Research has been conducted on the chiral secondary alcohols used as synthetic intermediates. The identification and isolation of the alcohol oxidases which can catalyze the secondary alcohol's irreversible oxidation and do not require the nicotinamide cofactors are described.

Abstract

The use of methyltransferases (MTs) in industrial biotechnology to replace toxic alkylating agents is of increasing interest. Carboxyl MTs (CMTs) are a subgroup of MTs that methylate the hydroxyl oxygen of carboxylic acids. Research initially focussed on their natural functions in protein regulation and production of volatile methyl esters in plants. In this review we highlight this potentially valuable group of enzymes that show promise for formation of a wide range of structurally diverse methyl esters from the parent acids under aqueous conditions. CMTs have been used to generate intermediates for biofuels, bioplastics and pharmaceuticals. These biocatalysts could also be integrated into cascades with other enzymes such as acyltransferases that function under aqueous conditions. Recent approaches for regenerating the required cofactor S‐adenosylmethionine (SAM) are discussed including in vitro recycling, improvement in in vivo production and the use of more stable analogues. Advances in these areas will further improve the potential to use carboxyl MTs in industrial biotechnology. [ABSTRACT FROM AUTHOR]

Abstract

For enzyme-catalysed biotransformations, continuous in situ detection methods minimise the need for sample manipulation, ultimately leading to more accurate real-time kinetic determinations of substrate(s) and product(s). We have established for the first time an on-line, real-time quantitative approach to monitor simultaneously multiple biotransformations based on UV resonance Raman (UVRR) spectroscopy. To exemplify the generality and versatility of this approach, multiple substrates and enzyme systems were used involving nitrile hydratase (NHase) and xanthine oxidase (XO), both of which are of industrial and biological significance, and incorporate multistep enzymatic conversions. Multivariate data analysis of the UVRR spectra, involving multivariate curve resolution-alternating least squares (MCR-ALS), was employed to effect absolute quantification of substrate(s) and product(s); repeated benchmarking of UVRR combined with MCR-ALS by HPLC confirmed excellent reproducibility.

Abstract

A one-pot tandem enzyme reaction using galactose oxidase M3–5 and aldehyde oxidase PaoABC was used to convert hydroxymethylfurfural (HMF) to the pure bioplastics precursor FDCA in 74% isolated yield. A range of alcohols was also converted to carboxylic acids in high yield under mild conditions.
