Passion fruit is widely cultivated in tropical, subtropical regions of the world. The attack of bacterial and fungal diseases, and environmental factors heavily affect the yield and productivity of the passion fruit. The CC-NBS-LRR (CNL) gene family being a subclass of R-genes protects the plant against the attack of pathogens and plays a major role in effector-triggered immunity (ETI). However, no information is available regarding this gene family in passion fruit. To address the underlying problem a total of 25 and 21 CNL genes have been identified in the genome of purple (Passiflora edulis Sims.) and yellow (Passiflora edulis f. flavicarpa) passion fruit respectively. Phylogenetic tree was divided into four groups with PeCNLs present in 3 groups only. Gene structure analysis revealed that number of exons ranged from 1 to 9 with 1 being most common. Most of the PeCNL genes were clustered at the chromosome 3 and underwent strong purifying selection, expanded through segmental (17 gene pairs) and tandem duplications (17 gene pairs). PeCNL genes contained ciselements involved in plant growth, hormones, and stress response. Transcriptome data indicated that PeCNL3, PeCNL13, and PeCNL14 were found to be differentially expressed under Cucumber mosaic virus and cold stress. Three genes were validated to be multi-stress responsive by applying Random Forest model of machine learning. To comprehend the biological functions of PeCNL proteins, their 3D structure and gene ontology (GO) enrichment analysis were done. Our research analyzed the CNL gene family in passion fruit to understand stress regulation and improve resilience. This study lays the groundwork for future investigations aimed at enhancing the genetic composition of passion fruit to ensure robust growth and productivity in challenging environments. Simple Summary: A group of blood diseases called myeloproliferative neoplasms (MPNs) cause a buildup of certain blood cell types in affected individuals. Each MPN category has a distinct clinical presentation and is associated with mutations in specific genes. The Clinical Nurse Leader (CNL) is a master's prepared nurse envisioned to provide clinical leadership at the microsystem level to ensure safe, high quality patient-centered care. The American Association of Colleges of Nurses defined ten 'fundamental aspects' of CNL practice, but as the certified CNL population grows, data suggest they are filling a variety of positions besides formally designated CNL roles. This article reports the results of a systematic review of CNL literature to better understand what roles and activities certified CNLs are enacting when not hired into formally designated CNL roles. Sixty-nine articles met inclusion criteria. Roles identified include: faculty, 62%; clinical management/executive, 12%; specialty clinician, 11%; and staff nurse, 9%. In these roles, certified CNLs are reviewing literature, conducting research, and/or writing commentaries on CNL education and practice and other health foci such as alarm fatigue, insulin practices, and physical exercise for cancer-related fatigue. Results indicate that despite a lack of formal tracking of certified CNLs over time, the available information identifies a variety of roles and job titles used by this group of professional nurses. The study findings add to the body of knowledge informing overall understanding of the CNL initiative.

Chronic neutrophilic leukemia (CNL) is a rare myeloproliferative neoplasm that is genetically characterized by the absence of both the Philadelphia chromosome and BCR-ABL1 fusion gene and the high prevalence of mutations in the colony-stimulating factor 3 receptor (CSF3R). Additional disease-modifying mutations have been recognized in CNL samples, portraying a distinct mutational landscape. Despite the growing knowledge base on genomic aberrations, further progress could be gained from the availability of representative models of CNL. To address this gap, we screened a large panel of available leukemia cell lines, followed by a detailed mutational investigation with focus on the CNL-associated candidate driver genes. The sister cell lines CNLBC-1 and MOLM-20 were derived from a patient with CNL and carry CNL-typical molecular hallmarks, namely mutations in several genes, such as CSF3R, ASXL1, EZH2, NRAS, and SETBP1. The use of these validated and comprehensively characterized models will benefit the understanding of the pathobiology of CNL and help inform therapeutic strategies.

The mechanism underlying the ability of rice to germinate underwater is a largely enigmatic but key research question highly relevant to rice cultivation. Moreover, although rice is known to accumulate salicylic acid (SA), SA biosynthesis is poorly defined, and its role in underwater germination is unknown. It is also unclear whether peroxisomes, organelles essential to oilseed germination and rice SA accumulation, play a role in rice germination. Here, we show that submerged imbibition of rice seeds induces SA accumulation to promote germination in submergence. Two submergence-induced peroxisomal Oryza sativa cinnamate:CoA ligases (OsCNLs) are required for this SA accumulation. SA exerts this germination-promoting function by inducing indole-acetic acid (IAA) catabolism through the IAA-amino acid conjugating enzyme GH3. The metabolic cascade we identified may potentially be adopted in agriculture to improve the underwater germination of submergence-intolerant rice varieties. SA pretreatment is also a promising strategy to improve submerged rice germination in the field. The clinical nurse leader (CNL) is a relatively new nursing role, introduced in 2003 through the American Association of Colleges of Nursing (AACN). A narrative review of the extant CNL literature was conducted with the aim of comprehensively summarizing the broad and methodologically diverse CNL evidence base. The review included 25 implementation reports, 1 CNL job analysis, 7 qualitative and/or survey studies, and 3 quantitative studies. All CNL implementation reports and studies described improved care quality outcomes after introduction of the role into a care delivery microsystem. Despite preliminary evidence supporting the CNL as an innovative new nursing role capable of consistently improving care quality wherever it is implemented, CNLs are still struggling to define the role to themselves and to the health care spectrum at large. Although the AACN CNL White Paper provides a concise model for CNL educational curriculum and end competencies, there is a compelling need for further research to substantively delineate the CNL role in practice, define care delivery structures and processes that influence CNL integration, and develop indicators capable of capturing CNL-specific contributions to improved care quality.

The study of plant diseases is almost as old as agriculture itself. Advancements in molecular biology have given us much more insight into the plant immune system and how it detects the many pathogens plants may encounter. Members of the primary family of plant resistance (R) proteins, NLRs, contain three distinct domains, and appear to use several different mechanisms to recognize pathogen effectors and trigger immunity. Understanding the molecular process of NLR recognition and activation has been greatly aided by advancements in structural studies, with ZAR1 recently becoming the first full-length NLR to be visualized. Genetic and biochemical analysis identified many critical components for NLR activation and homeostasis control. The increased study of helper NLRs has also provided insights into the downstream signaling pathways of NLRs. This review summarizes the progress in the last decades on plant NLR research, focusing on the mechanistic understanding that has been achieved.

To ensure Clinical Nurse Leaders (CNL) are prepared to function to the full scope of their master's level education, the American Association of Colleges of Nursing published a 2007 White Paper and a 2013 update articulating expected end-of-education and CNL practice competencies. The Commission on Nurse Certification published a CNL job analysis in 2016, which identified a core set of knowledge, skills, and abilities for entry-level practicing CNLs, as the basis for the CNL certification exam. While all share core themes, the language and organization of competencies differs across the three policy documents, resulting in potential ambiguity about expected CNL knowledge and practices, and how they differ from other nursing roles. This effort identified, analyzed and synthesized CNL education and practice concepts listed in each document to refine understanding about CNL competencies and harmonize concepts across documents. The product of this effort is a cohesive CNL competency framework that aligns across the education-to-practice trajectory. On the Cray XT, aprun is the command to launch an application to a set of compute nodes reserved through the Application Level Placement Scheduler (ALPS). At the National Energy Research Scientific Computing Center (NERSC), interactive aprun is disabled. That is, invocations of aprun have to go through the batch system. Batch scripts can and often do contain several apruns which either use subsets of the reserved nodes in parallel, or use all reserved nodes in consecutive apruns. In order to better understand how NERSC users run on the XT, it is necessary to associate aprun information with jobs. It is surprisingly more challenging than it sounds. In this paper, we describe those challenges and how we solved them to produce daily per-job reports for completed apruns. We also describe additional uses of the data, e.g. adjusting charging policy accordingly or associating node failures with jobs/users, and plans for enhancements.

The shear behaviour of concrete-rock interfaces has been the aim of extensive research in geotechnical engineering applications such as rock socketed piles, rock bolts and concrete dam arch bridge foundations. Several experimental studies through direct shear tests have been conducted to evaluate the shear behaviour of rock-concrete interfaces under CNL (Constant Normal Load) and CNS (Constant Normal Stiffness) conditions. In this paper, PFC2D numerical simulations of unbonded rock-concrete planar and saw-tooth triangular joints under CNL and CNS boundary conditions are conducted using the Shear Box Genesis (SBG) approach proposed by Bahaaddini et al. (2013b). The numerical simulation results are compared with experimental data published by Gutiérrez (2013) and Gu et al. (2003). Results indicate that the SBG approach reproduces suitably the shear behaviour, failure mode and asperity damage of unbonded (planar and triangular) rock-concrete interfaces, specially under CNL conditions. Despite their frequent natural occurrence, filled discontinuities under constant normal stiffness (CNS) boundary conditions have been studied much less systematically, perhaps because of the difficulties arising from the increased number of variable parameters. Because of the lack of reliable and realistic theoretical or empirical relations and the difficulties in obtaining and testing representative samples, engineers rely on judgment and often consider the shear strength of the infilled material itself as shear strength of rock joints. This assumption leads to uneconomical and also sometimes the unsafe design of underground structures, slopes, rock-socketed piles and foundations. To study the effect of infill on the shear behavior of rock joints, tests were performed on the modeled infilled rock joint having different joint roughness under constant normal load (CNL) and CNS boundary conditions at various initial normal stress and varying thickness of the infilled material. The test results indicate that shear strength decreases with an increase in t/ a ratio for both CNL and CNS conditions, but the reduction in shear strength is more for CNL than for CNS condition for a given initial normal stress. The detailed account of the effect of thickness of infilled material on shear and deformation behavior of infilled rock joint is discussed in this paper, and a model is proposed to predict shear strength of infilled rock joint. Plant nucleotide-binding leucine-rich repeat receptors (NLRs) are intracellular immune receptors for pathogen recognition and signaling. They include sensor NLRs (sNLRs) that detect pathogens, and helper NLRs, which transduce downstream immune signals. During immune responses, both membrane-localized pattern recognition receptors (PRRs) and sNLRs rely on helper NLRs for signal transduction. The Arabidopsis helper NLRs, ADR1s and NRG1s, along with their interacting lipase-like protein dimers, are differentially required by sNLRs. Recent structural and biochemical analyses suggest that they assemble into oligomeric resistosomes with lipase-like protein dimers upon perception of small molecules produced from enzymatic activities of upstream TIR-type sNLRs. As a result, ADR1s and NRG1s form membrane calcium channels to induce immune responses and cell death. In contrast, Solanaceous NRC clade helper NLRs transduce signals from many sNLRs and some PRRs. Since the mid-eighties, the Centre national du livre in Paris has developed a translation grant programme aimed at encouraging the translation into French of scientific or literary works of high quality. Almost unique in kind, this initiative has kept on supporting around 330 translation projects every year, 60 % of which are submitted to the Commission littératures étrangères. Far from systematic, the grants are allocated following an evaluation of the quality of both the original and its French translation. Following a Descriptive paradigm and based on a fieldwork within the Commission, the present article analyses this evaluation process in order to highlight its underlying dynamics and tensions. In doing so, the author questions what the notions of “ high literary representativeness” and “translation quality” refer to in this translating institution.

Authoring OWL ontologies is hard for both domain experts and ontology engineers unless they have good knowledge of mathematics and logic. Thus, controlled natural languages (CNLs) have been proposed for easier authoring of ontologies via statements that can be automatically translated to OWL. Unfortunately, most of these languages are controlled English languages. Though they typically allow using non-English identifiers for representing concepts, instances (of concepts), and relationships, keywords remain English. This makes statements awkward and both hard and slow to read and write. Besides, error messages are generated in English only. Thus, this paper proposes the first controlled Arabic language (CAL) for authoring ontologies using Arabic words only. CAL is accompanied by a tool to allow translating CAL statements to OWL. Detailed error messages are also generated in Arabic. Besides, CAL is based on and has a similar syntax as the controlled English language Rabbit, and thus, statements can be easily translated between both languages allowing the cooperation of Arabic-speaking and English-speaking domain experts and ontology engineers in ontology authoring and validation, a capability which is absent in other CNLs. The advantages of CAL have been empirically tested to support our claim regarding the need to develop a controlled language corresponding to each natural language other than English. Nevertheless, an additional layer, NCAL, allows developing ontologies in a more-natural CAL in comparison with CAL. Plant disease resistance genes (R-genes) play a critical role in the defense response to pathogens. Barley is one of the most important cereal crops, having a genome recently made available, for which the diversity and evolution of R-genes are not well understood. The main objectives of this research were to conduct a genome-wide identification of barley Coiled-coil, Nucleotide-binding site, Leucine-rich repeat (CNL) genes and elucidate their evolutionary history. We employed a Hidden Markov Model using 52 Arabidopsis thaliana CNL reference sequences and analyzed for phylogenetic relationships, structural variation, and gene clustering. We identified 175 barley CNL genes nested into three clades, showing (a) evidence of an expansion of the CNL-C clade, primarily due to tandem duplications; (b) very few members of clade CNL-A and CNL-B; and (c) a complete absence of clade CNL-D. Our results also showed that several of the previously identified mildew locus A (MLA) genes may be allelic variants of two barley CNL genes, MLOC_66581 and MLOC_10425, which respond to powdery mildew. Approximately 23% of the barley CNL genes formed 15 gene clusters located in the extra-pericentromeric regions on six of the seven chromosomes; more than half of the clustered genes were located on chromosomes 1H and 7H. Higher average numbers of exons and multiple splice variants in barley relative to those in Arabidopsis and rice may have contributed to a diversification of the CNL-C members. Capsule null locus ( cnl ) strains, one type of specific unencapsulated Neisseria spp., only have regions D and E of the capsule gene cluster which encodes the genes for capsule biosynthesis, modification, and transportation. Compared with encapsulated strains, regions A and C of cnl strains have been replaced by 113 or 114 bp conserved non-coding sequences. Cnl strains include multiple clonal complexes (CC). According to previous studies, CC198 is the major clonal lineage in both cnl patients and healthy cnl carriers. We hypothesized that CC198 possesses different genome characteristics compared with other cnl strains. In this study, we obtained the draft genomes of two CC198 strains from healthy carriers. Using 75071 single nucleotide polymorphisms located in 1163 core genes, we constructed the phylogenetic relationships between a batch of representative Neisseria meningitidis genomes. CC198 and CC1136 clustered together, but apart from other N. meningitidis strains including CC53. We also aligned the sequences of genes located in regions D and E of the capsule gene locus from encapsulated and unencapsulated strains. A number of possible recombination events were identified in the galE and tex genes between different serogroups of encapsulated N. meningitidis and CC53 strains, especially in tex . In contrast, there is almost no recombination in N. meningitidis CC198 strains. These results showed that CC198 belongs to a phylogenetically distinct group within the species N. meningitidis , which may be directly derived from the cnl -type ancestor of N. meningitidis . According to the 2008 WHO classification, the category of myelodysplastic/myeloproliferative neoplasms (MDS/MPN) includes atypical chronic myeloid leukaemia (aCML), chronic myelomonocytic leukaemia (CMML), MDS/MPN-unclassifiable (MDS/MPN-U), juvenile myelomonocytic leukaemia (JMML) and a 'provisional' entity, refractory anaemia with ring sideroblasts and thrombocytosis (RARS-T). The remarkable progress in our understanding of the somatic pathogenesis of MDS/MPN has made it clear that there is considerable overlap among these diseases at the molecular level, as well as layers of unexpected complexity. Deregulation of signalling plays an important role in many cases, and is clearly linked to more highly proliferative disease. Other mutations affect a range of other essential, interrelated cellular mechanisms, including epigenetic regulation, RNA splicing, transcription, and DNA damage response. The various combinations of mutations indicate a multi-step pathogenesis, which likely contributes to the marked clinical heterogeneity of these disorders. The delineation of complex clonal architectures may serve as the cornerstone for the identification of novel therapeutic targets and lead to better patient outcomes. The WHO 2008 definition of chronic neutrophilic leukemia (CNL) is based on clinical and laboratory parameters but not on molecular abnormalities. Mutations in CSF3R, SETBP1 and CALR are reported in patients with chronic neutrophilic leukemia (CNL). However, because CNL is rare, there are few large studies of this issue. We sequenced these genes in 14 patients who met the WHO-criteria of CNL. 8 subjects had CSF3RT618I, 6 SETBP1 mutations and 1 a CALR mutation. Our data suggest mutation analysis of CSF3R, SETBP1 and CALR should be included in the diagnostic criteria for CNL. These data may also have therapy implications. [ABSTRACT FROM AUTHOR]

The US Veterans Administration (VA) has implemented the clinical nurse leader (CNL) role nationwide. Nursing leaders at the Malcolm Randall VA Medical Center in Gainesville, Florida, implemented the development of the CNL role in the perioperative setting during the summer of 2012. The perioperative department developed the position in partnership with the University of Florida College of Nursing, Gainesville, Florida. The team developed a description of the roles and experiences of the preceptors, the clinical nurse leader resident, and the University of Florida faculty member. The clinical nurse leader resident's successes and the positive outcomes, such as improved patient outcomes, experienced by the perioperative department demonstrated the importance of the CNL role. Granulocyte colony-stimulating factor is a cytokine able to stimulate bothmyelopoiesis and hematopoietic stem cell mobilization, which has seen it used extensively in the clinic to aid hematopoietic recovery. It acts specifically via the homodimeric granulocyte colony-stimulating factor receptor (G-CSFR), which is principally expressed on the surface of myeloid and hematopoietic progenitor cells. A number of pathogenic mutations have now been identified in CSF3R, the gene encoding G-CSFR.These fall into distinct classes, each of which is associated with a particular spectrum of myeloid disorders, including malignancy. Abstract: In this paper, an empirical model based on an earlier model by Goodman and laboratory investigations of the shear behavior of limestone rock joints are presented. For developing such a model, a series of laboratory experiments were carried out on joint specimens under constant normal load (CNL). The model of shear behavior consists of two non-linear parts; the initial section includes the onset of shearing to the peak shear strength and the post-peak covering the behavior to the end of the shear test. Input data to the model include the peak and residual strength at the corresponding shear displacements. The proposed model shows very good agreement with the laboratory results and is capable of predicting the shear behavior of rock joints. Following the development of the empirical model, the investigations focused on the model by Saeb and Amadei (1992) and a mathematical model was proposed to consider both the shear and normal stiffness behavior of rock joints under constant normal stress and conditions. In this study, the proposed mathematical model was based on the aforesaid empirical model instead of the Goodman model. The capability of the proposed mathematical model to predict the behavior of rock joints under constant normal stiffness conditions was assessed using the laboratory results reported by Seidel and Haberfield (2002a,b). A case study was conducted to examine the appropriateness of the clinical nurse leader (CNL) role and its use within the Alabama Department of Public Health. The role of the CNL to provide leadership within the public health environment is examined. Using the aspects of the CNL training toward improvement of patient outcomes and prevention of errors through coordination was the key outcome of the case study. The role of the CNL was used to provide evaluation while promoting progress and improvement of planning efforts with the performance improvement planning microsystem. The article will discuss and evaluate various functions of the CNL role as it is adapted to the public health environment. The CNL role, as described in the case study furthermore, may be implemented in a wide variety of similar situations through the nation's health settings. [ABSTRACT FROM AUTHOR]

Abstract: Our understanding of inducible plant defense responses was greatly increased by the characterization of disease resistance genes from a variety of plant–pathogen interaction models. In the paper we aimed to characterize conserved motifs in the NBS domains, to determine their evolutionary fate. Using combined iterative data-mining approaches we effectively expanded the dataset, but the number of NBS-LRR sequences obtained was much lower than projected. We interpret this as evidence supporting low basal transcription levels as expected for R genes situated at the very start of signal amplification cascades. Motif analysis showed all key motifs of the NBS-LRR CNL domain present, with no evidence for TNL type sequences as previously observed for monocotyledonous taxa. Significant overlap between Triticeae CNL members and CNL R genes from other taxa was found. Three recently diverged clades of paralogous NBS-core sequences for barley was tested for gene-conversion events, but none was found, contradicting results from Arabidopsis where even ectopic gene-conversion events were detected. Non-synonymous to synonymous substitution rate ratio tests for comparing evolution of recent paralogous and homeologous duplications showed that the NBS-core domain of three barley paralogue clades were under strong purifying selection in contrast to the wheat go35 CNL gene, where four different non-synonymous substitutions were found. Only synonymous differences were seen between sequences obtained from two diploid wheat ancestors (A. tauschii and A. speltoides). We consider this as evidence supporting a divergence-before-duplication model of R gene evolution. [Copyright &y& Elsevier]

The study of plant diseases is almost as old as agriculture itself. Advancements in molecular biology have given us much more insight into the plant immune system and how it detects the many pathogens plants may encounter. Members of the primary family of plant resistance (R) proteins, NLRs, contain three distinct domains, and appear to use several different mechanisms to recognize pathogen effectors and trigger immunity. Understanding the molecular process of NLR recognition and activation has been greatly aided by advancements in structural studies, with ZAR1 recently becoming the first full-length NLR to be visualized. Genetic and biochemical analysis identified many critical components for NLR activation and homeostasis control. The increased study of helper NLRs has also provided insights into the downstream signaling pathways of NLRs.
