This study characterized the effects of artesunate on thyroid cancer and partially identified the related molecular mechanisms. We determined the effect of artesunate on the proliferation of thyroid cancer cells using the MTT assay, cell colony formation experiments, and western blotting, and used flow cytometry to detect the apoptosis of cancer cells. Using a wound healing assay, Transwell chamber experiments, and western blotting, we determined the effect of artesunate on cancer cell migration. We also partially identified the molecular mechanism by co-cultivation of artesunate with the PI3K agonist 740Y-P. Artesunate significantly inhibited the growth, proliferation, migration, and invasion of thyroid cancer cells and promoted the apoptosis of cancer cells. Using co-cultivation with a PI3K agonist, we found that the inhibitory effect of artesunate on cancer cells was mainly due to suppression of the PI3K/AKT/FKHR signaling pathway. By inhibiting the PI3K/AKT/FKHR signaling pathway, artesunate induced apoptosis in thyroid cancer cells and inhibited their proliferation and migration. Les auteurs ont determine l'effet de l'artesunate sur la proliferation des cellules cancereuses de la thyroide a l'aide du test au MTT, d'experiences de formation de colonies cellulaires et de transferts de type Western. Ils ont egalement utilise la cytometrie en flux pour detecter l'apoptose des cellules cancereuses. L'artesunate inhibait significativement la croissance, la proliferation, la migration et l'invasion des cellules cancereuses de la thyro'ide, et favorisait l'apoptose des cellules cancereuses. En ajoutant en culture un agoniste de PI3K, ils ont decouvert que l'effet inhibiteur de l'artesunate sur les cellules cancereuses etait principalement du a la suppression de la voie de signalisation PI3K/AKT/FKHR. En inhibant la voie de signalisation PI3K/AKT/FKHR, l'artesunate induisait l'apoptose des cellules cancereuses de la thyro'ide et inhibait leur proliferation et leur migration. The PI3K/AKT pathway plays an important role in the development of osteosarcoma. RNF38 interferes with activation of the AKT pathway. Cryptochrome1 (CRY1) inhibits osteosarcoma proliferation through the AKT pathway. We aimed to clarify whether RNF38 affects the proliferation of osteosarcoma cells by regulating the PI3K/AKT pathway through its interaction with CRY1. The mRNA levels of RNF38 were determined using qRT-PCR. Protein levels of RNF38, p-p70S6, p70S6, +p-AKT, AKT, p-mTOR, mTOR, and CRY1 were detected by western blotting. The proliferation of osteosarcoma cells was detected using CCK-8 and colony formation assays. The interaction between CRY1 and RNF38 was detected by co-immunoprecipitation and GST pull-down assays. RNF38 expression was higher in Saos-2 and U20S cells than in hFOB cells. Overexpression of RNF38 promoted the proliferation of osteosarcoma cells, the number of colonies, and p-AKT and p-mTOR levels, suggesting that overexpression of RNF38 activated the PI3K/AKT pathway. In addition, RNF38 directly binds to the N-terminal of CRY1. The simultaneous knockdown of RNF38 and CRY1 restored the level of p-AKT, which was reduced by RNF38 knockdown alone. En outre, RNF38 peut se lier directement à l'extrémité N-terminale de CRY1. Cervical cancer is one of the leading causes of mortality amongst women in developing countries, and resistance to therapy is the main reason for treatment failure. Recent advances suggest that cancer stem cells (CSCs) are critically involved in regulating the chemo-resistant behavior of cervical cancer cells. In our study, cells with the CSC phenotype were isolated, and we examined the expression levels of stem cell markers and genes associated with epithelial--mesenchymal transition (EMT) using different assays. However, the cells with the CSC phenotype could not be cultured for further cytotoxicity studies, so we established a model of CSC in cervical cancer cells. We performed siRNA-mediated knockdown of E-cadherin in these cells, and studied them for EMT-associated stem-cell-like properties. We also performed dose-dependent cell viability assays using clinically relevant drugs such as cisplatin, cyclopamine, and GANT58 to analyze the drug resistant behavior of these cancer cells. We found that knockdown of E-cadherin induces EMT in cervical cancer cells, imparting stem-cell like characteristics along with enhanced tumorsphere formation, cell migration, invasiveness, and drug resistance. This is the first study to establish a CSC model in cervical cancer cells by knockdown of E-cadherin, which can be used to develop anti-cancer therapies. Le cancer du col de l'utérus est l'une des principales causes de mortalité chez les femmes des pays en développement et la résistance aux traitements est la principale raison de son échec thérapeutique. Des avancées récentes suggèrent que les cellules souches cancéreuses (CSC) sont impliquées de manière importante dans la régulation du comportement chimiorésistant des cellules du cancer du col de l'utérus. Dans cette étude, les cellules du phénotype CSC ont été isolées et l'expression d'un marqueur de cellules souches et d'un gène associé à la transition épithélio-mésenchymateuse (TEM) a été confirmée par différents tests. Cependant, ces cellules de phénotype CSC ne peuvent pas être cultivées pour des études de cytotoxicité supplémentaires. Les auteurs ont donc essayé d'établir un modèle de CSC de cellules du cancer du col de l'utérus. Ils ont procédé à l'élimination de la E-cadhérine dans ces cellules à l'aide d'un ARNsi et étudié chez elle les propriétés associées à la TEM de type cellules souches. Ils ont également réalisé un test de viabilité cellulaire dépendant de la dose en utilisant des médicaments cliniquement pertinents tels que le cisplatine, la cyclopamine et le GANT58 pour analyser la résistance aux médicaments de ces cellules cancéreuses. Ils ont constaté que le knockdown de la E-cadhérine induit la TEM dans les cellules cancéreuses du col de l'utérus, ce qui leur confère des caractéristiques semblables à celles des cellules souches et accroît la formation de microsphères tumorales, la migration, la capacité d'invasion et la résistance aux médicaments. Macrophages have a key role in shaping the tumour microenvironment (TME), tumour immunity and response to immunotherapy, which makes them an important target for cancer treatment.sup.1,2. However, modulating macrophages has proved extremely difficult, as we still lack a complete understanding of the molecular and functional diversity of the tumour macrophage compartment. Macrophages arise from two distinct lineages. Tissue-resident macrophages self-renew locally, independent of adult haematopoiesis.sup.3-5, whereas short-lived monocyte-derived macrophages arise from adult haematopoietic stem cells, and accumulate mostly in inflamed lesions.sup.1. How these macrophage lineages contribute to the TME and cancer progression remains unclear. To explore the diversity of the macrophage compartment in human non-small cell lung carcinoma (NSCLC) lesions, here we performed single-cell RNA sequencing of tumour-associated leukocytes. We identified distinct populations of macrophages that were enriched in human and mouse lung tumours. Using lineage tracing, we discovered that these macrophage populations differ in origin and have a distinct temporal and spatial distribution in the TME. Tissue-resident macrophages accumulate close to tumour cells early during tumour formation to promote epithelial-mesenchymal transition and invasiveness in tumour cells, and they also induce a potent regulatory T cell response that protects tumour cells from adaptive immunity. Depletion of tissue-resident macrophages reduced the numbers and altered the phenotype of regulatory T cells, promoted the accumulation of CD8.sup.+ T cells and reduced tumour invasiveness and growth. During tumour growth, tissue-resident macrophages became redistributed at the periphery of the TME, which becomes dominated by monocyte-derived macrophages in both mouse and human NSCLC. This study identifies the contribution of tissue-resident macrophages to early lung cancer and establishes them as a target for the prevention and treatment of early lung cancer lesions.
