17. Drone Semen Cryopreservation with Protein Supplemented TL-Hepes Based Extender

Abstract

The aim of the current study was to determine the optimum concentration of bovine serum albumin for post-thawing quality of drone sperm and this is the first study to evaluate the effect of BSA supplemented TL-Hepes based extenders for drone semen cryopreservation. Sexually mature drones were used for semen collection. Pooled semen was diluted with TL-Hepes based extender supplemented with different concentrations of BSA (1 mg/mL, 3 mg/mL, and 5 mg/mL) or without BSA (control), at a final concentration of 100x10(6) spermatozoon/mL. Motility, plasma membrane functional integrity (HOST), and defected acrosome (PSA-FITC) were evaluated in the study. At post thaw, the highest sperm motility rates were obtained in the BSA5 group (P< 0.05). Functional integrity of sperm membrane was better preserved in the BSA3 and BSA5 groups compared to the other groups. The acrosomal integrity rates were higher in BSA5 group than in control group (P< 0.05). The study shows that bovine serum albumin supplemented TL-Hepes based extenders have beneficial effect on drone semen parameters at post-thaw. The results of the study demonstrated a notable advantage of using 5 mg/mL of BSA in TL-Hepes based extender.

19. Development And Life History Parameters Of Typhlodromus Recki (Acari: Phytoseiidae) Feeding On Tetranychus Urticae (Acari: Tetranychidae) At Different Temperatures

Abstract

Typhlodromus recki (Acari: Phytoseiidae) was collected from aubergines, pepper, black nightshade and jimsonweed plants infested with spider mites from a pesticide-free vegetable garden in Denizli, Turkey. The biology and life table parameters for T. recki feeding on Tetranychus urticae (Acari: Tetranychidae) at different temperatures (15, 20, 25, 30 and 35 degrees C) were determined. The results showed that total preadult periods of T. recki at 15, 20, 25, 30 and 35 degrees C were 19.6 +/- 0.51, 9.4 +/- 0.16, 7.6 +/- 0.14, 5.7 +/- 0.14 and 4.5 +/- 0.08 days, respectively. The longest oviposition period was 23.5 days at 20 degrees C. Total fecundity of T. recki significantly differed between the constant temperatures and the highest was observed at 20 degrees C (28.6 eggs/female) and 25 degrees C (23.5 eggs/female). Although females survived for 26 days at 35 degrees C, only four females laid eggs for 1 day. Thus, 35 degrees C is not suitable for the reproduction of T. recki. Total longevity of female was the longest at 15 degrees C (59.5 days) and 20 degrees C (51.3 days) whereas male total longevity was the longest at 15 degrees C (53.2 days). The intrinsic rate of increase (r) and finite rate of increase (lambda) were numerically the highest at 30 degrees C (0.17 and 1.18 d(-1)), but there was no statistical difference observed between 25 and 30 degrees C for both r and lambda values. The net productive rate was significantly highest at 20, 25 and 30 degrees C. The longest mean generation time occurred at 15 degrees C (42.6 days) and the shortest was at 35 degrees C (8.0 days). Abstract

Natural products (NPs) from microorganisms have been important sources for discovering new therapeutic and chemical entities. While their corresponding biosynthetic gene clusters (BGCs) can be easily identified by gene-sequencesimilarity-based bioinformatics strategies, the actual access to these NPs for structure elucidation and bioactivity testing remains difficult. Deletion of the gene encoding the RNA chaperone, Hfq, results in strains losing the production of most NPs. By exchanging the native promoter of a desired BGC against an inducible promoter in Dhfq mutants, almost exclusive production of the corresponding NP from the targeted BGC in Photorhabdus, Xenorhabdus and Pseudomonas was observed including the production of several new NPs derived from previously uncharacterized non-ribosomal peptide synthetases (NRPS). This easyPACId approach (easy Promoter Activated Compound Identification) facilitates NP identification due to low interference from other NPs. Moreover, it allows direct bioactivity testing of supernatants containing secreted NPs, without laborious purification.

Abstract

This study was carried out to determine the mites parasitizing aphids in the park and gardens in Aydin city between March 2019 and June 2020. As a result, a total of 1276 mite larvae were collected and 12 species were identified. Eight species, Allothrombium clavatum, A. polikarpi (Trombidiidae), Erythraeus (Erythraeus) phalangoides, E. (E.) uhadi, Erythraeus (Zaracarus) coleopterus, E. (Z.) iranicus, E. (Z.) kurdistaniensis (Erythraeidae) and Lassenia novoseljensis (Tanaupodidae), are new records to the Turkish fauna. The larvae of the mites were ectoparasites of 10 different aphid species: Aphis (Aphis) craccivora, A. (A.) nerii, Aulacorthum (Aulacorthum) solani, Brachycaudus (Appelia) tragopogonis, Hyalopterus pruni, Hyperomyzus (Hyperomyzus) lactucae, Macrosiphoniella (Macrosiphoniella) sanborni, Myzus (Nectarosiphon) persicae, Uroleucon (Uroleucon) sonchi, U. (U.) aeneum (Hemiptera: Aphididae). Erythraeus (E.) ankaraicus was the most common with a rate of 42.4% followed by A. triticium with 22.9% and E.(E.)phalangoideswith 19.7%. Most of the mites were collected from Uroleucon (U.) sonchi with a rate of 69.4%, followed by H. pruni with 8.5% and Aulacorthum (A.) solani with 6.0%. The mites were collected mostly in the spring with 86.9% and of which 51.9% was obtained in May. Allothrombium clavatumis also re-described and some data for collected species are corrected. 49. Biological Parameters Of The Predatory Mite Cheletomimus Bakeri (Acari: Cheyletidae) Feeding On Tetranychus Cinnabarinus (Acari: Tetranychidae)

Abstract

Developmental periods of egg, larva and nymphal stages and fecundity as well as predation of Cheletomimus bakeri (Acari: Cheyletidae) feeding on Tetranychus cinnabarinus (Acari: Tetranychidae) were evaluated at different temperatures (15, 20, 25, 30 and 35 degrees C) at 65 +/- 10% relative humidity and 16 L: 8D in the laboratory. The development periods of C. bakeri from egg through adult decreased significantly when the temperature was increased from 20 degrees C to 35 degrees C. Egg and total development periods of C. bakeri at 20, 25, 30 and 35 degrees C were 13.86, 7.98, 5.07, 4.08 days and 58.66, 41.51, 21.21, 22.92 days, respectively. The highest numbers of total and daily egg production were found at 20numbers of total and daily egg production were found at 20 degrees C and 30 degrees C, respectively. Net reproductive rate (R-0 = 13.29), mean generation time (T = 88.30), gross reproductive rate (GRR = 17.46) and doubling time (DT = 23.66) were the highest at 20 degrees C. The intrinsic rate of increase (r(m) = 0.0592) and finite capacity for increase (lambda = 1.061) for C. bakeri were the highest at 30 degrees C. Predation of C. bakeri increased throughout the range of prey densities. The highest consumption number of C. bakeri feeding on T. cinnabarinus males per day was 4.63, 4.70 and 4.60 when confined to 40, 80 and 160 individuals, respectively. Our data suggest that C. bakeri does not appear to have much promise for the control of spider mites because of the characteristics of the predator such as slow development period, poor searching capacity and low intrinsic rate of increase.
