As filter feeders, marine bivalves inhabiting estuarine and coastal areas are directly exposed to microplastics (MPs) in water. To assess whether MPs number, and their shape, size, colour, and polymer type present in mussels (Mytilus galloprovincialis) and cockles (Cerastoderma edule) varied over one year, bivalves were collected over the year of 2019 in the lower part of the coastal Aveiro lagoon, Portugal. After extraction from the bivalve's whole-body soft tissues, a subset of the visually inspected particles was randomly separated for identification using the Fourier-transform mid-infrared (FT-MIR) spectroscopy. A fraction of the inspected particles, 26-32% of particles >100 μm, and 59-100% of smaller ones were confirmed as MPs. Concentrations varied within the intervals of 0.77-4.3 items g-1 in mussels and 0.83-5.1 items g-1 in cockles, with the lowest values observed in January. In winter, the accumulation of large-sized fibers was composed of a mixture of plastic types, which contrasted against the most abundant MPs in summer consisting mainly of polyethylene of diverse size classes and shapes. Temperature decrease registered in winter might have triggered a lower filtration rate, resulting in lower MPs concentrations in the whole-soft body tissues of organisms. Different properties of MPs found in bivalves between January-February and August-September appear to reflect changes in the characteristics of MPs available in the Aveiro lagoon. published

- Abstract
Background: Results from retrospective studies suggest that use of neuromuscular blocking agents during general anaesthesia might be linked to postoperative pulmonary complications. We therefore aimed to assess whether the use of neuromuscular blocking agents is associated with postoperative pulmonary complications. Methods: We did a multicentre, prospective observational cohort study. Patients were recruited from 211 hospitals in 28 European countries. We included patients (aged ≥18 years) who received general anaesthesia for any in-hospital procedure except cardiac surgery. Patient characteristics, surgical and anaesthetic details, and chart review at discharge were prospectively collected over 2 weeks. Additionally, each patient underwent postoperative physical examination within 3 days of surgery to check for adverse pulmonary events. The study outcome was the incidence of postoperative pulmonary complications from the end of surgery up to postoperative day 28. Logistic regression analyses were adjusted for surgical factors and patients' preoperative physical status, providing adjusted odds ratios (ORadj) and adjusted absolute risk reduction (ARRadj). This study is registered with ClinicalTrials.gov, number NCT01865513. Findings: Between June 16, 2014, and April 29, 2015, data from 22 803 patients were collected. The use of neuromuscular blocking agents was associated with an increased incidence of postoperative pulmonary complications in patients who had undergone general anaesthesia (1658 [7·6%] of 21 694); ORadj 1·86, 95% CI 1·53–2·26; ARRadj −4·4%, 95% CI −5·5 to −3·2). Only 2·3% of high-risk surgical patients and those with adverse respiratory profiles were anaesthetised without neuromuscular blocking agents. The use of neuromuscular monitoring (ORadj 1·31, 95% CI 1·15–1·49; ARRadj −2·6%, 95% CI −3·9 to −1·4) and the administration of reversal agents (1·23, 1·07–1·41; −1·9%, −3·2 to −0·7) were not associated with a decreased risk of postoperative pulmonary

Detailed particle models by taking into account the material grain structure explicitly consider the material randomness, including a size limiter for damage localization. A VGMC3D contact model is presented that considers the polyhedral particle shape in an approximate way. The VGCM3D flexible contact model is validated against known experimental data on a granite rock, namely triaxial tests and Brazilian tests.

A oxidação de lípidos constitui a principal causa de degradação da carne, levando à perda do seu valor nutritivo. A quantidade intramuscular de agentes oxidantes é finamente regulada por agentes redutores endógenos como o ascorbato ou catalases que decrescem rapidamente durante a conversão do músculo em carne. Assim, os níveis de catalase ou o conteúdo lipídico podem ser bons indicadores da qualidade da carne. Nesse contexto, o desenvolvimento do estudo aqui descrito possibilitou aos alunos de uma turma-piloto da unidade curricular Bioquímica I do Mestrado Integrado em Medicina Veterinária, da Universidade de Évora, participarem ativamente no planeamento e na preparação de uma atividade laboratorial, com o objetivo de determinar marcadores nutricionais e de peroxidação lipídica da carne de Meleagris ocellata, Sus scrofa domesticus e Bos taurus. Os resultados obtidos na quantificação gravimétrica de lípidos, espetrométrica do colesterol e catalases, mobilizaram os alunos para uma aproximação bioquímica à caraterização de carnes de diferente origem animal. Lavenders belong to the family Labiatae and are one of the most popular aromatic plants widely used in food industry, perfumery and pharmaceutical preparations. Their oils are traditionally believed to be antibacterial, antifungal, carminative, sedative, antidepressive, disinfectant, and effective for burns and insect bites [1]. Previous studies have shown that the essentials oils of Lavandula stoechas subsp. luisieri Rozeira (L. luisieri) and Lavandula viridis L’Hér have a broad-spectrum antimicrobial activity [2]. In present work we have developed a set of studies in vivo, using animal model, in order to evaluate the toxicological proprieties of essential oils (EOs) of two aromatic plants endemic of south of Portugal, namely L. luisieri from Alentejo and L. viridis from Algarve. EOs were obtained from aerial part of plants by hydrodistillation and chemical composition was evaluated by GC-FID. Acute toxicity (LD50) was determined in Swiss mice according to Up-and-Down OECD procedure [3] and oral toxicity assays were performed based on hippocratic screening during 14 days [4], as required by the Direção Geral de Veterinária and FELASA. Histological analyses of the liver and kidney, as well as quantification of biomarkers: aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), gamma glutamyltransferase (γ-GT), creatinine and urea were performed.

Under normoxic conditions, living organisms can very efficiently prevent reactive oxygen (ROS) accumulation and attenuate oxidative damages, using various defensive strategies such as those involving the peptide glutathione. However, sudden exposure of eukaryotic cells to xenobiotic as herbicides may cause a redox imbalance of the cell, causing oxidative stress. The yeast Saccharomyces cerevisiae is a promising unicellular eukaryotic organism for the toxicological evaluation of xenobiotic as herbicides, because its cellular structure and functional organization have many similarities to those of higher-level organisms. However, the toxicity of phenylureas and triazines herbicides in yeast is still poorly understood. These photosystem II inhibitors herbicides are mainly used in agricultural applications for selective control of germinating grass and broad-leaved weeds in many crops (e.g. cereals). Diuron is also employed for total weed control on non-cultivated areas as maintenance of roads, railways and parks. On the other hand, atrazine (ATZ), diuron (DIU) and isoproturon (IPU) are the most used herbicides in Europe that are often found in contaminated groundwater, surface water, and effluents of wastewater treatment plants. Consequently, the main purpose of this study was to compare the effects of ATZ, DIU and IPU on yeast cell growth and glutathione cycle enzymes.

In nature and in industrial musts, pollutants are available in a complex mixture where some specific microorganisms are able to mineralize or degrade them in more basic and less harmful compounds to the environment. Isoproturon, a phenylurea herbicide, is an environmental contaminant poorly degraded in soils and aquifers, reaching levels considered toxic by European legislation. Thus, it is urgent to discover microorganisms able to degrade or eliminate these pollutants in situations of accidental or systematic contamination, establishing the best culture conditions. Therefore, in this work it was intended to evaluate if peptone, a nitrogen source, essential for yeast growth, amends the extension of fermentative phase of a wild-type strain S. cerevisiae UE-ME3 caused by isoproturon. The results show that this wild-type yeast grown in peptone starvation conditions, in presence of 100 µM isoproturon, started early the fermentative-respiratory transition.

The sea lamprey is an anadromous species that migrates twice during its life cycle between freshwater and seawater. Microphagous larvae generally spend 4–5 years burrowed in the substrate of rivers and streams before undergoing metamorphosis that ends with the beginning of the juvenile trophic migration. Once metamorphosis is complete, sea lamprey juvenile downstream migrants are fully tolerant to 35 PSU seawater. Pollution resulting from industrial effluents may disturb the seawater acclimatization causing oxidative damages, and ultimately may lead to a decrease of sea lamprey population. The aim of this study was to compare salt acclimation of sea lamprey juveniles captured in river basins with different levels of aquatic pollution, using mitochondrial glutathione and malondialdehyde of gills and liver as markers of physiological stress and cell damages. The results showed that juveniles from Lima basin exhibited the highest levels of mitochondrial malondialdehyde in gills, even though significant changes in the stress markers of mitochondrial gills of all animals subject to salt acclimation were not detected. In addition, an increase in the oxidative damages of hepatic mitochondria of macrophthalmia from Vouga basin suggests the occurrence of metabolic failures with the potential to disturb the capacity to adaptation to the marine environment.

Edible mushrooms, moreover to their gastronomic characteristics, are a source of valuable nutrients and bioactive compounds with potential beneficial effects on human health. Currently mushrooms become attractive as functional foods due to medicinal properties and useful in preventing diseases shown antitumour and immunomodulating properties, cardioprotective, hepatoprotective, antibacterial, antiviral and antifungal effects. In the near future, could play an important role as a nutraceutical and/or therapeutic agents [1, 2]. The southern of Portugal, namely in Alentejo region, due to its Mediterranean microclimate and flora diversity, is one of the European regions with a high predominance of wild edible mushrooms Amanita ponderosa. Although there are many studies about the beneficial properties of constituents and some edible wild mushrooms, there are no studies on the biological activity of A. ponderosa [3]. Thus, the main objective of this study was to evaluate the hepatoprotective effect of fruiting bodies and mycelia of liquid cultures of some strains of A. ponderosa, with enzymatic activity determinations of ALT, AST, ALP and G-GT, after intoxication with ethanol in Wistar rats, as an experimental model. It was also previously evaluated the toxicity of these cultures using biological models: Swiss mice and Artemia salina. The dried fruiting bodies of mushroom and mycelia of cultures obtained from A. ponderosa, showed no citotoxicity against A. salina and also not observed acute toxicity in Swiss mice. The administration of mycelia of A. ponderosa liquid cultures seems to indicate hepatoprotective activity equivalent to that observed for the standard drug, silymarin. Also is observable in all enzymes studied a decrease in their levels in the bloodstream, showing capacity to reduce the liver damage induced by ethanol-administration.

Lavenders belong to the family Labiatae and represent some of the most popular medicinal plants extensively used in aromatherapy. In this study, we selected Lavandula stoechas L. subsp. luisieri (Rozeira) Rozeira, endemic of Portugal, collected in Évora (Alentejo). The aim of this study was to evaluate the acute toxicity and the analgesic and anti-inflammatory activities of the essential oil and aqueous extract from leaves of L. luisieri. The essential oil was obtained by hydrodistillation in Clevenger type apparatus and the resulting hydrolate was lyophilized. Chemical composition of essential oil was evaluated by GC/FID and extract was characterized by phytochemical tests and total phenolics and flavonoids quantification. Acute toxicity was evaluated in vitro in brine shrimp (LC50) and in vivo in Swiss mice, by pharmacological screening, determination of LD50, according to Up-and-Down OECD Procedure and liver and kidney histopathological study. Pharmacological properties, namely, analgesic and anti- inflammatory activities were evaluated in rats.

As plantas do género Lavandula L. pertencentes à família Labiatae, são plantas aromáticas de grande importância económica muito usadas na medicina tradicional. Neste estudo, foi selecionada uma espécie natural do Alentejo, Lavandula stoechas L. subsps. luisieri (Rozeira) Rozeira, vulgarmente conhecida por rosmaninho, recolhida de uma mancha da Herdade da Mitra (Évora) O objetivo deste estudo foi avaliar as atividades analgésica e anti-inflamatória, e avaliar a toxicidade aguda do óleo essencial e extrato aquoso das folhas de L. luisieri. O óleo essencial foi obtido por hidrodestilação em aparelho tipo Clevenger e a sua composição química foi avaliada por GC/FID, o hidrolato resultante foi liofilizado e caracterizado por testes fitoquímicos, com quantificação dos compostos fenólicos totais e flavonoides. As propriedades farmacológicas, analgésica e anti-inflamatória, foram avaliadas em ratos Wistar pela prova de Amour-Smith e pelo teste da indução do edema na pata, respetivamente [1]. A toxicidade aguda foi avaliada in vitro em Artemia salina com determinação da CL50 [2] e in vivo, em murganhos Swiss, com avaliação do screening farmacológico, determinação da DL50 [3] e análise histopatológica do fígado e rins. O óleo essencial e o hidrolato apresentaram atividade analgésica e anti-inflamatória nas doses de 100 mg/Kg e 500 mg/Kg, respetivamente. O óleo essencial apresentou citotoxicidade para A. salina, com valores de CL50 inferiores a 500 µg/mL, enquanto o hidrolato não apresentou toxicidade, para concentrações até 3500 µg/mL. O óleo essencial e hidrolato de L. luisieri apresentaram baixa toxicidade aguda, com valores de DL50 muito superiores a 2000 mg/kg, sem sinais clínicos de toxicidade, no entanto, observou-se um comportamento passivo dos animais administrados com óleo essencial nas concentrações de 1000 e de 2000 mg/Kg. Nos estudos histopalógicos observaram-se pequenas alterações ao nível das estruturas do fígado e do rim apenas para as doses mais altas de óleo essencial. Os estudos prosseguem com a avaliação toxicológica das funções hepática e renal com marcadores enzimáticos e avaliação dos mecanismos de ação anti-inflamatória, com vista utilização do óleo essencial e hidrolato como agentes nutracêuticos e/ou fitoterapêuticos.

The isoproturon (IPU) is an herbicide used in Autumn-Winter crops that often persists in soils and aquifers at levels considered toxic by European legislation. In eukaryotic cells the IPU can generate ROS and consequently oxidative stress and it may be involved in triggering of serious illnesses like cancer. Accordingly, it is urgent to find microorganisms useful in bioremediation of this phenylurea. The aim of this study was to evaluate the effect of IPU on survival and antioxidant response of wild-type Saccharomyces cerevisiae UE-ME3 from Alentejo wines, Portugal, that present great resistant to metals and adverse cellar conditions. S. cerevisiae at mid-exponential phase were inoculated in absence (YEP) or presence of 2% glucose (YEP-D) liquid media without or with 100 µM isoproturon (YEP-IPU; YEP-D-IPU) and incubated at 28 ºC during 72 h. The cultures were used to draw growth curves (OD; cfu) and to estimate dry weight as well as to prepare post-12000 g supernatant for spectrophotometric determination of protein contents, CAT A, G6PD, and NADP-ME activities. Yeast cells grown in presence of IPU, using glucose as main carbon source or in glucose starvation conditions exhibit a similar growth pattern (OD). However, the number of viable cells (cfu) is greater whenever yeast cells were grown in presence of IPU, fact that reflects higher cell proliferation conditioned by IPU. The CAT A, G6PD and NADP-ME activities has showed higher values in S. cerevisiae grown in YEP-D medium than yeast cells grown in YEP-IPU media. The same response pattern was observed when compared YEP-D-IPU with YEP-IPU. Probably, this response can be related with a catabolic repression of oxidative metabolism concomitantly with a eventual enhances of glycolytic flux and fermentation even in the presence of oxygen, caused by IPU inhibition of cytoplasmic conversion malate-pyruvate of S. cerevisiae UE-ME3, which conferred a selective growth advantage useful in bioremediation.

The main purpose of this study was to evaluate the effect of IPU on the antioxidant response of the wine-wild Saccharomyces cerevisiae UE-ME3 from Alentejo, Portugal, that present great resistant to vanadium and adverse conditions of wine maker. Cells at mid-exponential phase were inoculated in 2% glucose YEPD or 0% glucose YEP media with or without 100µM IPU incubated at 28ºC, 72h. The culture were used to obtain growth curves (DO; cfu), dry weight and to prepare post-12000g supernatant for determination of protein and antioxidant power (DPPH), malonaldehyde (MDA), glutathione (GSH) and glutathione disulfide (GSSG) level by fluorescence and spectrophotometry. The results show that yeast grown in presence of IPU exhibit a similar growth profile of the cells grown in glucose starvation conditions. However, the number of viable cells is greater whenever yeast cells were grown in presence of IPU, either in the presence or absence of glucose, fact that reflects higher cell proliferation conditioned by IPU. The glucose starvation has also caused a decrease of post-12000g supernatant protein content and the capacity to scavenge free radicals estimated by DPPH, showing likewise an increase of supernatant glutathione and pellet protein content, which seems be essentials for survival to glucose deprivation. The differences in cfu, protein, GSH, GSSG and MDA contents and antioxidant power (DPPH; GSH/GSSG) of UE-ME3 grown in YEPD and YEP were attenuated by the presence of IPU, which may indicate that the presence of this phenylurea has decrease oxidative damages induced by glucose starvation. So we can presume that IPU may have been metabolized as carbon source to ensure the survival of this strain.

The isoproturon is a systemic herbicide applied in pre- and post-emergence control of annual grasses and broad-leaved weeds in winter crops that block photosynthesis, being listed by European Union as special substance that threaten the earth surface. Probably, can generate reactive oxygen species blocking the respiratory chain and cause oxidative stress and cell death. So, the main purpose of this study was to evaluate the effect of isoproturon on the antioxidant capacity of the wild-type Saccharomyces cerevisiae UE-ME3 and IGC-4072 strains. The UE-ME3 strain showed an adaptive response to this phenylurea, showing a growth similar to that control cells, an increase of non-protein thiols content, cell viability and glutathione antioxidant capacity, response that depend on glutathione reductase and glucose-6-phosphate dehydrogenase activities. However, the IGC-4072 strain in the presence of isoproturon showed cell death, exhibiting a poorly growth, dry weight and cell viability decrease, as well as a significant increase of free radicals scavenger (DPPH) and glutathione peroxidase activity.

35. Differential response to isoproturon by two strains of Saccharomyces cerevisiae

S. cerevisiae UE-ME3 grown in MB medium in the absence and presence of IPU show a similar growth profile and a stabilization of the biomass, non-protein thiols content, cell viability (cfu) and cell redox status estimated by GSH/GSSG ratio. This response seems depend on GR and G6PD activities increase, which probably activates the glutathione cycle, signs of adaptive response to phenylurea. However, S. cerevisiae IGC-4072 strain showed signs of cell death in the presence of IPU, exhibiting a more slowly growth profile, a decrease of dry weight and cell viability, as well as a significant increase of free radicals scavenger (DPPH) in presence of phenylurea, without significant changes of the GSH/GSSG ratio. As the value of the GSH/GSSG ratio is significant lower in IGC-4072 cells and GR activity undergoes a marked decrease in the presence of the IPU it is suspected that the worst IPU adaptive capacity displayed by this strain depends on a poor antioxidant power mediated by the glutathione.

Biotransformation enzymes catalyze oxidation of a diversity of xenobiotics, doing a key role in its detoxication. CYP1A and GST expression, induced by pollutants, seems to be highly conserved across vertebrate taxa. Since this system acts as a defense mechanism, the intra or interspecies differences have been attributed to adaptations to different habitats and trophic strategies. Thus, the main goal of this study was to evaluate the presence and inducibility of biotransformation enzymes in Petromyzon marinus (agnathan), because a clear causal link between toxicological effects on individuals and population responses have demonstrated reduced abundance in wild populations. Sampling occurred at the beginning of the sea lamprey downstream migration in three Portuguese river basins: Lima, Vouga, and Mondego.

Isoproturon, an herbicide of pre- and pos-emergence of Autumn-Winter crops, persists occasionally in soil, groundwater and biological systems at levels above those established by European Directives. Saccharomyces cerevisiae UE-ME3 exposed in stationary phase to 50 and 100 mM isoproturon exhibit growth rates higher than control or exposed cells to 5 and 25 mM of this phenylurea. However, in S.cerevisiae UE-ME3 grown in the presence of 5 mM isoproturon, were observed a decrease of GSH/GSSG ratio, an increase of cytoplasmatic MDA level, GR and GPx activities, usual markers of cell damage and oxidative stress. Nevertheless, S.cerevisiae grown at 25, 50 and 100 mM isoproturon, have developed adaptive responses to this phenylurea by stabilization of its reducing environment, lipid peroxidation decrease and GR, GPx activities increase, events regulated by isoproturon level in culture medium, facts which suggest that this yeast strain can be useful on bioremediation.

39. Avaliação nutricional de citrinos em agricultura biológica

Neste trabalho apresentam-se alguns resultados preliminares referentes à caracterização e avaliação do estado nutricional de um pomar de laranjeiras (Citrus sinensis (L.) Osb. cv. Valencia Late instalado em agricultura biológica num solo com elevado teor de calcário activo do Centro de Experimentação Agrária de Tavira da Direcção Regional de Agricultura do Algarve. Com o objectivo de caracterizar o solo do pomar em estudo foram realizadas colheitas de solo a duas profundidades (0-25 e 25-50 cm) e efectuadas diversas determinações, nomeadamente: textura, matéria orgânica, fósforo e potássio assimiláveis, e calcário total e activo. Durante 2 anos (2002 e 2003) foram efectuadas colheitas de flores na plena floração e colheitas de folhas de Abril a Outubro e foi determinada a sua composição mineral (macro e micronutrientes). Nas mesmas datas de amostragem fizeram-se estimativas do teor de clorofila total das folhas através da utilização do aparelho de SPAD-502. Os resultados são discutidos com base nas variações sazonais dos nutrientes nas folhas e flores e comparados com os valores referenciados para a agricultura convencional. The atrazine attracts special attention as pollutant because of itspersistence in the aquatic environment. Although this herbicide has been studied in teleost, its toxicity in the sea lamprey, Petromyzon marinus is still poorly understood. Oxidative stress may occur if chemical pollutants contribute to block the capacity of mitochondria to generate ATP with continuous production of reactive oxygen species (ROS), disturbing the success of P. marinus seawater acclimation. So, the aim of this study was to evaluate how atrazine influences the malondialdehyde (MDA), glutathione (GSH) and glutathione disulfide (GSSG) contents of gills and liver mitochondria of juveniles from Lima river basin, Portugal during salt acclimation. Sampling occurred at the beginning of the P. marinus downstream migration. The sampled juveniles were transported alive to the laboratory and maintained in 200 l tanks with LSS 8 life support system. Two groups of 40 specimens were hold in tanks with 50 or 100 lg/l atrazine, during 30 days. The salinity was gradually increased from 0 to 35 psu,following a three step procedure during a 30 days period. The control group was maintained in freshwater without atrazine. Mitochondria obtained by centrifugation at 15000 g, 30 min, 4°C, of tissues homogenates prepared in 50 mM Tris-HCl pH 7.5 buffer were used in determination of ROS, MDA, GSH and GSSG by fluorescence. The statistical analysis were performed by ANOVA I and Duncan (p < 0.05), using SPSS 22 for Windows.The results showed that in P. marinus juveniles, no significant changes in the markers of oxidative stress and cell damages were detected in the mitochondrial gills. Nevertheless, in the animals exposed to 50 lg/l atrazine the content in glutathione and GSSG increased. A similar pattern of stress markers was detected in hepatic mitochondria. However, in the presence of atrazine, the MDA level of the mitochondria of liver increased threefold in the animals during salt acclimation. The high level of mitochondrial damages, detected in the hepatic mitochondria of macrophthalmia treated with atrazine, suggests that herbicide exposure caused metabolic failures which can disturb the adaptation of these specimens to the oceanic feeding phase. The hepatic mitochondrial MDA levels of P. marinus, may eventually detect sea lamprey contact with chlorine herbicides.
