Abstract

Chorus is one of the strongest electromagnetic emissions naturally occurring in space, and can cause hazardous radiations to humans and satellites1-3. Although chorus has attracted extreme interest and been intensively studied for decades4-7, its generation and evolution remain highly debated, due to the complexity of the underlying physics and the limited capacity of previous spacecraft missions7. Chorus has also been believed to be governed by planetary magnetic dipolar fields5,7. Contrary to such conventional expectation, here we report unexpected observations of chorus in the terrestrial neutral sheet where magnetic dipolar effect is absent. Using unprecedentedly high-cadence data from the Magnetospheric Multiscale Mission, we present the first, ultrafast measurements of the wave dispersion relation and electron three-dimensional distributions within the waves, showing smoking-gun evidences for chorus-electron interactions and development of electron holes in the wave phase space. We estimate field-particle energy transfer inside the waves and find that the waves were extracting energy from local thermal electrons, in line with the wave positive growth rate derived from instability analysis. Our observations, opening new pathways for resolving long-standing controversies regarding the chorus emissions, are crucial for understanding nonlinear energy transport ubiquitously observed in space and astrophysical environments., Comment: under review; comments and suggestions are welcomed

Abstract

The global linear behaviors of 2/1 DTM in the collisional regime are investigated based on a concisely resistive drift-MHD model. Besides DTM, extra normal modes including EDW and SAW are coupled together and destabilized in different parameter regimes by considering resistivity in this system. The EVP approach is applied for solving the eigenstate spectra with the distribution of all unstable solutions. It is found that in the small EDD frequency (omega_*e) regime, DTM growth rate agrees well with local theory that is reduced with increasing omega_*e. However, when omega_*e exceeds a critical threshold omega_*crit, the strongly linear coupling between DTM and other discretized EDW instabilities happens so that the free energies from current and pressure channels can be released together and thus enhance the DTM, of which growth rate increases with increasing omega_*e and deviates from local theory results qualitatively. Correspondingly, a cross-scale mode structure forms with mixed polarization, namely, phi perturbation is dominated by electrostatic polarized short-wavelength oscillation as EDW instability character, and A_para perturbation remains typical tearing mode solution of Alfvenic polarized macroscopic structure. Within omega_*e > omega_*crit, the additional IDD causes phi oscillating structure to shift towards small density gradient domain, which cancels the extra drive from ion channel and thus DTM growth rate is insensitive to IDD frequency. Compared to EDD effects, the IDD effect alone with zero-omega_*e only leads to the stabilization of RTM that shows agreements between global simulation and local theory, which is no longer the condition for DTM regime. These results are useful for clarifying the DTM global properties with underlying physics mechanisms, which occurs in the regime of omega_*e >> gamma_c that is relevant to nowadays tokamak discharges with hot plasmas., Comment: 23 pages, 15 figues

Abstract

The Cryogenic Underground Observatory for Rare Events (CUORE) is a detector array comprised by 988 5$\;$cm$\times$5$\;$cm$\times$5$\;$cm TeO$_2$ crystals held below 20 mK, primarily searching for neutrinoless double-beta decay in $^{130}$Te. Unprecedented in size amongst cryogenic calorimetric experiments, CUORE provides a promising setting for the study of exotic through-going particles. Using the first tonne-year of CUORE's exposure, we perform a search for hypothesized fractionally charged particles (FCPs), which are well-motivated by various Standard Model extensions and would have suppressed interactions with matter. No excess of FCP candidate tracks is observed over background, setting leading limits on the underground FCP flux with charges between $e/24-e/5$ at 90\% confidence level. Using the low background environment and segmented geometry of CUORE, we establish the sensitivity of tonne-scale sub-Kelvin detectors to diverse signatures of new physics., Comment: 7 pages, 5 figures

Abstract

This study evaluates more than 4,000 tiles made of Hamamatsu visual-sensitive silicon photomultipier (SiPM), each with dimensions of 5 $\times$ 5 cm$^2$, intended for the central detector of the Taishan Anti-neutrino Observatory (TAO), a satellite experiment of the Jiangmen Underground Neutrino Observatory (JUNO) aimed at measuring the reactor anti-neutrino energy spectrum with unprecedented energy resolution. All SiPM tiles underwent a room temperature burn-in test in the dark for two weeks, while cryogenic testing analyzed the thermal dependence of parameters for some sampled SiPMs. Results from these comprehensive tests provide crucial insights into the long-term performance and stability of the 10 square meters of SiPMs operating at -50{\deg}C to detect scintillation photons in the TAO detector. Despite some anomalies awaiting further inspection, all SiPMs successfully passed the burn-in test over two weeks at room temperature, which is equivalent to 6.7 years at -50{\deg}C. Results are also used to guide optimal SiPM selection, configuration, and operation, ensuring reliability and sustainability in reactor neutrino measurements. Abstract

This Letter reports the first measurement of the oscillation amplitude and frequency of reactor antineutrinos at Daya Bay via neutron capture on hydrogen using 1958 days of data. With over 3.6 million signal candidates, an optimized candidate selection, improved treatment of backgrounds and efficiencies, refined energy calibration, and an energy response model for the capture-on-hydrogen sensitive region, the relative $\overline{\nu}_{e}$ rates and energy spectra variation among the near and far detectors gives $\mathrm{sin}^22\theta_{13} = 0.0759_{-0.0049}^{+0.0050}$ and $\Delta m^2_{32} = (2.72^{+0.14}_{-0.15})\times10^{-3}$ eV$^2$ assuming the normal neutrino mass ordering, and $\Delta m^2_{32} = (-2.83^{+0.15}_{-0.14})\times10^{-3}$ eV$^2$ for the inverted neutrino mass ordering. This estimate of $\sin^2 2\theta_{13}$ is consistent with and essentially independent from the one obtained using the capture-on-gadolinium sample at Daya Bay. The combination of these two results yields $\mathrm{sin}^22\theta_{13}= 0.0833\pm0.0022$, which represents an 8% relative improvement in precision regarding the Daya Bay full 3158-day capture-on-gadolinium result.

Abstract

We present the model we developed to reconstruct the CUORE radioactive background based on the analysis of an experimental exposure of 1038.4 kg yr. The data reconstruction relies on a simultaneous Bayesian fit applied to energy spectra over a broad energy range. The high granularity of the CUORE detector, together with the large exposure and extended stable operations, allow for an in-depth exploration of both spatial and time dependence of backgrounds. We achieve high sensitivity to both bulk and surface activities of the materials of the setup, detecting levels as low as 10 nBq kg$^{-1}$ and 0.1 nBq cm$^{-2}$, respectively. We compare the contamination levels we extract from the background model with prior radio-assay data, which informs future background risk mitigation strategies. The results of this background model play a crucial role in constructing the background budget for the CUPID experiment as it will exploit the same CUORE infrastructure.

Abstract

The matter-antimatter asymmetry underlines the incompleteness of the current understanding of particle physics. Neutrinoless double-beta ($0\nu \beta\beta$) decay may help explain this asymmetry, while unveiling the Majorana nature of the neutrino. The CUORE experiment searches for $0\nu \beta\beta$ decay of $^{130}$Te using a tonne-scale cryogenic calorimeter operated at milli-kelvin temperatures. We report no evidence for $0\nu \beta\beta$ decay and place a lower limit on the half-life of T$_{1/2}$ $>$ 3.8 $\times$ 10$^{25}$ years (90% C.I.) with over 2 tonne$\cdot$year TeO$_2$ exposure. The tools and techniques developed for this result and the 5 year stable operation of nearly 1000 detectors demonstrate the infrastructure for a next-generation experiment capable of searching for $0\nu \beta\beta$ decay across multiple isotopes.

Abstract

This Letter presents results of a search for the mixing of a sub-eV sterile neutrino with three active neutrinos based on the full data sample of the Daya Bay Reactor Neutrino Experiment, collected during 3158 days of detector operation, which contains $5.55 \times 10^{6}$ reactor \anue candidates identified as inverse beta-decay interactions followed by neutron-capture on gadolinium. The analysis benefits from a doubling of the statistics of our previous result and from improvements of several important systematic uncertainties. No significant oscillation due to mixing of a sub-eV sterile neutrino with active neutrinos was found. Exclusion limits are set by both Feldman-Cousins and CLs methods. Light sterile neutrino mixing with $\sin^2 2\theta_{14} \gtrsim 0.01$ can be excluded at 95\% confidence level in the region of $0.01$ eV$^2 \lesssim |\Delta m^{2}_{41}| \lesssim 0.1 $ eV$^2$. Abstract

Daya Bay presents the first measurement of cosmogenic $^8$He isotope production in liquid scintillator, using an innovative method for identifying cascade decays of $^8$He and its child isotope, $^8$Li. We also measure the production yield of $^9$Li isotopes using well-established methodology. The results, in units of 10$^{-8}\mu^{-1}$g$^{-1}$cm$^{2}$, are 0.307$\pm$0.042, 0.341$\pm$0.040, and 0.546$\pm$0.076 for $^8$He, and 6.73$\pm$0.73, 6.75$\pm$0.70, and 13.74$\pm$0.82 for $^9$Li at average muon energies of 63.9~GeV, 64.7~GeV, and 143.0~GeV, respectively. The measured production rate of $^8$He isotopes is more than an order of magnitude lower than any other measurement of cosmogenic isotope production. It replaces the results of previous attempts to determine the ratio of $^8$He to $^9$Li production that yielded a wide range of limits from 0 to 30\%. The results provide future liquid-scintillator-based experiments with improved ability to predict cosmogenic backgrounds.

Abstract

The full data set of the Daya Bay reactor neutrino experiment is used to probe the effect of the charged current non-standard interactions (CC-NSI) on neutrino oscillation experiments. Two different approaches are applied and constraints on the corresponding CC-NSI parameters are obtained with the neutrino flux taken from the Huber-Mueller model with a $5\%$ uncertainty. For the quantum mechanics-based approach (QM-NSI), the constraints on the CC-NSI parameters $\epsilon_{e\alpha}$ and $\epsilon_{e\alpha}^{s}$ are extracted with and without the assumption that the effects of the new physics are the same in the production and detection processes, respectively. The approach based on the weak effective field theory (WEFT-NSI) deals with four types of CC-NSI represented by the parameters $[\varepsilon_{X}]_{e\alpha}$. For both approaches, the results for the CC-NSI parameters are shown for cases with various fixed values of the CC-NSI and the Dirac CP-violating phases, and when they are allowed to vary freely. We find that constraints on the QM-NSI parameters $\epsilon_{e\alpha}$ and $\epsilon_{e\alpha}^{s}$ from the Daya Bay experiment alone can reach the order $\mathcal{O}(0.01)$ for the former and $\mathcal{O}(0.1)$ for the latter, while for WEFT-NSI parameters $[\varepsilon_{X}]_{e\alpha}$, we obtain $\mathcal{O}(0.1)$ for both cases., Comment: 25 pages, 16 figures, 6 tables; 36 pages, format changed, references added

13. Analysis to closed surface-wave photonic crystal waveguides based on coupled-resonator optical waveguide theory

Abstract

Traditionally, one can construct a waveguide by introduce defects into surface-wave photonic crystals (SPCs). Here we propose a new structure named closed SPC that can introduce waveguide modes out of photonic bandgap of surface-wave photonic crystal. In this paper, we have comprehensively analyzed dispersion relation, group velocity, normalized transmission and electric field distribution of closed SPC waveguides, and propose several methods to improve performance of the waveguide based on coupled-resonator optical waveguide theory. These methods can improve transmission efficiency from 10% to 60% and eliminate intraband oscillation by adjusting eigenfrequency and coupling factor . These methods are applicable to both single mode and multi mode situations. This letter also paves a new way for improving the performance of other coupled-resonator waveguides.

This study aimed to compare the ability of nucleotide MALDI-TOF MS to detect rifampicin (RIF) resistance in drug-resistant TB (DR-TB) patients with Xpert MTB/RIF and to analyze the disparate results individually. Additionally, potential factors associated with rifampicin resistance among DR-TB patients in Qingdao were investigated.Patients and Methods: A retrospective study was conducted at Qingdao Chest Hospital, and patients with DR-TB were enrolled. Corresponding frozen isolates were recovered and subjected to nucleotide MALDI-TOF MS, Xpert MTB/RIF, and phenotypic drug susceptibility testing (pDST). Sanger sequencing was performed for the discordant results of nucleotide MALDI-TOF MS and Xpert MTB/RIF. Univariate and multivariate logistic regression analyses were used to identify potential factors associated with rifampicin resistance among patients with DR-TB.Results: A total of 125 patients with DR-TB (18.8%, 125/668) were enrolled in this study from May 1 to July 31, 2023. Rifampicin-resistant (DR-TB/RR, 29) and rifampicin-sensitive (DR-TB/RS, 96) groups were divided according to the pDST results. Nucleotide MALDI-TOF MS performed better than Xpert MTB/RIF in terms of sensitivity, specificity, accuracy, and agreement with pDST. Only six cases had inconsistent results, and the sequencing results of five cases were identical to nucleotide MALDI-TOF MS. Furthermore, chest pain (aOR=12.84, 95% CI, 2.29– 91.97, p=0.005), isoniazid sensitivity (aOR=0.14, 0.02– 0.59, p=0.013), and ethambutol sensitivity (aOR=0.02, 0.00– 0.10, p=0.000) were potential factors associated with rifampicin resistance among DR-TB patients in Qingdao.Conclusion: The overall concordance between nucleotide MALDI-TOF MS and Xpert MTB/RIF was 95.2%, with the former performing better in determining rifampicin susceptibility among DR-TB cases in Qingdao. The aim of this study was to develop a novel assessment system based on laboratory indicators and frailty quantification to better infer short-term prognosis and risk indication in patients with AMI.Methods: A total of 365 patients with MI from January 2022 to June 2023 in Northern Jiangsu Province Hospital were included. The primary endpoint was all-cause mortality and major adverse cardiac events (MACE) during follow-up. A novel scoring model ranging from 0 to 12 was constructed, and the predictive ability of this scoring system was evaluated using the area under the receiver operating characteristic curve (AUC).Results: During follow-up, 68 patients experienced MACE. Five scoring indicators were selected through multivariate logistic regression analysis, resulting in a composite score with an AUC of 0.925, demonstrating good prognostic accuracy.Conclusion: The novel prognostic assessment system, which integrates age, Stress Hyperglycemia Ratio (SHR), Neutrophil to Lymphocyte Ratio (NLR), lactate, and frailty score, exhibits good predictive value for short-term MACE in patients with acute myocardial infarction and may enable more accurate risk classification for future use in MI patient risk management.Keywords: myocardial infarction, frailty index, major adverse cardiac events, prognosis, cohort study

Graves’ disease (GD), as an autoimmune thyroid disease (AITD), is attributed to the production of thyroid-stimulating hormone receptor (TSHR) autoantibodies that bind to the thyroid follicular endothelial cells. It is well known that genetic factors, environmental factors, and immune disorders count for much in the development of GD. So far, the pathogenesis of GD is not elucidated. Emerging research reveals that the change in gut microbiota composition and its metabolites are related to GD. The gut microbial diversity is reduced in GDs compared with healthy controls (HCs). Firmicutes and Bacteroidetes account for a large proportion at the genus level. It is found that phyla Bacteroidetes increased while phyla Firmicutes decreased in Graves’ Disease patients (GD patients). Moreover, gut microbiota modulates the immune system to produce cytokines through bacterial metabolites. This article aims to find out the relation between gut microbiota dysbiosis and the development of GD. Firstly, we use network pharmacology methods to collect core targets, and then validate their effects on diseases through experiments.Results: Five core targets were screened, Threonine kinase 1 (AKT1), Cellular tumor antigen p53 (TP53), Tumor necrosis factor (TNF), Albumin (ALB) and Vascular endothelial growthfactor A (VEGFA). KEGG analysis showed that YJKL treatment for PCOS mainly include AGE-RAGE signaling pathway in diabetic complications, TNF signaling pathway and HIF-1 signaling pathway. The molecular docking results showed that compounds have higher affinity with targets. However, the associations between genetic variants in the p53 signaling pathway and prognosis in hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC) remain unknown. The current study aims to analyze associations between the single nucleotide polymorphisms (SNPs) in p53 pathway-related genes and survival of patients with HBV-HCC.Methods: We evaluated the associations between 4698 SNPs in 70 genes of the p53 pathway and overall survival (OS) of 866 patients in additive genetic models by using Cox proportional hazards regression analysis. Stepwise multivariable Cox regression analysis was conducted to determine the independent effects of identified SNPs in single-locus analyses. The expression of quantitative trait loci (eQTL) was also analyzed using data from GTEx and 1000 Genomes Project, and functional prediction of SNPs was performed by using RegulomeDB v2.2, 3DSNP v2.0, HaploReg v4.2 and VannoPortal.Results: We found that two novel SNPs of CD82 rs7925603 A > G and PMAIP1 rs4396625 A > T, were significantly and independently associated with OS [adjusted hazards ratios (HRs) and 95% confidence intervals (CI) were 1.27 (1.10– 1.48) and 0.77 (0.66– 0.91), respectively; P = 0.001 and = 0.002, respectively] and that the combined risk genotypes of these SNPs showed a significant association with OS in patients with HBV-HCC (Ptrend < 0.001). Further eQTL analysis in the GTEx dataset showed that the rs7925603 G allele was associated with lower CD82 mRNA expression levels, while the rs4396625 T allele was associated with higher PMAIP1 mRNA expression levels in whole blood cells.Conclusion: We identified two observed survival-associated SNPs in CD82 and PMAIP1 in the p53 pathway, which influenced HBV-HCC survival possibly through a mechanism of altering mRNA expression. However, its role in colorectal cancer remains unknown.Methods: Bioinformatic methods were used to analyze gene expression, correlation and patient survival. Western blotting, colony formation assays and animal experiments were used to evaluate the function of VPS72 in colon cancer in vivo and in vitro.Results: VPS72 was highly expressed in colon cancer tissues and correlated with poor overall survival (P< 0.05) and relapse free survival (P< 0.01). Furthermore, patients with III/IV clinical stage (P< 0.001), N1 nodal metastasis (P< 0.001) or N2 nodal metastasis (P< 0.05) status had poor overall survival. Further analysis showed that VPS72 is correlated with proliferation and EMT biomarkers. Western blotting, colony formation assays and animal experiments showed that VPS72 overexpression promoted colon cancer proliferation and EMT progress.Conclusion: Our study found that VPS72 was correlated with poor overall survival in colon cancer patients, and high expressed level of VPS72 promoted colon cancer progression, indicating its role as a potential prognosis biomarker.Keywords: VPS72, colon cancer, biomarker, proliferation, epithelial–mesenchymal transformation

Abstract

The Super $\tau$-Charm facility (STCF) is an electron-positron collider proposed by the Chinese particle physics community. It is designed to operate in a center-of-mass energy range from 2 to 7 GeV with a peak luminosity of $0.5\times 10^{35}{\rm cm}^{-2}{\rm s}^{-1}$ or higher. The STCF will produce a data sample about a factor of 100 larger than that by the present $\tau$-Charm factory -- the BEPCII, providing a unique platform for exploring the asymmetry of matter-antimatter (charge-parity violation), in-depth studies of the internal structure of hadrons and the nature of non-perturbative strong interactions, as well as searching for exotic hadrons and physics beyond the Standard Model. The STCF project in China is under development with an extensive R\&D program. This document presents the physics opportunities at the STCF, describes conceptual designs of the STCF detector system, and discusses future plans for detector R\&D and physics case studies.

Necroptosis, a form of programmed necrotic cell death, occurs in many solid tumors, including breast cancer, and influences anti-tumor immunity. The role of necroptosis in managing breast cancer recurrence remains unclear.Methods: Gene expression profiles and clinical data of breast cancer patients were obtained from the GEO (GSE20685, GSE21653, GSE25055) and TCGA databases. Data analysis and visualization were performed using R. Unsupervised Consensus Clustering and LASSO-COX regression stratified breast cancer patients. GO, KEGG, GSVA, ESTIMATE, and ROC analyses were used to investigate necroptotic signatures. In vitro and in vivo experiments validated necroptosis’s role in breast cancer immunity.Results: The potential function of necroptotic signature in immunity was first indicated with GO analysis in BRCA cohort. Next, two prognostic models based on the necroptotic profiles both suggested a link between low-risk group with a particular necroptotic immune signature. And a variety of immune cells and immune pathways were shown to be positively associated with a patient’s risk score. As an altered immune checkpoint pattern was observed after regulating necroptotic genes, where TIM-3 and LAGLS9 elevated significantly in low-risk group, further validation in vitro and in vivo demonstrated that manipulating a subset of necroptotic gene set could sensitize tumor response to the co-blockade immunotherapy of anti-TIM-3 and anti-PD-1.Conclusion: We demonstrated two strategies to stratify breast cancer patients based on their necroptotic profiles and showed that necroptotic signature could assign patients with different tumor immune microenvironment patterns and different recurrence-related prognosis. A subset of necroptotic gene set, composed of TLR3, RIPK3, NLRP3, CASP1, ALDH2 and EZH2, was identified as a biomarker set for predicting immunotherapy-response and recurrence-related prognosis. The purpose of this study is to evaluate the biodistribution and toxicity of MPs/NPs with different particle sizes comprehensively and thoroughly.Methods: The purpose of this study was to investigate the biodistribution and in vivo toxicity of polystyrene (PS) MPs/NPs with different sizes (50 nm, 100 nm, and 500 nm). The BALB/c mice were given 100 μL of PS50, PS100 and PS500 at the dosage of 1 mg/kg BW or 10 mg/kg BW, respectively, by gavage once a day. After 28 consecutive days of treatment, the biodistribution of differently sized PS MPs/NPs was determined through cryosection fluorescence microscopy and fluorescent microplate reader analysis, and the subsequent effects of differently sized PS MPs/NPs on histopathology, hematology and blood biochemistry were also evaluated.Results: The results showed that the three different sizes of PS MPs/NPs were distributed in the organs of mice, mainly in the liver, spleen, and intestine. At the same time, the smaller the particle size, the more they accumulate in the body and more easily penetrate the tissue. During the whole observation period, no abnormal behavior and weight change were observed. The results of H&E staining showed that no severe histopathological abnormalities were observed in the main organs in the low-dose exposure group, while. Exposure of three sizes of PS MPs/NPs could cause some changes in hematological parameters or biochemical parameters related to heart, liver, and kidney function; meanwhile, there were size- and dose-dependencies.Conclusion: The biological distribution and toxicity of plastic particles in mice were more obvious with the decrease of particle size and the increase of concentration of plastic particles. Compared with MPs, NPs were easier to enter the tissues and produce changes in liver, kidney, and heart functions. The clinical characteristics, laboratory results, causative pathogen and other indicators were collected and compared, and risk factors were identified by applying Cox proportional hazard regression model to the data.Results: Community acquired-bloodstream infection is most commonly caused by Escherichia coli, Klebsiella species and Staphylococcus hominis. Among the total of 940 selected patients, 52 (5.5%) died during hospitalization. The demographic parameters like age and gender, clinical protocols like maintenance hemodialysis, glucocorticoid use during hospitalization, catheter placement, procaicitonin, total protein, albumin, creatinine, uric acid contents and Sequential Organ Failure Assessment scores were significantly different between the survivor and non-survivor groups. The survival analysis results revealed that age (HR=1.02, 95% CI: 1.00– 1.05, P=0.002), glucocorticoid use during hospitalization (HR=3.69, 95% CI: 1.62– 8.37, P=0.021) and Sequential Organ Failure Assessment score (HR=1.10, 95% CI: 1.03– 1.18, P=0.004) might be the risk factors affecting 30-day mortality in patients with community acquired-bloodstream infection.Conclusion: The identified risk factors may help guide clinical treatment protocol for patients with community acquired-bloodstream infection, providing more effective treatment strategy selection with improved clinical outcomes.Keywords: prognosis, pathogen, clinical characteristics, treatment

The present research focuses on the clinical significance of miR-379 in obesity-PCOS and attempts to elucidate its potential mechanisms.Methods: Healthy individuals (n = 46), obesity-PCOS (n = 92), and non-obesity PCOS (n = 31) subjects were enrolled. Quantitative real-time polymerase chain reaction (qRT-PCR) was conducted to examine the level of serum miR-379. The receiver operating characteristic (ROC) curve and logistic regressions were applied to reveal the diagnostic significance. Dual luciferase reporters were performed to validate the targeting relationships. And cell count kit (CCK-8) assay was used to detect cell proliferation.Results: Serum miR-379 was highly expressed in PCOS patients (P < 0.05), in especially obesity-PCOS patients. Higher miR-379 was associated with greater body mass index (BMI), higher bioavailable testosterone (bT), and greater insulin resistance (IR). Additionally, miR-379 was an independent risk factor for the development of obesity-PCOS. The sensitivity of miR-379 in identifying patients with obesity-PCOS from healthy or non-obesity-PCSO patients was 81.52% and 72.83%, and the specificity was 86.96% and 80.65%. Semaphorin 3 A (SEMA3A) was identified as a target of miR-379 and was reduced in the patients with obesity PCOS (P < 0.05). Inhibition of miR-375 reduced KGN proliferation, but this reduction was partially restored by silencing of SEMA3A (P < 0.05).Conclusion: Elevated miR-379 assists the diagnosis of obesity-PCOS and regulates the proliferation of KGN by targeting SEMA3A engaged in disease development.Keywords: obesity, PCOS, miR-379, diagnostic, SEMA3A

The synthetic control method (SCM) has emerged as a robust tool for evaluating the efficacy of health policies worldwide.Methods: This study focused on Panzhihua city in Sichuan Province, a pilot city for DRG payment reform implementation, serving as the treatment group. In contrast, 20 nonpilot cities within the province were utilized as potential control units. A counterfactual control group was constructed to evaluate the changes in average inpatient stay duration and health care organization costs following the DRG payment reform initiated in 2018.Results: Focusing on Panzhihua, Sichuan Province, the analysis reveals that following the reform in March 2018, the average length of hospital stay in Panzhihua decreased by 1.35 days during 2019– 2021. Additionally, the average cost per hospitalization dropped by 855.48 RMB, the average cost of medication per hospitalization decreased by 68.51 RMB, and the average cost of diagnostic and therapeutic procedures per hospitalization declined by 136.37 RMB. While global evidence backs DRGs for efficiency and cost reduction, challenges persist in addressing emerging issues like new conditions.Conclusion: Since its introduction in 2018, the DRG payment reform in Sichuan Province has effectively reduced both the duration of hospital stays and the operational costs of health care facilities. Meanwhile, the study included 18 cases with UDEC in our center and explored their molecular characteristics and prognosis.Results: Between 2000 and 2019, a total of 443 patients were included from the SEER database. The median CSS duration was 14 months, with corresponding 3- and 5-year CSS rates of 45.9% and 44.0%, respectively. Factors such as pTNM stage, surgical resection of primary lesion, and chemoradiation independently influenced CSS. Postoperative chemotherapy alone improved CSS in patients with initial tumor spread beyond the uterus (pT3 and pT4), or lymph node (LN) invasion, or distant metastases. Additionally, postoperative radiotherapy enhanced CSS in patients who had undergone postoperative chemotherapy, those with primary tumors progressing to stage pT3, and those with LN involvement but without distant metastases. Of the 18 patients diagnosed at our center, with a median follow-up of 15.5 months, one experienced relapse and two succumbed to UDEC, who exhibited aberrant p53 expression in immunohistochemical staining.Conclusion: Postoperative chemotherapy and radiotherapy are beneficial for UDEC patients with tumors extending beyond the uterus or involving lymph nodes.Keywords: undifferentiated and dedifferentiated endometrial carcinoma, cancer-specific survival, prognostic factors, postoperative adjuvant therapy

However, the specific mechanisms and roles of the intestine in islets regulation remain unclear. Recent studies investigated that exosomes derived from gut microbiota can transport signals to remotely regulate islets β-cell function, suggesting the possibility of novel signaling pathways mediated by gut exosomes in the regulation of the “gut-islet axis.”.Methods: The exosomes were isolated from the intestinal enteroendocrine cell-line STC-1cells culture supernatants treated with palmitate acid (PA) or BSA. Metabolic stress models were established by separately subjecting MIN6 cells to PA stimulation and feeding mice with a high-fat diet. Intervention with exosomes in vitro and in vivo to assess the biological effects of exosomes on islets β cells under metabolic stress. The Mas receptor antagonist A779 and ACE2ko mice were used to evaluate the role of exosomal ACE2.Results: We found ACE2, a molecule that plays a crucial role in the regulation of islets function, is abundantly expressed in exosomes derived from STC-1 under physiological normal condition (NCEO). These exosomes cannot only be taken up by β-cells in vitro but also selectively transported to the islets in vivo. Following intervention with NCEXO, both Min6 cells in a lipotoxic environment and mice on a high-fat diet exhibited significant improvements in islets β-cell function and β-cell mass. Further investigations demonstrated that these protective effects are attributed to exosomal ACE2, as ACE2 inhibits NLRP3 inflammasome activation and reduces β-cell pyroptosis.Conclusion: ACE2-enriched exosomes from the gut can selectively target islets, subsequently inhibiting NLRP3 inflammasome activation and β cell pyroptosis, thereby restoring islets β cell function under metabolic stress. The neutrophil-lymphocyte ratio (NLR) and platelet-lymphocyte ratio (PLR) were assessed, and appropriate cut-off values were established. The NLR-PLR score ranged from 0 to 2 as follows: score of 2, high NLR (≥ 3.37) and high PLR (≥ 105.96); score of 1, either high NLR or high PLR; score of 0, neither high NLR nor high PLR.Results: The median overall survival (OS) of patients with NLR-PLR score of 0, 1 and 2 was 27, 26.5, and 6 months, respectively. The median OS of patients with NLR-PLR score of 2 was shorter than those with 0 (P < 0.001) and 1 (P < 0.001). The median disease-free survival (DFS) time of patients with NLR-PLR score of 0, 1 and 2 was 24.5, 24, and 6 months, The median DFS of patients with NLR-PLR score of 2 was shorter than those with 0 (P = 0.001) and 1 (P = 0.015). Abstract

Reactor neutrino experiments play a crucial role in advancing our knowledge of neutrinos. In this Letter, the evolution of the flux and spectrum as a function of the reactor isotopic content is reported in terms of the inverse-beta-decay yield at Daya Bay with 1958 days of data and improved systematic uncertainties. These measurements are compared with two signature model predictions: the Huber-Mueller model based on the conversion method and the SM2018 model based on the summation method. The measured average flux and spectrum, as well as the flux evolution with the ^{239}Pu isotopic fraction, are inconsistent with the predictions of the Huber-Mueller model. In contrast, the SM2018 model is shown to agree with the average flux and its evolution but fails to describe the energy spectrum. Altering the predicted inverse-beta-decay spectrum from ^{239}Pu fission does not improve the agreement with the measurement for either model. The models can be brought into better agreement with the measurements if either the predicted spectrum due to ^{235}U fission is changed or the predicted ^{235}U, ^{238}U, ^{239}Pu, and ^{241}Pu spectra are changed in equal measure.

Abstract

We present a new determination of the smallest neutrino mixing angle ${\theta}_{13}$ and the mass-squared difference ${\Delta}{\rm m}^{2}_{32}$ using a final sample of $5.55 \times 10^{6}$ inverse beta-decay (IBD) candidates with the final-state neutron captured on gadolinium. This sample was selected from the complete data set obtained by the Daya Bay reactor neutrino experiment in 3158 days of operation. Compared to the previous Daya Bay results, selection of IBD candidates has been optimized, energy calibration refined, and treatment of backgrounds further improved. Abstract

Abstract

HERMES (High Energy Rapid Modular Ensemble of Satellites) is a space-borne mission based on a constellation of nano-satellites flying in a low-Earth orbit (LEO). The six 3U CubeSat buses host new miniaturized instruments hosting a hybrid Silicon Drift Detector/GAGG:Ce scintillator photodetector system sensitive to X-rays and gamma-rays. HERMES will probe the temporal emission of bright high-energy transients such as Gamma-Ray Bursts (GRBs), ensuring a fast transient localization (with arcmin-level accuracy) in a field of view of several steradians exploiting the triangulation technique. With a foreseen launch date in late 2023, HERMES transient monitoring represents a keystone capability to complement the next generation of gravitational wave experiments. Moreover, the HERMES constellation will operate in conjunction with the Space Industry Responsive Intelligent Thermal (SpIRIT) 6U CubeSat, to be launched in early 2023. SpIRIT is an Australian-Italian mission for high-energy astrophysics that will carry in a Sun-synchronous orbit (SSO) an actively cooled HERMES detector system payload. Few tools and indicators are available to assess TAO, restricting personalized diagnosis and treatment.Aim: To identify an aptamer targeting thyroid-stimulating hormone receptor (TSHR) and utilize this aptamer to evaluate clinical activity in patients with TAO.Methods: An aptamer targeting TSHR was developed by exponential enrichment and systematic evaluation of TSHR ligands. After truncation and optimization, the affinity, equilibrium dissociation constant, and serum stability of this aptamer were evaluated. The affinity of the TSHR-targeting aptamer to isolated fibrocytes was assessed, as was aptamer internalization by fibrocytes. The mechanism of binding was determined by molecular docking. The correlation between disease manifestations and the percentage of TSHR-positive cells was assessed by correlation analysis.Results: The aptamer TSHR-21-42 was developed to bind to TSHR, with the equilibrium dissociation constant being 71.46 Kd. Isolated fibrocytes were shown to bind TSHR-21-42 through TSHR, with its affinity maintained at various temperatures and ion concentrations. TSHR-21-42 could compete with anti-TSHR antibody, both for binding site to TSHR and uptake by cells after binding. In addition, TSHR-21-42 could bind to leukocytes in peripheral blood, with this binding differing in patients with TAO and healthy control subjects. The percentage of TSHR-positive monocytes, as determined by binding of TSHR-21-42, correlated positively with clinical activity score in patients with TAO, indicating that TSHR-21-42 binding could assess the severity of TAO.Conclusion: This aptamer targeting TSHR may be used to objectively assess disease activity in patients with TAO, by evaluating the percentages of TSHR positive cells in peripheral blood.Keywords: thyroid-associated ophthalmopathy, thyroid-stimulating hormone receptor, aptamer, clinical activity score, precision medicine

It exhibits high resistance to many antibiotics, including ceftazidime/avibactam and cefteolozane/tazobactam. The presence of carbapenem-resistant genes and co-existence Klebsiella pneumoniae carbapenemase (KPC) and metallo-β-lactamases (MBLs) further inactivated all β-lactams. Understanding the resistance genes of CRPA can help in uncovering the resistance mechanism and guiding anti-infective treatment. Herein, we reported a case of perianal infection with hypervirulent ST463 Pseudomonas aeruginosa.Case Presentation: The case is a 32-year-old acute myeloid leukemia (AML) patient with fever and septic shock during hematopoietic stem cell transplantation (HSCT), and the pathogen was finally identified as a highly virulent sequence type 463 (ST463) P. aeruginosa harboring carbapenem-resistant genes blaAFM-1 and blaKPC-2, which was detected in the bloodstream and originated from a perianal infection. The strain was resistant to ceftazidime/avibactam but successfully treated with polymyxin B, surgical debridement, and granulocyte engraftment after HSCT. The AML was cured during the 19-month follow-up.Conclusion: This case emphasizes the importance of metagenomic next-generation sequencing (mNGS) and whole-genome sequencing (WGS) in identifying microbes with rare resistant genes, and managing CRPA, especially in immunocompromised patients. An eosinophil-related index, the neutrophil to eosinophil ratio (NER), can be used to predict the prognosis of patients with tumors. However, there is still a lack of efficient prognostic biomarkers for HCC. In this study, we aimed to investigate the predictive value of the NER and develop an optimal machine learning model for the recurrence of HCC patients. Patients and methods: A retrospective collection of 562 patients who underwent hepatectomy with a pathologic diagnosis of HCC was performed. The relationship between NER and progression-free survival (PFS) was investigated. We developed a new machine learning framework with 10 machine learning algorithms and their 101 combinations to select the best model for predicting recurrence after hepatectomy. The performance of the model was assessed by the area under the curve (AUC) of characteristics and calibration curves, and clinical utility was evaluated by decision curve analysis (DCA).Results: Kaplan‒Meier curves showed that the PFS in the low NER group was significantly better than that in the high NER group. Multivariate Cox regression analysis showed that NER was an independent risk factor for recurrence after surgery. The random survival forests (RSF) model was selected as the best model that had good predictive efficacy and outperformed the TNM, BCLC, and CNLC staging systems.Conclusion: The NER has good predictive value for postoperative recurrence in patients with hepatocellular carcinoma. The heterogeneity of CMS distribution in metastases and the concordance of CMS between PT and metastases still lack sufficient study. We used CMS to classify CRC metastases and combine it with histopathological analysis to explore differences between PT and distant metastases.Patients and Methods: We obtained gene expression profiles for 942 PT samples from TCGA database (n=376) and GEO database (n=566), as well as 442 metastasis samples from GEO database. Among these, 765 PT samples and 442 metastasis samples were confidently identified with CMS using the “CMS classifier” and enrolled for analysis. Clinicopathological manifestation and CMS classification of CRC metastases were assessed with data from GEO, TCGA, and cBioPortal. Overall, 105 PT-metastasis pairs were extracted from 10 GEO datasets to assess CMS concordance. Tumor microenvironment (TME) features between PT and metastases were analyzed by immune-stromal infiltration with ESTIMATE and xCell algorithms. Finally, TME features were validated with multiplex immunohistochemistry in 27 PT-metastasis pairs we retrospectively collected.Results: Up to 64% of CRC metastases exhibited concordant CMS groups with matched PT, and the TME of metastases was similar to that of PT. For most common distant metastases, liver metastases were predominantly CMS2 and lung and peritoneal metastases were mainly CMS4, highlighting “seed” of tumor cells of different CMS groups had a preference for metastasis to “soil” of specific organs. Compared with PT, cancer-associated fibroblasts (CAF) reduced in liver metastases, CD4+T cells and M2-like macrophages increased in lung metastases, and M2-like macrophages and CAF increased in peritoneal metastases.Conclusion: Our findings underscore the importance of CMS-guided specific organ monitoring and treatment post-primary tumor surgery for patients. Abstract

Employing two frequency combs with a slight difference in repetition frequencies, the dual-comb source shows unique advantages in high precision spectroscopy, imaging, ranging, communications, etc. In the terahertz (THz) frequency range, the electrically pumped quantum cascade laser (QCL) offers the possibility of realizing the compact dual-comb source due to its semiconductor-based chip-scale configuration. Although the active stabilization of a THz QCL dual-comb source was demonstrated by phase locking one of the dual-comb lines, the full stabilization of all dual-comb lines is still challenging. Here, we propose a self-reference method to obtain a fully stabilized dual-comb signal on a pure THz QCL platform. Without using any external locking components, we filter out one dual-comb line and beat it with the whole dual-comb signal, which eliminates the common carrier offset frequency noise and reduces the dual-comb repetition frequency noise. It is experimentally demonstrated that the self-reference technique can significantly improve the long-term stability of the dual-comb signal. A record of the ``maxhold" linewidth of 14.8 kHz (60 s time duration) is obtained by implementing the self-reference technique, while without the self-reference the dual-comb lines show a ``maxhold" linewidth of 2 MHz (15 s time duration). The method provides the simplest way to improve the long-term stability of THz QCL dual-comb sources, which can be further adopted for high precision measurements., Comment: 15 pages, 4 figures

Abstract

Ultra-broadband imaging devices with high performance are in great demand for a variety of technological applications, including imaging, remote sensing, and communications. An ultra-broadband up-converter is realized based on a p-GaAs homojunction interfacial workfunction internal photoemission (HIWIP) detector-light emitting diode (LED) device. The device demonstrates an ultra-broad response ranging from visible to terahertz (THz) with good reproducibility. The peak responsivity in the mid-infrared (MIR) region is 140 mA/W at 10.5 microns. The HIWIP-LED shows enormous potential for ultra-broadband up-conversion covering all infrared atmospheric windows, as well as the THz region, and the pixel-less imaging of the MIR spot from the CO2 laser is further demonstrated. In addition, the proposed up-converter also performs as a near-infrared and visible detector under zero bias by using a bi-functional LED. Thanks to its ultra-wide response, the HIWIP-LED up-converter has great promise for stable, high-performance ultra-broadband pixel-less imaging and multi-functional analysis systems., Comment: 23 pages, 5 figures

49. Stopping time detection of wood panel compression: A functional time series approach

Abstract

We consider determining the optimal stopping time for the glue curing of wood panels in an automatic process environment. Using the near-infrared spectroscopy technology to monitor the manufacturing process ensures substantial savings in energy and time. We collect a time series of curves from a near-infrared spectrum probe consisting of 72 spectra and aim to detect an optimal stopping time. We propose an estimation procedure to determine the optimal stopping time of wood panel compression and the estimation uncertainty associated with the estimated stopping time. Our method first divides the entire data set into a training sample and a testing sample, then iteratively computes integrated squared forecast errors based on the testing sample. We then apply a structural break detection method with one breakpoint to determine an estimated optimal stopping time from a univariate time series of the integrated squared forecast errors. Abstract

This Letter reports the first measurement of high-energy reactor antineutrinos at Daya Bay, with nearly 9000 inverse beta decay candidates in the prompt energy region of 8-12~MeV observed over 1958 days of data collection. A multivariate analysis is used to separate 2500 signal events from background statistically. The hypothesis of no reactor antineutrinos with neutrino energy above 10~MeV is rejected with a significance of 6.2 standard deviations. A 29\% antineutrino flux deficit in the prompt energy region of 8-11~MeV is observed compared to a recent model prediction. We provide the unfolded antineutrino spectrum above 7 MeV as a data-based reference for other experiments. This result provides the first direct observation of the production of antineutrinos from several high-$Q_{\beta}$ isotopes in commercial reactors., Comment: 7 pages, 4 figures, accepted by Physical Review Letters
