Improving the synchronization between the pattern of milk synthesis and nutrient availability throughout the day could enhance production efficiency. In this study, we evaluated the effects of changing feed delivery time on milk production, feeding behavior, and the daily rhythms of blood biomarkers. Eight multiparous Holstein cows housed in a tie-stall barn with controlled environmental conditions were enrolled in a crossover experimental design with three periods of 14 days and three treatments each. Cows were milked twice daily (0530 and 1730 h) and were individually fed with two equal meals of forage and eight equal meals of concentrate during the day. Forage meals were provided at 12-hour intervals either: (i) 5 h before each milking (0030 and 1230 h; BM), (ii) at the end of each milking (0530 and 1730 h;ME), (iii) or 2 h after (0730 and 1930 h; AM). Feed intake and feeding behavior were monitored, and milk production and composition were measured. Blood samples were collected every 4 days at 0700 h and, during the last day of each period, 15 times daily to determine metabolic profiles, hormones, and their daily rhythmicity by the cosinor analysis. Changing forage delivery time did not affect milk yield and dry matter intake. No difference was observed in feeding behavior when expressed relative to the first meal. There were no significant differences in milk component contents and yields. In samples collected at 0700 h, ME had reduced plasma calcium (Ca), magnesium (Mg), and potassium (K) and increased sodium (Na). AM had increased inflammation, as suggested by the greater blood globulin and ceruloplasmin. The patterns of metabolic biomarkers had limited variations when expressed relative to the first forage meal. Nevertheless, the daily rhythms of these biomarkers were remarkably different. Under our conditions, feeding forage meals to cows at different times of the day did not influence productive performance but highlighted the importance of considering the sampling time when interpreting metabolic profiles. Bactrian camel (BC) milk has gained increasing attention due to its unique nutritional profile and potential bioactivities. This comprehensive review explores the chemical composition, bioactivities, processing techniques, and economic potential of BC milk in China. The distinctive chemical composition of BC milk, including protein, lipid, carbohydrate, vitamin, and mineral content, is discussed, emphasizing its differences from other mammalian milk. The review highlights the various bioactivities of BC milk, such as anti-inflammatory, antidiabetic, lipid-lowering, and anticancer properties, as well as its modulatory effects on intestinal microbiota. The technological properties of BC milk, focusing on its heat stability, coagulation behavior, and potential for product development, are examined. The review also addresses current processing techniques and their impact on milk quality. Finally, the economic potential and future perspectives of BC milk in China are evaluated. This review provides valuable insights into the multifaceted aspects of BC milk, serving as a foundation for future research and development in this emerging field. The motivation for this review stems from the growing interest in BC milk as a functional food and the need for a comprehensive understanding of its properties, applications, and market potential to guide future research and industry development. Simple Summary: Fourier Transform Infrared Spectroscopy (FT-MIRS) is widely used in milk quality detection, dairy herd improvement (DHI), and other fields. It is an economical, fast, accurate, and nondestructive batch tool for determining production performance phenotypes of dairy cows. Milk is one of the most important ways to provide the human body with the fatty acids it needs. There are a huge number of dairy cows in China. Therefore, it is possible to control fatty acid production in the milk source through targeted husbandry and breeding on large pastures to improve the quality of milk production. However, this work has not yet officially begun in China. In summary, our work uses FT-MIRS for the first time to study the phenotypic properties of milk fatty acid content and the genetic mechanism of its formation and to estimate genetic parameters. At the same time, SNPs significantly related to fatty acid content were discovered and the genes or adjacent genes had critical regulatory effects on milk fat synthesis, milk protein synthesis, adipocyte differentiation, mammary gland development, milk synthesis, and growth and development in dairy cows, thus providing a new perspective for cow genetic selection in China. Fourier Transform Mid-Infrared Spectroscopy (FT-MIRS) can be used for quantitative detection of milk components. Here, milk samples of 458 Chinese Holstein cows from 11 provinces in China were collected and we established a total of 22 quantitative prediction models in milk fatty acids by FT-MIRS. The coefficient of determination of the validation set ranged from 0.59 (C18:0) to 0.76 (C4:0). The models were adopted to predict the milk fatty acids from 2138 cows and a new high-throughput computing software HiBLUP was employed to construct a multi-trait model to estimate and analyze genetic parameters in dairy cows. Finally, genome-wide association analysis was performed and seven novel SNPs significantly associated with fatty acid content were selected, investigated, and verified with the FarmCPU method, which stands for "Fixed and random model Circulating Probability Unification". The findings of this study lay a foundation and offer technical support for the study of fatty acid trait breeding and the screening and grouping of characteristic dairy cows in China with rich, high-quality fatty acids. Simple Summary: This study reports the results of two studies aiming to determine, under commercial farm conditions, the effects of electronic sow feeders on the production and economic yields of lactating sows. The results indicate remarkable technical and economic outputs compared with traditional feeders due to the weaning of heavier piglets with a lower amount of feed per kg of weaned piglet. Adequate nutritional management in maternities is one of the most challenging aspects of swine production. This study reports the results of two studies aiming to determine, under commercial farm conditions, the effects of precision feeding (electronic sow feeders, ESFs) on the production and economic yields of lactating sows and possible nutritional and metabolic differences when compared to a control group fed with traditional feeders. The first trial showed that sows fed with ESFs weaned heavier piglets than sows fed with traditional feeders. Feed intake during the lactation period was similar in the sows of both groups; consequently, the amount of feed per kg of weaned piglet was lower in the sows fed with ESFs, which is a remarkable economic output. The second trial confirmed these findings and showed that, despite similar feed intakes, the sows fed with ESFs had lower bodyweight losses during the lactation period, but there were no major differences in milk composition or metabolic traits of sows and piglets. Simple Summary: This study evaluated the effects of heat stress on the productivity of dairy cows. During the experimental period, THI values ranged from 46.9 to 81.0, with the most severe heat stress occurring in August. A decrease in productivity was observed when the THI exceeded 72. It was found that milk production took about one month to recover after the high-temperature period ended. The THI ratio, calculated based on the daily THI distribution, showed a strong negative correlation with rumen temperature (− 0.744; p < 0.001). During periods of significant productivity decreases due to high-temperature stress, rumen temperatures were higher than 39.15 °C. Based on the rumen temperature and daily THI distribution, it is suggested that these findings could serve as foundational data for developing technologies to manage high-temperature environments for dairy cows. This study was conducted to evaluate the effects of high-temperature stress on dairy cow productivity and the correlation between rumen sensors. The data were collected on the temperature, humidity, milk productivity, milk components, blood components, and rumen sensor data from 125 dairy cows during the experimental period (1 May 2020 to 30 October 2020). High-temperature stress of dairy cows was evaluated based on the temperature–humidity index (THI). The correlations between the high-temperature stress, productivity, and sensor data were analyzed using SAS and R programs. The THI ranged from 46.9 to 81.0 during the experimental period, and a significant decrease was observed in the milk production of dairy cows during August (p < 0.05). Milk production was evaluated to decrease by 1.8% because of high-temperature stress during the experimental period. There was a significantly high negative correlation between the THI ratio of day and rumen temperature (r = 0.744; p < 0.001). The other rumen sensor data did not show a significant correlation with the productivity of dairy cows. Subclinical mastitis (SCM) is a serious production and financial health issue facing small ruminant farms. Shami dairy does, and Rahmani ewes were chosen for this study. The udder dimensions of the test animals were evaluated, while the taken milk samples were analysed for chemical composition and the somatic cell count (SCC). The presence milk lactate dehydrogenase (LDH) enzyme in milk samples, haemogram and leukogram were determined, too. The incidence of SCM in goats and sheep was 53.33 % and 41.67 %, respectively. In both ovine and caprine milk, there was an increase in LDH concerning the increase in SCC. There was a significant difference (p<0.05) between the decrease in milk fat and lactose contents (%) and the high SCC. The coagulase-negative staphylococci in sheep presented a significant difference in the high protein and low fat content, asymmetrical udder, low udder depth, and increased LDH. SCM in dairy ewes was detected with a rising serum C-reactive protein (CRP), while SCM in goats caused by S. aureus was associated to a reduction in lactose and fat but a high LDH, with a weak udder attachment. Nutritional strategies that improve an animal's resilience to various challenges may improve animal health and welfare. One such nutrient is niacin, which has reduced inflammation in mice, humans, and swine; however, niacin's anti-inflammatory effects have not been investigated in cattle. Our objective was to determine whether rumen-protected niacin (RPN) alters lactating dairy cows' inflammatory response to intramammary LPS challenges, whether RPN resulted in any carryover effects, and whether repeated LPS challenges result in signs of immune tolerance or innate immune training. Twenty healthy, late-lactation Holstein cows (232 ± 65 DIM; 39 ± 5.8 kg/d of milk) were enrolled in a randomized complete block experiment that lasted 70 d. Cows received 26 g/d of RPN or no top-dress (CON) for the first 42 d of the experiment. During the final milking of d 27 and 55, cows were challenged in their rear right (RR) mammary gland with 100 µg of LPS suspended in 5 mL of PBS. Milk yield, milk conductivity, and feed intake were measured daily. Milk composition was measured on d 14, 23, 24, 30, 37, 45, and 52. Blood samples were collected at 0, 8, 12, 24, 48, 72, 96, and 120 h after each LPS challenge, whereas RR quarter milk samples were collected at 0, 8, 16, 24, 48, 72, 96, 120, 144, and 168 h after each LPS challenge. Body temperature was measured continuously during each challenge with an intravaginal thermometer. Linear mixed models with repeated measures were used to analyze the results. Before LPS challenge, RPN did not affect feed intake or milk production, but it reduced SCS (1.24 ± 0.41 [SE] vs. 0.05 ± 0.45). After challenge, RPN did not affect feed intake, milk production, milk composition, SCS, body temperature, plasma glucose, or plasma insulin concentrations. Our results suggest RPN reduced peak plasma haptoglobin and lipopolysaccharide binding protein during the first LPS challenge. Plasma haptoglobin tended to be less after the second challenge for cows previously supplemented RPN, and lipopolysaccharide binding protein was similar for each treatment group after the second challenge. The second LPS challenge resulted in decreased plasma haptoglobin compared with the first LPS challenge, suggestive of tolerance, but it also induced a greater peak SCS than the first LPS challenge. Our results suggest that repeated LPS challenges promote a systemic tolerance but heightened local response to LPS-induced mastitis. Feeding RPN reduced SCS before challenge and reduced plasma acute phase proteins after challenge, suggesting that RPN may reduce systemic inflammation without altering the local inflammatory responses. The objectives of this study were to determine the effects of 2 dietary microbial additives on productive performance and feed efficiency when supplemented to diets of Holstein cows. One hundred seventeen Holstein cows were enrolled at 61 d (31–87 d) postpartum in a randomized complete block design experiment. Cows were blocked by parity group, as nulliparous or multiparous cows and, within parity, by pretreatment ECM yield. Within block, cows were randomly assigned to one of 3 treatments administered as top-dress for 140 d. Treatments consisted of either 100 g of corn meal containing no microbial additive (CON; 15 primiparous and 25 multiparous), 100 g of corn meal containing 5 g of a mixture of Clostridium beijerinckii and Pichia kudriavzevii (G1; 4 × 107 cfu of C. beijerinckii and 1 × 109 cfu of P. kudriavzevii ; 14 primiparous and 24 multiparous), or 100 g of corn meal containing 5 g of a mixture of C. beijerinckii , P. kudriavzevii , Butyrivibrio fibrisolvens , and Ruminococcus bovis (G2; 4 × 107 cfu of C. beijerinckii, 1 × 109 cfu of P. kudriavzevii, 1 × 108 cfu of B. fibrisolvens , and 1 × 108 cfu of R. bovis ; 15 primiparous and 24 multiparous). Intake of DM, milk yield, and BW were measured daily, whereas milk composition was analyzed at each milking 2 d a week, and body condition was scored twice weekly. Milk samples were collected on d 60 and 62 in the experiment and analyzed for individual fatty acids. The data were analyzed with mixed-effects models with orthogonal contrast to determine the effect of microbial additive (MA; CON vs. 1/2 G1 + 1/2 G2) and type of microbial additive (TMA; G1 vs. G2). Results are described in sequence as CON, G1, and G2. Intake of DM (22.2 vs. 22.4 vs. 22.4 kg/d), BW (685 vs. 685 vs. 685 kg) and the daily BW change (0.40 vs. 0.39 vs. 0.39 kg/d) did not differ among treatments; however, feeding MA tended to increase BCS (3.28 vs. 3.33 vs. 3.36). Supplementing MA increased yields of milk (39.9 vs. 41.3 vs. 41.5 kg/d), ECM (37.9 vs. 39.3 vs. 39.9 kg/d), fat (1.31 vs. 1.37 vs. 1.40 kg/d), TS (4.59 vs. 4.75 vs. 4.79 kg/d), and ECM per kg of DMI (1.72 vs. 1.76 vs. 1.80 kg/kg). Cow milk microbiota has received increased attention in recent years, not only because of its importance for human health but also because of its effect on the quality and technological properties of milk. Several studies, therefore, have investigated the effect of various production factors on the microbial composition of milk. However, most of the previous studies considered a limited number of animals from experimental or single farm, which could have biased the results. Therefore, this study aimed to understand the effect of different alpine production systems in Italy on the compositional and microbiological quality of milk, considering commercial herds with different feeding intensities and cattle breeds. The results obtained in this work indicated that the month and season of sampling (July for summer or February for winter) more than farm, breed, and cow diet exerted significant effects on cow milk parameters and microbiota. In particular, significant differences were observed for urea content in milk between sampling seasons. Differences in milk fat were mainly related to breed specific effects. From a microbiological point of view, statistically significant differences were found in presumptive lactic acid bacteria counts. Based on a culture-independent method, milk obtained in February harbored the highest number of Firmicutes (e.g., Lactobacillus) and the lowest number of Actinobacteria (e.g., Corynebacterium). Moreover, bacterial richness and diversity were higher in July during alpine pasture season indicating a significant effect of pasture feed on the growth of bacterial communities. The results of this study highlighted the effect of month or season mainly related to differences in feeding management (e.g., access to pasture during vegetation period, concentrates supplementation) on composition and microbiota in milk. Mare milk has a unique protein composition that makes it a preferred option for adult and infant nutrition. Several functional properties have been attributed to this milk, but with little current evidence. In fact, knowledge on mare milk composition is still limited. In particular, studies addressing the performance of mare milk proteins during human gastrointestinal digestion are scarce, which limits the understanding of mare milk nutritional quality and functionality. For this reason, the present study describes the digestibility of mare milk proteins and the release of peptides as affected by management and lactation stage, factors which are known to affect milk composition. Mare milk samples from 3 different farms collected during 6 mo of lactation (n = 54) were subjected to a static in vitro gastrointestinal model to measure peptide release and protein digestibility. In the present study, a detailed description of protein and individual amino acid behavior during the digestion process was given. For the first time, digestion of the 2 equine β-lactoglobulin isoforms (I and II) was described individually. In addition, it was found that lactation stage and management system can significantly affect protein digestibility and peptide release during gastrointestinal digestion of mare milk. Presumably, differences in the composition of mare milk influence the protein structure and enzyme accessibility, which might have an impact on digestion behavior. Although no specific bioactive peptides were identified, several precursors of previously described bioactive peptides were found. Determination of energy requirements for growth depends on measuring the composition of BW gain. Previous studies have shown that the composition of gain can be altered in young dairy calves by the composition of the milk replacer diet. Here, our objective was to determine body composition and the composition of empty body gain in young calves fed increasing amounts of a milk replacer containing adequate CP. Male Holstein calves underwent an adjustment period of 14 d after birth in which they were fed whole-waste milk at 10% of BW. Calves were then stratified by BW and randomly assigned to either an initial slaughter group (n = 11) or to groups fed 1 of 3 milk replacer amounts and slaughtered after 35 d of growth. Calves in all treatments consumed the same milk replacer containing 24.8% CP (DM basis; from all milk proteins) and 18.9% fat, reconstituted to 12.5% solids. Treatments were milk replacer fed at 1.25% of BW (DM basis; n = 6), 1.75% of BW (n = 6), or 2.25% of BW (n = 8), adjusted weekly as calves grew. Calves fed at 1.25% or 1.75% of BW were fed twice daily and those fed 2.25% of BW were fed 3 times daily. No starter was offered. Postslaughter, the bodies of calves were separated into 4 fractions: carcass; total viscera minus digesta; head, hide, feet, and tail;, and blood. The sum of those 4 fractions was empty BW (EBW), which increased linearly as the amount of milk replacer increased. Final heart girth and body length, but not withers height, increased linearly as intake increased. Gain:feed increased linearly with increasing milk replacer. Feeding more milk replacer increased the amounts of lean tissue and fat in the body. The percentages of water and protein in the final body decreased linearly, whereas fat percentage and energy content increased linearly as intake increased. As gain increased, the percentage of protein in gain decreased and the percentage of fat increased, resulting in an increase of energy content of EBW gain. Efficiency of energy use (retained energy:gross energy intake) increased linearly but retained energy:ME available for growth was not different among treatments. Efficiency of protein use increased quadratically as feeding rate increased; there was no further increase at 2.25% of BW. Plasma insulin-like growth factor 1, insulin, and glucose increased linearly, whereas urea-N decreased linearly, as milk replacer intake increased. Our data document changes in body composition that affect estimates of retained energy in the bodies of calves slaughtered at a common age. ABSTRACT Previous studies have suggested that maternal childhood trauma (MCT) may influence infant temperament, but the underlying physiological mechanisms remain unclear. This study sought to confirm the involvement of breast milk cortisol in the link between MCT and infant temperament. The study sample included 90 mother–infant dyads recruited from the urban Polish population. MCT was assessed based on the Early Life Stress Questionnaire (ELSQ) and infant temperamental factors (surgency/extraversion, negative affectivity, and orienting/regulation) using the Infant Behavior Questionnaire‐Revised at 12 months of life. Cortisol was assayed in milk samples collected at 5 months of life using the ELISA method. Based on the ELSQ median, the sample was divided into low and high MCT groups. The ANCOVA models with milk cortisol as a covariant were run to check the effect of low versus high MCT on infant temperament. We found a positive association between milk cortisol and orienting/regulation. Surprisingly, the low and high MCT groups did not significantly differ in milk cortisol. Furthermore, we found that MCT was unrelated to any infant temperamental factor. While recent literature on the association between milk cortisol and infant temperament is inconsistent, our results suggest that high orienting/regulation might be an adaptation to adverse environments such as stress. Moreover, the infant's temperament appears to be more responsive to the current exposition to maternal stress than her experience of traumatic stress. [ABSTRACT FROM AUTHOR]

In breastfeeding mothers, managing medical conditions presents unique challenges, particularly concerning medication use and breastfeeding practices. The transfer of drugs into breast milk and subsequent exposure to nursing infants raises important considerations for drug safety and efficacy. Modeling approaches are increasingly employed to predict infant exposure levels, crucial for assessing drug safety during breastfeeding. Physiologically‐based pharmacokinetic (PBPK) modeling provides a valuable tool for predicting drug exposure in lactating individuals and their infants. This tutorial offers an overview of PBPK modeling in lactation research, covering key concepts, prediction approaches, and best practices for model development and application. We delve into milk composition dynamics and its influence on drug transfer into breast milk, addressing modeling considerations, knowledge gaps, and future research directions. Practical examples and case studies illustrate PBPK modeling application in lactation studies. We demonstrate how prediction algorithms for Milk‐to‐Plasma (M/P) ratios within a PBPK framework can support scenarios lacking clinical lactation data or extend the utility of available lactation clinical data to support further untested clinical scenarios. This tutorial aims to assist researchers and clinicians in understanding and applying PBPK modeling to understand and support clinical scenarios in breastfeeding mothers. Background: Proportion of roughage and concentrate supply to dairy animal is a major challenge in commercial dairying of Bangladesh that directly correlated with quality and quantity of milk. Several studies revealed that, level of milk urea nitrogen (MUN) is an indication of proper feed supply of the animals. Therefore, the study was the attempt to find, the relationship between the level of MUN with metabolic energy (ME) and crude protein (CP) by supplied ratio of roughage, concentrate and effect of altered level MUN with milk quality and quantity in commercial dairy farms of Chittagong, Bangladesh. Methods: For the study, feed and milk samples were taken from randomly selected three different farm categories for the period of July 2014 to June 2015 monthly. The herd average milk production was taken from the recorded data. The feed and milk composition were analyzed in the dairy lab of Chittagong Veterinary and Animal Sciences University (CVASU). MUN was determined by spectrophotometric method in the Poultry Research and Tanning Laboratory (PRTC) of CVASU, Chittagong, Bangladesh. Observed data was analyzed by using Pearson's correlation methods to find out the correlation among MUN level, supplied source of ME and CP and milk production and composition. Result: The results suggested that there was significant level difference of MUN (P<0.05) irrespective of seasons and farm categories with the sources of ME and CP of supplied feed. Milk yield relate significantly (P<0.05) with MUN level when there was lowest supply of roughage and maximum M.E comes from concentrate feed irrespective of farm categories. So, determination of MUN level will be helpful to commercial dairy farmers when the issues related to milk quality and quantity. [ABSTRACT FROM AUTHOR]

Objective. To evaluate the physicochemical composition and nutritional value of goat milk produced in three different small-scale production management systems in Merida, Yucatan in Mexico, to produce milk for human consumption for people with lactose intolerance problems. Materials and methods. Samples were collected every two weeks for four months, and their fat, protein, lactose, total solids, ash, calcium, and phosphorus contents were quantified. Density, pH, acidity, fatty acid profiles by gas chromatography, and the electrophoretic pattern of caseins were determined. Results. The average composition was 2.69% fat, 4.10% protein, 4.12% lactose, 12.96% total solids, 0.83% ash, 139 mg/100g calcium, and 105 mg/100g phosphorus. The density, acidity as lactic acid, and pH obtained were 1.032 g/cm3, 1.36 g/L, and 6.51, respectively. The milk provided MUFAs, such as oleic and gadoleic acids, and PUFAs, such as linoleic and arachidonic acid, involved in metabolic processes that reduce the risk of metabolic syndrome. The electrophoretic pattern of the caseins showed more abundant bands, with molecular weights of 20 to 36 kDa. Conclusions. Background: Immunomodulatory proteins in human milk (HM) can shape infant immune development. However, strategies to modulate their levels are currently unknown. This study investigated whether maternal prebiotic supplementation alters the levels of immunomodulatory proteins in HM. Methods: The study was nested within the SYMBA double‐blind randomized controlled trial (ACTRN12615001075572), which investigated the effects of maternal prebiotic (short‐chain galacto‐oligosaccharides/long‐chain fructo‐oligosaccharides) supplementation from <21 weeks gestation during pregnancy until 6 months postnatal during lactation on child allergic disease risk. Mother–child dyads receiving prebiotics (n = 46) or placebo (n = 54) were included in this study. We measured the levels of 24 immunomodulatory proteins in HM collected at 2, 4, and 6 months. Results: Cluster analysis showed that the overall immunomodulatory protein composition of milk samples from both groups was similar. At 2 months, HM of prebiotic‐supplemented women had decreased levels of TGF‐β1 and TSLP (95% CI: −17.4 [−29.68, −2.28] and −57.32 [−94.22, −4.7] respectively) and increased levels of sCD14 (95% CI: 1.81 [0.17, 3.71]), when compared to the placebo group. At 4 months, IgG1 was lower in the prebiotic group (95% CI: −1.55 [−3.55, −0.12]) compared to placebo group. Conclusion: This exploratory study shows that prebiotic consumption by lactating mothers selectively alters specific immunomodulatory proteins in HM. This finding is crucial for understanding how prebiotic dietary recommendations for pregnant and lactating women can modify the immune properties of HM and potentially influence infant health outcomes through immune support from breastfeeding. Background The AnticFast® Beta-lactams & Tetracyclines Combo Test Kit is a qualitative two-step (2 min + 5 min) rapid lateral flow assay to detect β-lactam (penicillins and cefalosporins) and tetracycline antibiotic residues in raw commingled cows' milk. Objective The method performance was evaluated according to Commission Implementing Regulation 2021/808 and Community Reference Laboratories Residues Guidelines for the Validation of Screening Methods for Residues of Veterinary Medicines and submitted for AOAC Performance Tested Methods SM certification. Methods The AnticFast Beta-lactams & Tetracyclines Combo Test Kit was evaluated for detection capability (CCβ), selectivity, false-positive results, repeatability, robustness, suitability for various milk types and milk compositions, milks from various species, and test kit consistency and stability. Samples included milks spiked at concentrations bracketing the EU maximum residue limits (MRLs) for β-lactams and tetracyclines as well as bulk farm and tanker milks. Results The AnticFast Beta-lactams & Tetracyclines Combo Test Kit is specific for the detection of residues of β-lactams and tetracyclines in milk and does not detect residues from other antibiotic families. Interference was seen with clavulanic acid, a β-lactamase inhibitor, which was expected. The test can detect (minimum a 95% detection) all residues of β-lactams (penicillins and cepfalosporins) and tetracyclines (parent drugs and their 4-epimers) present on the EU-MRL list for milk at their respective MRL except for desfuroylceftiofur and cefalexin, with a 95% detection only above the MRL. No false positives were detected in 599 blank samples (out of 305 farm and 301 tanker load samples) tested on both channels. Five real positives were detected and confirmed on the tetracycline channel for the farm milk samples, and two positives were detected and confirmed on the β-lactam channel for the tanker samples. Robustness testing indicated that the detection in high-protein raw cows' milk and heat-treated milk types (UHT, sterilized, and reconstituted milk powder) may be slightly hampered. For substances with a detection capability well below the MRL, this interference does not cause problems since detection at MRL remains guaranteed, but care should be taken for substances with a CCβ at or near their MRL. Diminished sample flow was seen for high-fat raw cows' milk and for all other cows' milk types other than raw milk and blank ewes' milk, so sample flow should always be verified for these milk types. Conclusions Results of this validation show that the AnticFast Beta-lactams & Tetracyclines Combo Test Kit is a reliable test for rapid screening of raw cows' milk for residues of β-lactam and tetracycline antibiotics. Objective: We investigated the effects of outdoor access for one-hour per day (ODA) on milk yield and composition and behaviors of tethered dairy cows. Methods: Eleven all-day tethered dairy cows were treated with ODA for two weeks. To evaluate the effect of ODA on milk yield, we first calculated the average daily milk yield of each cow for three days during two weeks before the ODA, three days before the ODA, three days at the end of the ODA, and three days during two weeks after the ODA. We then compared the milk yield change during the ODA with that for two weeks before and two weeks after the ODA. The effects of ODA on milk compositions and behaviors were evaluated by comparing the average values for each composition and behavior for the three days before the ODA and the last three days of the ODA. Results: The decrease of milk yield during the two weeks of ODA was significantly higher than that during the two weeks before ODA (p<0.01). The milk fat rate was significantly higher during ODA than before ODA (p<0.05). Lactose rate was significantly lower during ODA than before ODA (p<0.05). The concentrations of milk urea nitrogen, ketone bodies, and free fatty acids in the milk were significantly higher during ODA than before ODA (p<0.05). The mean total duration per day of lying during ODA was significantly lower than that before ODA (p<0.05). The walking steps per one-hour outdoor access were 158.4±54.7. The social behavior during the one-hour outdoor access of the 11 cows was 53 times/h/herd. Conclusion: Our results suggested that ODA promotes the expression of normal behavior in dairy cows, but even one hour of ODA decreases milk production in cows, which may drop producers’ profits without some financial supplementation. Moving from conventional (CMS) to automatic (AMS) milking systems could affect milk quality. Moreover, the type and preservation methods of the forages used in the TMR, such as alfalfa hay (HTMR) or corn silage (STMR) have been demonstrated to modify milk composition. Thus, this study investigated the effect of implementing AMS and different diet forage types on the quality of Italian Holstein-Friesian bulk milk. Milk samples (n = 168) were collected monthly from 21 commercial farms in northern Italy during a period of 8 mo. Farms were categorized into 4 groups according to their milking system (CMS vs. AMS) and diet forage type (HTMR vs. STMR). Milk quality data were analyzed through the mixed procedure for repeated measurement of SAS with the milking system, diet forage type, and sampling day as fixed effects. Milking through the AMS led to lower milk fat, freezing point, and β-LG A; longer coagulation time; and higher K content, pH, and β-LG B than CMS. Cows fed STMR produced milk with greater fat, protein, casein, Mg content, titratable acidity, and β-LG A, but with reduced curd firming time, freezing point, and β-LG B than those fed HTMR. The presence of an autocrine factor in milk that can trigger mammary gland involution was proposed more than 50 yr ago. To provide evidence for the existence of one or more autocrine factors, 10 multiparous cows in late lactation were quarter-milked for 7 d. Following this baseline period, the right front quarter of each cow was left unmilked, and the other quarters were milked for 7 d. Before the last milking of that period, milk (mammary secretions) was collected aseptically from both front quarters. After that milking, 250 mL of the collected samples were infused in the cows' respective rear quarters. No quarters were milked for the following 7 d (milk stasis period), and quarter milking was then resumed in all quarters for the last 7 d of the experiment (remilking period). Quarter milk samples were collected during the baseline period, before the milk stasis period, and during the remilking period. These samples were used for measuring milk components and the concentration of involution markers (SCC, BSA, and lactoferrin). Samples of mammary secretions were collected manually from the quarters during the milk stasis period for involution marker determination. We extracted RNA from samples collected from front quarters before the last milking before the milk stasis period for microRNA (miRNA) determination. As anticipated, the longer milk stasis period implemented for the right front quarter resulted in a more advanced involution than in the left front quarter, based on the concentration of involution markers in the mammary secretions, lower milk production recovery, and changes in milk composition during the remilking period. All 3 involution marker concentrations in the mammary secretions increased in both rear quarters, but were greater in the right quarter secretions than in the left quarter secretions. Resuming milking reinitiated milk production in all quarters, but milk production recovery in the right rear quarters was less robust than that in the left rear quarters (54.3 ± 1.4% vs. 61.6 ± 1.4%, respectively). Milk from the quarters infused with mammary secretions (right rear) had a lower lactose content, but a higher milk protein content and higher SCC than the quarters infused with milk. We detected a total of 359 miRNAs, 76 of which were differentially expressed in milk and mammary secretions. Expression of bta-miR-221 and bta-miR-223 was upregulated in mammary secretions 34- and 40-fold, respectively. The results of the present experiment support the contention that milk stasis leads to the accumulation of one or more factors that trigger involution. The results also indicate that milk stasis leads to changes in the miRNA profile of the milk, but whether such changes are a cause or a consequence of the involution process remains to be established. Ruminant milk composition can be affected by many factors, primarily interspecies differences, but also environmental factors (e.g., season, feeding system, and feed composition). Pasture-based feeding systems are known to be influenced by seasonal effects on grass composition. Spring pasture is rich in protein and low in fiber compared with late-season pasture, potentially inducing variability in the composition of some milk metabolites across the season. This study aimed to investigate interspecies and seasonal differences in the milk metabolome across the 3 major commercial ruminant milk species from factories in New Zealand: bovine, caprine, and ovine milk. Samples of bovine (n = 41) and caprine (n = 44) raw milk were collected monthly for a period of 9 mo (August 2016–April 2017), and ovine milk samples (n = 20) were collected for a period of 5 mo (August 2016–January 2017). Milk samples were subjected to biphasic extraction, and untargeted metabolite profiling was performed using 2 separate liquid chromatography high-resolution mass spectrometry analytical methods (polar metabolites and lipids). Major differences in the milk metabolome were observed between the 3 ruminant species, with 414 of 587 (71%) polar metabolite features and 210 of 233 (87%) lipid features being significantly different between species. Significant seasonal trends were observed in the polar metabolite fraction for bovine, caprine, and ovine milk (17, 24, and 32 metabolites, respectively), suggesting that the polar metabolite relative intensities of ovine and caprine milk were more susceptible to changes within seasons than bovine milk. We found no significant seasonal difference for the triglycerides (TG) species measured in bovine milk, whereas 3 and 52 TG species changed in caprine and ovine milk, respectively, across the seasons. In addition, 4 phosphatidylcholines and 2 phosphatidylethanolamines varied in caprine milk within the season, and 8 diglycerides varied in ovine milk. The interspecies and seasonal metabolite differences reported here provide a knowledge base of components potentially linked to milk physiochemical properties, and potential health benefits of New Zealand pasture-fed dairy ingredients. Connections between the 2 scientific fields of physiology and dairy science have existed for a very long time—since the 19th century. One possible explanation for this circumstance could have been the necessity for financial support, which might have played a significant role for many theoretical physiologists to conduct studies on dairy products. This study discusses a correspondence written by the physiologist R. Gscheidlen to J. A. Winter, the main editor of the German (previously Saxon) journal "Schmidt's Yearbooks on the Entire Field of Domestic and Foreign Medicine" ("Schmids Jahrbücher der in–und ausländischen gesammten Medicin"). Gscheidlen submitted a review manuscript on the composition of milk to Winter's journal. In that work, which was published later as the first issue's very first article of the first volume (of in total 4 quarterly volumes) in 1871, Gscheidlen mainly refers to Eduard Kemmerich, who later became a pioneer of cattle breeding in Argentine. Therefore, studies on dairy products and on dairy science, although regularly not very significant from a theoretical and physiological point of view, obviously had concrete implications for practical purposes. Furthermore, other parts of Winter's journal, volume 1871, indicate that Gscheidlen tried to connect these studies with his early theoretical works on physiological metabolism. These theoretical studies included experiments that already explored basic principles of the urea cycle from today's point of view. Of course, these works, which were partly carried out on animals, must have been extravagant and expensive. For that reason, it is possible to assume that early dairy science and other scientific fields in Germany around 1870, which were linked to the developing food industry, had a very significant influence on the advances in theoretical metabolism physiology. Automated measurements of the ratio of concentrations of methane and carbon dioxide, [CH 4 ]:[CO 2 ], in breath from individual animals (the so-called "sniffer technique") and estimated CO 2 production can be used to estimate CH 4 production, provided that CO 2 production can be reliably calculated. This would allow CH 4 production from individual cows to be estimated in large cohorts of cows, whereby ranking of cows according to their CH 4 production might become possible and their values could be used for breeding of low CH 4 -emitting animals. Estimates of CO 2 production are typically based on predictions of heat production, which can be calculated from body weight (BW), energy-corrected milk yield, and days of pregnancy. The objectives of the present study were to develop predictions of CO 2 production directly from milk production, dietary, and animal variables, and furthermore to develop different models to be used for different scenarios, depending on available data. An international dataset with 2,244 records from individual lactating cows including CO 2 production and associated traits, as dry matter intake (DMI), diet composition, BW, milk production and composition, days in milk, and days pregnant, was compiled to constitute the training dataset. Research location and experiment nested within research location were included as random intercepts. The method of CO 2 production measurement (respiration chamber [RC] or GreenFeed [GF]) was confounded with research location, and therefore excluded from the model. In total, 3 models were developed based on the current training dataset: model 1 ("best model"), where all significant traits were included; model 2 ("on-farm model"), where DMI was excluded; and model 3 ("reduced on-farm model"), where both DMI and BW were excluded. Evaluation on test dat sets with either RC data (n = 103), GF data without additives (n = 478), or GF data only including observations where nitrate, 3-nitrooxypropanol (3-NOP), or a combination of nitrate and 3-NOP were fed to the cows (GF+: n = 295), showed good precision of the 3 models, illustrated by low slope bias both in absolute values (−0.22 to 0.097) and in percentage (0.049 to 4.89) of mean square error (MSE). However, the mean bias (MB) indicated systematic overprediction and underprediction of CO 2 production when the models were evaluated on the GF and the RC test datasets, respectively. To address this bias, the 3 models were evaluated on a modified test dataset, where the CO 2 production (g/d) was adjusted by subtracting (where measurements were obtained by RC) or adding absolute MB (where measurements were obtained by GF) from evaluation of the specific model on RC, GF, and GF+ test datasets. With this modification, the absolute values of MB and MB as percentage of MSE became negligible. In conclusion, the 3 models were precise in predicting CO 2 production from lactating dairy cows. The objective of this experiment was to investigate the effect of lipid from rapeseed cake and oats on ruminal CH 4 emission and lactational performance of dairy cows. Twelve lactating Nordic Red cows, of which 4 were primiparous, and averaging (±SD) 48 ± 22.9 DIM, 37.8 ± 7.14 kg/d milk yield were enrolled in a switch-back design experiment with 3 periods of 4 wk each. The cows were assigned into 6 pairs based on parity, DIM, milk yield, and BW at the beginning of the experiment. The experimental treatments were (1) rapeseed cake and oats (RSC+O), and (2) rapeseed meal and barley (RSM+B) as the concentrate feeds. Cows in each pair were randomly assigned to 1 of the 2 groups, which received the treatments in 2 different sequences (i.e., group 1 received RSC+O in period 1 and 3, and RSM+B in period 2, whereas group 2 was fed RSM+B in period 1 and 3, and RSC+O in period 2). The diets consisted of a partially mixed ration with grass silage mixed with either oats or barley, according to the treatment sequence, and the rapeseed cake or meal being mixed into a pellet with either oats or barley according to the treatments, and a mineral mix. The pellet was delivered at a fixed amount (i.e., 6 kg/d for multiparous and 5 kg/d for the primiparous cows) from the milking robot. The actual forage to concentrate ratios for RSC+O and RSM+B were 51:49 and 52:48, respectively, with NDF concentrations of 41.5% and 36.0% and CP concentrations of 17.0% and 16.7% of diet DM. Dry matter intake, milk yield, and gas exchange (with a GreenFeed system attached to the milking robot) were recorded daily, and milk composition and spot fecal samples were collected during the last week of each period. Based on feed analysis, and DMI of the cows during the experiment, the total fat content of the experimental diets was 4.1% and 2.7% of DM for RSC+O and RSM+B diets, respectively. Dry matter intake was 1.6 kg/d lower, and milk yield tended to be 1.0 kg/d greater for RSC+O versus RSM+B. There were no differences in ECM yield and milk composition between the treatments, whereas milk ME efficiency was greater for cows fed RSC+O than RSM+B. Methane yield (g/kg DMI) did not differ between treatments, but CH 4 production (g/d) was 9.4% and CH 4 intensity as g/kg ECM was 11.7% lower for RSC+O versus RSM+B. The lower CH 4 production was likely caused by the lower DMI and fiber digestibility, observed with the RSC+O diet. In addition, the greater lipid intake also contributed to lower rate of fermentation and subsequent decrease in CH 4 production. Overall, feeding rapeseed cake with oats in a grass silage–based diet increased feed efficiency while decreasing CH 4 emission intensity in lactating cows. The effect of the horn status of cows on their milk composition and quality is a controversial research topic. In this study, 128 milk samples from 64 horned and 64 disbudded Brown Swiss and Original Braunvieh cows were collected from alpine farms where both horned and disbudded cows were grazing on mountain pastures. The samples were analyzed for their detailed composition and protein digestion in a simulated in vitro digestion (INFOGEST). To exclude probable influences on digestion, the β-CN genotype with its variants A1 and A2 was also included in the study. The effects of horn status and β-CN genotype were investigated in linear mixed models, which included additional influencing random factors such as Original Braunvieh blood proportion, stage of lactation, and farm. Horn status did not have any effect on milk composition or digestion. In contrast, milk from A1A1 cows showed a different protein digestion than milk of A1A2 and A2A2 cows in the gastric phase, including smaller amounts of β-casomorphin(BCM)21-associated peptides and larger amounts of BCM11-associated peptides. This study evaluated the effect of tannin extract from Acacia mearnsii on intravaginal temperature, production and milk composition of Holstein cows in a subtropical environment. The study period was 29 days and 20 multiparous Holstein cows were selected with an average milk production of 50 ± 6 kg/day and 147 ± 83 days in milk. The cows were separated into two treatments: Control (n = 10) did not receive dietary supplementation. Tannin (n = 10) received Condensed Tannin (Tanac S.A., Montenegro, RS, Brazil) at a dose of 0.19% dry matter (40 g/cow/day). Animals were monitored for milk production and composition as well as intravaginal temperature. The environmental temperature and humidity index (THI) were monitored. The mean THI was 72.39 ± 0.69. The mean intravaginal temperature was 39.17 ± 0.14 °C in the Tannin treatment and 39.03 ± 0.14°C in the Control treatment (P > 0.05). The animals in the Tannin treatment exhibited an intravaginal temperature of 39.2 °C at THI 69.38, and the Control treatment exhibited the same temperature at THI 70.16 (receiver operating characteristic (ROC); P < 0.01). Milk production was higher in the Tannin treatment than in the Control treatment (51.38 ± 0.90 kg/day and 49.94 ± 1.05 kg/day, respectively; P < 0.01). The protein content in milk was higher in the Tannin treatment than in the Control treatment (3.20 ± 0.01% and 3.04 ± 0.01%; respectively; P < 0.01). Simple Summary: Mastitis in dairy cows is a major problem for livestock production. In this study, we investigated the effects of exogenous melatonin on the milk somatic cell count and yield and the serum anti-inflammatory factors, antioxidant indices, and melatonin in dairy cows. Exogenous melatonin was shown to increase the milk component content, reduce the somatic cell count, and improve the antioxidant capacity and immune responses. Mastitis is an important factor affecting the health of cows that leads to elevated somatic cell counts in milk, which can seriously affect milk quality and result in huge economic losses for the livestock industry. Therefore, the aim of this trial was to investigate the effect of melatonin on performance and mastitis in dairy cows. Forty-eight Holstein cows with a similar body weight (470 ± 10 kg), parity (2.75 ± 1.23), number of lactation days (143 ± 43 days), BCS (3.0–3.5), milk yield (36.80 ± 4.18 kg), and somatic cell count (300,000–500,000 cells/mL) were selected and randomly divided into four groups: control (CON group), trial Ⅰ (T80 group), trial Ⅱ (T120 group), and trial Ⅲ (T160 group). Twelve cows in trial groups I, II, and III were pre-dispensed 80, 120, and 160 mg of melatonin in edible glutinous rice capsules along with the basal ration, respectively, while the control group was fed an empty glutinous rice capsule along with the ration. The trial period was 37 days, which included a 7-day adaptive phase followed by a 30-day experimental period. At the end of the trial period, feeding was ended and the cows were observed for 7 days. Milk samples were collected on days 0, 7, 14, 21, 28, and 37 to determine the somatic cell number and milk composition. Blood samples were collected on days 0, 15, 30, and 37 of the trial to determine the serum biochemical indicators, antioxidant and immune indicators, and the amount of melatonin in the blood. The results showed that the somatic cell counts of lactating cows in the CON group were lower than those in the T120 group on days 14 (p < 0.05) and 28 (p < 0.01) at 1 week after melatonin cessation. The milk protein percentage and milk fat percentage of cows in the T120 group were higher than those in the CON group (p < 0.01). The total protein and globulin content in the T120 group were higher than those in the CON group (p < 0.01). In terms of antioxidant capacity and immunity, the cows 1 week after melatonin cessation showed higher superoxide dismutase activity and interleukin-10 contents (p < 0.01) compared with the CON group and lower malondialdehyde and tumor necrosis factor-alpha contents (p < 0.01) compared with the T120 group. The melatonin content in the T120 group was increased relative to that in the other groups. In conclusion, exogenous melatonin can increase the content of milk components, reduce the somatic cell count, and improve the antioxidant capacity and immune responses to a certain extent. The objective was to conduct a systematic review to clarify the effects of l‐arginine supplementation in pregnant and lactating sows on plasma hormone levels, milk production and composition, the body condition of sows and piglet performance. In April 2023, an online search and a systematic search were performed in the following databases: Embase, Scopus, SciELO, Web of Science, PubMed and Science Direct. The combinations of keywords used were sow and arginine and lactation; sow and arginine and lactating; sow and arginine and gestation; sow and arginine and gestating; sow and arginine and pregnancy; sow and arginine and reproduction; piglet and arginine; and sow and arginine and mammary gland. In total, 21 scientific articles with original data were selected according to preestablished criteria. Among the 21 articles, seven (33%) reported measurements of some plasma hormones, and among these, six reported an increase in the levels of at least one hormone, namely, estradiol, insulin‐like growth factor 1 (IGF‐1), insulin, follicle stimulating hormone, growth hormone or prolactin, with l‐arginine supplementation. The parameters of milk were evaluated in 11 studies (52%), one reported an increase in protein content, and one reported an increase in IGF‐1 content in milk with supplementation of this amino acid. Of the 14 studies that evaluated the performance parameters of piglets, only four reported improvements in some parameters of piglets from sows that received supplementation. Dietary supplementation of arginine for sows in the final third of gestation and/or lactation may alter the plasma levels of some hormones, which may reflect in greater development of the mammary gland tissue and, consequently, promote benefits on the performance of piglets. However, more studies are needed to evaluate the real impact of this amino acid supplementation on the physiology of the sows, in general, and the performance of suckling piglets. Simple Summary: The increasing use of automated milking systems (AMS) in the dairy industry has led to the necessity for changes in management practices implemented by producers. This study investigated the productive and metabolic impact of three versus six milkings per day (MPD) during early lactation on multiparous Holstein cows managed in an AMS. Mature cows (third and greater parity) with six MPD had greater milk fat production during the experimental phase (4 to 29 days in milk; DIM) and milk production during the carryover phase (30 to 90 DIM) than mature three MPD cows. The composition of milk fatty acids differed between MPD groups with cows, with three MPD having greater short-, medium-, odd- and branched-chain fatty acids during the experimental phase compared to cows with six MPD. Based on greater blood fatty acid and β-hydroxybutyrate concentrations, the mature six MPD cows may have been in a greater postpartum nutrient deficit and at greater risk of metabolic illness. Our findings suggest a need to strategically manage the MPD or nutrition of mature, early lactation dairy cows in an AMS. Automated milking systems (AMS) are increasingly adopted for dairy cow production, promoting individualized cow management dependent on factors like lactation stage, age, and productivity. The study objective was to investigate the effects of early lactation milking frequency on cows milked via AMS. Multiparous Holstein cows blocked by parity and due date were randomly assigned to treatments (n = 8 per treatment): three (3X) or six (6X) milkings per day (MPD). The experimental phase (EXP) was defined as 4 to 29 days in milk (DIM). The AMS settings were programed so 3X cows were limited to three MPD while 6X cows were allowed six MPD. Afterwards was the carry over phase (CO) ranging from 30 to 90 DIM; all cows were allowed up to six MPD. Measurements by the AMS included bodyweight, milk yield (MY), and pellet intake. Weekly composite milk samples were analyzed for macronutrient composition and fatty acid (FA) profile. Coccygeal blood was sampled at 3, 8 ± 1, and 13 ± 1 DIM; concentrations of blood plasma analytes were quantified. Greater MPD was achieved for 6X cows versus 3X cows during EXP, but similar during the CO. Daily MY was non-separable during the EXP while 6X cows in their third or greater lactation group (3 + LG) had greater MY than 3X cows of the same LG during the CO. Milk fat content and 4% fat-corrected MY were both greater for 6X, 3 + LG cows during the EXP compared to 3X, 3 + LG cows. Milk FA methyl esters (FAME) proportions were different between MPD groups, with 6X, 3 + LG cows having the lowest short, even-chain FA from de novo or post-absorptive origin. Differences in analytes indicated that 6X, 3 + LG cows experienced metabolic stress and incorporated greater FA from adipose tissue. This study evaluated the effects of supplementing the diet of lactating cows with Acremonium terrestris culture (ATC) on milk production, serum antioxidant capacity, inflammatory indices, and serum lipid metabolomics. Over 90 days, 24 multiparous Chinese Holstein cows in mid-lactation (108 ± 10.4 days in milk, 637 ± 25 kg body weight, 30.23 ± 3.7 kg/d milk yield) were divided into either a control diet (CON) or a diet supplemented with 30 g of ATC daily. All the data were analyzed using Student's t test with SPSS 20.0 software. The results showed that compared with CON feeding, ATC feeding significantly increased milk yield, antioxidant capacity, and immune function. Lipidome screening identified 143 lipid metabolites that differed between the two groups. Further analysis using "random forest" machine learning revealed three glycerophospholipid serum metabolites that could serve as lipid markers with a predictive accuracy of 91.67%. This study suggests that ATC can be a useful dietary supplement for improving lactational performance in dairy cows and provides valuable insights into developing nutritional strategies to maintain metabolic homeostasis in ruminants. Graviera Naxos, a renowned cheese with Protected Designation of Origin status, is crafted from a blend of cow, goat, and sheep milk. This study focused on assessing the Sr isotopic and multi-elemental composition of both the processed cheese and its ingredients, as well as the environmental context of Naxos Island, including samples of milk, water, soil, and feed. The objective was to delineate the geochemical signature of Graviera Naxos cheese and to explore the utility of Sr isotopes as indicators of geographic origin. The 87Sr/86Sr values for Graviera Naxos samples ranged from 0.70891 to 0.70952, indicating a relatively narrow range. However, the Sr isotopic signature of milk, heavily influenced by the feed, which originates from geologically distinct areas, does not always accurately reflect the local breeding environment. Multi-elemental analysis revealed variations in milk composition based on type and season; yet, no notable differences were found between raw and pasteurized milk. A mixing model evaluating the contributions of milk, sea salt, and rennet to the cheese's Sr isotopic signature suggested a significant average contribution of 73.1% from sea salt. Among other bioactive molecules, milk contains high amounts of sialylated milk oligosaccharides (MOs) that influence numerous processes in the offspring. For instance, sialylated MOs inhibit the invasion of pathogens and positively influence the gut microbiome to support the optimal development of the offspring. For these reasons, sialylated MOs are also used in infant formula as well as food supplements and are potential therapeutic substances for humans and animals. Because of the high interest in sialylated bovine MOs (bMOs), we used several analytical approaches, such as gas and liquid chromatography in combination with mass spectrometry, to investigate in detail the profile of sialylated bMOs in the milk of Holstein Friesian cows during early lactation. Most of the 40 MOs identified in this study were sialylated, and a rapid decrease in all detected sialylated bMOs took place during the first day of lactation. Remarkably, we observed a high variance within the sialylation level during the first two days after calving. Therefore, our results suggest that the content of sialylated MOs might be an additional quality marker for the bioactivity of colostrum and transitional milk to ensure its optimized application for the production of milk replacer and food supplements. Background: This study aims to determine the relationship between milk composition traits and breed in the Akkaraman and Awasi sheep as well as to provide ease of interpretation by showing the relationships structure between variables and between categories of variables in two-dimensional space with Categorical principal component analysis. Methods: Categorical principal component analysis determines relationships between continuous and categorical variables as well as ordinal variables. It aims to reduce system dimensionality through optimal scaling while maintaining variable measurement levels (nominal, multiple nominal, ordinal and interval). In this research, data obtained from Akkaraman and Awasi Breed Sheep Raised by Public Hands in Tuşba District of Van Province were used. In order to determine relationship with breed, the traits were divided into two categories, "low" and "high" and all variables (9 variables) were considered together and a Categorical principal components analysis was performed. Result: As a results, Dimension 1 accounted for 35.58% of the total variation while dimension 2 accounted for 15.21%. Two dimensions together accounted for 50.79% of the variation. Thus it can be noted that Categorical principal component analysis can be used in the analysis of data sets containing a large number of different types of variables with linear or non-linear relationships between them. Subclinical mastitis reduces milk yield and elicits undesirable changes in milk composition, but the mechanisms resulting in reduced milk production in affected mammary glands are incompletely understood. This study investigated the effects of sterile inflammation on mammary gland metabolism by assessing changes in milk and venous blood composition. Mid-lactation primiparous Holstein cows (n = 4) had udder halves randomly allocated to treatments; quarters of 1 udder half were infused with 2 billion cfu of formalin-fixed Staphylococcus aureus (FX-STAPH) and quarters of the opposite udder half were infused with saline (SAL). Blood samples were collected from the right and left subcutaneous abdominal veins in 2.6 h intervals until 40 h postchallenge and analyzed for blood gas and metabolite concentrations. Milk from FX-STAPH udder halves had significantly increased SCS by the first milking at 8 h postchallenge. By 16 h postchallenge, FX-STAPH udder halves had increased concentrations of protein and lactate and lower lactose concentrations than SAL udder halves. Milk fat concentrations, milk yields, ECM yields, and the ferric reducing antioxidant power of milk were not significantly different between SAL and FX-STAPH udder halves. Venous blood of FX-STAPH halves had marginally greater concentrations of saturated O 2 , partial pressures of O 2 , and glucose concentrations than SAL halves. Conversely, total and partial pressures of CO 2 did not differ between udder half treatments, suggesting a shift in local metabolite utilization in FX-STAPH udder halves. These results indicate that changes in milk composition resulting from mastitis are accompanied by changes in some key blood metabolite concentrations. The shift in venous blood metabolite concentrations, along with the marked increase in milk lactate, suggests that local mammary tissue or recruited immune cells, or both, alter metabolite usage in mammary tissues. A total of 18 Italian Mediterranean buffalo cows were subjected to fortnightly milk sampling from May to July 2022. Air temperature and relative humidity were monitored throughout the trial. Temperature-humidity index (THI) was calculated and ranged from 69 to 79, so that 3 classes were set to study the effect of different THI on milk quality: THI less than 72 (THI <72), THI ranging from 72 to 76 (THI 72–76), and THI over 76 (THI >76). Individual milk samples from buffalo cows were analyzed for milk composition and coagulation traits, fatty acid and amino acid composition, and enzymatic and mineral profiles. The THI >76 class registered higher lactose content and poorer milk clot characteristics in comparison with THI <72. Exposure to a THI over 76 resulted in increased SFA and short-chain fatty acid contents in milk, as well as in the highest atherogenic and thrombogenic index values and the lowest essential amino acid content. Moreover, the lowest values of calcium, copper, and selenium contents were found in the milk of buffalo cows exposed to a THI over 72. Results confirm that Italian Mediterranean buffalo express a good level of thermo-tolerance, even though exposure to a daily THI over 76 has a deleterious effect on some nutritional and technological properties of milk. The aim of this study was to determine the individual and combined effects of supplementing fat with whole-cracked rapeseed (FAT), nitrate (NITRATE), and 3-nitrooxypropanol (3-NOP) on compositional and functional properties of milk fat. An 8 × 8 incomplete Latin square design was conducted with 48 lactating Danish Holstein cows over 6 periods of 21 d each. Eight diets were 2 × 2 × 2 factorially arranged: FAT (30 or 63 g crude fat/kg DM), NITRATE (0 or 10 g nitrate/kg DM), and 3-NOP (0 or 80 mg 3-NOP/kg DM), and cows were fed ad libitum. Milk samples were analyzed for general composition, fatty acids (FA) and thermal properties of milk fat. Milk fat content was decreased by supplementing fat but increased by 3-NOP. The changes in FA composition were mainly driven by the FAT × 3-NOP interaction. Fat supplementation shifted milk FA composition toward a lower content of SFA and greater contents of MUFA and PUFA, whereas these effects became smaller in combination with 3-NOP. However, 3-NOP had no effects on SFA, MUFA, or PUFA in low-fat diets. Fat supplementation lowered solid-fat content in milk fat because of the decreased SFA content. The onset crystallization temperature of milk fat was decreased by 3-NOP when supplemented in low-fat diets. According to the FAT × 3-NOP interaction, supplementation of fat without 3-NOP shifted the peak temperature of the low-melting fraction of milk fat toward low temperature as a result of a decreased proportion of C16:0 and increased proportions of C18:1 cis -9, C18:1 trans -11, C18:2 cis -9, and CLA cis -9, trans -11. In conclusion, no additive effects were observed among FAT, NITRATE, and 3-NOP on chemical and thermal properties of milk fat, and fat supplementation largely changed milk FA composition, which in turn affected the thermal properties of milk fat. This study examined the effects of spread formulation and the structural/lubricant properties of six different commercial hazelnut and cocoa spreads on sensory perception. Rheology, tribology, and quantitative descriptive analysis (QDA) was assessed by also evaluating the correlation coefficients between the quality descriptor and the rheological and textural parameters. The viscosity was evaluated at different temperatures to better simulate conditions before and after ingestion. Tribological analysis was executed at 37°C to mimic the human oral cavity. The effect of saliva presence and the number of runs on tribological behaviors was investigated. Moreover, textural, calorimetric, and particle size distribution measurements were performed to reinforce the correlation between structural/thermal parameters (e.g., firmness, stickiness, sugar melting point) and sensory aspects. "Visual viscosity," defined as a sensory attribute evaluated prior to consumption, negatively correlated with apparent viscosity measured at 20°C and 10 s−1, whereas "body," defined during oral processing and related to creaminess, positively correlated with apparent viscosity measured at 37°C and 50 s−1. These attributes were mainly influenced by particulate microstructure and solid volume fraction within the formulation. Textural stickiness positively correlated with sensory "adhesiveness" and was related to fat composition and milk powder addition, while "sweetness" was related to sucrose content and sugar melting enthalpy. Tribological data provided meaningful information related to particle‐derived attributes, as well as after‐coating perception (fattiness/oiliness), thus better predicting food evolution during oral consumption. Subclinical mastitis (SCM) has a detrimental impact on cow's milk characteristics. The study was to investigate the consequences of SCM on milk composition and blood biochemistry, oxidative stress biomarkers, and the correlation between different milk and serum parameters in SCM-affected cows. The multiparous Holstein cows were grouped as 16 subclinical infected and nine healthy according to milk somatic cell count and California Mastitis Test scores. The blood and milk samples were gathered from each animal and evaluated for compositional, biochemical, and oxidative stress marker alterations. SCM-affected milk samples showed a significant (P < 0.05) decrease in fat, protein, lactose, total solids, solids-non-fat percentages, and total antioxidant capacity (TAC) and a significant (P < 0.05) increase in ash %, catalase, and malondialdehyde (MDA). Milk somatic cell count showed a positive correlation with MDA and catalase. Milk fat showed a positive correlation with lactose, total protein, and albumin. A significant increase in catalase and total protein associated with a significant decrease in Ca level was recorded in SCM blood samples. The results indicate that a significant correlation between different parameters in blood and milk can be useful as a proper clinical indicator for the early detection of SCM in cows. The fatty acid (FA) composition of milk fat can be positively influenced by the modification of dairy farming and the possible selection of raw material for processing. The question arises as to whether the benefits of a better FA composition will be maintained even after processing dairy products. Thus, the aim was to obtain a more favourable FA composition by a targeted selection of cow's milk (grazing vs stabled herds) and then compare FAs of milk and fermented product (yoghurt). Bulk tank milk of grazing herds had significantly better FA composition than milk of stabled herds (for example, C16:0 28.07% vs 32.27%, P < 0.001; C18:3n-3 0.83% vs 0.41%, P < 0.001; C18:2 cis-9, trans-11 (conjugated linoleic acid; CLA) 1.02% vs 0.41%, P < 0.01). The differences between the FAs in milk and yoghurt samples were negligible (in relative values from 0.04% to 5.21%). The correlations between milk and yoghurt for nutritionally important FAs were high, from 0.925 0 (C18:2n-6) to 0.998 8 (CLA; both P < 0.001). The minimal effect of milk fermentation on the original FA composition of milk fat was found. The present study investigated the effects of various inclusion levels of dietary Chinese herbal medicine (CHM) preparation on feed consumption, milk yield and milk composition, serum biochemistry, hematological profile, and reproductive efficiency of Holstein dairy cows. A total of 117 lactating Holstein cows were randomly divided into four groups as control (n = 27; without CHM supplementation) and treatment groups CHM-0.5 (n = 31), CHM-0.75 (n = 29), and CHM-1 (n = 30) fed diet supplemented with 0.5, 0.75, and 1 kg/cow/d for 30 days, respectively. The study began at d 20 postpartum (d 0 of the study). At d 50 postpartum, the cows in all groups were subjected to estrus synchronization using a modified Ovsynch protocol (GPGMH) and observed for reproductive variables. Feed intake, milk yield and milk composition, serum biochemistry and hematological profile, and reproductive efficiency were measured. A significantly higher milk yield with improved milk lactose, milk protein and milk fat were found in the CHM-0.75 group compared to the other groups (p < 0.05). Besides, the estrus response, ovulation rate, ovulatory follicle diameter, and pregnancy rate increased in CHM-0.75 compared to CHM-0 or CHM-0.5 group (p < 0.05). The serum metabolites (glucose, AST, arginine, BUN, and NO) showed variations among the treatment groups at different time points (synchronization, AI, or post-AI). In conclusion, CHM supplementation improves the milk yield, milk composition, and serum metabolites in dairy cows. The current study was done to evaluate prevalence of mastitis in Lassi camel breed in Lasbela, Pakistan. A total of 100 camel cows were selected for milk sampling for isolation and identification of mastitis causing bacteria and biochemical tests. A questionnaire was used to know prevalence of mastitis associated with species, breed, age, parity, and udder hygiene condition, frequency of milking, practice of teat dipping, and feeding frequency of the camel cows. A t-test was used to anylyse the results and prevalence data was expressed in percentage. The total 43% prevalence of mastitis was observed in Lassi camel of of Lasbella district Balochistan, Pakistan. It was observed that prevalence of mastitis 38%, 41.18%, and 47.5% was increased with advancement in the age 4-7, 7-10, and above 10 years, respectively. It was also observed that some associated factors such as milking and feeding frequency, and udder hygine play important role in mastitis. During mastitis, significantly increased in fat, protein, total solids, solids not fat, and moisture contents, and pH and significantly declined in lactose indices was observed. Staphylococcus aureus species were dominantly found followed with Streptococcus agalactiea and Streptococcus dysagalctiea. This study will helpful to the local farmers and veterinarian regarding proper use of antimicrobial agents, and maintenance of udder hygiene to control mastitis in Lassi camel breed in Lasbela, Pakistan. The current study was designed to explore all possible etiological features including mineral composition of soil and their effects on hematobiochemical profile in buffaloes suffering from post-parturient hemoglobinuria. The total 384 buffaloes were randomly selected from Tehsil Bhalwal, District Sargodha, Pakistan. The post parturient hemoglobinuria was observed during the period of four months, from November 2016 to February 2017. The parameters including point prevalence of PPHU and hemato-biochemical profile were investigated. The data was analyzed using T-test and regression analysis through SPSS software. Out of 384 animals, 40 (10.4%) were confirmed with PPHU. All hemato-biochemical profile showed significant difference between healthy and affected groups instead mean corpuscular hemoglobin concentration (MCHC), serum calcium and glucose, which were not significant. Vital signs including pulse and respiration showed significant difference but heart rate was found non-significant between both groups. The results regarding mineral analysis of the soil showed significant difference in phosphorus and copper and non-significant difference in calcium and molybdenum. Moreover, strong positive correlation was associated with phosphorus and molybdenum level while moderate positive correlation was attributed to the calcium and copper level in the soil. It is concluded that post parturient hemoglobinuria in buffaloes of Tehsil Bhalwal was associated with variation of soil composition, particularly the deficiency of phosphorus which is main cause of hemoglobinuria. Introduction: Breastfeeding is considered the best nutrition for infants, including born preterm due to containing optimal ingredients for development and growth. Breast milk is a rich source of nutrients, it also contains diverse types of bacteria, which play a role in infant's gastrointestinal tract colonization and preserving mammary health. Composition of human milk varies depending on numerous maternal and infant factors. Aim of the study: The aim of the study was to summarize the current knowledge about the content of nutrients, bioactive factors and microbiota diversity in human milk depending on various factors. Materials and methods: The work is a review of 26 mainly English-language scientific publications from 1989-2024 posted on the PubMed platform. Results: Milk composition changes in addition to several factors. Depending on postnatal age: preterm milk has higher content of protein, fat and immunological factors and lower concentration of lactose than term milk. Depending on milk maturity: Colostrum is higher in protein but lower in lactose, fat and energy than mature milk. Human milk is also a source of viable bacteria important for developing infant's gut microbiota, but the diversity of bacterial strains in milk may vary. Depending on mode of delivery: milk of mothers after spontaneous labour contents more probiotic vaginal- and enteric-associated bacteria, while milk of mothers after C-section has more skin-associated microbiota. Depending on mother's weight: milk of overweight mothers is lower in probiotic- and higher in pathogenic bacteria. Alcohol, nicotine, and some medications also have negative impact on produced milk. Conclusions: Human milk varies from woman to woman and its composition depends on several factors. Postnatal and gestational stage, delivery mode, mother's weight and certain substances have been found to be important factors influencing the constitution of breast milk, including nutrients, microbiota and volume of milk. Simple Summary: Pre-weaning piglet diarrhea is one of the major concerns in the swine industry. Recently, various probiotics have been applied to improve animal gut health and performance. This study investigated the impact of C. butyricum probiotic feed additive on sow and piglet performance, together with alterations in lipidomic and metabolomic profiles of sow milk. Results showed that sows given the probiotics had lower backfat loss and their piglets experienced less diarrhea incidence, although there were no other significant benefits for piglet growth. Changes in certain fatty acids and metabolites present in sow colostrum and milk, which could impact their nutritional profiles and the health of the piglets, were significantly observed. This study provided new insights regarding the impacts of probiotics application that could potentially lead to better outcomes in swine farms. The present study aimed to investigate the impact of Clostridium butyricum probiotic feed additive on sow and piglet performances, together with alterations in the lipidomic and metabolomic profiles of sow milk. Sixty-four Landrace × Yorkshire crossbred sows and 794 piglets were included. Sows were divided into two groups; i.e., (i) conventional gestation diet (control; n = 35) and (ii) conventional diet added with 10 g/sow/day of probiotic C. butyricum spores (treatment; n = 29) from one week before the estimated farrowing day until weaning (29.6 ± 4.8 days). The sow and piglet performances and incidence of piglet diarrhea were recorded. Changes in gross chemical composition, fatty acid and non-volatile polar metabolite profiles of sow colostrum, transient milk and mature milk were evaluated. The results showed that relative backfat loss in the treatment group (−2.3%) was significantly lower than in control group (11.6%), especially in primiparous sows (p = 0.019). The application of C. butyricum probiotics in sows significantly reduced the incidence of diarrhea in piglets (p < 0.001) but no other effect on piglet performance was found. Lipidomic and metabolomic analyses revealed variations in sow colostrum and milk biomolecular profiles, with indicative compounds significantly altered by feeding with the C. butyricum probiotics. In conclusion, the use of C. butyricum probiotics in sows may improve sow body condition and reduce diarrhea incidence in piglets, with underlying changes in milk composition that warrant further investigation. There are concerns pertaining to their suitability for vulnerable people as they differ from natural milk in nutritional composition, flavour and digestibility. Currently, there exists no clear‐cut definition/identity/standards in almost all countries (except Canada) for them as well as for lab‐grown milk. The present paper focuses on novel areas of issues and challenges associated with their suitability to feed infants. It also raises concerns and priority areas for the regulators to develop policies regarding establishing standards and regulations for varied parameters to assure safety, health and wellness of consumers. This study investigates the dynamic changes in milk nutritional composition and microbial communities in Tibetan sheep and goats during the first 56 days of lactation. Milk samples were systematically collected at five time points (D0, D7, D14, D28, D56) post-delivery. In Tibetan sheep, milk fat, protein, and casein contents were highest on D0, gradually decreased, and stabilized after D14, while lactose and galactose levels showed the opposite trend. Goat milk exhibited similar initial peaks, with significant changes particularly between D0, D7, D14, and D56. 16S rRNA gene sequencing revealed increasing microbial diversity in both species over the lactation period. Principal coordinates analysis identified distinct microbial clusters corresponding to early (D0–D7), transitional (D14–D28), and mature (D56) stages. Core phyla, including Proteobacteria, Firmicutes, Bacteroidetes, and Actinobacteria, dominated the milk microbiota, with significant temporal shifts. Core microbes like Lactobacillus, Leuconostoc, and Streptococcus were common in both species, with species-specific taxa observed (e.g., Pediococcus in sheep, Shewanella in goats). Furthermore, we observed a highly shared core microbiota in sheep and goat milk, including Lactobacillus, Leuconostoc, and Streptococcus. Spearman correlation analysis highlighted significant relationships between specific microbial genera and milk nutrients. For instance, Lactobacillus positively correlated with total solids, non-fat milk solids, protein, and casein, while Mannheimia negatively correlated with protein content. This study underscores the complex interplay between milk composition and microbial dynamics in Tibetan sheep and goats, informing strategies for livestock management and nutritional enhancement. This paper aimed to estimate the heritability and genetic correlations for processability (milk coagulation properties, cheese yield, and heat stability), milk production, and milk composition of dairy sheep in a New Zealand flock. Test-day records from 169 ewes were obtained during the 2021–2022 season. Heritability estimates for yield traits (milk, fat, protein, and lactose yield) were moderate and ranged from 0.33 to 0.38. Whereas heritability estimates for processability traits were lower and ranged from 0.17 to 0.27. The genetic correlations indicated that poorer milk coagulation properties were associated with lower mammary health indicated by higher SCS, higher pH, and lower lactose content. Additionally, a higher ratio of casein to calcium i.e. lower availability of soluble calcium was associated with poorer milk coagulation properties. Higher yields of milk, fat, protein, and lactose were associated, but to a smaller degree, with better MCP. It is proposed that genetic improvement of dairy sheep for higher yields of fat and protein, and for lower SCS, could indirectly improve milk processability for cheese-making. However, it is recommended that larger studies involving more dairy sheep flocks are conducted to validate the present results before developing a selection index for this industry. The composition of milk lipids varies across different ethnic sources. The lipidome profiles of Chinese Han human milk (HHM) and Chinese Korean human milk (KHM) were investigated in this study. A total of 741 lipids were identified in HHM and KHM. Twenty-eight differentially expressed lipids (DEL) were screened between the 2 milk groups; among these, 6 triacylglycerols (TG), 13 diacylglycerols (DG), 7 free fatty acids (FFA), and 1 monoglyceride (MG) were upregulated in KHM. Carnitine (CAR) was upregulated in HHM. Most DEL showed a single peak distribution in both groups. The correlations, related pathways and diseases of these DEL were further analyzed. The results demonstrated that DG, MG, and FFA showed highly positive correlations with each other (r > 0.8). The most enriched Kyoto Encyclopedia of Genes and Genomes (https://www.kegg.jp/kegg/) and Human Metabolome Database (http://www.hmdb.ca) pathways were inositol phosphate metabolism, and α-linolenic acid and linolenic acid metabolism, respectively. Major depressive disorder-related FFA (20:5) and FFA (22:6) were more abundant in KHM, whereas HHM showed more obesity-related CAR.
