Abstract Background Cormus domestica (L.) is a monophyletic wild fruit tree belonging to the Rosaceae family, with well-documented use in the Mediterranean region. Traditionally, these fruits are harvested and stored for at least 2 weeks before consumption. During this period, the fruit reaches its well-known and peculiar organoleptic and texture characteristics. However, the spread of more profitable fruit tree species, resulted in its progressive erosion. In this work we performed proteomic and metabolomic fruit analyses at three times after harvesting, to characterise postharvest physiological and molecular changes, it related to nutritional and organoleptic properties at consumption. Results Proteomics and metabolomics analysis were performed on fruits harvested at different time points: freshly harvested fruit (T0), fruit two weeks after harvest (T1) and fruit four weeks after harvest (T2). Proteomic analysis (Shotgun Proteomic in LC-MS/MS) resulted in 643 proteins identified. Most of the differentially abundant proteins between the three phases observed were involved in the softening process, carbohydrate metabolism and stress responses. Enzymes, such as xyloglucan endotransglucosylase/hydrolase, pectin acetylesterase, beta-galactosidase and pectinesterase, accumulated during fruit ripening and could explain the pulp breakdown observed in C. domestica. At the same time, enzymes abundant in the early stages (T0), such as sucrose synthase and malic enzyme, explain the accumulation of sugars and the lowering of acidity during the process. The metabolites extraction from C. domestica fruits enabled the identification of 606 statistically significant differentially abundant metabolites. Some compounds such as piptamine and resorcinol, well-known for their antimicrobial and antifungal properties, and several bioactive compounds such as endocannabinoids, usually described in the leaves, accumulate in C. domestica fruit during the post-harvest process. Conclusions The metabolomic and proteomic profiling of the C. domestica fruit during the postharvest process, evaluated in the study, provides a considerable contribution to filling the existing information gap, enabling the molecular and phytochemical characterisation of this erosion-endangered fruit. Data show biochemical changes that transform the harvested fruit into palatable consumable product.

Abstract Nowadays, secondary raw materials (SRM) obtained from plant matrices are of great interest for circular economy, suitable for sustainable measures to reduce environmental impact. This work focused on the extraction, characterization and quantification of compounds obtained from leaves and fruits of the Sicilian sumac, Rhus coriaria L. and their application as natural dyes on textile fibres. Extractions were performed with Extractor Naviglio®, maceration and ultrasound assisted methods and food-grade solvents (aqueous and hydroalcoholic) to evaluate the yields for dye compounds. The presence of colouring molecules was evaluated by UV–Vis spectrophotometer, and the extracts selected for colouring were quantified and characterized by LC–MS. The results showed that Extractor Naviglio® achieved the best extraction yield, and the ethanol–water mixture extracts had a higher amount of total phenolic compounds (TPC) and a higher content of total colouring compounds (TCC). These extracts were selected for subsequent applications as dyes for linen, cotton and wool. The chemical profile of selected extracts was rich in compounds such as gallotannin and anthocyanin class. Fibre dyeing was verified by recording CIELAB colouring coordinates. The results suggest that the dyes obtained from R. coriaria can be of great interest for artisanal and industrial processes, in accordance with environmental sustainability.

The present study gathers mineralogical and isotopic geochemical data of 45 soil samples collected from the La Guardiense and Cantina di Solopaca wineries (Southern Italy) in order to provide useful information for the geotraceability of Falanghina white wine terroir. Mineralogical analyses show the ubiquitous presence of quartz, frequent and moderate contents of feldspars and phyllosilicates (i.e., illite/mica, kaolinite, and smectite). Rhizosphere soils from La Guardiense vineyards show a strontium isotope signature (87Sr/86Sr) ranging from 0.708604 to 0.711234 (average 0.710334; 1σ = 0.000943; n = 7) for the total fraction and a narrower range (from 0.706907 to 0.708807; average 0.708120; 1σ = 0.000641; n = 7) for the bioavailable one. For the rhizosphere soils collected from the vineyards of Cantina di Solopaca winery, 87Sr/86Sr ratios range from 0.708268 to 0.712413 (average 0.710145; 1σ = 0.001622; n = 8) and from 0.707968 to 0.709157 (average 0.708384; 1σ = 0.000409; n = 8) for the total and the bioavailable fraction, respectively. Actually, the bioavailable Sr fraction shows a narrower range of 87Sr/86Sr ratios compared to that of the total fraction, making the former more suitable for any study of wine traceability.

Vitis vinifera L. is a natural source of bioactive compounds that is already used for cosmeceutical and nutraceutical approaches. However, their phytochemical and antioxidant properties, although studied, have not been fully explored. We aimed to characterize V. vinifera L. cv. Falanghina seed extracts in different polarity solvents (hexane, ethyl acetate, ethanol, and a mixture of acetone–water) for their phytochemical contents, including the total phenolic compound content (TPC), free radical scavenging capacities, and antioxidant ability on HepG2 cells. We directly profiled the functional quality of V. vinifera seed extracts against H2O2-induced oxidative stress in HepG2 cells, focusing on mitochondrial functions. The content of bioactive compounds was characterized by LC-MS. To assess the cytocompatibility of the extracts, a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was conducted. Results showed that extraction with ethyl acetate (18.12 mg GAE·g−1) and ethanol solvents (18.07 mg GAE·g−1), through Soxhlet, and with an acetone–water mixture (14.17 mg GAE·g−1), through maceration, yielded extracts rich in (poly)phenols, with good scavenging and antioxidant activity (98.32 I% for ethanol solvents and 96.31 I% for acetone–water mixture). The antioxidant effect of polyphenols is at least partially due to their capacity to maintain mitochondrial biogenesis and mitophagy, which elevates mitochondrial efficiency, resulting in diminished ROS production, hence re-establishing the mitochondrial quality control. These findings highlight the valorization of Vitis by-products to improve food functional characteristics.

The valorisation and conservation of plant genetic resources (PGRs) and wild fruit PGRs are critical to ensure the maintenance of genetic and cultural heritage and to promote new perspectives on resource use. New strategies to characterize PGRs are needed, and the omics approach can provide information that is still largely unknown. The Strawberry tree (Arbutus unedo L.) is an underutilized, drought and fire-resistant species distributed in the Mediterranean area and its berries have large ethnobotanical use. Although their phenolic profile and antioxidant capacity are known, they are not well characterised, particularly from a proteomic perspective. The aim of this work is the characterisation of two ecotypes of A. unedo (Campania and Sicily) from a molecular viewpoint to valorise and encourage the preservation of this wild fruit. Samples were collected from two different geographical areas to assess whether different geographical conditions could influence the characteristics of leaves and fruits at the three stages of ripening (green, veraison, red). Proteomic analysis identified 904 proteins, of which 122 showed significance along the ripening. Some of these differentially abundant proteins, such as chalcone synthase, show a marked increase during ripening. The protein functional classes with the highest representation are involved in protein and amino acid metabolism, glycolysis and in secondary metabolism. From a proteomic perspective, there are no differences between the fruits from the two regions compared by the ripening stage. However, the pedoclimatic metabolic imprinting allowed the observation of good diversity in the metabolomic profiles between the two ecotypes, especially for anthocyanins, 4 times more abundant in the Sicilian veraisoned fruit than in the Campania one, and catechins, with double the abundance in the Campania ecotype compared to the Sicilian ecotype in the green phase, but more abundant (3x) in the Sicilian veraisoned fruit. Phenolic compounds show a 20% greater abundance in the Campania green arbutus fruit than in the Sicilian one, values that then equalise as ripening progresses. Multi-omic characterisation enhanced the knowledge on a wild fruit plant species which shows specific adaptations and responses to the environment to be considered when addressing the issue of local agrobiodiversity.

Abstract A tool for urban forest Ecosystem services (ES) and disservices (ED) assessment has been developed to visualize (i) overall ES and ED value, (ii) ES–ED trade-off and (iii) explore principal influences in ES and ED provision. The Real Bosco di Capodimonte (RBC) (Naples, Southern Italy) has been chosen as a case study. ES and ED linked to urban forest plant cover were: biodiversity, carbon storage, gross and net carbon sequestration, lessen runoff, oxygen production, air pollution removal, UV effects reduction, pollen-related allergenicity risk, and volatile organic compounds (VOCs) emissions. A phytosociological survey was conducted and biodiversity value was evaluated. ES and ED were assessed by i-Tree Eco model and Index of Urban Green Zones Allergenicity (IUGZA). Results showed that 441 different plant species occur in the RBC and the most represented genera are Quercus and Trifolium, while the largest family was Asteraceae. Carbon storage and pollution removal were highest in natural forest, while remaining ES were greater in managed forest areas. Highest value for VOCs emission and allergenicity were assigned to managed and natural forest, respectively. Managed forest scored the highest ES–ED value, while managed grassland scored the lowest. Results highlighted the greater influence of plant cover structure in overall ES and ED provision levels, and management influence considering the same type of plant cover. The model could be a valuable tool for ES and ED effective management generally applicable in urban forests.

The rhizosphere effect occurring at the root-soil interface has increasingly been shown to play a key role in plant fitness and soil functionality, influencing plants resilience. Here, for the first time, we investigated whether the rootstock genotype on which Vitis vinifera L. cultivar Falanghina is grafted can influence the rhizosphere microbiome. Specifically, we evaluated to which extent the 5BB and 1103P rootstocks are able to shape microbial diversity of rhizosphere environment. Moreover, we explored the potential function of microbial community and its shift under plant genotype influence. We investigated seven vineyards subjected to the same pedo-climatic conditions, similar age, training system and management and collected twelve rhizosphere soil samples for metagenomic analyses and composite soil samples for physical-chemical properties. In this study, we used 16S rRNA gene-based metagenomic analysis to investigate the rhizosphere bacterial diversity and composition. Liner discriminant analysis effect size (LEFSe) was conducted for metagenomic biomarker discovery. The functional composition of sampled communities was determined using PICRUSt, which is based on marker gene sequencing profiles. Soil analyses involved the determination of texture, pH, Cation Exchange Capacity (CSC), Organic Carbon (OC), electrical conductivity (EC), calcium (Ca), magnesium (Mg), potassium (K) content, Phosphorous (P), nitrogen (N). The latter revealed that soil features were quite homogenous. The metagenomic data showed that the bacterial alpha-diversity (Observed OTUs) significantly increased in 1103P rhizosphere microbiota. Irrespective of cultivar, Pseudomonadota was the dominant phylum, followed by Actinomycetota > Bacteroidota > Thermoproteota. However, Actinomycetota was the major marker phyla differentiating the rhizosphere microbial communities associated with the different rootstock types. At the genus level, several taxa belonging to Actinomycetota and Alphaproteobacteria classes were enriched in 1103P genotype rhizosphere. Investigating the potential functional profile, we found that most key enzyme-encoding genes involved in N cycling were significantly more abundant in 5BB rootstock rhizosphere soil. However, we found that 1103P rhizosphere was enriched in genes involved in C cycle and Plant Growth Promotion (PGP) functionality. Our results suggest that the different rootstocks not only recruit specific bacterial communities, but also specific functional traits within the same environment.

This work focused on the extraction, quantification, and characterization of bioactive compounds of Arbutus unedo L. fruits, comparing the results obtained from the different ripening states. Extractions were performed by different methods (such as maceration extraction and ultrasonic extraction) and food grade solvents (aqueous and hydroalcoholic solvents) in each of the all ripening states (four states considered, associated with four different colors, i.e., green, yellow, orange, and red). The presence of (poly)phenols was quantified and characterized, and scavenging activity was determined by the Folin–Ciocâlteu reagent and the DPPH method, respectively. The content of bioactive compounds was characterized by LC-MS/MS, such as multiple reaction monitoring (MRM) mass spectrometry. The results showed that ultrasound-assisted extraction (UAE) performed better than maceration extraction; ethanol–water mixture extracts showed a more positive effect than the use of aqueous extracts regarding the content of total phenolic compounds. Overall, the total phenolic compounds in the EtOH:H2O mixture at a ratio of 7:3 (v:v) were higher than that of the other solvents for both extraction methods. Some bioactive molecules were characterized for the first time in the extracts of A. unedo. The chemical profile of the strawberry tree extracts depended on the degree of fruit ripeness. The results suggest that A. unedo fruits may be of great interest for food and nutraceutical applications.

Soil pollution is a pressing problem requiring solutions that can be applied without large-scale side effects directly in the field. Phytoremediation is an effective strategy combining plant and root-associated microbiome to immobilize, degrade, and adsorb pollutants from the soil. To improve phytoremediation, it is necessary to think of plants, fungi, and bacteria not as individual entities, but as a meta-organism that reacts organically, synergistically, and cooperatively to environmental stimuli. Analyzing the tripartite enzymatic activity in the rhizosphere is necessary to understand the mechanisms underlying plant–microorganism communication under abiotic stress (such as soil pollution). In this work, the potential of a microbial consortium along with a plant already known for its phytoremediation capabilities, Schedonorus arundinaceus (Scheb.) Dumort., was validated in a mesocosm experiment with pluricontaminated soil (heavy metals, PAHs, and PCBs). Chemical analyses of the soil at the beginning and end of the experiment confirmed the reduction of the main pollutants. The microscopic observation and chemical analyses confirmed the greater root colonization and pollutant removal following the microbial treatment. To obtain a taxonomic and functional picture, tripartite (plant, fungi, and bacteria) enzyme activity was assessed using a metatranscriptomic approach. Total RNA was extracted from a sample of rhizosphere sampled considering 2 centimeters of root and soil attached. From the total reads obtained, mRNAs were filtered, and analysis focused on reads identified as proteins with enzymatic activity. The differential analysis of transcripts identified as enzymes showed that a general increase in potential enzyme activity was observed in the rhizosphere after our biotechnological treatment. Also from a taxonomic perspective, an increase in the activity of some Phyla, such as Actinobacteria and Basidiomycota, was found in the treated sample compared to the control. An increased abundance of enzymes involved in rhizospheric activities and pollutant removal (such as dehydrogenase, urease, and laccase) was found in the treated sample compared to the control at the end of the experiment. Several enzymes expressed by the plant confirmed the increase in metabolic activity and architectural rearrangement of the root following the enhancement of the rhizospheric biome. The study provides new outcomes useful in rhizosphere engineering advancement.

In this contribution, new data concerning bryophytes, fungi, and lichens of the Italian flora are presented. It includes new records and confirmations for the bryophyte genera Encalypta, Grimmia, and Riccia, for the fungal genera Hericium, Inocybe, Inocutis, Pluteus, and Russula, and for the lichen genera Bryoria, Farnoldia, Hypocenomyce, Lecania, Paracollema, Peltigera, Sarcogyne, and Teloschistes.

A 59-year-old female ex-smoker with 40 pack year smoking history and a 5-year current e-cigarette (EC) use history, presented with progressive dyspnea on exertion and daily cough for 2 months. A CT scan showed a consolidation area with air bronchogram in the middle lobe and non-calcific bilateral nodules, which could be attributed to community-acquired pneumonia. The patient was treated with empiric antibiotics and systemic steroids for 10 days. Infectious, neoplastic and autoimmune pathologies were excluded, whereas a broncho-alveolar lavage revealed an accumulation of lipids in the cytoplasm of the alveolar macrophages. Despite the recommendation of vaping cessation, the patient continued to use EC. A new CT exam, carried out after 18 months, showed reversed halo sign (RHS), patchy ground-glass opacity (GGO), pleuro-parenchymal bands, and indeed perilobular pattern, suggestive of organizing pneumonia (OP). The final diagnosis was E-cigarette, or vaping, product use Associated Lung Injury (EVALI)- related OP.

Pulmonary hamartomas represent the most frequent family of benign lung tumors that typically involve the lung parenchyma and only rarely grow as endobronchial tumors. The elective treatment of endobronchial hamartoma is the bronchoscopic resection, and in those cases in which tumor extension and localization makes it not possible, surgical treatment must be evaluated. Patients with symptomatic COVID-19, hospitalized, frequently undergo a chest CT scan and in some cases, occasional findings may emerge, requiring diagnostic investigations such as bronchoscopy and interventional pulmonology procedures. Therefore, in such a delicate pathological condition, such as COVID-19, the need to perform bronchoscopy and interventional pulmonology procedures, minimizing the risk of viral transmission and ensuring necessary assistance, represents a great challenge for pulmonologists. In this article authors describe, for the first time in literature, a rare case of endobronchial hamartoma, radically resected using a single use bronchoscope, in a young female patient hospitalized for symptomatic COVID-19.

The intra- and interdomain phyllosphere microbiome features of Quercus ilex L. in a Mediterranean context is reported. We hypothesized that the main driver of the phyllosphere microbiome might be the season and that atmospheric pollutants might have a co-effect. Hence, we investigated the composition of epiphytic bacteria and fungi of leaves sampled in urban and natural areas (in Southern Italy) in summer and winter, using microscopy and metagenomic analysis. To assess possible co-effects on the composition of the phyllosphere microbiome, concentrations of particulate matter and polycyclic aromatic hydrocarbons (PAHs) were determined from sampled leaves. We found that environmental factors had a significative influence on the phyllosphere biodiversity, altering the taxa relative abundances. Ascomycota and Firmicutes were higher in summer and in urban areas, whereas a significant increase in Proteobacteria was observed in the winter season, with higher abundance in natural areas. Network analysis suggested that OTUs belonging to Acidobacteria, Cytophagia, unkn. Firmicutes(p), Actinobacteria are keystone of the Q. ilex phyllosphere microbiome. In addition, 83 genes coding for 5 enzymes involved in PAH degradation pathways were identified. Given that the phyllosphere microbiome can be considered an extension of the ecosystem services offered by trees, our results can be exploited in the framework of Next-Generation Biomonitoring.

A full-scale mesocosm study was conducted to depict how integrated biological systems interact to adapt to contaminant stress and improve remediation of polycyclic aromatic hydrocarbons (PAHs)contaminated soils. The combination of Verbascum sinuatum L. and microbial consortium (fungi and bacteria) was employed along with three differently contaminated soils. After 240 days the highest PAHs removal (up to 68 %) and 6-rings compounds decrease was found in soil with lower pollution and cation exchange capacity. V. sinuatum showed a significant adaptability over time in terms of redox biology. Soil enzyme activities and microscopic evidences proved a rising plant-microorganisms association and a successful mycorrhization, arising from the inoculation of our consortia. In addition, an enhanced richness of PAHs degrading genes was achieved. Microbial co-metabolism, helped by the establishment of complex relationships with hosting plant, demonstrated to be suitable for the degradation of high molecular weight PAHs and represents a biotechnology with great prospects.

Strawberry (Fragaria ananassa Duch.) is one of most important fruit crops cultivated widely in world. In the present study, we have investigated the proteome variation to different commercially available red strawberry varieties, to gain better insight into the variation in allergen content between the different varieties, as well as their proteome variation. In fact, a combination of proteomic tools have been used to build a complete allergen map of strawberry. The water-soluble fraction of a strawberry extract was precipitated using a phenol-based procedure and separated by 2-DE. Further, all known strawberry allergens were localized on a 2-DE map and they were matched with spots recognized by sera of patients with different allergic patterns. A number of constitutive or differentially accumulated proteins were found. The role of the identified proteins, in particular of the allergen proteins, is discussed in relation to the different variety of strawberry. Fruit ripening is a physiologically complex process altering texture, color, flavor, nutritional value, and aroma. However, some fruits are consumed at an early stage of ripening due to the very peculiar characteristics varying during ripening. An example is a particular ecotype of pepper, the Friariello pepper, among the most important representatives of Campania (Southern Italy) agro-alimentary culture. In this study, for the first time, the physiological variations during Friariello ripening (green, veraison, and fully ripe) were evaluated by hyphenated mass spectrometric techniques in a proteomic and metabolomic approach. We found that Lutein and Thaumatin are particularly abundant in the green Friariello. Friariello at an early stage of ripening, is rich in volatile compounds like butanol, 1 3 5-cycloheptatriene, dimethylheptane, α-pinene, furan-2-penthyl, ethylhexanol, 3-carene, detected by gas chromatography-mass spectrometry (GC-MS) analysis, which give it the peculiar fresh and pleasant taste. The detected features of Friariello may justify its preferential consumption in the early ripening stage and outline new knowledge aimed at preserving specific agro-cultural heritage.

Severe Acute Respiratory Syndrome due to Coronavirus-19 (SARS-CoV-2) is caused by combined alveolar-capillary lung damage, with bilateral pneumonia and thrombosis, which often causes respiratory failure. Proper COVID-19 management requires high skills in airway control and the need to perform aerosol-generating procedures such as bronchoscopy, which can increase the possibility of virus spreading among healthcare professionals. In an epidemiologically delicate moment, the multidisciplinary decision on “WHEN, HOW and WHY” to perform bronchoscopies minimizing the risk of COVID-19 transmission, represented a great challenge for all specialists engaged in bronchoscopic procedures. In this work authors want to share all technical aspects of 87 videobronchoscopies performed in confirmed or suspected COVID-19 patients, from 3rd to 6th January 2020, describing the reason, the organizational and operational model and patients characteristics. Was also evaluated the impact of high-risk procedures such as bronchoscopy on the personnel involved. The disclosure of all technical details, represents, in the opinion of the authors, an important contribution, capable of providing support to all physicians engaged in bronchoscopy procedures in confirmed or suspected COVID-19 patients.

The contamination of soil with total petroleum hydrocarbons (TPH) may result in dramatic consequences and needs great attention, as soil rehabilitation would need more effort from a sustainability perspective. However, there is still no known general method since the remediation technology is strictly site-specific. Adaptive biological system dynamics can play a key role in understanding and addressing the potential of situ-specific biological combinations for soil pollutants removal. The potential worst-case of TPH contamination reflects soil affected by heavy industrial activities, such as oil refineries. Therefore, the experimental trial was conducted on a 2,000 m2 area from a contaminated site located in northern Italy. We evaluated the remediation potential over time (270 days) assessing (i) the phytoremediation efficiency of two species of Poaceae (Festuca arundinacea Schreb. and Dactylis glomerata L.) and two species of Fabaceae (Medicago sativa L. and Lotus corniculatus L.) and (ii) the role of the indigenous bacteria flora and endo-mycorrhizae consortium addition in plant growth promotion. We also induced resistance to contamination stress in a field experiment. Thirty-three indigenous bacteria selected from the contaminated soils showed marked plant growth promotion. Moreover, functional metagenomics confirmed the metabolic capability of hydrocarbon-degrading microorganisms living in the polluted soil. Our data showed that soil enzymatic activities increased with hydrocarbon degradation rate after 60 days. Both Poaceae and Fabaceae resulted in remarkable remediation potential. Stress markers and antioxidant activity indicated that the selected plant species generally need some time to adapt to TPH stress. In conclusion, our evaluation implied both the rhizosphere effects and functional features of the plant and suggested that plants should (i) have marked tolerance to specific contaminants, (ii) be characterized by an extensive root system, and (iii) be susceptible to arbuscular mycorrhizal fungi (AMF) infection.

Trans-bronchial needle aspiration allows lymph node sampling in several thoracic conditions; the ability of Rapid On-Site Evaluation (ROSE) to predict the final diagnosis in this setting has not been well characterized. We performed a retrospective study to establish the utility of ROSE in the diagnosis of thoracic diseases with mediastinal lymph node involvement. We retrospectively reviewed 297 patients with hilar-mediastinal lymph node enlargement detected at CT scan from January 2013 to April 2016. 201 patients underwent conventional TBNA; in 96 patients, TBNA procedure was performed by on-site presence of a team of pathologists and research morphologists. Lung neoplasms, sarcoidosis, infections and lymphoma were the most common diseases diagnosed with TBNA samples. This work focused on the recovery and application of bioactive and functional compounds (BFC) from a waste matrix, the Opuntia ficus-indica (L.) Miller fruit peel (OFP), using different extraction methods: maceration and Extractor Naviglio®. The presence of BFC was initially evaluated by following the extraction yields and antioxidant activity using the Folin-Ciocȃlteu reagent and the DPPH method, respectively. The quali-quantitative content of BFC was also verified with UHPLC/UV-ESI-HRMS profile of OFP extracts. The evaluation of the BFC yields is aimed at using extract components, such as betanins, in the technological application as textile dye based on natural pigments: we evaluated the stability of the extracted dyes, tested the dyeing power and assessed the stability of the dye on the textile material. The results showed different compositions depending on the polarities of the solvents. The ethanol-water mixture extracts obtained using Naviglio method revealed an important presence in terms of total phenolic compound yields. Wool, linen and cotton fibers were chosen for the dyeing tests. Rock alum and lemon juice, two etching methods, were used. Results suggest that the OFP may be of great interest as a natural source of BFC for food, nutraceutical and technological applications in the sustainability context.

This article offers statistical analyses of trace elements (TEs) in soils and plants through a Pearson correlation matrix. The main objectives were the assessment of soil TEs sources and the evaluation of native plant response to physical and chemical characteristics of a TEs contaminated soil. Data were collected from Bagnoli brownfield site (Southern Italy). Interpretation of the data, can be found in “Identification of native-metal tolerant plant species in-situ: environmental implications and functional traits” [1]. Nowadays, anthropic pressures are continuously transforming the landscape mosaic, leading to issues related to habitat fragmentation and the loss of ecosystem functions. In this study, a landscape-change analysis over the 1990–2018 timeframe of the case study area of Metropolitan City of Naples (MCN) (southern Italy) was performed to evaluate trends, causes, and results of the landscape transformations. We preliminary performed a spatial–temporal fragmentation-expansion analysis in the MCN through the landscape expansion index (LEI), and subsequently determined the impacts on the eco-mosaic. The multitemporal analysis (1990–2018) highlighted an urban expansion in the MCN. The LEI analysis depicted a dual tendency in the increased fragmenting of the MCN’s eco-mosaic. The urban landscape has compacted in the hollow areas, and in the meantime there has been a sprawling expansion of the urban fabric. The most impacted land-use category was “Agricultural areas”, of which 57.42 km2 have been lost in 28 years, with negative impacts on ecological connectivity. Finally, we investigated the relationship between the increase and type of landscape fragmentation and the demographic flows over time. We found that the areas affected by demographic growth were the same ones characterized by an edge expansion of the urban patterns. Conversely, where there has been a demographic decrease, an infilling behavior of urban fabric has been noted. This study highlights a possible correlation between the expansion type of the urban fabric and the demographic flows in a region as a main driver. The outcome of 30-year transformations is the actual ecological network in which Core Areas, Buffer Zones, and Primary Connections covered 35.3%, 11.1%, and 39.6%, respectively, of the total surface of the MCN, covering an overall area of 1008.4 km2.

In this work, the effect of dyes extracted through the Naviglio method (an eco-innovative solid-liquid extraction technique) was tested, which proved interesting for such an extraction. The dyes extracted from Opuntia ficus-indica (L.) Miller waste were treated to maximize the extraction of the coloring molecules. The extraction method was compared with conventional methods under the same conditions. The results showed that the extracts obtained with the different techniques, in the same extraction conditions (solvent, pH, temperature, time), were richer in the pigments for the extract obtained with the Naviglio method. The stability of the dyes was tested before the staining. A plant-based fiber, cotton, as well as an animal-based fiber, wool, were chosen for the staining tests. For the two types of fiber, two etching methods were used: one with rock alum, a method widely used to fix a color and one with lemon juice, a method used for a long time by rural populations who practiced the coloring of hides and tissues. In addition, the qualitative content of the pigments was also verified with an HPLC profile of the Opuntia ficus-indica (L.) Miller extracts. Ultimately, the results suggest that the Opuntia ficus-indica (L.) Miller fruit can be of great interest as a natural source of dyes and be used for tinctures.

Total petroleum hydrocarbons (TPHs) are a persistent environmental organic contaminant. The possibility of obtaining synergistic effects between plants and microorganisms has further increased the possibility of alternative techniques for bioremediation. Oloptum miliaceum (L.) Röser & H.R. Hamasha and Pennisetum setaceum (Forssk.) Chiov. are, undoubtedly, good model plants for phytoremediation because they have large roots, leaf biomass, and a fast and effective renewal capacity, also, they have a great capacity to host endophytes in their roots. Gas chromatography-mass spectrometry (GC-MS) based on carbon fraction number was a basic technique used to determine the hydrocarbon degradation, and microorganism’s population was identified by high-throughput sequencing of 16s rRNA. The microbial consortium used allows the plant to increase overall biomass, adapt more in terms of redox biology (Superoxide dismutase SOD, catalase CAT, ascorbate peroxidase APX, guaiacol peroxidase GPX), and stress markers (Glutathione S-transferase GST, Phenylalanine Ammonia Lyase PAL, Proline content, and lipid peroxidation MDA). In addition, the photosynthetic efficiency and the soil dehydrogenase activity were monitored. After 240 days, the percentage of TPHs removed in Group 2 was 94%, whereas in Group 1, it was 78% in Oloptum miliaceum and Pennisetum setaceum. The removal of aliphatic hydrocarbons (C13–C36) was observed in Oloptum miliaceum and Pennisetum setaceum inoculated with the consortium of indigenous bacteria selected from rhizosphere soil and mycorrhizae strains. Our data demonstrate that, the Poaceae, in relation to its great ecological and vegetative potential, could be a great candidate for extensive remediation of soils contaminated by TPHs.

We applied an integrated phytoremediation system by inoculation of rhizosphere microbes on an area of 2,000 m2 that was contaminated by heavy metals. Hybrid poplar (Populus deltoids × Populus nigra) and willow (Salix purpurea subsp. lambertiana) were planted on four parcels of 500 m2. Of these, two parcels were inoculated with an autochthonous consortium specially selected and the other two were not inoculated. The analyses were carried out on plant and soil samples collected at different times: month 0 (T0), 12 (T1), 24 (T2), and 36 (Tf). Total concentrations of HMs in the roots of Populus and Salix showed significant increase in the plants with rhizosphere microflora consortium. The plants treated with the consortium have an increased enzymatic activity of antioxidants and of stress markers. All enzymes showed an important contribution in enzymatic thiol-dependent ROS scavening mechanisms, through its over-expression at time T1 and T2 and a consequent stabilization at Tf which results in adaptive mechanisms. The plant-fungi-bacterium system was a valid candidate for the implementation of phytoremediation systems in the field, since the plants showed a greater capacity for adaptation and accumulation.

A fast and sensitive ultra-performance liquid chromatography tandem mass spectrometry method was developed for the measurement of caffeine in caffeine-laced pants and in urine and skin of a pants user. The substance and its internal standard (N-ethylnorcotinine) were separated by reversed phase chromatography with 5 mM ammonium formate pH 3.0 and 0.3% formic acid in acetonitrile mobile phase (83:17 v/v) by isocratic elution and detected by tandem mass spectrometry operated in multiple reaction monitoring mode via positive electrospray ionization. Linearity was studied from 1.4 to100 ng/mL range for urine, from 5 to 100 ng/cotton swab for skin caffeine and from 1.3 to 100 µg/samples for 4 cm2 textile samples. Good determination coefficients (r2 = 0.99) were found in all cases. At three concentrations spanning the linear dynamic ranges of different samples mean recoveries of caffeine were always higher than 80% and intra-assay and inter-assay imprecision and inaccuracy were always better than 105%. For the first time, caffeine content in this cosmetotextile was determined together with the measurement of caffeine released on the user skin, the absorbed amount with resulting urinary concentrations.

Most common prostaglandin analogs, bimatoprost, latanoprost and travoprost, are licensed for the reduction of elevated intraocular pressure in patients with open angle glaucoma and ocular hypertension, but their non approved use as eyelash enhancers is becoming popular, especially in patients with eyelashes hypotrichosis. A fast and sensitive high performance liquid chromatography tandem mass spectrometry method was developed for the measurement of bimatoprost, latanoprost and travoprost in cosmetic serums freely web-sold to increase eyelash length, thickness and darkness. The analytes and the internal standard (reserpine) were separated by reversed phase chromatography with 5 mM ammonium acetate with 0.02% formic acid (mobile phase A) and 5 mM ammonium acetate in acetonitrile/water (95/5; v/v) with 0.02% formic acid (mobile phase B) by gradient elution and detected with tandem mass spectrometry operated in multiple reaction monitoring mode. Linearity between 1 and 500 μg/g shows good correlation coefficients (r2 = 0.99) for all substances. Analytical recovery of analytes under investigation were always higher than 90% and intra-assay and inter-assay precision and accuracy always better than 11%. This method was successfully applied to analyze cosmetic serums freely sold on the Internet websites.
