Heavy metal contamination is a global environmental concern due to its persistence and toxicity. To explore soil microbial interaction mechanisms and their association with heavy metals on a Pb-Zn waste heap, ecological network analysis tools were used to analyze high-throughput data in microbiology. The microbial network was divided into several modules, but heavy metals were associated with specific modules. The heavy metal-tolerant module (M2) had a more negative than positive relationship with the heavy metal-mid-tolerant modules (M1 and M3). Tight coupling between fungal and bacterial operational taxonomic units (OTUs) within M2 was critical for module stability and heavy metal bioremediation. Additionally, members within M2 needed to form a positive relationship to cope with heavy metal contamination (As, Pb, Zn, Cu, and Cd). The study provides fundamental information for a deeper understanding of heavy metal bioremediation mechanisms in the Pb-Zn waste heap.

The predominant use of junk food in our societies is continuously held responsible for the obese body physiques and overweight among the kids and adolescents. The current supportive environments where organic foods are limited, and new processed foods have been brought to the market with more variant tastes and acceptability for the kids and adolescents that have diverged their eating patterns. It has significantly contributed to the health issues and growth discrepancies of the users. However, the awareness of the food contents is an important milestone for understanding the risks associated with the usage of junk foods. A quantitative approach has been used in this study to measure the effect of perceived severity, vulnerability and fear on the junk food eating behaviors and ultimately on the obesity. The moderating role of product knowledge hiding has also been measured on the relationship of junk food eating and obesity. Structural equation modeling is used using the software Smart-PLS for measuring the hypothesis with a sample size of 228 selected through purposive sampling. The sample consisted of kids and adolescents who were reached on purpose for data collection. The current study has explored the role of perceived severity, vulnerability and the fear of using junk foods which have been found as a negative effect on junk food eating behavior which is positively associated with obesity among the kids and adolescents. The result of study shows that perceived threat has a negative effect on the junk food eating behavior of the adolescents. However, the positive relationship of junk food eating behavior with obesity can be decreased if the information about the products is not hidden. This study will be useful for making the consumers aware of the product knowledge hiding of the junk food usage. Moreover, it will help the users in creating understanding of risks allied with the use of junk food which may be addressed in order to avoid obesity issues in the kids and adolescents globally.

The spiking market for natural canthaxanthin promoted researchers toward genetic engineering of heterologous hosts for canthaxanthin production. Mucor circinelloides is a dimorphic fungus that produces β-carotene as the major carotenoid and is considered as a model organism for carotenogenic studies. In this study, canthaxanthin-producing M. circinelloides strain was developed by integrating the codon-optimized β-carotene ketolase gene (bkt) of the Haematococcus pluvialis into the genome of the fungus under the control of strong promoter zrt1. First, a basic plasmid was constructed to disrupt crgA gene, a negative regulator of carotene biosynthesis resulted in substantial β-carotene production, which served as the building block for canthaxanthin by further enzymatic reaction of the ketolase enzyme. The genetically engineered strain produced a significant amount (576 ± 28 μg/g) of canthaxanthin, which is the highest amount reported in Mucor to date. Moreover, the cell dry weight of the recombinant strain was also determined, producing up to more than 9.0 g/L, after 96 h. The mRNA expression level of bkt in the overexpressing strain was analyzed by RT-qPCR, which increased by 5.3-, 4.1-, and 3-folds at 24, 48, and 72 h, respectively, compared with the control strain. To identify novel lipases with stability and long-chain fatty acids preference by phylogenetic evolution analysis methods from database. Thermo-stable Candida antarctica Lipase-A (CALA) was set as a template for gene mining by PSI-BLAST. Based on phylogenetic analysis, three candidate lipases exhibiting 97%, 55%, and 35% identities with CALA, respectively, were selected for overexpression and characterization. Lipase, PhLip from Pseudozyma hubeiensis SY62 showed highest activity towards long-chain fatty acids, and showed maximum activity at pH 9.0 and 60 °C, and stability between 40 and 50 °C for 4 h and at pH 7–10 for 12 h. Enzymatic hydrolysis of Mucor circinelloides WJ11 oils by PhLip was about twofold higher than that by CALA, with respect to hydrolysis of long-chain fatty acids. Besides, fatty acids with 18 carbons, including oleic acid, linoleic acid, and linolenic acid, were preferred as substrates. The current investigation discovered a stable lipase PhLip with long-chain fatty acids preference. PhLip may be a potential candidate for producing polyunsaturated fatty acids from natural oils.

Mucor circinelloides is an oleaginous fungus that utilizes a wide variety of carbon substrates for its growth. The different sources of carbon strongly influence the total lipid content of the fungus. These different carbon substrates are assimilated and dissimilated through different metabolic pathways before entering into the TAG synthesis pathway. In the present study, we attempted to explore the mechanism of ex-novo lipid biosynthesis in M. circinelloides WJ11 in response to exogenous plant oil as a carbon source through transcriptomic analysis. The lipid content of WJ11 grown in a media containing mixed soybean oil with glucose as a carbon source was up to 43.8%, an increase of 13.9% as compared to glucose alone as the carbon source. RNA-Seq analysis was performed to investigate global gene expression patterns in the oil-treated WJ11. Based on RNA-seq analysis, among the 4646 differentially expressed genes (DEGs), 2379 were up-regulated and 2267 down-regulated. The expression of acetyl-CoA synthetase, 6-phosphofructokinase, alcohol dehydrogenase (NADP+), fructose-bisphosphate aldolase, and pyruvate kinase was down-regulated while genes related to triglyceride synthesis were up-regulated. The majority of genes and pathways related to lipid biosynthesis were up-regulated indicating a diversion of metabolic pathways towards lipid biosynthesis. The data generated advance the genomic resources and provide insights into the mechanisms of ex-novo lipid accumulation in fungi that use exogenous oil as a carbon source.

Understanding the influence of long-term fertilization on the multifractal characteristics and erodibility of soil of cultivated land on the Loess Plateau of China could help prevent soil erosion and promote the sustainable development of agriculture. We collected 27 soil samples from 0 to20 cm layers of nine fertilizer treatments. Renyi spectrum (Dq) and singularity spectrum (ƒ(α)) were applied to characterize soil particle-size distribution (PSD). The multifractal parameters of capacity dimension (D0), entropy dimension (D1), Holder exponent of order zero (α0), and erodibility K and anti-erodibility indices (aggregate state and degree) were used to determine the effect of fertilization on soil properties and erodibility. The multifractal models could characterize the PSDs in the various fertilizer treatments well. Knowledge of the changes of soil properties and soil particle-size distribution (PSD) is important for quantifying soil quality and dynamics during natural succession. We applied multifractal theory to characterize the PSDs in five soil layers of nine croplands abandoned for less than 1 year, 2, 5, 7, 10, 15, 20, 30 and 35 years and evaluated the changes of soil organic carbon (SOC), total nitrogen (total N), total phosphorus (total P), soil texture and multifractal parameters [capacity dimension (D 0), entropy dimension (D 1), correlation dimension (D 2) and Holder exponent of order zero (α 0)] during natural succession on the Loess Plateau of China. Renyi spectra (D q ) and singularity spectra f(α) characterized the PSDs well and sensitively reflected the changes of heterogeneity of the surface soil (0–20 cm) during natural succession. SOC, total N and clay contents and the multifractal parameters increased significantly by the late stage of succession, mostly in the topsoil (0–10 cm). Natural succession thus effectively improved the soil physicochemical properties on the Loess Plateau of China, even though it was time-consuming. SOC and total N contents decreased with depth throughout the natural succession, but the multifractal parameters were higher in the topsoil only in the late stage. D 1 and D 2 were strongly and positively correlated with SOC and total N contents in the surface layers and with fine particles in all layers, suggesting that D 1 and D 2 may be sensitive and practical indices for quantifying changes in soil properties and erosion.

This paper attempts to explore the characteristics of the spatial structure and development dynamics of three predominant tree populations, Quercus aliena var. acutiserrata Maxim.(QA), Acer caesium subsp. giraldii (Pax) E. Murr.(AC), and Ulmus propinqua Koidz (U. japonica) Sag (UG), in a virgin old-growth oak broad-leaved mixed forest in the Qinling Mountains, China. We employed a bivariate distribution of the structural parameters (mingling, dominance, and uniform angle index) based on the relationship between nearest neighbor tree groups and found that all predominant populations with a high species mixture and random distribution pattern. The oxidizability of soil organic carbon (SOC) influences soil quality and carbon sequestration. Four fractions of oxidizable organic carbon (very labile (C1), labile (C2), less labile (C3) and non-labile (C4)) reflect the status and composition of SOC and have implications for the change and retention of SOC. Studies of the fractions of oxidizable organic carbon (OC) have been limited to shallow soil depths and agroecosystems. How these fractions respond at deep soil depths and in other types of land-use is not clear. In this study, we evaluated the vertical distributions of the fractions of oxidizable organic carbon to a soil depth of 5.0 m in 10 land-use types in the Zhifanggou Watershed on the Loess Plateau, China. Along the soil profile, C1 contents were highly variable in the natural grassland and shrubland I (Caragana microphylla), C2 and C4 contents were highly variable in the natural grassland and two terraced croplands, respectively, and C3 contents varied little. Among the land-use types, natural grassland had the highest C1 and C2 contents in the 0–0.4 m layers, followed by shrubland I in the 0–0.1 m layer. Natural grassland had the highest C4 contents in the 1.0–4.5 m layers. Natural grassland and shrubland I thus contributed to improve the oxidizability of SOC in shallow soil, and the deep soil of natural grassland has a large potential to sequester SOC on the Loess Plateau.

BackgroundLong-term fertilisation has a large influence on soil physical and chemical properties in agro-ecosystems. The effects on the distribution of aggregates, however, are not fully understood. We determined the dynamic change of the distribution of aggregates and soil organic carbon (SOC) content over time in a long-term field experiment established in 1998 on the Loess Plateau of China and illustrated the relationship between them.MethodsWe determined SOC content and the distribution of aggregates in nine fertiliser treatments: manure (M); nitrogen (N); phosphorus (P); M and N; M, N, and P; M and P; N and P; bare land; and an unfertilised control. These parameters were then used for a path analysis and to analyse the fractal dimension (Dv).ResultsThe organic fertiliser increased SOC content. The proportions of 0.1–0.25 mm microaggregates and 0.25–0.5 mm macroaggregates were higher and the proportion of the 0.01–0.05 mm size class of the silt + clay fraction was lower in the treatments receiving organic fertiliser (M, MN, MNP, and MP) than that in the control, indicating that the addition of organic fertiliser promoted aggregation. Analyzing the dynamic patterns of species diversity and spatial heterogeneity of vegetation in grasslands during secondary succession could help with the maintenance and management of these ecosystems. Here, we evaluated the influence of secondary succession on grassland plant diversity and spatial heterogeneity of abandoned croplands on the Loess Plateau (China) during four phases of recovery: 1–5, 5–10, 10–20, and 20–30 years. The species composition and dominance of the grassland vegetation changed markedly during secondary succession and formed a clear successional series, with the species assemblage dominated by Artemisia capillaris→ Heteropappus altaicus→ A. sacrorum. The diversity pattern was one of low–high–low, with diversity peaking in the 10–20 year phase, thus corresponding to a hump-backed model in which maximum diversity occurring at the intermediate stages. A spatially aggregated pattern prevailed throughout the entire period of grassland recovery; this was likely linked to the dispersal properties of herbaceous plants and to high habitat heterogeneity. We conclude that natural succession was conducive to the successful recovery of native vegetation. From a management perspective, native pioneer tree species should be introduced about 20 years after abandoning croplands to accelerate the natural succession of grassland vegetation.

Land use conversion could directly or indirectly influence heavy metal geochemistry by changing soil properties. The aim of this study was to explore the effect of land use conversion on surface soil heavy metal contamination in the karst plateau lakeshore wetlands of Southwest China. Based on this, a total of 120 soil samples were collected from 30 sites from different types of land uses (farmlands, grasslands and woodlands) around a lake in Suohuangcang National Wetland Park in August 2017. Contents of As, Cd, Cu, Cr, Hg, Pb and Zn were analyzed, and soil heavy metal contamination was assessed in all three land use types. Results showed that land use transformation from farmland to grassland or woodland was not conducive to the release of soil heavy metal. Surface soil of all three land use types have been moderately polluted by As, Cr, Pb, and Zn, and grassland and woodland also had moderate Cd contamination. The pollution load index (PLI) results revealed low heavy metal contamination in grassland and woodland but no contamination in farmland. Although the integrated contamination in the studied region did not pose a serious potential ecological risk (RI &lt, 150), it might affect human health through the water supply and food chain. Therefore, it is necessary to monitor and control As, Cd, Cr, Pb, and Zn concentrations of surface soil through controlling pollutants, improving waste treatment, as well as strengthening supervision and management in the vicinity of the Suohuangcang National Wetland Park.

Soil organic carbon (SOC) is an important component of agricultural soil. Understanding the storage of carbon (C) and nitrogen (N) helps us understand how ecosystems would respond to natural and anthropogenic disturbances under different management strategies. Although the quantity of SOC stocks in the Loess Plateau has been previously investigated, the distribution and storage of C in its deep soil layers has been poorly studied to date. The present study was conducted in the Zhifanggou Watershed on the Loess Plateau to evaluate the effect of the types of land use on the distribution and storage of SOC and total nitrogen (TN) at different soil depths. Ten types of land use were investigated: woodland (Robinia pseudoacacia), shrubland (Caragana korshinskii; Hippophae rhamnoides), natural grassland, artificial grassland (Medicago sativa), orchard, check-dam cropland, terraced cropland (Setaria italica; Zea mays), and sloped cropland. The results show that land use significantly affects the SOC and TN content. The reconversion of sloped croplands into forestlands and grasslands improve their SOC and TN content. Natural grassland, terraced cropland with Z. mays, and the artificial grassland had the highest SOC content within 0-60 cm, 60-100 cm, and 150-500 cm layers, respectively. The SOC and TN content in the top layer were higher than those in the deep layer. Natural grassland had the highest SOC and TN storage within 0-40 cm layer, followed by shrubland I (C. korshinskii), check-dam cropland, woodland, and shrubland II (H. rhamnoides), orchard and artificial grassland, whereas the sloped cropland, and the terraced cropland with S. italica had the lowest values. The SOC and TN storage between the top (0 -20 cm) and deep layers (0-100 cm, 0-300 cm, and 0-500 cm) can be modeled using linear function (y = ax + b). The results suggest that natural grasslands may be the optimal choice for SOC and TN sequestration in the Zhifanggou Watershed. Shrublands, woodland, artificial grasslands, orchards, terrace cropland with Z. mays and check-dam croplands are also recommended for their great contribution to SOC and TN storage in the area. Objective To observe the influence of different load on the rat gastrocnemius muscle spindle of shape and structure. Methods Adopt swimming training model. Medium load group swim 60 min each day. Over load group, loaded with the weight of 5 %, swim 60 min each day, 6 days a week. Adopt muscle spindle of Faworky’s display and electron microscopy techniques; observe changes of different load training on the rat gastrocnemius muscle spindle shape and structure. Results show that gastrocnemius muscle spindle form is smaller, while muscle spindle structure is normal. In medium load group muscle spindle structure is integrate, spindle extrinsic is aligned, muscle spindle in neural myelin structure are clear and complete, nuclear bag fiber, nuclear chain fiber diameter than the control group significantly increases. In over load group spindle body increased obviously, inside and outside is separated by capsule seriously. Muscle fibrils quantity is less, sac cells are swollen, axis clearance increased obviously, spindle in neural axis lining and myelin has separated. These preliminary findings indicate medium load movement could significantly improve muscle spindle shape and structure, while over load sports training could lead to pathological changes of muscle spindle structure.

Analyzing the dynamics of soil particle-size distributions (PSDs), soil nutrients, and erodibility are very important for understanding the changes of soil structure and quality after long-term land-use conversion. We applied multifractal Rényi spectra (Dq) and singularity spectra (f(α)) to characterize PSDs 35 years after conversions from cropland to shrubland with Caragana microphylla (shrubland I), shrubland with Hippophae rhamnoides (shrubland II), forested land, and grassland on the Loess Plateau of China. Multifractal parameters (capacity dimension (D0), entropy dimension (D1), D1/D0, correlation dimension (D2), and Hölder exponent of order zero (α0)) were used to analyze the changes of PSDs. Dq and f(α) characterized the PSDs well and sensitively represented the changes in PSDs after conversion. All types of land-use conversion significantly improved the properties of the topsoil (0–10 cm), but the effect of shrubland I and even forested land decreased with depth. All types of land-use conversion significantly increased D1 and D2 in the topsoil, and D1 and D2 in the 10–50 cm layers of shrubland II, forested land, and grassland and D1 in the 50–100 cm layers of shrubland II were significantly higher relative to the control. Both D1 and D2 were positively correlated with the contents of soil nutrients and fine particles and were negatively correlated with soil erosion, indicating that D1 and D2 were potential indices for quantifying changes in soil properties and erosion. In conclusion, all types of land-use conversion significantly improved the conditions of the topsoil, but conversion from cropland to shrubland II, forested land, and grassland, especially shrubland II and grassland, were more effective for improving soil conditions in deeper layers.

Background Understanding interspecific associations in old-growth forests will help to reveal mechanisms of interspecific replacement in the process of forest development and provide a theoretical basis for vegetation restoration and reestablishment. In this study, we analyzed interspecific associations of eleven dominant tree populations of varying development stages in an old-growth oak forest stand in the Qinling Mountains, China. We examined overall interspecific associations (multiple species) and pairwise interspecific associations (two species). Results Interspecific competition was intense during forest development and was the main factor driving succession. Community structure appears to become more stable over time which supports the harsh-benign hypothesis that interspecific competition is more common in stable sites. Conclusion Old growth oak (Quercus spp.) forests are distributed widely around the world in part due to oak being a typical K-selected species. K-selected species produce fewer, high-quality offspring with higher survival rates, strong competitive ability, and longevity. The resulting distribution shifted from clumped to random, likely as a result of intense interspecific competition creating ecological niche differentiation.
