13. Transcriptome Analysis Reveals Cross-Talk between the Flagellar Transcriptional Hierarchy and Secretion System in Plesiomonas shigelloides.

Abstract

Plesiomonas shigelloides, a Gram-negative bacillus, is the only member of the Enterobacteriaceae family able to produce polar and lateral flagella and cause gastrointestinal and extraintestinal illnesses in humans. The flagellar transcriptional hierarchy of P. shigelloides is currently unknown. In this study, we identified FlaK, FlaM, FliA, and FliAL as the four regulators responsible for polar and lateral flagellar regulation in P. shigelloides. To determine the flagellar transcription hierarchy of P. shigelloides, the transcriptomes of the WT and ΔflaK, ΔflaM, ΔfliA, and ΔfliAL were carried out for comparison in this study. Quantitative Real-Time Polymerase Chain Reaction (qRT-PCR) and luminescence screening assays were used to validate the RNA-seq results, and the Electrophoretic Mobility Shift Assay (EMSA) results revealed that FlaK can directly bind to the promoters of fliK, fliE, flhA, and cheY, while the FlaM protein can bind directly to the promoters of flgO, flgT, and flgA. Meanwhile, we also observed type VI secretion system (T6SS) and type II secretion system 2 (T2SS-2) genes downregulated in the transcriptome profiles, and the killing assay revealed lower killing abilities for ΔflaK, ΔflaM, ΔfliA, and ΔfliAL compared to the WT, indicating that there was a cross-talk between the flagellar hierarchy system and bacterial secretion system. Invasion assays also showed that ΔflaK, ΔflaM, ΔfliA, and ΔfliAL were less effective in infecting Caco-2 cells than the WT. Additionally, we also found that the loss of flagellar regulators causes the differential expression of some of the physiological metabolic genes of P. shigelloides. Overall, this study aims to reveal the transcriptional hierarchy that controls flagellar gene expression in P. shigelloides, as well as the cross-talk between motility, virulence, and physiological and metabolic activity, laying the groundwork for future research into P. shigelloides' coordinated survival in the natural environment and the mechanisms that infect the host. 17. Exploring the Performance of Hall Sensor With Substrate Bias Modulation in SOI Process

Abstract

This article proposed a Hall cross plate for magnetic field detection with substrate biasing to improve the sensitivity. By adjusting the bias voltage of the reversed-biased p-n junction ( ${V} _{\text {PN}}\text {)}$ formed by the n-well and the p-well underneath, we can modulate the thickness of the depletion layer and, thus, the effective thickness of the cross plate formed by the n-well, eventually ameliorating the sensitivity. We verified the proposed substrate-biased Hall sensor with finite-element analysis and measurement from the cross plate prototyped in the 180-nm silicon-on-insulator (SOI) CMOS process. From the measurement, the resistance of the cross-plate changes from 2.13 to 2.39 k $\Omega $ when the ${V} _{\text {PN}}$ decreases from 0 to -0.6 V, indicating the variation in the sheet resistance, induced by the change in the thickness of the cross-plate. Moreover, the current-related sensitivity increased by 28.4% in the current mode when ${V} _{\text {PN}}$ changed from 0 to -0.6 V. The Hall plate achieves a current-related sensitivity of 0.3 V/mA $\cdot $ T with a power consumption of 6.5 mW.

31. Impact of Biogenic Amines on the Growth of Green Microalgae

Abstract

Background: The goal of this research project was to test various neuroactive amines in the capacity of growth stimulators/accelerators of the green microalgae Scenedesmus quadricauda and Chlorella vulgaris that have much biotechnological potential because they can be used for producing drugs, food ingredients, cosmetics, and biofuel. The issue of the ecological role of the biogenic amines in terms of interspecies communication in aqueous ecosystems was also addressed in this work. Methods: S. quadricauda strain GEHD and C. vulgaris strain ALP were cultivated in the light with constant aeration at 24oC in a minerals-containing medium. Experimental systems contained 1, 10 or 100 mM of dopamine hydrochloride, histamine hydrochloride, norepinephrine hydrochloride, or serotonin hydrochloride that were added at inoculation as freshly prepared aqueous solutions. Algal cells were counted using a light microscope , and their number in 1 mL of culture was calculated. The culture liquid and sonicated biomass of S. quadricauda and C. vulgaris were tested for the presence of endogenous amines using high-performance liquid chromatography (HPLC) with an amperometric detector. Results: The biogenic amines serotonin, norepinephrine, dopamine, and histamine significantly stimulated the growth of S. quadricauda, at concentrations of 1 and/or 10 mM but not 100 mM. Histamine was the most efficient stimulator, causing an average 65% increase in biomass accumulation at the end of the cultivation period. The effects of serotonin, dopamine and histamine on C. vulgaris were reported in our previous publication [1], but this work contains the results of our experiments with the previously untested norepinephrine that slightly stimulated the growth of C. vulgaris. HPLC analysis failed to reveal any endogenous amines in the culture liquid and biomass of both microalgae. Conclusions: Since biogenic amines stimulate the growth of the microalgae S. quadricauda and C. vulgaris but are not synthesized by them, we suggest that the algae normally respond to amines produced by other components of aqueous ecosystems, including zooplankton and fish that are known to release significant amounts of biogenic amines into the environment. The data obtained hold some promise with regard to developing a relatively economical technique of boosting algal biomass production.

Abstract

The present work aims to develop a new approach enabling biotechnologists to increase the yield of Chlorella vulgaris biomass by means of biogenic amines (serotonin, dopamine, and histamine) that are known to stimulate growth of various unicellular organisms. C. vulgaris strain ALP was cultivated in the light with constant aeration at 24oC in a minerals-containing medium. Experimental systems contained 1, 10, or 100 mM of dopamine, histamine, or serotonin. Algal cells were counted using a light microscope. Serotonin caused a slight increase in biomass yield at a concentration of 10 mM, but not at the other tested concentrations. 1 and 10 mM (but not 100 mM) dopamine increased the cell number in the C. vulgaris culture at early cultivation stages. Histamine is the most efficient growth stimulator at concentrations of 1 and 10 mM, but not at a concentration of 100 mM, which even proved inhibitory to the algal culture. The data obtained demonstrate that the neurochemicals exert a stimulatory influence on the growth of the Chlorella culture at relatively low (micromolar) concentrations. Since animals often produce biogenic amines in response to stress or injury, the data give grounds for the suggestion that planktonic algae can benefit, in terms of growth rate, from the substances released by stressed or wounded representatives of aquatic fauna. In biotechnological terms, the data obtained hold some promise with regard to developing a relatively economical technique of boosting Chlorella biomass production.

This is due to their importance as efficient and economical producers of food additives, cosmetics, pharmaceuticals, biofertilizers, biofuels, and wastewater bioremediation agents. Recently, the role of microalgae has increasingly been considered in terms of their probiotic function, i.e., of their ability to normalize the functioning of the microbiota of humans and agricultural animals and to produce biologically active substances, including hormones, neurotransmitters, and immunostimulators. A separate brief subsection of the review deals with the potential functions of microalgae with respect to the brain and psyche, i.e., as psychobiotics. Moreover, algal polysaccharides and some other compounds can be broken down to short fragments that will stimulate the development of useful intestinal microorganisms, i.e., function as efficient prebiotics. Finally, many components of microalgal cells and chemical agents produced by them can exert important health-promoting effects per se, which indicates that they are as potentially valuable metabiotics (the term preferred by late Prof. B.A. Shenderov), which are alternatively denoted as postbiotics in the literature.

Abstract

Based on the analysis of traditional state maintenance strategy and big data-based state maintenance strategy, this paper builds a transformer state duration simulation model based on Monte Carlo method and builds an annual cost model based on the running state of the transformer. Finally, a typical case is selected to simulate a time series of 8760 hours of transformer operation under two operational inspection strategies, and the annual cost is calculated separately. The example shows that the annual cost of the transformer is lower under the big data-based inspection strategy.

Abstract

Circulating tumor cells (CTCs) have important application prospects in the early diagnosis, treatment evaluation, and prognostic prediction of tumors. In this study, we enrolled a total of 65 patients with different stages and molecular subtypes of breast cancer and isolated and enriched for CTCs from peripheral blood using the ClearCell FX1 platform, which is based on a label-free spiral microfluidic method. The ClearCell platform can successfully isolate CTCs from peripheral blood with different detection rates in breast cancer patients. We also compared the difference between the ClearCell and CellSearch platforms for isolating CTCs. To further determine the genetic information of CTCs, we performed single-cell whole-exome sequencing (WES) in three CTCs isolated from one patient. The sequencing results indicated the presence of a few hundreds of single-nucleotide variants (SNVs) in each CTC, with only 16 SNVs being shared by all three CTCs. These shared SNVs may have a crucial impact on the development of breast cancer. Here, we report, for the first time, the complete process and results of performing single-cell WES on CTCs isolated by the ClearCell FX1 platform.
