Classically, vicariant phenomena have been essentially identified on the basis of biogeographical and ecological data. Here, we report unequivocal evidences that demonstrate that a physical-chemical characterization of the epicuticular waxes of the surface of plant leaves represents a very powerful strategy to get rich insight into vicariant events. We found vicariant similarity between Cercis siliquastrum L. (family Fabaceae, subfamily Cercidoideae) and Ceratonia siliqua L. (family Fabaceae, subfamily Caesalpinoideae). Both taxa converge in the Mediterranean basin (C. siliquastrum on the north and C. siliqua across the south), in similar habitats (sclerophyll communities of maquis) and climatic profiles. These species are the current representation of their subfamilies in the Mediterranean basin, where they overlap. Because of this biogeographic and ecological similarity, the environmental pattern of both taxa was found to be very significant. The physical-chemical analysis performed on the epicuticular waxes of C. siliquastrum and C. siliqua leaves provided relevant data that confirm the functional proximity between them. A striking resemblance was found in the epicuticular waxes of the abaxial surfaces of C. siliquastrum and C. siliqua leaves in terms of the dominant chemical compounds (1-triacontanol (C30) and 1-octacosanol (C28), respectively), morphology (intricate network of randomly organized nanometer-thick and micrometer-long plates), wettability (superhydrophobic character, with water contact angle values of 167.5 ± 0.5° and 162 ± 3°, respectively), and optical properties (in both species the light reflectance/absorptance of the abaxial surface is significantly higher/lower than that of the adaxial surface, but the overall trend in reflectance is qualitatively similar). These results enable us to include for the first time C. siliqua in the vicariant process exhibited by C. canadensis L., C. griffithii L., and C. siliquastrum. [ABSTRACT FROM AUTHOR]

Reconstructions of vascular plant mitochondrial genomes (mt-genomes) are notoriously complicated by rampant recombination that has resulted in comparatively few plant mt-genomes being available. The dearth of plant mitochondrial resources has limited our understanding of mt-genome structural diversity, complex patterns of RNA editing, and the origins of novel mt-genome elements. Here, we use an efficient long read (PacBio) iterative assembly pipeline to generate mt-genome assemblies for Leucaena trichandra (Leguminosae: Caesalpinioideae: mimosoid clade), providing the first assessment of non-papilionoid legume mt-genome content and structure to date. The efficiency of the assembly approach facilitated the exploration of alternative structures that are common place among plant mitochondrial genomes. A compact version (729 kbp) of the recovered assemblies was used to investigate sources of mt-genome size variation among legumes and mt-genome sequence similarity to the legume associated root holoparasite Lophophytum. The genome and an associated suite of transcriptome data from select species of Leucaena permitted an in-depth exploration of RNA editing in a diverse clade of closely related species that includes hybrid lineages. RNA editing in the allotetraploid, Leucaena leucocephala, is consistent with co-option of nearly equal maternal and paternal C-to-U edit components, generating novel combinations of RNA edited sites. A preliminary investigation of L. leucocephala C-to-U edit frequencies identified the potential for a hybrid to generate unique pools of alleles from parental variation through edit frequencies shared with one parental lineage, those intermediate between parents, and transgressive patterns. O objetivo do presente trabalho foi definir a melhor dose de Osmocote® para a produção de mudas de Apuleia leiocarpa para 60 e 90 dias de permanência no viveiro após o transplantio e acompanhar a resposta do índice de clorofila das mudas durante o período em estudo. O delineamento experimental utilizado foi o inteiramente casualizado, sendo constituído de cinco tratamentos, e dez repetições. Os tratamentos consistiram em doses crescentes de Osmocote® ( 0, 3, 6, 9 e 12 kg m-3 de substrato).Avaliou-se o índice de clorofila, diâmetro do colo e altura das mudas aos 60 e 90 dias após transplante (DAT), área foliar, massa seca da parte aérea, comprimento e massa seca das raízes aos 90 DAT. A partir dos valores médios dos pontos de máximaeficiência técnica, de diâmetro de colo e altura das mudas, concluiu-se que a melhor dose de Osmocote® para a produção de mudas de A. leiocarpa é de 7,94 e 7,73 kg de Osmocote® m-3 de substrato, para um período de permanência em viveiro de 60 e 90 diasapós o transplantio, respectivamente. O índice de clorofila demonstrou comportamento quadrático frente às doses de Osmocote® e correlação positiva com as variáveis altura e diâmetro do colo das mudas.

Lectins are a structurally heterogeneous group of highly specific carbohydrate-binding proteins. Due to their great biotechnological potential, lectins are widely used in biomedical research. The purpose of the present study was to evaluate the healing potential of the lectin of Bauhinia variegata (nBVL) and its recombinant isoform (rBVL-1). Following surgical creation of dorsal skin wounds, seven groups of mice were submitted to topical treatment for 12 days with lectin, D-galactose, BSA and saline. The animals were anesthetized and euthanized on POD 2, 7 and 12 in order to evaluate the healing potential of each treatment. The parameters considered included wound size, contraction rate, epithelialization rate and histopathological findings. Wound closure was fastest in animals treated with rBVL-1 (POD 7). nBVL was more effective than the controls. All skin layers were reconstructed and keratin deposition increased. Our findings indicate that the lectin of Bauhinia variegata possesses pro-healing properties and may be employed in the treatment of acute skin wounds.

PREMISE OF THE STUDY: A diverse range of pollen morphologies occurs within the large, paraphyletic legume subfamily Caesalpinioideae, especially among early-branching lineages. Previous studies have hypothesized an association between surface ornamentation and pollination syndrome or other aspects of pollen function such as desiccation tolerance and adaptations to accommodate volume changes. METHODS: We reviewed caesalpinioid pollen morphology using light microscopy, scanning and transmission electron microscopy, in combination with a literature survey of pollination vectors. KEY RESULTS: Pollen structural diversity is greatest in the early-branching tribes Cercideae and Detarieae, whereas Cassieae and Caesalpinieae are relatively low in pollen diversity. Functional structures to counter desiccation include opercula (lids) covering apertures and reduced aperture size. Structures preventing wall rupture during dehydration and rehydration include different forms of colpi (syncolpi, parasyncolpi, pseudocolpi), striate supratectal ornamentation, and columellate or granular wall structures that resist tensile or compressive forces respectively. Specialized aperture structures (Zwischenkorper) may be advantageous for efficient germination of the pollen tube. CONCLUSIONS: In Detarieae and Cercideae in particular, there is potential to utilize pollen characters to estimate pollination systems where these are unknown. Supratectal verrucae and gemmae have apparently evolved iteratively in Cercideae and Detarieae. Carob is a caesalpinoid tree species widespread in the Mediterranean. In this paper, carob EST-SSR (Expressed Sequence Tags-Simple Sequence Repeat) markers were used to assess the level of genetic diversity among 71 cultivars and accessions collected in three Mediterranean countries (Italy, Malta and Spain). Starting from 20 microsatellite loci, we identified a set of 9 polymorphic EST-SSRs, with a number of alleles per locus ranging from 2 to 7, and a number of genotypes from 3 to 13. The nine polymorphic EST-SSRs discriminated most of the analyzed genotypes with the exceptions of some cultivated clones having similar phenotypic features and probably belonging to cultivar-populations, and clarified some cases of homonymy. A neighbor joining dendrogram generated three main clusters which did not show a correlation with the distribution areas of the analyzed genotypes. A Bayesian analysis evidenced a high degree of admixture among the gene pools from the three countries. The low level of diversification among geographical areas reveals that the asexual propagation of selected clones played a major role in the diffusion of the species in the western Mediterranean. Haematoxylum calakmulense Cruz Durán & M. Sousa (Leguminosae, Caesalpinoideae), a new Mesoamerican species from the Yucatan Peninsula, is described and illustrated. This new species is similar to H. campechianum L., but differs by having more leaflets, up to six pairs, that are smaller and thick, with secondary venation non-evident; the rachis and petiole always with uncinate white trichomes, while the leaflets present with white trichomes on the abaxial surface toward the base; the inflorescences are shorter, no more than 4.5 cm, the inflorescence rachis, pedicels, and calyx are always pilose and the petals are smaller. Abstract: An early Eocene (52±2Ma) flora from the Mwadui kimberlite pipe in Tanzania includes ten leaf morphotypes, small seeds, fossil wood putatively related to Cynometra (Detarieae, Fabaceae), and a sparse palynoflora. The leaf flora is characterised by microphyllous and notophyllous entire margined leaves that are suggested to be related to the Euphorbiaceae and Fabaceae, although many cannot be firmly placed in modern families. The wood is the oldest member of Cynometroxylon and confirms that the Detarieae were diverse by the early Paleogene. The palynoflora is dominated by Ailanthipites daedaleus and Triporopollenites mwaduiensis with lesser amounts of small tricolpate (Tricolpites) and monosulcate (Lilliacidites) grains together with spores (Cyathidites spp., Triporoletes laevigatus) and freshwater algal cysts. Other components of the palynoresidue include abundant charcoalified tracheids and fungal hyphae. The low diversity and sparse palynoflora suggests a restricted catchment with little input from the regional vegetation. The sediments accumulated in a freshwater lacustrine environment with palynological input from the crater walls that were covered by low diversity vegetation. This is supported by the leaf floras. Although the leaf floras are more diverse with nine morphotypes identified, the small leaves (microphyll to notophyll) suggest a water-limited palaeoenvironment. This is supported by the low diversity and abundance of fern and bryophyte spores. The presence of a single wood taxon not only indicates that there were large trees around the crater-lake but that the aboreal component was of low diversity. These observations suggest an open woodland-like setting, while the taxonomic affinities of the wood (Cynometra) suggest a humid microclimate possibly with marked wet–dry seasonal contrasts. A new mannose/glucose-specific lectin, named DigL, was purified from seeds of Dialium guineense by a single step using a Sepharose 4b-Mannose affinity chromatography column. DigL strongly agglutinated rabbit erythrocytes and was inhibited by d-mannose, d-glucose, and derived sugars, especially α-methyl- d-mannopyranoside and N-acetyl- d-glucosamine. DigL has been shown to be a stable protein, maintaining its hemagglutinating activity after incubation at a wide range of temperature and pH values and after incubation with EDTA. DigL is a glycoprotein composite by approximately 2.9% of carbohydrates by weight. By sodium dodecyl sulfate polyacrylamide gel electrophoresis analysis, the purified DigL exhibited an electrophoretic profile consisting of a broad band of 28-30 kDa. Analysis using electrospray ionization mass spectrometry indicated that purified DigL possesses a molecular average mass of 28 452 ± 2 Da and shows the presence of possible glycoforms. In addition, DigL exhibited an intermediary toxic effect on Artemia sp. nauplii, and this effect was both dependent on native structure and mediated by a carbohydrate-binding site. Abstract: A new lectin, BfL, was purified from Bauhinia forficata seeds by ammonium sulfate fractionation, DEAE-Sephadex ion exchange chromatography, Sepharose-4B and chitin affinity chromatographies and Superdex 75 size exclusion chromatography. The molecular homogeneity and purity of BfL were assessed by reversed-phase HPLC. BfL appeared as a single band of approximately 27.0kDa on SDS-PAGE under non-reducing and reducing conditions, and its molecular weight was determined to be 27,850Da by LC/ESI-MS. BfL is a glycoprotein with a carbohydrate content of 6.24% determined by the phenol–sulfuric acid method. Fetuin, asialofetuin, thyroglobulin and azocasein inhibited the hemagglutinating activity of BfL, whereas saccharides did not. BfL hemagglutinating activity was stable at 100°C for 30min, pH-dependent, with the highest activity at pH 6.0, and metal-independent. The primary structure of BfL shows similarity with other lectins from the genus Bauhinia. Deconvolution of the BfL circular dichroism (CD) spectrum indicated the presence of α-helix and β structures. BfL increases coagulation time, but this effect is not related to human plasma kallikrein or human factor Xa inhibition. BfL also inhibits ADP- and epinephrine-induced platelet aggregation in a dose-dependent manner and is the only currently described lectin from Bauhinia that exhibits anticoagulant and antiplatelet aggregating properties. Abstract: A new fossil species of Leguminosae is described from Neogene of northwest of Argentina. The good preservation of tissues and the diagnostic characters present in this fossil wood let assigning it to a new species of Gleditisioxylon Müller-Stoll and Madel. Gleditisioxylon riojana nov. sp. (Caesalpinoideae) has diagnostic features such as: growth rings distinct, semi-annular porosity, vestured pits, helical thickenings, simple plates, paratracheal parenchyma and rays 1-6 seriate. The possible climatic conditions of Toro Negro Formation were inferred by the use of xylological characters presents in this fossil wood. The presence of Gleditsioxylon added to other data, suggest a new hypothesis to explain the disjunction of Gleditsia L. genus and the occurrence of a single extant species in tropical and subtropical South America. A new galactose-specific lectin was purified from seeds of a Caesalpinoideae plant, Bauhinia variegata, by affinity chromatography on lactose-agarose. Protein extracts haemagglutinated rabbit and human erythrocytes (native and treated with proteolytic enzymes), showing preference for rabbit blood treated with papain and trypsin. Among various carbohydrates tested, the lectin was best inhibited by D-galactose and its derivatives, especially lactose. SDS-PAGE showed that the lectin, named BVL, has a pattern similar to other lectins isolated from the same genus, Bauhinia purpurea agglutinin (BPA). The molecular mass of BVL subunit is 32 871 Da, determined by MALDI-TOF spectrometry. DNA extracted from B. variegata young leaves and primers designed according to the B. purpurea lectin were used to generate specific fragments which were cloned and sequenced, revealing two distinct isoforms. The bvl gene sequence comprised an open reading frame of 876 base pairs which encodes a protein of 291 amino acids. The protein carried a putative signal peptide. Two new trypsin inhibitors, TDI-I and TDI-II, were purified from the seeds of the native Brazilian tree Copaifera langsdorffii (Caesalpinoideae, Leguminosae). The purification procedure involved ammonium sulfate fractionation, ion-exchange chromatography on DEAE-Sepharose, affinity chromatography on trypsin-Sepharose, and reversed-phase (RP) HPLC. RP-HPLC yielded two forms (TDI-I and TDI-II), as confirmed by isoelectric focusing, with pI values between 7.0 and 8.1. The molecular mass of the TDI forms was 24 kDa based on FPLC gel filtration on Superdex 75. Under reducing conditions in tricine SDS-PAGE the molecular masses of TDI-I and TDI-II were 12 and 10 kDa, respectively. The Ki values were 1.1 and 1.2 nM for TDI-I and TDI-II, respectively, and there was no inhibitory effect on chymotrypsin. Amino acid analysis revealed high levels of aspartic acid, glutamic acid, serine, glycine, proline, and lysine but low levels of methionine and aromatic amino acids in both inhibitors; the calculated molecular masses were 11,456 and 10,008 for TDI-I and II, respectively. Based on the N-terminal sequences of TDI-I and TDI-II, TDI-I belongs to the Kunitz family of trypsin inhibitors, whereas TDI-II showed no homology to any other protein. This observation suggests that TDI-II belongs to a new inhibitor subclass of low-molecular mass proteins in the subfamily Caesalpinoideae. [ABSTRACT FROM AUTHOR]
