Abstract

Phytotherapy is based on the use of plants to prevent or treat human and animal diseases. Recently, the use of essential oils and polyphenol-enriched extracts is also rapidly increasing in the aquaculture sector as a means of greater industrial and environmental sustainability. Previous studies assessed the antibacterial and antiparasitic effects of these bioactive compounds on fish. However, studies on the modulation of oxidative stress biomarkers are still scant to date. Thus, in this study, the modulation of antioxidant defense against oxidative stress exerted by fish diets supplemented with a basil supercritical extract (F1-BEO) was assessed in rainbow trout Oncorhynchus mykiss. The F1-BEO extracted with supercritical fluid extraction was added to the commercial feed flour (0.5, 1, 2, 3% w/w) and mixed with fish oil to obtain a suitable compound for pellet preparation. Fish were fed for 30 days. The levels of stress biomarkers such as superoxide dismutase, catalase, glutathione peroxidase, glutathione S-transferase, glutathione reductase, glyoxalase I, glyoxalase II, lactate dehydrogenase, glutathione and malondialdehyde showed a boost in the antioxidant pathway in fish fed with a 0.5% F1-BEO-supplemented diet. Higher F1-BEO supplementation led to a failure of activity of several enzymes and the depletion of glutathione levels. Malondialdehyde concentration suggests a sufficient oxidative stress defense against lipid peroxidation in all experimental groups, except for a 3% F1-BEO-supplemented diet (liver 168.87 ± 38.79 nmol/mg prot; kidney 146.86 ± 23.28 nmol/mg prot), compared to control (liver 127.76 ± 18.15 nmol/mg prot; kidney 98.68 ± 15.65 nmol/mg prot). Our results suggest supplementing F1-BEO in fish diets up to 0.5% to avoid potential oxidative pressure in farmed trout.

Abstract

Invasive species threaten native biodiversity, potentially leading to a decline in endemic aquatic species. Thus, understanding their physiological responses and evaluating ecosystem impacts is crucial. The study investigates adaptive responses to oxidative stress in the invasive Sinanodonta woodianain Lake Candia (northwest Italy). This approach represents a novel perspective in environmental monitoring, utilizing invasive species to gain insights into ecosystem health and potential ecological threats. The investigation delved into oxidative stress biomarkers, including superoxide dismutase, catalase, glutathione S-transferase, glyoxalase I, glyoxalase II, and lactate dehydrogenase, in mussel tissues (gills and digestive gland). Concurrently, environmental factors such as temperature, dissolved oxygen, pH, and conductivity were measured at four stations in Lake Candia during the summer. Environmental factors exhibited homogeneity across selected stations, with positive correlations observed only for temperature and conductivity with catalase and glyoxalase I. A distinct pattern of oxidative stress response was consistently observed in both mussel tissues throughout the sampling period across all monitoring stations, with more pronounced alterations observed in one specific station. This observation aligns with IBRv2 scores, indicating the station's highest impact. Gills exhibited a transient and severe oxidative stress response, while heightened antioxidant defenses in digestive glands suggested the adaptive ability of Chinese pond mussels to environmental disturbances. Proactive inclusion of invasive species in biomonitoring programs reduces reliance on native populations, enhancing strategies for controlling non-native mussels in protected areas like Lake Candia.

Abstract

High-mountain lakes are pristine ecosystems characterized by extreme environmental conditions. The atmospheric transport of pollutants from lowlands may add further stress to organisms inhabiting these environments. We investigated the environmental stress pressure on brook trout (Salvelinus fontinalis) from a high-mountain lake in the Cottian Alps (Piedmont, northwest Italy). To do this, males and females of brook trout were sampled from Balma Lake in summer (August) and autumn (October) 2017 in order to assess the influence of trace elements accumulation and environmental parameters (physicochemical parameters and nutrient characteristics of water) on oxidative stress biomarkers. Bioaccumulation of Al, As, Cd, Cr, Cu, Fe, Hg, Pb, Ni, Se, and Zn and metallothionein levels were measured in muscle tissue of males and females. Liver, gills, kidney, and spleen tissue samples were analyzed for superoxide dismutase, catalase, total glutathione peroxidase, selenium-dependent glutathione peroxidase, glutathione reductase, and glutathione S-transferase activity. Analysis of environmental parameters showed changes in biomarker levels with seasonal variations. Water temperature was significantly higher in summer than autumn (Wilcoxon test; p = .0078), while pH was significantly higher in autumn than in summer (Wilcoxon test; p = .0078). Sex-related differences in oxidative stress biomarkers in tissues were unremarkable, whereas seasonal variability of oxidative stress biomarkers was observed, with major differences occurred for liver in summer and for gills, kidney, spleen and muscle in autumn. Positive correlations between environmental parameters and biomarkers were noted. Major fluctuations in water temperature, pH, Cu, Pb and Hg produced changes in biomarker levels; however, increased food intake during the ice-free season was probably the main factor that influenced changes in oxidative stress biomarker levels in brook trout in this extreme ecosystem.

Abstract

Insect protein meals are considered as a promising alternative feedstuff in aquafeeds, but until now the effects induced by its dietary inclusion on histological features, gut mucin composition and oxidative stress in fish are still not assessed. In the present study, we describe these effects after inclusion of a partially defatted Hermetia illucens (HI) larvae meal in Oncorhynchus mykiss diets. Three isoproteic, isolipidic and isoenergetics diets were formulated containing HI larvae meal in partial substitution of fish meal: 0% (HI0, control diet), 25% (HI25) and 50% (HI50), corresponding to dietary inclusion levels of 0, 20% and 40% of HI. Fish were fed for 78 days with the experimental diets and at the end of the growing trial, histological and histochemical evaluations were performed on spleen, liver and gut samples of control and treatment groups. Oxidative stress biomarkers, such as superoxide dismutase, catalase, glutathione peroxidases, ethoxyresorufin O-deethylase, glutathione S-transferase, glutathione reductase, total glutathione and malondialdehyde were investigated in liver and kidney of experimental groups. Histopathological examination of liver, spleen and gut revealed no adverse effects following increasing levels of insect meal inclusions. Hermetia illucens consumption stimulated higher production of neutral than acidic mucins. Altered values of several antioxidant biomarkers indicated an unbalance of oxidative homeostasis in liver and mainly in kidney, linked to the feeding conditions. While lowering of glutathione peroxidase activity may reflect slight adverse effect of HI meals in both tissues, strengthening in levels of ethoxyresorufin O-deethylase, glutathione S-transferase and total glutathione may indicate the shielding of HI meals-mediated oxidative process. Although the administration of both inclusion levels of insect meal did not negatively affect the histological features in fish, the changes of oxidative stress biomarkers advise for an HI dietary inclusion lower than 20%.

Abstract

Understanding oxidative stress in high-mountain lake fish offers crucial insights into their health, resilience, and adaptation to extreme environmental changes. This study investigates the oxidative stress response of Cottus gobio in a high-mountain lake (Dimon Lake) located in the northeast Italy during the ice-free season, focusing on the relationship between oxidative stress biomarkers and physicochemical water parameters, as well as persistent and emerging contaminants. Significant seasonal variations were observed in water parameters, with lower oxygen, pH, conductivity, and phosphate levels in summer compared to autumn, while temperature, ammonium, and nitrate were higher in summer. Metal concentrations in C. gobio muscle were higher in autumn, with Zn showing the most significant increase. PAHs, NDL-PCBs, and pesticides were all below the limit of quantification in the fish muscle samples. No microplastics items were found in the gastrointestinal tracts of fish. Oxidative stress biomarkers revealed organ-specific and seasonal variations. The liver exhibited the highest activities of catalase (CAT), glutathione peroxidase (GPx), glutathione S-tranferase (GST), and glutathione reductase (GR), highlighting its central role in detoxification and metabolic processes. Superoxide dismutase (SOD) activity was notably higher in muscle tissue during summer, suggesting increased metabolic activity. A strong correlation was found between pH and the activities of SOD, CAT, GPx, GR, and metallothioneins (MTs), emphasizing the importance of water chemistry in influencing oxidative stress responses. This approach not only aids in understanding the local adaptations of these fish but also highlights the impacts of environmental stressors on high-mountain ecosystems.
