Abstract This study investigates the efficiency in methane production of lab-scale mesophilic (37 °C) and thermophilic (54 °C) continuous stirred tank reactors fed with cheese whey at different operational conditions. Results showed that whey mono-digestion was feasible at mesophilic conditions, while at thermophilic conditions frequent acidification incidents were recorded. The limited buffer capacity of the influent feedstock was responsible for the unstable anaerobic digestion process. The co-digestion of cheese whey with cattle manure maintained the pH levels higher than 7.0, and therefore, stable methane production rates were achieved without any significant accumulation of volatile fatty acids. An additional enhancement of the methane productivity was achieved by in-situ H 2 dispersion. Microbial community composition was investigated using high-throughput 16S rRNA gene amplicon sequencing and results were correlated with process parameters. Highlights • Social isolation is stressful for cattle yet practiced frequently in dairy farms. • 18 cows were isolated for 30 min with a mirror and/or an automated grooming brush. • Physiological and behavioural signs of stress were assessed throughout isolation. • Presence of an automated grooming brush/mirror did not reduce signs of stress. • We found no indication of immediate stress reduction through brush/mirror usage. Abstract In dairy farming, social isolation of cattle is commonly practiced for husbandry procedures such as artificial insemination, claw trimming and at times, for provision of medical treatment. When isolated, cows express physiological and behavioural signs of stress, such as elevated heart rate, hypothalamic-pituitary-adrenocortical activity and increased vocalisation rate. The aim of this study was to examine whether enriching the environment of the isolation pen using both tactile (i.e. an automated grooming brush) and visual (i.e. a mirror) stimulation could alleviate stress induced in socially isolated dairy cows. Eighteen cows (9 lactating and 9 dry cows) were subjected to four isolation conditions of 30 min each; isolation in the presence of a mirror, in the presence of an automated grooming brush, in the presence of both a mirror and an automated grooming brush, and in a non-enriched environment (without brush and mirror) that served as a control condition. Physiological (heart rate and heart rate variability) and behavioural indicators of stress (locomotion, vocalizations, attempts to escape the isolation pen and ear position of the cows) were measured during three phases throughout the isolation period (0–5 min, 10–15 min, 20–25 min). Our results show that, first, the heart rate of cows kept in social isolation, as well as the time cows spent in locomotion and exploration of the pen, decreased throughout the isolation period, regardless of treatment. Second, the presence of an automated grooming brush, a mirror or both an automated grooming brush and mirror in the isolation pen was not associated with reduced indicators of stress (physiological and behavioural measures) compared to the non-enriched environment. The results of our study are not in agreement with the findings of previous studies showing reduced levels of stress among socially isolated heifers/cows kept in the presence of visual enrichment (i.e. mirror/picture of a conspecific), and illustrate the need to further explore practices to reduce stress during social isolation. In Turkey, the use of robotic milking technology is still very new. The scientific data showing the effects on animal health are important as well as the studies related to the economic performance of milking robots. The aim of this study was to evaluate the hoof diseases encountered on dairy cattle farms working with robotic milking system and to determine the causes and to provide the necessary precautions to prevent and control hoof health problems. In the study, 93 Holstein dairy cattle aged between 35-54 months were evaluated in a dairy farm with two robotic milking system (system capacity of 120 cows/day). The locomotion scores were recorded and the cows' feet were examined in hoof trimming chute and claws of all cows were trimmed according to biomechanical characteristics of foot. The treatment method was determined according to the condition of the lesion in cases of hoof disease and the results of recovery were followed. In the herd, cases of claw deformation (6.4%; n=6), sole ulcer (4.3%; n=4), white line disease (3.2%; n=3), digital dermatitis (8.6%; n=8), heel lesion (6.4%; n=6), foot rot (1.0%; n=1), limax (2.1%; n=2), hoof crack (2.1%; n=2) and coroner inflammation (1.0%; n=1) were recorded. Evaluated etiologically, it has been found that mistakes in the application of bath solutions used in the single footbath at the exit of the robotic milking unit in the formation of infectious foot diseases, feeding mistakes made without regard to cow traffic in the case of non infectious foot diseases and exposure of the foot to traumas due to not paying attention to comfort in the feeding areas are effective. As a result, to prevent management malfunctions causing metabolic problems, stress and traumas in animal, the use of senseless technology should be avoided in order to ensure sustainable hoof health on robotic milking farms. We examined the hypothesis that rumination time (RT) could serve as a useful predictor of various common diseases of high producing dairy cows and hence improve herd management and animal wellbeing. We measured the changes in rumination time (RT) in the days before the recording of diseases (specifically: mastitis, reproductive system diseases, locomotor system issues, and gastroenteric diseases). We built predictive models to assess the association between RT and these diseases, using the former as the outcome variable, and to study the effects of the latter on the former. The average Pseudo-R2 of the fitted models was moderate to low, and this could be due to the fact that RT is influenced by other additional factors which have a greater effect than the predictors used here. Although remaining in a moderate-to-low range, the average Pseudo-R2 of the models regarding locomotion issues and gastroenteric diseases was higher than the others, suggesting the greater effect of these diseases on RT. Colonial waterbirds such as herons, egrets and spoonbills exhibit ecological characteristics that could have promoted the evolution of conspecific brood parasitism and extra-pair copulation. However, an adequate characterization of the genetic mating systems of this avian group has been hindered by the lack of samples of elusive candidate parents which precluded conducting conventional parentage allocation tests. Here, we investigate the genetic mating system of the invasive cattle egret using hematophagous insects contained in fake eggs to collect blood from incubating adults in a wild breeding colony. We tested a protocol with a previously unused Neotropical Triatominae, Panstrongylus megistus, obtained blood samples from males and females in 31 nests built on trees, drew blood from 89 nestlings at those nests, and genotyped all samples at 14 microsatellite loci, including six new species-specific loci. We comparatively addressed the performance of parentage allocation versus kinship classification of nestlings to infer the genetic mating system of cattle egrets. In line with previous behavioral observations, we found evidence in support of a non-monogamous genetic mating system, including extra-pair paternity (EPP) and conspecific brood parasitism (CBP). Parentage allocation tests detected a higher percentage of nests with alternative reproductive tactics (EPP: 61.7%; CBP: 64.5%) than the kinship classification method (EPP: 50.0%; CBP: 43.3%). Overall, these results indicate that rates of alternative reproductive tactics inferred in the absence of parental genetic information could be underestimated and should be interpreted with caution. This study highlights the importance of incorporating samples from candidate parents to adequately determine the genetic mating system of a species. We expand knowledge on the reproductive tactics of colonial waterbirds, contributing novel data on the genetic mating system of the cattle egret, valuable for the design of management strategies for this invasive bird. Background: Commercial cuts yield is an important trait for beef production, which affects the final value of the products, but its direct determination is a challenging procedure to be implemented in practice. The measurement of ribeye area (REA) and backfat thickness (BFT) can be used as indirect measures of meat yield. REA and BFT are important traits studied in beef cattle due to their strong implication in technological (carcass yield) and nutritional characteristics of meat products, like the degree of muscularity and total body fat. Thus, the aim of this work was to study the Longissimus dorsi muscle transcriptome of Nellore cattle, associated with REA and BFT, to find differentially expressed (DE) genes, metabolic pathways, and biological processes that may regulate these traits. Results: By comparing the gene expression level between groups with extreme genomic estimated breeding values (GEBV), 101 DE genes for REA and 18 for BFT (false discovery rate, FDR 10%) were identified. Functional enrichment analysis for REA identified two KEGG pathways, MAPK (Mitogen-Activated Protein Kinase) signaling pathway and endocytosis pathway, and three biological processes, response to endoplasmic reticulum stress, cellular protein modification process, and macromolecule modification. The MAPK pathway is responsible for fundamental cellular processes, such as growth, differentiation, and hypertrophy. For BFT, 18 biological processes were found to be altered and grouped into 8 clusters of semantically similar terms. The DE genes identified in the biological processes for BFT were ACHE, SRD5A1, RSAD2 and RSPO3. RSAD2 has been previously shown to be associated with lipid droplet content and lipid biosynthesis. Conclusion: In this study, we identified genes, metabolic pathways, and biological processes, involved in differentiation, proliferation, protein turnover, hypertrophy, as well as adipogenesis and lipid biosynthesis related to REA and BFT. The objective of the present study was to estimate genetic parameters and evaluate the genetic trends of 100-day milk yield (MY), fat percentage (FP) and protein percentage (PP) in three government dairy cattle farms in Thailand, located in the north and north-east of the country. The data consisted of records from 1745 first-lactation cows that calved between 1986 and 2015, which were the progeny of 287 sires and 1237 dams. Most of the animals were crossbred Holstein-Friesian with native or zebu animals. The total number of animals in the pedigree was 4753. Univariate and bivariate linear mixed models were used to estimate genetic parameters and breeding values by using ASReml-R. The fixed effects in the model were herd, year of calving, season of calving, breed group and the interaction between year and season of calving, and the random effects were animal and residual. All fixed effects had significant effects on the three traits, except breed group. The estimated heritabilities for MY, FP and PP were 0.242 ± 0.061, 0.066 ± 0.048 and 0.329 ± 0.072 respectively. The genetic correlation for MY and FP, MY and PP, and FP and PP were 0.219 ± 0.352, -0.271 ± 0.177 and 0.452 ± 0.255 respectively. The genetic trends based on estimated breeding values over year of birth did not show consistent improvement in all three traits, although there was a slight declining trend in MY in the later years. In this paper, an open algorithm was developed for the detection of cattle’s grass intake and rumination activities. This was done using the widely available inertial measurement unit (IMU) from a smartphone, which contains an accelerometer, a gyroscope, a magnetometer and location sensors signals sampled at 100 Hz. This equipment was mounted on 19 grazing cows of different breeds and daily video sequences were recorded on pasture of different forage allowances. After visually analyzing the cows’ movements on a calibration database, signal combinations were selected and thresholds were determined based on 1-s time windows, since increasing the time window did not increase the accuracy of detection. The final algorithm uses the average value and standard deviation of two signals in a two-step discrimination tree: the gravitational acceleration on x-axis (Gx) expressing the cows’ head movements and the rotation rate on the same x-axis (Rx) expressing jaw movements. Threshold values encompassing 95% of the normalized calibrated data gave the best results. Validation on an independent database resulted in an average detection accuracy of 92% with a better detection for rumination (95%) than for grass intake (91%). The detection algorithm also allows for characterization of the diurnal feeding activities of cattle at pasture. Any user can make further improvements, for data collected at the same way as the iPhone’s IMU has done, since the algorithm codes are open and provided as supplementary data . Poor soil structural properties and nutrient status, and low enzyme activities are common in the solonetz. This is caused by excessive exchangeable Na+ and high soil pH value in soils. Long-term application of cattle manure is an important management practice that can affect soil quality in the solonetzic soils. Experiments were carried out in a randomized complete block design comprising 5 treatments according to the cattle manure application history: Corns (Zea mays L.) with manure applied for 2, 6, 13, and 18 years were used as the experimental treatments and corn without manure application was used as a control treatment. Soil physic-chemical properties and enzyme activities were measured across all treatments. The effects of long-term manure application on soil quality were assessed using factor analysis. Results indicated that long-term manure application significantly improved soil physic-chemical properties and enhanced soil enzyme activities. Two factors were selected for the measured soil attributes, which were “soil structural properties” (Factor 1) and “saline-alkaline properties” (Factor 2), respectively. Compared to untreated soils, soils treated with manure for 13 and 18 years were characterized by improved soil structural properties (Factor 1) and decreased soil saline-alkaline properties (Factor 2), while soils treated with manure for 2 and 6 years were characterized by decreased soil saline-alkaline properties (Factor 2). Soil quality was increased with the number of years of manure application judging from the soil quality indices, with the highest value observed for 18 years of manure application. We conclude that soil structural and saline-alkaline properties are the key factors that limit soil quality, and promotion of soil quality is characterized by the decrease in bulk density, pH value and electrical conductivity in the solonetz, especially for bulk density. Background: The availability of dense genotypes and whole-genome sequence variants from various sources offers the opportunity to compile large datasets consisting of tens of thousands of individuals with genotypes at millions of polymorphic sites that may enhance the power of genomic analyses. The imputation of missing genotypes ensures that all individuals have genotypes for a shared set of variants. Results: We evaluated the accuracy of imputation from dense genotypes to whole-genome sequence variants in 249 Fleckvieh and 450 Holstein cattle using Minimac and FImpute. The sequence variants of a subset of the animals were reduced to the variants that were included on the Illumina BovineHD genotyping array and subsequently inferred in silico using either within- or multi-breed reference populations. The accuracy of imputation varied considerably across chromosomes and dropped at regions where the bovine genome contains segmental duplications. Depending on the imputation strategy, the correlation between imputed and true genotypes ranged from 0.898 to 0.952. The accuracy of imputation was higher with Minimac than FImpute particularly for variants with a low minor allele frequency. Using a multi-breed reference population increased the accuracy of imputation, particularly when FImpute was used to infer genotypes. When the sequence variants were imputed using Minimac, the true genotypes were more correlated to predicted allele dosages than best-guess genotypes. The computing costs to impute 23,256,743 sequence variants in 6958 animals were ten-fold higher with Minimac than FImpute. Association studies with imputed sequence variants revealed seven quantitative trait loci (QTL) for milk fat percentage. Two causal mutations in the DGAT1 and GHR genes were the most significantly associated variants at two QTL on chromosomes 14 and 20 when Minimac was used to infer genotypes. Conclusions: The population-based imputation of millions of sequence variants in large cohorts is computationally feasible and provides accurate genotypes. However, the accuracy of imputation is low in regions where the genome contains large segmental duplications or the coverage with array-derived single nucleotide polymorphisms is poor. Using a reference population that includes individuals from many breeds increases the accuracy of imputation particularly at low-frequency variants. Considering allele dosages rather than best-guess genotypes as explanatory variables is advantageous to detect causal mutations in association studies with imputed sequence variants. OBJECTIVE To describe the incidence of specific causes of lameness and the associations of cause and severity of lameness on the outcome for cattle on commercial feedlots. DESIGN Dynamic population longitudinal study. ANIMALS Cattle on 6 commercial feedlots in Kansas and Nebraska during a 12-month period (mean daily population, 243,602 cattle; range, 223,544 to 252,825 cattle). PROCEDURES Feedlot personnel were trained to use a standardized diagnostic algorithm and locomotion score (LMS) system to identify and classify cattle by cause and severity of lameness. Information regarding lameness cause, severity, and treatments was recorded for individual cattle. Cattle were monitored until they left the feedlot (ie, outcome; shipped with pen mates [shipped], culled prematurely because of lameness [realized], or euthanized or died [died]). Incidence rates for various causes of lameness, LMSs, and outcomes were calculated. The respective associations of cause of lameness and LMS with outcome were evaluated. RESULTS Lameness was identified in 2,532 cattle, resulting in an overall lameness incidence rate of 1.04 cases/100 animal-years. Realized and mortality rates were 0.096 cattle/100 animal-years and 0.397 deaths/100 animal-years, respectively. Injury to the proximal portion of a limb was the most frequently identified cause of lameness followed by undefined lameness, septic joint or deep digital sepsis, and interdigital phlegmon (foot rot). As the LMS (lameness severity) at lameness detection increased, the percentage of cattle that died but not the percentage of cattle that were realized increased. Recently, dysfunction of antigen-specific T cells is well documented as T-cell exhaustion and has been defined by the loss of effector functions during chronic infections and cancer in human. The exhausted T cells are characterized phenotypically by the surface expression of immunoinhibitory receptors, such as programmed death 1 (PD-1), lymphocyte activation gene 3 (LAG-3), T-cell immunoglobulin and mucin domain-containing protein 3 (Tim-3) and cytotoxic T-lymphocyte antigen 4 (CTLA-4). However, there is still a fundamental lack of knowledge about the immunoinhibitory receptors in the fields of veterinary medicine. In particular, very little is known about mechanism of T cell dysfunction in chronic infection in cattle. Recent our studies have revealed that immunoinhibitory molecules including PD-1/programmed death-ligand 1 (PDL1) play critical roles in immune exhaustion and disease progression in case of bovine leukemia virus (BLV) infection, Johne's disease and bovine anaplasmosis. To minimise soil compaction, land managers should limit cow grazing periods when soil water content (SWC) is greater than a critical water content (CWC) at which point soils are prone to treading damage. A laboratory-based method, the ‘proctor test’, has been used to identify the CWC in two intact Pallic soils collected from North and South Otago, New Zealand. We have attempted to relate the CWC to a simple field-based metric—the soil plastic limit—which can be measured by farmers as a proxy for CWC. Plastic limit was a suitable proxy for CWC in the North Otago soil only. In the South Otago soil, the CWC occurred at a lower SWC than the plastic limit. Although CWC varied across the two soil types, in both instances, it was reached at an SWC less than field capacity, while this was not always the case for plastic limit. [ABSTRACT FROM AUTHOR]

The aim of this study is to provide insight the network of cattle movement in Kampong Cham, Kampong Speu and Takeo, Cambodia. A cross-sectional study was carried out from July 2014 to August 2014, using questionnaires. It was implemented with 435 interviewees (24.4%, 24.6% and 51.0% from Kampong Cham, Kampong Speu and Takeo, respectively) using one-step snowball sampling. The findings suggest that the key players in all three provinces are producers who raise their cattle as backyard animals. In all three provinces the key players in spreading disease are probably the middlemen, collectors, brokers or traders. The network of cattle movement is presented as a strong component of varying size in each location. In this network we found three cut-points in both Kampong Cham and Kampong Speu. The network in each province indicates a random pattern of node distribution. The results of our study are useful to relevant authorities and researchers to understand the spread of infectious diseases into different areas. The middlemen, collectors, brokers and traders need to be controlled as first priority in order to reduce the magnitude of the spread of disease. [ABSTRACT FROM AUTHOR]

Feed efficiency, simply expressed as less feed inputs versus animal production outputs, can be measured in several ways, such as feed conversion ratio (FCR) and residual feed intake (RFI). FCR is a common measurement in beef cattle operations, and is the ratio of feed intake to live-weight gain. RFI is defined as the difference between actual and predicted feed intake after taking into account variability in maintenance and growth requirements. Rumen microbiota, which inludes bacteria, archaea, protozoa, and fungi, play an essential role in the digestion of lignocellulosic plant biomass, and can provide more than 70% of the host ruminants energy requirements via the production of volatile fatty acids (VFAs). Methane, a potent greenhouse gas (GHG), is produced in large quantities by the rumen microbiota, and is a known contributor to the global increase in GHG emissions. Studies have shown a negative relationship between methane emission and feed efficiency. Therefore, there is a need to study the feed efficiency from a rumen microbiome perspective and explore the probability of improving feed efficiency and hence reduce methane production in cattle by manipulating the rumen microbiome. The development of highthroughput sequencing technologies incuding metagenomics and metatranscriptomic analyses in the past decade has led to a sharp increase in understanding the rumen microbiota and associated function. The aim of this study was to evaluate the genome integrity so as to assess the adaptability of three breeds of indigenous cattle reared under arid and semi-arid regions of Rajasthan (Bikaner) and Haryana (Karnal) India. The cattle were of homogenous group (same age and sex) of indigenous breeds viz. Sahiwal, Tharparkar and Kankrej. A total of 100 animals were selected for this study from both climatic conditions. The sister chromatid exchanges (SCE's), chromosomal gaps and chromatid breaks were observed in metaphase plates of chromosome preparations obtained from in vitro culture of peripheral blood lymphocytes. The mean number of breaks and gaps in Sahiwal and Tharparkar of semi-arid zone were 8.56 ± 3.16, 6.4 ± 3.39 and 8.72 ± 2.04, 3.52 ± 6.29, respectively. Similarly, the mean number of breaks and gaps in Tharparkar and Kankrej cattle of arid zone were 5.26 ± 1.76, 2.74 ± 1.76 and 5.24 ± 1.84, 2.5 ± 1.26, respectively. The frequency of SCEs in chromosomes was found significantly higher ( P < 0.05) in Tharparkar of semi-arid region (4.72 ± 1.55) compared to arid region (2.83 ± 1.01). Similarly, the frequency of SCEs was found to be 4.0 ± 1.41 in the Sahiwal of semi-arid region and 2.69 ± 1.12 in Kankrej of arid zone. Statistical analysis revealed significant differences ( P < 0.05) amongst the different zones, i.e. arid and semi-arid, whereas no significant difference ( P > 0.05) was observed in the same zone. The analysis of frequency of CAs and SCEs revealed significant effects of environmental conditions on the genome integrity of animals, thereby indicating an association with their adaptability. This study was carried out to detect differences in locomotion and feeding behavior in lame (group L; n = 41; gait score ≥ 2.5) and non-lame (group C; n = 12; gait score ≤ 2) multiparous Holstein cows in a cross-sectional study design. A model for automatic lameness detection was created, using data from accelerometers attached to the hind limbs and noseband sensors attached to the head. Each cow’s gait was videotaped and scored on a 5-point scale before and after a period of 3 consecutive days of behavioral data recording. The mean value of 3 independent experienced observers was taken as a definite gait score and considered to be the gold standard. For statistical analysis, data from the noseband sensor and one of two accelerometers per cow (randomly selected) of 2 out of 3 randomly selected days was used. For comparison between group L and group C, the T-test, the Aspin-Welch Test and the Wilcoxon Test were used. The sensitivity and specificity for lameness detection was determined with logistic regression and ROC-analysis. Group L compared to group C had significantly lower eating and ruminating time, fewer eating chews, ruminating chews and ruminating boluses, longer lying time and lying bout duration, lower standing time, fewer standing and walking bouts, fewer, slower and shorter strides and a lower walking speed. The model considering the number of standing bouts and walking speed was the best predictor of cows being lame with a sensitivity of 90.2% and specificity of 91.7%. Sensitivity and specificity of the lameness detection model were considered to be very high, even without the use of halter data. It was concluded that under the conditions of the study farm, accelerometer data were suitable for accurately distinguishing between lame and non-lame dairy cows, even in cases of slight lameness with a gait score of 2.5. Play behaviour is an indicator of good welfare in young calves and is reduced by low energy intake and weaning off milk. There is renewed interest in keeping calves longer with the cow but separation leads to signs of distress, such as vocalizations. Providing calves with an alternative milk source prior to separation helps them adapt to the separation. We hypothesized that (1) locomotor play of nursed calves separated from their mothers will reflect their energy intake after separation, and (2) calves’ prior access to automated milk feeders will increase locomotor play and reduce vocalizations after separation due to an increased energy intake. 30 Holstein cows and their calves were kept in adjacent pens. “Suckling-only” calves were allowed to suckle the cow during the night and received no other milk, “Milk-feeder only” calves could obtain 12 L/day milk only from an automated milk feeder while “Suckling-and-milk-feeder” calves could suckle during the night and were allowed 12 L/day of milk from an automated milk feeder. At 6 weeks of age, calves were not allowed to enter the cow pen during the night but had access to automated milk and grain feeders. To measure locomotor play, we placed the calves individually in a 9.5 m × 2 m arena for 10 min, 10 times before and after separation. The frequency of jumping and vocalization were scored. Digestible energy (DE) intakes of the calves after separation were calculated from combined milk and grain intake. Before separation, there were no differences between the treatment groups on any behavioural measure ( P > 0.10). For Suckling-only calves, there was a marked decrease in the frequency of jumping (Wilcoxon test, P = 0.02) and a marked increase in the frequency of vocalization during the first 3 days after separation ( P = 0.004). During the first 3 days of separation, Suckling-only calves had a lower frequency of jumping (Mann–Whitney test, P = 0.009) and vocalized more frequently (Mann–Whitney test, P = 0.009) than did calves of the other treatment groups. For all calves, the number of vocalization was negatively correlated with frequency of jumping ( r s = −0.51; P = 0.005). After separation, Suckling-only calves had lower energy intakes than calves of the other treatment groups (Mann–Whitney test, P = 0.01) and the digestible energy intake of the calves was positively correlated with the frequency of jumping ( r s = 0.75; P < 0.001), and negatively correlated with the frequency of vocalization ( r s = −0.59; P < 0.001). There were no differences between treatment groups five or more days after separation. Our results show that a low energy intake of calves after separation from the mothers is associated with reduced locomotor play and increased vocalization and that prior access to an automated milk feeder helps maintain energy intake after separation, which results in increased locomotor play. The association between vocalization and locomotor play suggests that the reduction in play at separation may be related to the emotional response of the calves to the decrease in energy intake. Background: Bovine progressive degenerative myeloencephalopathy (Weaver syndrome) is a neurodegenerative disorder in Brown Swiss cattle that is characterized by progressive hind leg weakness and ataxia, while sensorium and spinal reflexes remain unaffected. Although the causal mutation has not been identified yet, an indirect genetic test based on six microsatellite markers and consequent exclusion of Weaver carriers from breeding have led to the complete absence of new cases for over two decades. Evaluation of disease status by imputation of 41 diagnostic single nucleotide polymorphisms (SNPs) and a common haplotype published in 2013 identified several suspected carriers in the current breeding population, which suggests a higher frequency of the Weaver allele than anticipated. In order to prevent the reemergence of the disease, this study aimed at mapping the gene that underlies Weaver syndrome and thus at providing the basis for direct genetic testing and monitoring of today's Braunvieh/Brown Swiss herds. Results: Combined linkage/linkage disequilibrium mapping on Bos taurus chromosome (BTA) 4 based on Illumina Bovine SNP50 genotypes of 43 Weaver-affected, 31 Weaver carrier and 86 Weaver-free animals resulted in a maximum likelihood ratio test statistic value at position 49,812,384 bp. The confidence interval (0.853 Mb) determined by the 2-LOD drop-off method was contained within a 1.72-Mb segment of extended homozygosity. Exploitation of wholegenome sequence data from two official Weaver carriers and 1145 other bulls that were sequenced in Run4 of the 1000 bull genomes project showed that only a non-synonymous SNP (rs800397662) within the PNPLA8 gene at position 49,878,773 bp was concordant with the Weaver carrier status. Targeted SNP genotyping confirmed this SNP as a candidate causal mutation for Weaver syndrome. Genotyping for the candidate causal mutation in a random sample of 2334 current Braunvieh animals suggested a frequency of the Weaver allele of 0.26 %. Conclusions: Through combined use of exhaustive sequencing data and SNP genotyping results, we were able to provide evidence that supports the non-synonymous mutation at position 49,878,773 bp as the most likely causal mutation for Weaver syndrome. The increased summertime prevalence of cattle carriage of enterohemorrhagic Shiga toxin-producing Escherichia coli O157:H7 (STEC O157) is associated with the increased summertime incidence of human infection. The mechanism driving the seasonality of STEC O157 carriage among cattle is unknown. We conducted experimental challenge trials to distinguish whether factors extrinsic or intrinsic to cattle underlie the seasonality of STEC O157 colonization. Holstein steers (n=20) exposed to ambient environmental conditions were challenged with a standardized pool of STEC O157 strains four times at 6-month intervals. The densities and durations of rectoanal junction mucosa (RAJ) colonization with STEC O157 were compared by season (winter versus summer), dose (109 CFU versus 107 CFU), and route of challenge (oral versus rectal). Following summer challenges, the RAJ STEC O157 colonization density was significantly lower (P=0.016) and the duration was shorter (P=0.052) than for winter challenges, a seasonal pattern opposite to that observed naturally. Colonization was unaffected by the challenge route, indicating that passage through the gastrointestinal microbiome did not significantly affect the infectious dose to the RAJ. A 2-log reduction of the challenge doses in the second-year trials was accompanied by similarly reduced RAJ colonization in both seasons (P< 0.001). These results refute the hypothesis that cattle are predisposed to STEC O157 colonization during the summer months, either due to intrinsic factors or indirectly due to gastrointestinal tract microbiome effects. The Italian Piedmontese cattle breed is traditionally housed in tie-stalls and, to a lesser extent, in loose housing systems with free stalls. The present study has compared the same group of 15 cattle before and after stable renovation (tie-stall vs free-stall housing) funded by Regulation (EU) No. 1305/2013. All the animals remained healthy during the trial, no clinical signs were observed and no mastitis occurred. The tested parameters were: locomotion scoring system, salivary cortisol, blood parameters, serum acute phase protein (albumin, haptoglobin, serum amyloid A and lysozyme). Samples were collected 3 times: before the change (T1), 3 days later (T2), 40 days later (T3). The change in housing determined noteworthy variations in the stress parameters: albumin and total protein displayed the lowest value at T2, while lysozyme displayed the lowest value at T3. Among the App, SAA and Hp were not affected by the sampling time. Salivary cortisol displayed the highest value at T1. The Dutch Five Step trimming method (Toussaint Raven, 1985) has been an internationally recognised standard for cattle foot trimming for over 30 years. However, with the increasing size and productivity of UK dairy cattle, modifications have been proposed. Research evidence suggests that some modifications may be beneficial, particularly a deeper concavity of the sole of the lateral hind claw to reduce peak force and compression of the corium which can lead to injury during standing and locomotion. There remains considerable debate over optimal toe length and lack of clarity about from where the initial measurement is made. Furthermore, there is limited evidence on what is the optimal frequency of routine trimming. Current research supports the view that routine trimming using best practice standards is beneficial in terms of reducing lameness and promoting welfare, but this may not always be cost beneficial in herds with a low lameness incidence. Lameness is still an important problem in modern dairy farming. Human observation of locomotion, by looking at different traits in one go, is used in practice to assess locomotion. The objectives of this article were to determine which individual locomotion traits are most related to locomotion scores in dairy cows, and whether experienced raters are capable of scoring these individual traits consistently. Locomotion and 5 individual locomotion traits (arched back, asymmetric gait, head bobbing, reluctance to bear weight, and tracking up) were scored independently on a 5-level scale for 58 videos of different cows. Videos were shown to 10 experienced raters in 2 different scoring sessions. Relations between locomotion score and traits were estimated by 3 logistic regression models aiming to calculate the size of the fixed effects on the probability of scoring a cow in 1 of the 5 levels of the scale (model 1) and the probability of classifying a cow as lame (locomotion score ≥3; model 2) or as severely lame (locomotion score ≥4; model 3). Fixed effects were rater, session, traits, and interactions among fixed effects. Odds ratios were calculated to estimate the relative probability to classify a cow as lame when an altered (trait score ≥3) or severely altered trait (trait score ≥4) was present. Overall intrarater and interrater reliability and agreement were calculated as weighted kappa coefficient (κw) and percentage of agreement, respectively. Specific intrarater and interrater agreement for individual levels within a 5-level scale were calculated. All traits were significantly related to the locomotion score when scored with a 5-level scale and when classified as (severely) lame or nonlame. Odds ratios for altered and severely altered traits were 10.8 and 14.5 for reluctance to bear weight, 6.5 and 7.2 for asymmetric gait, and 4.8 and 3.2 for arched back, respectively. Raters showed substantial variation in reliability and agreement values when scoring traits. The acceptance threshold for overall intrarater reliability (κw ≥0.60) was exceeded by locomotion scoring and all traits. Overall interrater reliability values ranged from κw = 0.53 for tracking up to κw = 0.61 for reluctance to bear weight. Intrarater and interrater agreement were below the acceptance threshold (percentage of agreement <75%). Most traits tended to have lower specific intrarater and interrater agreement in level 3 and 5 of the scale. In conclusion, raters had difficulties in scoring locomotion traits consistently, especially slight alterations were difficult to detect by experienced raters. Yet, the locomotion traits reluctance to bear weight, asymmetric gait, and arched back had the strongest relation with the locomotion score. A participatory epidemiological (PE) study was conducted with livestock keepers in Moroto and Kotido districts, Karamoja Region, Uganda, between October and December 2013 to determine the management options and relative importance of tick-borne diseases (TBDs) amongst transhumant zebu cattle. Data collection involved 24 focus group discussions (each comprising 8–12 people) in 24 settlement areas (manyattas), key informant interviews (30), direct observation, a review of surveillance data, clinical examination, and laboratory confirmation of cases of TBDs. Methods used in group discussions included semi-structured interviews, simple ranking, pairwise ranking, matrix scoring, proportional piling and participatory mapping. The results of pairwise comparison showed the Ngakarimojong-named diseases, lokit (East Coast fever, ECF), lopid (anaplasmosis), loukoi (contagious bovine pleuropneumonia, CBPP), lokou (heartwater) and lokulam (babesiosis), were considered the most important cattle diseases in Moroto in that order, while ECF, anaplasmosis, trypanosomosis (ediit), CBPP and nonspecific diarrhoea (loleo) were most important in Kotido. Strong agreement between informant groups (Kendall’s coefficient of concordance W = 0.568 and 0.682; p < 0.001) in pairwise ranking indicated that the diseases were a common problem in selected districts. East Coast fever had the highest median score for incidence (18% [range: 2, 33]) in Moroto, followed by anaplasmosis (17.5% [8,32]) and CBPP (9% [1,21]). Most animals that suffered from ECF, anaplasmosis, heartwater and babesiosis died, as the respective median scores for case fatality rates (CFR) were 89.5% (42, 100), 82.8% (63, 100), 66.7% (20, 100) and 85.7% (0, 100). In Kotido, diseases with high incidence scores were ECF (21% [6,32]), anaplasmosis (17% [10,33]) and trypanosomosis (8% [2,18]). The CFRs for ECF and anaplasmosis were 81.7% (44, 100) and 70.7% (48, 100), respectively. Matrix scoring revealed that disease indicators showed strong agreement ( W = 0.382–0.659, p < 0.05– p < 0.001) between informant groups. Inadequate knowledge, poor veterinary services and limited availability of drugs were the main constraints that hindered the control of TBDs. Hand picking of ticks was done by all pastoralists while hand spraying with acaricides was irregular, often determined by availability of drug supplies and money. It was concluded that TBDs, particularly ECF and anaplasmosis were important diseases in this pastoral region. Results from this study may assist in the design of feasible control strategies. Objective To identify patterns of cattle movement that could influence disease spread in the Australian cattle population. Methods Records from the National Livestock Identification System database for the period January 2008 to December 2012 were accessed and analysed. Postcodes were used to allocate each individual property to one of 12 livestock production regions. National movement patterns and the characteristics of each livestock production region were quantified in terms of the number of consignments and animals moved, and seasonality of movements. Results The majority of cattle movements remained within a single livestock production region, while those that did not, usually remained within the same state or territory. Producers were the most common source of cattle, and abattoirs and other producers the most common destinations, with approximately 40% of animals moving via a saleyard. The northern regions generally moved larger consignments than the southern regions and were less connected to other regions. The eastern and south-eastern regions were very well connected by cattle movements. Seasonal patterns were seen for some regions, particularly the northern regions where weather patterns strongly influence the ability of producers to muster and transport stock. Conclusions The movement patterns observed provide quantitative support for previous information based on surveys and expert opinion, and capture more of the variability in Australian cattle production. This study studied the relation between longevity, the interval from last insemination to culling in days (IL2C) with 7 different cull classes; 1. Reproduction cull class, 2. Production cull class, 3. Locomotion cull class, 4. Accident cull class, 5. General disorder cull class, 6. Peri-partum cull class and 7. Unknown cull class. Overall, the most important cull classes were reproduction (19%) and production (50%). The IL2C for production (146.3±1.2) and reproduction (87.4±1.9) were significantly different from each other but also from locomotion (127.0±3.6), accident (120.1±7.3), general disorder (105.4±5.5), peri-partum (109.6±5.7) cull classes. Sows that were culled for production reasons had significantly the highest average parity number at culling (4.6±0.2) compared to all other culling reasons such as reproduction (3.1±0.2), locomotion (2.3±0.3), accident (2.9±0.5), general disorder (3.3±0.4) and peri-partum (3.3±0.4) cull classes. Furthermore, it was found that age in days, parity number at culling and IL2C showed to be heritable traits (0.13±0.05, 0.16±0.05 and 0.10±0.04 respectively). The genetic correlation between parity number at culling and IL2C was not significantly different from 0 (−0.04±0.28). Because IL2C is a heritable trait and sows that are culled within the production cull class obtain on average a higher parity and show the longest IL2C, this study showed the potential to select on longer IL2C and thereby select against sows to be culled within the reproduction, locomotion, accident, general disorder and peri-partum cull class and to be culled at the end of a parity. Due to its detrimental effect on cow welfare, health and production, lameness in dairy cows has received quite a lot of attention in the last few decades-not only in terms of prevention and treatment of lameness but also in terms of detection, as early treatment might decrease the number of severely lame cows in the herds as well as decrease the direct and indirect costs associated with lameness cases. Generally, lame cows are detected by the herdsman, hoof trimmer or veterinarian based on abnormal locomotion, abnormal behavior or the presence of hoof lesions during routine trimming. In the scientific literature, several guidelines are proposed to detect lame cows based on visual interpretation of the locomotion of individual cows (i.e., locomotion scoring systems). Researchers and the industry have focused on automating such observations to support the farmer in finding the lame cows in their herds, but until now, such automated systems have rarely been used in commercial herds. This review starts with the description of normal locomotion of cows in order to define 'abnormal' locomotion caused by lameness. Cow locomotion (gait and posture) and behavioral features that change when a cow becomes lame are described and linked to the existing visual scoring systems. In addition, the lack of information of normal cow gait and a clear description of 'abnormal' gait are discussed. Claw health, an important dairy cow welfare parameter, may be affected by early-life foot/leg stresses. To investigate this, groups of pregnant heifers were allocated to deep straw bedding (Soft) or cubicles (Hard), both with scraped concrete feeding alleys. After the grazing season, they were re-housed in cubicle systems, half on slatted concrete (Hard) and half on slatted rubber (Soft) alleys. Claw measurements, contact area and pressure distribution claw/flooring, claw disorders and leg lesions were recorded at the start and end of each housing season. Locomotion and leg lesions were also scored monthly after calving. Prevalence of sole haemorrhages was higher among pregnant heifers in cubicles than in deep straw. After calving, first-calvers on Hard floors had higher odds for lameness (OR = 3.6; p < 0.01), sole haemorrhages/ulcers (OR = 2.2; p < 0.05), white-line haemorrhages (OR = 2.8; p < 0.01) and leg lesions (OR = 2.6; p < 0.02) than those on Soft floors. Lowest prevalence and severity of sole and white-line haemorrhages (non-significant) in first-calvers was found in those on Soft floors and reared on Hard floors and the highest prevalence and severity on those on Hard floors reared on Soft floors. Assessment of lameness prevalence and severity requires visual evaluation of the locomotion of a cow. Welfare schemes including locomotion assessments are increasingly being adopted, and more farmers and their veterinarians might implement a locomotion-scoring routine together. However, high within-observer agreement is a prerequisite for obtaining valid mobility scorings, and within-observer agreement cannot be estimated in a barn, because the gait of cows is dynamic and may change between 2 occasions. The objective of this study was to estimate the within-observer agreement according to the observers' educational background and experience with cattle, based on video recordings with very diverse types of gait. Groups of farmers, bovine veterinarians, first- and fourth-year veterinary students, researchers, and cattle-inexperienced sensory assessors evaluated mobility using a 5-point mobility score system developed specifically for walking cows (n = 102 observers). The evaluation sessions were similar for all groups, lasted 75 min, and were organized as follows: introduction, test A, short training session, break, and test B. In total, video recordings of 22 cows were displayed twice in a random order (11 cows in each test x 2 replicates). Data were analyzed applying kappa coefficient, logistic regression, and testing for random effects of observers. The crude estimates of 95% confidence interval for weighted kappa in test A and B ranged, respectively, from 0.76 to 0.80 and 0.70 to 0.75. When adjusting for the fixed effects of video sample and gait scoring preferences, the probability of assigning the same mobility score twice to the same cow varied from 55% (sensory assessors) to 72% (fourth-year veterinary students). The random effect of the individual observers was negligible. That is, in general observers could categorize the mobility characteristics of cows quite well. Observers who preferred to assess the attributes back arch or the overall mobility score (based on uneven gait) had the highest agreement, respectively, 69 or 68%. The training session seemed insufficient to improve agreement. Nonetheless, even novice observers were able to achieve perfect agreement up to 60% of the 22 scorings with merely the experience obtained during the study (introduction and training session). The relatively small differences between groups, together with a high agreement, demonstrate that the new system is easy to follow compared with previously described scoring systems. The mobility score achieves sufficiently high within-observer repeatability to allow between-observer agreement estimates, which are reliable compared with other more-complex scoring systems. Consequently, the new scoring scale seems feasible for on-farm applications as a tool to monitor mobility within and between cows, for communication between farmers and veterinarians with diverse educational background, and for lameness benchmarking of herds. Lameness is one of the most significant endemic disease problems facing the dairy industry. Claw horn lesions (principally sole hemorrhage, sole ulcer, and white line disease) are some of the most prevalent conditions. Despite the fact that thousands of animals are treated for these conditions every year, experimental evidence is limited on the most effective treatment protocols. A randomized, positively controlled clinical trial was conducted to test the recovery of newly lame cows with claw horn lesions. Animals on 5 farms were locomotion scored every 2 wk. Cows were eligible for recruitment if they had 2 nonlame scores followed by a lame score and had a claw horn lesion on a single claw of a single foot. Following a therapeutic trim, enrolled cows were randomly allocated to 1 of 4 treatments: treatment 1--no further treatment (positive control; TRM), treatment 2--trim plus a block on the sound claw (TB), treatment 3--trim plus a 3-d course of the nonsteroidal anti-inflammatory drug (NSAID) ketoprofen (TN), treatment 4--trim plus a block plus ketoprofen (TBN). The primary outcome measure was locomotion score 35 d after treatment, by an observer blind to treatment group. Descriptive statistics suggested that treatment groups were balanced at the time of enrollment, that is, randomization was successful. Based on a sound locomotion score (score 0) 35 d after treatment, the number of cures was 11 of 45 (24.4%) for TRM, 14 of 39 (35.9%) for TB, 12 of 42 (28.6%) for TN, and 23 of 41 (56.1%) for TBN. The difference between TBN and TRM was significant. To test for confounding imbalances between treatment groups, logistic regression models were built with 2 outcomes, either sound (score 0) or nonlame (score 0 or 1) 35 d after treatment. Compared with TRM, animals that received TBN were significantly more likely to cure to a sound outcome. Farm, treatment season, lesion diagnosis, limb affected, treatment operator, and stage of lactation were included in the final models. Our work suggests that lameness cure is maximized with NSAID treatment in addition to the common practices of therapeutic trimming and elevation of the diseased claw using a block when cows are newly and predominantly mildly lame. This study characterizes the physiological and morphological changes related to partial luteolysis in bovine corpus luteum (CL) after challenges with sub-doses of cloprostenol sodium on Day 6 (D6) of the estrous cycle. Cows ( n = 12/treatment) were treated as follows: Control (2 mL, saline, i.m.); 2XPGF (two treatments i.m. 500 μg of cloprostenol sodium 2 h apart) and 1/6PGF (83.3 μg of cloprostenol sodium, i.m., once). Plasma progesterone (P4) concentration, CL volume and blood flow were measured immediately before the treatments, then every 8 h (h) for 48 h. In the Control, P4 concentrations were higher at 48 h than at 0 h. P4 decreased 8 h after 2XPGF treatment ( P < 0.05), and remained low until the end of the trial. P4 decreased in 1/6PGF between 8 and 16 h ( P < 0.05), then began to rebound at 24 h. Luteal volume was higher in Controls at 48 h than at 0 h. Under 1/6PGF, luteal volume decreased at 24 h ( P < 0.05) and began to rebound at 32 h. Luteal volume and blood flow were reduced starting at 24 and 32 h, respectively, after 2XPGF treatment ( P < 0.05). Background Foot-and-mouth disease (FMD) vaccines applied for prophylactic use in endemic areas provide short-lived immunity requiring regular boosters. Indian FMD control program recommends twice a year vaccination. Development of high potency vaccines that provide better immune response can singificantly contribute to control programme by reducing the frequency of vaccination. The present study explores new adjuvants to enhance the protective efficacy of inactivated trivalent FMD vaccines. Methodology and principal findings VacciMax ® is a novel adjuvant which uses a liposome-based oil emulsion platform. Cattle were immunized using VacciMax-A and VacciMax-B FMD vaccines and evaluated for protective efficacy. Similar groups of animals were also boosted after 6 months to study the effect of booster immunisation on protection against homologous challenge. Serum samples from immunized animals were tested by virus neutralization test (VNT) and liquid phase blocking ELISA (LPBE). After challenge, animals were screened for virus load by real-time PCR and reactivity in non-structural protein (3ABC) antibody detection ELISA to corroborate the protection data. A single dose of VacciMax-A formulation elicited higher percentage protection (63%) in VacciMax-A compared to 25% in VacciMax-B upon challenge at one-year post-vaccination. Upon boosting at 6 months also, VacciMax-A group showed higher levels of protection (100%) compared to VacciMax-B (86%), even though both the groups elicited comparable VNT titre ( p = 0.4964). The results also demonstrated that intramuscular route was preferrable over subcutaneous route of administration. Conclusion The study demonstrates that immunization with VacciMax-A-IM adjuvanted FMD vaccine with high antigen payload under boosting regimen could effectively be used as potent vaccine to maintain herd immunity. The comfort of dairy cattle while standing has important implications for housing design. Research has examined how cattle respond to standing surfaces by presenting options under all 4 legs or under a single leg, but no work, to date, has compared presentation methods. This study examined behavior and muscle activity when cows stood on rough floors under all 4 legs or just 1 hind leg. Three treatments were tested: smooth con crete under all 4 legs (0-ROUGH), a rough surface under all 4 legs [2 cm x 2 cm x 4 cm trapezoidal protrusions (4-ROUGH)], and a rough surface only under 1 hind leg, with other legs on smooth concrete (1-ROUGH). Twenty-four healthy Holstein cows stood on each surface for 1 h/d in a repeated-measures design. Surface electromyograms (SEMG) were used to evaluate muscle fatigue and total activity. Muscle fatigue was measured using SEMG to evaluate (1) static contractions when cows were continuously weight bearing on each hind leg, before and after 1 h of standing, and (2) dynamic contractions associated with steps during 1 h of standing. Behavioral measures included steps per minute, time between each consecutive step, and the latency to lie down after testing. The number of legs affected by roughness influenced both behavioral and physiological responses to flooring. Cows on 1-ROUGH stepped twice as often with the rough-treated leg and one-half as much with the hind leg on smooth concrete com pared with other surfaces. Similarly, on the 1-ROUGH surface, total muscle activity was reduced in the leg on the rough surface, and muscle activity was more sustained (3x higher) in the other hind leg, suggesting that cows avoid possible discomfort under 1 leg by us ing muscles in the other. In the 4-ROUGH treatment, time between steps was more variable than on the other 2 treatments (coefficient of variation, 4-ROUGH: 245; 1-ROUGH: 208; 0-ROUGH: 190 ± 5.8%), likely because cows could not move away from this uneven flooring. Thus, the method of presentation of stimuli used to evaluate comfort while standing altered the response. Stepping rate and SEMG changed when roughness was under a single leg, whereas timing between consecutive steps was more variable with rough flooring under all 4 legs. Background Atypical bovine spongiform encephalopathies (BSEs), classified as H-type and L-type BSE based on the Western immunoblot profiles, are naturally occurring diseases in cattle, which are phenotypically different to classical BSE. Transmission studies in cattle using the intracerebral route resulted in disease where the phenotypes were maintained irrespective of BSE type but clinically affected cattle with a shorter survival time displayed a nervous form whereas cattle with a longer survival time displayed a dull form. A second transmission study is reported here where four cattle were intracerebrally inoculated with brain tissue from experimentally infected cattle presenting with either the nervous or dull form of H- or L-type BSE to determine whether the phenotype is maintained. Results The four inoculated cattle were culled at 16.5-19.5 months post inoculation after presenting with difficulty getting up, a positive scratch response (all) and dullness (three cattle), which was not observed in two non-inoculated control cattle, each housed with either group of inoculated cattle. Only the inoculated cattle had detectable prion protein in the brain based on immunohistochemical examination, and the Western immunoblot profile was consistent with the H-type or L-type BSE of the respective donor cattle. Conclusions Second passage of H-type and L-type BSE in cattle produced a TSE where the majority of cattle displayed the dull form regardless of clinical disease form of the donor cattle. Background Perosomus elumbis (PE) is a congenital defect that has been observed sporadically in Holstein cattle for many years. However, several cases have been reported in recent years and this may indicate an unrecognised spread of a mutant allele in the Holstein population worldwide. Two cases in Danish Holstein calves are reported to provide details on the phenotype. Case presentation Two full-term Holstein calves were born after assisted delivery due to dystocia with breech presentation. External morphological examination indicated that the lumbar, sacral and coccygeal vertebrae were absent and the abdominal region was just present as a floppy sac covered by skin and enclosing the abdominal organs. The hind limbs were hypoplastic with bilateral symmetric arthrogryposis and muscular atrophy. Radiographs, computed tomography scan and necropsy confirmed these findings. The caudal part of the thoracic spinal cord showed myelodysplasia. A range of abdominal organ malformations were found at necropsy. Inbreeding was not found during genealogical examination, but remote shared ancestors were present in the pedigrees. Conclusion The addition of these further cases of PE to the in recent years reported record of cases should draw more attention to this defect in the Holstein breed. PE may be an emerging genetic defect in the Holstein population worldwide and cases should be sampled to enable genetic mapping of the gene possibly underlying the disease. Locomotion scores are used for lameness detection in dairy cows. In research, locomotion scores with 5 levels are used most often. Analysis of scores, however, is done after transformation of the original 5-level scale into a 4-, 3-, or 2-level scale to improve reliability and agreement. The objective of this study was to evaluate different ways of merging levels to optimize resolution, reliability, and agreement of locomotion scores for dairy cows. Locomotion scoring was done by using a 5-level scale and 10 experienced raters in 2 different scoring sessions from videos from 58 cows. Intra- and interrater reliability and agreement were calculated as weighted kappa coefficient (KW) and percentage of agreement (PA), respectively. Overall intra- and interrater reliability and agreement and specific intra- and interrater agreement were determined for the 5-level scale and after transformation into 4-, 3-, and 2-level scales by merging different combinations of adjacent levels. Intrarater reliability (KW) ranged from 0.63 to 0.86, whereas intrarater agreement (PA) ranged from 60.3 to 82.8% for the 5-level scale. Interrater KW = 0.28 to 0.84 and interrater PA = 22.6 to 81.8% for the 5-level scale. The specific intrarater agreement was 76.4% for locomotion level 1, 68.5% for level 2, 65% for level 3, 77.2% for level 4, and 80% for level 5. Specific interrater agreement was 64.7% for locomotion level 1, 57.5% for level 2, 50.8% for level 3, 60% for level 4, and 45.2% for level 5. Specific intra- and interrater agreement suggested that levels 2 and 3 were more difficult to score consistently compared with other levels in the 5-level scale. The acceptance threshold for overall intra- and interrater reliability (KW and K ≥0.6) and agreement (PA ≥75%) and specific intra- and interrater agreement (≥75% for all levels within locomotion score) was exceeded only for the 2-level scale when the 5 levels were merged as (12)(345) or (123)(45). In conclusion, when locomotion scoring is performed by experienced raters without further training together, the lowest specific intra- and interrater agreement was obtained in levels 2 and 3 of the 5-level scale. The objective of this review was to describe, compare and evaluate agreement, reliability, and validity of manual and automatic locomotion scoring systems (MLSSs and ALSSs, respectively) used in dairy cattle lameness research. There are many different types of MLSSs and ALSSs. Twenty-five MLSSs were found in 244 articles. MLSSs use different types of scale (ordinal or continuous) and different gait and posture traits need to be observed. The most used MLSS (used in 28% of the references) is based on asymmetric gait, reluctance to bear weight, and arched back, and is scored on a five-level scale. Fifteen ALSSs were found that could be categorized according to three approaches: (a) the kinetic approach measures forces involved in locomotion, (b) the kinematic approach measures time and distance of variables associated to limb movement and some specific posture variables, and (c) the indirect approach uses behavioural variables or production variables as indicators for impaired locomotion. Agreement and reliability estimates were scarcely reported in articles related to MLSSs. When reported, inappropriate statistical methods such as PABAK and Pearson and Spearman correlation coefficients were commonly used. Some of the most frequently used MLSSs were poorly evaluated for agreement and reliability. Agreement and reliability estimates for the original four-, five- or nine-level MLSS, expressed in percentage of agreement, kappa and weighted kappa, showed large ranges among and sometimes also within articles. After the transformation into a two-level scale, agreement and reliability estimates showed acceptable estimates (percentage of agreement ≥75%; kappa and weighted kappa ≥0.6), but still estimates showed a large variation between articles. Agreement and reliability estimates for ALSSs were not reported in any article. Several ALSSs use MLSSs as a reference for model calibration and validation. However, varying agreement and reliability estimates of MLSSs make a clear definition of a lameness case difficult, and thus affect the validity of ALSSs. MLSSs and ALSSs showed limited validity for hoof lesion detection and pain assessment. The utilization of MLSSs and ALSSs should aim to the prevention and efficient management of conditions that induce impaired locomotion. Long-term studies comparing MLSSs and ALSSs while applying various strategies to detect and control unfavourable conditions leading to impaired locomotion are required to determine the usefulness of MLSSs and ALSSs for securing optimal production and animal welfare in practice. Movement of animals from one farm to another is a potential risk and can lead to the spreading of livestock diseases. Therefore, in order to implement effective control measures, it is important to understand the movement network in a given area. Using the SANITEL data from 2005 to 2009, around 2 million cattle movements in Belgium were traced. Exploratory analysis revealed different spatial structures for the movement of different cattle types: fattening calves are mostly moved to the Antwerp region, adult cattle are moved to different parts in Belgium. Based on these differences, movement of cattle would more likely cause a spread of disease to a larger number of areas in Belgium as compared to the fattening calves. A closer inspection of the spatial and temporal patterns of cattle movement using a weighted negative binomial model, revealed a significant short-distance movement of bovine which could be an important factor contributing to the local spreading of a disease. The model however revealed hot spot areas of movement in Belgium; four areas in the Walloon region (Luxembourg, Hainaut, Namur and Liege) were found as hot spot areas while East and West Flanders are important "receivers" of movement. This implies that an introduction of a disease to these Walloon regions could result in a spread toward the East and West Flanders regions, as what happened in the case of Bluetongue BTV-8 outbreak in 2006. The temporal component in the model also revealed a linear trend and short- and long-term seasonality in the cattle movement with a peak around spring and autumn. The result of this explorative analysis enabled the identification of "hot spots" in time and space which is important in enhancing any existing monitoring and surveillance system. Ruminal impaction comprises 19.4% of cattle diseases in the Jiangsu province of China. The authors treated ruminal impaction in 55 cattle with electro-acupuncture (EA) of the classical acupoints Pi-shu and Du-jiao. Recovery occurred after 1 EA treatment in 40/55 (73%), 2 EA treatments in 9/50 (18%) and 3 EA treatments in 4/50 (8%) cattle. The final 2 animals recovered after 3 EA treatments and Chinese herbal medicine, Jian Pi Li Qi San. To determine the mechanisms of action of EA of Pi-shu and Du-jiao, 8 cattle with rumen impaction were randomly selected to compare rumen contractions, pH, fermentation and ciliate concentrations before and after EA. Compared to normal cattle, the frequency and duration of ruminal contractions greatly decreased, the pH increased and fermentation and number of ciliates decreased during ruminal impaction. After EA, contraction frequency and duration increased very significantly (p<0.001), the pH significantly decreased (p=0.02), the fermentation significantly increased (p=0.004) and the number of ciliates very significantly increased (p=0.0006). To investigate the effect of vagus nerve integrity on EA effectiveness, EA of Pi-shu and Du-jiao was performed on 8 clinically normal cattle and the frequency and duration of ruminal contractions increased. Following bilateral cervical vagectomy, ruminal contractions ceased and EA had no effect to restore ruminal movements. Based on the findings of this study and the successful treatment of 53 cattle with EA alone, it can be concluded that left unilateral EA of Pi-shu and Du-jiao provides an effective treatment for ruminal impaction. Forestomach atony from simple indigestion was treated in 54 cattle with 2-4 daily doses of a Spleen-invigorating and Qi regulating Chinese herbal medicine, Jian Pi Li Qi San. The ingredients of Jian Pi Li Qi San included: 50-100 g each of Bai Zhu (Atractylodes), Fu Ling (Poria) and Mu Xiang (Aucklandia), 50 g Bing Lang (Areca), 40 g Gan Cao (Glycyrrhiza) and 100-200 g each of Shan Zha (Crataegus), Shen Qu (Massa) and Mai Ya (Hordeum). The first 6 herbal ingredients were ground, decocted in boiling water and the last 2 herbs were ground and added after the solution had cooled, so as not to deactivate important enzymes. The solution was administered via a tube into the rumen. Rumination began 1-2 days after dosing in 50/54 (92.5%) cattle. From the 54 affected cattle, 5 were randomly selected to study rumen contractions and ruminal pH, fermentation and ciliated protozoa before and after Jian Pi Li Qi San treatment. Before treatment, rumen contractions were reduced, pH was elevated and fermentation and ciliate numbers were reduced, compared to normal reference levels. After treatment, the number of rumen contractions increased above normal, the contraction amplitude quadrupled and the contraction duration more than doubled. Further, there was a slight reduction in rumen pH and fermentation and ciliates tripled. Although there were insufficient numbers of animals to make meaningful statistical comparisons, the trends were consistent. Jian Pi Li Qi San appears to be an effective treatment for bovine forestomach atony from simple indigestion and further clinical studies are warranted. [ABSTRACT FROM AUTHOR]

The movements of replacement breeding cattle have been implicated in the spread of many economically important cattle diseases. In this analysis, records from the Cattle Tracing System database were used to investigate the frequency and characteristics of replacement breeding cattle trade in Great Britain. During the 2006 calendar year, an estimated 48.7 per cent of beef herds and 47.8 per cent of dairy herds purchased at least one replacement breeding animal. Open beef herds purchased an average of 7.2 replacement animals (median: 4, range: 1-819) from 3.6 source herds (median: 2, range: 1-114), while open dairy herds purchased an average of 13.7 replacement animals (median: 7, range: 1-827) from 5.2 source herds (median: 3, range: 1-146). The most common animal types purchased by beef and dairy herds were open heifers and open lactating cows, respectively. Although the movements of purchased replacement breeding cattle accounted for only 13 per cent of individual movements in the between-herd contact network, they had a disproportionately strong influence on the risk of disease spreading through the industry as evidenced by their high betweenness centrality scores. Background: Cryptosporidiosis is one of the most important parasitic infections in human and animals. This study was designed for survey on the prevalence of Cryptosporidium infection in farms of Ilam, west of Iran, using parasitology method and genotyping by Nested PCR-RFLP. Methods: Fecal samples of 217 cattle were collected fresh and directly from the rectum of cattle. All of the samples were examined by microscopic observation after staining with modified Ziehl-Neelsen (MZN). Genomic DNA extracted by using EURx DNA kit. A Nested PCR-RFLP protocol amplifying 825 bp fragment of 18s rRNA gene conducted to differentiate species and genotyping of the isolates using SspI and VspI as restriction enzymes. Results: The prevalence of Cryptosporidium infection in cattle using both methods is 3.68%. Most of the positive cattle were calves under six months. Species diagnosis carried out by digesting the secondary PCR product with SspI that C. parvum generated 3 visible bands of 448, 247 and 106 bp and digested by VspI restriction enzyme generated 2 visible bands of 628 and 104bp. In this investigation all of the positive samples were Cryptosporidium parvum. Conclusion: C. parvum (bovine genotype) detected in all positive cattle samples in Ilam, west of Iran. The results of the present study can help for public health care systems to prevention and management of cryptosporidiosis in cattle and the assessment of cattle cryptosporidiosis as a reservoir for the human infection. Dairy welfare assessment programs are becoming more common on US farms. Outcome-based measurements, such as locomotion, hock lesion, hygiene, and body condition scores (BCS), are included in these assessments. The objective of the current study was to investigate the proportion of cows in the pen or subsamples of pens on a farm needed to provide an accurate estimate of the previously mentioned measurements. In experiment 1, we evaluated cows in 52 high pens (50 farms) for lameness using a 1- to 5-scale locomotion scoring system (1 = normal and 5 = severely lame; 24.4 and 6% of animals were scored ≥3 or ≥4, respectively). Cows were also given a BCS using a 1- to 5-scale, where 1 = emaciated and 5 = obese; cows were rarely thin (BCS ≤2; 0.10% of cows) or fat (BCS ≥4; 0.11% of cows). Hygiene scores were assessed on a 1- to 5-scale with 1 = clean and 5 = severely dirty; 54.9% of cows had a hygiene score ≥3. Hock injuries were classified as 1 = no lesion, 2 = mild lesion, and 3 = severe lesion; 10.6% of cows had a score of 3. Subsets of data were created with 10 replicates of random sampling that represented 100, 90, 80, 70, 60, 50, 40, 30, 20, 15, 10, 5, and 3% of the cows measured/pen. In experiment 2, we scored the same outcome measures on all cows in lactating pens from 12 farms and evaluated using pen subsamples: high; high and fresh; high, fresh, and hospital; and high, low, and hospital. For both experiments, the association between the estimates derived from all subsamples and entire pen (experiment 1) or herd (experiment 2) prevalence was evaluated using linear regression. To be considered a good estimate, 3 criteria must be met: R ² > 0.9, slope =1, and intercept = 0. In experiment 1, on average, recording 15% of the pen represented the percentage of clinically lame cows (score ≥3), whereas 30% needed to be measured to estimate severe lameness (score ≥4). Only 15% of the pen was needed to estimate the percentage of the herd with a hygiene score ≥3, whereas 30% to estimate the prevalence of severe hock lesions. Estimating very thin and fat cows required that 70 to 80% of the pen be measured. In experiment 2, none of the pen subsamples met our criteria for accurate estimates of herd prevalence. In conclusion, we found that both a higher percentage of the pen must be sampled to generate accurate values for relatively rare parameters and that the population measured plays an important role in prevalence estimates. More than 400 serotypes of Shiga toxin-producing Escherichia coli (STEC) have been implicated in outbreaks and sporadic human diseases. In recent years STEC strains belonging to serogroup O178 have been commonly isolated from cattle and food of bovine origin in South America and Europe. In order to explore the significance of these STEC strains as potential human pathogens, 74 German and Argentinean E. coli O178 strains from animals, food and humans were characterized phenotypically and investigated for their serotypes, stx-genotypes and 43 virulence-associated markers by a real-time PCR-microarray. The majority (n = 66) of the O178 strains belonged to serotype O178:H19. The remaining strains divided into O178:H7 (n = 6), O178:H10 (n = 1), and O178:H16 (n = 1). STEC O178:H19 strains were mainly isolated from cattle and food of bovine origin, but one strain was from a patient with hemolytic uremic syndrome (HUS). Genotyping of the STEC O178:H19 strains by pulsed-field gel electrophoresis revealed two major clusters of genetically highly related strains which differ in their stx-genotypes and non-Stx putative virulence traits, including adhesins, toxins, and serine-proteases. Cluster A-strains including the HUS-strain (n = 35) carried genes associated with severe disease in humans (stx2a, stx2d, ehxA, saa, subAB1, lpfAO113, terE combined with stx1a, espP,iha). Cluster B-strains (n = 26) showed a limited repertoire of virulence genes (stx2c, pagC, lpfAO113,espP, iha). Among O178:H7 strains isolated from deer meat and patients with uncomplicated disease a new STEC variant was detected that is associated with the genotypestx1c/stx2b/ehxA/subAB2/espI/[terE]/espP/iha. None of the STEC O178 strains was positive for locus of enterocyte effacement (LEE)- and nle-genes. Results indicate that STEC O178:H19 strains belong to the growing group of LEE-negative STEC that should be considered with respect to their potential to cause diseases in humans. Globalization has increased the potential for the introduction and spread of novel pathogens over large spatial scales necessitating continental-scale disease models to guide emergency preparedness. Livestock disease spread models, such as those for the 2001 foot-and-mouth disease (FMD) epidemic in the United Kingdom, represent some of the best case studies of large-scale disease spread. However, generalization of these models to explore disease outcomes in other systems, such as the United States’s cattle industry, has been hampered by differences in system size and complexity and the absence of suitable livestock movement data. Here, a unique database of US cattle shipments allows estimation of synthetic movement networks that inform a near-continental scale disease model of a potential FMD-like (i.e., rapidly spreading) epidemic in US cattle. The largest epidemics may affect over one-third of the US and 120,000 cattle premises, but cattle movement restrictions from infected counties, as opposed to national movement moratoriums, are found to effectively contain outbreaks. Slow detection or weak compliance may necessitate more severe state-level bans for similar control. Such results highlight the role of large-scale disease models in emergency preparedness, particularly for systems lacking comprehensive movement and outbreak data, and the need to rapidly implement multi-scale contingency plans during a potential US outbreak. Abstract: The objective of this study was to determine the influence of weekly locomotion scoring and, thus, early detection and treatment of lame cows by a veterinarian on lameness prevalence, incidence, duration of lameness, fertility and milk yield on one dairy farm in Northern Germany. Cows were distributed to two groups. Cows in Group A (n = 99) with a locomotion score (LS) > 1 were examined and treated. In Group B (n = 99), it was solely in the hands of the farmer to detect lame cows and to decide which cows received treatment. Four weeks after the beginning of the experimental period, the prevalence of cows with LS = 1 was higher in Group A compared with Group B. Prevalence of lame cows (LS > 1) increased in Group B (47.6% in Week 2 to 84.0% in Week 40) and decreased in Group A from Week 2 to Week 40 (50% to 14.4%; P < 0.05). Within groups, the monthly lameness incidence did not differ. The average duration of lameness for newly lame cows was 3.7 weeks in Group A and 10.4 weeks in Group B (P < 0.001). There was no effect on fertility and incidence of puerperal disorders. The 100-day milk yield was calculated from cows having their first four Dairy Herd Improvement (DHI) test day results during the experimental period. The objectives of this research were to estimate genetic parameters for body condition score (BCS) and locomotion (LOC), and to assess their relationships with angularity (ANG), milk yield, fat and protein content, and fat to protein content ratio (F:P) in the Italian Holstein Friesian breed. The Italian Holstein Friesian Cattle Breeders Association collects type trait data once on all registered first lactation cows. Body condition score and LOC were introduced in the conformation scoring system in 2007 and 2009, respectively. Variance (and covariance) components among traits were estimated with a Bayesian approach via a Gibbs sampling algorithm and an animal model. Heritability estimates were 0.114 and 0.049 for BCS and LOC, respectively. The genetic correlation between BCS and LOC was weak (-0.084) and not different from zero; therefore, the traits seem to be genetically independent, but further investigation on possible departures from linearity of this relationship is needed. Angularity was strongly negatively correlated with BCS (-0.612), and strongly positively correlated with LOC (0.650). The genetic relationship of milk yield with BCS was moderately negative (-0.386), and was moderately positive (0.238) with LOC. These results indicate that high-producing cows tend to be thinner and tend to have better locomotion than low-producing cows. The genetic correlation of BCS with fat content (0.094) and F:P (-0.014) was very weak and not different from zero, and with protein content (0.173) was weak but different from zero. Locomotion was weakly correlated with fat content (0.071), protein content (0.028), and F:P (0.074), and correlations were not different from zero. Phenotypic correlations were generally weaker than their genetic counterparts, ranging from -0.241 (BCS with ANG) to 0.245 (LOC with ANG). Before including BCS and LOC in the selection index of the Italian Holstein breed, the correlations with other traits currently used to improve type and functionality of animals need to be investigated. The objectives of this study were to estimate the prevalence of lameness and determine locomotion score distribution in dairy cows housed in free-stalls. The study was realized in 24 selected free-stall farms in Transylvania. Lameness was assessed based on the locomotion score devised by Sprecher et al. The percentage of cows with different locomotion scores was established. The lameness prevalence was calculated as the proportion of cows with locomotion score 3 or more. The results were statistically processed using the SPSS software. Of the 2519 cows assessed, normal locomotion was found in 27.04% in winter (24.69% in summer), slight lameness in 40.69% (45.30% in summer), moderate lameness in 27.87% (25.96% in summer), lameness in 3.33% (2.82% in summer) and severe lameness in 1.07% (1.23% in summer). The prevalence of lameness in the investigated barns varied between 5% and 49.35% with a mean prevalence of 31.04% in winter (28.54% in summer). Although the value obtained in the warm season is lower, the difference between the two seasons was insignificant (P > 0.05). Early social development in long-lived social mammals has important implications for adult behaviour, particularly in taxa that exhibit stable long-term bonds. In one such species, bottlenose dolphins, calves have precocious locomotion, enabling them to separate from their mothers soon after birth and associate with other dolphins in the absence of direct maternal influence. To investigate mother and calf social patterns while together and separated, we analysed the ego networks of 27 mother–calf pairs constructed using group composition data from focal follows and evaluated differences based on calf sex. When separated, all calves had larger, less dense ego networks than their mothers, whereas ego networks of mother and calf when together were similar in size to those of calves, but significantly less dense. Most intriguingly, during separations, male calves’ relationships with other male calves were stronger than expected, foreshadowing the long-term bonds between adult male alliance partners. Female–female calf relationships were not stronger than expected, but when together, mothers and female calves had strong relationships with juvenile females. These results support the social bonds hypothesis and suggest that temporary separations allow calves to build and strengthen their social networks. Since bottlenose dolphins show bisexual philopatry and infant relationships can persist into adulthood, calf independence and early social development probably have implications for future success.
