Aiming to strengthen the performance of waste activated sludge digestion, pretreatment is a prerequisite for keeping operations within an industrially acceptable time- frame. In this study, the performance of microaeration coupled with thermophilic (MT) pretreatment on waste activated sludge solubilization and acidification was investigated. The results showed that the maximum soluble organics concentrations, including soluble proteins and carbohydrates, reached 2290 mg COD/L in 24 h when the ventilation rate and temperature were 0.05 vvm and 70 °C. SDS-polyacrylamide gel electrophoresis (SDS-PAGE) test on pure-culture (E.coli) and scanning electron microscopy analysis indicated that MT pretreatment effectively destroyed microbial cell wall and resulted in an increase in soluble proteins. Polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) analysis revealed that MT pretreatment reduced the diversity of the bacterial community during pretreatment. Along with the dissolution of a large quantity of organic matter, microbial species such as Lactococcus and Methyloversatilis bloomed at the end of acidification period, which contributed to SCFAs production. This study revealed that MT pretreatment facilitated the WAS hydrolysis efficiency and enhanced SCFAs (3149 mg COD/L with fermentation for 96 h) production, especially for acetic acid (50%) accumulation, which provides a new perspective for the application and recovery of sludge resources.

Objective To evaluate 4Rs principle applied in animal experiments for research and to optimize training outcomes with animal model. Methods The 4Rs principle was applied in four parts of training process with animal models including “replacing animal experiments with human physiological experiments” “integrating experimental projects, reducing the use of animals” “optimizing experimental process for minimizing animal suffering” and “strengthening ethical concepts, responsibility in research with experimental animals”. After class, feedbacks from students were collected through questionnaire survey and the data was analyzed. Results There were 92.0% medical students showed sympathize for the laboratory animals in the process of animal experiments; took care of the welfare of experimental animals while achieving training objectives. Conclusions The 4Rs principle provides a new idea and method for animal experiment and cultivates learners' animal ethics.

Background: Type 2 diabetes mellitus (T2DM) is a metabolic syndrome characterized by chronic inflammation, insulin resistance, and islet cell damage. The prevention of T2DM and its associated complications is an urgent public health issue that affects hundreds of millions of people globally. Numerous studies suggest that disturbances in gut metabolites are important driving forces for the pathogenesis of diabetes. However, the functions and mechanisms of action of most commensal bacteria in T2DM remain largely unknown. Methods: The quantification of bile acids (BAs) in fecal samples was performed using ultra-performance liquid chromatography–tandem mass spectrometer (UPLC-MS/MS). The anti-diabetic effects of Bacteroides uniformis (B. uniformis) and its metabolites cholic acid (CA) and chenodeoxycholic acid (CDCA) were assessed in T2DM mice induced by streptozocin (STZ) plus high-fat diet (HFD). Results: We found that the abundance of B. uniformis in the feces and the contents of CA and CDCA were significantly downregulated in T2DM mice. B. uniformis was diminished in diabetic individuals and this bacterium was sufficient to promote the production of BAs. Colonization of B. uniformis and intragastric gavage of CA and CDCA effectively improved the disorder of glucose and lipid metabolism in T2DM mice by inhibiting gluconeogenesis and lipolysis in the liver. CA and CDCA improved hepatic glucose and lipid metabolism by acting on the Takeda G protein-coupled receptor 5 (TGR5)/adenosine monophosphate-activated protein kinase (AMPK) signaling pathway since knockdown of TGR5 minimized the benefit of CA and CDCA. Furthermore, we screened a natural product—vaccarin (VAC)—that exhibited anti-diabetic effects by promoting the growth of B. uniformis in vitro and in vivo. Gut microbiota pre-depletion abolished the favorable effects of VAC in diabetic mice. Conclusions: These data suggest that supplementation of B. uniformis may be a promising avenue to ameliorate T2DM by linking the gut and liver. [ABSTRACT FROM AUTHOR]

Biogenetic nanopalladium (bio-Pd) has attracted increasing attention recently due to its economical and environmental friendly synthesis route. However, traditional bacteria suspensions formed palladium (Sus-Pd) is limited to be the electrochemical catalyst owing to the poor conductivity of bacterial cells. Herein, we demonstrated Pd nanoparticles, synthesized by electroactive Geobacter biofilm, can form a three-dimensional conductive network (EAB-Pd) that is beneficial to the electrons transfer. As a result, the EAB-Pd delivered an over 5-fold increase of current compared to the Sus-Pd in hydrogen evolution and the reductive degradation of nitro-, azo- and chloroaromatics. Superior performance of EAB-Pd was also observed in comparison with the commercial Pd catalyst. A good stability of EAB-Pd has been further confirmed under electrochemical and mechanical stresses as well as through the reuse after over 3 months of storage. This novel proposed method enables the direct electrochemical application of bio-Pd without the previous required cell carbonization and chemical binders.

High prevalence and metastasis rates are characteristics of lung cancer. Glycolysis provides energy for the development and metastasis of cancer cells. The 1,25‐dihydroxy vitamin D3 (1,25(OH)2D3) has been linked to reducing cancer risk and regulates various physiological functions. We hypothesized that 1,25(OH)2D3 could be associated with the expression and activity of Na+/H+ exchanger isoform 1 (NHE1) of Lewis lung cancer cells, thus regulating glycolysis as well as migration by actin reorganization. Followed by online public data analysis, Vitamin D3 receptor, the receptor of 1,25(OH)2D3 has been proved to be abundant in lung cancers. We demonstrated that 1,25(OH)2D3 treatment suppressed transcript levels, protein levels, and activity of NHE1 in LLC cells. Furthermore, 1,25(OH)2D3 treatment resets the metabolic balance between glycolysis and OXPHOS, mainly including reducing glycolytic enzymes expression and lactate production. In vivo experiments showed the inhibition effects on tumor growth as well. Therefore, we concluded that 1,25(OH)2D3 could amend the NHE1 function, which leads to metabolic reprogramming and cytoskeleton reconstruction, finally inhibits the cell migration. The spacecraft relative motion trajectory planning is one of the enabling techniques for autonomous proximity operations, especially in the increasingly complicated mission environments. Most traditional trajectory planning methods focus on improving the performance criteria in the deterministic conditions, whereas various uncertain elements in practice would significantly degrade the trajectory performance. Considering the uncertainties underlying the collision avoidance constraints, this paper suggests a model predictive control based online trajectory planning framework in which the obstacle information in higher-precision would be consistently updated by the onboard sensor. To improve the computational efficiency of the online planning framework, the rotating hyperplane (RH) technique is utilized to transform the nonlinear ellipsoidal keep-out zone constraints into convex formulations. And the concept of rotation window is introduced to eliminate the unexpected mismatch between the spacecraft motion and hyperplane rotation in the conventional RH method, which in sequence improves the RH method's capability for multiple obstacle avoidance problem. Moreover, a three-dimensional (3-D) extension strategy is proposed to simplify the computation procedure when applying the RH method for a 3-D collision avoidance problem. Numerical simulations are carried out to validate the performance of the proposed online trajectory planning framework in addressing the uncertain collision avoidance constraints.
