2. Zinc in the retinal pigment epithelium and choriocapillaris interface

The highest concentration of zinc in human tissues is found in the retinal pigment epithelium (RPE)-choroid complex. Despite the association of zinc deficiency with age-related macular degeneration (AMD) and the widespread use of zinc supplementation to slow the progression of AMD, very little is known about how zinc affects the RPE and the choroid. Molecular and cell biology techniques were used to uncover how changes in zinc levels could play a role in regulating the RPE-choroid complex. First, QRT-PCR was used to assess the expressions of all 24 known zinc transporters in cadaveric human RPE, cultured RPE cells and cells isolated from other parts of the retina, ZIP12 was identified as a potentially important transporter to regulate zinc levels at the RPE-choroid interface. As there is very little published about ZIP12, bioinformatics and data mining were used to understand how this protein might function. Confirmation of these predictions was achieved through the cloning and expression of V5-tagged ZIP12 protein in different cell lines. Based on these experiments, we concluded that ZIP12 is a plasma membrane transporter that mediates zinc influx. In parallel, we tested the hypothesis that extracellular zinc levels in Bruch’s membrane might be involved in regulating both the RPE as well as the fenestrated choroidal capillaries using cultures of ARPE19 and bEND5 cells respectively. The presence of extracellular zinc in the growth media affected the characteristics of ARPE19 cells as well as fenestrae formation in bEND5 cells. In summary, the range of zinc transporter at the RPE-choroid interface was defined and properties of one particular transporter, ZIP12 which may have a specific role at this site, were elucidated. Using cellular systems some of the effects of zinc on the RPE-choroid complex were investigated. Future studies are required to elucidate the role of zinc in the AMD pathogenesis.

Penelitian ini bertujuan untuk mengidentifikasi sejauh mana permainan edukatif Stop Disasters Game mampu menstimulasi pengetahuan dan pemahaman peserta didik SMAN 98 Jakarta pada materi pengurangan risiko bencana banjir. Penelitian ini dilaksanakan pada bulan November 2023 – Juni 2024. Metode yang digunakan dalam penelitian ini adalah eksperimen dengan menggunakan One Group Pre test-Post test Design. Populasi dalam penelitian ini adalah seluruh siswa kelas 12 SMAN 98 yang mengambil mata pelajaran geografi tahun ajaran 2024/2025 dengan jumlah 179 peserta didik. Teknik pengambilan sampel menggunakan random sampling sehingga jumlah sampel yang diujikan adalah sebesar 64 peserta didik. Pengambilan data menggunakan pre test dan post test dengan tes yang digunakan dalam bentuk pilihan ganda untuk mengukur tingkat pemahaman dan esai mengukur pengetahuan. Teknik analisis data untuk mengukur pengetahuan secara deskriptif persentase, sedangkan tingkat pemahaman menggunakan uji Normalized Gain (N Gain). Hasil analisis data rata-rata pengetahuan peserta didik sebelum diberikan perlakuan hanya sebesar 28%. Sementara itu, setelah adanya perlakuan dalam pembelajaran menggunakan Stop Disasters Game, rata-rata pengetahuan peserta didik meningkat menjadi 82%. Hal ini menunjukkan terdapat peningkatan sebesar 54% dan peningkatan tersebut berada pada kategori sedang, yang berarti terdapat kemajuan dan perkembangan yang positif. Hasil uji N Gain mendapat skor 0,67 dan berada pada kriteria sedang. N Gain skor yang memiliki nilai makin positif menunjukkan adanya peningkatan pemahaman pada peserta didik setelah pembelajaran. Sementara itu, persentase rata-rata nilai N Gain sebesar 67% dan berada pada kriteria cukup efektif. Dengan demikian, pemanfaatan Stop Disasters Game dapat meningkatkan pengetahuan dan pemahaman peserta didik pada pembelajaran pengurangan risiko bencana banjir.

DKI Jakarta was very disaster-prone area, therefore it necessary to strive for strategic steps to protect every citizen with disaster management efforts. Potential disasters in urban areas other than flooding are building and residential fires. This study was aim to determine the impact of fire disaster mitigation socialization on increasing students' knowledge at SMP Muttaqien, South Jakarta. Impact is calculated by knowing the difference in knowledge before and after socialization. The analysis technique used paired sample T-test. The sample was determined using random sampling, with total of 41 respondents. Based on the results of data processing, it was foundthat there was an increase in knowledge on students about fire disaster mitigation. This can be seen through the increased of the student's mean score, from 79.27 to 85.12. So it can be concluded that this socialization activity has a positive impact on increasing students' knowledge in dealing with fire disasters.

8. Study of the Motion Performance of Marine Current Power Plant Turbine Floaters Due To Ocean Current Forces under Moored Conditions

Indonesia targets carbon emissions to reach 0% in 2060 and is replaced by optimizing the use of renewable energy sources. Indonesia as an archipelago country, with the potential of thousands of straits can be utilized as a source of ocean currents as a source of electrical energy. The electricity generated is obtained from a turbine rotor that rotates due to the force of the ocean current flow. To support the turbine rotor to move in the sea, a floating support structure is needed. In this study, a trimaran tipe support structure is used where on the left and right sides are installed 2 (pieces) turbine rotors @ 50 kW each, so that the total has a capability of 200 kW (@4 x 50 kW). The novelty of this study is the utilization of Trimaran technology in marine current power generation turbines, which has good stability, low resistance, and a wider deck area rather than monohull structures. A numerical study using Computational Fluid Dynamics (CFD) is used to calculate the program. The results showed that the floater only moves backward and then is pulled forward with a small amplitude of movement in the X-direction, while those on the Y and Z axes are insignificant. The turbine floater can be immediately stabilized and the turbine rotor will rotate due to the force of the ocean current received. Therefore, in this study, the marine current turbine using trimaran type is showing good ability to survive in Indonesian waters even in high current areas.

Kebudayaan Islam berbasis kearifan lokal sangat kental dengan masyarakat Gorontalo. Keberagaman budaya Gorontalo yang lekat kaitannya dengan nuansa islami ini memberi dampak juga terhadap sektor pariwisata, salah satu ada di desa Bubohu, Kabupaten Gorontalo yang mengusung konsep desa wisata religius. Promosi pariwisata jika tidak dibarengi dengan strategi komunikasi yang baik, maka akan mempengaruhi eksistensi dari objek wisata itu sendiri. Berdasarkan itu, tujuan dari penelitian ini adalah untuk menganalisis strategi promosi pariwisata melalui media sosial yang dilakukan oleh pengelola dalam meningkatkan pengunjung di desa wisata religi Bubohu serta peran dari masyarakat setempat dalam mengenalkan potensi wisata di desa wisata religi Bubohu. Penelitian ini menggunakan data primer dan data sekunder. Dalam penelitian ini data primernya adalah hasil wawancara dengan pemerintah setempat dan pengelola desa wisata religi Bubohu di Kabupaten Gorontalo, data sekundernya adalah konten media sosial dari desa wisata religi Bubohu serta penelitian terdahulu yang terkait dengan penelitian ini. Dalam penelitian ini metode penarikan informan menggunakan Teknik purposive sampling. Informan dalam penelitian ini adalah pemerintah setempat dan pengelola objek wisata. Hasil penelitian menunjukan bahwa dalam melakukan promosi pariwisata dibutuhkan sinergi antara pemerintah, pengelola, serta masyarakat agar pesan promosi bisa lebih menjangkau masyarakat luas. Media sosial memberikan dampak yang besar dalam proses promosi serta dapat meningkatkan jumlah pengunjung objek wisata religi Bubohu.

The impact of Covid-19 does not only occur in the health sector, but also on other sectors such as social, economic, environmental and even geopolitical and including the education sector. Family socioeconomic conditions have an important role in distance learning process which is carried out during a pandemic so that this research aims toanalyze the family's economic condition of the distance learning process (PJJ) during the Covid-19 pandemic in Cibodas Jonggol Village, West Java. This type of research is quantitative research with a survey approach. The research sample from this study was a family that had family members who were still attending elementary school in Cibodas Village with a total sample of 82 respondents. The research instrument used is an interview with data analysis using descriptive quantitative. The results showed that the socioeconomic conditions of families in the Cibodas Jonggol Village area showed that there were still many who were at a very poor and poor level so that there was an inability to fulfill supporting facilities and infrastructure to carry out PJJ for families.

The 1945 Constitution states that education is a right for every citizen. Education is a conscious and planned effort to realize the atmosphere of learning and the learning process so that students can develop their potential. Education in Indonesia aims to educate the nation's life. Indonesia's education system in its development continues to make changes. The zoning policy in education is one form of positive response from the government to equalize education in each region. The zoning policy requires each student to go to school the closest distance from their residence. Schools provide about 90% of the student capacity measured by distance. The zoning policy in its development still leaves various problems. A common problem is the unavailability of educational facilities in a sub-district area so that some students have the potential to not go to school. Bekasi City is a metropolitan city that serves as a buffer for the capital city. In its development, Bekasi City has the potential to experience problems of inequality in the number of students and absorption in schools. This study aims to project the number of students and their absorption at various levels of education in Bekasi until 2025.

The purpose of this study was to determine the extent of changes in land cover around the Mount Sinabung area after the 2009-2019 eruption by monitoring through remote sensing imagery and GIS. The method used in this research is descriptive quantitative. The technique of data collection used document study techniques by collecting Landsat images are among the widely used satellite remote sensing data and their spectral, spatial and temporal resolution made them useful input for mapping and planning projects (Sadidy et al. 1981). Changes in land cover that occurred around the Mount Sinabung area were dominated by pyroclastic material due to eruption. In addition, changes in land cover also occur due to the abandonment of potential lands, such as local residents who work in the plantation sector are forced to leave that, so they eventually turn into shrubs. The direction of the dominant pyroclastic material slides was directed towards the east-south and southeast of Mount Sinabung, where the area was dominated by the plantation sector. The impact of the eruption of Mount Sinabung was directly and indirectly. The total land cover changes due to pyroclastic material in 2010 was an area of 26.27 Ha, in 2014 it was 475.82 Ha, 2017 was 1339.75 Ha, and 2019 was 1196.11 Ha.

Approximately 40% of Jakarta is below sea level when the tide is in, which is referred to as a fluviomarine landform. This study aims: (a) to analyse the relationship between total income and household water demand, and (b) to analyse the relationship between total income and the proportion of groundwater utilization. It uses quantitative and qualitative analysis survey methods, as well as sampling methods, to represent the population. The population of this research is comprised of households that use groundwater on land units made from two classes of landform, two classes of settlement pattern, and three classes of settlement density. To determine the 30 wells, samples with proportional random sampling of the land units formed with groundwater samples have been taken at a radius of 100m from each well sample of 110 households. Quantitative and qualitative approaches have been used to prove the research aims. The analysis of this study indicates that the total income is proportional to household water demand but that it is inversely proportional to the share of groundwater utilization. The results also show that groundwater is not the only source to fulfil household water demand, and that it is necessary to utilize other sources of water.

- Abstract
Age-related macular degeneration (AMD) is a common blinding disease in the western world that is linked to the loss of fenestration in the choriocapillaris that sustains the retinal pigment epithelium and photoreceptors in the back of the eye. Changes in ocular and systemic zinc concentrations have been associated with AMD; therefore, we hypothesized that these changes might be directly involved in fenestrae formation. To test this hypothesis, an endothelial cell (bEND.5) model for fenestrae formation was treated with different concentrations of zinc sulfate (ZnSO
4 ) solution for up to 20 h. Fenestrae were visualized by staining for Plasmalemmal Vesicle Associated Protein-1 (PV-1), the protein that forms the diaphragms of the fenestrated endothelium. Size and distribution were monitored by transmission electron microscopy (TEM). We found that zinc induced the redistribution of PV-1 into areas called sieve plates containing ~70-nm uniform size and typical morphology fenestrae. As AMD is associated with reduced zinc concentrations in the serum and in ocular tissues, and dietary zinc supplementation is recommended to slow disease progression, we propose here that the elevation of zinc concentration may restore choriocapillaris fenestration resulting in improved nutrient flow and clearance of waste material in the retina.- Published
