscos e promover a segurança dos recém-nascidos. As intoxicações em animais domésticos é algo crescente, principalmente por produtos tóxicos como raticidas, sanitizantes, medicações, agrotóxicos e plantas. Muitos tutores desconhecem os perigos destes produtos e o risco à saúde dos seus animais. Objetiva-se com este trabalho relatar um caso de um canino intoxicado espontaneamente por SOLUPAN. Uma cadela, sem raça definida de 1 ano de idade, apresentou sinais clínicos sugestivos de intoxicação, após ingerir detergente desengraxante alcalino (SOLUPAN), sendo encaminhado ao atendimento veterinário para realização de exame clínico e laboratorial, que demonstraram intensa alteração hepática e anemia severa. Foi instaurado protocolo terapêutico sintomático com o qual foi possível a reversão total do quadro clínico do paciente.

In addition to being an environmentally healthy and inexpensive technique, the production of land ink can be a facilitator in the teaching-learning process in science. This work presents an experience in the History of Chemistry, in a course in Licentiate in Rural Education, Federal University of Maranhão (UFMA-Bacabal), in which the central theme "the chemistry of pigments" was worked in a contextualized way. The contextualization of the content was carried out through the three categories of the contextualization process (punctual exemplification and motivational character, teaching-learning strategy, formation of critical citizen in decision-making), culminating in the development of a critical-reflective consciousness in the CTSA perspective. As methodological procedures were used: expository class; reading and interpretation of scientific articles; debates; land ink office; painting and exposition of ceramic artifacts; and evaluation. In this context, the land ink workshop gains featured because it brought scientific, technological and social contexts, contributing to the valorization and recovery of facts and experiences of everyday life, discussing and experiencing the meaning of sustainability, with technology and technique environmentally healthy and low thus increasing the potential of the teaching-learning process.

Em tumores mamários, é comum alterações inflamatórias, causadas principalmente em neoplasias que metastatizam. Este trabalho tem por objetivo avaliar a concentração de Proteína C Reativa, correlacionada as alterações de proteínas totais e albumina em cadelas com carcinoma ductal in situ e demonstrar as alterações hematológicas encontradas. Foram utilizadas 13 cadelas diagnosticadas com carcinoma ductal in situ, triados pelo exame citopatológico e confirmado pela histopatologia. Conclui-se que a PCR é uma proteína de fase aguda que se eleva independente das proteínas totais e albumina e as alterações hematológica são de pouca relevância para o diagnóstico de carcinoma ductal in situ.

Critérios de inclusão: disponíveis na íntegra, em português, inglês e espanhol, sem limitação de ano; e os de exclusão: artigos que não responderam à questão de pesquisa, repetidos e provenientes de outras fontes científicas.Resultados as produções científicas revelaram que a estimulação sensório-motora-oral, sucção não nutritiva e avaliação de parâmetros hemodinâmicos favorecem a transição sonda-oral para o aleitamento materno.Conclusão foram identificadas evidências que comprovam que o uso da estimulação e o desenvolvimento das habilidades facilitam, em menor tempo, o início da alimentação oral.

O exame citopatológico foi determinante para a visualização de formas amastigota de Leishmania spp, sendo possível o diagnóstico de leishmaniose.

O mastocitoma é o tumor cutâneo que mais acomete os cães. A citopatologia é um exame que tem por finalidade diferir processos infecciosos de processos neoplásicos. A histopatologia diferencia os mastocitomas em diferentes graus. No entanto, há a necessidade de uma padronização do exame citopatológico para a diferenciação dos mastocitomas em benignos ou malignos. Este trabalho teve como objetivo demonstrar a capacidade de diferenciação de mastocitomas benignos e malignos pelo exame citopatológico, comparando com o resultado da histopatologia. Foram utilizados 19 cães com nódulos cutâneos, diagnosticados com mastocitoma. As lâminas citológicas e histológicas foram avaliadas separadamente para não haver influência no diagnóstico. O exame citológico demonstrou ser capaz de demonstrar diferentes graus de malignidade, limitando-se aos critérios nucleares e de invasividade.

O autocriticismo pode ser entendido como uma autoavaliação prejudicial que implica sentimentos de inutilidade e culpa quando o sujeito se depara com situações de fracasso. Objetivos: explorar a estrutura latente e qualidades psicométricas da Escala das Formas do Autocriticismo e Autotranquilização (FSCRS-C) da população infantil portuguesa. Método: o FSCRS-C foi administrado a 249 crianças açorianas. Resultados: verifica-se uma estrutura bidimensional que incluiu o autocriticismo e autotranquilização. As subescalas apresentaram uma boa consistência interna. O FSCRS-C apresentou uma boa validade divergente e convergente. Conclusão: a versão adaptada para crianças fornece uma medida robusta a ser utilizada em estudos visando o estudo do desenvolvimento deste construto.

ABSTRACT Objective: to investigate protocols used by Speech Therapists to evaluate breastfeeding. Methods: this is an integrative literature review that aims to synthesize available evidence on the evaluation of breastfeeding, from 2002 to 2015. The inclusion criteria were scientific articles that approached breastfeeding evaluation performed by Speech Therapists, published in English and Portuguese. Papers that did not present summaries in their entirety were discarded. Results: a total of 140 studies were found, 134 being excluded. The remaining six publications evaluated breastfeeding and were included in the study with four instruments. The articles selected aimed at evaluating the mothers' knowledge about breastfeeding and its relation to language, the first feeding of the premature baby, describing the initial conditions of breastfeeding, relating breastfeeding to the cup/bottle supply and evaluating orofacial characteristics and breastfeeding. The protocols evaluated aspects of the infant's state of consciousness, behavior and positioning, mother's breastfeeding behavior, breast characteristics, latching and milking, and conditions after breastfeeding. Conclusion: there are few studies in Speech Therapy that use standardized and validated protocols for breastfeeding evaluation. The major focus is on the evaluation of preterm infants. It is suggested that new studies be performed, especially with a focus on validating instruments.

A aplicação de nitrogênio (N) no momento e dose adequados pode aumentar sua eficiência na cultura da aveia, incrementando o rendimento de grãos por meio de alterações nos componentes de produção. No entanto, altas doses deste nutriente podem resultar no acamamento das plantas, o que pode ser contornado com uso de redutores de crescimento. As respostas dos cereais as doses de N e ao redutor são variáveis de acordo com o genótipo e o ambiente de cultivo. Neste sentido, objetivou-se avaliar os componentes de rendimento, o acamamento e a produtividade de grãos de cultivares de aveia branca granífera cultivadas em dois ambientes, sob diferentes doses de nitrogênio, associadas ao redutor de crescimento trinexapac-ethyl. Os experimentos foram conduzidos nos municípios de Londrina e Mauá da Serra, Paraná, sob o delineamento experimental de blocos casualizados com parcelas subdivididas, com quatro repetições. As parcelas constaram de quatro cultivares de aveia branca (IPR Afrodite, IPR Artemis, URS Corona e URS Guria) e as subparcelas de quatro doses de nitrogênio (N) em cobertura (0, 30, 60 e 90 kg ha-1). Em cada local foram conduzidos dois experimentos, com e sem aplicação do redutor de crescimento. Foram avaliados: altura de plantas, número de panículas m-2, número de grãos por panícula, massa de mil grãos, acamamento de plantas e produtividade de grãos. Nas condições em que os experimentos foram conduzidos, a aplicação do redutor de crescimento diminui a altura e o acamamento de plantas e, quando associado à adubação nitrogenada, eleva o número de panículas m-2. O aumento da produtividade de grãos, com a utilização de redutor de crescimento, depende das características do ambiente de cultivo, sendo observado somente em Mauá da Serra. A resposta das cultivares à fertilização nitrogenada de cobertura também depende das características edafoclimáticas dos locais de cultivo.

This article reports on the execution of the #Useful Science project, part of the National Science Science Week (SNCT) of 2017. This project was executed in five rural schools in the micro-region of the Middle Mearim (MA), by teachers and students of the Federal University of Maranhão. The activities developed consisted of eleven workshops and a photographic exhibition, through which information and applications of scientific research associated with popular knowledge were presented to the public formed by the school and local communities, seeking to arouse their attention to the resulting effects of the natural sciences and mathematics in your daily life. In addition, children and young people were encouraged to seek new knowledge and scientific and technological careers. The project had a significant participation of the faculty, students and administrators of the schools served, as well as the community in its surroundings. During its execution, 715 citizens were served. After the final evaluations, we verified that the activities accomplished reached their objectives, contributing to the expansion of the understanding about science and technology, detected through the recognition of their relevance and the connections made with their realities.

O contexto atual da alimentação encontra-se em meio a sistemas agroalimentares cada vez mais segmentados, o que põe em risco as tradições relacionadas à alimentação. Ao se conferir à comida e à culinária o status de bem cultural e identitário, elas passam a ser instrumentos de transmissão, de vivência e de valorização das tradições. Esse aparente estratégico papel de receitas e produtos típicos para o desenvolvimento regional impulsionou a instituição dodecreto nº 4868, Programa RS MAIS GASTRONOMIA/Casa Civil, do Governo do Estado do Rio Grande do Sul com a finalidade de resgatar, pesquisar, catalogar e divulgar os assuntos referentes à gastronomia gaúcha, que atuou de 2011 a 2015. Este artigo deriva de um projeto de pesquisa vinculado a este programa e teve como objetivo verificar o cenário das tradições gastronômicas dos principais grupos étnicos do estado, tendo como recorte para a pesquisa de campo, a região centro sul, composta pelas cidades: Charqueadas, Jaguarão, Pelotas, Piratini, Rio Grande, Santa Vitória do Palmar, Turuçu e Porto Alegre. Para tanto, utilizou-se uma abordagem qualitativa, com pesquisa documental e entrevistas semiestruturadas. Foram realizadas 50 entrevistas com donos de restaurantes, agricultores, representantes de grupos étnicos e agentes do poder público. Os dados foram analisados com auxílio do software NVIVO versão 10. A partir das entrevistas infere-se que, em meio ao processo de globalização e consumo cotidiano de produtos processados, muitas receitas, técnicas e pratos tradicionais foram se modificando ao longo do tempo pela busca de praticidade e pela dificuldade de adquirir os ingredientes típicos, principalmente no meio urbano, onde se nota uma desconexão na relação produtor-ingrediente-consumidor. No entanto, essas tradições mantêm-se vivas nos núcleos familiares e nas comunidades, preparadas principalmente aos domingos e dias de festa, manifestando, assim, as tradições que compõem a identidade dos grupos.
