Background: Multidrug‐resistant (MDR) bacteria have significantly affected the management and treatment of wound infections globally. Data on the prevalence of MDR bacterial profiles that cause wound infections in Ethiopia are scarce. Therefore, this study aimed to determine MDR as well as extended‐spectrum beta‐lactamase production profiles of Gram‐negative rods that are difficult to treat with conventional antibiotics and that cause wound infections. Objective: The aim of the study was to determine the prevalence of extended‐spectrum beta‐lactamase producer Gram‐negative rods and associated factors among patients with wound infection at University of Gondar Comprehensive Specialized Hospital, northwest Ethiopia. Materials and Methods: This hospital‐based cross‐sectional study was conducted at University of Gondar Comprehensive Specialized Hospital between May and July 2022. Convenience sampling was used to recruit 228 participants. Swabs from different wound types were inoculated onto the MacConkey agar and blood agar plates and incubated overnight at 37°C for 24 h. Biochemical tests were performed on isolated colonies for the identification of bacterial species based on their biochemical reaction. Antimicrobial susceptibility tests were performed using the disk diffusion technique as per the standard Kirby–Bauer method by using Muller–Hinton agar, and the zone of inhibition was interpreted as resistant, intermediate, and sensitive as per the recommendation of Clinical Laboratory Standard Institute. Isolates were tested against ceftriaxone, cefotaxime, and ceftazidime for extended‐spectrum beta‐lactamase screening using the Kirby–Bauer disk diffusion method, and combined disk tests were applied for phenotypic confirmatory test of extended‐spectrum beta‐lactamase producing isolates. Result: Of 228 study participants, 162 (71.1%) were culture‐positive. Among culture‐positive patients, 165 Gram‐negative bacteria were recovered. The most common Gram‐negative isolates were Pseudomonas aeruginosa (47; 28.5%), followed by Escherichia coli (43; 26.1%) and Klebsiella pneumoniae (24; 14.5%). The susceptibility rates of the isolate for imipenem and tobramycin were 97.0% and 73.2%, respectively, and the overall multidrug resistance rate was 80.5%. Extended‐spectrum beta‐lactamase producer bacteria were also isolated. Besides, all (100%) of extended‐spectrum beta‐lactamase producer bacteria were MDR. Living in rural areas (AOR 5.8; 95% CI [2.01–16.7]), hospital admission (AOR 3.95; 95% CI [1.13–13.83]), antibiotic use (AOR 2.83; 95% CI [1.03–7.72]), and comorbidity (AOR 0.25; 95% CI [0.07–0.59]) were significantly associated with wound infection. Conclusions and Recommendations: There was a high prevalence of Gram‐negative bacterial isolates in this study. Pseudomonas aeruginosa (28.5%) was the predominant isolate. In addition, high rates of multidrug resistance were observed. The high level of multidrug resistance in this study implies that definitive therapy should be based on culture and susceptibility analysis to promote the rational use of antibiotics and to reduce the emergence of bacterial resistance to antimicrobials. Fibrinolytic agents are essential in treating thrombosis, playing a critical role in improving survival rates in cardiovascular diseases. Microbial fibrinolytic proteases have emerged as promising alternatives due to their affordability, specificity, lower toxicity, and reduced side effects. Consequently, the search for microorganisms capable of producing these enzymes has gained significant economic importance in the pharmaceutical industry. This study reports and characterizes a novel fibrinolytic enzyme produced by Penicillium citrinum CFAM 521, a strain isolated from the Amazon region. The enzyme was purified using a polyethylene glycol (PEG)–phosphate salt aqueous two‐phase system (ATPS). The effects of PEG molecular weight, PEG concentration, and phosphate concentration on the protease partition coefficient (K) were evaluated through a 22 full factorial design. The enzyme exhibited both fibrinolytic and fibrinogenolytic activities. After partitioning in a two‐phase system with 10% (w/w) PEG and 15% (w/w) sodium phosphate, the fibrinolytic proteases were predominantly retained in the salt‐rich bottom phase (K = 0.33). The enzyme has a molecular weight of 34 kDa, with optimal pH and temperature at 9°C and 37°C, respectively. Inhibitory analysis confirmed that it is a serine protease, and its activity was enhanced by the addition of Mn2+. Notably, the enzyme exhibited no hemolytic activity. Therefore, P. citrinum CFAM 521 represents a novel source of fibrinolytic enzymes, highlighting its potential as an alternative for the development of thrombolytic agents. [ABSTRACT FROM AUTHOR]

Aflatoxins, primary foodborne mycotoxins, come from Aspergillus flavus and Aspergillus parasiticus fungi. They pose significant health risks to humans and animals, creating a major challenge in the dairy sector. The objective of this study is to evaluate the knowledge, attitudes, and practices (KAP) of dairy farmers regarding aflatoxin contamination in milk and feeds. Conducted as a cross‐sectional study in Bahir Dar city between November 2019 and February 2020, this investigation randomly selected 106 dairy farms for data collection. Face‐to‐face interviews, facilitated by a semistructured questionnaire, were employed. Findings indicate that 59.4% of respondents displayed good knowledge, while a substantial 94.3% exhibited a favorable attitude. Intriguingly, only 1.9% implemented good practices. Notably, the educational background of dairy farmers emerged as a significant factor influencing their KAP (p < 0.05). Conversely, various sociodemographic factors did not yield a significant impact on the KAP of dairy farmers. Despite a robust knowledge base and favorable attitudes towards aflatoxin among dairy farmers, the study highlights a significant gap in the implementation of recommended practices. This finding emphasizes the necessity for increased efforts to cultivate and reinforce good practices. Bluetongue (BT) is an arthropod‐borne viral disease that primarily affects ruminants in tropical and temperate regions. In the present study, a cross‐sectional survey was conducted to define the seroprevalence of Bluetongue virus and to identify the possible risk factors correlated with BTV seropositivity among cattle, sheep, and goats during the period 2015‐2016 in Gadarif State. A total of 420 cattle, 877 sheep, and 641 goat serum samples were collected randomly from 12 localities. Information about age, sex, breed, area ecology, and location was obtained for each sample. Bluetongue seroprevalence was estimated using competitive enzyme‐linked immunosorbent assay (cELISA). The overall seroprevalence of BTV was 92.9% (390/420), 76.4% (670/877), and 85.3% (547/641) among cattle, sheep, and goats, respectively. Multivariate analysis followed univariate analysis showed that there was a significant difference (p < 0.05) between location, area ecology and age groups of cattle, sheep, and goats, and seropositivity to BTV. In addition, a significant association (p < 0.05) was observed between sex and seropositivity to BTV in sheep. In conclusion, BTV antibodies are highly prevalent in Gadarif State and susceptible livestock are at risk of exposition with BTV. Infectious bronchitis virus (IBV) is a significant threat to poultry worldwide, but its status in Ethiopia remains understudied. Thus, this study aimed to detect the virus and associated risk factors in South West Ethiopia. Ninety oropharyngeal swab samples were purposively collected from symptomatic chickens located in Jimma town, Seqa Chekorsa, and Tiro Afeta woredas of the Jimma zone between November 2021 and April 2022 to detect IBV virus by using RT‐PCR. A side‐by‐side questionnaire was administered to assess risk factors. Total RNA was extracted, reverse transcription polymerase chain reaction (RT‐PCR) was conducted, and products were visualized under UV light. The overall proportion of IBV was 16.6% (15/90). No statistical association was observed between any of the animal risk factors and the detection of the virus (P = 0.57, 0.586, and 1). However, the proportion of birds infected by the virus was higher in males, exotic breeds, and adults compared to females, local breeds, and young birds. Similarly, none of the management risk factors had a significantly different effect on virus detection (P = 0.25, 0.09, 0.088, and 0.726). However, improper carcass disposal (OR = 0.43, 95% CI: 0.13–1.4), lack of veterinary services (OR = 2.7, 95% CI: 0.8–8.3), and the presence of wild birds/rodents (OR = 4.4, 95% CI: 0.88‐22.3) were associated with increased IBV risk but not cleaning of feeders/drinkers (OR = 1.1, 95% CI: 0.2–4.8). The fungus Phyllosticta citricarpa is a quarantine phytopathogen responsible for causing citrus black spot (CBS) disease. To export fruits to CBS‐free countries, they must undergo a sanitation process to ensure disease control. In this study, neem essential oil (NEO) was tested against P. citricarpa for the first time as an alternative sanitizer. In vitro experiments were conducted to determine the inhibition concentration of NEO for P. citricarpa, and the mode of action of the essential oil was evaluated. In vivo assays were performed to simulate the sanitization process used in packinghouses. NEO was characterized by GC‐MS/MS. The results revealed that NEO at 100 μL·mL−1 exhibited a similar inhibitory effect as copper oxychloride, suppressing 89.68 ± 1.14% of fungal mycelium growth. Fluorescence microscopy experiments demonstrated that NEO functions by disrupting the cytoplasmic membrane of fungal hyphae, leading to their death within 30 minutes of contact with NEO. GC‐MS/MS characterization revealed a high presence of phenolic compounds, which serve as the primary antifungal agents responsible for the action against fungal hyphae. In vivo assays showed that NEO at 100 μL·mL−1 also reduced microorganisms (CFU mL−1) by 93.00 ± 3.88% compared to the negative control. Overall, the results demonstrate that NEO can effectively serve as an alternative sanitizer against P. citricarpa in citrus packinghouses. Our findings allow future studies to explore the use of NEO for sanitizing other fruits and combating different phytopathogens to broaden its potential application in fruit sanitation for export. Boza, a cereal‐based beverage popular in southeast Europe, is fortified with probiotics and is believed to positively impact the composition of the gut microflora. This investigation focused on fermented cereal‐based beverage boza to identify strains of probiotic Lactobacillus spp. capable of inhibiting carbohydrate‐hydrolysing enzymes α‐glucosidase (AG) and α‐amylase (AA). The isolated bacterial strains underwent a comprehensive assessment, including biochemical, molecular, and probiotic trait analyses such as tolerance survivability, adhesion, safety, and health‐promoting attributes. We evaluated the inhibitory potential of the supernatant, cell lysate, and intact cells of Lactobacillus spp. Molecular analysis has revealed that isolates RAMULAB30 and RAMULAB29 exhibit a significant genetic similarity (>97%) to Lacticaseibacillus paracasei and Limosilactobacillus fermentum, respectively. These findings are documented in the NCBI database. They exhibited significant resistance to gastrointestinal and intestinal fluids, also indicating their potential for adhesion. Additionally, the isolates showed a significant antibacterial activity, particularly against Micrococcus luteus. They showed resistance to vancomycin and methicillin antibiotics but were more susceptible to streptomycin and ampicillin. Furthermore, the strains demonstrated antioxidant properties. To ensure their safety, a haemolytic assay was conducted despite their general recognition as safe (GRAS) status. The study primarily aimed to evaluate the inhibitory effects of the extract on enzymes AG and AA. Bacterial isolates demonstrated a significant inhibitory activity against both enzyme AG (32%–67% inhibition) and enzyme AA (18%–46% inhibition) in different forms, including supernatant (CS), lysed extract (CE), and intact cell (IC). These findings underscore the potential of bacterial isolates to inhibit the enzyme activity effectively. Furthermore, the L. fermentum RAMULAB29 and L. paracasei RAMULAB30 strains exhibit remarkable antidiabetic potential. Introduction: Medicinal plants, rich in phytochemicals like phenolic acids, flavonoids, and tannins, offer potential benefits in enhancing productivity, quality, and animal health. Amla fruit (Phyllanthus emblica) is one such plant with promising attributes. This study aimed to investigate the impact of fresh Amla fruit (FAF) supplementation on ruminal microbial composition and its correlation with rumen fermentation in lactating dairy cows. Methods: The study employed a repeated crossover design involving eight ruminally cannulated mid-lactation Holstein dairy cows. Animals received varying levels of fresh Amla fruit supplementation (0, 200, 400, and 600 g/d). Results: When 400 g/d of FAF was added to the diet, there was a significant increase in the relative abundance of Firmicutes (p = 0.02). However, at 200 g/d, the relative abundance of ruminal Bacteroidota was higher than the 0 and 400 g/d FAF supplementation (p < 0.01). LEfSe analysis identified distinct taxa, such as Clostridia vadinBB60 in the 200 g/d group, Oscillospiraceae in the 400 g/d group, and Elusimicrobium in the 600 g/d group. Notably, the random forest species abundance statistics identified Oscillospiraceae V9D2013 as a biomarker related to milk yield. Oscillospiraceae, Bacilli RF39, norank_f Prevotellaceae, and Bifidobacterium were positively correlated with ruminal total VFA and molar proportion of propionate, while Rikenellaceae RC9 gut group and Clostridia vadinBB60 were negatively correlated. Discussion: FAF supplementation affects the abundance of beneficial microbes in a dose-dependent manner, which can improve milk yield, efficiency, rumen health, desirable fatty acids, and animal health. Mushrooms are a valuable source of food and medicine that have been used for centuries in various cultures. They contain a variety of phytochemicals, such as terpenoids and polysaccharides, that exhibit diverse biological activities, such as antioxidant, anti‐inflammatory, anticancer, antimicrobial, immunomodulatory, and antidiabetic effects. However, mushroom's phytochemical composition and bioactivity vary depending on their species, cultivation conditions, processing methods, and extraction techniques. Therefore, using reliable analytical methods and standardized protocols is important for systematically evaluating the quality and quantity of mushroom phytochemicals and their therapeutic potential. This review provides a bibliometric analysis of the recent literature on biological activities, highlights trends in the field, and highlights the countries and journals with the highest contribution. Livestock producers need new technologies to maintain the optimal health and well-being of their animals while minimizing the risks of propagating and disseminating pathogenic and antimicrobial-resistant bacteria to humans or other animals. Where possible, these interventions should contribute to the efficiency and profitability of animal production to avoid passing costs on to consumers. In this study, we examined the potential of nitroethane, 3-nitro-1-propionate, ethyl nitroacetate, taurine and L-cysteinesulfinic acid to modulate rumen methane production, a digestive inefficiency that results in the loss of up to 12% of the host's dietary energy intake and a major contributor of methane as a greenhouse gas to the atmosphere. The potential for these compounds to inhibit the foodborne pathogens, Escherichia coli O157:H7 and Salmonella Typhimurium DT104, was also tested. The results from the present study revealed that anaerobically grown O157:H7 and DT104 treated with the methanogenic inhibitor, ethyl nitroacetate, at concentrations of 3 and 9 mM had decreased (p < 0.05) mean specific growth rates of O157:H7 (by 22 to 36%) and of DT104 (by 16 to 26%) when compared to controls (0.823 and 0.886 h−1, respectively). The growth rates of O157:H7 and DT104 were decreased (p < 0.05) from controls by 31 to 73% and by 41 to 78% by α-lipoic acid, which we also found to inhibit in vitro rumen methanogenesis up to 66% (p < 0.05). Ethyl nitroacetate was mainly bacteriostatic, whereas 9 mM α-lipoic acid decreased (p < 0.05) maximal optical densities (measured at 600 nm) of O157:H7 and DT104 by 25 and 42% compared to controls (0.448 and 0.451, respectively).
