Along with the transition to cleaner energy production methods, closing the processing loop on batteries is becoming one of the significant issues to tackle in this decade. The most promising recycling technique consists in the leaching of crushed cathode material, i.e. the dissolution of the solid battery material in an acid solution, to recover valuable metals from spent batteries. To lower process time and to use green organic solvents, ultrasonically enhanced leaching is a valid alternative to conventional processing. The mechanism of action of ultrasound during leaching is still unclear, and yet to be directly observed on solid particles. Therefore, this work aims to shed light on the underlying phenomena in the ultrasonically enhanced leaching process, by directly observing leached material. In particular, the focus is placed on the combined effect of ultrasound and acetic acid on NMC particles. Residual material from conventional and ultrasonically enhanced leaching was analyzed with inductively coupled plasma - optical emission spectrometry (ICP-OES), dynamic light scattering (DLS) and scanning electron microscopy (SEM). Conventional and ultrasonically enhanced leaching techniques were thus compared in terms of leaching efficiency, particle size distribution and morphological changes, demonstrating the beneficial effect of ultrasonic cavitation on mass transfer. Additionally, the NMC particles were exposed to ultrasound in water, to confirm that standalone ultrasonic cavitation does not lead to particles breakage. The understanding of the effect of ultrasound enables their targeted application in leaching processes and allows a deeper understanding of ultrasound in heterogeneous systems.

En este artículo estudiamos la carrera militar de Benito Machado en la frontera sur de Buenos Aires entre 1852 y 1872. Fue un modesto propietario y arrendatario, que desde la comandancia de la Guardia Nacional y del ejército de la frontera, logró transformarse en un importante dirigente mitrista en el sur de la campaña. No construyó su autoridad por ser un militar de carrera, sino más bien se destacó por su pericia en la guerra de frontera, por su capacidad para reunir y dirigir hombres, por tener aptitudes para el liderazgo y por poseer un amplio conocimiento del territorio. Tras el alejamiento del mitrismo de los gobiernos nacional y provincial, fue apartado del ejército y de la comandancia de la Guardia Nacional. A través de este caso, problematizaremos ciertos aspectos del orden social y militar en la frontera, como así también algunas cuestiones vinculadas a la relación entre lo nacional, lo provincial y lo local durante el contexto de la construcción estatal.

Strategies to repair the meniscus have achieved limited success; thus, a cell-based therapy combined with an appropriate biocompatible scaffold could be an interesting alternative to overcome this issue. The aim of this project is to analyze different cell populations and a collagen gel scaffold as a potential source for meniscus tissue engineering applications. Dermal fibroblasts (DFs) and mesenchymal stem cells (MSCs) isolated from adipose tissue (ASCs) or bone marrow (BMSCs) were analyzed. Two different fibro-chondrogenic media, M1 and M2, were tested, and qualitative and quantitative analyses were performed. Significant increases in glycosaminoglycans (GAGs) production and in fibro-cartilaginous marker expression were observed in MSCs in the presence of M1 medium. In addition, both ASCs and BMSCs cultured in M1 medium were used in association with the collagen hydrogel (MSCs-SCF) for the development of an in vitro meniscal-like tissue. Significant up-regulation in GAGs production and in the expression of aggrecan, collagen type I, and collagen type II was observed in BMSCs-SCF. This study improves knowledge of the potential of combining undifferentiated MSCs with a collagen gel as a new tissue engineering strategy for meniscus repair.

The coupling of hydrodynamic cavitation (HC) and photocatalysis (PC) predominantly exhibits a complementary nature, which is highlighted through a synergistic index. However, this calculation, based on pseudo-first order kinetics, fails to accurately represent the concentration data, often skewed by the formation of reaction intermediates. To address this, an ‘extended’ kinetic approach previously developed with two parameters, which accounts for the formation of intermediates, was adopted. The PC process was optimized for a simulated wastewater containing phenol of concentration (C0) of 100 ppm by applying a UV-A light source of intensity 175 ± 8 W/cm2 on an operating volume (VL) of 200 mL. Catalyst loading, solution pH and initial concentration (C0) were optimized. These optimal parameters were used to operate HC-PC (VL = 3500 mL) at a comparable illumination intensity across the 2 techniques’ reactors. It was observed that phenol conversion was observable only with halved C0 (= 50 ppm) for the HC-PC system, increasing catalyst loading from 0.5 to 1.0 g/L exhibited no increase in phenol conversion. The obtained results from experiments were interpreted with the help of the two-parameter model. For the PC system, the initial rate constant (k0) exhibits a similar trend to the final oxidation extent, however, it did not compare quantitatively. The second parameter – ‘y’ showed a high finite value, denoting the need for dosing external oxidants. The trends of the two-parameters with respect to different relevant parameters presented will help leverage this kinetic approach for AOPs, while optimizing and translating processes to larger scales of operation.

Nowadays, the research in the fields of conservation and restoration is focusing on recording large amounts of data and information in order to obtain highly accurate surveys that include most details of constructions. Documenting and surveying are fundamental activities in achieving the successful preservation of cultural heritage, but also in implementing efficient strengthening interventions of structures. The applications of methods and techniques that link the disciplines of geometrical surveys and structural analyses are still fragmented, and further research is necessary to fully transfer site information into structural models. In this article, the authors propose a methodology with which to link different restoration fields, with the aim of highlighting the strengths and weaknesses of the interoperability between surveys (carried out through detailed restitution from point clouds), heritage building information modeling (HBIM), and structural analyses to connect three different disciplines. The goal is to find a synthesis model that brings together historical and geometric characteristics, but also physical–mechanical properties, for a structural evaluation through a finite element model (FEM). The case study of the Escuela José Mariano Méndez is analyzed, an extremely damaged historical adobe building in Santa Ana, the second largest city of El Salvador; it is considered fundamental to the national economy, particularly after the “coffee boom” in the late 1800s. The site, located near the historic center, is also known as “Antigua Escuela de Artes y Oficios” and embodies the architectural traditions of the region, being built in adobe and stone. Roma Tre University and the Secretaría de Cultura de la Presidencia (SECULTURA) are partners in this research project to document and restore the historical and cultural value of this site.

Sickle cell disease (SCD) is an inherited blood disorder, due to a single point mutation in the β-globin gene (HBB) leading to multisystemic manifestations and it affects millions of people worldwide. The monogenic nature of the disease and the availability of autologous hematopoietic stem cells (HSCs) make this disorder an ideal candidate for gene modification strategies. Notably, significant advances in the field of gene therapy and genome editing that took place in the last decade enabled the possibility to develop several strategies for the treatment of SCD. These curative approaches were firstly based on the correction of disease-causing mutations holding the promise for a specific, effective and safe option for patients. Specifically, gene-editing approaches exploiting the homology directed repair pathway were investigated, but soon their limited efficacy in quiescent HSC has curbed their wider development. On the other hand, a number of studies on globin gene regulation, led to the development of several genome editing strategies based on the reactivation of the fetal γ-globin gene (HBG) by nuclease-mediated targeting of HBG-repressor elements. Although the efficiency of these strategies seems to be confirmed in preclinical and clinical studies, very little is known about the long-term consequences of these modifications. Moreover, the potential genotoxicity of these nuclease-based strategies must be taken into account, especially when associated with high targeting rates. The recent introduction of nuclease-free genome editing technologies brought along the potential for safer strategies for SCD gene correction, which may also harbor significant advantages over HBG-reactivating ones. In this Review, we discuss the recent advances in genome editing strategies for the correction of SCD-causing mutations trying to recapitulate the promising strategies currently available and their relative strengths and weaknesses.

During endochondral bone development, a complex process that leads to the formation of the majority of skeletal elements, mesenchymal cells condense, differentiating into chondrocytes and producing the foetal growth plate. Chondrocytes progressively hypertrophy, induce angiogenesis and are then gradually replaced by bone. Epidermal Growth Factor (EGF), one of many growth factors, is the prototype of the EGF-ligand family, which comprises several proteins involved in cell proliferation, migration and survival. In bone, EGF pathway signalling finely tunes the first steps of chondrogenesis by maintaining mesenchymal cells in an undifferentiated stage, and by promoting hypertrophic cartilage replacement. Moreover, EGF signalling modulates bone homeostasis by stimulating osteoblast and osteoclast proliferation, and by regulating osteoblast differentiation under specific spatial and temporal conditions. This evidence-based narrative review describes the EGF pathway in bone metabolism and endochondral bone development. This comprehensive description may be useful in light of possible clinical applications in orthopaedic practice. A deeper knowledge of the role of EGF in bone may be useful in musculoskeletal conditions which may benefit from the modulation of this signalling pathway.Key messages The EGF pathway is involved in bone metabolism. EGF signalling is essential in the very early stages of limb development by maintaining cells in an undifferentiated stage. EGF pathway positively regulates chondrocyte proliferation, negatively modulates hypertrophy, and favours cartilage replacement by bone. EGF and EGF-like proteins finely tune the proliferation and differentiation of bone tissue cells, and they also regulate the initial phases of endochondral ossification.

Despite the transition toward carbon-free energy carriers, liquid fossil fuels are expected to occupy an important market share in the future. Therefore, it is crucial to develop innovative technology for better combustion reducing the emissions of pollutants associated with their utilization. Water in oil (w/o) emulsions contribute to greener combustion, increasing carbon efficiency and reducing emissions. Water content, emulsions stability, and droplet size distributions are key parameters in targeting the efficient use of emulsions as combustibles. In particular, for fixed water content, the finer the emulsion, the better its beneficial effect on combustion. In this work, two emulsions, mechanically and ultrasonically generated, were compared. Cryogenic scanning electron microscopy (cryo-SEM) allowed the visualization of water droplets inside the oily matrix. No surfactants were added to the oil, due to its high asphaltenic content. Asphaltene molecular aggregates, namely clusters, act as natural surfactants stabilizing the emulsions by arranging at w/o interface and forming a rigid film. The asphaltenic rigid film is clearly visualized in this work and compared for the two emulsions. The results showed finer water droplets in the ultrasonically generated emulsion, together with a reduction in the thickness of the asphaltenic film. Ultrasonically induced cavitation favored the de-clustering (breakage of intermolecular forces) of asphaltene molecules. Thus, smaller clusters allowed to stabilize smaller water droplets resulting in an ultra-fine emulsion, which improves the combustion performances of the fuel.

Mechanotransduction refers to the ability of cells to sense mechanical stimuli and convert them into biochemical signals. In this context, the key players are focal adhesions (FAs): multiprotein complexes that link intracellular actin bundles and the extracellular matrix (ECM). FAs are involved in cellular adhesion, growth, differentiation, gene expression, migration, communication, force transmission, and contractility. Focal adhesion signaling molecules, including Focal Adhesion Kinase (FAK), integrins, vinculin, and paxillin, also play pivotal roles in cardiomyogenesis, impacting cell proliferation and heart tube looping. In fact, cardiomyocytes sense ECM stiffness through integrins, modulating signaling pathways like PI3K/AKT and Wnt/β-catenin. Moreover, FAK/Src complex activation mediates cardiac hypertrophic growth and survival signaling in response to mechanical loads. This review provides an overview of the molecular and mechanical mechanisms underlying the crosstalk between FAs and cardiac differentiation, as well as the role of FA-mediated mechanotransduction in guiding cardiac muscle responses to mechanical stimuli.

Abstract Bitter taste receptor TAS2R38 is expressed in the respiratory tract and can respond to quorum-sensing molecules produced by pathogens, stimulating the release of nitric oxide, with biocidal activity. TAS2R38 presents two main high-frequency haplotypes: the “taster” PAV and the “non-taster” AVI. Individuals carrying the AVI allele could be at greater risk of infections, including SARS-CoV-2. The aim of this study was to assess the frequency of PAV and AVI alleles in COVID-19 patients with severe or non-severe symptoms compared to healthy subjects to further corroborate, or not, the hypothesis that the PAV allele may act as a protecting factor towards SARS-CoV-2 infection while the AVI one may represent a risk factor. After careful selection, 54 individuals were included in the study and underwent genetic analysis and PROP phenotype assessment. Our investigation could not point out at a significant relationship between single nucleotide polymorphisms responsible for PROP bitterness and presence/severity of SARS-CoV-2 infection, as previous studies suggested. Our results uncouple the direct genetic contribution of rs10246939, rs1726866 and rs713598 on COVID-19, calling for caution when proposing a treatment based on TAS2R38 phenotypes.

Bioengineered materials represent an innovative option to support the regenerative processes of damaged tissues, with the final objective of creating a functional environment closely mimicking the native tissue. Among the different available biomaterials, hydrogels represent the solution of choice for tissue regeneration, thanks to the easy synthesis process and the highly tunable physical and mechanical properties. Moreover, hydrogels are biocompatible and biodegradable, able to integrate in biological environments and to support cellular interactions in order to restore damaged tissues' functionality. This review offers an overview of the current knowledge concerning hydrogel synthesis and characterization and of the recent achievements in their experimental use in supporting skin, bone, cartilage, and muscle regeneration. The currently available in vitro and in vivo results are of great interest, highlighting the need for carefully designed and controlled preclinical studies and clinical trials to support the transition of these innovative biomaterials from the bench to the bedside.
