The evolution of herbicide resistance in weeds has emerged as one of the most serious threats to sustainable food production systems, which necessitates the evaluation of herbicides to determine their efficacy. The first herbicide resistance case in the Iberian Peninsula was reported about 50 years ago, wherein Panicum dichotomiflorum was found to be resistant (R) to atrazine in Spanish maize fields. Since then, herbicide resistance has evolved in 33 weed species, representing a total of 77 single-herbicide-resistance cases in this geographic area: 66 in Spain and 11 in Portugal. Changes in agricultural practices, namely the adoption of non-tillage systems and the increased use of herbicides, led to the selection of weed biotypes resistant to a wide range of herbicides. Nowadays the most important crops in Spain and Portugal (maize, winter cereals, rice, citrus, fruits, and olive orchards) are affected, with biotypes resistant to several mechanisms of action (MoAs), namely: ALS inhibitors (20 species), ACCase inhibitors (8 species), PS II inhibitors (18 species), and synthetic auxin herbicides (3 species). More recently, the fast increase in cases of resistance to the EPSPS-inhibiting herbicide glyphosate has been remarkable, with 11 species already having evolved resistance in the last 10 years in the Iberian Peninsula. The diversity of resistance mechanisms, both target-site and non-target-site, are responsible for the resistance to different MoAs, involving point mutations in the target site and enhanced rates of herbicide detoxification, respectively. More serious are the 13 cases reported with multiple-herbicide resistance, with three cases of resistance to three–four MoAs, and one case of resistance to five MoAs. Future research perspectives should further study the relationship between management strategies and the occurrence of TSR and NTSR resistance, to improve their design, develop monitoring and diagnostic tools for herbicide resistance, and deepen the study of NTSR resistance

In Portugal, glyphosate is widely used for weed control on perennial crops and non-cropping areas. It is considered an herbicide of high risk of resistance worldwide. Glyphosate resistance in Portugal was recently confirmed inConyza bonariensispopulations from intensive olive groves. This paper presents the results of a survey conducted for detecting glyphosate resistance in populations ofConyza canadensis. Whole plant dose-response bioassays were conducted to confirm resistance. ED50values ranged from 32.72 g.ha-1in susceptible population to 77.91 g.ha-1for the most resistant population. The resistance factor (RF = ED50R/ED50S) was around 2 for all populations. These values reflect a trend that all populations studied were resistant to glyphosate and laboratory tests to determine the mechanism of resistance are in course., O glifosato é o herbicida mais importante a nível nacional, utilizado principalmente para controlar infestantes em culturas perenes e em zonas não cultivadas. Neste trabalho apresentam-se os resultados da prospecção realizada para despistagem de resistência ao glifosato em populações deConyza canadensis. Para confirmação da resistência, realizaram-se ensaios de dose-resposta com planta inteira. Estudaram-se as condições mais adequadas à germinação de sementes de Alisma plantagoaquatica, susceptíveis (S) e resistentes (R) ao herbicida bensulfurão-metilo, de diferente origem. Analisou-se a taxa de germinação de 11 amostras de sementes provenientes de arrozais do Sado (2), do Sorraia (3) e do Baixo Mondego (6) em dois regimes de temperatura (15 ºC e 15/30 ºC); os estudos foram realizados com amostras de dois anos e incluíram uma amostra com três anos proveniente de arrozal do Sorraia. A regressão não linear pelo modelo log-logístico (SSlogis) permitiu caracterizar as populações em estudo e compará-los quanto à idade, origem geográfica e resistência. Verificou-se que as sementes com três anos apresentaram menor capacidade germinativa (CG) do que as sementes com dois anos. Não foi possível ajustar o modelo à população S do Sado nem a duas populações do Baixo Mondego, pela falta de maturidade das sementes. Verificou-se que nas populações S a germinação das sementes era independente do regime de temperatura, enquanto nas populações R, a germinação era favorecida por determinado regime de temperatura, consoante a sua origem geográfica. Assim a germinação das sementes da população do Baixo Mondego (região Centro) foi favorecida pelo regime de alternância (15/30 ºC), enquanto a populações das bacias hidrográficas dos rios Sorraia e Sado (região Centro-Sul) foram favorecidas pelo regime de temperatura constante (15 ºC). Considerando que, à temperatura de 15 ºC, as sementes das populações R dos arrozais do Centro-Sul do País apresentaram maior CG do que as da região Centro, esta característica pode ser aproveitada para implementar, naquela região, medidas de gestão da resistência durante a fase inicial do ciclo cultural do arroz quando as temperaturas são mais baixas., Revista de Ciências Agrárias, vol. 31 n.º 1 (2008)

El desarrollo de modelos predictivos de la emergencia de malas hierbas puede facilitar la toma de decisiones de control óptimas por parte de agricultores y técnicos. Se ha especulado con la posibilidad de desarrollar modelos globales que permitan su utilización en diferentes localidades. Sin embargo existe poca información a este respecto. El objetivo de este trabajo fue validar y comparar dos modelos termales (Weibull y Logístico) de la emergencia de A. teophrastri con datos independientes obtenidos en localidades de EEUU, Portugal y España. Los resultados muestran que el modelo Weibull fue el más apropiado para predecir la emergencia de A. teophrastri de manera global. Abutilon theophrasti causes -severe yield losses in maize and soybean (70 % - 72%). A. theophrasti management is crucial for optimum crop production. Prevention is a pre-requisite to reduce weed pressure and investigation into weed bioecology is needed. This study characterized the morphological and genetic variation of eleven A. theophrasti accessions representative of a climatic gradient distribution range from north (47° N) to southeast Europe(39 ° N) and north America (43ºN). To evaluate interpopulation variability, seed were characterized with morphological parameters (length; width; thickness; weight and volume of 100 seeds) and biomolecular markers: trnL-trnF; trnH-trnK; trnT-psbC and matK. Multivariate and univariate analyses were performed to estimate genetic diversity within and among populations. Populations from north and southern Europe were separated in two groups according to seed morphology. However it was not possible to associate seed morphological traits to genetic variability. All gene sequences (cpDNA genome) presented the same nucleotide sequence.

The insects present in the blackberry and blueberry crops are still almost unknown in Portugal. Thrips and their natural enemies were surveyed in these crops, during the flowering period, in Odemira region, as well as in the weeds inside and around the crops areas. This allows an evaluation of the entomological interaction cropweeds, for a future management of weeds taking also into account crops protection against pests. The preliminary results obtained are presented. Thrips flavus and T. tabaci were the most abundant thrips in the crops. Weeds where Thrips sp. adults were more abundant were Arctotheca calendula, Crepis capillaris, Senecio gallicus, Geranium molle, Malva parviflora, Lavatera cretica and Lupinus luteus. In this last weed species, Aeolothrips spp. predators were abundant. The heteropterans Orius sp., predators of thrips, were detected in the blackberry crop, and also on Conyza bonariensis, Conyza sumatrensis, Sonchus tenerrimus and Echium plantagineum

Acarological surveys were carried out fortnightly from March to June 2011 on blackberries and weeds in a greenhouse at Fataca Experimental Farm in Odemira, in order to study species diversity and population interaction. This study focused on two blackberry cultivars, ‘Ouachita’ and ‘Karaka Black’. Sixteen mite species were identified on blackberries. The redberry mite Acalitus essigi (Hassan) was the most noxious mite in both cultivars, causing severe damage. However, the late maturing Ouachita cultivar was the most attacked by this eriophyid mite. The most common predacious mites were the stigmaeid Agistemus longisetus Gonzalez and the phytoseiids Amblyseius stipulatus Athias-Henriot and Amblyseius californicus (McGregor). From the 36 weed species identified, 22 were host plants of mites with agricultural interest, 14 mite species being identified. Phytoseiids, especially A. stipulatus, observed on 13 weed species, indifferent tydeids, in particular Orthotydeus californicus (Banks), observed on 10 weed species, and the stigmaeid A. longisetus, observed on seven weed species, were the predominant mites. These mite species also occurred on blackberries. The most important host weeds of mites were Amaranthus deflexus L., Chenopodium murale L., Conyza bonariensis (L.) Cronq., Conyza canadensis (L.) and Solanum nigrum L., mainly associated to predators, A. stipulatus and A. longisetus. From acarological point of view, safeguarding the competition relatively to some vegetal species, ground cover vegetation can be an important component of conservation biological control in blackberry crop, because host weeds of mites are mostly repositories of predacious species

The repetitive use of bensulfuron-methyl in rice fields from Sorraia, Sado and Mondego river valleys had selected resistant (R) populations of Alisma plantago-aquatica in Portugal, identified as So306, So307, Sa88, Mo190; Mo260, respectively. The response of R-biotypes to bensulfuron-methyl at 26 days after seeding (DAS) and 42 DAS and the crossresistance either to ALS- inhibitors or to herbicides with other mode of action, was investigated in dose-response experiments. The results confirmed bensulfuron-methyl resistance in four biotypes, but not in Mo190, which had been also treated with bentazon every year. Younger plants (6 linear phyllodium, BBCH 116) were more susceptible to bensulfuron-merthyl than were older plants (6 expanded phyllodium, BBCH 126). R-biotypes presented cross-resistance to all ALS-inhibitors studied except for imazethapyr and metsulfuron-methyl. Resistance indeces (RI= ED50R /ED50S) ranged frome 3 to 313 for the three sulfonylurea herbicides. Herbicides with different mode of action including bentazon, MCPA, oxadiazon and propanil showed resistance indices of one, indicating that all herbicides tested had equal activity to both Rand S-biotypes. It was concluded that these herbicides could be effective for the control of resistant A. plantago-aquatica biotypes in rice paddy fields., A aplicação repetida de bensulfurão-metilo em arrozais dos vales dos rios Sorraia, Sado e Mondego seleccionaram populações resistentes (R) de Alisma plantago-aquatica (Mo260, Sa88, So306 e So307). Determinou-se o nível de resistência (NR= ED50R/ ED50S) ao bensulfurão-metilo em plantas com 26 dias (6 filódios lineares) e com 42 dias (6 filódios expandidos) e a resistência cruzada a 14 herbicidas. Confirmou-se que quatro populações eram resistentes e uma (Mo190) era susceptível. Esta população provinha de um campo tratado com bensulfurão-metilo e bentazona anualmente. Os resultados dos ensaios de doseresposta confirmam a maior susceptibilidade ao bensulfurão-metilo, no estado fenológico 116. Apresentaram resistência cruzada a todos os herbicidas inibidores da ALS excepto ao metsulfurão-metilo e ao imazetapir. Obtiveram-se valores de NR de 3 a 315 para azimsulfurão, cinosulfurão e etoxisulfurão. Bentazona, MCPA, oxadiazão e propanil apresentaram NR próximos da unidade, indicando que herbicidas com diferente modo de acção mantiveram a sua eficácia e podem constituir alternativa no combate a populações de A. plantago-aquatica resistentes ao bensulfurão-metilo.

Seed germination of bensulfuron-methyl resistant (R) and susceptible (S) Alisma plantagoaquatica biotypes collected in rice fields from Baixo Mondego (6 samples); Sorraia river valey (2 samples) and Sado river valey (2 samples) were compared at 15 ºC and 15/30 ºC. All seed samples were two years old and an extra sample three years old was also included to assess the effect of seed age on germination. Three year seeds reached higher germination rates than two year seeds at both temperature conditions. Cumulative germination characterized for each biotype using non linear regression (SSlogis) was different according to geographic origin, resistance trait and seed age. The model could not fit data from Sado S biotype and two Baixo Mondego R biotypes due to immature seed. Samples with the highest cumulative germination also have the highest germination rate. Seed germination of S populations was similar at both temperature conditions, whilst for R populations seed germination was favoured by different temperature depending on geographic origin: temperature of 15/30 ºC gave the best results for Mondego river valley (Centre region) and temperature of 15 ºC for Sorraia and Sado river valleys (Centre-South region of Portugal). The resistant trait affects germination of A. plantago-aquatica seeds and may affect resistant-weed management strategies implemented early in the growing season when temperatures are lower

Universidade de Lisboa O arroz é um dos cereais com maior área de cultivo e maior consumo no mundo e em Portugal. As infestantes são o principal problema fitossanitário desta cultura, devido à dificuldade na gestão de infestações, por aspetos relacionados com a sua biologia e com a ocorrência de resistência a herbicidas. Leptochloa fusca subsp. fascicularis (erva-fina ou leptocloa) é uma gramínea anual semiaquática, norte-americana, considerada uma infestante emergente do arroz em Portugal. Cyperus difformis (negrinha) é uma ciperácea anual, causando elevados prejuízos na maioria das regiões orizícolas mundiais e em Portugal. Há suspeitas de populações resistentes a herbicidas de ambas as espécies em arrozais portugueses. Este trabalho teve como primeiro objetivo eleger o tratamento mais eficaz de quebra de dormência das sementes de leptocloa, devido à grande dificuldade na obtenção de plantas para ensaios de resistência devido a fenómenos de dormência. Realizaram-se ensaios de germinação de sementes colhidas na campanha de 2020 e provenientes de várias populações de arroz do Baixo Mondego, Ribatejo e Alentejo. Foram testados 24 tratamentos. Um segundo objetivo focou-se na resistência de uma população de C. difformis, proveniente de um arrozal do Alentejo, com o objetivo de validar suspeitas de resistência às substâncias ativas bentazona (inibidor da fotossíntese no fotossistema II) e ao penoxsulame (inibidor da enzima acetolactato sintase), através de testes rápidos de despiste. Os ensaios de quebra de dormência em leptocloa evidenciaram que o tratamento misto de estratificação húmida a 3 ⁰C, na obscuridade, seguido de punção é o mais eficaz para as várias populações. Este resultado indica uma possível dormência combinada, fisiológica e física. Os ensaios na população de negrinha evidenciaram resistência às substâncias ativas estudadas, mas será aconselhável a validação em vaso com planta inteira em condições de campo. Universidade de Lisboa Esta dissertação consistiu na caracterização do género Echinochloa spp. em arrozais portugueses, quanto à identificação dos táxones presentes e à sua suscetibilidade a herbicidas da família dos arilpicolinatos. Utilizou-se material vegetal, correspondente a 24 populações de Echinochloa, colhido em arrozais nas bacias hidrográficas respetivas às três regiões orizícolas do país: rio Mondego, rios Tejo e Sorraia, e rio Sado, nas campanhas de 2014 e 2015. A identificação dos táxones foi realizada através da caracterização biométrica das espiguetas e confirmada in vivo. Realizou-se a avaliação da capacidade germinativa das populações através de ensaios de germinação. A avaliação da suscetibilidade de Echinochloa spp. à família dos arilpicolinatos foi realizada através de ensaios de dose-resposta com planta inteira realizados em estufa. Estimaram-se os valores de ED50 através de modelos lineares do tipo logístico. Foram identificados três táxones: Echinochloa crusgalli subs. hispidula (ECHHI), E. colona (ECHCO) e E. phyllopogon (ECHPH). Com o ensaio de germinação, selecionaram-se as populações com taxa de germinação superior a 85 % para o ensaio de dose-resposta. Os resultados do ensaio dose-resposta, evidenciaram suscetibilidades diferentes das populações de Echinochloa, nas três regiões orizícolas: maior suscetibilidade no Alentejo, resposta intermédia no Baixo Mondego e tolerância no Ribatejo. Colocou-se a hipótese destes resultados poderem estar relacionados com diferentes sensibilidades das espécies de Echinochloa ao herbicida ou ao historial dos campos de arroz. Não foi possível confirmar nenhuma destas hipóteses. A eventual manifestação de resistências cruzadas só poderá ser confirmada em futuros ensaios N/A

Mestrado em Engenharia Agronómica - Especialização em Proteção das plantas - Instituto Superior de Agronomia - UL Este estudo consistiu na prospeção de resistência a dois herbicidas em 12 populações de Echinochloa spp. de vários arrozais nas regiões dos vales dos rios Tejo e Sorraia ,em Outubro de 2014, e de levantamentos florísticos em arrozais das mesmas zonas. A identificação das espécies de Echinochloa de cada amostra foi feita através de uma análise biométrica da cariopse recorrendo ao software Smartgrain. Determinou-se o melhor prétratamento para quebra de dormência, necessária à obtenção de plântulas para os estudos posteriores. Em placa de Petri efetuou-se a prospeção de resistência, para a definição de uma população suscetível de referência e seleção de outras amostras consideradas como resistentes. Efetuaram-se também três ensaios em estufa: para confirmação da resistência ao penoxsulame, para deteção da resistência múltipla ao profoxidime e aferir se a resistência ao penoxsulame era metabólica. A análise biométrica das cariopses de cada população indicou a presença dos táxones E. crus-galli subsp. hispidula (ECHHI) e E. phyllopogon (ECHPH). Nos estudos de germinação, o tratamento com ácido sulfúrico foi o que induziu maior percentagem de germinação. No estudo de prospeção de resistência nas 12 populações, a suscetível de referência foi a ECHPH1 e as resistentes para os estudos com planta inteira em estufa foram as ECHPH5, ECHPH6, ECHHI1, ECHHI2 e ECHIHI4. Os ensaios com planta inteira em estufa evidenciam a existência de resistência ao penoxsulame, embora não tenha sido detetada resistência ao profoxidime, o que contraria a hipótese de resistência múltipla. O terceiro ensaio demonstrou que a resistência ao penoxsulame não era metabólica. Nos levantamentos florísticos as espécies com maior infestação foram Oryza sativa e E. phyllopogon

Mestrado em Engenharia Agronómica - Proteção de Plantas - Instituto Superior de Agronomia This dissertation reports the study of two resistance mechanisms in a glyphosate-resistant Conyza canadensis population (B15), comparatively with a susceptible population (B), from intensive olive orchards in Alentejo (south of Portugal). Initially, the presence of resistance was confirmed by a shikimate leaf disc assay. Then, the sequence of the gene encoding the enzyme 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) was determined and, lastly, were analysed the expression levels of the gene encoding this enzyme. The shikimate leaf disc assay showed a higher accumulation of shikimate in the population B, relatively to population B15, 24 hours after being exposed to glyphosate. The EPSPS-2 cDNA sequencing didn’t reveal differences in enzyme’s constitution between the two populations. The analysis of gene expression also revealed no differences in expression levels of this enzyme, 24 hours after glyphosate application. These results suggest that the studied population has no EPSPS enzyme mutation or overexpression as a mechanism of resistance to glyphosate

Mestrado em Engenharia Agronómica - Proteção de plantas - Instituto Superior de Agronomia The objective of this study was to assess efficacy of the glyphosate in three populations of Conyza canadensis, two suspected of resistance (B15-1 and B15-2) and a susceptible one (B), from intensive olive orchards in Alentejo, south of Portugal. There were performed: seedling-Petri plate assays, greenhouse dose-response assays and a shikimate accumulation assay. The seedling-petri plate assays showed that B population had significant higher germination (60-80%) than B15-1 (31.3%) and B15-2 (52.7%) populations. There was evidence of resistance in the B15-1 population and susceptibility of the B15-2 population. Mortality was the key parameter to distinguish between populations by the discriminating concentration: 0.3 mM glyphosate. The dose-response assays revealed a resistance level of 0.52 and ED50 of 59.2 g e.a. ha-1 (Bioassay I) and 87.2 g e.a. ha-1 (Bioassay II) for B15-2 population. The B population showed an ED50 of 113.3 g e.a. ha-1. The shikimate accumulation assay indicated susceptibility of both populations (B and B15-2), although the accumulation was higher in the B population (0.69 μg mg-1) than in the B15-2 population (0.49 μg mg-1), six days after application (720 g ha-1). The results showed evidence of resistance in the B15-1 population and confirmed the susceptibility of B and B15-2 populations.

Mestrado em Engenharia Agronómica - Protecção das Plantas - Instituto Superior de Agronomia The objective of this study was to determine the efficacy of this herbicide in two populations of Conyza bonariensis, one suspected of resistance (B8) and a susceptible one (C), from olive groves and citrus orchards in Alentejo, south of Portugal. Assays were performed on germination agar medium, glyphosate dose-response assays in petri dishes with different concentrations of glyphosate and a greenhouse (1) and laboratory (2) study were conducted (1) to confirm and characterize glyphosate resistance and (2) to determine whether resistant and susceptible populations have differential shikimate accumulation. Population B8 showed significant higher germination and emergence than population C but lower mortality rate. Dose–response studies revealed that the suspected resistant population was two- to three-fold less sensitive to glyphosate than the susceptible population. The dose-response flazasulfuron assay with whole plant in the rosette stage revealed the doses were completely effective on both populations. Shikimate accumulation was higher in resistant population at 72-h after treatment, for the recommended concentration of glyphosate. However, it was lower for concentrations of 360 g ha-1. These results pointed that population B8 is resistant to glyphosate and flazasulfuron is an option for the chemical control of resistant populations.

Mestrado em Produção Agrícola Tropical - Instituto Superior de Agronomia Nowadays with the problems in public and environmental health, associate with biotypes of species resistant to herbicides, occur the development of the concept of integrated weed management. This concept is based in different methods of weed management in the weed seed bank. The weed seed bank is a viable seed reservoir present in the top soil profile, capable to germinate with adequate conditions. In this study was evaluate the weed seed bank in a citrus orchard for determine the possible influence of four different methods. The methods were: establishment of sown cover crops, cover crop with natural flora, and application of herbicide in the inter-row and in the row. The soil samples were collected at a depth of 0-10 cm; the samples were placed in glasshouse. The weed emerged were observed and identified once a week. A higher diversity of species was assessed in the sown cover crop, but the natural cover crops showed a higher abundance. Piptaterum milliaceum showed a higher density of plants per m².------------------------------------Actualmente devido aos problemas de saúde pública e ambiental, associados a fenómenos de resistências das espécies infestantes aos herbicidas, desenvolveu-se o conceito de gestão integrada das infestantes. Este conceito engloba os diferentes métodos de gestão da flora infestante do banco de sementes do solo. O banco de sementes do solo é definido como o agregado de sementes não germinadas existentes na camada superficial do solo, potencialmente capazes de germinarem sempre que se verifiquem condições adequadas. Neste estudo foi avaliado o banco de sementes de solo num pomar de citrinos, para determinar as possíveis influências de quatros métodos de gestão da flora infestante. Os diferentes métodos consistiram no estabelecimento de um enrelvamento semeado, um enrelvamento formado pela vegetação natural, aplicação de herbicida na entrelinha e aplicação de herbicida na linha. Foram retiradas amostras de solo de cada um dos métodos de gestão, a uma profundidade de 0-10 cm, estas amostras foram colocadas em estufa de vidro. Avaliou-se as emergências das plantas com a periodicidade de 7 dias, tendo-se efectuado a contagem e identificação das espécies. A maior diversidade de espécies foi observada no enrelvamento semeado, mas na vegetação natural registou-se uma maior abundância. A espécie com maior densidade de plantas/m² foi Piptaterum miliaceum.
