For the development of a competitive ELISA (cELISA) to detect serum antibodies against the Mycoplasma mycoides subsp. Mycoides (Mmm) (strain PG1), the causative agent of contagious bovine pleuropneumonia (CBPP), all the proteins of this pathogen were analyzed. Then, a specific extracellular region of a transmembrane protein with the potential for diagnosis was identified. After that, a monoclonal antibody (Mab) named 3A8 was obtained using this extracellular region as an immunogen. Finally, a cELISA was established with the extracellular domain of this transmembrane protein as the coating antigen, Mab 3A8 as the competitive antibody, and HRP-labeled goat anti-mouse IgG as the enzyme-labeled antibody. This established method was used to detect the antibody dynamic regularity of goats which are artificially immunized Mmm and was also compared with a commercial ELISA kit. Further, the sera of 1011 different cattle from border provinces of China were monitored using a candidate Mab 3A8 cELISA. The detection results of known background sera used in this study indicate that a candidate diagnostic marker was successfully identified by analyzing all the coding proteins of Mmm in this research, and the cELISA established based on the Mab 3A8 against this protein can detect CBPP-positive serum with specificity and has no cross-reaction with other related epidemic disease-positive sera. In addition, we tested the sera collected from the border areas of China using the established ELISA, and no positive sample was detected. The research protocol of the CBPP cELISA established in this study is different from the traditional method, which can greatly reduce the investment of manpower and capital and save development time. We believe that this study's protocol could serve as a reference for the development of detection methods for mycoplasma and other complex pathogens. Vaccination is the most cost-effective tool to control contagious bovine pleuropneumonia. The vaccines currently used in Africa are derived from a live strain called T1, which was attenuated by passage in embryonated eggs and broth culture. The number of passages is directly correlated to the degree of attenuation of the vaccinal strains and inversely correlated to their immunogenicity in cattle. Current quality control protocols applied to vaccine batches allow the assessment of identity, purity, and titers, but cannot assess the level of genetic drift form the parental vaccine strains. Deep sequencing was used to assess the genetic drift generated over controlled in vitro passages of the parental strain, as well as on commercial vaccine batches. Signatures of cloning procedures were detected in some batches, which imply a deviation from the standard production protocol. The use of effective vaccines is among the most important strategies for the prevention and progressive control of transboundary infectious animal diseases. However, the use of vaccine is often impeded by the cost, a lack of cold chains and other factors. In resource-limited countries in Africa, one approach to improve coverage and reduce cost is to vaccinate against multiple diseases using combined vaccines. Therefore, the objective of this study was to evaluate a combined vaccine for the prevention and control of Lumpy Skin Disease (LSD), Contagious Bovine Pleuropneumonia (CBPP) and Rift Valley fever (RVF). The LSD and CBPP were formulated as a combined vaccine, and the RVF was formulated separately as live attenuated vaccines. These consisted of a Mycoplasma MmmSC T1/44 strain that was propagated in Hayflick-modified medium, RVF virus vaccine, C13T strain prepared in African green monkey cells (Vero), and the LSDV Neethling vaccine strain prepared in primary testis cells. The vaccines were tested for safety via the subcutaneous route in both young calves and pregnant heifers with no side effect, abortion or teratogenicity. The vaccination of calves induced seroconversions for all three vaccines starting from day 7 post-vaccination (PV), with rates of 50% for LSD, 70% for CBPP and 100% for RVF, or rates similar to those obtained with monovalent vaccines. The challenge of cattle vaccinated with the LSD/CBPP and the RVF vaccine afforded full protection against virulent strains of LSDV and RVFV. A satisfactory level of protection against a CBPP challenge was observed, with 50% of protection at 6 months and 81% at 13 months PV. A mass vaccination trial was performed in four regions of Burkina Faso that confirmed safety and specific antibody responses induced by the vaccines. The multivalent LSD/CBPP+RVF vaccine provides a novel and beneficial approach to the control of the three diseases through one intervention and, therefore, reduces the cost and improves vaccination coverage. Contagious bovine pleuropneumonia (CBPP) and Contagious caprine pleuropneumonia (CCPP) are insidious diseases that affect cattle and goats respectively and are caused by highly infectious bacteria of the genus Mycoplasma - Mycoplasma mycoides subsp. mycoides (Mmm) and Mycoplasma capricolum subsp. capripneumoniae (Mccp) respectively. These contagious Mycoplasma spp are responsible for trans-national animal illness, wreaking havoc on cattle and small ruminant farmers across Africa, and causing immense veterinary health concern and a huge economic loss. This review aimed at highlighting the trends of CBPP and CCPP, from different studies carried out in various locations in Nigeria. In order to focus on current research works with specific concentration on CBPP and CCPP prevalence, epidemiology and impacts in Nigeria, we used a purposive selection method in identifying studies conducted between 2014 to 2021. This review confirms the increasing prevalence of CBPP and CCPP in Nigeria, with high endemicity in the Northern parts of Nigeria, as reported in all research work reviewed. Likewise, it was clearly identified that these diseases have been consistently left unmonitored in developing countries like in Nigeria. Hence, CCPP continues to ravage among cattle and small ruminant, particularly in the nomadic areas and farms where large numbers of these ruminants are kept for economic and commercial purposes. This review therefore recommends that more studies be conducted on CBPP and CCPP prevalence and transmission foci, particularly in the Southern parts of Nigeria, while effective control measures such as continuous vaccination, disease surveillance, quarantine, as well as early diagnosis and reporting of cases should be prioritized, implemented and sustained. In this study, the transmission dynamics of Contagious Bovine Pleuropneumonia (CBPP) by finite difference method are presented. This model is made up of sensitive, exposed, vaccinated, infectious, constantly infected, and treated compartments. The model is studied by the finite difference method. Firstly, the finite difference scheme is constructed. Then the stability estimates are proved for this model. As a result, several simulations are given for this model on the verge of antibiotic therapy. From these figures, the supposition that 50% of infectious cattle take antibiotic therapy or the date of infection decrease to 28 days, 50% of susceptible obtain vaccination within 73 days.

We present and analyze a mathematical model of the transmission dynamics of Contagious Bovine Pleuropneumonia (CBPP) in the presence of antibiotic treatment with limited medical supply. We use a saturated treatment function to model the effect of delayed treatment. We prove that there exist one disease free equilibrium and at most two endemic equilibrium solutions. A backward bifurcation occurs for small values of delay constant such that two endemic equilibriums exist if Rt ∈ (R∗t,1); where, Rt is the treatment reproduction number and R∗t is a threshold such that the disease dies out if and persists in the population if Rt > R∗t. However, when a backward bifurcation occurs, a disease free system may easily be shifted to an epidemic. The bifurcation turns forward when the delay constant increases; thus, the disease free equilibrium becomes globally asymptotically stable if Rt < 1, and there exist unique and globally asymptotically stable endemic equilibrium if Rt > 1. However, the amount of maximal medical resource required to control the disease increases as the value of the delay constant increases. Thus, antibiotic treatment with limited medical supply setting would not successfully control CBPP unless we avoid any delayed treatment, improve the efficacy and availability of medical resources or it is given along with vaccination.

Abstract Contagious bovine pleuropneumonia (CBPP) is a respiratory disease caused by Mycoplasma mycoides subsp. mycoides. Infection occurs via Mycoplasma-containing droplets and therefore requires close contact between animals. The current infection models are suboptimal and based on intratracheal installation of mycoplasmas or in-contact infection. This work tested the infection of adult cattle via aerosols containing live mycoplasmas mimicking the infection of cattle in the field. Therefore, we infected six cattle with aerosolized Mycoplasma mycoides subsp. mycoides strain Afadé over seven consecutive days with altogether 109 colony forming units. All animals seroconverted between 11–24 days post infection and five out of six animals showed typical CBPP lesions. One animal did not show any lung lesions at necropsy, while another animal had to be euthanized at 25 days post infection because it reached endpoint criteria. Seroconversion confirmed successful infection and the spectrum of clinical and lesions observed mirrors epidemiological models and the field situation, in which only a fraction of animals suffers from acute clinical disease post infection.

This opinion belongs to a series of opinions where these control measures will be assessed, with this opinion covering the assessment of control measures for Contagious Bovine Pleuropneumonia (CBPP). In this opinion, EFSA and the AHAW Panel of experts review the effectiveness of: (i) clinical and laboratory sampling procedures, (ii) monitoring period, (iii) the minimum radius of the protection and surveillance zones, and (iv) the minimum length of time the measures should be applied in these zones. The general methodology used for this series of opinions has been published elsewhere. Several scenarios for which these control measures had to be assessed were designed and agreed prior to the start of the assessment. Different clinical and laboratory sampling procedures are proposed depending on the scenarios considered. The monitoring period of 45 days was assessed as not effective and at least 90 days (3 months) is recommended in affected areas where high awareness is expected; when the index case occurs in an area where the awareness is low the monitoring period should be at least 180 days (6 months). Since transmission kernels do not exist and data to estimate transmission kernels are not available, the effectiveness of surveillance and protection zones for CBPP was based on expert knowledge. A surveillance zone of 3 km was considered effective, while a protection zone including establishments adjacent to affected ones is recommended. Recommendations, provided for each of the scenarios assessed, aim to support the European Commission in the drafting of further pieces of legislation, as well as for plausible ad hoc requests in relation to CBPP. In this review, basic epidemiological features of CBPP, complicated by existing different strains of Mycoplasmas with similar biochemical characteristics, with preference to Sub-Saharan Africa are discussed. Many sub-Saharan African countries are challenged by variable gaps that include diagnostic tools and control strategies. Science-based issues on diagnostic procedures, vaccination, treatment, and other control strategies are discussed. Participatory epidemiology (PE), a diagnostic technique used in the identification and solving of animal health problems in rural communities, was also discussed. PE application, in conjunction with conventional diagnostic tools, will improve CBPP identification in pastoral rural communities and promote control of the disease in Africa. Furthermore, adequate CBPP control can be achieved through stronger political commitments from governments by prioritizing the disease among major diseases of high economic importance to the livestock industry for surveillance and control. Abstract Background Contagious bovine pleuropneumonia (CBPP) caused by Mycoplasma mycoides subspecies mycoides (Mmm) is an important disease of cattle that causes serious economic losses. With the known effectiveness of new generation macrolides, tulathromycin and gamithromycin were assessed in comparison with oxytetracycline as a positive control and saline as a negative control for effectiveness in inhibiting lung lesion development, promoting resolution, preventing spread and bacteriological clearance in susceptible local cattle breeds in two separate studies in Kenya and Zambia. Animals were monitored for clinical signs, sero-conversion as well as detailed post-mortem examination for CBPP lesions. Results Using the Hudson and Turner score for lesion type and size, tulathromycin protected 90%, gamithromycin 80%, and oxytetracycline 88% of treated animals in Kenya. In Zambia, all animals (100%) treated with macrolides were free of lung lesions, while oxytetracycline protected 77.5%. Using the mean adapted Hudson and Turner score, which includes clinical signs, post-mortem findings and serology, tulathromycin protected 82%, gamithromycin 56% and oxytetracycline 80% of the animals in Kenya whereas in Zambia, tulathromycin protected 98%, gamithromycin 94% and oxytetracycline 80%. The saline-treated groups had 93 and 92% lesions in Kenya and Zambia respectively, with Mmm recovered from 5/14 in Kenya and 10/13 animals in Zambia. Whereas the groups treated with macrolides were free from lesions in Zambia, in Kenya 5/15 tulathromycin-treated animals and 6/15 gamithromycin-treated animals showed lesions. Oxytetracycline-treated animals showed similarities with 3/14 and 4/15 showing lesions in Zambia and Kenya respectively and Mmm recovery from one animal in Kenya and six in Zambia. Contagious bovine pleuropneumonia (CBPP) is one of the six high impact diseases for which currently World Organization for Animal Health (WOAH) has established an official procedure for recognition of disease-free status for trade purposes. Although its exact distribution, incidence, and impact have not been well established, the disease is known to be endemic in sub-Saharan Africa, where it has a severe socio-economic impact on people's livelihoods. Control or elimination of the disease can be achieved by various strategies including vaccination, movement control, and stamping-out. Due to many factors, countries in sub-Saharan Africa are still struggling with this disease, compromising continental and global eradication. Mycoplasma mycoides ssp. capri (Mmc) is one of the etiological microorganisms of contagious agalactia, which is among the diseases causing the highest economical losses in small ruminants. We report a disease outbreak in a German flock that led to significant suffering of goats characterized by mastitis, arthritis, pleuropneumonia and sudden deaths. Mmc was persistently isolated from many animals both from milk, and from a number of different swab and tissue samples. A number of closely related Mycoplasma spp. have to be taken into consideration to rule out important animal epizootics listed by European Animal Health Law and the World Organisation for Animal Health (WOAH). Some goats developed cross-reacting antibodies against Mycoplasma mycoides ssp. mycoides. Although Mmc is believed to be an uncommon microorganism in Germany, this study highlights that veterinarians should consider this pathogen in their work during herd health monitoring in Central Europe. Although eradication was not fully achieved, autogenous vaccination significantly seemed to improve animal health and welfare. • Mycoplasma mycoides subspecies capri is an uncommon pathogen in Germany and other Central European countries. • Outbreaks with this pathogen can have high clinical and economic impact. • Not only veterinarians in Southern Europe need to take this pathogen into consideration during herd health monitoring. • Signs of disease and serological diagnostics might give a suspect of important notifiable pathogens like contagious caprine and bovine pleuropneumonia, respectively. Contagious bovine pleuropneumonia (CBPP) is an infectious and highly contagious respiratory disease of cattle and water buffalo, which is caused by the Mycoplasma mycoides subspecies mycoides small colony. It induces significant economic losses and leads to a serious food security problem, negatively influencing peoples' livelihoods in affected countries. The disease has been reported in different parts of Ethiopia with prevalence ranging from 1.78 to 96%. However, there is not enough epidemiological information about CBPP in the northwestern part of the country, particularly in North Gondar Administrative Zone. This cross-sectional study, therefore, was conducted in four selected districts (Metema, Alefa, Quara, and Dembia) of North Gondar Administrative Zone to detect the incursion and estimate the seroprevalence of CBPP and to identify the potential predisposing factors associated with the spread and occurrence of CBPP in the area. A total of 751 serum samples were collected from 41 herds (villages) having no history of CBPP vaccination. Information like age, sex, breed, herd size, and management were collected during blood sample collection. Data related to agro-climatic zone, trade trekking route from or through CBPP endemic/epidemic zone, transhumance grazing route, vehicle route, and bordering with CBPP epidemic/endemic zone were obtained from district agricultural offices. The serum was screened for CBPP using competitive enzyme-linked immunosorbent assay (c-ELISA) test. The animal level and herd level apparent seroprevalences were 12.92% (95% CI: 10.70–15.52) and 65.85% (95% CI: 49.57–79.10), respectively. The true animal level and herd level prevalences were estimated at 20.13% (95% CI: 16.64–24.21) and 65.22% (95% CI: 48.64–78.72%), respectively. At the animal level, trekking route from or through CBPP endemic/epidemic zone to the study area [OR = 4.77 (95% CI: 1.92–11.84) compared to without trekking route] was identified as the most important risk factor for CBPP spread and seropositivity. In general, epidemiological evidence of the present study indicates that CBPP is a prevalent disease, and animal trekking is an important risk factor for spread of the disease in the study area. This needs due attention from the government and other concerned bodies for its prevention and control to mitigate its economic impact.

Contagious bovine pleuropneumonia is a trans-boundary animal disease caused by Mycoplasma mycoides subsp. mycoides. This study was designed to determine the seroprevalence of contagious bovine pleuropneumonia (CBPP) in three selected south eastern states (Anambra, Enugu and Imo) of Nigeria using competitive enzyme-linked immunosorbent assay (c-ELISA). A total of 438 bovine sera samples were collected randomly for four months (December 2019 to March 2020) and screened for antibodies to Mycoplasma mycoides subsp. mycoides (Mmm) using IDEXX CBPP antibody ELISA kit (CIRAD, France). Results showed an overall prevalence of 59.4% for the three states screened. Antibodies to Mmm were detected in all the three states. Enugu state had the highest prevalence (64.3%) followed by Imo state (63%) and Anambra state (50.7%). Female animals had higher prevalence of CBPP than male. However, it was not statistically significant (P> 0.05). This study confirms the presence of CBPP in south eastern Nigeria, and could be used as a base line data for future studies in this region. Mycoplasma mycoides subsp. mycoides (Mmm) is the etiological agent of contagious bovine pleuropneumonia (CBPP), one of the major diseases affecting cattle in sub-Saharan Africa. Some evidences suggest that the immune system of the host (cattle) plays an important role in the pathogenic mechanism of CBPP, but the factors involved in the process remain largely unknown. The present study aimed to investigate the cell response of bovine polymorphonuclear neutrophils (PMNs) after Mmm in vitro exposure using one step RT-qPCR and Western blotting. Data obtained indicate that gene and protein expression levels of some pro-inflammatory factors already change upon 30 min of PMNs exposure to Mmm. Of note, mRNA expression level in Mmm exposed PMNs increased in a time-dependent manner and for all time points investigated; targets expression was also detected by Western blotting in Mmm exposed PMNs only. These data demonstrate that when bovine PMN cells are triggered by Mmm, they undergo molecular changes, upregulating mRNA and protein expression of specific pro-inflammatory factors. These results provide additional information on host-pathogen interaction during CBPP infection.

Contagious bovine pleuropneumonia (CBPP) is a highly contagious transboundary disease of ruminants which is currently widely spread on the African continent. Morbidity rate in a susceptible herd reaches 100% and mortality rate varies from 10% to 90% depending on the breed, individual susceptibility and animal resistance. Today, there is a huge risk of the agent introduction with animals and raw material exported from affected endemic areas of Africa to the areas free from contagious bovine pleuropneumonia (including Eurasia).

Contagious bovine pleuropneumonia (CBPP) and contagious caprine pleuropneumonia (CCPP) are major infectious diseases of ruminants caused by mycoplasmas in Africa and Asia. In contrast with the limited pathology in the respiratory tract of humans infected with mycoplasmas, CBPP and CCPP are devastating diseases associated with high morbidity and mortality. Beyond their obvious impact on animal health, CBPP and CCPP negatively impact the livelihood and wellbeing of a substantial proportion of livestock-dependent people affecting their culture, economy, trade and nutrition. The causative agents of CBPP and CCPP are Mycoplasma mycoides subspecies mycoides and Mycoplasma capricolum subspecies capripneumoniae, respectively, which have been eradicated in most of the developed world. The current vaccines used for disease control consist of a live attenuated CBPP vaccine and a bacterin vaccine for CCPP, which were developed in the 1960s and 1980s, respectively. Both of these vaccines have many limitations, so better vaccines are urgently needed to improve disease control. In this article the research community prioritized biomedical research needs related to challenge models, rational vaccine design and protective immune responses. Therefore, we scrutinized the current vaccines as well as the challenge-, pathogenicity- and immunity models. Contagious bovine pleuropneumonia (CBPP) is a respiratory disease of cattle that is listed as notifiable by the World Organization for Animal Health. It is endemic in sub-Saharan Africa and causes important productivity losses due to the high mortality and morbidity rates. CBPP is caused by Mycoplasma mycoides subsp. mycoides (Mmm) and is characterized by severe fibrinous bronchopneumonia and pleural effusion during the acute to subacute stages and by pulmonary sequestra in chronic cases. Additional lesions can be detected in the kidneys and in the carpal and tarsal joints of calves. Mmm infection occurs through the inhalation of infected aerosol droplets. After the colonization of bronchioles and alveoli, Mmm invades blood and lymphatic vessels and causes vasculitis. Moreover, Mmm can be occasionally demonstrated in blood and in a variety of other tissues. In the lung, Mmm antigen is commonly detected on bronchiolar and alveolar epithelial cells, in lung phagocytic cells, within the wall of blood and lymphatic vessels, inside necrotic areas, and within tertiary lymphoid follicles. Mmm antigen can also be present in the cytoplasm of macrophages within lymph node sinuses, in the germinal center of lymphoid follicles, in glomerular endothelial cells, and in renal tubules. A complete pathological examination is of great value for a rapid presumptive diagnosis, but laboratory investigations are mandatory for definitive diagnosis. The purpose of this review is to describe the main features of CBPP including the causative agent, history, geographic distribution, epidemiology, clinical course, diagnosis, and control. Background: Contagious bovine pleuropneumonia (CBPP) caused by Mycoplasma mycoides subspecies mycoides (Mmm) is an important disease of cattle that causes serious economic losses. With the known effectiveness of new generation macrolides, tulathromycin and gamithromycin were assessed in comparison with oxytetracycline as a positive control and saline as a negative control for effectiveness in inhibiting lung lesion development, promoting resolution, preventing spread and bacteriological clearance in susceptible local cattle breeds in two separate studies in Kenya and Zambia. Animals were monitored for clinical signs, sero-conversion as well as detailed post-mortem examination for CBPP lesions. Results: Using the Hudson and Turner score for lesion type and size, tulathromycin protected 90%, gamithromycin 80%, and oxytetracycline 88% of treated animals in Kenya. In Zambia, all animals (100%) treated with macrolides were free of lung lesions, while oxytetracycline protected 77.5%. Using the mean adapted Hudson and Turner score, which includes clinical signs, post-mortem findings and serology, tulathromycin protected 82%, gamithromycin 56% and oxytetracycline 80% of the animals in Kenya whereas in Zambia, tulathromycin protected 98%, gamithromycin 94% and oxytetracycline 80%. The saline-treated groups had 93 and 92% lesions in Kenya and Zambia respectively, with Mmm recovered from 5/14 in Kenya and 10/13 animals in Zambia. Whereas the groups treated with macrolides were free from lesions in Zambia, in Kenya 5/15 tulathromycin-treated animals and 6/15 gamithromycin-treated animals showed lesions. Oxytetracycline-treated animals showed similarities with 3/14 and 4/15 showing lesions in Zambia and Kenya respectively and Mmm recovery from one animal in Kenya and six in Zambia. In both studies, lesion scores of saline-treated groups were significantly higher than those of the antibiotic treated groups (p < 0.001). In sentinel animals, CBPP lesions were detected and Mmm recovered from one and two animals mixed with the saline-treated groups in Kenya and Zambia respectively. Conclusions: This study demonstrated that tulathromycin, a mycoplasmacidal, can achieve metaphylactic protection of up to 80%, while non-recovery of Mmm from sentinels suggests macrolides effectiveness in preventing spread of Mmm. Fasciolosis and Contagious Bovine Pleuropneumonia (CBP) are characterized by gross pathognomonic lesions in organ/offal of slaughtered cattle which result in condemnation of unsafe meat. Both are the major causes of bovine organ/offal condemnation in Nigeria. Postmortem detection with focus on Fasciola and CBP infections were made in cattle slaughtered at Ikpa market abattoir, Nsukka between May -- July 2018. In addition, the economic losses associated with the disease conditions were assessed. Out of 474 cattle examined, 114(24 %) and 200(42.2 %), had gross lesions of fasciolosis and CBP respectively, while 66(13.9 %) were positive for the both conditions. Both lesions were observed in the three breeds of slaughtered cattle (White Fulani, Sokoto gudali and Ndama) irrespective of sex and age. The disease lesions were not significantly associated (p<0.05) with sex, age and breed of cattle. However, there was a significant relationship (p>0.05) between the prevalence of CBP and fasciolosis lesions in the slaughtered cattle. The economic loss due to condemnation of affected organs was estimated at ₦ 1, 716,900.00 (US$ 4,769) in 24 days, approximately equivalent to ₦ 71, 537.50 (US$ 198.8) on daily basis. The findings have revealed the high level of organ damages and financial losses due to fasciolosis and CBP in the study area. This portends dangers not limited to livestock production and the economy, but zoonosis and other public health issues. Abstract The study estimates cattle owners' willingness-to-pay (WTP) for Contagious Bovine Pleuropneumonia (CBPP) vaccine in Samburu county, Kenya. Of particular policy relevance, the study presents findings on WTP for i) improved access to vaccines and ii) timely access to disease-risk information. The mean price for a CBPP vaccine was estimated at KES 66 (USD 0.64). This price relates to a CBPP vaccine that requires a 1.8 h commute, cattle owners' receipt of timely information that the CBPP disease risk is low-moderate and the vaccine lowers the risk of either tail-drop or post-vaccine abortion. The conditional WTP for mean travel duration and high-risk information are similar at KES 53.9 and KES 51.5. The marginal effect on demand for a 1 h additional travel duration and provision of CBPP disease risk information was estimated as a 1.5 per cent reduction and 2.3 increase. The results of this study indicate that cattle owners value greater levels of knowledge concerning the changing risk profile of CBPP in their community and improved access to CBPP vaccination services. In this paper we present a mathematical model for the transmission dynamics of Contagious Bovine Pleuropneumonia (CBPP) by considering antibiotic treatment and vaccination. The model is comprised of susceptible, vaccinated, exposed, infectious, persistently infected, and recovered compartments. We analyse the model by deriving a formula for the control reproduction number Rc and prove that, for Rc<1, the disease free equilibrium is globally asymptotically stable; thus CBPP dies out, whereas for Rc>1, the unique endemic equilibrium is globally asymptotically stable and hence the disease persists. Thus, Rc=1 acts as a sharp threshold between the disease dying out or causing an epidemic. As a result, the threshold of antibiotic treatment is αt⁎=0.1049. Thus, without using vaccination, more than 85.45% of the infectious cattle should receive antibiotic treatment or the period of infection should be reduced to less than 8.15 days to control the disease. Similarly, the threshold of vaccination is ρ⁎=0.0084. Therefore, we have to vaccinate at least 80% of susceptible cattle in less than 49.5 days, to control the disease. Using both vaccination and antibiotic treatment, the threshold value of vaccination depends on the rate of antibiotic treatment, αt, and is denoted by ραt. Hence, if 50% of infectious cattle receive antibiotic treatment, then at least 50% of susceptible cattle should get vaccination in less than 73.8 days in order to control the disease. Livestock vaccination coverage rates remain low in many lower and middle income countries despite effective vaccines being commonly available. Consequently, many preventable infectious livestock diseases remain highly prevalent, causing significant animal mortalities and threatening farmers' livelihood and food security. This study sought to assess farmers' maximum willingness to pay (WTP) for contagious bovine pleuropneumonia (CBPP), and peste-des-petits-ruminants (PPR) vaccination of cattle, and sheep and goats, respectively. Overall, 350 ruminant livestock farmers were randomly selected from three districts located in the northern, middle and southern farming belts of Ghana. We implemented a double-bounded dichotomous contingent valuation experiment, where farmers indicated their WTP for vaccinating each livestock specie(s) owned at randomly assigned price points. WTP responses were analyzed using maximum likelihood estimation, and factors influencing WTP were assessed using censored regression analysis accounting for village-level clustering. Mean WTP for CBPP vaccination was USD 1.43 or Ghanaian Cedi (GHC) 8.63 (95% CI: GHC 7.08–GHC 10.19) per cattle. Mean WTP for PPR vaccination was USD 1.17 or GHC 7.02 (95% CI: GHC 5.99–GHC 8.05) per sheep, and USD 1.1 or GHC 6.66 (95% CI: GHC 5.89–GHC 7.44) per goat. WTP was positively associated with resilience, limited knowledge about vaccines (assessed prior to WTP experiment), farmland size, and male gender, after adjusting for other covariates. To attain 70% vaccination coverage in Ghana, vaccination costs should be no larger than GHC 5.30 (USD 0.88) for CBPP per cattle and GHC 3.89 (USD 0.65) and GHC 3.67 (USD 0.61), respectively, for PPR vaccines per sheep and goat. Ruminant livestock farmers in Ghana value vaccination highly, and are, on average, willing to pay vaccination costs that exceed the prevailing market prices (GHC 6 for CBPP and GHC 5 for PPR vaccination) to protect their livestock resources. To achieve 70% coverage, only minor subsidies would likely be required. Bacterial pathogen-host interactions are a complex process starting with adherence and colonization followed by a variety of interactions such as invasion or cytotoxicity on one hand and pathogen recognition, secretion of proinflammatory/antibacterial substances and enhancing the barrier function of epithelial layers on the other hand. Therefore, a variety of in vitro , ex vivo and in vivo models have been established to investigate these interactions. Some in vitro models are composed of different cell types and extracellular matrices such as tissue explants or precision cut lung slices. These complex in vitro models mimic the in vivo situation more realistically, however, they often require new and more sophisticated methods for quantification of experimental results. Here we describe a multiplex qPCR-based method to quantify the number of bacteria of Mycoplasma (M.) mycoides interacting with their hosts in an absolute manner as well as normalized to the number of host cells. We choose the adenylate kinase (adk) gene from the pathogen and the Carcinoembryonic antigen-related cell adhesion molecule 18 (CEACAM18) gene from the host to determine cell numbers by a TaqMan-based assay system. Absolute copy numbers of the genes are calculated according to a standard containing a defined number of plasmids containing the sequence which is amplified by the qPCR. The new multiplex qPCR therefore allows the quantification of M. mycoides interacting with host cells in suspension, monolayer, 3D cell culture systems as well as in host tissues. • The new TaqMan-based qPCR was developed for absolute quantification of M. mycoides. • The duplex qPCR quantifies Mmm and Mmc normalized to the interacting host cells. • Assays are suitable for various in vitro models and tissue samples. Contagious bovine pleuropneumonia (CBPP) is a highly contagious disease of cattle caused by Mycoplasma mycoides subsp. mycoides Biotype Small Colony (MmmSC). The disease currently occurs in most of sub-Saharan Africa and where it is endemic and a major constraint for improving pastoral productivity. Following the persistence of this scourge, and in order to control this disease, a serological survey was conducted to determine the prevalence of CBPP in Niger. In fact, 1,590 sera were collected following a stratified sampling plan based on the risk factor of dissemination of CBPP. The analysis were performed at the Central Livestock Laboratory using the c-Elisa test. The results obtained show a wide distribution of the disease with an overall prevalence of 4.15% at individual level. The highest prevalences were recorded in the South-East regions [Zinder (7.5%), Diffa (7.5%)] and the West part [Tahoua (6.9%)]. The prevalence at the commune level was about 36.55%, which was relatively high. The prevalence at strata level was 36.55% (95% PI 0.2428–0.4882). The expected prevalences did not match those found. The results of this serological survey are considered the reference situation (T0) of CBPP in Niger with the PRAPS project, and allowed to the country to redefine control policies for better control of the disease at national and sub-regional level.

The current study was conducted on 110 lung samples from 2 - 3 years old imported beef cattle slaughtered at the Middle East abattoirs of Abu-Simbel city -Aswan Governorate- Egypt, in the period from July 2017 to March 2018. The aim of the present work was to characterize and detect the incidence rate of the pulmonary affections. During the postmortem examination, the pulmonary affections were recorded grossly and specimens were taken for histopathological examinations. The histopathological examinations revealed that 36 samples (32.7%) showed fibrinous pleuropneumonia. According to the characteristic features of lesions in the microscopical studies, we grouped them into (a) Acute fibrinous pleuropneumonia in 31cases (28.1 %) and (b) Organized fibrinous pleuropneumonia in 5 cases (4.5 %). When contagious bovine pleuropneumonia (CBPP) was first detected on a farm north of Melbourne, at Bundoora, in 1858, the predominant theory of miasma was being challenged by contagionist theories of disease transmission. This well‐documented case was recorded during a period of change in the scientific assessment of disease and therefore affords an exploration of what aspects of the landscape were considered important for livestock health at the time. Although the introduction, vaccination programs and eventual eradication of CBPP on mainland Australia has been well explored, scholars have neglected this aspect of the disease's history. By comparing 19th century records of farmland with how the site appears today, it is also possible to highlight the limited information provided by contemporary texts, while at the same time developing an appreciation of the ways in which the perception of the rural landscape has changed. This differing perception has implications for the utilisation of these sources for veterinary and environmental historians seeking to understand the mid‐19th century agricultural landscape and how it relates to animal health. Reactive oxygen species (ROS) are suggested to play a role in the pathogenesis of contagious bovine pleuropneumonia, a severe respiratory disorder caused by Mycoplasma mycoides subsp. mycoides ( Mmm ). The present study investigated the generation of ROS by different strains of Mmm , as well as their effect on the oxidative response of bovine neutrophils. The production of ROS was indirectly measured using a luminol-based chemiluminescence assay. Our results confirm that Mmm can produce ROS via the metabolism of glycerol, significant differences existing between African and European strains. Mmm was capable of adhering to the external surface of neutrophils. Interestingly, Mmm enhanced the respiratory burst of bovine neutrophils. This activity was particularly pronounced with the African field strain and in presence of glycerol. Taken together, our data argue in favour of a major role for neutrophils as the main source of ROS in contagious bovine pleuropneumonia. [ABSTRACT FROM AUTHOR]

Contagious bovine pleuropneumonia is an important economic disease of cattle caused by Mycoplasma mycoides subspecies mycoides and manifested by anorexia, fever, dyspnoea, polypnoea, cough and nasal discharges. An eleven years (2006-2016) abattoir-based retrospective data were collated and analysed from Adamawa state Ministry of Livestock Production. Out of 241,700 cattle examined at post-mortem, 8,429 had CBPP-like lesions giving a prevalence of 3.49% (95% CI: 3.41-3.56). The overall prevalence for each local government area were 2.42% (95% CI: 2.35-2.49), 5.53% (95% CI: 5.30-5.74) and 8.97% (95% CI: 8.56-9.41) for Yola, Mubi and Ganye abattoirs respectively. There was significant association (p<0.05) between Local Government Areas and occurrence of CBPP. The annual highest prevalence of 5.75% (95% CI: 5.43-6.09) was recorded in 2010 with 1,128 cases and lowest prevalence of 2.43% (95% CI: 2.23-2.66) was recorded in 2007 with 505 cases. Based on season, highest prevalence rate of 3.85% (95% CI: 3.58-4.14) was recorded in February with 709 cases and lowest prevalence rate of 3.03% (95% CI: 2.80-3.28) was recorded in December with 605 cases. Both annual and monthly prevalence rates varied significantly (p<0.05). It was concluded that CBPP is endemic and widespread in Adamawa state. Hence, we recommend increased funding in the control of the disease, restriction of cattle movement and provision of cattle ranches. [ABSTRACT FROM AUTHOR]

Contagious bovine pleuropneumonia (CBPP) is an infectious disease of cattle which substantially contributes to poor productivity of the sub-Saharan pastoral livestock sector. In Nigeria and most of the West African countries, limited public funding for CBPP control have necessitated farmers to bear a bigger burden of managing the disease. Understanding the factors influencing decision of farmers to implement disease control programmes is therefore a key element in informing future policies aimed at improving CBPP management. This study explored perceptions of Nigerian Fulani pastoral herdsmen on the responsibility for cattle healthcare, and identified their circumstances and motivations in implementing CBPP management programmes. Field data were collected from 191 pastoral farmers using a semi-structured, interviewer-administered questionnaire. The results indicated that younger farmers were more likely than their older counterpart to accept the responsibility for CBPP management (p < 0.01). This may signal future prospects for improved CBPP management where upcoming farmers could be encouraged to implement CBPP control programmes and uphold the costs. 13.6% of the farmers had no intention of implementing any CBPP control programme on farm, while 81.2% either had a positive intention or implemented at least one programme aimed at controlling CBPP. Intention to implement CBPP control programmes was significantly associated with educational attainment of farmers (p < 0.01) and their access to CBPP control services offered by trained veterinarians (p < 0.01). Farmers with negative attitudes towards implementing CBPP control programmes could be motivated to change their perspectives by advice from trusted sources and improved access to veterinary services. Conversely, farmers with positive attitudes towards implementing CBPP control programmes were more likely to be motivated by affordable veterinary services and advocacy on specific CBPP control programmes. As such, the former group of farmers will be more likely to benefit from programmes which focus on providing credible information from trusted sources, such as extension agents, veterinarians or successful peers. Contagious bovine pleuropneumonia (CBPP) is a severe infectious disease caused by Mycoplasma mycoides subsp. mycoides. The peculiar pathological features of CBPP make desirable the assessment of ad hoc score methods to grade the disease in the affected animals.
