This canine in vivo study assessed the effect of recombinant human platelet-derived growth factor (rhPDGF) on the healing of periapical tissues following apical surgery. From a total of 96 premolar teeth, 64 teeth from six beagle dogs (2 years old) were classified as experimental and were randomly assigned to four experimental groups (16 teeth per group). After having the pulp extirpated, leaving teeth open to the oral cavity for 1 week, and sealing with an immediate restorative material for 8 weeks, nonsurgical endodontic treatment was performed. A split-mouth design was used, and intra-animal randomization of treatment sides was applied to the groups as follows: apical curettage + 1.5-mm root-end resection (Group 1); apicoectomy + mineral trioxide aggregate (MTA) root-end filling (Group 2); apicoectomy + MTA root-end filling + rhPDGF (Group 3); and apical curettage + rhPDGF (Group 4). The animals were sacrificed 24 months after apical surgery, and histologic and µCT analyses were performed for bone volume loss (BVL). Group 1 showed partial resolution of the periapical lesions without signs of tissue regeneration (BVL: 49.09 ± 10.97 mm3). Group 2 had minimal bone regeneration and showed cementum reformation in 9 teeth, with no direct attachment to the MTA (BVL: 35.34 ± 10.97 mm3). Group 3 showed regeneration of all damaged apical tissues without direct contact between the cementum and MTA (BLV: 4.51 ± 1.55 mm3). Group 4 showed regeneration of PDL, bone, and cementum and attachment of functional cementum fibers (BVL: 2.82 ± 2.3 mm3). This study aimed to evaluate the periodontal responses of subgingival proximal margins elevated using different restorative materials. A total of 120 proximal cavities with dentin/cementum gingival margins were elevated using one of four materials (resin-modified glass ionomer, glass hybrid, flowable bulk-fill resin composite, or bioactive ionic resin) and completed with the same overlaying resin composite. At 2 weeks (baseline), 6 months, 1 year, and 2 years postrestoration, periodontal parameters and the radiographic distance between the restoration margin and bone crest were evaluated. Appropriate statistical analyses were performed. The values of all periodontal parameters increased with time, although only the increases in Plaque Index and probing depth within each material group were statistically significant. There were no statistically significant differences in any of the periodontal parameters between the different materials within the same evaluation period, nor between different time points or material groups in the radiographic distances. Objective: Our research was aimed to establish the equation and to determine its precision for dental age estimation in adolescents, adults and the elderly based on the thickness of deposited dental cement. Design: The study sample consisted of 206 donor teeth of both sexes (10 to 82 years of age). The roots were transversely cut (6 cuts) on an ISOMET 1000 cutter at apical, middle, and cervical parts (slice thickness 0.3 to 0.5 μm). Measurements of cement thickness on incisions were made with a light microscope and an Olympus EP50 camera with previous microscope calibration for each measurement episode. Measurements were carried out clockwise at 4 measuring points on each incision. Statistical analyses were conducted using statistical software packages STATISTICA version 12.0 StatSoft, Inc. 2013, and MedCalc® version 22.0. Results: Mean cement thickness (MCT) significantly declined from apical to cervical cuts (P < 0.001, ANOVA) and was thicker in teeth with a destroyed tooth crown (P < 0.001, ANOVA) and in molars compared to incisors (P < 0.001, ANOVA). MCT was greater in men and in teeth with more than one root but without reaching statistical significance. MCT showed a significant increase from the second decade of chronological age with a peek at sixth decade and declined to the ninth decade (P < 0.001, ANOVA). Piecewise linear regression defined a breakpoint age of 53 years, so two different equations were calculated (≤ 53 years and > 53 years). Conclusions: The resulting equation is recommended for estimating dental age in forensic analyses. Highlights: Tooth cement is created during growth and development and throughout life. Determining the dental age is an important part of determining the dental profile. The thickness of the deposited tooth cement is different in male and female. The Scythian archaeological culture is a component of the Scythian-Siberian community of equestrian nomadic pastoralists located in the northern Black Sea region and Ciscaucasia. This paper explores whether the Scythians were present in the central Pre-Caucasus all year round or only during a part of the year and were migrating seasonally. Since Scythian settlements are not known in this region, a possible clue may come from determining the season of the year in which the Scythians buried their deceased tribesmen and performed memorial events. In this paper, we determine the seasonality of the burials and memorial events at the mound burial ground Novozavedennoye-III (southern Russia), dated to 430–300 b.c., by applying cementum increment analysis to horse teeth. This analysis has shown that the studied horses died during May–October. This result indicates that the Scythians were present in the central Pre-Caucasus foothills during some part of the warm season. [ABSTRACT FROM AUTHOR]

Treatment strategies for hard tissue defects aim to establish a mineralized microenvironment that facilitates tissue remodeling. As a mineralized tissue, cementum shares a similar structure with bone and exhibits an excellent capacity to resist resorption under compression. Macrophages are crucial for mineralized remodeling; however, their functional alterations in the microenvironment of cementum remain poorly understood. Therefore, this study explores the mechanisms by which cementum resists resorption under compression and the regulatory roles of cementoblasts in macrophage functions. As a result, extracellular vesicles from compression‐loaded cementoblasts (Comp‐EVs) promote macrophage M2 polarization and enhance the clearance of apoptotic cells (efferocytosis) by 2‐ to 3‐fold. Local injection of Comp‐EVs relieves cementum destruction in mouse root resorption model by activating the tissue repair function of macrophages. Moreover, Comp‐EV‐loaded hydrogels achieve significant bone healing in calvarial bone defect. Unexpectedly, under compression, EV secretion in cementoblasts is reduced by half. RNA‐Seq analysis and verification reveal that Rab35 expression decreases by 60% under compression, thereby hampering the release of EVs. Rab35 overexpression is proposed as a modification of cementoblasts to boost the yield of Comp‐EVs. Collectively, Comp‐EVs activate the repair function of macrophages, which will be a potential therapeutic strategy for hard tissue repair and regeneration. [ABSTRACT FROM AUTHOR]

Vital pulp therapy aims to preserve the vitality of dental pulp exposed due to caries, trauma, or restorative procedures. The aim of the present review was to evaluate the clinical, radiographic, and histological outcomes of different calcium silicate-based cements used in vital pulp therapy for both primary and permanent teeth. The review included 40 randomized controlled trials from a search across PubMed, LILACS, and the Cochrane Collaboration, as well as manual searches and author inquiries according to specific inclusion and exclusion criteria. A critical assessment of studies was conducted, and after data extraction the results were submitted to a quantitative statistical analysis using meta-analysis. The studies, involving 1701 patients and 3168 teeth, compared a total of 18 different calcium silicate-based cements in both dentitions. The qualitative synthesis showed no significant differences in short-term outcomes (up to 6 months) between different calcium silicate-based cements in primary teeth. ProRoot MTA and Biodentine showed similar clinical and radiographic success rates at 6 and 12 months. In permanent teeth, although the global results appeared to be well balanced, ProRoot MTA generally seemed to perform better than other calcium silicate-based cements except for Biodentine, which had comparable or superior results at 6 months. Meta-analyses for selected comparisons showed no significant differences in clinical and radiographic outcomes between ProRoot MTA and Biodentine over follow-up periods. The present review highlights the need for standardized definitions of success and follow-up periods in future studies to better guide clinical decisions. Despite the introduction of new calcium silicate-based cements aiming to address limitations of the original MTA. ProRoot MTA and Biodentine remain the most used and reliable materials for vital pulp therapy, although the results did not deviate that much from the other calcium silicate-based cements. Objectives: Age at death estimation is a key element to many research questions in biological anthropology, archeology, and forensic science. Dental cementum is a tissue of choice for the estimation of age at death in adult individuals as it continues deposition for the entirety of an individual's life. Previous works have devised regression formulas correlating cementum thickness to age at death. However, interpopulation variances are unknown, and it is therefore not clear whether regressions based on a single population are applicable to individuals with different ancestries. Materials and Methods: Here, we use a sample (n = 52) of teeth from individuals with known age at tooth extraction/death of European, African, and East Asian ancestry to assess whether there are interpopulations differences in cementum growth rate. We measured growth rate in four different areas (2nd and 5th decile of both the lingual and buccal aspect of the root) of each tooth and used nonparametric tests to evaluate population differences in growth rate between homologous regions of the teeth. Results: The results of the analyses show that, even after controlling for tooth size, individuals of European ancestry have significantly lower growth rates than those of both African and East Asian ancestry across all four tooth areas. Cementum plays a crucial role in linking periodontal and dental tissues and maintaining tooth root stability. Regenerating cementum is essential for functional periodontal tissue regeneration. It is a significant focus in tissue engineering. Periodontal disease, prevalent among the middle-aged and elderly, can severely impact overall health. Effective periodontal tissue regeneration is vital for enhancing the elderly's health and quality of life. Despite the progress, the lack of ideal scaffold materials and methods makes cementum regeneration a persistent challenge. This review discusses the current state of cementum tissue engineering materials and addresses existing challenges, providing a foundation for future scaffold material development.

We use synchrotron x-ray tomography of annual growth increments in the dental cementum of mammaliaforms (stem and crown fossil mammals) from three faunas across the Jurassic to map the origin of patterns of mammalian growth patterns, which are intrinsically related to mammalian endothermy. Although all fossils studied exhibited slower growth rates, longer life spans, and delayed sexual maturity relative to comparably sized extant mammals, the earliest crown mammals developed significantly faster growth rates in early life that reduced at sexual maturity, compared to stem mammaliaforms. Estimation of basal metabolic rates (BMRs) suggests that some fossil crown mammals had BMRs approaching the lowest rates of extant mammals. We suggest that mammalian growth patterns first evolved during their mid-Jurassic adaptive radiation, although growth remained slower than in extant mammals. [ABSTRACT FROM AUTHOR]

Carbon nanotubes (CNT) are 1 of the allotropes of carbon with unique properties. CNT shows good bone-tissue compatibility and has been reported to induce osteogenesis; therefore, it is regarded as an ideal material in a wide range of applications. However, the therapeutic effect of CNT-containing materials in the healing of apical periodontal tissue is unknown. The purpose of this study was to clarify the effect of CNT on the proliferation and mineralization of the human cementoblast cell line (HCEM). The proliferation of HCEM cells with CNT stimulation was assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay performed from 24–72 hours. Calcium deposition levels were evaluated by alizarin red S staining on days 7 and 10, and mineralization-related gene expression was examined by quantitative real-time polymerase chain reaction on days 3, 7, and 10. Scanning electron microscopy was used to observe the culture with CNT on day 14. CNT showed no cytotoxicity to HCEM cell proliferation. Treatment was performed with mineralization medium, CNT-induced HCEM mineralization on day 7, and increased calcium deposition on days 7 and 14. Messenger RNA expression of alkaline phosphatase was significantly increased throughout the experimental period, and bone sialoprotein was significantly increased on day 3 by CNT, whereas no effect was found on mRNA expression of type I collagen. CNT was observed in attachment to the cell surface on day 14. CNT promotes the mineralization of HCEM cells, indicating the potential as a new bioactive component for apical periodontal tissue regeneration materials through the regulation of cementoblast mineralization. Background The robustness and credibility of RT-qPCR results are critically dependent on the selection of suitable reference genes. However, the mineralization of the extracellular matrix can alter the intracellular tension and energy metabolism within cells, potentially impacting the expression of traditional reference genes, namely Actb and Gapdh. Objective To methodically identify appropriate reference genes for research focused on mouse cementoblast mineralization. Materials and methods Time-series transcriptomic data of mouse cementoblast mineralization were used. To ensure expression stability and medium to high expression levels, three specific criteria were applied to select potential reference genes. The expression stability of these genes was ranked based on the DI index (1/coefficient of variation) to identify the top six potential reference genes. RT-qPCR validation was performed on these top six candidates, comparing their performance against six previously used reference genes (Rpl22 , Ppib , Gusb , Rplp0 , Actb , and Gapdh). Cq values of these 12 genes were analyzed by RefFinder to get a stability ranking. Results A total of 4418 (12.27%) genes met the selection criteria. Among them, Rab5if , Chmp4b , Birc5 , Pea15a , Nudc , Supt4a were identified as candidate reference genes. RefFinder analyses revealed that two candidates (Birc5 and Nudc) exhibited superior performance compared to previously used reference genes. Limitations RefFinder's stability ranking does not consider the influence of primer efficiency. Objectives: This study aimed to evaluate and compare the impact of cigarette smoking (CS) and heated tobacco (HT) on the alteration of color and ultrastructural characteristics of human enamel and cementum. Background: According to tobacco companies, a less harmful substitute for CS is HT products. Nevertheless, comprehensive research on the effects of HT on tooth structures has been lacking. This study aimed to evaluate and compare the impact of CS and HT on the alteration of color and ultrastructural characteristics of human enamel and cementum. Materials and Methods: Thirty intact and noncarious human maxillary premolars extracted for orthodontic treatment purposes, previously disinfected, were used in the study. The specimens were randomly separated into six groups (n = 10), as follows: Group 1: enamel without smoking exposure; Group 2: enamel exposed to CS; Group 3: enamel exposed to HT; Group 4: cementum without smoking exposure; Group 5: cementum exposed to CS; and Group 6: cementum exposed to HT. The measurement of color change was conducted using a spectrophotometer. The surface alterations and mineral composition of enamel and cementum were evaluated using scanning electron microscopy and energy‐dispersive X‐ray spectroscopy. ANOVA test followed by Tukey's post hoc test was used to determine significant differences between groups. Results: Results showed that CS had a more pronounced effect on enamel and cementum color changes than HT. The impact of CS and HT on color changes was more evident in cementum than in enamel. Surface morphology of enamel and cementum showed alterations in histology following exposure to both smoking types. Moreover, the mineral content experienced a significant reduction after using CS and HT. The reduction in calcium content after CS and HT exposure was similar. However, HT led to a significant decrease in the phosphorus content of enamel when compared with CS. At the same time, CS exposure in cementum resulted in a more significant reduction in Ca/P ratio than HT. Conclusions: Although HT may appear to present a lower danger to hard dental tissues than CS, it is not entirely harmless. CS results in more color changes on the enamel and cementum of teeth. Both smoking methods affected the mineral content of teeth, with CS having a significant effect on the roots, while HT significantly affected the crowns' mineral composition. Clinical Relevance: The study addresses a knowledge gap on how smoking and HT products affect hard tooth tissues. Few studies compared the histological and colorimetric effects of these two smoking strategies before this study. Our research shows that cigarette smoke and HT products affect dental enamel and cementum differently, revealing their potential tooth harm. The use of HT products may cause tooth cavities and periodontal disease. This research can help clinicians advise patients on the oral health effects of these smoking alternatives. Background Materials and Methods Results Conclusion The aim of this study was to calculate the stress distribution of fiberglass post associated with resin composite crown restoration and fiberglass posts with zirconia restorations in mature and immature endodontically treated central maxillary incisor under various loading conditions.The study created six different study models in a virtual environment: healthy mature maxillary central teeth, intact immature maxillary central teeth, mature maxillary central teeth with fiberglass post associated with resin composite crown restoration, immature maxillary central teeth with fiberglass post associated with resin composite crown restoration, mature maxillary central teeth with fiberglass posts and zirconia restoration, and immature maxillary central teeth with fiberglass posts and zirconia restoration. Loading conditions simulating mastication, trauma, and bruxism were applied to each of the models at different angles and amounts. The von Mises and the maximum and minimum principal stress values in tooth structures (dentin) and support structures (bone, PDL) and materials were observed using finite element stress analysis.The highest stress values in the tissue and the restoration structure were observed for masticating force and crowns rehabilitated with zirconia restorations. None of the compared loading conditions and restorations showed destructive stress values on periodontal ligament or bone.The mature and immature endodontically treated central maxillary incisors can be better rehabilitated using fiberglass post associated with resin composite crown restoration and may be preferred to zirconia restorations in order to reduce the stresses on the surrounding tissues and teeth. Introduction: P. gingivalis are the main bacteria that cause periodontal diseases. One of the clinical signs of periodontal disease is alveolar bone resorption. Tofu liquid waste is a typical high water-containing biomass and isoflavones. Isoflavones is a polyphenol that has an anti-inflammatory effect by reducing pro-inflammatory cytokine expression. This study aims to evaluate whether systemic administration of different tofu liquid waste doses has effects on decreasing alveolar bone resorption of the Wistar rats' periodontal tissues induced by P. gingivalis. Materials and methods:: This is true experimental research, with a randomized posttest-only group design. The groups are K (-) for the normal group, K (+) for the negative control group, and K-1, K-2, and K-3 for group induced with P.gingivalis [0.03 mL(1x 109CFU/mL)] and administrated with 6, 12, and 18 mL/kgBW tofu liquid waste respectively every day for 28 days. Results: Freeze-dried technique result showed tofu liquid waste still contains isoflavone (daidzin and glycitin). There is no difference in mean in all parameters between K(-) and K-3. K(+) has difference mean with K-1, K-2, and K-3. Correlation test showed there was a positive correlation between the dose levels of tofu liquid waste with osteoblasts and OPG, and a negative correlation between the dose levels of tofu liquid waste with lymphocytes, neutrophils, osteoclasts, RANKL, alveolar bone resorption, and periodontal ligament space. Conclusion: Tofu liquid waste administration has effects on decreasing the alveolar bone resorption of Wistar rat (Rattus norvegicus) periodontal tissues induced by P. gingivalis and 18 mL/kgBW has the highest effect. Introduction. Periodontitis is a serious gum infection that disrupts the soft tissue around teeth. This study aimed to identify the most effective fraction of the Chinese medicine Kangfuxin for periodontitis treatment in a rat model. Material and methods. Kangfuxin solution was subjected to sequential extraction using chloroform, ethyl acetate, n-butanol, and water. The extracts were evaporated, dissolved in DMSO, diluted in water, and administered to rats via gavage (0.5 mL/day) for 2 weeks. The n-butanol extract was further fractionated using macroporous resin chromatography with 10%, 30%, 50%, 70%, and 90% ethanol elution. Levels of inflammatory cytokines IL-6, IL-1β, and TNF-α in periodontitis samples were examined by ELISA. Leukocyte infiltration in the cementum was analysed by haematoxylin and eosin (H&E) staining. Results. The n-butanol extract showed the best anti-inflammatory effect, reducing IL-6, IL-1β, and TNF-α levels in periodontitis samples and alleviating tissue damage and leukocyte infiltration in the cementum. Further fractionation revealed that the 50% ethanol fraction of the n-butanol extract had the most potent action in attenuating inflammation. This fraction suppressed the activation of the PI3K-AKT-mTOR signalling pathway in periodontitis samples. Application of a PI3K activator counteracted the anti-inflammatory effect of the 50% ethanol fraction. Conclusions. We identified a potent anti-inflammatory fraction (50% ethanol fraction of the n-butanol extract) of Kangfuxin for periodontitis treatment. This fraction suppressed the activity of the PI3K-AKT-mTOR signalling pathway in periodontitis samples. Establishing the identity of a person is important in the field of forensic odontology. This field can be helpful in identifying charred corpses in cases involving intense heat. Bones and body tissues can be extremely damaged due to heat, in such cases dental records have become a reliable source of identification. Dental Identification of charred human remains begins with the examination of dental features such as teeth anatomy, morphology, dental implants, oral health, restorations, fillings, dental anomalies and overall dental structure. These features remain resistant to extreme heat and can thus be compared with that of the ante-mortem records. When the postmortem and ante-mortem records match a successful identification is done. Dental features may also be damaged and become less effective for identification when temperatures rise above a particular range. In order to help with the identification of charred human remains, this research intends to give forensic experts a clear understanding of the circumstances in which dental records can be used by examining the effects of extreme temperature within the 400--800°C range. By gaining insights into this particular range and its effects on dental tissues and records gives forensic experts a chance to solve complex cases involving burned corpses. [ABSTRACT FROM AUTHOR]

In this paper, we describe a new ophthalmosaurid specimen found in the lower Barremian beds of the Paja Formation at Villa de Leiva, Boyacá, Colombia. The specimen represents a new species of Platypterygius, P. elsuntuoso sp. nov. defined by three unique features within the genus: a small ventral peg in the anterior ventral border of the extracondylar area of the basioccipital; a supratemporal groove in the quadrate; and a shallow neck at the base of the acellular cementum ring on the teeth root. In support of our taxonomic determination, in this contribution we also assess the species composition of the genus Platypterygius, comparing the known anatomy of the type species P. platydactylus with that of the other species previously assigned to the genus. We found that a great morphological affinity in the forelimb of the species P. americanus, P. australis, P. hercynicus, and P. sachicarum (lower Barremian to lower Cenomanian) with that of the type species (Aptian) is unquestionable and differentiate them from all other ophthalmosaurids. Three new diagnostic features (synapomorphies of the genus) support their inclusion in Platypterygius. Consequently, the genus name "Kyhytysuka", recently proposed to differentiate P. sachicarum from the genus Platypterygius is here rejected. A careful comparison of the cranial characteristics of Platypterygius specimens preserving associated cranial and fin remains demonstrate that Platypterygius is a genus distinguishable from other ophthalmosaurids not only by a typical forelimb but also by a particular combination of skull features. In this context, the skull morphology of "Simbirskiasaurus" concurs with that defining Platypterygius and its minor differences are not sufficient to support its generic distinction. The morphological comparisons and our phylogenetic analysis show the new species P. elsuntuoso more closely related to the Colombian species P. sachicarum than to other species of Platypterygius. Given that the new species came from the lower Barremian and P. sachicarum came from the upper Barremian, the minor differences in dentition found between the two species suggest a speciation by adaptation to new food sources in the Cretaceous epicontinental sea of Colombia. We aimed to evaluate the materials based on 4‐methacryloxyethyl trimellitate anhydride/methyl methacrylate tri‐n‐butylborane (Super‐bond [SB]) and nano hydroxyapatite (naHAp) for the repair of perforation at pulp chamber floor (PPF) in vitro and in vivo models. SB and naHAp were mixed in the mass ratio of 10% or 30% to produce naHAp/SB. Human periodontal ligament stem cells (HPDLSCs) were cultured on resin discs of SB or naHAp/SB to analyze the effects of naHAp/SB on cell adhesion, proliferation, and cementoblastic differentiation. A rat PPF model was treated with SB or naHAp/SB to examine the effects of naHAp/SB on the healing of defected cementum and periodontal ligament (PDL) at the site of PPF. HPDLSCs were spindle‐shaped and adhered to all resin discs. Changing the resin from SB to naHAp/SB did not significantly alter cell proliferation. Both 10% and 30% naHAp/SB were more effective than SB in promoting cementoblastic differentiation of HPDLSCs. In the rat PPF model, 30% naHAp/SB was more effective than SB in promoting the formation Sharpey's fiber‐like structures with expression of the PDL‐related marker and cementum‐like structures with expression of cementum‐related markers. In conclusion, 30% naHAp/SB can be the new restorative material for PPF because it exhibited the abilities of adhering to dentin and healing of defected periodontal tissue. Significance Statement: Perforation at pulp chamber floor (PPF) induces inflammatory reactions in surrounding periodontal tissues and leads to tooth loss. An ideal restorative material should exhibit biocompatibility, provide an adequate bond strength to dentin, and induce the healing of defected periodontal tissues. We aimed to develop and evaluate new materials based on 4‐methacryloxyethyl trimellitate anhydride/methyl methacrylate tri‐n‐butylborane (super‐bond [SB]) and nano hydroxyapatite (naHAp) and mixed them (naHAp/SB). The naHAp/SB exhibited biocompatible properties with adhering ability to dentin and induced the healing of natural periodontal tissues. Liver‐X receptors (LXRs) are essential nuclear hormone receptors involved in cholesterol and lipid metabolism. They are also believed to regulate inflammation and physiological and pathological bone turnover. We have previously shown that infection with the periodontal pathogen Porphyromonas gingivalis (Pg) in mice increases the abundance of CD11b+c‐fms+Ly6Chi cells in bone marrow (BM), spleen (SPL), and peripheral blood. These cells also demonstrated enhanced osteoclastogenic activity and a distinctive gene profile following Pg infection. Here, we investigated the role of LXRs in regulating these osteoclast precursors (OCPs) and periodontal bone loss. We found that Pg infection downregulates the gene expression of LXRs, as well as ApoE, a transcription target of LXRs, in CD11b+c‐fms+Ly6Chi OCPs. Activation of LXRs by treatment with GW3965, a selective LXR agonist, significantly decreased Pg‐induced accumulation of CD11b+c‐fms+Ly6Chi population in BM and SPL. GW3965 treatment also significantly suppressed the osteoclastogenic potential of these OCPs induced by Pg infection. Furthermore, the activation of LXRs reduces the abundance of OCPs systemically in BM and locally in the periodontium, as well as mitigates gingival c‐fms expression and periodontal bone loss in a ligature‐induced periodontitis model. Objective: The use of immediate dentin sealing (IDS) following tooth preparation for indirect restorations is well‐supported by laboratory studies. The inclusion of IDS as a mandatory clinical step, it must be firmly supported by evidence from randomized clinical trials (RCT). The objective of this study is to assess whether IDS should be added as a mandatory procedure in all indirect restorations, taking into consideration clinically significant outcomes for the patient, such as restoration longevity and hypersensitivity. Materials and Methods: A bibliographic search was carried out in the PubMed. In addition, a manual search was conducted in the references of literature reviews. Results: Data from four RCT were included. Two studies assessed preparations for full crowns, and two assessed preparations for partial restorations. Data from full crown preparations were subjected to a meta‐analysis, revealing a reduction in hypersensitivity incidence in the IDS group 1 week after restoration cementation. With respect to hypersensitivity, IDS seems to offer advantages exclusively during the provisional phase and up to 1 week following the cementation of the final restoration. One study has shown no difference on longevity. Conclusion: The use of IDS should be considered as an elective clinical step during the rehabilitation with indirect restorations. Periodontal tissue destruction in periodontitis is a consequence of the host inflammatory response to periodontal pathogens, which could be aggravated in the presence of type 2 diabetes mellitus (T2DM). Accumulating evidence highlights the intricate involvement of macrophage-mediated inflammation in the pathogenesis of periodontitis under both normal and T2DM conditions. However, the underlying mechanism remains elusive. Alpha-2-glycoprotein 1 (AZGP1), a glycoprotein featuring an MHC-I domain, has been implicated in both inflammation and metabolic disorders. In this study, we found that AZGP1 was primarily colocalized with macrophages in periodontitis tissues. AZGP1 was increased in periodontitis compared with controls, which was further elevated when accompanied by T2DM. Adeno-associated virus-mediated overexpression of Azgp1 in the periodontium significantly enhanced periodontal inflammation and alveolar bone loss, accompanied by elevated M1 macrophages and pyroptosis in murine models of periodontitis and T2DM-associated periodontitis, while Azgp1 -/- mice exhibited opposite effects. In primary bone marrow–derived macrophages stimulated by lipopolysaccharide (LPS) or LPS and palmitic acid (PA), overexpression or knockout of Azgp1 markedly upregulated or suppressed, respectively, the expression of macrophage M1 markers and key components of the NLR Family Pyrin Domain Containing 3 (NLRP3)/caspase-1 signaling. Moreover, conditioned medium from Azgp1 -overexpressed macrophages under LPS or LPS+PA stimulation induced higher inflammatory activation and lower osteogenic differentiation in human periodontal ligament stem cells (hPDLSCs). Furthermore, elevated M1 polarization and pyroptosis in macrophages and associated detrimental effects on hPDLSCs induced by Azgp1 overexpression could be rescued by NLRP3 or caspase-1 inhibition. Collectively, our study elucidated that AZGP1 could aggravate periodontitis by promoting macrophage M1 polarization and pyroptosis through the NLRP3/casapse-1 pathway, which was accentuated in T2DM-associated periodontitis. This finding deepens the understanding of AZGP1 in the pathogenesis of periodontitis and suggests AZGP1 as a crucial link mediating the adverse effects of diabetes on periodontal inflammation. Objective: This study aimed to evaluate the regenerative capacities of octacalcium phosphate collagen composite (OCP/Col) in one‐wall intrabony defects in dogs. The background data discuss the present state of the field: No study has assessed the efficacy of OCP/Col for periodontal regeneration therapy despite the fact that OCP/Col has proved to be efficient for bone regeneration. Methods: In six beagle dogs, the mandibular left third premolars were extracted 12 weeks before the experimental surgery. Standardized bone defects (5 mm in height and 4 mm in width) were simulated on the distal surface of the second premolars and mesially on the fourth premolars. The defect was filled with either OCP/Col (experimental group) or left empty (control group). Histological and histomorphometric characteristics were compared 8 weeks after surgery. Results: No infectious or ankylotic complications were detected at any of the tested sites. The experimental group exhibited a significantly greater volume, height, and area of newly formed bone than the control group. The former also showed a greater height of the newly formed cementum than the latter, although the results were not statistically significant. The newly formed periodontal ligaments were inserted into newly formed bone and cementum in the experimental group. Objective: The purpose of this study is to investigate regenerative process by immunohistochemical analysis and evaluate periodontal tissue regeneration following a topical application of BDNF to inflamed 3‐wall intra‐bony defects. Background: Brain‐derived neurotrophic factor (BDNF) plays a role in the survival and differentiation of central and peripheral neurons. BDNF can regulate the functions of non‐neural cells, osteoblasts, periodontal ligament cells, endothelial cells, as well as neural cells. Our previous study showed that a topical application of BDNF enhances periodontal tissue regeneration in experimental periodontal defects of dog and that BDNF stimulates the expression of bone (cementum)‐related proteins and proliferation of human periodontal ligament cells. Methods: Six weeks after extraction of mandibular first and third premolars, 3‐wall intra‐bony defects were created in mandibular second and fourth premolars of beagle dogs. Impression material was placed in all of the artificial defects to induce inflammation. Two weeks after the first operation, BDNF (25 and 50 μg/mL) immersed into atelocollagen sponge was applied to the defects. As a control, only atelocollagen sponge immersed in saline was applied. Two and four weeks after the BDNF application, morphometric analysis was performed. Localizations of osteopontin (OPN) and proliferating cell nuclear antigen (PCNA)‐positive cells were evaluated by immunohistochemical analysis. Results: Two weeks after application of BDNF, periodontal tissue was partially regenerated. Immunohistochemical analyses revealed that cells on the denuded root surface were positive with OPN and PCNA. PCNA‐positive cells were also detected in the soft connective tissue of regenerating periodontal tissue. Four weeks after application of BDNF, the periodontal defects were regenerated with cementum, periodontal ligament, and alveolar bone. Along the root surface, abundant OPN‐positive cells were observed. Morphometric analyses revealed that percentage of new cementum length and percentage of new bone area of experimental groups were higher than control group and dose‐dependently increased. Conclusion: These findings suggest that BDNF could induce cementum regeneration in early regenerative phase by stimulating proliferation of periodontal ligament cells and differentiation into periodontal tissue cells, resulting in enhancement of periodontal tissue regeneration in inflamed 3‐wall intra‐bony defects. Periodontal Disease (PD) is one of the most common inflammatory oral diseases, affecting majority of the population who are 30 years and older. Improper treatment of periodontal disease leads to degradation of periodontal tissues, eventually leading to tooth loss. Conventional clinical therapy for PD aims at eliminating infectious sources and reducing inflammation to arrest disease progression, which cannot achieve the regeneration of lost periodontal tissues. Over the past two decades, various regenerative periodontal therapies, such as Guided Tissue Regeneration (GTR), enamel matrix derivative, bone grafts, growth factor delivery, and the combination of cells and growth factors with matrix-based scaffolds, have been developed to target the restoration of lost tooth-supporting tissues, including the periodontal ligament, alveolar bone, and cementum. This review discusses recent progress in periodontal regeneration using tissueengineering and regenerative medicine approaches. Specifically, the focus is on the advances in biomaterials and controlled drug delivery for periodontal regeneration in recent years. Special attention is given to the development of advanced bio-inspired scaffolding biomaterials and temporospatial control of multi-drug delivery for the regeneration of the cementum-periodontal ligament-alveolar bone complex. Challenges and future perspectives are presented to provide inspiration for the design and development of innovative biomaterials and delivery systems for new regenerative periodontal therapy. Introduction: The periodontium is a connective tissue which consists of periodontal ligament, alveolar bone, cementum and gingiva. Periodontal ligament (PDL) is a specialized connective tissue that connects the cementum – coating the surface of the tooth – to the alveolar bone. Mohawk homeobox (Mkx) is a transcription factor that is expressed in PDL, that is known to play a vital role in the development and homeostasis of PDL. A detailed functional analysis of Mkx in the periodontal ligament for alveolar bone and cementum metabolism has not yet been conducted. Materials and methods: Alveolar bone height, bone mineral density (BMD) and bone volume fractions (Bone volume/Total volume: BV/TV) were measured and analyzed using micro-computed tomography (Micro-CT) and 3DBon on 7-week-old male wild-type (WT) (Mkx+/+) (n = 10) and Mkx-knockout (Mkx−/−) (n = 6) rats. Hematoxylin and Eosin (H&E), tartrate-resistant acid phosphatase (TRAP), alkaline phosphatase (ALP) and Masson Trichrome staining were performed on 5, 6, and 7-week-old Mkx+/+ and Mkx−/− rats. Cementum surface area and the number of TRAP-positive osteoclasts/mm were quantified, measured, and compared for 5,6 and 7-week-old Mkx+/+ and Mkx−/− rats (n = 3 each). Results: The level of alveolar bone height was significantly higher in Mkx−/− rats than in Mkx+/+ rats. On the other hand, there was significantly less BMD in Mkx−/− alveolar bone. A significant increase in cellular cementum could be observed as early as 5 weeks in Mkx−/− rats when compared with Mkx+/+ rats of the same age. More TRAP-positive osteoclasts were observed in Mkx−/− rats. Conclusion: Our findings further reveal the essential roles of Mkx in the homeostasis of the periodontal tissue. Mkx was found to contribute to bone and cementum metabolism and may be essential to the prevention of diseases such as periodontitis, and could show potential in regenerative treatments.

Background: Mesenchymal stromal cells (MSCs) isolated from the periodontal ligament (hPDL-MSCs) have a high therapeutic potential, presumably due to their immunomodulatory properties. The interaction between hPDL-MSCs and immune cells is reciprocal and executed by diverse cytokine-triggered paracrine and direct cell-to-cell contact mechanisms. For the first time, this study aimed to directly compare the contribution of various mechanisms on this reciprocal interaction using different in vitro co-culture models at different inflammatory milieus. Methods: Three co-culture models were used: indirect with 0.4 μm-pored insert, and direct with or without insert. After five days of co-culturing mitogen-activated CD4+ T lymphocytes with untreated, interleukin (IL)-1β, or tumor necrosis factor (TNF)-α- treated hPDL-MSCs, the CD4+ T lymphocyte proliferation, viability, and cytokine secretion were investigated. The gene expression of soluble and membrane-bound immunomediators was investigated in the co-cultured hPDL-MSCs. Results: Untreated hPDL-MSCs decreased the CD4+ T lymphocyte proliferation and viability more effectively in the direct co-culture models. The direct co-culture model without inserts showed a strikingly higher CD4+ T lymphocyte cell death rate. Adding IL-1β to the co-culture models resulted in substantial CD4+ T lymphocyte response alterations, whereas adding TNF resulted in only moderate effects. The most changes in CD4+ T lymphocyte parameters upon the addition of IL-1β or TNF-α in a direct co-culture model without insert were qualitatively different from those observed in two other models. Additionally, the co-culture models caused variability in the immunomediator gene expression in untreated and cytokine-triggered hPDL-MSCs. Conclusion: These results suggest that both paracrine and cell-to-cell contact mechanisms contribute to the reciprocal interaction between hPDL-MSCs and CD4+ T lymphocytes. The inflammatory environment affects each of these mechanisms, which depends on the type of cytokines used for the activation of MSCs' immunomodulatory activities. Objectives: The analysis of dental wear provides a useful approach for dietary and cultural habit reconstructions of past human populations. The analysis of macrowear patterns can also be used to better understand the individual chewing behavior and to investigate the biomechanical responses during different biting scenarios. The aim of this study is to evaluate the diet and chewing performance of the adult Neanderthal Bourgeois‐Delaunay 1 (BD 1) and to investigate the relationship between wear and cementum deposition under mechanical demands. Materials and methods: The macrowear pattern of BD 1 was analyzed using the occlusal fingerprint analysis method. We propose a new method for the bilateral measurement of the cementum volume along both buccal and lingual sides of the molar root. Results: BD 1's anterior dentition is more affected by wear compared to the posterior one. The macrowear pattern suggest a normal chewing behavior and a mixed‐diet coming from temperate environments. The teeth on the left side of the mandible display greater levels of wear, as well as the buccal side of the molar crowns. The cementum analysis shows higher buccal volume along the molar roots. Discussion: BD1 could have been preferably chewing on the left side of the mandible. The exploitation of various food resources suggested by the macrowear analysis is compatible with the environmental reconstructions. Finally, the greater wear on the buccal side of the molar occlusal surface and the greater volume of cementum in that side of the molar roots offers a preliminary understanding about the potential correlation between dental wear and cementum deposition. Objectives: Exosomes derived from stem cells are a potential cell‐free tool for tissue regeneration with therapeutic potential. However, its application in cementum repair is unclear. This study aimed to investigate the effect of human periodontal ligament stem cell‐derived exosomes on the biological activity of cementoblasts, the main effector cells in cementum synthesis. Materials and Methods: OCCM‐30 cementoblasts were cultured with various human periodontal ligament stem cell‐derived exosome concentrations. OCCM‐30 cells proliferation, migration, and cementogenic mineralization were examined, along with the gene and protein expression of factors associated with cementoblastic mineralization. Results: Exosomal promoted the migration, proliferation, and mineralization of OCCM‐30 cells. The exosome‐treated group significantly increased the expression of cementogenic‐related genes and proteins. Furthermore, the expression of p‐PI3K and p‐AKT was enhanced by exosome administration. Treatment with a PI3K/AKT inhibitor markedly attenuated the gene and protein expression of cementoblastic factors, and this effect was partially reversed by exosome administration. Conclusions: Human periodontal ligament stem cell‐derived exosomes can promote the activity of cementoblasts via the PI3K/AKT signaling pathway, providing a scientific basis for promoting the repair process in orthodontically induced inflammatory root resorption. Periodontitis is known to be affected by high-glucose conditions, which poses a challenge to periodontal tissue regeneration, particularly in bone formation. In this study, the potential effects of resveratrol (3,5,4′-trihydroxystilbene, RSV) in facilitating bone formation under high-glucose conditions after periodontitis has been investigated. We focused on the analysis of osteoblasts and periodontal ligament cells, which are essential for bone formation including cell proliferation and differentiation. And we aimed to investigate the impact of RSV on bone healing, employed diabetic mouse model induced by streptozotocin and confirmed through histological observation. High-glucose conditions adversely affected cell proliferation and ALP activity in both MC3T3-E1 and hPDLF in vitro, with more significant impact on MC3T3-E1 cells. RSV under high-glucose conditions had positive effects on both, showing early-stage effects for MC3T3-E1 cells and later-stage effects for hPDLF cells. RSV seemed to have a more pronounced rescuing role in MC3T3-E1 cells. Increased ALP activity was observed and the expression levels of significant genes, such as Col 1, TGF-β1, ALP, and OC, in osteogenic differentiation were exhibited stage-specific expression patterns. Upregulated Col 1 and TGF-β1 were detected in the early stage, and then ALP and OC expressions became more pronounced in the later stages. Similarly, stronger positive reactions against RUNX2 were detected in the RSV-treated group compared to the control. Furthermore, in in vivo experiment, RSV stimulates the growth and differentiation of osteoblasts, thereby promoting bone formation. Highglucose levels have the potential to impair cellular functions and the regenerative capacity to facilitate bone formation with MC3T3-E1 rather than hPDLF cells. Resveratrol appears to facilitate the inherent abilities of MC3T3-E1 cells compared with hPDLF cells, indicating its potential capacity to restore functionality during periodontal regeneration. Background: Osteogenic differentiation of human periodontal ligament stem cells (hPDLSCs) is an essential event in alveolar bone regeneration. Oxidative stress may be the main inhibiting factor of hPDLSC osteogenesis. Superoxide dismutase 2 (SOD2) is a key antioxidant enzyme, but its effect on hPDLSC osteogenic differentiation is unclear. Methods: Several surface markers were detected by flow cytometry, and the differentiation potential of hPDLSCs was validated by alkaline phosphatase (ALP), Alizarin Red S, and Oil Red O staining. Osteogenic indicators of hPDLSCs were detected by real‐time quantitative polymerase chain reaction (RT‐qPCR), Western blotting, and ALP staining. Furthermore, alveolar bone defect rat models were analyzed through micro‐CT, hematoxylin and eosin, and Masson staining. The intracellular reactive oxygen species (ROS) level was evaluated by a ROS assay kit. Finally, the expression of SOD2, Smad3, and p‐Smad3 in hPDLSCs was detected by RT‐qPCR and Western blotting (WB). Results: SOD2 positively regulated the gene and protein expressions of ALP, BMP6, and RUNX2 in hPDLSCs (p < 0.05). Ideal bone formation and continuous cortical bone were obtained by transplanting LV‐SOD2 hPDLSCs (lentivirus vector for overexpressing SOD2 in hPDLSCs) in vivo. Exogenous H2O2 downregulated osteogenic indicators (ALP, BMP6, RUNX2) in hPDLSCs (p < 0.05); this was reversed by overexpression of SOD2. WB results showed that the Smad3 and p‐Smad3 signaling pathways participated in the osteogenic process of SOD2 in hPDLSCs. Conclusion: SOD2 positively regulated hPDLSC osteogenic differentiation in vitro and in vivo. Mechanistically, SOD2 promotes hPDLSC osteogenic differentiation by regulating the phosphorylation of Smad3 to scavenge ROS. The regeneration of periodontal bone defects continues to be an essential therapeutic concern in dental biomaterials. Numerous biomaterials have been utilized in this sector so far. However, the immune response and vascularity in defect regions may be disregarded when evaluating the effectiveness of biomaterials for bone repair. Among several regenerative treatments, the most recent technique of in situ tissue engineering stands out for its ability to replicate endogenous restorative processes by combining scaffold with particular growth factors. Regenerative medicine solutions that combine biomaterials/scaffolds, cells, and bioactive substances have attracted significant interest, particularly for bone repair and regeneration. Dental stem cells (DSCs) share the same progenitor and immunomodulatory properties as other types of MSCs, and because they are easily isolable, they are regarded as desirable therapeutic agents in regenerative dentistry. Recent research has demonstrated that DSCs sown on newly designed synthetic bio-material scaffolds preserve their proliferative capacity while exhibiting increased differentiation and immuno-suppressive capabilities. As researchers discovered how short peptide sequences modify the adhesion and proliferative capacities of scaffolds by activating or inhibiting conventional osteogenic pathways, the scaffolds became more effective at priming MSCs. Introduction: nanoparticles are tiny-sized materials whose characteristics and properties mean that their association with dental materials is being investigated to ascertain their effects and possible benefits on tooth structures. This systematic review aimed to qualitatively collect in vitro studies that address the potential application of different nanoparticles in dental regeneration. Following an exhaustive search and article selection process, 16 in vitro studies that met our eligibility criteria were included. BG-NPs were analyzed across five studies, with three demonstrating their impact on the growth and differentiation of human hDPSCs. CS-NPs were examined in three studies, with findings from two indicating a significant effect on the differentiation of SCAPs. Nanoparticles' therapeutic potential and their stimulatory effect on promoting the regeneration of cells of the dentin-pulp complex have been proven. Their effect is altered according to the type of nanoparticle, concentration, and substances associated with them and, depending on these variables, they will affect the pulp, dentine, and dental cementum differently. Introduction: Intense histological research has been conducted on human bone and teeth for a long period of time. Histological evaluation of these highly mineralised tissues demands a thorough decalcification process using different chemical agents, which might alter some aspects of tissue architecture and staining properties. Aim: To assess the impact of four decalcifying agents on rat teeth and bone, focusing on the rate of decalcification, staining effectiveness, and tissue structure preservation. Materials and Methods: An in-vitro animal study was conducted in the Department of Oral Diagnosis at the College of Dentistry, University of Baghdad, Bab Almoaadim, Baghdad, Iraq from October 2022 to March 2023. Four decalcification agents, namely 10% Neutral Ethylenediaminetetraacetic Acid (EDTA), 10% formic acid, 3% nitric acid, and 5% nitric acid, were used to decalcify 24 molar teeth with their surrounding alveolar bone obtained from healthy male rats, which were randomly divided into four groups, each group consisting of six teeth. The decalcified sections underwent regular processing, and staining was performed using Haematoxylin and Eosin (H&E). Grading was conducted after two separate observers examined the stained sections under a light microscope. Data were presented using Mean±Standard Deviation (SD), number, and percentage. A one-way Analysis of Variance (ANOVA) (F-test) was used to compare the decalcification times within groups. Several group comparisons, including categorical data, were examined using the Chi-square test. Results: Nitric acids (3% and 5%) exhibited the fastest decalcification with a mean value of (2±0.89) days, while Formic acid required (3.5±0.54) days, and EDTA was the slowest, with (19±1.09) days. These differences in decalcification time were found to be statistically significant (p<0.00001). Formic acid 10% demonstrated superior tissue preservation and staining quality, with excellent staining results, minimal dentin-pulp separation, and preservation of pulp zones, cementum, and osteoblasts. In contrast, nitric acid 5% resulted in severe dentinpulp separation, absence of pulp zones, absence of osteoblast, and significant osteocyte retraction. Statistical significance was observed across all agents for dentin-pulp separation, pulp organisation, cementum destruction, Periodontal Ligament (PDL) separation, and osteocyte retraction (p-values of 0.004, 0.02, 0.02, 0.04, and 0.03, respectively). Conclusion: Consequently, 10% formic acid emerged as the most efficient decalcifying solution, ensuring rapid decalcification with favourable staining intensity and tissue architecture. Root fractures are more prevalent in teeth that have fully erupted and have closed apices, where the entire root is firmly anchored in the bone and surrounding tissues. The results can become complex because of the pulp, dentine, cementum, bone, and periodontium injury. The handling of horizontal root fractures encompasses numerous factors, resulting in the suggestion of different treatment methods. This report presents the steps and post-treatment period of two lower central incisors with root fractures at the apical third, which were treated using root canal therapy and mineral trioxide aggregate, respectively. Objective: The aim of this study was to investigate the thickness of acellular extrinsic fibre cementum (AEFC) at four root positions of anterior and posterior teeth with special focus on functional aspects. Furthermore, the correlations between cementum thickness and chronological age and sex are investigated. Background: While numerous studies confirm continuous cementum apposition with age, masticatory forces as well as physiological and orthodontically induced tooth movements also have the potential to affect tooth cementum thickness. Materials and Methods: Undecalcified teeth were embedded in resin and transverse‐sectioned in the cervical third of the root. Two sections per root were selected, and digital images at four positions were obtained (mesial, distal, oral, and vestibular) using light microscopy. The AEFC thickness of 99 teeth (anterior = 66, posterior = 33, male = 54, female = 45) were measured in both sections. The differences in mean values between root positions and the association of root position variation with tooth type, age, sex, and subject as well as the overall effects of age and sex were analysed using a mixed model. Results: First incisors and canines showed the greatest mean AFEC thickness, in contrast to premolars which had the lowest values. Differences were found across the four root positions, with a pattern varying considerably between anterior and posterior teeth and between maxilla and mandible in the anterior teeth. An interaction between root position and subject pointed to the existence of an individual component in the variation of AEFC thickness across the four root positions. There was an age trend with an almost linear increase in cementum thickness of 1 μm per year. Overall, females tended to exhibit a significantly lesser AEFC thickness compared to males. Conclusions: Distinct differences in the pattern of thickness values across the four root positions in anterior and posterior teeth support the assumption that the AEFC is strongly affected by functional processes. Background: This animal study sought to evaluate two novel nanomaterials for pulpotomy of primary teeth and assess the short-term pulpal response and hard tissue formation in dogs. The results were compared with mineral trioxide aggregate (MTA). Methods: This in vivo animal study on dogs evaluated 48 primary premolar teeth of 4 mongrel female dogs the age of 6–8 weeks, randomly divided into four groups (n = 12). The teeth underwent complete pulpotomy under general anesthesia. The pulp tissue was capped with MCM-48, MCM-48/Hydroxyapatite (HA), MTA (positive control), and gutta-percha (negative control), and the teeth were restored with intermediate restorative material (IRM) paste and amalgam. After 4–6 weeks, the teeth were extracted and histologically analyzed to assess the pulpal response to the pulpotomy agent. Results: The data were analyzed using the Kruskal‒Wallis, Fisher's exact, Spearman's, and Mann‒Whitney tests. The four groups were not significantly different regarding the severity of inflammation (P = 0.53), extent of inflammation (P = 0.72), necrosis (P = 0.361), severity of edema (P = 0.52), extent of edema (P = 0.06), or connective tissue formation (P = 0.064). A significant correlation was noted between the severity and extent of inflammation (r = 0.954, P < 0.001). The four groups were significantly different regarding the frequency of bone formation (P = 0.012), extent of connective tissue formation (P = 0.047), severity of congestion (P = 0.02), and extent of congestion (P = 0.01). No bone formation was noted in the gutta-percha group. The type of newly formed bone was not significantly different among the three experimental groups (P = 0.320). Conclusion: MCM-48 and MCM-48/HA are bioactive nanomaterials that may serve as alternatives for pulpotomy of primary teeth due to their ability to induce hard tissue formation. The MCM-48 and MCM-48/HA mesoporous silica nanomaterials have the potential to induce osteogenesis and tertiary (reparative) dentin formation. [ABSTRACT FROM AUTHOR]

Background: Chronic periodontitis is a prevalent dental condition characterized by the progressive loss of periodontal tissues, including alveolar bone. Conventional treatments have limitations in achieving complete periodontal regeneration, especially in cases with horizontal bone defects. Objective: This study aimed to evaluate the efficacy of diode laser therapy for periodontal regeneration in chronic periodontitis patients with horizontal bone defects. Methods: A prospective study was conducted at a Tertiary Care Center, involving 50 participants diagnosed with chronic periodontitis and horizontal bone defects. Diode laser therapy was administered, and clinical parameters (periodontal probing depth and clinical attachment level) were assessed pre- and post-treatment. Radiographic evaluation of bone fill and adverse events associated with laser therapy were also recorded. Results: Significant reductions in periodontal probing depth and gains in clinical attachment level were observed post-diode laser therapy (p < 0.001). Radiographic assessment revealed substantial bone fill in the horizontal defects (mean fill: 47.5% to 50.0%). Adverse events associated with laser therapy were minimal, with 80% of participants reporting no adverse events. Conclusion: Diode laser therapy demonstrates efficacy in promoting periodontal regeneration and osseous fill in chronic periodontitis patients with horizontal bone defects. These findings support the integration of diode laser therapy as an adjunctive treatment modality in periodontal therapy protocols. [ABSTRACT FROM AUTHOR]

ABSTRACT Objectives This study aimed to evaluate and compare the impact of cigarette smoking (CS) and heated tobacco (HT) on the alteration of color and ultrastructural characteristics of human enamel and cementum. Background According to tobacco companies, a less harmful substitute for CS is HT products. Nevertheless, comprehensive research on the effects of HT on tooth structures has been lacking. This study aimed to evaluate and compare the impact of CS and HT on the alteration of color and ultrastructural characteristics of human enamel and cementum. Materials and Methods Thirty intact and noncarious human maxillary premolars extracted for orthodontic treatment purposes, previously disinfected, were used in the study. The specimens were randomly separated into six groups (n = 10), as follows: Group 1: enamel without smoking exposure; Group 2: enamel exposed to CS; Group 3: enamel exposed to HT; Group 4: cementum without smoking exposure; Group 5: cementum exposed to CS; and Group 6: cementum exposed to HT. The measurement of color change was conducted using a spectrophotometer. The surface alterations and mineral composition of enamel and cementum were evaluated using scanning electron microscopy and energy‐dispersive X‐ray spectroscopy. ANOVA test followed by Tukey's post hoc test was used to determine significant differences between groups. Results Results showed that CS had a more pronounced effect on enamel and cementum color changes than HT. The impact of CS and HT on color changes was more evident in cementum than in enamel. Surface morphology of enamel and cementum showed alterations in histology following exposure to both smoking types. Moreover, the mineral content experienced a significant reduction after using CS and HT. The reduction in calcium content after CS and HT exposure was similar. However, HT led to a significant decrease in the phosphorus content of enamel when compared with CS. At the same time, CS exposure in cementum resulted in a more significant reduction in Ca/P ratio than HT. Conclusions Although HT may appear to present a lower danger to hard dental tissues than CS, it is not entirely harmless. CS results in more color changes on the enamel and cementum of teeth. Both smoking methods affected the mineral content of teeth, with CS having a significant effect on the roots, while HT significantly affected the crowns' mineral composition.

This case report describes the diagnosis and multidisciplinary treatment of a clinically indiscoverable cementodentinal tear associated with a periodontalendodontic combined lesion. The tear site was located at the palatal root surface of the maxillary left canine. Due to its position and concomitant periapical periodontitis, it was not noticed at the initial visit until a 3D CBCT examination was conducted. Through combined endodontic-periodontal therapy (which included root canal therapy, root debridement, and periodontal flap surgery), the tear fragment was removed, and the periapical lesion healed gradually. A histologic examination confirmed the definitive diagnosis of a cementodentinal tear. After 14 months, the periodontal and endodontic status of the maxillary left canine were stable. According to these results, CBCT examination and multidisciplinary cooperation seem to be effective and necessary for the diagnosis and treatment of such clinically indiscoverable cementodentinal/cemental tears. Objective: To evaluate the effect of different CAD/CAM materials and occlusal thicknesses on the fracture resistance of primary molar crowns. Methods: Sixty extracted primary molar teeth were prepared and randomly divided into six experimental groups according to the material and thickness. Primary molar crowns with a central groove thickness of 0.3 and 0.5 mm were fabricated from CAD/CAM zirconia (group Z), zirconia-reinforced lithium silicate (group ZLS), and pre-polymerized composite resin blocks (group C). Each crown was cemented with self-adhesive resin cement on the prepared tooth. All specimens were subjected to fracture tests until fracture. Fracture load values were recorded in Newtons (N). Data were statistically analyzed using a two-way analysis of variance (ANOVA) followed by Tukey multiple comparison test. Results: The highest fracture load values were obtained in group Z at 0.5 mm occlusal thickness and were significantly higher compared with the other experimental groups (p < 0.05). Although the lowest fracture load values were obtained in group ZLS at 0.3 mm occlusal thickness, all the tested CAD/CAM primary molar crowns at both thicknesses demonstrated fracture load values exceeding reported chewing force in pediatric patients. Conclusion: CAD/CAM primary molar crowns with reduced occlusal thickness may be used for the full-coverage restoration of primary molar teeth. [ABSTRACT FROM AUTHOR]

Periodontitis-induced alveolar bone defects necessitate the use of bone grafts for optimal therapeutic outcomes. Therefore, synthetic bone grafts exhibiting consistent properties, sufficient amounts, and multiple application forms have been extensively studied. Our previously designed Ca–Si–Mg cement has demonstrated benefits regarding cell proliferation, osteogenesis, and angiogenesis. Similarly, Sr exhibits osteogenic, anti-osteoclastogenic, and angiogenic properties by influencing several intracellular signaling pathways. In this study, we modified the previously established Ca–Si–Mg cement by incorporating Sr, which resulted in the formation of Mg–Sr-calcium silicate (CS) cement. Furthermore, we evaluated its biocompatibility and efficacy in promoting osteogenesis and angiogenesis. The results revealed that the setting time of the cement was extended with the individual incorporation of either Mg or Sr; notably, the setting time of M5S5 (integrating both Sr and Mg in equal proportions) was 2.7 min shorter than that of M5S0 (integrating only Mg). In addition, the presence of Sr enhanced the mechanical strength to an ultimate strength of 1.89 MPa and reduced the biodegradation rate of the Mg-doped CS cement from a weight loss of 13.9 %–11.5 %. Sr and Mg also demonstrated a synergistic effect on cell proliferation, with no observed cytotoxicity in human periodontal ligament cells (hPDLs). Osteogenic markers such as alkaline phosphate, bone sialoprotein, and osteocalcin were significantly expressed in hPDLs under the combined influence of Mg and Sr on osteogenic activity. Similarly, the angiogenesis-related proteins, vascular endothelial growth factor and angiopoietin-1 exhibited considerable enhancement, indicating the angiogenic capability of M5S5. In summary, our in vitro experiments confirmed that M5S5 is a potential candidate for osteogenic substitutes and alveolar bone regeneration, suggesting its potential applicability in clinical practice. The cementum is a highly mineralized tissue that covers the tooth root. The regional differences among the types of cementum, especially in the extrinsic fibers that contribute to tooth support, remain controversial. Therefore, this study used second harmonic generation imaging in conjunction with automated collagen extraction and image analysis algorithms to facilitate the quantitative examination of the fiber characteristics and the changes occurring in these fibers over time. Acellular extrinsic fiber cementum (AEFC) was invariably observed in the superficial layer of the apical cementum in mouse molars, indicating that this region of the cementum plays a crucial role in supporting the tooth. The apical AEFC exhibited continuity and fiber characteristics comparable with the cervical AEFC, suggesting a common cellular origin for their formation. The cellular intrinsic fiber cementum present in the inner layer of the apical cementum showed consistent growth in the apical direction without layering. This study highlights the dynamic nature of the cementum in mouse molars and underscores the requirement for re-examining its structure and roles. The findings of the present study elucidate the morphophysiological features of cementum and have broader implications for the maintenance of periodontal tissue health. [ABSTRACT FROM AUTHOR]

Background/Objectives: It has been repeatedly demonstrated that cementum formation is a crucial step in periodontal regeneration. Hyaluronic acid (HA) is an important component of the extracellular matrix which regulates cells functions and cell–cell communication. Hyaluronic acid/derivatives have been used in regenerative periodontal therapy, but the cellular effects of HA are still unknown. To investigate the effects of HA on cementoblast functions, cell viability, migration, mineralization, differentiation, and mineralized tissue‐associated genes and cementoblast‐specific markers of the cementoblasts were tested. Materials and Methods: Cementoblasts (OCCM‐30) were treated with various dilutions (0, 1:2, 1:4, 1:8, 1:16, 1:32, 1:64, 1:128) of HA and examined for cell viability, migration, mineralization, and gene expressions. The mRNA expressions of osteocalcin (OCN), runt‐related transcription factor 2 (Runx2), bone sialoprotein (BSP), collagen type I (COL‐I), alkaline phosphatase (ALP), cementum protein‐1 (CEMP‐1), cementum attachment protein (CAP), and small mothers against decapentaplegic (Smad) ‐1, 2, 3, 6, 7, β‐catenin (Ctnnb1) were performed with real‐time polymerase chain reaction (RT‐PCR). Total RNA was isolated on days 3 and 8, and cell viability was determined using MTT assay on days 1 and 3. The cell mineralization was evaluated by von Kossa staining on day 8. Cell migration was assessed 2, 4, 6, and 24 hours following exposure to HA dilutions using an in vitro wound healing assay (0, 1:2, 1:4, 1:8). Results: At dilution of 1:2 to 1:128, HA importantly increased cell viability (p <.01). HA at a dilution of 1/2 increased wound healing rates after 4 h compared to the other dilutions and the untreated control group. Increased numbers of mineralized nodules were determined at dilutions of 1:2, 1:4, and 1:8 compared with control group. mRNA expressions of mineralized tissue marker including COL‐I, BSP, RunX2, ALP, and OCN significantly improved by HA treatments compared with control group both on 3 days and on 8 days (p <.01). Smad 2, Smad 3, Smad 7, and β‐catenin (Ctnnb1) mRNAs were up‐regulated, while Smad1 and Smad 6 were not affected by HA administration. Additionally, HA at dilutions of 1:2, 1:4, and 1:8 remarkably enhanced CEMP‐1 and CAP expressions in a dilution‐ and time‐dependent manner (p <.01). Conclusions: The present results have demonstrated that HA affected the expression of both mineralized tissue markers and cementoblast‐specific genes.
