BACKGROUND: Colonization of large part of Europe by the Asian tiger mosquito Aedes albopictus is causing autochthonous transmission of chikungunya and dengue exotic arboviruses. While pyrethroids are recommended only to reduce/limit transmission, they are widely implemented to reduce biting nuisance and to control agricultural pests, increasing the risk of insurgence of resistance mechanisms. Worryingly, pyrethroid resistance (with mortality < 70%) was recently reported in Ae. albopictus populations from Italy and Spain and associated with the V1016G point mutation in the voltage-sensitive sodium channel gene conferring knockdown resistance (kdr). Genotyping pyrethroid resistance-associated kdr mutations in field mosquito samples represents a powerful approach to detect early signs of resistance without the need for carrying out phenotypic bioassays which require availability of live mosquitoes, dedicated facilities and appropriate expertise. METHODS: Here we report results on the PCR-genotyping of the V1016G mutation in 2530 Ae. albopictus specimens from 69 sampling sites in 19 European countries. RESULTS: The mutation was identified in 12 sites from nine countries (with allele frequencies ranging from 1 to 8%), mostly distributed in two geographical clusters. The western cluster includes Mediterranean coastal sites from Italy, France and Malta as well as single sites from both Spain and Switzerland. The eastern cluster includes sites on both sides of the Black Sea in Bulgaria, Turkey and Georgia as well as one site from Romania. These results are consistent with genomic data showing high connectivity and close genetic relationship among West European populations and a major barrier to gene flow between West European and Balkan populations. CONCLUSIONS: The results of this first effort to map kdr mutations in Ae. albopictus on a continental scale show a widespread presence of the V1016G allele in Europe, although at lower frequencies than those previously reported from Italy. This represents a wake-up call for mosquito surveillance programs in Europe to include PCR-genotyping of pyrethroid resistance alleles, as well as phenotypic resistance assessments, in their routine activities.

Mosquito control remains a central pillar of efforts to reduce malaria burden in sub-Saharan Africa. However, insecticide resistance is entrenched in malaria vector populations, and countries with a high malaria burden face a daunting challenge to sustain malaria control with a limited set of surveillance and intervention tools. Here we report on the second phase of a project to build an open resource of high-quality data on genome variation among natural populations of the major African malaria vector species Anopheles gambiae and Anopheles coluzzii. We analyzed whole genomes of 1142 individual mosquitoes sampled from the wild in 13 African countries, as well as a further 234 individuals comprising parents and progeny of 11 laboratory crosses. The data resource includes high-confidence single-nucleotide polymorphism (SNP) calls at 57 million variable sites, genome-wide copy number variation (CNV) calls, and haplotypes phased at biallelic SNPs. We use these data to analyze genetic population structure and characterize genetic diversity within and between populations. We illustrate the utility of these data by investigating species differences in isolation by distance, genetic variation within proposed gene drive target sequences, and patterns of resistance to pyrethroid insecticides. This data resource provides a foundation for developing new operational systems for molecular surveillance and for accelerating research and development of new vector control tools. It also provides a unique resource for the study of population genomics and evolutionary biology in eukaryotic species with high levels of genetic diversity under strong anthropogenic evolutionary pressures.

Accurate estimation of population size is key to understanding the ecology of disease vectors, as well as the epidemiology of the pathogens they carry and to plan effective control activities. Population size can be estimated through mark-release-recapture (MRR) experiments that are based on the assumption that the ratio of recaptured individuals to the total captures approximates the ratio of marked individuals released to the total population. However, methods to obtain population size estimates usually consider pooled data and are often based on the total number of marked and unmarked captures. We here present a logistic regression model, based on the principle of the well-known Fisher-Ford method, specific for MRR experiments where the information available is the number of marked mosquitoes released, the number of marked and unmarked mosquitoes caught in each trap and on each day, and the geographic coordinates of the traps. The model estimates population size, taking into consideration the distance between release points and traps, the time between release and recapture, and the loss of marked mosquitoes to death or dispersal. The performance and accuracy of the logistic regression model has been assessed using simulated data from known population sizes. We then applied the model to data from MRR experiments with Aedes albopictus Skuse performed on the campus of "Sapienza" University in Rome (Italy). © 2013 Entomological Society of America.

Speciation with gene flow may be aided by reduced recombination helping to build linkage between genes involved in the early stages of reproductive isolation. Reduced recombination on chromosome-X has been implicated in speciation within the Anopheles gambiae complex, species of which represent the major Afrotropical malaria vectors. The most recently diverged, morphologically-indistinguishable, species-pair, An. gambiae and An. coluzzii, ubiquitously display a ‘genomic island of divergence’ spanning over 4Mb from chromosome-X centromere, which represents a particularly promising candidate region for reproductive isolation genes, in addition to containing the diagnostic markers used to distinguish the species. Very low recombination makes the island intractable for experimental recombination studies, but an extreme hybrid zone in Guinea Bissau offers the opportunity for natural investigation of X-island recombination. SNP-analysis of chromosome-X hemizygous males revealed: (i) strong divergence in the X-island despite a lack of autosomal divergence; (ii) individuals with multiple-recombinant genotypes, including likely double crossovers and localized gene conversion; (iii) recombination-driven discontinuity both within and between the molecular species markers, suggesting that the utility of the diagnostics is undermined under high hybridization. The largely-, but incompletely-protected nature of the X-centromeric genomic island is consistent with a primary candidate area for accumulation of adaptive variants driving speciation with gene flow, whilst permitting some selective shuffling and removal of genetic variation.

Background The main constraint to the fight against container-breeding mosquito vectors of human arboviruses is the difficulty in targeting the multiplicity of larval sources, mostly represented by small man-made water containers. The aim of this work is to assess the feasibility of the “auto-dissemination” approach, already tested for Aedes aegypti, as a possible alternative to traditional, inefficient control tools, against Ae. albopictus in urban areas. The approach is based on the possibility that wild adult females, exposed to artificial resting sites contaminated with pyriproxyfen, can disseminate this juvenile hormone analogue to larval habitats, thus interfering with adult emergence. Methodology We carried out four field experiments in two areas of Rome that are typically highly infested with Ae. albopictus, i.e. the main cemetery and a small green area within a highly urbanised neighbourhood. In each area we used 10 pyriproxyfen “dissemination” stations, 10 “sentinel” sites and 10 covered, control sites. The sentinel and control sites each contained 25 Ae. albopictus larvae. These were monitored for development and adult emergence. Principal Findings When a 5% pyriproxyfen powder was used to contaminate the dissemination sites, we observed significantly higher mortality at the pupal stage in the sentinel sites (50–70%) than in the controls (, Author Summary Aedes albopictus (the Asian Tiger mosquito) is one of the most invasive and aggressive disease vectors in the world. It is a serious public nuisance and a public health risk, due to its ability to transmit pathogens to humans. The control of this mosquito is complicated by the difficulty in targeting either juveniles (due to their ability to colonise myriad and often cryptic domestic and natural water containers) or adults (due to their diurnal and outdoor resting and biting patterns). The aim of our work was to assess the feasibility of a novel approach to control Ae. albopictus in urban areas. This was based on the possibility that wild adult females, exposed to artificial resting sites contaminated with an insect growth regulator, could subsequently contaminate their breeding sites and kill both the larvae and pupae developing therein. The results obtained from field experiments carried out in highly infested areas in Rome demonstrate that this approach has potential as a valid alternative to traditional (and ineffective) larval control efforts in urban areas. We therefore introduce a new perspective in the fight against the Asian Tiger Mosquito.

Visual concept detection is important to access visual information on the level of objects and scene types. The current state–of–the–art in visual concept detection and annotation tasks is based on the bag–of–words model. Within the bag–of–words model, points are first sampled according to some strategy, then the area around these points are described using color descriptors. These descriptors are then vector–quantized against a codebook of prototypical descriptors, which results in a fixed–length representation of the image. Based on these representations, visual concept models are trained. In this chapter, we discuss the design choices within the bag–of–words model and their implications for concept detection accuracy.

This book constitutes the thoroughly refereed proceedings of the 10th Workshop of the Cross Language Evaluation Forum, CLEF 2010, held in Corfu, Greece, in September/October 2009. The volume reports experiments on various types of multimedia collections. It is divided into three main sections presenting the results of the following tracks: Interactive Cross-Language Retrieval (iCLEF), Cross-Language Image Retrieval (ImageCLEF), and Cross-Language Video Retrieval (VideoCLEF).
