Background:Staphylococcus saprophyticus is the second most frequent community-acquired causative agent of urinary tract infection (UTI). The objective of this study was to evaluate the susceptibility profile and resistance detection in Staphylococcus species. isolated from patients with UTI. Materials and Methods: The isolates were investigated using the disk diffusion method, Vitek I system, E-test®, and detection of the mecA gene. Results: Most isolates (76.2%) were resistant to oxacillin by the disk diffusion method, followed by those resistant to penicillin (72.2%). The oxacillin disk diffusion method, E-test, and Vitek I method showed higher sensitivity (94.4%) and lower specificity (28.9, 26.5, and 24.0%, respectively) than the cefoxitin disk diffusion test (sensitivity: 83.5%, specificity: 85.5%) for the detection of oxacillin resistance. Conclusions: The large number of oxacillin-resistant isolates indicates that the breakpoint value recommended by the Clinical and Laboratory Standards Institute may overestimate oxacillin resistance in S. saprophyticus. Background: Cervical dystonia (CD) involves clinical and motor manifestations, and visual and cognitive dysfunctions may also be frequent., Objective: To evaluate functional vision, visual attention, and cognitive aspects in patients with CD compared with a control group., Methods: Fifty patients with CD were assessed using the Useful Field of View Test (UFOV), the Mini Mental State Examination (MMSE), and the Trail Making Tests (TMT-A and TMT-B), and compared with an identical number of health controls matched by sex, age, and educational level., Results: No differences were seen between the groups in terms of MMSE score ( P  = 0.481), but the CD patient group had poorer scores for the TMA-A ( P  = 0.004) and TMT-B ( P  = 0.004). For the UFOV subtests, a decrease was found for visual processing speed ( P  < 0.001), divided attention ( P  < 0.001), and selective attention ( P  = 0.001), as well as higher frequency in the categories with higher risk index in the UFOV test ( P  < 0.001)., Conclusion: Patients with CD may exhibit decreased functional vision and visual attention, as well as higher risk in performing complex activities., Competing Interests: The authors declare that there are no funding sources or conflicts of interest relevant to this work., (© 2021 International Parkinson and Movement Disorder Society.)

Poly(ε-caprolactone) (PCL) is a hydrophobic and cytocompatible aliphatic polyester that has been used to produce PCL-based nanofibrous for both wound healing and tissue repair. However, the high hydrophobicity and low water adsorptive have been challenges for developing PCL-based materials for use in tissue engineering field. Here, we report a new polymer (a hydrophilic amino-functionalized tannin (TN)) that is associated with PCL for developing PCL-TN blends at different PCL:TN weight ratios (100:0, 95:5, 85:15 and 78:22). PCL:TN ratio may be tuned to modulate hydrophilicity and cytocompatibility of the nanofibers. The neutralization step and surface wettability played an important role in the attachment of human adipose-derived stem cells (ADSC cells) on PCL-TN membranes. Also, fluorescence images confirmed great proliferation of ADSC cells on the PCL-TN electrospun surfaces. Yet, neutralized PCL-TN nanofibers promoted bactericidal activity against Pseudomonas aeruginosa. The aim of this study was to evaluate the effects of Wnt signaling through lipoprotein receptor-related protein 6 (LRP6) and Frizzled6 on the endothelial differentiation of dental pulp stem cells (DPSCs). DPSCs were stably transduced with enhanced green fluorescent protein (EGFP)-tagged lentiviral vectors (short hairpin RNA-LRP6, short hairpin RNA-Frizzled6, or empty vector controls). We evaluated the effects of LRP6 and Frizzled6 on expression of endothelial markers and on capillary tube formation mediated by DPSCs induced with recombinant human Wnt1 (rhWnt1) and/or recombinant human vascular endothelial growth factor 165 (rhVEGF 165 ). In vivo, tooth slices/scaffolds were seeded with LRP6-silenced, Frizzled6-silenced, or vector control DPSC cells and transplanted into immunodeficient mice. The density of blood vessels generated by DPSCs differentiated into vascular endothelial cells was analyzed by immunohistochemistry for EGFP. The rhWnt1 and rhVEGF 165 induced expression of active β-catenin in control DPSCs and in Frizzled6-silenced DPSCs, but not in LRP6-silenced DPSCs. Furthermore, VEGF and interleukin-8 were downregulated in LRP6-silenced DPSCs, but not in control DPSCs or in Frizzled6-silenced DPSCs (P < .05). Likewise, rhWnt1 and rhVEGF 165 induced expression of the endothelial marker VEGF receptor-2 in control DPSCs and in Frizzled6-silenced DPSCs, but not in LRP6-silenced DPSCs. These data correlated with a trend for lower density of capillary sprouts generated by LRP6-silenced DPSCs when compared with control DPSCs in Matrigel. In vivo, tooth slice/scaffolds seeded with DPSC-short hairpinRNA-LRP6 cells showed lower density of human blood vessels (ie, EGFP-positive blood vessels), when compared with tooth slice/scaffolds seeded with vector control cells (P < .05). Root perforations may lead to a loss of integrity in the root and periodontium, violations of the biologic periodontal distance, and injuries to periodontal tissue. This study sought to analyze the effect of root canal biomechanical preparation on the microhardness and the marginal sealing ability of different materials used to treat root perforations. Standard root perforations were performed in 96 bovine incisors. The teeth were divided into four groups (n = 24), based on the material used to treat those teeth: Mineral trioxide aggregate (MTA) (Group 1), MTA protected with cyanoacrylate (Group 2), MTA protected with glass ionomer (GI) cement (Group 3), and castor oil bean (COB) cement (Group 4). After root perforations were closed, the root canals were prepared biomechanically and teeth were sectioned longitudinally. Microleakage and microhardness of sealed perforations were assessed; microleakage data were submitted to analysis of variance (ANOVA) testing, while microhardness data were submitted to Dunnet and Tukey tests (p < 0.05). Group 4 reported the lowest amount of microleakage (0.65 mm), followed by Group 3 (1.02 mm), Group 1 (1.14 mm), and Group 2 (1.30 mm); however, no difference was detected among the groups. Groups 1-3 demonstrated significantly higher microhardness values compared to COB. It was concluded that the chemical and mechanical agents used during root canal preparation did not affect the sealing procedures. Administering surface protection to MTA did not improve microhardness or sealing.

Objective: To analyze the prevalence, severity and functional interference of movement disorders (MD) secondary to chronic use of cyclosporine A (CsA)., Method: We conducted a cross-sectional study of 60 patients (58.3% male) with mean age 23.1 (3-75) years, followed at the Bone Marrow Transplantation Service of the Hospital de Clínicas of the Federal University of Paraná, Brazil, taking CsA for at least six months. Our protocol included clinical data, assessment of functional interference of symptoms and neurological examination including observation and grading of MD., Results: Eight (13.3%) subjects reported the presence of tremor at the moment of interview and 29 (48.3%) recalled this symptom at some point during treatment. Neurological examination identified 14 (23.3%) subjects with MD: upper limb symmetric action tremor in 13 (21.6%) and parkinsonism (rigidity and bradykinesia) in 1 (1.7%). No other MD was detected. The mean scores indicated mild clinical signs in all cases. Symptoms were considered subjectively mild with no functional interference., Conclusion: Almost one quarter of patients using CsA chronically presented MD, almost always mild and transitory action tremor, with minimal interference on daily living activities, not requiring any form of intervention in the majority of cases.

Background: Blepharospasm (BS) is a form of central focal dystonia recently associated with psychiatric disorders, particularly obsessive and compulsive symptoms. Hemifacial spasm (HFS) represents a focal myoclonus with peripheral origin in the facial nerve., Objective: To determine the frequency of obsessive and compulsive symptoms in patients with BS in comparison with patients with HFS., Methods: 30 patients from each group (BS and HFS) followed by the botulinum toxin clinic at the HC-UFPR were evaluated using a structured interview based on the DSM-IV criteria and the Yale-Brown scale. Results were compared by the mean two-tailed t test., Results: We found obsessive or compulsive symptoms in 20 (66.6%) patients with BE and 21 (70%) with HFS. Yale-Brown scale scores for each group were higher among BS patients; however, diferences were not statisticaly significant., Conclusion: Our study did not show a significant diference in the comparison of the prevalence of obsessive and compulsive symptoms among patients with BS and HFS.
