1. In vitro bioassays for monitoring drinking water quality of tap water, domestic filtration and bottled water

Abstract

Location-specific patterns of regulated and non-regulated disinfection byproducts (DBPs) were detected in tap water samples of the Barcelona Metropolitan Area. However, it remains unclear if the detected DBPs together with undetected DPBs and organic micropollutants can lead to mixture effects in drinking water.To evaluate the neurotoxicity, oxidative stress response and cytotoxicity of 42 tap water samples, 6 treated with activated carbon filters, 5 with reverse osmosis and 9 bottled waters. To compare the measured effects of the extracts with the mixture effects predicted from the detected concentrations and the relative effect potencies of the detected DBPs using the mixture model of concentration addition.Mixtures of organic chemicals in water samples were enriched by solid phase extraction and tested for cytotoxicity and neurite outgrowth inhibition in the neuronal cell line SH-SY5Y and for cytotoxicity and oxidative stress response in the AREc32 assay.Unenriched water did not trigger neurotoxicity or cytotoxicity. After up to 500-fold enrichment, few extracts showed cytotoxicity. Disinfected water showed low neurotoxicity at 20- to 300-fold enrichment and oxidative stress response at 8- to 140-fold enrichment. Non-regulated non-volatile DBPs, particularly (brominated) haloacetonitriles dominated the predicted mixture effects of the detected chemicals and predicted effects agreed with the measured effects. By hierarchical clustering we identified strong geographical patterns in the types of DPBs and their association with effects. 10.1002/jms.3377 This paper presents a comparison of triple quadrupole tandem mass spectrometry (MS/MS) and Orbitrap high-resolution mass spectrometry (HRMS) combined to ultrahigh performance liquid chromatography for the determination of glucocorticoids and polyether ionophores in sewage, in order to show the major benefits and drawbacks for each mass spectrometry analyser. Overall, HRMS measurements have enhanced performance in terms of confirmatory capabilities than MS/MS measurements. Moreover, similar limits of quantification, limits of detection, linear range and repeatability for glucocorticoids with both the MS/MS and HRMS methods were compared, but in the case of polyether ionophores, slightly better limits of detection and limits of quantification were obtained with the HRMS method because of the high sensitivity obtained when diagnostic ions are used for quantification instead of selected reaction monitoring transitions for these compounds. The two methods have been applied to the analysis of several influent and effluent sewage samples from sewage treatment plants located in the Tarragona region (Catalonia, Spain), showing an excellent correlation between the two methods

Abstract

Static and dynamic headspace techniques have been applied to the extraction and concentration of volatile organic compounds (VOCs) from paprika oleoresin. In dynamic headspace, both purge and trap and sweeping techniques have been used. In order to show the influence of temperature on the extraction techniques, three differents temperatures have been applied to each technique. The separation and identification of the extracted VOCs has been carried out through HRGC/MS. The extraction techniques are simple and easily reproducible. Se han aplicado las técnicas de espacio de cabeza estático y dinámico para la extracción y concentración de los compuestos orgánicos volátiles de la oleorresina de pimentón. En espacio de cabeza dinámico se han utilizado las variantes de arrastre con borboteo y espacio de cabeza dinámico propiamente dicho. Con el fin de estudiar la influencia de la temperatura en ambas técnicas de extracción, se han realizado pruebas a tres temperaturas diferentes en cada una de ellas, estudiando la influencia de la misma en el proceso de aislamiento de los CVOs. La separación e identificación de los CVOs extraídos se ha llevado a cabo mediante HRGC/MS. Todas las técnicas presentadas son de aplicación simple y reproducibles con facilidad.
