Mobility management is a regulatory framework designed to streamline systematic mobility and mitigate energy, environmental and economic impacts. In this work, we propose a flexible methodology for evaluating the sustainability of home-to-work travel, providing a comprehensive and detailed ex post cost–benefit assessment. Specifically, we analyzed the effectiveness of the shuttle service operating in the ENEA “Casaccia” Research Centre in pre-pandemic times. Initially, we conducted an online survey to collect data with the aim of characterizing the travel behavior of the staff and reconstructing the multi-modal individual mobility patterns. Over 70% of the recipients, which amounted to about 950 individuals, completed the survey. Subsequently, we studied two alternative scenarios—with and without the shuttle service—comparing their total mileage, energy consumption, and pollutant emissions and performing an economic analysis. Our findings suggest that operating the service has a significant impact on air pollutants and greenhouse gas emissions, with reductions of 97% for volatile organic compounds, 72% for particulate matter, and 60% for carbon dioxide. Moreover, the cost–benefit analysis reveals that both users and the community reaped benefits from the provision of the collective service. These benefits are estimated to be almost EUR 1.35 M per year.

Abstract Rexinoids are ligands which activate retinoid X receptors (RXRs), regulating transcription of genes involved in cancer-relevant processes. Rexinoids have anti-neoplastic activity in multiple preclinical studies. Bexarotene, used to treat cutaneous T cell lymphoma, is the only FDA-approved rexinoid. Bexarotene has also been evaluated in clinical trials for lung and metastatic breast cancer, wherein subsets of patients responded despite advanced disease. Abstract Mutations in BRCA genes are the leading cause of hereditary breast cancer. Current options to prevent cancer in these high-risk patients, such as anti-estrogen drugs and radical mastectomy, are limited by lack of efficacy, undesirable toxicities, or physical and emotional challenges. We have previously shown that PARP inhibitors can significantly delay tumor development in BRCA1-deficient mice. Here, we fabricated the PARP inhibitor talazoparib (TLZ) into spacer implants (InCeT-TLZ) for localized and sustained delivery. We hypothesized that this novel formulation will provide an effective chemopreventive strategy with minimal toxicity. TLZ was released gradually over 30 days as implants degraded. InCeT-TLZ significantly decreased proliferation and increased DNA damage in the mammary glands of BRCA1-deficient mice. Notably, the number of mice that developed hyperplasia in the mammary glands was significantly lower with InCeT-TLZ treatment compared to the control group. Meanwhile, InCeT-TLZ was also better tolerated than oral TLZ, without loss of body weight or anemia. This study provides proof of concept for a novel and safe chemopreventive strategy using localized delivery of a PARP inhibitor for high-risk individuals. Future studies will directly evaluate the effects of InCeT-TLZ for preventing tumor development.

Abstract Effective drugs are needed for lung cancer, as this disease remains the leading cause of cancer-related deaths. Rexinoids are promising drug candidates for cancer therapy because of their ability to modulate genes involved in inflammation, cell proliferation or differentiation, and apoptosis through activation of the retinoid X receptor (RXR). The only currently FDA-approved rexinoid, bexarotene, is ineffective as a single agent for treating epithelial cancers and induces hypertriglyceridemia. Here, we used a previously validated screening paradigm to evaluate 23 novel rexinoids for biomarkers related to efficacy and safety. These biomarkers include suppression of inducible nitric oxide synthase (iNOS) and induction of sterol regulatory element-binding protein (SREBP). Because of its potent iNOS suppression, low SREBP induction, and activation of RXR, MSU-42011 was selected as our lead compound. We next used MSU-42011 to treat established tumors in a clinically relevant Kras-driven mouse model of lung cancer. KRAS is one of the most common driver mutations in human lung cancer and correlates with aggressive disease progression and poor patient prognosis. Ultrasound imaging was used to detect and monitor tumor development and growth over time in the lungs of the A/J mice. MSU-42011 markedly decreased the tumor number, size, and histopathology of lung tumors compared to the control and bexarotene groups. Territorial energetic and environmental planning provides operational solidity to the concept of sustainable development, in particular in energy-related issues, where recent attention to and social awareness of climate change are driving actions and policies at local and international levels. The goals of the United Nations Agenda 2030 can be reached through the strategy of glocalization, giving more responsibility to local administrations like municipalities. In this work, a scientific methodology is developed and validated to revise Sustainable Energy Action Plans (SEAP) and the monitoring phase of municipalities. The methodology starts from measured data in the territory considered and makes use of specific statistical models in order to estimate the needed data. The methodology considers the energy consumption of the main sectors: residential, transportation, tertiary, and commercial, with a particular focus on municipal competences (public lighting, urban transport, municipal fleet, etc.). Renewable energy is also considered due to its importance in local energy production. In order to go deeper into SEAPs, in this paper, the authors describe the quantitative analysis of the Baseline Emission Inventory, the quantification of the SEAP planning actions, and the definition of the Monitoring Emission Inventory, which is the final step of the planning process. This step was done for nine municipalities of the Abruzzo region with different characteristics (size, population, climate, geographical position, economy, etc.) in order to widen the results of the analysis and test the robustness of the methodology. Indeed, it gave a quantitative dimension to the primary energy consumption and CO2 emissions for 2018, compared with the 2005 baseline values, and the final results are related to the reduction commitments planned for 2020. All the municipalities were considered to have achieved this goal, surpassing the 20% emissions reduction. This validated methodology is also the basis for the development of the Sustainable Energy and Climate Action Plans (SECAPs), which integrate adaptation actions and mitigation ones.

Reforestation is the appropriate natural tool to mitigate the climate change. The authors illustrate how to manage cuts and plantations of trees making profit from unused lands, to reach different carbon capture goals. Unused lands, moreover, are frequently in the neighbouring of Municipalities which often are politically responsible of these territories. Their destination as carbon sink inside the implementation of the Sustainable Energy and Climate Action Plans (SECAPs) is particularly suitable, participating in a synergic way to reduce the CO2 municipal emissions by 55 % in 2030 with respect to the 1990 datum. In the framework of a SECAP, this level is mandatory and participates to the common huge effort to decarbonize energy needs. After having modelled some intrinsic aspects related to the dynamics of the carbon capture due to the growth of trees, the research demonstrated how, with a proper management of cuts and plantations, a new concept of mutualism between city and territory can be designed, recognizing the crucial role of neighbouring unused lands alongside those direct actions usually implemented to reduce the carbon intensity of a city.

Suitability to off-design operation, applicability to combined thermal and electrical generation in a wide range of low temperatures and pressures and compliance with safety and environmental limitations qualify small-scale Organic Rankine Cycle plants as a viable option for combined heat and power generation in the residential sector. As the plants scale down, the electric and thermal output maximization has to account for issues, spanning from high pump power absorption, compared to the electric output of the plant, to intrinsically low plant permeability induced by the expander, to the intermittent availability of thermal power, affected by the heat demand for domestic hot water (DHW) production. The present paper accounts for a flat-plate solar thermal collector array, bottomed by an ORC unit featuring a sliding vane expander and pump and flat-plate heat exchangers. A high-temperature buffer vessel stores artificially heated water – electric heaters, simulating the solar collector - and feeds either the hot water line for domestic use or the ORC evaporator, depending on the instantaneous demand (i.e., domestic hot water or electric power), the temperature conditions inside the tank and the stored mass availability. A low-temperature receiver acts like the heat sink of the ORC unit and harvests the residual thermal power, downstream the expander: a dedicated control, modelled to properly modulate the mass addition/subtraction to this storage unit allows to restore the operating points of the cycle and to limit the incidence of off-design operation, via real-time adjustment of the cycle operating parameters. Indeed, the possibility of continuous ORC generation depends on (i) the nature of the demand and (ii) the amount of hot water withdrawn from the high-temperature buffer vessel. The time-to-temperature for the mass stored inside the buffer affects the amount of ORC unit activations and eventually the maximum attainable generation of electric energy. The plant energy performance is experimentally assessed, and various characteristic operating points are mapped, based on test runs carried out on a real-scale ORC pilot unit.
