Synthetic cannabinoids are the most popular psychoactive compounds on the illegal market. In the gas chromatographic determination, some synthetic cannabinoids undergo chemical transformations because of their thermal interaction with the chromatographic system. This paper is devoted to the elucidation of the structure of a gas chromatographic artifact formed from synthetic cannabinoid N-(1-carbamoyl-2-methylpropyl)-1-(cyclohexylmethyl)-1H-indazol-3-carboxamide as a result of dehydration of its terminal carbamoyl moiety. The chemical structure of the artifact is determined by high-resolution mass spectrometry. The solid state NMR spectroscopy was used for (super 31)P and (super 2)H relaxation studies of helix VI and the cytoplasmic helix of human cannabinoid receptor. This technique can be used to sense the physical state of the lipid bilayer as well as the molecular motions associated with acyl chains in the presence of macromolecules.

Circular dichroism and nuclear magnetic resonance spectroscopy are utilized to perform the conformational analysis of hemopressin and its truncated, biologically active fragments and molecular docking calculations are used to study the binding modes of hemopressin peptide to the cannabinoid [CB.sub.1] receptor. The conformational data indicated that regular turn structures in the central portion of hemopressin and its fragments are critical for an effective interaction with the receptor molecular docking calculations provides new insight into new chemical scaffolds for the design of new [CB.sub.1].

Thermal isomerization of [CBC.sub.an] to [THC.sub.an] (nonaromatic analogues of plant cannabinoids cannabichromene (CBC) and [[DELTA].sup.1]-tetrahydrocannabinol (THC)) is predicted in silico. Density functional theory calculations have supported a similar isomerization mechanism for plant cannabinoids and docking studies have shown that [THC.sub.an] has a C[B.sub.1] receptor binding affinity similar to that of natural THC.

A solid-state NMR and site-directed spin labeling/EPR with a synthetic peptide are used to study the influence of membrane environment on human cannabinoid 1 (h[CB.sub.1]) receptor transmembrane helix (TMH) conformational dynamics. The data obtained indicated that h[CB.sub.1](T377-E416) backbone dynamics as well as spin-label rotameric freedom are sensitive to and altered by the phospholipid bilayer environment of peptide, which exerted a dynamic influence on the conformation of a TMH critical to signal transmission by the h[CB.sub.1] receptor.
