Abstract

The soluble epoxide hydrolase (sEH) has been suggested as a pharmacological target for the treatment of several diseases, including pain-related disorders. Herein, we report further medicinal chemistry around new benzohomoadamantane-based sEH inhibitors (sEHI) in order to improve the drug metabolism and pharmacokinetics properties of a previous hit. After an extensive in vitro screening cascade, molecular modeling, and in vivo pharmacokinetics studies, two candidates were evaluated in vivo in a murine model of capsaicin-induced allodynia. The two compounds showed an anti-allodynic effect in a dose-dependent manner. Moreover, the most potent compound presented robust analgesic efficacy in the cyclophosphamide-induced murine model of cystitis, a well-established model of visceral pain. Overall, these results suggest painful bladder syndrome as a new possible indication for sEHI, opening a new range of applications for them in the visceral pain field.

Abstract

Objective: Idiopathic central precocious puberty (CPP) precipitates epiphyseal fusion of growth plates in long bones, leading to reduced adult stature. Gonadotropin-releasing hormone analogues (GnRHa) are the treatment of choice for idiopathic CPP, but their benefit on height gain is unclear. We aimed to elucidate the effects of GnRHa treatment on adult height in girls with idiopathic CPP. Design: This prospective observational descriptive study analyzed data of girls with idiopathic CPP diagnosed at 55 centers in Spain between January 1, 1998 and December 31, 2012 included in the Spanish Society for Pediatric Endocrinology's national registry. Methods: We included girls with idiopathic CPP (thelarche < 8 years, positive LHRH stimulation test, bone age > 1 year older than chronological age, and normal brain imaging) treated with triptorelin (3.75 mg monthly, adjusted according to LHRH test results and clinical findings). We assessed weight, height, BMI, and secondary sexual characteristics every 6 months and bone age every 12 months until adult height (AH) was attained. The primary outcome was the difference between AH and target height (TH). Results: A total of 465 girls (18.90% adopted) were included; we analyzed data recorded at treatment end in 358 girls and at AH in 216. Mean difference between AH and TH was -1.5 (95%CI: -2.56− -0.45) cm and between AH and PAH 2,57 (95%CI:-3.56− -1.58) cm. Conclusions: GnRHa treatment helps preserve genetic growth potential in girls with idiopathic CPP. [ABSTRACT FROM AUTHOR]

Abstract

Chronic neuropathic pain is a major morbidity of neural injury, yet its mechanisms are incompletely understood. Hypersensitivity to previously non-noxious stimuli (allodynia) is a common symptom. Here, we demonstrate that the onset of cold hypersensitivity precedes tactile allodynia in a model of partial nerve injury, and this temporal divergence was associated with major differences in global gene expression in innervating dorsal root ganglia. Transcripts whose expression change correlates with the onset of cold allodynia were nociceptor related, whereas those correlating with tactile hypersensitivity were immune cell centric. Ablation of TrpV1 lineage nociceptors resulted in mice that did not acquire cold allodynia but developed normal tactile hypersensitivity, whereas depletion of macrophages or T cells reduced neuropathic tactile allodynia but not cold hypersensitivity. We conclude that neuropathic pain incorporates reactive processes of sensory neurons and immune cells, each leading to distinct forms of hypersensitivity, potentially allowing drug development targeted to each pain type.

Abstract

[Simple Summary] The Spanish cattle breeds “Berrenda en Colorado” and “Berrenda en Negro” are considered endangered breeds. For years, breeders’ associations have been implementing breeding strategies in herds to address the presence of the Robertsonian translocation (rob (1;29)), which is a disease that causes a decline in reproductive performance. Single-nucleotide polymorphism (SNP) genotyping detection techniques are presented as an appropriate means to evaluate genetic diversity. However, previous research on genetic variability in both breeds has been carried out using DNA microsatellites. Therefore, we studied the genetic diversity, population structure, and potential genetic differences among individuals of both Berrenda breeds and groups based on the presence of the Robertsonian chromosomal translocation, rob (1;29). The genetic diversity in terms of expected heterozygosity was significantly lower in those subpopulations containing rob (1;29) within the breed, but in general, the four subpopulations considered showed minor genetic differences. The presence of this Robertsonian translocation did not result in sub-structuring associated with this chromosomal disorder within either of the breeds. The improvement of the reproductive performance of the Berrenda breeds requires the implementation of breeding strategies based on the contributions to the populations of the breeding stock carrying rob (1;29)., [Abstract] Most of the previous studies on the genetic variability in Spanish “Berrenda” breeds have been carried out using DNA microsatellites. The present work aimed to estimate the genetic diversity, population structure, and potential genetic differences among individuals of both Berrenda breeds and groups based on the presence of the Robertsonian chromosomal translocation, rob (1;29). A total of 373 samples from animals belonging to the two breeds, including 169 cases diagnosed as rob (1;29)-positive, were genotyped using an SNP50K chip. The genetic diversity at the breed level did not show significant differences, but it was significantly lower in those subpopulations containing the rob (1;29). Runs of homozygosity identified a region of homozygosity on chromosome 6, where the KIT (KIT proto-oncogene, receptor tyrosine kinase) gene, which determines the typical spotted coat pattern in both breeds, is located. The four subpopulations considered showed minor genetic differences. The regions of the genome that most determined the differences between the breeds were observed on chromosomes 4, 6, 18, and 22. The presence of this Robertsonian translocation did not result in sub-structuring within each of the breeds considered. To improve the reproductive performance of Berrenda breeds, it would be necessary to implement strategies considering the involvement of potential breeding stock carrying rob (1;29).

Abstract

In the prize‐collecting geometric enclosure problem (PCGEP), a set S$S$ of points in the plane is given, each with an associated benefit. The goal is to find a simple polygon P$\mathcal {P}$ with vertices in S$S$ that maximizes the sum of the benefits of the points of S$S$ enclosed by P$\mathcal {P}$ minus the perimeter of P$\mathcal {P}$ multiplied by a given nonnegative cost. The PCGEP is NP‐complete and has applications to land surveying for exploration or preservation of natural resources. In this paper, we develop the first heuristic, called PCGEP‐GR, for the PCGEP and revisit a previously proposed integer linear programming (ILP) model to solve it to optimality. We conducted a comprehensive experimental study of that heuristic and an exact algorithm based on the ILP model. We show that a new set of constraints, together with the previous set, is necessary to guarantee the correctness of the ILP model and introduce preprocessing strategies that allow us to prove optimality 40% faster on average. The proposed heuristic is able to reach the optimum in 32% of our benchmark instances and, for those with unknown optima, PCGEP‐GR was found better than or at least as good solutions as the ones obtained by the cplex ILP solver in 54% of the cases. Notwithstanding these positive results, the design of effective heuristics for the PCGEP proved to be very challenging, which also led us to obtain a result that provides the theoretical foundation for future advances in the study of this problem. Abstract

The rediscovery of Erigeron morelensis, not collected since 1898, is reported. Between 2021 and 2022, E. morelensis was recorded at two sites in the northwestern canyons of Cuernavaca, Morelos, Mexico, which are subjected to degradation pressure. The morphological description is updated and complemented, and a key for E. sect. Erigeridium is included. Data about the known distribution, ecology and habitat of the species are provided. Based on the IUCN criteria, it is recommended that the species be listed as Critically Endangered (CR).
