Abstract Population pharmacokinetic (PK) modeling serves as the cornerstone for understanding drug behavior within a specific population. It integrates subject covariates to elucidate the variability in PK parameters, thus enhancing predictive accuracy. However, covariate modeling within this framework can be intricate and time‐consuming due to the often obscure structural relationship between covariates and PK parameters. Previous attempts, such as deep compartment modeling (DCM), aimed to streamline this process using machine learning techniques. Nonetheless, DCM fell short in assessing residual errors and interindividual variability (IIV), potentially leading to model misspecification and overfitting. Furthermore, DCM lacked the ability to quantify model uncertainty. To address these limitations, we introduce hierarchical deep compartment modeling (HDCM) as an advancement of DCM. HDCM harnesses machine learning to discern the interplay between covariates and PK parameters while simultaneously evaluating diverse levels of random effects and quantifying uncertainty through Bayesian inference. This tutorial provides a comprehensive application of the HDCM workflow using open‐source Julia tools.

Abstract: Aims We assessed the beta diversity of macroinvertebrates associated with aquatic macrophytes in a dam-regulated river floodplain. We tested two hypotheses: (1) macroinvertebrates associated with aquatic macrophytes have higher beta diversity (higher turnover and nestedness components) in the downstream areas closer to the São Francisco River compared to upstream sites; (2) closer lakes share a higher similarity of aquatic macroinvertebrates, disregarding their position relative to the dam. Methods We sampled four lakes, including two upstream (L1- natural and L2- regulated by a Small Hydropower Dam - SHD) and two downstream (L3, L4, unregulated) by the SHD. Results We corroborated our first hypothesis because we found a higher turnover of macroinvertebrates associated with aquatic macrophytes close to the São Francisco River (L4 downstream), where higher non-native mollusk species richness occurs, while we found higher nestedness upstream and downstream of the SHD (L1, L2, L3). We corroborated our second hypothesis, as closer lakes are more similar than distant lakes. Finally, these differences were consistent between lakes for emergent, floating leaf, and submerged aquatic macrophytes. Conclusions Aquatic macrophytes are essential habitats for macroinvertebrates in the Pandeiros River basin, with higher beta diversity in the lake closer to the São Francisco River.

BackgroundSexual and gender minority (SGM) young people are disproportionately affected by HIV in the United States, and substance use is a major driver of new infections. People who use web-based venues to meet sex partners are more likely to report substance use, sexual risk behaviors, and sexually transmitted infections. To our knowledge, no machine learning (ML) interventions have been developed that use web-based and digital technologies to inform and personalize HIV and substance use prevention efforts for SGM young people. ObjectiveThis study aims to test the acceptability, appropriateness, and feasibility of the uTECH intervention, a SMS text messaging intervention using an ML algorithm to promote HIV prevention and substance use harm reduction among SGM people aged 18 to 29 years who have sex with men. This intervention will be compared to the Young Men’s Health Project (YMHP) alone, an existing Centers for Disease Control and Prevention best evidence intervention for young SGM people, which consists of 4 motivational interviewing–based counseling sessions. The YMHP condition will receive YMHP sessions and will be compared to the uTECH+YMHP condition, which includes YMHP sessions as well as uTECH SMS text messages. MethodsIn a study funded by the National Institutes of Health, we will recruit and enroll SGM participants (aged 18-29 years) in the United States (N=330) to participate in a 12-month, 2-arm randomized comparison trial. All participants will receive 4 counseling sessions conducted over Zoom (Zoom Video Communications, Inc) with a master’s-level social worker. Participants in the uTECH+YMHP condition will receive curated SMS text messages informed by an ML algorithm that seek to promote HIV and substance use risk reduction strategies as well as undergoing YMHP counseling. We hypothesize that the uTECH+YMHP intervention will be considered acceptable, appropriate, and feasible to most participants. We also hypothesize that participants in the combined condition will experience enhanced and more durable reductions in substance use and sexual risk behaviors compared to participants receiving YMHP alone. Abstract We tested interactive effects of warming (+2°C) and browning on periphyton accrual and pigment composition when grown on a synthetic substrate (plastic strips) in the euphotic zone of 16 experimental ponds. We found that increased colored dissolved organic matter (cDOM) and associated nutrients alone, or in combination with warming, resulted in a substantially enhanced biomass accrual of periphyton, and a comparatively smaller increase in phytoplankton. This illustrates that periphyton is capable of using nutrients associated with cDOM, and by this may affect nutrient availability for phytoplankton. However, warming weakened the positive impact of browning on periphyton accrual, possibly by thermal compensation inferred from altered pigment composition, and/or changes in community composition. Our results illustrate multiple impacts of climate change on algal growth, which could have implications for productivity and consumer resource use, especially in shallow areas in northern lakes.

ABSTRACT Environmental degradation from human pressures includes the conversion of native vegetation cover into pastures and cropland, as well as riparian deforestation, leading to river siltation, biotic homogenization, and loss of ecosystem services. The objective of our study was to evaluate water quality and benthic macroinvertebrate assemblage structure in response to changes in land use at local and buffer spatial extents. We assumed that human disturbances negatively affect water quality and macroinvertebrate assemblage condition. Greater human influence was observed at the local extent (Local Disturbance Index – LDI) than at the buffer (Buffer Disturbance Index – BDI) extent. Likewise, biological metric responses were stronger relative to the LDI than to the BDI or to the Integrated Disturbance Index (IDI). These results support establishing a biomonitoring program for assessing water body quality in the Doce River basin to facilitate conserving aquatic biodiversity and ecosystem services in the upper Graipu River.

OBJECTIVES:. ICU patients have an increased risk of joint stiffness because of their critical illness and reduced mobility. There is a paucity of evidence evaluating the efficacy of passive movements (PMs). We investigated whether PMs prevent or reduce joint stiffness in ICU patients. DESIGN:. A randomized, controlled, within-participant, assessor-blinded study. SETTING:. A 48-bed tertiary care adult ICU. PATIENTS:. Intubated patients who were expected to be invasively mechanically ventilated for greater than 48 hours with an ICU length of stay greater than or equal to 5 days, and unable to voluntarily move their limbs through full range of motion (ROM). INTERVENTIONS:. The ankle and elbow on one side of each participant’s body received PMs (10 min each joint, morning and afternoon, 5 d/wk). The other side acted as the control. The PMs intervention continued for as long as clinically indicated to a maximum of 4 weeks. MEASUREMENTS:. The primary outcome was ankle dorsiflexion ROM at cessation of PMs. Plantarflexion, elbow flexion and extension ROM, and participant-reported joint pain and stiffness (verbal analog scale [VAS]) were also measured. Outcomes were recorded at baseline and cessation of PMs. For participants whose PMs intervention ceased early due to recovery, additional post-early-cessation of PMs review measurements were undertaken as near as possible to 4 weeks. MAIN RESULTS:. We analyzed data from 25 participants with a median (interquartile range) ICU stay of 15.6 days (11.3–25.4). The mean (95% CI) between-side difference for dorsiflexion ROM (with knee extension) at cessation of PMs was 0.4 degrees (–4.4 to 5.2; p = 0.882), favoring the intervention side, indicating there was not a clinically meaningful effect of 5 degrees. No statistically significant differences were found between the intervention and control sides for any ROM or VAS data. CONCLUSIONS:. PMs, as provided to this sample of medium to long-stay ICU patients, did not prevent or reduce joint stiffness.

Increasingly, scientists and non-scientists, especially employees of government agencies, tend to use weak or equivocal language when making statements related to science policy and governmental regulation. We use recent publications to provide examples of vague language versus examples of strong language when authors write about regulating anthropogenic pressures on natural resources. Lifeless language is common in agency reports, policy documents, and even scientific papers published by academics. Such language limits success in regulating anthropogenic pressures on natural resources. This challenge must be recognized and countered as a driver of the condition of water and associated resources. We also list sources of vague wording, provide global examples of how ambiguous language and political influences have contributed to water resource degradation, discuss the recent history of science censorship, and offer possible solutions for more direct scientific discourse. We found that: (1) equivocal language was especially common in concluding statements and not only by government employees; (2) authors discussed confusing language concerns in an agency publication; and (3) agency employees sometimes used active, strong language. Key drivers of weak language include: (1) holding on to old paradigms and resisting new knowledge; (2) scientific uncertainty; (3) institutional manuscript review policies; (4) employment and funding insecurity; and (5) avoiding the appearance of advocacy. Examples associated with euphemistic language included climate change, flow and physical habitat alteration, dams, agriculture, mining, forestry, and fisheries, as well as resistance towards monitoring, assessing, and reporting ecological conditions. Suggestions for mitigating equivocal language involve employment protections and greater focus on scientific ethics. We conclude that natural resource scientists should resist calls to employ imprecise language. Instead, they should be strong advocates for prescriptive and protective natural resource actions—based on their science—to halt and reverse the systemic degradation of those resources.

Tropical and sub-tropical streams harbor great biodiversity and are responsible for crucial ecosystem services. Intending to contribute to the understanding of how anthropogenic stressors and pressures alter functional or taxonomic diversity in biotic assemblages or processes in these systems, Water Biology and Security published eight papers. The research originated from Cameroon (Central Africa), southeastern Brazil, eastern Amazonia, and eastern China, representing eight river basins: Huai, Sui and Yishusi from east Asia; São Francisco, Doce, Jequitinhonha, and Amazon from South America; and Congo from Africa. The last two are the most biodiverse on the planet. The studied biomes included tropical rainforest and savanna, neotropical savanna and rainforest, neotropical savanna mountains, and subtropical monsoon forest. The response variables included fish assemblages (3 papers), entire macroinvertebrate assemblages or sub-groups (4 papers), and stream physical habitat structure (1 paper).

Motivation: Freshwater ecosystems have been heavily impacted by land‐use changes, but data syntheses on these impacts are still limited. Here, we compiled a global database encompassing 241 studies with species abundance data (from multiple biological groups and geographic locations) across sites with different land‐use categories. This compilation will be useful for addressing questions regarding land‐use change and its impact on freshwater biodiversity. Main Types of Variables Contained: The database includes metadata of each study, sites location, sample methods, sample time, land‐use category and abundance of each taxon. Spatial Location and Grain: The database contains data from across the globe, with 85% of the sites having well‐defined geographical coordinates. Major Taxa and Level of Measurement: The database covers all major freshwater biological groups including algae, macrophytes, zooplankton, macroinvertebrates, fish and amphibians.
