Objective: Various stress factors have been shown to cause significant and permanent injury to both mother and foetus. The histological examination of the brains of pregnant mice exposed to chronic immobilization stress in terms of the mechanisms of cell death was the premise of this study.

Aim: Traumatic brain injury (TBI) is the leading cause of death and disability in childhood. The aim of the present study is to determine the acute effects of resveratrol (Res) in the hippocampus and parietal cortex in 7-day-old rat pups subjected to contusion injury.

Uterus leiomyomaları, üreme çağındaki kadınlarda % 77 gibi yüksek sıklıkla meydana gelen, iyi huylu myometrial tümörlerdir. Leiomyomalar sıklıkla menoraji, dismenore, pelvik basınç ve ağrı, infertilite ve tekrarlayan gebelik kayıplarına neden olmaktadır. Bu tümörler premenopozal kadınlarda histeroktemi için önemli bir göstergedir. Son birkaç yıl içerisinde bazı medikal ve minimal invazif cerrahi girişimler, uterusunun korunmasını isteyen kadınlarda histerektomiye alternatif olarak geliştirilmektedir. Sıklıkla görülen uterus leiomyomalarının kadın sağlığı ve yaşam kalitesi üzerine büyük bir etkisi vardır. Bu nedenle tedavi ve bu hastalıkları önlemek için yeni yollar tanımlamak oldukça önemlidir.Bizde çalışmamızda uterus leiomyoması üzerine kurkuminin etkisini histokimyasal, immunohistokimyasal ve biyokimyasal analizler ile incelemeyi amaçladık.Yöntem:Çalışmada gruplar aşağıdaki gibi düzenlenmiştir.1.Kontrol Grubu: Hiçbir işlem uygulanmadı, uterusları alındı. Bazal değerler için kullanıldı.2.Etanol Grubu: Etanol grubuna 30 gün boyunca günde birer defa intraperitoneal olarak 0,05 ml etanol enjekte edildi ve deney sonuna kadar başka bir işlem uygulanmadı.3.Leiomyoma Grubu: 25 gün boyunca günde birer defa intraperitoneal olarak 0,5 mg/kg estradiol benzoat enjekte edildi. Daha sonra 5 gün boyunca günde birer defa intraperitoneal enjeksiyon ile 4 mg/kg progesteron enjekte edildi.Deney sonuna kadar başka bir işlem uygulanmadı.4.Leiomyoma+mısıryağı (Çözgen) Grubu: Leiomyoma oluşturulduktan sonra 15 gün boyunca günde birer defa oral gavaj ile 2 cc mısır yağı verildi.5.Leiomyoma+kurkumin (Antioksidan) Grubu: Leiomyoma oluşturulduktan sonra15 gün boyunca günde birer defa 200mg/kg kurkumin 2 cc mısır yağında çözdürülürek oral gavaj ile verildi.Deney başlangıcı, leiomyoma oluşumu sonrası ve deney sonu deneklerin ağırlıkları ölçüldü ve kan örnekleri alındı. Deney sonunda tüm grupların eter anestezisi altında uterusları alındı.Histokimyasal incelemeler için; Hematoksilen & Eozin, Periyodik Asit Schiff ve Masson Trikrom, immünohistokimyasal incelemeler için; Ki-67, PCNA, VEGF ve α-SMA boyamaları yapıldı. Biyokimyasal incelemeler için östradiol ve progesteron düzeyleri incelendi.Bulgular:Histokimyasal incelemeler sonucunda leiomyoma grubu ve leiomyoma oluşturulup mısıryağı verilen gruplarda atipik leiomyoma, hücresel leiomyoma ve epiteloid leiomyoma olgularına rastlandı. Kollajen artışına bağlı olarak myometriyal genişlemeler gözlendi. Bazal membran ondülasyonunda bozulma olduğu gösterildi. Leiomyoma+kurkumin grubunda bu bulgulara rastlanmadı ve bulguların kontrol grubuna yakın olduğu izlendi. Etanol grubunda kontrol grubuna göre vasküler yapılarda artış olduğu gözlendi.İmmunohistokimyasal incelemeler sonucunda kontrol ve etanol gruplarına göre Ki-67, PCNA, VEGF ve α-SMA pozitif hücre sayısı leiomyoma ve leimyoma+mısıryağı grubunda anlamlı derecede artış gösterdi. Leiomyoma+kurkumin grubunda ise leiomyoma ve leiomyoma+mısıryağı grubuna göre immün pozitif hücre sayısında anlamlı azalma gözlendi.Biyokimyasal olarak östradiol(E2) ve progesteron(P) seviyeleri değerlendirildi.Kontrol ve etanol gruplarına göre leiomyoma ve leiomyoma+mısıryağı grubunda E2 ve P seviyeleri anlamlı derecede artış gösterdi. Leiomyoma+kurkumin grubunda ise leiomyoma ve leiomyoma+mısıryağı grubuna göre E2 ve P seviyelerinde anlamlı azalma gözlendi.Sonuç:Elde ettiğimiz sonuçlar kurkuminin leiomyomalar üzerine etkilerini histokimyasal,immünohistokimyasal ve biyokimyasal olarak göstermiştir. Leiomyomas frequently cause menorrhagia, dismenore, pelvic pressure and pain, infertility and repeating loss of pregnancy. These tumors are important indicators of hystorectomy in premenopausal women. In the last few years, some medical and minimal surgical attempts were developed to protect the uteruses of women who desire to keep them. Uterus leiomyomas which can be seen frequently has a big effect on women health and life quality. For this reason cure and the preventative attempts are extremely important.In our study, we aimed to investigate the effect of curcumin on uterine leiomyoma by histochemical, immunohistochemical and biochemical analyzes.Method:Groups are arranged within this study as the following.1. Control Group: No procedure was performed, the uterus was removed. Used for the basal values.2. Ethanol Group: Ethanol was injected intraperitoneally with 0.05 ml ethanol once a day for 30 days and no further treatment was applied until the end of the experiment.3. Leiomyoma Group: 0.5 mg / kg estradiol benzoate was injected intraperitoneally once a day for 25 days. Then, 4 mg / kg progesterone was injected intraperitoneally once a day for 5 days. No further treatment was performed until the end of the experiment.4. Leiomyoma+corn oil (Solvent) Group: After the formation of leiomyoma, 2 cc of corn oil was given by oral gavage once a day for 15 days.5. Leiomyoma+curcumin (Antioxidant) Group: After leiomyoma formation, they were given oral gavage by dissolving 200 mg /kg curcumin in 2cc corn oil once a day for 15 days. The weights of the test subjects, the end of the leiomyoma formation and the end of the experiment were measured and blood samples were taken. At the end of the experiment, uterus was taken under ether anesthesia in all groups. For the histochemical studies; Hematoxylin and Eosin, Periodic Acid Schiff and Masson's trichrome, for the immunohistochemical studies; Ki-67, PCNA, VEGF and α-SMA staining methods were used. Results: Histochemical studies revealed atypical leiomyoma, cellular leiomyoma and epithelioid leiomyoma in the leiomyoma group and the leiomyoma group and the corn oil group. Myometrial enlargements were observed due to increased collagen. It was shown that basal membrane degeneration was deteriorated. Leiomyoma + curcumin group did not show these findings and it was observed that the findings were close to the control group. In the ethanol group, there was an increase in vascular structures compared to the control group. As a result of immunohistochemical studies, Ki-67, PCNA, VEGF and α-SMA positive cell numbers increased significantly in the leiomyoma and leiomyoma + corn oil groups according to the control and ethanol groups. Leiomyoma + curcumin group showed a significant decrease in the number of immunocompetent cells compared to leiomyoma and leiomyoma + corn oil group. Biochemically estradiol (E2) and progesterone (P) levels were evaluated. E2 and P levels in leiomyoma and leiomyoma + corn oil group increased significantly according to the control and ethanol groups. Leiomyoma + curcumin group showed a significant decrease in E2 and P levels compared to leiomyoma and leiomyoma + corn oil group. Conclusion: Our results showed histochemical, immunohistochemical and biochemical effects of curcumin leiomyomas. Amaç:Çalı?mamızda, formaldehitin testisler üzerine olan toksik etkileri ve bu toksik etkilere kar?ı N-asetilsistein?in koruyucu etkisi histokimyasal, immünohistokimyasal ve biyokimyasal düzeylerde ara?tırıldı.Yöntem:Çalı?mamızda, 21 adet Wistar-Albino cinsi erkek sıçan üç gruba ayrıldı. Grup I?deki sıçanlar sham olarak kullanıldı ve bu gruptaki sıçanlara her gün fizyolojik serum enjekte edildi. Grup II?deki sıçanlara gün a?ırı olarak formaldehit enjekte edildi. Grup III?deki sıçanlara ise gün a?ırı olarak formaldehit enjeksiyonu ile birlikte her gün N-asetilsistein uygulandı. Bir aylık deney süresi sonunda, bütün sıçanlar eter anestezisi altında sakrifiye edildi. Daha sonra, sıçanların testisleri çıkartılarak çevre dokulardan ayrıldı. Testis örneklerinin bir bölümü histokimyasal ve immunohistokimyasal incelemeler için kullanıldı. Testis örneklerinin diğer kısmında ise süperoksit dismutaz (SOD) enzim aktivitesi ile malondialdehit (MDA) ve glutatyon (GSH) seviyeleri belirlendi.Bulgular:Yapılan histokimyasal incelemelerde kontrol grubunda normal testis yapısı gözlendi. Formaldehit grubunda seminifer tübüllerinde düzensiz dizilmi? germ hücreleri, germ hücre kaybı, intertisyel alanda bağ doku artı?ı ve bazal membrankalınla?ması gözlendi. N-asetilsistein grubunda ise normale yakın morfolojide testis yapısı gözlendi. Yapılan immunohistokimyasal incelemeler sonucu formaldehit grubu seminifer tübüllerinde sham grubuna oranla TUNEL (+) boyanan hücre sayısının ve aktif kaspaz 3 immunpozitif boyanan hücre sayısının arttığı, koruyucu olarak N-asetilsistein verilen grupta ise immunpozitif boyanan hücre sayısının azaldığı gözlendi. Biyokimyasal olarak ölçülen MDA, GSH ve SOD düzeyleri gruplar arasında istatistiksel olarak anlamlı değildi.Sonuç:Sonuç olarak, formaldehit maruziyetine bağlı olarak testisde olu?an histokimyasal hasarın ve apoptozisin N-asetilsistein uygulamasıyla baskılandığı belirlendi.Anahtar Sözcükler:Testis, Formaldetit, N-asetilsistein, Sıçan Objective:In our study, toxic effects of formaldehyde on testises and protective effects of n-acetylcysteine against these toxic effects were investigated at histolochemically, imunohistochemically and biochemically levels.Method:In our study, 21 male Wistar-Albino rats were divided into three groups. Rats in group I were used as sham and this rats in grup were injected with physiological saline every day. Rats in group II were injected every other day with formaldehyde. Rats in group III were administered n-acetylcysteine everyday with every other day injection of formaldehyde. At the end of one month experimental period, all rats were sacrificed under anesthesia of ether. Then the testes of rats were removed and dissected from the surrounding tissue. The some of testis specimens were used for histochemical and immunohistochemical examination. In the other part of the testis samples, superoxide dismutase (SOD) enzyme activity and malondialdehyde (MDA) and glutathione (GSH) levels were determined.Results:Histochemically investigations showed that on Formaldehyde group?s seminifer tubules showed irregularly arranged group of germ cells, germ cell deprivation, increase of connective tissue on interstitial area and thickening basement membrane. No significant changes were seen in N-acetylcysteine Protective Effect group. After immunohistochemical investigations apoptotic cell death was increased in Formaldehyde group compared with Sham group. N-acetylcysteine group decreased TUNEL (+) cells and Active Caspase 3 immunoreactive cells comparedwith Formaldehyde group. As measured by biochemical MDA, GSH and SOD levels between all the groups were not statistically significant.Conclusion:In conclusion, it was determined that histochemical damage and apoptosis in testis caused by exposure of formaldehyde were suppressed by administration of N-acetylcysteine.Key Words:Testis, Formaldehyde, N-acetylcysteine, Rat 115

Amaç:Çalışmamızda ağırlık atışı modelini kullanarak oluşturduğumuz travmatik beyin hasarının testis üzerine etkisi olup olmadığını araştırmayı amaçladık.Yöntem:Deney gruplarımız aşağıdaki gibi oluşturuldu:Grup I, Kontrol grubu: Hiçbir hasara maruz bırakılmayan deney grubudur. (n=7)Grup II, Travma grubu: Ağırlık atışı modeliyle travmatik beyin hasarına maruz bırakılan grup. (n=7)Travmanın kanıtlanması amacıyla, deneyden 24 saat sonra sakrifiye edilen deneklerden alınan beyin dokularından kesitler alınarak, cresyl violet boyaması yapıldı. Boyama yapılan kesitlerde hücre sayımı gerçekleştirildi. Alınan diğer doku olan testislere ise histokimyasal değerlendirme amacıyla H&E, PAS ve Masson Trikrom boyamaları yapıldı. İmmünohistokimyasal olarak TUNEL ve Aktif Kaspaz 3 boyaması yapılarak apoptoz değerlendirildi. Elektron mikroskobik olarak testislerin ince yapısı incelendi.Sakrifikasyon esnasında deneklerden alınan kan örnekleriyle GnRH, testosteron, LH, FSH hormon seviyeleri ve MDA, GPx seviyeleri değerlendirildi. Sonuçlar istatistiksel olarak gruplar arasında karşılaştırıldı.Bulgular:Kontrol grubunun histolojik olarak değerlendirilmesinde testis dokusu normal gözlendi. Seminifer tübüller içinde tabakalanmış spermatogenik seri hücreleri, intertisiyel alanda bulunan leydig hücreleri ve bağ doku hücreleri, dokuyu etrafından saran düzensiz sıkı bağ dokusu tunika albuginea gözlemlendi.Travma grubunda ise histolojik olarak değerlendirme yapıldığında spermatogenik seri hücre sayılarında azalma gözlendi.TUNEL boyamasında DNA fragmantasyonu değerlendirildiğinde kontrol grubu da dahil olmak üzere apoptoz görüldü. Grup II'de Grup I'e kıyasla daha fazla sayıda TUNEL pozitif hücrelerin bulunduğu saptandı.Sonuç:Çalışmamızda travmatik beyin hasarının testis üzerine olan etkisi histokimyasal, immünohistokimyasal, ultrastrüktürel ve biyokimyasal olarak incelendi. Travmatik beyin hasarının tübüllerde hasar oluşturduğu ve apoptoz oranında artışa neden olduğu gözlemlendi.Ancak travmatik beyin hasarının testise olası etkileriyle ilgili literatürde çalışma bulunmadığından daha farklı ağırlıklar kullanılarak çalışmaların artırılması ve araştırılması gerekmektedir. Purpose:We aimed to show the effects of traumatic brain injury on the testis.In this study, we aimed to investigate the impact of travmatic brain injury which was formed by weight drop model on testis.Material and Method:Animal groups were designed as showed belowGrup I, Control group: No application was performed to this animals (n=7).Grup II, Trauma group: Traumatic brain injury which is done by weight drop model (n=7).24 hours later trauma, animals were sacrified. Testis tissue slides were stained with Hematoxylene&Eosine (H&E), Masson Trichrome and Periodic Acid Schiff (PAS) for light microscopy. Aktif Kaspaz 3 and Immunohistochemical TUNEL stain (terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick-end labeling) were performed to determine apoptosis.And GnRH, testosterone, LH, FSH hormone levels and MDA, GPx levels were observed and statistically groups were compared.In histological evaluation, control group?s testis tissue was observed normal.Testis had an connective tissue capsule which is called tunica albuginea.On the other hand, in traumatic group there was a diminution in serial spermatogenic cells when histologically evaluated.In Tunel analysis, we evaluated DNA fragmentation and there was apoptosis in all the groups including control group. In group two we observed higher TUNEL positive cells when compared to group one cells.Result:In our study, we observed the impact of traumatic brain injury which is turned out by weight drop model on testis using immunohistochemistry, ultrastructural and biochemistry techniques. These results suggest that, traumatic brain injury causes damage in the tubulles and increase in ratio of apoptosis.In our opinion, more studies are needed to understand the mechanism of traumatic brain injury on testis damage. 81

Çalışmamızda yardımlı üreme teknikleri sikluslarında follikül aspirayonuyla elde edilen granüloza hücrelerindeki apopitoz oranlarının immunohistokimyasal ve ultrastrüktürel incelemesi ve folliküler sıvılarda ROS düzeylerinin karşılaştırılması amaçlanmaktadır.Çalışmamız prospektif klinik gözlemsel bir çalışmadır. Dokuz Eylül Üniversitesi Kadın Hastalıkları ve Doğum Anabilim Dalı, Tüp Bebek Merkezi'ne çocuk sahibi olamama şikayeti ile müracaat eden primer yada sekonder infertil tanısına ek olarak PCOS, erkek faktör ve açıklanamayan infertilite tanısı konan ve yardımla üreme tekniği siklusuna alınan 30 hasta katıldı. Bütün olgulardan toplanan folliküler sıvılarındaki ROS spektrofotometrik yöntemle incelendi, ışık mikroskopu değerlendirmesi ile granüloza hücrelerindeki TUNEL pozitif hücre insidansı değerlendirildi ve aynı zamanda granüloza hücrelerinde meydana gelebilecek organel düzeyindeki değişiklikler Transmisyon Elektron Mikroskobu ile incelendi.Çalışmamızda granüloza hücrelerindeki ışık mikroskopi değerlendirmesi sonucunda açıklanamayan infertil grubunda TUNEL pozitif hücre ortalaması PCOS ve erkek infertil gruplarına göre istatistiksel olarak anlamlı şekilde yüksek bulundu. Folliküler sıvı MDA düzeyleri açıklanamayan infertilite olgularında PCOS'lu olgulara ve erkek faktör grubuna göre yüksek olduğu tespit edildi. Folliküler sıvı GPx düzeylerinin ise PCOS'lu olgularda, açıklanamayan infertilite ve erkek faktör olgularına göre yüksek olduğu tespit edildi. Ancak gruplardaki folliküler sıvı MDA ve GPx seviyeleri arasındaki bu fark istatistiksel olarak anlamlı bulunamadı. Elektron mikroskopisi değerlendirmelerinde ise hem DNA fragmantasyonunun değerlendirildiği TUNEL testi sonuçları ile hem de folliküler sıvı MDA seviyelerinin değerlendirilmesi ile elde edilen sonuçlara paralel olarak ince yapı değişiklikleri gözlendi.Çalışmamızda TUNEL tekniği, oositi destekleyen granüloza hücrelerindeki DNA kırıklarının belirlenmesi ve böylece hücrelerin apopitotik yola girdiğinin gösterilmesi için kullanılmıştır. Buna ek olarak ultrastrüktürel olarak süreçteki dejenerasyon değerlendirilmiş ayrıca bu duruma follikül sıvısındaki oksidatif stresin etkisi de araştırılmıştır. Böylece çalışmamızda PCOS'lu olgularla açıklanamayan infertilite olgularının karşılaştırılması, bu 3 farklı teknik kullanılarak ilk kez değerlendirilmiştir.Anahtar kelimeler: Granüloza Hücresi, PCOS, Açıklanamayan İnfertilite, TUNEL, Apopitoz, Ultrastrüktür, ROS In this study, we wanted to investigate to compare the percentage of apopitosis in granulosa cells from follicular fluid use immunohistochemistry technique and ultrastructure of granulosa cells and also assessment for ROS respectively cycles in assisted reproductive technology.Our study was a prospective, nonrandomized clinical study. Dokuz Eylül University Institute of Health Sciences Ethic committee approved our application with 13 numbered decision at 19th March 2008 and also Dokuz Eylül University Ethics Committee of Clinic And Laboratory Researches, Faculty of Medicine, approved our application with 120 numbered and 28 March 2008. This study is a workshop comprising 31 IVF cycles which has applied to 11 PCOS infertile patients, 10 unexplaned infertile patients and 10 male factor infertil patients between April 2008 and December 2008, in Dokuz Eylul Unıversıty Faculty Of Medıcıne Department Of Obstetrıcs and Gynecology ,IVF Center.Follicular fluid from each follicle were separated to three groups after each case. TUNEL assay was used to detect of DNA fragmentation in granulosa cells, MDA assessment for ROS, GPx assessment for antioxidant enzyme respectively and apoptosis in granulosa cells was evaluated ultrastructurally by transmission electron microscopy.There was statistically significant difference the percentage of apopitotic cells in granulosa cells between PCOS and unexplaned infertility group. There was no statistically significant difference between the levels of MDA and GPx in follicular fluid in theree groups.With this work we have done, it was investigated immunohistochemistry and also ultrastructural apopitosis of granulosa cells which are promoting oocytes , besides the follicle fluid was investigated for the effects of oxidative stress in this situation.
