Abstract

Despite the ubiquity of pollutants in the environment, their long-term ecological consequences are not always clear and still poorly studied. This is the case concerning the radioactive contamination of the environment following the major nuclear accident at the Chernobyl nuclear power plant. Notwithstanding the implications of evolutionary processes on the population status, few studies concern the evolution of organisms chronically exposed to ionizing radiation in the Chernobyl exclusion zone. Here, we examined genetic markers for 19 populations of Eastern tree frog (Hyla orientalis) sampled in the Chernobyl region about thirty years after the nuclear power plant accident to investigate microevolutionary processes ongoing in local populations. Genetic diversity estimated from nuclear and mitochondrial markers showed an absence of genetic erosion and higher mitochondrial diversity in tree frogs from the Chernobyl exclusion zone compared to other European populations. Moreover, the study of haplotype network permitted us to decipher the presence of an independent recent evolutionary history of Chernobyl exclusion zone's Eastern tree frogs caused by an elevated mutation rate compared to other European populations. By fitting to our data a model of haplotype network evolution, we suspected that Eastern tree frog populations in the Chernobyl exclusion zone have a high mitochondrial mutation rate and small effective population sizes. These data suggest that Eastern tree frog populations might offset the impact of deleterious mutations because of their large clutch size, but also question the long-term impact of ionizing radiation on the status of other species living in the Chernobyl exclusion zone.

Abstract

High levels of ionizing radiation (IR) are known to induce neurogenesis defects with harmful consequences on brain morphogenesis and cognitive functions, but the effects of chronic low to moderate dose rates of IR remain largely unknown. In this study, we aim at defining the main molecular pathways impacted by IR and how these effects can translate to higher organizational levels such as behavior. Adult zebrafish were exposed to gamma radiation for 36 days at 0.05 mGy/h, 0.5 mGy/h and 5 mGy/h. RNA sequencing was performed on the telencephalon and completed by RNA in situ hybridization that confirmed the upregulation of oxytocin and cone rod homeobox in the parvocellular preoptic nucleus. A dose rate-dependent increase in differentially expressed genes (DEG) was observed with 27 DEG at 0.05 mGy/h, 200 DEG at 0.5 mGy/h and 530 DEG at 5 mGy/h. Genes involved in neurotransmission, neurohormones and hypothalamic-pituitary-interrenal axis functions were specifically affected, strongly suggesting their involvement in the stress response behavior observed after exposure to dose rates superior or equal to 0.5 mGy/h. At the individual scale, hypolocomotion, increased freezing and social stress were detected. Together, these data highlight the intricate interaction between neurohormones (and particularly oxytocin), neurotransmission and neurogenesis in response to chronic exposure to IR and the establishment of anxiety-like behavior.

Abstract

International audience; This study examined the short term transfer of carbon-14 (14C) in the common carp Cyprinus carpio under laboratory conditions. Various experiments were achieved in order to investigate direct or trophic transfer for 4 days, using waterborne 14C-labelled arginine or 14C-labelled food pellets respectively. Radiolabelled food was prepared with 14C-labelled arginine or glucose in order to test how transfer kinetics might vary with the biochemical form of 14C. Elimination experiments were achieved using fish fed for 5 days on radiolabelled food and then placed under starvation for 4 days. In all experiments, water, food and fish activities were monitored every day. Different fish fractions (whole body, muscle) were sampled in order to elucidate the role of muscle as a potential storage. Results suggested that direct water-to-fish absorption rate was 20% d-1 per fish. Carps incorporated 14.3% of the absorbed 14C. Fish activity did not increase over days, due to a strong decrease in 14C concentration in the water (resulting from aquarium sorption). During trophic transfer experiments, food was entirely ingested and 14C sources rapidly assimilated. For either arginine or glucose, results suggested that 19-20% of ingested 14C was incorporated, yielding a significant increase in fish activity over days. No difference in mass-specific activity was observed among muscle and whole body. Total activity in the muscle represented 29%-32% of whole body activity, this proportion reflecting the contribution of muscle to whole body weight. During elimination experiments, results showed a significant decrease in whole body total activity and mass-specific activity with arginine. The decrease was not significant with glucose due to a great variability among fish. Results suggested that an essential amino-acid like arginine can be used as an energy source under starvation and that muscles can act as a storage for essential amino-acids.

Abstract

International audience; Le contexte des effets biologiques de l’exposition aux rayonnements ionisants est une préoccupation majeure pour la radioprotection de l’homme et de l’environnement. Ceci a récemment été mis en exergue après l’accident de Fukushima, notamment dans le contexte de la quantification du risque pour la santé des individus et populations exposées aux faibles doses. Augmenter la connaissance des effets des faibles doses sur les espèces humaines et non-humaines permettrait de gagner en précision pour la prédiction des effets et la prise de décision.L’industrie nucléaire rejette dans l’environnement différents types de radionucléides, principalement des émetteurs gamma (137Cs, 60Co, 54Mn), mais aussi du tritium, le plus important en termes de production (200g correspondant à 7,2.106 Bq/an dans le monde). Le tritium est un isotope radioactif de l’hydrogène qui émet des rayonnements beta. Sa demi-vie est de 12,3 ans. Les rejets de tritium sont supposés augmenter dans le futur avec le développement de réacteurs basés sur la fusion nucléaire (ITER). Les écosystèmes aquatiques, habités pas de nombreuses espèces animales et végétales, représentent généralement le récepteur final d’un grand nombre de polluants, incluant des radionucléides dont le tritium. L’exposition à des niveaux élevés de radionculéides est connue pour induire de nombreuses altérations sur différentes fonctions de l’organisme, de la molécule (intégrité de l’ADN) à la population (effets sur la survie et la reproduction). Cependant, les effets de faibles niveaux d’exposition, plus pertinents d’un point de vue environnemental, sont peu connus.Cette présentation résume les différents travaux menés depuis plusieurs années à l’IRSN sur le tritium, sur différents modèles biologiques : le poisson zèbre, Danio rerio, espèce modèle facile à maintenir en laboratoire, afin de comprendre les mécanismes d’action du tritium dans le cadre d’expositions contrôlées ; et le tête de boule, Pimephales promelas, espèce autochtone du continent Nord-Américain, afin d’étudier les effets du tritium seul ou un mélange dans un contexte réel de contamination aux effluents d’un centre de recherche. L’évaluation des effets du tritium a été réalisée par l’analyse d’une batterie de biomarqueurs à différentes échelles biologiques, de la molécule à l’individu. L’analyse de l’ensemble des résultats permettra de mieux comprendre les effets du tritium, seul ou en mélange, sur la physiologie du poisson.

Abstract

Uranium (U) toxicity patterns for fish have been mainly determined under laboratory‐controlled waterborne exposure conditions. Because fish can take up metals from water and diet under in situ exposure conditions, a waterborne U exposure experiment (20 μg L−1, 20 d) was conducted in the laboratory to investigate transfer efficiency and target organ distribution in zebrafish Danio rerio compared with combined waterborne exposure (20 μg L−1) and diet‐borne exposure (10.7 μg g−1). 233Uranium was used as a specific U isotope tracer for diet‐borne exposure. Bioaccumulation was examined in the gills, liver, kidneys, intestine, and gonads of D. rerio. Concentrations in the organs after waterborne exposure were approximately 500 ng g−1 fresh weight, except in the intestine (> 10 μg g−1 fresh wt) and the kidneys (200 ng g−1 fresh wt). No significant difference was observed between waterborne and diet‐borne conditions. Trophic U transfer in organs was found but at a low level (< 10 ng g−1 fresh wt). Surprisingly, the intestine appeared to be the main target organ after both tested exposure modalities. Abstract

ABSTRACT Due to a lack of information on the assessment of uranium's (U) toxicity, our work aimed to compare the effects of U on the crayfish Procambarus clarkii with those of the well documented metal: cadmium (Cd). Accumulation and impacts at different levels of biological organization were assessed after acute (40 µM Cd or U; 4-10 days) and chronic (0.1 µM Cd or U; 30-60 days) exposures. The survival rates demonstrated the high tolerance of this species toward both metals and showed that Cd had a greater effect on the sustainability of crayfish. The concentration levels of Cd and U accumulated in gills and hepatopancreas were compared between both conditions. Distinctions in the adsorption capacities and the mobility of the contaminants were suspected. Differences in the detoxification mechanisms of both metals using transmission electron microscopy equiped with an energy dispersive X-ray were also pointed out. In contrast, comparison between the histological structures of contaminated hepatopancreas showed similar symptoms. Principal component analyses revealed different impacts of each metal on the oxidative balance and mitochondria using enzymatic activities and gene expression levels as endpoints.
