Abstract

Magnetotactic bacteria (MTB) sequester iron from the environment to biomineralize magnetite or greigite nanoparticles in magnetosome organelles, though the necessity of intracellular iron storage for the formation process is still in question. Understanding the role of iron storage would make clear the contribution of MTB in geochemical iron cycling and its potential importance during the biosynthesis of application‐relevant magnetic nanoparticles. Herein, how scanning X‐ray fluorescence microscopy (SXFM) and nanoscale X‐ray absorption near‐edge structure (nano‐XANES) mapping can spatially and chemically identify intracellular iron species is reported, creating an opportunity to examine the role of iron storage in magnetite biomineralization at the single‐cell level. Fe K‐edge nano‐XANES measurements of Magnetospirillum gryphiswaldense in varied iron media conditions and iron storage capacity reveal a significant quantity of intracellular iron heterogeneities through a distinction between formed magnetosomes and intracellular iron material. This intracellular iron component is found in both early and late stages of biomineralization. The capabilities of nano‐XANES in providing an experimental advantage in the multidisciplinary field of biomineralization are highlighted. [ABSTRACT FROM AUTHOR]

Abstract

Monolayers of ligand-grafted nanoparticles at fluid interfaces exhibit a complex response to deformation due to an interplay of particle rearrangements within the monolayer, and molecular rearrangements of the ligand brush on the surface of the particles. We use grazing-incidence small-angle X-ray scattering (GISAXS) combined with pendant drop tensiometry to probe in situ the dynamic organization of ligand-grafted nanoparticles upon adsorption at a fluid-fluid interface, and during monolayer compression. Through the simultaneous measurements of interparticle distance, obtained from GISAXS, and of surface pressure, obtained from pendant drop tensiometry, we link the interfacial stress to the monolayer microstructure. The results indicate that, during adsorption, the nanoparticles form rafts that grow while the interparticle distance remains constant. For small-amplitude, slow compression of the monolayer, the evolution of the interparticle distance bears a signature of ligand rearrangements leading to a local decrease in thickness of the ligand brush. For large-amplitude compression, the surface pressure is found to be strongly dependent on the rate of compression. Two-dimensional Brownian dynamics simulations show that the rate-dependent features are not due to jamming of the monolayer, and suggest that they may be due to out-of-plane reorganization of the particles (for instance expulsion or buckling). The corresponding GISAXS patterns are also consistent with out-of-plane reorganization of the nanoparticles. [ABSTRACT FROM AUTHOR]

Abstract

Due to its remarkable mechanical and biological properties, there is considerable interest in understanding, and replicating, spider silk's stress-processing mechanisms and structure-function relationships. Here, we investigate the role of water in the nanoscale mechanics of the different regions in the spider silk fibre, and their relative contributions to stress processing. We propose that the inner core region, rich in spidroin II, retains water due to its inherent disorder, thereby providing a mechanism to dissipate energy as it breaks a sacrificial amide-water bond and gains order under strain, forming a stronger amide-amide bond. The spidroin I-rich outer core is more ordered under ambient conditions and is inherently stiffer and stronger, yet does not on its own provide high toughness. The markedly different interactions of the two proteins with water, and their distribution across the fibre, produce a stiffness differential and provide a balance between stiffness, strength and toughness under ambient conditions. Under wet conditions, this balance is destroyed as the stiff outer core material reverts to the behaviour of the inner core.

Abstract

Liquid jets are of interest, both for their industrial relevance and for scientific applications (more important, in particular for X-rays, after the advent of free-electron lasers that require liquid jets as sample carrier). Instability mechanisms have been described theoretically and by numerical simulation, but confirmed by few experimental techniques. In fact, these are mainly based on cameras, which is limited by the imaging resolution, and on light scattering, which is hindered by absorption, reflection, Mie scattering and multiple scattering due to complex air/liquid interfaces during jet break-up. In this communication it is demonstrated that synchrotron small-angle X-ray scattering (SAXS) can give quantitative information on liquid jet dynamics at the nanoscale, by detecting time-dependent morphology and break-up length. Jets ejected from circular tubes of different diameters (100-450 mm) and speeds (0.7-21 m s-1) have been explored to cover the Rayleigh and first wind-induced regimes. Various solvents (water, ethanol, 2-propanol) and their mixtures have been examined. The determination of the liquid jet behaviour becomes essential, as it provides background data in subsequent studies of chemical and biological reactions using SAXS or X-ray diffraction based on synchrotron radiation and free-electron lasers. Abstract

Nanoscience and nanotechnology are broad domains of research and innovation that represent a major fi eld of activity at global level. This originates from the outstanding demonstrations obtained in the last two decades of the possibility to control the organization of matter at the scale of few atoms and molecules, to address their properties and to obtain functional systems with potential exploitation in the fields of health, environment, energy and communication. Research in nanoscience requires direct sensitivity to the atomic scale and to the fundamental time scale of the processes ruling the assembly of matter and the interactions with light, electric current, and molecules at all levels of complexity, from the gases of the atmosphere to proteins and DNA. Nanoscience therefore necessitates special research infrastructures like “atomically clean environments” for the synthesis and the assembling of nanosystems on the one hand, and advanced radiation sources for the direct analysis of their properties (Large Scale Facilities for fine analysis of matter, LSFs) and for controlling the functionalities at the relevant space and time scale of the fundamental processes on the other hand. Both these infrastructures, which require substantial investment in terms of human and financial resources, are most often not co-located and not operated in close synergy. While the European offer of LSFs is adequate, the availability of open access nanoscience centres is overall undersized, and the link to LSF is weak. This is a severe limitation for the optimal development of nanoscience and the ensuing nanotechnologies, as well as a limiting factor for a scientifi c return from the LSFs. State of the art synthesis and characterization laboratories lack the direct use of numerical modelling, fine analysis, in-situ and in-operando experiments. Advanced beamlines on synchrotron radiation sources, free electron lasers and neutron sources generally lack adequate synthesis and sample preparation and delivery tools, or atomic resolution microscopy. Numerical simulations are most often performed on model systems and are not on-line with synthesis and experimental work. The Nanoscience Foundries & Fine Analysis project (NFFA) addresses the integration of all these competences and methods by setting up a European Distributed Research Infrastructure that will co-locate 3-6 Nanoscience Centres with LSFs and thus shall offer a unique environment for advanced research by providing open access to European and international scientists. NFFA intends to play a key role in the construction of the European Research Area by creating a unique environment for the support of leading research by users from academy and institutes, by offering advanced training to young researchers and engineers and by providing an effective support to innovation projects by diverse stakeholders. 35. Free jet micromixer to study fast chemical reactions by small angle X-ray scattering

Abstract

We present the design, fabrication process, and the first test results of a high aspect ratio micromixer combined with a free jet for under 100 µs time resolved studies of chemical reactions. The whole system has been optimized for synchrotron small angle X-ray scattering (SAXS) experiments. These studies are of particular interest to understand the early stages of chemical reactions, such as the kinetics of nanoparticle formation. The mixer is based on hydrodynamic focusing and works in the laminar regime. The use of a free jet overcomes the fouling of the channels and simultaneously circumvents background scattering from the walls. The geometrical parameters of the device have been optimized using finite element simulations, resulting in smallest features with radius <1 µm, and a channel depth of 60 µm, thus leading to an aspect ratio >60. To achieve the desired dimensions deep X-ray lithography (DXRL) has been employed. The device has been tested. First the focusing effect has been visualized using fluorescein. Then the evolution and stability of the jet, which exits the mixer nozzle at 13 m s−1, have been characterized. Finally SAXS measurements have been conducted of the formation of calcium carbonate from calcium chloride and sodium carbonate. The fastest measurement is 75 µs after the beginning of the mixing of the reagents. The nanostructural evolution of chemical reactions is clearly discernible.
