ABSTRACT Knowledge on the viability of the seeds is important in the management of the Germplasm Bank, as it allows monitoring the longevity and the need for regeneration of the accessions. The objective was to validate the efficiency of the Gomez-Campo methodology for seed conservation by evaluating physical dormancy and physiological quality of 11 Stylosanthes spp. accessions stored between seven and 13 years in the Forage Germplasm Bank of the State University of Feira de Santana (FGB-UEFS). One-hundred seed were used to determine moisture degree (%) and the viability with the descriptors: percentage of germination (G), germination speed index (GSI), synchronicity (Z) and average germination time (AGT). Sowing was done in plates moistened with water, arranged under controlled conditions (BOD chambers: at 20/30 °C and 12 hours/light) and in a completely randomized design in a 2 x 11 factorial scheme (scarified/non-scarified seeds and 11 accessions). After 14 days, it was observed that most of the accessesion presented high G (greater than 70%), GSI less than 2 seeds.day-1, Z with a mean value of 0,52 and AGT with a mean value of 2 days, characterizing a synchronous and rapid event. Thus, the methodology used in storage is efficient for the conservation of Stylosanthes seeds in the medium term.

Tetrazolium test (TZT) can quickly evaluate in detail the viability and vigor of seeds. This study aimed to determine the optimal conditions for conducting the TZT on seeds of Handroanthus spongiosus. For this purpose, seeds from three lots were pre-soaked in water for 16 h, followed by extraction of the tegument and immersion in tetrazolium salt solutions at different concentrations (0.01–0.1%), for increasing periods (1–4 h) and at 30 °C in the dark. The experimental design was completely randomized with a 4 × 4 factorial scheme with 25 seeds per repetition. We applied generalized linear models and the Tukey test for pairwise comparisons of the means at 5% probability. The viability/vigor results were compared with data obtained from the germination test at 25 °C using a subsample of seeds from the same lots. The time (1 h to 4 h) of immersion of the seeds in tetrazolium salt solutions did not cause a clear coloration difference. The seeds subjected to all treatment concentrations for 3 h presented average viability greater than 60%, with no difference in germination percentage. The TZT at 0.01% tetrazolium salt solution for 3 h was most efficient in assessing the viability of the Handroanthus spongiosus seeds.

Abstract: Erythrina velutina Willd is a native Fabaceae with wide occurrence, mainly in the Caatinga Domain with multiple uses. This study aimed to compare the vulnerability/tolerance of seeds of four accessions of E. velutina to osmotic stresses. Four replications were conducted with 25 seeds, germinated in osmotic solutions of NaCl or PEG up to -1.73 MPa. Seeds that did not germinate after 14 days were recovered in distilled water. Obtained data were fitted in Boltzmann curves and t50 and synchrony were evaluated. A probit analysis was performed, and hydrotime and halotime models were constructed. In distilled water, the germination curve was similar for all accessions. When the concentration of salt or PEG in the substrate increased, the accessions differentiated into two groups. Two accessions - Jutaí 2012 and Jutaí 2015 - maintained high germination up to -0.86 MPa NaCl, while Caboclo 2008 and Caiçara 2011 accessions presented a lower germination speed. The hydrotime and halotime analyses separated the germination response of accessions according to their physiological quality and tolerance to osmotic and saline conditions. Seeds viability was maintained at both stresses, since germination was reestablished at distilled water, thus constituting a crucial response for this species’ survival and conservation.

Estudos de maturação são importantes para definir o momento de colheita, para tanto objetivou-se avaliar a maturação de frutos e sementes de Lecythis pisonis. Estudou-se frutos e sementes 1, 2, 3, 4 e 5 meses de idade, avaliou-se a morfologia e a morfometria, a massa das sementes nos frutos (%), o teor de água (%) e a massa seca (g) e a composição química das reservas nas sementes com 4 e 5 meses. No teste de emergência determinou-se as plântulas emergidas, a protrusão das raízes, a mortalidade e a dormência. Os frutos tornaram-se elítico-ovóides após 2 meses, distinguindo a região mediana em que está presente a calicina e uma constrição que marca o opérculo. As sementes ariladas se formaram em quatro lóculos. As sementes inicialmente hialinas, tornaram-se marrons aos 4 meses e o embrião foi observado após 3 meses, mantendo-se indiferenciado e envolto pelo endosperma, com reservas lipídicas. Sementes com 4 meses apresentaram 60,3% de água, a máxima massa seca (3 g), a máxima emergência (67,0 ± 15,3%) e a menor mortalidade (9%), contudo, exibiram 9% de dormência. A maturidade morfológica de frutos e sementes é alcançada aos 4 meses, mas a maturidade fisiológica das sementes ocorre aos 5 meses.

As espécies nativas carecem de informações sobre a maturação de seus frutos e sementes. Objetivou-se avaliar a maturação de frutos e sementes de inhaíba (Lecythis lurida). Fez-se a marcação de flores em antese e a posterior colheita de frutos e sementes após 1, 2, 3, 4 e 5 meses, quando foram analisados e feita a descrição morfológica. Aos 4 meses de idade, os frutos de semiglobosos passaram a ter o formato cilíndrico-comprimidos. As sementes apresentaram um arilo vestigial, além do tegumento inicialmente de cor bege e que tornou-se marrom a partir do terceiro mês. O embrião branco manteve-se envolto pelo endosperma e não apresentou diferenciação morfológica. O máximo desenvolvimento morfológico das sementes ocorreu aos 4 meses, quando apresentaram as maiores dimensões (50,9 x 50,8 x 43,3 mm), as máximas germinação (84,8%) e massa seca (32 g), a menor mortalidade das sementes (19%), caracterizando a maturidade fisiológica e o momento de colheita. Nesse momento, os teores de N P K eram menores que 5% e o amido tornou-se o componente predominante nas reservas (47,8% da massa seca). As sementes maduras representam mais de 50% da massa dos frutos e apresentam dormência embrionária e resistência tegumentar que atrasam a emergência das plântulas.

A disponibilidade hídrica é um dos principais fatores ambientais capaz de influenciar o processo de germinação de sementes. Nessas condições, a habilidade das sementes em manter sua viabilidade durante o armazenamento pode ser uma vantagem adaptativa de espécies encontradas em diferentes biomas, especialmente na caatinga. Objetivou-se, com este trabalho, avaliar a influência do armazenamento na tolerância das sementes de catingueira (Caesalpinia pyramidalis Tul.) à restrição hídrica, utilizando-se como agente osmótico o polietilenoglicol 6000 (PEG 6000). Sementes de catingueira foram armazenadas por 0, 3 e 6 meses em temperatura ambiente e acondicionadas em sacos de papel kraft, avaliando-se a influência dos diferentes potenciais osmóticos (0,0; -0,2; -0,4; -0,6; -0,8; -1,0 e -1,2 MPa) das soluções de PEG 6000 sobre a porcentagem de germinação (G%), no tempo médio de germinação (Tm), na velocidade média de germinação (Vm) e no índice de velocidade de germinação (IVG) das sementes. Observou-se que a interação entre o armazenamento e os tratamentos osmóticos influenciou significativamente as variáveis cinéticas de germinação avaliadas, exceto a Vm. Em todos os meses avaliados, a redução da água disponível no meio afetou no decréscimo da G%, da Vm e do IVG e na elevação do Tm das sementes. A tolerância à deficiência hídrica nas sementes de catingueira foi relativamente alta, tendo como limite a faixa entre -0,8 e -1,0 MPa. A identificação dos fatores ambientais que governam a germinação das sementes, como a disponibilidade hídrica, representa uma importante ferramenta na interpretação do comportamento ecológico das espécies em hábitats naturais.

Objetivou-se com este trabalho apontar as melhores condições de armazenamento das sementes de C. pyramidalis (catingueira), visando ao estabelecimento de protocolos que possam servir de base para outras espécies do semiárido nordestino brasileiro. Sementes de catingueira foram armazenadas por 0, 3, 6 e 9 meses em ambiente de laboratório e em geladeira, nas embalagens saco de papel Kraft e saco de polietileno. A cada período de armazenamento, as sementes foram avaliadas quanto ao percentual de germinação (G%), ao tempo médio de germinação (Tm), à velocidade média de germinação (Vm), ao índice de velocidade de germinação (IVG) e ao conteúdo de água (Ca). Observou-se elevada taxa de germinação das sementes nos diferentes tratamentos propostos aos três (89,25%), seis (88,5%) e nove (91%) meses de armazenamento, sendo tais índices, inclusive, muito próximos ou ligeiramente superiores à taxa de germinação inicial (90%). O armazenamento favoreceu significativamente a elevação das taxas de IVG das sementes para índices superiores àquele verificado nas sementes recém-coletadas (11,24 aos 3 meses; 9,79 aos 6 meses; 10,29 aos 9 meses e 7,70 no mês zero). Os resultados indicaram que as sementes de catingueira possuem comportamento ortodoxo quanto à tolerância à secagem e ao armazenamento e que a possibilidade de manutenção de sementes viáveis em diferentes ambientes a um baixo custo pode ser uma boa estratégia para fornecimento de sementes em qualquer época do ano.

Abstract: Myracrodruon urundeuva, a native species from the Brazilian Caatinga, is widely distributed across its endemic region, where it also plays an essential socioeconomic role. The objective of this study was to evaluate the influence of environmental stress on the germination of M. urundeuva seeds harvested in different years (2010 to 2013). Seeds were germinated at constant temperatures between 10 to 40 °C, osmotic potentials from 0 to -0.8 MPa (in polyethylene glycol 6000 solutions), and from 0 to -0.5 MPa (in NaCl solutions). The experiment was conducted according to a completely randomized design, with three replicates of 50 seeds, in a factorial scheme (harvest year x stress intensity) for each environmental stress. Germination data were then analysed using thermal, hydro and halo time models, and future germination responses projected according to climate change scenarios. The germination thermal thresholds ranged from 7.4 to 53.3 oC. The germination base osmotic threshold (using polyethylene glycol) was -0.6 MPa and the base osmotic threshold in NaCl was -0.43MPa. Seeds from different harvest years showed distinct tolerance to environmental stresses. The thermal, hydro and halo-time models were efficient to describe the germinative response of seeds, and the climate models allowed to identify the germination responses of M. urundeuva in future climate. According to the models for future climate (RCP 8.5), the reduction of rainfall by 2100 will directly affect seed germination and seedling recruitment of M. urundeuva.

Abstract: The aim of this study was to evaluate the effects of storage conditions on the germination of A. cearensis seeds. The experimental design was completely randomized into split plots over time with four replicates. The storage conditions of the airtight containers in the refrigerator and laboratory, paper bags in the laboratory and liquid nitrogen were assessed for 27 months. In the laboratory, we evaluated the germination, the germination rate, uniformity of germination, and total soluble and reducing sugars in the radicle. In the greenhouse, we evaluated seedling emergence, emergence rate and height of 30-day-old seedlings. Seeds stored in the refrigerator maintained a high initial germination rate, which decreased from the 21st month. Seeds stored in paper bags in the laboratory showed low emergence and small seedlings. Total soluble sugars and reducing sugars were mobilized when the seeds were stored at low temperatures. Thus, it is not advisable to store A. cearensis seeds in a laboratory environment without airtight containers. A. cearensis seeds kept in a refrigerated environment maintained their viability for at least two years.

Physalis ixocarpa is an edible fruit with great economic importance in many countries. The use of high-quality seeds is essential for the success of the crop, so their harvest should be carried out at their maximum point of physiological maturity. This study aimed to verify the influence of the growing season and fruit maturation stage on the seed physiological maturity of two P. ixocarpa varieties (‘green’ and ‘purple’) grown in the Brazilian semi-arid region. The water content, dry weight, germination percentage and seedling emergence were evaluated. The growing season influenced the seed maturation process of the two evaluated varieties, with a more pronounced effect for the ‘purple’ one. To obtain higher-quality seeds, the cultivation in the Brazilian semi-arid region can be carried out from May to August and the collection of fruits to obtain seeds from the stage 4 of maturation.

Abstract: Handroanthus spongiosus (Rizzini) S. Grose is an endangered tree species. However, its seed quality, storage, and conservation strategies are issues still unexplored. This study aimed to evaluate the physiological quality of H. spongiosus seeds subjected to different storage times, packaging, and environments for their conservation. A completely randomized experimental design was used, in a double factorial arrangement with an additional treatment (recently-harvested seeds), consisted of five storage times (up to 24 months) and six storage conditions, combining packaging types (permeable and impermeable) and environments (room, cold chamber, freezer, and liquid nitrogen conditions). Seed germination percentage and normal seedling percentage, shoot length, root length, and root to shoot dry weight ratio were evaluated. The seed germination and normal seedling percentages of H. spongiosus seeds conserved under room conditions decreased over the storage time. Normal seedling percentages decreased from the 12th month of storage onwards. Low and ultralow temperatures are recommended for short and medium-term conservation of H. spongiosus seeds, since they did not affect the growth of seedlings.

The Physalis ixocarpa species has an interesting performance in hot and dry regions. However, there has been a lack of information about the influence of edaphoclimatic conditions, during the growing period, on the physical and physicochemical characteristics of the fruits. This study aimed to evaluate the characteristics of ‘purple' and ‘green' fruits of the P. ixocarpa species, according to the growing season, in the semi-arid region of the Bahia state, Brazil. A randomized block design was used, in a 2 x 2 x 5 factorial scheme (variety x growing season x fruit maturation stages), with three replications. The following characteristics were evaluated: color, fresh weight, length, diameter and total soluble solids contents of the fruits. The changes in the characteristics of the fruits follow the same tendency during maturation, for both evaluated varieties, with the best characteristics being observed when the calyx is green, with dry parts, completely expanded and ruptured. The growing season has a significant effect on the characteristics of the fruits, with May to August presenting the best environmental conditions for growing in the semi-arid region of the Bahia state.

The important chemical and food properties conferred to P. angulata make it necessary to conduct studies in seed conservation. This study evaluated the effect of priming of P. angulata seed for varying periods and storage conditions. Lots of seeds were stored in a refrigerator and in ambient conditions for up to 24 months. Some of these seeds were primed before or after storage. The results show that there was variation in moisture content. The germination rate and germination rate index remained high in ambient conditions when primed up to 24 months.

Also, different temperatures were combined with different osmotic potentials. The seed germination Myracrodruon urundeuva starts in two days. At the temperature of 250C, it was obtained for maximum seed germination of the species at 330C for 50% of the seeds germinated and at 400C, the germination was zero. Regarding to water restriction induced -0.2 MPa, it was an optimum germination potential, the potential of -0.7 MPa 50% germination and from -0.9 MPa and germination was nil. The osmotic potentials of -0.9, -1.0 and -1.2 MPa, can be used for priming this species. The seeds of Myracrodruon urundeuva not tolerate the stresses of high temperatures and decreased water potential, concurrently, as they reduce the germination and seed vigor.

Studies of seed physiological processes represent the starting point for the sustainable utilization of native or cultivated plant species from the caatinga biome for which germination studies are still scarce. In order to determine adequate methods for the conservation of Gliricidia sepium seeds, the physiological quality of propagules exposed to different conditions and times of storage were examined. Recently collected seeds of G. sepium were analyzed immediately, while the other sample was stored for three, six, nine, or twelve months in plastic or paper bags under refrigeration or at room temperature for further analysis. The germination rates of seeds from each storage regime were determined and these rates compared to their water content. It was observed that germination capacity was directly related to changes in seed humidity. Storing seeds in plastic bags yielded the best conservation, mainly when they were stored at low temperatures. However, even at temperatures near 25°C it was possible to prolong seed viability when their water content was controlled through the use of impermeable packing.

O caroá é uma Bromeliaceae nativa da Caatinga brasileira cujas fibras retiradas das folhas geram trabalho e renda para diversas famílias nordestinas com a fabricação de vários produtos artesanais. A propagação ocorre via sementes, que podem servir como alimento para muitos animais e pássaros. Diante de sua importância econômica regional, a planta do caroá tem sido coletada na caatinga de forma extrativista, já tendo praticamente desaparecido em algumas regiões da Bahia. Objetivou-se, com o desenvolvimento deste trabalho, avaliar o efeito da temperatura e da restrição de água na germinação de sementes de Caroá. A germinabilidade foi avaliada sob as temperaturas de 25, 30, 34 e 37° C, sendo que a temperatura mais favorável à germinação foi utilizada no ensaio seguinte, combinando com diferentes soluções-teste de polietilenoglicol. As maiores taxas de sementes germinadas (> 66%) ocorreram quando as temperaturas do meio eram mais elevadas, sendo que a de 30° C foi a que proporcionou a maior velocidade média de germinação (0,12 dias-1) e índice de velocidade de germinação; por sua vez a germinação decresceu com a diminuição do potencial osmótico do meio. Sob condições de restrição hídrica, as sementes de caroá mostraram capacidade germinativa até o potencial de -0,6 MPa.Caroá is a native Bromeliaceae of the Brazilian savanna. It generates work and income for many Northeastern families when used as source of fiber for producing craft products. The species can be propagated from seeds and it serves as food for a variety of animals. As a result of its economic importance to the region, the caroá plant has been indiscriminately harvested in the savanna and has practically disappeared in some areas of Bahia State, Brazil. The objective of this research was to study the effect of hydric restriction on the seed germination of caroá, as a proposed means of establishing a cultivation system and more rational production methods. Germination was tested at temperatures of 25, 30, 34 and 37° C, with the optimum temperature for germination used in a subsequent test, combining different test solutions of polyethylene glycol. The highest rates of seed germination (> 66%) occurred when the ambient temperature was higher, and 30° C yielded the highest average speed of germination (0.12 day-1) and germination speed index. On the other hand, germination rates decreased as osmotic potential of the environment decreased. Under conditions of hydric restriction, seed germination of caroá showed the potential to reach up to -0.6 MPa.

O gênero Physalis pertencente à família Solanaceae, possui 110 espécies, das quais 11 ocorrem no Brasil. Physalis angulata L. destaca-se no gênero pela presença de vitaesteróides, principalmente as fisalinas, encontradas em raízes, caules e folhas, com propriedades imunomoduladora, antimicrobiana e anticancerígena. Este trabalho teve por objetivo avaliar a germinabilidade de sementes osmocondicionadas e não osmocondicionadas e o crescimento inicial de P. angulata em ambientes salinos. Foram realizados experimentos nos quais as sementes foram submetidas ao osmocondicionamento (PEG 6000) e colocadas para germinar em soluções com diferentes concentrações de NaCl (2, 4, 6, 10, 12, 14 e 16 dS m-1). O controle (0 dS m-1) foi realizado com água destilada. Os parâmetros avaliados foram a germinabilidade das sementes, o índice de velocidade de germinação das sementes (IVG), o coeficiente de uniformidade de germinação e a massa seca total das plântulas. Houve diminuição da germinabilidade e do IVG, em ambos os tratamentos. No entanto, as sementes osmocondicionadas apresentaram maior tolerância ao estresse salino. Apesar da redução significativa da massa seca em relação ao aumento da salinidade do meio, o osmocondicionamento como tratamento pré-germinativo poderia ser utilizado para favorecer a tolerância ao estresse salino, uma vez que a germinabilidade foi maior para sementes osmocondicionadas.The genus Physalis belongs to the Solanaceae family, with 110 species, of which 11 occur in Brazil. Physalis angulata L. stands out in the genus due to the presence of vitasteroids, especially physalins, found in roots, stems and leaves, showing antimicrobial, anticancer and immunomodulatory properties. This study aimed to evaluate the germination of osmotic priming treatment and non-primed seeds and the initial growth of P. angulata in saline environments. The seeds were subjected to priming (PEG 6000) and incubated in solutions with increasing concentration of NaCl (2, 4, 6, 10, 12, 14 and 16 dS m-1). Distilled water was used as control (0 dS m-1). The parameters evaluated were seed germinability, rate of germination, coefficient of uniformity and total dry mass of seedlings. A decrease in germinability and IVG was observed in both treatments. However, primed seeds showed higher tolerance of salt stress. Despite the significant reduction in dry mass with increasing salinity, priming as a pre-germinative treatment could be used to foster tolerance, since germinability was higher for primed seeds.

O gênero Calliandra Benth. (Leguminosae) possui 134 espécies exclusivamente neotropicais, das quais 42 ocorrem na Chapada Diamantina, sendo 32 endêmicas da área. O objetivo deste trabalho foi avaliar o efeito da luz e substrato na germinação e desenvolvimento inicial de C. viscidula e C. hygrophila. As sementes foram submetidas a três tratamentos em laboratório: luz, escuro e luz/escuro (12 h/12 h). Em viveiro foram utilizados três substratos: (I) areia; (II) terra vegetal + areia; (III) vermiculita + terra vegetal + areia, mantidos a 100%; e 70% de luminosidade. As avaliações da germinação foram diárias durante 21 dias, utilizando-se os parâmetros: germinabilidade, índice de velocidade de germinação (índice de velocidade de emergência para viveiro), tempo médio de germinação e frequência relativa. Os parâmetros analisados no desenvolvimento inicial foram: comprimentos da parte aérea e raiz, número de folhas, massa seca, relação parte aérea/sistema radicular e razão do peso da folha. C. viscidula e C. hygrophila foram classificadas como fotoblásticas neutras e apresentaram melhor desenvolvimento a 70% de luminosidade nos substratos III e I e II , respectivamente.

Physalis angulata é conhecida como camapu, mullaca ou juá-de-capote e pertence à família Solanaceae. Esta espécie possui importância medicinal, pela presença dos vitaesteróides conhecidos como fisalinas e, atualmente, na alimentação. Essa espécie se propaga facilmente por sementes, motivo pelo qual ela é tida como infestante de outras culturas. Nesse sentido, a identificação da espécie ainda nos estágios iniciais de crescimento confunde com as demais infestantes, tornando difícil a sua caracterização taxonômica. Assim, o objetivo deste trabalho foi caracterizar morfologicamente as sementes e o desenvolvimento pós-seminal de Physalis angulata. Os ensaios foram desenvolvidos no Laboratório de Germinação da Unidade Experimental Horto Florestal, Feira de Santana - BA. Para tanto foram realizadas análises morfométricas das sementes (4 repetições de 25) e analisadas suas características externas e internas com auxilio de microscópico estereoscópico. Para a descrição das plântulas da espécie foram utilizadas placas de petri contendo duas folhas de papel filtro (4 repetições de 20 sementes) e umedecidos com água destilada. As sementes foram colocadas em câmara de germinação à 35ºC (fotoperíodo de 12 horas) até a emissão dos cotilédones. A metodologia para descrição do desenvolvimento pós-seminal foi realizada em condições de casa de vegetação e consistiu em avaliações realizadas cinco dias após a semeadura (emissão dos cotilédones) e no intervalo de até 15 dias (emissão de eófilos e metáfilos) no qual foram utilizados 2 repetições de 20 vasos. As sementes apresentaram em média 1,55 mm de comprimento, 1,26 mm de largura e 0,43 mm de espessura. O período de análise foi suficiente para caracterizar as sementes e plântulas da espécie, permitindo a adoção de critérios de anormalidade.Physalis angulata is also known as camapu, mullaca or juá-de-capote it belongs to the Solanaceae family. This species has its medicinal importance due to vitasteroids presence known as physalins, and currently due to its use as food. It propagates readily by seed, which is the reason why it is regarded as weeds in other crops. Accordingly, the identification of that species in the early stages of growth is indistinguishable from other weeds, hindering taxonomic characterization. Thus, the objective of this study was to morphologically characterize seeds and post-seminal development of Physalis angulata. The tests were developed in the Laboratory of Germination of the Experimental Unit/Nursery, in Feira de Santana/BA. Therefore, we have performed morphometric analysis of the seeds (4 replicates of 25 seeds) and we have analyzed their external and internal features with the aid of a stereoscopic microscope. For the description of the species seedlings it was used Petri dishes containing two sheets of filter paper (4 replicates of 20 seeds) and moistened with distilled water. The methodology for describing the post-seminal developmente was conduced under field conditions and consisted of evaluetions performed five days after sowing (inssuance of cotyledons) and the range of up to 15 days (and inssuing eophylls metaphylls) in which two replicates were used 20 pots. Assessments were performed five days after sowing (issuance of cotyledons) and ranging of up to 15 days (emission of eophylls and metaphylls). The seeds had an average of 1.55 mm long, 1.26 mm wide and 0.43 mm thick. The period of analysis was sufficient to characterize the seeds and seedlings of the species, applied as a criteria for abnormality.

Neoglaziovia variegata (Arr. Cam.) Mez is a Bromeliaceae native to the Caatinga, used for fiber extraction in the Northeast Region of Brazil. The antropic activity has place this species among the threatened ones. The objective of the work was to establish an in vitro propagation and conservation of caroá. Seeds were cultivated in MS medium in the presence or absence of light. In vitro germinated seedlings were multiplied in MS medium supplemented with the combinations 0.05 and 0.50 µM NAA and 2.2 and 4.4 µM BAP and KIN. The best percentages of germination were obtained with the seeds incubated in the presence of light. The highest multiplication ratio was obtained for the NAA (0,5 µM) + BAP (4,4 µM) treatment and the number of roots, with NAA (0.5 µM) + KIN (2.2 µM). Plant acclimatization presented differentiated results regarding the substrates tested. Plântulas germinadas in vitro foram multiplicadas em meio MS suplementado com as combinações de 0,05 e 0.50 µM de NAA e 2.2 e 4.4 µM de BAP e KIN. Foram obtidas as melhores porcentagens de germinação com sementes incubadas na presença de luz. A taxa de multiplicação mais alta foi obtida no tratamento NAA (0,5 µM) + BAP (4,4 µM) e, o número de raízes, com NAA (0.5 µM) + KIN (2.2 µM). Aclimatização das plantas apresentou resultados diferenciados em relação aos substratos testados. A conservação foi estabelecida.

Este trabalho teve como objetivo avaliar o estabelecimento in vitro de Anadenanthera colubrina var. cebil, através das respostas morfofisiológicas das plântulas submetidas a várias condições de cultivo. As sementes foram desinfestadas e inoculadas em placas de Petri contendo papel germtest previamente esterilizado e umedecido com água estéril. As placas ficaram no escuro por dois dias até que ocorresse a germinação das sementes e, em seguida, transferidas para tubo de ensaio contendo meio de cultura WPM. Foram avaliados três tipos de fechamentos dos tubos de ensaio: filme de PVC, tampa plástica sem filme de PVC e tampão de algodão. No segundo experimento, o meio de cultura foi suplementado com carvão ativado (0,0; e 82,2 mM) e diferentes concentrações de sacarose (0,0; 29,21; 58,43; e 87,64 mM). Ao final de 30 dias, observaram-se efeitos altamente significativos dos tipos de fechamento sobre todas as variáveis analisadas: comprimento da parte aérea (CPA), número de folhas (NF), número de pinas (NP), matéria seca da parte aérea (MSPA), abscisão foliar (AF), comprimento da raiz (CR), matéria seca da raiz (MSR) e número de raízes secundárias (NRS). A AF foi reduzida em 7,3 vezes, quando os tubos de ensaio foram fechados com tampão de algodão em relação ao fechamento com filme de PVC. Os maiores valores de CPA (102,5 mm), NF (3,0), CR (194,7 mm) e NRS (47,88) foram obtidos com 29,21 mM de sacarose; a MSPA (59,1 mg), quando utilizou 58,43 mM e MSR (33,37 mg), com 87,64 mM de sacarose. O carvão ativado apresentou efeito significativo apenas em NRS, com maior valor (45,79) na presença desse composto.This work aimed to evaluate the in vitro establishment of the species Anadenanthera colubrina var. cebil, throug the morphophysiological answers of the seedlings subjected to several culture conditions. The seeds were desinfested and inoculated in Petri plates containing germtest paper, previously sterilized and imbibed with sterile water. The plates were kept in the dark for two days, until the seeds germinated. Then, they were transferred to the test tube containing WPM medium. Three types of test tube closings were evaluated - PVC film, plastic lid without PVC film and cotton lid. In the second experiment, the culture medium was supplied with activated charcoal (0,0 and 82,2mM) and different sucrose concentrations (0,0; 29,21; 58,43; 87,64mM). After 30 days, highly significant effects of the types of closure were observed on all the variables analysed - aereal part length (APL), number of leaves (NL), number of pinnas (NP), dry matter of aereal part (DMAP), leaf abscission (LA), root length (RL), root dry matter (RDM) and number of secondary roots (NSR). The LA was reduced in 7,3 times when the cotton lid was used instead of the PVC film. The greatest values for APL (102,5 mm), NP (3,0), RL (194,7 mm) and NRS (47,88) were obtained with 29,21mM of sucrose. For DMAP (59,1 mg), when it used 58,43mM, and for RDM (33,37 mg), with 87,64mM of sucrose. The activated charcoal showed significant effects only for the variable SRN, presenting the greatest value (45,79) in the presence of this compound.

Plantas de mandioca (Manihot esculenta Crantz) apresentam redução no acúmulo de matéria seca quando cultivadas com altos níveis de amônio na solução do solo. A razão para esse efeito do amônio ainda permanece pouco estudada. O presente trabalho foi desenvolvido visando avaliar a influência do NO3- e do NH4+ sobre alguns aspectos relacionados à fotossíntese e à concentração de alguns compostos nitrogenados em plantas de mandioca. Para esse estudo, utilizou-se a variedade Cigana Preta, que foi cultivada em solução nutritiva, tendo areia grossa lavada como substrato. Os tratamentos consistiram de três proporções entre NO3-:NH4+, em mM (12:0, 6:6, e 0:12). Plantas cultivadas apenas com NO3- ou com NO3- + NH4+ não apresentaram diferenças na taxa fotosssintética. No entanto, observou-se redução desse evento quando o NH4+ foi a única fonte nitrogenada. O valor da condutância estomática obtido para as plantas cultivadas apenas com NO3- foi de 0,49mol m-2s-1, enquanto que para as plantas cultivadas apenas com NH4+, o valor foi de apenas 0,16mol m-2s-1 (três vezes menor). Não houve diferenças significativas na concentração foliar de amônio livre, sugerindo não ter ocorrido efeito tóxico direto do NH4+ sobre a fotossíntese. A concentração de proteínas foi maior para as plantas cultivadas apenas com NH4+. Os resultados sugerem que o menor acúmulo de matéria seca das plantas de mandioca cultivadas exclusivamente com NH4+ é decorrente, dentre outros fatores, do efeito desse íon sobre a atividade fotosssintética, em virtude, principalmente, de sua ação negativa sobre a condutância estomática.Cassava (Manihot esculenta Crantz) plants present reduction in dry matter accumulation when grown at high levels of ammonium in the soil solution. The reason for that still remains to be determined. The present research was carried out in order to evaluate the influence of nitrate and ammonium on aspects related to photosynthesis and the concentration of nitrogen compounds of cassava plants. Cultivar Cigana Preta plants were grown in nutritive solution with sand as substrate. Treatments consisted of three ratios of NO3-:NH4+, in mM (12:0, 6:6, e 0:12). Plants grown only on NO3- or NO3- + NH4+ did not show differences in photosynthetic rates, but reduction of them were observed for NH4+ as the only nitrogen source. Stomatal conductance of plants grown on nitrate was 0.49mol m-2s-1 and three times less for plants grown only on ammonium (0.16mol m-2s-1). There were no significant differences in leaf concentration of free ammonium among treatments, suggesting no direct toxic effect of ammonium on photosynthesis. Protein concentration was higher for plants grown only on ammonium. The results strongly suggest that the lower dry matter accumulation of cassava plants grown just on ammonium is due, among other factors, to its effect on photosynthetic activity, mainly because of its negative influence on stomatal conductance.

The work had as objective to evaluate the germination of seeds of sugar apple (Annona squamosa L.), submitted the treatments daily pre-germinative, under controlled conditions. One evaluated the percentage of germination and the index of germination speed (IVG), using the delineation entirely casualizado with four repetitions of 25 seeds, in factorial project 2x5, being the factor scarification mechanics in two levels (with and without scarification) and factor imbibition in five levels: water, Stimulate® the 20 mL L-1, acid giberélico 50 and 750 GA3 mg L-1, all absorbing for 12 hours, and a treatment without imbibition. The scarification mechanics was carried through with sandpaper for wood. The results had shown that the scarification mechanics separately, did not make possible the overcoming of the dormancy in seeds of sugar apple. Significant alterations with the use of the sandpaper had been verified with the embebição of seeds scarification in water and Stimulate®, with increases of 22 and 43 %, respectively. The scarification with sandpaper revealed unnecessary with the imbibitions of the seeds in 750 dosages of 50 and mg L- 1 of acid gibberellic, with percentage of germination varying of 83 and 98%. The Stimulate®, in the used concentration, was inferior to the GA3 use, providing, however, increase of 43 % in relation to the germination of scarification seeds, without imbibitions the biggest indices of germination speed had been verified in the treatments with GA3, observing itself rise of the values with the increase of the dosage of acid gibberellic, and also with the use of the sandpaper.

O trabalho teve por objetivos controlar a abscisão foliar de plântulas de angico utilizando AgNO3 e CoCl2 e avaliar o efeito do paclobutrazol sobre o comportamento in vitro das plântulas. As sementes foram desinfestadas e inoculadas em placas de Petri contendo papel germtest, previamente esterilizado e umedecido com água estéril. As placas ficaram no escuro por dois dias até que ocorresse a germinação das sementes e, em seguida, foram transferidas para tubo de ensaio contendo meio WPM. Todo esse material foi mantido em sala de crescimento com temperatura de 25 ± 2 ºC. No primeiro experimento, o meio de cultura foi suplementado com dois tipos de inibidores de etileno (AgNO3 e CoCl2) em diferentes concentrações (0,0; 5,0; 10,0; 20,0; e 40,0 µM). No segundo, o meio foi suplementado com diferentes concentrações (1,7; 3,4; 6,8; e 13,6 µM) de paclobutrazol. Verificou-se que houve aumento no número de folhas e redução na abscisão foliar com a utilização de 10 e 20 µM (respectivamente), independentemente do inibidor de etileno utilizado. Tanto o AgNO3 quanto o CoCl2 contribuíram com acréscimos no número de gemas e no número de brotações. O aumento na concentração de paclobutrazol reduziu o crescimento das plântulas, obtendo-se menores médias das seguintes variáveis: comprimento da parte aérea, número de folhas, número de folhas senescentes e matéria seca da parte aérea na maior concentração (13,6 µM) de paclobutrazol. Esse inibidor de crescimento teve efeito também no sistema radicular, reduzindo o comprimento da raiz e o número de raízes secundárias, no entanto promoveu o engrossamento das raízes.This work aimed to control leaf abscission of seedlings of angico using AgNO3 and CoCl2 and evaluate the effect of the paclobutrazol on the in vitro behavior of seedlings. The seeds were disinfected and inoculated in Petri plates containing germtest paper, previously sterilized and imbibed in sterile water. The plates were kept in the dark for two days, until seed germination. Seeds were transferred to test tubes containing WPM medium. In the first experiment, the culture medium was supplemented with different concentrations of (0.0; 5.0; 10.0; 20.0; 40.0 µM) and types (AgNO3 and CoCl2) of ethylene inhibitors. In the second experiment, the medium was supplemented with different concentrations (1.7; 3.4; 6.8; 13.6 µM) of paclobutrazol. There was increase in the number of leaves and reduction in foliar abscission with 10 and 20 µM respectively, independent of the used ethylene inhibitor. Both the AgNO3 and the CoCl2 helped to increase the number of buds and shoots. The increase in the concentration of paclobutrazol reduced seedling growth, with lower means for the variables - length of aerial part, number of leaves, number of senescent leaves and dry matter of the aerial part in the highest concentration (13.6 µM) of paclobutrazol. This growth inhibitor had also effects on the root system, reducing root length and number of secondary roots, however promoting root thickening.

Objetivou-se neste trabalho, avaliar o efeito da nutrição nitrogenada sobre alguns aspectos relacionados à taxa fotossintética em plantas de mamão da variedade "Golden". As plantas foram cultivadas em vasos sob condições de casa de vegetação, em soluções nutritivas contendo três concentrações de N-N3- (1,0; 5,0 e 8,0molm-3). O delineamento experimental foi em blocos casualizados, com sete repetições. As avaliações foram realizadas 61 dias após o início do experimento. Observou-se que a deficiência de nitrogênio reduziu a matéria seca total; no entanto, a relação raiz:parte aérea foi maior nessas plantas. A taxa fotossintética líquida foi reduzida pela menor disponibilidade de nitrogênio. Os resultados observados para a condutância estomática e a correlação negativa encontrada entre a taxa fotossintética e a concentração interna de CO2 claramente indicam que a menor taxa fotossintética não foi devida à limitação estomática. Adicionalmente, a redução verificada nas concentrações de pigmentos foliares, como as clorofilas a e b e os carotenóides, também pode ter contribuído para a menor taxa fotossintética e o menor crescimento das plantas. A taxa transpiratória foi maior para as plantas cultivadas sob estresse; como conseqüência, observou-se, para essas plantas, menor ganho de CO2 por unidade de água transpirada.This research was aimed at evaluating the effect of nitrogen (N) levels on some aspects related to the photosynthetic rate in plants of papaya Cv. Golden. The plants were cultivated under greenhouse conditions on different nutritional solutions with three concentrations of N-N3- (1.0; 5.0; 8.0molm-3). The experiment was carried out under a randomized blocks experimental design, with seven repetitions. The evaluations occurred on the 61st day after the beginning of the experiment. The nitrogen deficiency reduced the total dry mass; however, the relation root:shoot was highest for these plants. The photosynthetic rate was reduced by the lesser nitrogen availability. The results observed for the stomatal conductance and the negative correlation found between the photosynthesis and the internal concentration of CO2, indicate clearly that the reduced photosynthetic rate was not due to stomatal limitation. Additionally, the reduction verified in leaf pigments, such as chlorophylls a and b and carotenoids also could have been contributed for the reduction on photosynthesis rate and growth of these plants. Transpiratory rate was higher for the plants cultivated under stress; as consequence it was observed, for these plants, minor CO2 profit for unit of transpirated water.

Foi estudado o efeito de níveis de sombreamento sobre o desenvolvimento de mudas de maracujazeiro, no período de 12/05 a 29/06 de 2004, na Universidade Estadual do Sudoeste da Bahia, em Vitória da Conquista, Estado da Bahia. Mudas com 45 dias após aemergência foram transplantadas e submetidas a diferentes níveis de redução da radiação solar, quais sejam: 0% (pleno sol), 30%, 50% e 70%. Foi utilizado o delineamento experimental em blocos casualizados, com quatro tratamentos, cinco repetições e seis plantas úteis por parcela. Foram avaliadas as características: massa seca de raízes, caules e folhas, razão parte aérea/raiz, área foliar, área foliar específica, razão de área foliar, razão de massa foliar e teor de nitrogênio total. Foi observada tendência ao acúmulo de massa secanas mudas menos sombreadas, sendo que houve aumento da razão parte aérea/raiz nas mudas sob maiores níveis de sombreamento. A área foliar, área foliar específica e razão de área foliar foram maiores nas plantas mais sombreadas, refletindo uma adaptação destas paramaximizar a absorção de luz. A razão de massa foliar não diferiu entre os tratamentos. O teor de nitrogênio total por unidade de massa foi maior nas plantas mais sombreadas, enquanto por unidade de área, decresceu.The effect of shading levels on the development of yellow passion fruit seedlings was studied, from May 12 to June 29 (2004), at theUniversidade Estadual do Sudoeste da Bahia, in Vitória da Conquista, Bahia State, Brazil. Seedlings aged 45 days after the emergency, were cultivated and submitted to different levels of solar radiation reduction: 0%, 30%, 50% and 70%. The experimental design was in randomized blocks, with four treatments, five replications and six plants for plot. The evaluation parameters were: dry mass of roots, stems and leaves, shoot/root ratio, leaf area, specific leaf area, leaf area ratio, leaf mass ratio and nitrogen content. A trend to the accumulation of dry mass in the less shaded plants was observed. Also, there was an increase of the aerial part/root ratio in the plants under higher shade levels. The leaf area, specific leaf area and leaf area ratio were higher in the shaded plants, reflecting an adaptation of these to maximize the light absorption. The leaf massratio did not differ between the treatments. The total nitrogen content per unit of mass was higher in the shaded plants, while per unit of area, a decrease was observed.

Modificações nos níveis de luminosidade, aos quais uma espécie está adaptada, podem condicionar diferentes respostas fisiológicas em suas características bioquímicas, anatômicas e de crescimento. Este trabalho teve como objetivo avaliar o crescimento inicial de plantas de licuri (Syagrus coronata (Mart.) Becc.) submetidas a diferentes condições de luminosidade. Neste estudo foram utilizadas plântulas de licuri com cinco meses de idade, cultivadas em substrato composto de terra + adubo orgânico e mantidas a 30 e 100% de luminosidade. Mensalmente foram tomadas medidas de altura, diâmetro do colo e número de folhas das plantas, durante um período de 12 meses. Ao final do experimento, determinaram-se a massa seca, a área foliar e o teor de clorofila. O maior crescimento em altura, diâmetro do colo, número de folhas emitidas e massa seca total foram verificados nas plantas submetidas a 30% de luz. A proporção de massa seca direcionada para as raízes foi maior à medida que houve aumento da intensidade luminosa. Já a parte aérea, diferentemente do sistema radicular, apresentou uma diminuição com o aumento da luminosidade. Em 100% de luz, as plantas apresentaram maior área foliar total, bem como maior razão das clorofilas a/b, enquanto a área foliar específica mostrou-se inversamente proporcional à intensidade luminosa. Com os resultados obtidos, pode-se inferir que a condição de sombreamento favoreceu o crescimento inicial das plantas de licuri.Changes in light intensity, to which a species is adapted, may condition different physiological responses in its biochemical, anatomical and growing features. This paper aims at assessing the initial growth of licuri plants (Syagrus coronata (Mart.) Becc.) subjected to different light conditions. Five-month-old licuri plants, grown in substrates of soil + organic fertilizer, were used. Plants were kept in 30% and 100% light intensity. Measurements of height, stem diameter, and number of leaves were taken monthly over a 12-month period. At the end of the experiment, dry mass, foliar area and chlorophyll contents were evaluated. Those plants submitted to 30% light intensity showed improvement in height, stem diameter, number of produced leaves, and total dry mass. Proportion of dry mass directed towards the roots was higher as light intensity increased, however, aerial part, differently from the radicular system, decreased. Under 100% light intensity, the plants showed a larger total leaf area, as well as a higher chlorophyll a/b ratio. Specific leaf area was inversely proportional to the light intensity. With the obtained results, it can be inferred that shade favored initial growth of licuri plants.

O presente trabalho teve como objetivo avaliar a influência da salinidade sobre alguns parâmetros fisiológicos do limoeiro 'Cravo' (Citrus limonia Osbeck), principal porta-enxerto da citricultura baiana. Esses estudos são importantes, porque podem viabilizar a utilização de água de baixa qualidade para irrigação, com vistas a melhoria da produtividade dessa cultura. Sementes de limão 'Cravo' foram selecionadas quanto ao tamanho e colocadas para germinar em areia lavada. Três dias após a germinação as plantas foram transferidas para vasos plásticos contendo solução nutritiva. Após um período de adaptação de dez dias, os tratamentos salinos foram induzidos pela adição de quantidades de NaCl à solução de crescimento para a obtenção das concentrações finais de 0, 20, 40 e 80 mM de NaCl. O experimento foi montado em um delineamento inteiramente casualizado, com quatro tratamentos (concentrações de NaCl) e cinco repetições. Quarenta e dois dias após o início da imposição do estresse salino o experimento foi encerrado. Observou-se que a salinidade reduziu as produções das matérias secas totais, do caule e das raízes. Nos níveis intermediários de NaCl (20 e 40 mM) nem a área foliar e nem a matéria seca das folhas foram afetadas; esses caracteres foram afetados apenas no nível de 80 mM. O aumento dos níveis de salinidade determinou reduções na relação raiz:parte aérea das plantas, na condutância estomática, na transpiração e na temperatura foliar.

Foi estudado o efeito de níveis de sombreamento sobre o desenvolvimento de mudas de maracujazeiro, no período de 12/05 a 29/06 de 2004, na Universidade Estadual do Sudoeste da Bahia, em Vitória da Conquista, Estado da Bahia. A razão de massa foliar não diferiu entre os tratamentos. O teor de nitrogênio total por unidade de massa foi maior nas plantas mais sombreadas, enquanto por unidade de área, decresceu.

O gênero Melocactus (Cactaceae) conhecido como cabeça-de-frade ou coroa-de-frade possui 36 espécies, sendo 11 endêmicas da Bahia, região onde ocorre maior concentração das espécies do gênero. O alto grau de endemismo associado a fatores como exploração de areais, degradação de habitats, queimadas na caatinga e coleta destas plantas para comercialização como ornamental tem levado algumas espécies ao risco de extinção. No habitat natural, os membros desse gênero se reproduzem exclusivamente por sementes, visto que essas espécies não ramificam, nem emitem brotos laterais. O crescimento é lento e a floração inicia-se após uma década de existência da planta, quando o cefálio desenvolve-se e começa sua fase reprodutiva. A técnica de cultura de tecidos é uma alternativa viável à propagação convencional dessas espécies por proporcionar altas taxas de multiplicação. O objetivo deste trabalho foi avaliar o potencial de formação de brotações a partir de diferentes explantes de M. glaucescens e M. paucispinus. Plantas com um ano de idade, germinadas in vitro e estioladas durante 3 meses no escuro, foram seccionadas transversalmente originando 3 tipos de explantes: apical, mediano e basal. Os explantes foram inoculados em meio de cultura MS½ sem regulador de crescimento, suplementado com 15g.L-1 de sacarose e 6g.L-1 de agar. Utilizou-se o delineamento experimental inteiramente casualizado em arranjo fatorial 2 X 3 (duas espécies e três tipos de explantes), com dez repetições de 1 explante cada. Aos 150 dias da inoculação avaliou-se a porcentagem de explantes responsivos e o número de brotos e raízes por explantes. As raízes surgiram após a 3ª semana de inoculação e as brotações após a 4ª semana. Em M. paucispinus foram obtidos 100% de explantes responsivos diferindo significativamente de M. glaucescens (50%). Apesar desse resultado, não houve diferença entre as espécies com relação à produção total de brotos, o que pode estar relacionado a presença de múltiplas brotações em M. glaucescens. Com relação ao tipo de explante, não houve diferença estatística para o número de brotações nas duas espécies. Observou-se formação de raízes em 86,67% dos explantes de M. paucispinus, enquanto que em M. glaucescens o enraizamento ocorreu em apenas 40% dos explantes. M. paucispinus apresentou um maior número de raízes em todos os tipos de explantes havendo uma tendência a maiores valores para o explante apical. Estes resultados indicam que a indução de brotações em M. glaucescens e M. paucispinus por organogênese direta pode ser obtida em meio de cultura sem regulador de crescimento a partir da fragmentação das plântulas.

The aim of this study was to evaluate the effect of different abiotic stresses on the activity of antioxidant enzymes and on accumulation of proline in Erythrina velutina Willd. seeds during germination. Mulungu seeds were scarified and placed to germinate at constant temperatures of 15, 25, and 35 ºC, moistened with distilled water, and exposed to 12 h of light. Other seeds were exposed to solutions of NaCl (EC of 0, 4, and 8 dS.m-1) and polyethylene glycol (osmotic potentials of 0.0, -0.2, and - 0.6 MPa) and maintained in a germination chamber set at 25 ºC and 12 h photoperiod for seven days. At the end of each period of imbibition, the embryonic axis and cotyledons of the seedlings were collected separately and used to quantify proline content and the activity of antioxidant enzymes. These were detected in both the cotyledons and embryonic axis of the mulungu seeds. Antioxidant activity varied depending upon the type and degree of stress applied. It was concluded that under the aspect of the detoxification process, the mechanism found in mulungu seeds is more efficient when subjected to different temperatures followed by salt stress and water stress.

ABSTRACT Physalis is represented by species that produce fruits with relevant potential for national and international marketing. The species are normally grown from seeds, so maturation studies are important to assist the point of harvest, as well as seeds with a high physiological potential, since the fruits remain enclosed within a cup throughout their development. Despite the nutraceutical importance of fruits and medicinal potential of many species of Physalisinformation on the physiological seeds quality are scarce. This study aimed to monitor the physical and physiological changes of maturation and the viability of seeds during fruit development stages. The fruits of P. ixocarpa were collected in 15, 25, 35, 45, 55 days after anthesis (DAA). Length, diameter, fresh weight, skin and cup coloring characterization, TSS content, number and weight of seeds per fruit were obtained at each development stage. We also evaluated and correlated the coloring and pigment content of the cup (chlorophyll a and b). Water content, dry matter weight and germination rate were measured from fresh and dry seeds (room conditions, 72 hours). The main physical and physiological changes during maturation occurred within 35 DAA. The maximum physiological seeds quality was achieved within 45 DAA when the cup is completely or partially broken with coloring between yellowish-green and light-yellowish-brown and the ripe fruit in dark-green.
