Introduction.Recently, the emphasis has been on the problems faced by performance athletes with physical deficiencies of the spine. The aim of the paper is to train coaches in awareness of physical problems, especially of the spine in swimming athletes aged 10-12 years and the development of kinetic programs on land in order to correct them. Material and method. Research on the effectiveness of kinetic programs as means of correction are very numerous and, in this regard, we aim to select the most effective exercises to correct deficiencies acquired by swimmers and change the functional parameters of the cardio-respiratory system during their training. In order to highlight the need for kinetic correction programs, the coach must work in collaboration with the sports doctor and the physiotherapist in order to prevent possible deviations from normal somatic-functional values. Results and discussions. Regarding the estimation of the adaptive possibilities of the cardio-respiratory capacity, hence the need to apply a complex of tests to assess the effort capacity of athletes. Conclusions. In this sense, we will submit to the study the performance group from the University Sports Club from Suceava, and the recovery programs will take place within the Swimming and Kinesiology Complex, Suceava.

The study included the results obtained after questioning the students from the Faculty of Physical Education and Sports, Suceava. In terms of the transition from the onsite teaching system to the online teaching system, it has raised major issues in learning and how they have been informed. In this regard, we developed a questionnaire with closed questions (with answer options) as well as open-ended questions. We believe that online teaching is a major solution in times of pandemic crisis. This crisis has highlighted many problems, which have imposed on teachers and students a radical change in behavior, mentality.

The case study presented highlights the pathological physiological genesis and evolutionary dynamics of different neurogenic diseases which in most cases can cause changes in the neuro-myo-arthro-kinetic apparatus such as functional incapacity through installed paralysis. Repercussions of neurological conditions are also harmful to the normal function and n development of the neurological child, the earlier the disease is established or the greater the complexity of the neurological pathology. Physical therapy through the variety of methods and techniques used judiciously can lead to the realization of a program for the preservation of somatic-functional structures, preventing their deconditioning

Children’s respiratory physiology during their period of growth and development, present some aspects specific to the age of the childhood period, where we have some specific characteristics regarding the respiratory rate, heart rate and the dynamics of morphological changes assigned to every period. If during children’s growth and development period the functional capacities and possible acquired deficits are ignored, through overload, the growth process is disrupted and the consequences are able to cause harm through the rest of the life regarding health status.

Introduction. In the literature, nutrition is considered to be the basis for the energy supply in all the forms of physical activity. Energy needs depend on gender, age, body weight and possibly the type of the physical activity. Inadequate energy intake can lead to weight loss, fatigue, lack of concentration and no athletic performance. The constant physical activity involves the consumption of carbohydrates, useful for the formation and storage of muscle glycogen. The development of healthy eating and physical activity behaviors in childhood and adolescence are very important for adult life. That is why an early intervention can lead to healthy behaviors and it can prevent the occurrence of overweight and obesity. Objectives. The purpose of this study was to demonstrate how important a controlled diet is correlated with regular physical activity, in the normal development of young people aged between 19 and 26 years. Conclusions. The attitude is the one that should lead to a food program with a balanced diet that ensures good health, without increasing the percentage of fat, and to cause a weight loss for the young people who want to lose weight.

It is known, certainly, that it is much easier to form a correct attitude than to correct the vicious one, a reason that must determine the approach of prophylactic means of physical therapy and therapeutic swimming. The identification, at an early stage, of the etiological factors responsible for the installation of postural deficiencies, is a determining factor in the management of programs for their maintenance or recovery in children of prepubertal age. The study took place at the Constantinescu Mihai Physiotherapy Office of Suceava and at the Cornișa Botoșani Swimming Complex. It is known that the correct position is formed from childhood, and movement and exercise are the main factors of its construction. In this sense, the paper proposes kinetic recovery and therapeutic swimming programs in order to strengthen the postural status in children during growth and development. In this regard, we have set the following objectives: identification of children with vicious postural attitudes because of the overloads in the prepubertal period and analysis of the scoliosis mechanisms in children of prepubertal age on a vulnerable somato-functional status.

The purpose of this paper is to point out and propose the most effective means of therapeutic swimming in the use of work programs for patients with dorsalgia in spinal static disorders. The knowledge degree of therapeutic swimming is not sufficiently promoted and used, which is why we propose the following hypothesis: we start from the premise that therapeutic swimming can reduce the dorsalgia in the spine, improve lung capacity and physical condition in young adults. The objectives of this work are to identify subjects with spinal static dorsalgia and to establish the sample for the experiment; in the selection of therapeutic swimming means for the elaboration of the work program; in the registration and interpretation of the data in the form of tables and charts. The experiment took place at the Kinetic Therapy Practice of Mr. Constantinescu Mihai, as well as at the swimming pool in Cornisa, Botoşani county, with a sample consisting of 15 subjects aged between 18 and 22, all of whom suffered dorsalgia (in the spine). In this respect, we set out to implement a therapeutic swimming program for 6 months to achieve positive results.

Due to the accelerated process of growth during the pre-puberty regular physical functional deficiencies known find the proper field for the onset and evolution. The muscle - skeletal disorders become more common in the Children and teenagers in schools. If it is not detected in time, will it lead to severe damage of the process of growth and harmonious development of the spine. With minimal effort from the educational factors like family, teachers, teachers in physical education, these deficiencies could be timely detected and corrected by kinesiology.

The current article presents several general aspects based on a research in progress and desires argument the necessity for the paraclinical investigation, respectively imagistic examination of the type of radiography for the establishment of the positive diagnosis in the treatment of functional physical deficiencies, of scoliosis in particular.

Background: Ischemia/reperfusion injury (IRI) is a complex pathological process, triggered by the restoration of blood flow following an interrupted blood supply. While restoring the blood flow is the only option to salvage the ischemic tissue, reperfusion after a prolonged period of ischemia initiates IRI, triggering a cascade of inflammatory responses ultimately leading to neutrophil recruitment to the inflamed tissue, where they release neutrophil extracellular traps (NETs). NETs are web-like structures of decondensed chromatin and neutrophilic proteins, including peptidyl-arginine deiminase 2 and 4 (PAD2, PAD4), that, once outside, can citrullinate plasma proteins, irreversibly changing their conformation and potentially their function. While the involvement of NETs in IRI is known mainly from rodent models, we aimed to determine the effect of NET formation and especially PADs-mediated extracellular protein citrullination in a porcine model of limb IRI. Methods: We conducted our study on amputated pig forelimbs exposed to 1 h or 9 h of ischemia and then reperfused in vivo for 12 h. Limb weight, edema formation, compartmental pressure were measured, and skeletal muscle was analyzed by immunofluorescence (TUNEL assay and dystrophin staining) to evaluate tissue damage. Fibrin tissue deposition, complement deposition and NETs were investigated by immunofluorescence. Citrullinated plasma proteins were immunoprecipitated and citrullinated fibrinogen was identified in the plasma by Western blot and in the tissue by immunofluorescence and Western blot. Results: Our data consolidate the involvement of NETs in a porcine model of limb IRI, correlating their contribution to damage extension with the duration of the ischemic time. We found a massive infiltration of NETs in the group subjected to 9 h ischemia compared to the 1 h and citrullinated fibrinogen levels, in plasma and tissue, were higher in 9 h ischemia group. We propose fibrinogen citrullination as one of the mechanisms contributing to the worsening of IRI. NETs and protein citrullination represent a potential therapeutic target, but approaches are still a matter of debate. Here we introduce the idea of therapeutic approaches against citrullination to specifically inhibit PADs extracellularly, avoiding the downstream effects of hypercitrullination and keeping PADs' and NETs' intracellular regulatory functions. Simple Summary: A positive sentinel lymph node biopsy of cutaneous melanoma patients has a substantial impact on subsequent treatment decisions. The standard of care approach to identify the sentinel lymph node is technetium (Tc)-based lymphoscintigraphy. This technique comes with a radiation exposure and high costs. Indocyanine green (ICG)-based near-infrared fluorescence imaging could be an alternative if demonstrated to have a comparable diagnostic accuracy. Therefore, a systematic literature review and meta-analysis were conducted considering studies comparing the accuracy of ICG and Tc for intraoperative guidance. Within the seven included studies, no significant differences between the two modalities were found regarding the identification of metastatic patients or the false negative rate. ICG may be a non-inferior alternative to Tc for intraoperative identification of the sentinel lymph node in cutaneous melanoma patients. The standard of care approach to identify sentinel lymph nodes (SLNs) in clinically non-metastatic cutaneous melanoma patients is technetium (Tc)-based lymphoscintigraphy. This technique is associated with radiation exposure, a long intervention time, high costs, and limited availability. Indocyanine green (ICG)-based near-infrared fluorescence imaging offers a potential alternative if proven to be of comparable diagnostic accuracy. While several clinical cohorts have compared these modalities, no systematic review exists that provides a quantitative analysis of their results. Hence, a systematic literature review was conducted in December 2023 considering clinical studies comparing the diagnostic accuracy of ICG and Tc for sentinel lymph node biopsy in cutaneous melanoma patients. Three hundred nineteen studies were identified and further screened in accordance with the PRISMA 2020 guidelines, resulting in seven studies being included in the final meta-analysis. Tc identified a significantly higher number of SLNs and metastatic SLNs in prospective studies only. However, in the overall meta-analysis of all included comparative studies, no significant differences were found regarding the identification of metastatic patients or the false negative rate (FNR). ICG may be a non-inferior alternative to Tc for intraoperative guidance in sentinel lymph node biopsy in cutaneous melanoma patients. Future randomized controlled trials are needed, especially regarding the preoperative, transcutaneous identification of the affected lymph node basin. [ABSTRACT FROM AUTHOR]

Introduction: The standard treatment for preventing rejection in vascularized composite allotransplantation (VCA) currently relies on systemic immunosuppression, which exposes the host to well-known side effects. Locally administered immunosuppression strategies have shown promising results to bypass this hurdle. Nevertheless, their progress has been slow, partially attributed to a limited understanding of the essential mechanisms underlying graft rejection. Recent discoveries highlight the crucial involvement of innate immune components, such as neutrophil extracellular traps (NETs), in organ transplantation. Here we aimed to prolong graft survival through a tacrolimus-based drug delivery system and to understand the role of NETs in VCA graft rejection. Methods: To prevent off-target toxicity and promote graft survival, we tested a locally administered tacrolimus-loaded on-demand drug delivery system (TGMS-TAC) in a multiple MHC-mismatched porcine VCA model. Off-target toxicity was assessed in tissue and blood. Graft rejection was evaluated macroscopically while the complement system, T cells, neutrophils and NETs were analyzed in graft tissues by immunofluorescence and/or western blot. Plasmatic levels of inflammatory cytokines were measured using a Luminex magnetic-bead porcine panel, and NETs were measured in plasma and tissue using DNA-MPO ELISA. Lastly, to evaluate the effect of tacrolimus on NET formation, NETs were induced in-vitro in porcine and human peripheral neutrophils following incubation with tacrolimus. Results: Repeated intra-graft administrations of TGMS-TAC minimized systemic toxicity and prolonged graft survival. Nevertheless, signs of rejection were observed at endpoint. Systemically, there were no increases in cytokine levels, complement anaphylatoxins, T-cell subpopulations, or neutrophils during rejection. Yet, tissue analysis showed local infiltration of T cells and neutrophils, together with neutrophil extracellular traps (NETs) in rejected grafts. Interestingly, intra-graft administration of tacrolimus contributed to a reduction in both Tcellular infiltration and NETs. In fact, in-vitro NETosis assessment showed a 62– 84% reduction in NETs after stimulated neutrophils were treated with tacrolimus. Conclusion: Our data indicate that the proposed local delivery of immunosuppression avoids off-target toxicity while prolonging graft survival in a multiple MHC-mismatch VCA model. Background: Vascularized composite allotransplantation (VCA) offers the potential for a biological, functional reconstruction in individuals with limb loss or facial disfigurement. Yet, it faces substantial challenges due to heightened immune rejection rates compared to solid organ transplants. A deep understanding of the genetic and immunological drivers of VCA rejection is essential to improve VCA outcomes Methods: Heterotopic porcine hindlimb VCA models were established and followed until reaching the endpoint. Skin and muscle samples were obtained from VCA transplant recipient pigs for histological assessments and RNA sequencing analysis. The rejection groups included recipients with moderate pathological rejection, treated locally with tacrolimus encapsulated in triglycerol-monostearate gel (TGMS-TAC), as well as recipients with severe end-stage rejection presenting evident necrosis. Healthy donor tissue served as controls. Bioinformatics analysis, immunofluorescence, and electron microscopy were utilized to examine gene expression patterns and the expression of immune response markers. Results: Our comprehensive analyses encompassed differentially expressed genes, Gene Ontology, and Kyoto Encyclopedia of Genes and Genomes pathways, spanning various composite tissues including skin and muscle, in comparison to the healthy control group. The analysis revealed a consistency and reproducibility in alignment with the pathological rejection grading. Genes and pathways associated with innate immunity, notably pattern recognition receptors (PRRs), damage-associated molecular patterns (DAMPs), and antigen processing and presentation pathways, exhibited upregulation in the VCA rejection groups compared to the healthy controls. Our investigation identified significant shifts in gene expression related to cytokines, chemokines, complement pathways, and diverse immune cell types, with CD8 T cells and macrophages notably enriched in the VCA rejection tissues. Mechanisms of cell death, such as apoptosis, necroptosis and ferroptosis were observed and coexisted in rejected tissues. Conclusion: Our study provides insights into the genetic profile of tissue rejection in the porcine VCA model. We comprehensively analyze the molecular landscape of immune rejection mechanisms, from innate immunity activation to critical stages such as antigen recognition, cytotoxic rejection, and cell death. This research advances our understanding of graft rejection mechanisms and offers potential for improving diagnostic and therapeutic strategies to enhance the long-term success of VCA. Background. The study aims to perform a somatoscopic analysis on a sample of 100 prepubertal children aged 10-14 years, girls and boys, who are diagnosed with scoliosis. Somatoscopic assessment is one of the most commonly used, therefore we thought that making a way of interpreting the results could be useful to monitor postural attitude in children diagnosed with scoliosis. Identifying the early onset of physical impairment in the spine with somatoscopy can be a key factor in maintaining optimal postural status. This approach may help in achieving a way of working that limits the negative effects of scoliosis on the subjects. Materials and methods. The selection of subjects is based on exclusion and inclusion criteria related to age, comorbidities, therapeutic approach, and other criteria. Also, aspects that accompany the growth and development process of children in this prepubertal period and factors that may disturb this process are described in detail. Also in this context, we will present basic notions of body posture and etiopathogenesis of the onset of scoliotic deficiencies in children during this period of growth and development. Somatoscopic analysis is performed in the orthostatic position and is based on the identification of anatomical landmarks of the body concerning body posture. Results. From the total analyzed scoliosis, n = 100, 74% are type „C" scoliosis and only 26% are type „S" and in terms of gender, 59% are girls and 41% boys; type „C1" (Cobb angle <100) scoliosis n = 60, with an angulation < 100 are 43.24% stabilized, therefore type „C1" scoliosis are more likely to be stabilized. „S"-type scoliosis with an angulation < 150 is only 3 cases representing 11.53%, and those with an angulation from < 200 to < 400 are number 23 cases 88.64% which concludes that „S" type scoliosis is more aggressive and more difficult to manage. Conclusions. Our study show that only 13% of the subjects had a regression of the angulation following the kinetotherapy program, therefore the main objective remains to stabilize the scoliosis attitude and limit the effects of this pathology. A regular assessment and somatotopic analysis resulting in a physiotherapy exercises program containing corrective postural postures and postural education is the most beneficial approach to stabilize the postural deficit. Background: Total hip arthroplasty (THA) is a procedure that removes damaged bone and cartilage and replaces it with prosthetic components and is performed by orthopedics. Objective: management of post-operative rehabilitation of THA patients in terms of content and completion criteria of physiotherapy sessions. The research aims to explore the experience of Romanian physiotherapists about the content and completion criteria of physiotherapy sessions involved in the management of postoperative rehabilitation after THA. Methods: The method used in this study was a questionnaire-based survey, with 120 Romanian specialists in the field completing an online screening tool that contained, along with a socio-demographic questionnaire, a series of questions about their professional experience. Results: One of the most important results of the study is the identification of statistically significant associations between the completion criteria of physiotherapy sessions and three techniques included in the rehabilitation program after THA: edema management, gait re-education exercises, and exercises to increase hip mobility. Conclusion: The the research revealed that the experience of Romanian specialists regarding postoperative rehabilitation after THA is concordant with the experience of specialists in the field, and can be used to develop and test an individualized therapeutic protocol and to design continuing professional training programs for physiotherapists in Romania.
