Abstract

Lake Lugano is increasingly experiencing new processes and dynamics due to the combined effects of climate change and human activities. In recent years, abnormal phenomena of algal proliferation, known as Harmful Algal Blooms (HABs), have appeared on the lake. Particularly in 2023, after several critical events, public awareness of this issue has risen, as it can potentially impact socio-economic aspects related to the use of this locally important water resource. In this study, we aim to share the development process of a monitoring system designed to promptly inform about the increase in concentration levels, thereby initiating a protocol to support decision-makers. This system, based on open-source and cost-effective devices, is designed to gather information on these blooms, which pose potential toxicity risks to humans and animals. The system uses a Raspberry Pi connected to a Trilux fluorimeter, measuring algal pigments and transmitting data via NB-IoT. The data is processed using istSOS software, stored locally, and transmitted to a data warehouse. Alerts are set up based on phycocyanin (PC) levels, indicating cyanobacterial biomass and potential microcystin toxicity. Abstract

Plastic debris is widespread in freshwater ecosystems, but a rigorous assessment of its global distribution has been hindered by a lack of comprehensive and comparable data. We performed the first standardized global survey of lakes to assess the quantity and type of plastics (>250μm). We included 38 lakes located in 23 different countries distributed across five continents, spanning different environmental gradients and varying levels of anthropogenic stress. All samples were collected by horizontal trawling of a plankton net and subsequently treated with hydrogen peroxide. We identified 9425 plastic particles, which were classified based on shape, color, and size. Polymer identification was carried out using Raman micro-spectroscopy. Our results showed that the concentration of plastics spanned four orders of magnitude (10-3-101 particles/m3). Fibers (49%) and fragments (41%) were the most frequently detected particles, suggesting a secondary origin of plastic contamination. The most commonly identified polymers were polyester (30%), polypropylene (20%), and polyethylene (16%), which are widely used in short life-cycle products and account for the majority of global plastic production. Further, we found that urban-related attributes of lakes/watersheds influenced the occurrence and type of plastics in lentic systems and larger and deeper lakes with higher retention times are accumulating plastic debris at higher concentrations. info:eu-repo/semantics/publishedVersion

Abstract

The majority of microplastic research has focused on seawater, with fewer than 4% of microplasticsrelated studies occurring on freshwaters. The limited available information suggests that the abundance of microplastics in freshwaters is often as high or even higher than marine environments. However, comprehensive investigations on occurrence and fate of microplastics in freshwaters are scarce and highly fragmented, partly because detection and identification of microplastic particles is rather complex. In addition, up to now, harmonized and standardized protocols for the sampling and analysis of microplastics in freshwaters do not exist, and studies with different research aims and hypotheses often report unstandardized results, making comparison among studies difficult. In the present study, we performed the first global standardized sampling and analysis effort to investigate the occurrence and distribution of microplastics in surface water of lakes and reservoirs with different anthropogenic impacts. Participants aim to collect water samples of freshwater systems with different features (e.g., area, depth, thermal behavior, watershed), following a common protocol. This 18 establishes the collection of samples by horizontal trawling of a plankton net and, after treatment with hydrogen peroxide, the polymer identification through micro-Raman spectroscopy. This GLEON project will allow obtaining comparable data about microplastic contamination in different freshwater systems around the globe. Abstract

Telonemiaare one of the oldest identified marine protists that for most part of their history have been recognized as a distinct incertae sedislineage. Today, their evolutionary proximity to the SAR supergroup (Stramenopiles, Alveolates, and Rhizaria) is firmly established. However, their ecological distribution and importance as a natural predatory flagellate, especially in freshwater food webs, still remain unclear. To unravel the distribution and diversity of the phylum Telonemiain freshwater habitats, we examined over a thousand freshwater metagenomes from all over the world. In addition, to directly quantify absolute abundances, we analyzed 407 samples from 97 lakes and reservoirs using Catalyzed Reporter Deposition-Fluorescence in situHybridization (CARD-FISH). We recovered Telonemia18S rRNA gene sequences from hundreds of metagenomic samples from a wide variety of habitats, indicating a global distribution of this phylum. However, even after this extensive sampling, our phylogenetic analysis did not reveal any new major clades, suggesting current molecular surveys are near to capturing the full diversity within this group. We observed excellent concordance between CARD-FISH analyses and estimates of abundances from metagenomes. Both approaches suggest that Telonemiaare largely absent from shallow lakes and prefer to inhabit the colder hypolimnion of lakes and reservoirs in the Northern Hemisphere, where they frequently bloom, reaching 10%–20% of the total heterotrophic flagellate population, making them important predatory flagellates in the freshwater food web.

41. Historical colonisation patterns of dolichospermum lemmermannii (Cyanobacteria) in a deep lake south of the alps

Abstract

Since the beginning of the 1990s, Lake Garda showed the appearance of extended surface water blooms of Dolichospermum lemmermannii (Nostocales, Cyanobacteria). Between the 1990s and the 2000s, the blooms appeared also in the other large lakes south of the Alps (Iseo, Como and Maggiore). Despite the sudden appearance of the blooms, the correct identification of the establishment time of the populations of Dolichospermum in the southern subalpine lake district remained unclear. In this work, the establishment of the populations of D. lemmermannii in Lake Garda has been evaluated by the direct counting of sub-fossil akinetes extracted from sediment cores, and by estimating the abundance of filaments germinated from sub-fossil viable akinetes. The two techniques provided comparable results, allowing locating the beginning of the establishment of Dolichospermum around the middle of the 1960s. Four strains of Dolichospermum germinated from akinetes isolated from the core sediments between around the 1989 and 2012 did not show any mutation or recombination signal in the rpoB gene sequences, suggesting a strong founder effect. The establishment of Dolichospermum coincided with the beginning of the rapid increase of total phosphorus as inferred from the distribution of sub-fossil diatoms in the sediment core. These results supported the hypothesis of a strong link between the shift of Lake Garda from ultraoligotrophy/oligotrophy to oligo-mesotrophy and the development of Dolichospermum. This colonisation pattern was possibly reinforced by the increase in the water temperatures in the subalpine lake district during the last 3 decades. In warmer lakes, gas-vacuolated Nostocales are favoured by high replication rates and, in particular, by their ability to control vertical movements in stratified water columns. Abstract

This work allowed assessing a widespread occurrence of Tychonema bourrellyi in the largest lakes south of the Alps (Garda, Iseo, Como and Maggiore). The taxonomy of the species was confirmed adopting a polyphasic approach, which included microscopic examinations, molecular (16S rRNA and rbcLX sequences) and (Lake Garda) ecological characterisations. Over 70% of the 36 isolates of Tychonema sampled from the four lakes tested positive for the presence of genes implicated in the biosynthesis of anatoxins (anaF and/or anaC) and for the production of anatoxin-a (ATX) and homoanatoxin-a (HTX). A detailed analysis carried out in Lake Garda showed strong ongoing changes in the cyanobacterial community, with populations of Tychonema developing with higher biovolumes compared to the microcystins (MCs) producer Planktothrix rubescens. Moreover, the time × depth distribution of Tychonema was paralleled by a comparable distribution of ATX and HTX. The increasing importance of Tychonema in Lake Garda was also suggested by the opposite trends of ATX and MCs observed since 2009. These results suggest that radical changes are occurring in the largest lakes south of the Alps.
