ABSTRACT Parasitic infections in dairy cattle reduce herd immunity, milk production, and conception rates. This leads to higher production costs, compromised animal welfare, and increased interest in extralabel drug use. The extralabel use of anthelmintics poses food safety risks for consumers since appropriate withdrawal intervals in milk have yet to be established. Although topical eprinomectin has no milk withdrawal time, more research is needed to determine the residues present in milk after subcutaneous administration. This study aimed to characterize the pharmacokinetics of injectable eprinomectin in dry dairy cows. We hypothesized that, when given at the labeled dose, eprinomectin residues in dry dairy cattle would be below the FDA milk tolerance at the onset of lactation. Plasma was collected daily from 13 mature dairy cattle for 7 days postadministration, followed by periodic samples for 90 days. After calving, milk was collected daily until 90 days. Eprinomectin concentrations were measured using HPLC‐fluorescence detection. The maximum eprinomectin concentration in plasma and milk was approximately 36 ng/mL 43 h after administration and 3 ng/mL at the onset of lactation, respectively. The low eprinomectin levels in milk collected from these lactating dairy cattle suggest that administering eprinomectin at dry‐off is unlikely to result in violative residues. However, subcutaneous eprinomectin in lactating dairy cattle would be hard to justify unless there is evidence that the approved topical formulation is clinically ineffective. [ABSTRACT FROM AUTHOR]

African animal trypanosomosis (AAT) affects the livestock industry, impacting protein intake and herders' livelihoods. Despite the presence of vector flies, livestock owners often rely solely on chemotherapy, with diminazene aceturate being a common choice, particularly among non-elite herders. Cattle herds were sampled during an outbreak and re-sampled 8 weeks post-treatment with diminazene aceturate (Nonazin®) in Ogun and Osun states, Nigeria. Initial sampling was conducted in October 2019 and January 2020 for farms A and B, respectively. A total of 316 cattle, comprising 20 males and 296 females, were sampled through convenient sampling of the entire herd. Nzi traps were deployed to capture Trypanosoma-transmitting vectors. Haematological parameters, microscopic, and molecular assessments were conducted. The total transmitting vectors captured averaged 3.74 fly/trap/day, with Stomoxys niger being the most captured biting fly. Post-treatment prevalence rates were 21.5% and 2.5% with PCR and thin blood smear, respectively, with Trypanosoma congolense and T. vivax being the species detected by both diagnostic methods. The average parasitaemia before treatment was recorded at 1.95 × 104 ± 1.3 × 104, signifying a high parasitic infection, while post-treatment parasitaemia was 1.5 × 102 ± 1.4 × 102. The PCV, Hb, and RBC counts decreased, causing macrocytic normocytic anaemia before treatment, while post-treatment values suggested lower haemoglobin and MCH values with less-characteristic anaemia. Post-treatment trypanosome prevalence suggests the existence of Trypanosoma-resistant strains, potentially widespread in southwest Nigeria. The present study demonstrates successful herd sanitation and eradication of contagious mastitis caused by Staphylococcus aureus genotype B (GTB) in an entire Swiss district (Ticino) including 3,364 dairy cows from 168 farms. Herd sanitation included testing of all cows using a highly GTB-specific and sensitive real-time quantitative PCR (qPCR) assay, implementation of related on-farm measures, appropriate antibiotic therapy of GTB-positive cows, and culling of therapy-resistant animals, respectively. A treatment index was used as an objective criterion to select GTB-positive cows eligible for culling and replacement payment. Sixty-two herds (37%) were initially GTB-positive with a cow prevalence between 10% and 100% and were submitted to sanitation. Twenty months after the start of the campaign, all of these herds were free from S. aureus GTB, whereby 73% of them were sanitized during the first 7 mo. At the cow level, a total of 343 animals were infected. Fifty of them were immediately culled and farmers were financially compensated based on their treatment index value. The remaining 293 cows were intramammarily treated with antibiotics either during lactation using the combination of cephalexin-kanamycin or penicillin-gentamicin or at dry-off using cloxacillin. Out of these cows, 275 (93.9%) were treated successfully, meaning that their milk was twice GTB-negative by qPCR after therapy. For lactational treatment, control samples were taken ≥10 and ≥20 d after treatment, for dry-off treatment ≥14 and ≥24 d after parturition. Neither lactation number nor SCC before treatment of the cow nor the type of therapy was associated with therapeutic cure. Using data of 30 GTB-positive and 71 GTB-negative herds (1,855 observations), the effect of GTB sanitation on bulk tank milk SCC (BTSCC) was evaluated by applying a linear mixed statistical model. In the year before sanitation, BTSCC was always higher in GTB-positive than in GTB-negative herds. After the start of the campaign, BTSCC declined rapidly in the herds under GTB sanitation and achieved values that no longer differed statistically from those of GTB-free herds after only 2 mo, remaining very similar for the rest of the campaign. The farmers were very satisfied with the outcome of the campaign because all GTB-positive herds could be sanitized rapidly, sanitation was sustainable, and milk quality increased. This study determined the frequency of positive tests for bovine tuberculosis in animals and breeders/establishments in Rio Grande do Norte, Brazil. Data were provided by the Instituto de Defesa e Inspeção Agropecuária do Rio Grande do Norte (IDIARN), from its Unidade Local de Sanidade Animal e Vegetal (ULSAV'S), acquired in monthly reports issued by qualified Veterinarians under the National Program for the Control and Eradication of Brucellosis and Animal Tuberculosis (PNCEBT), from June 2012 to December 2021. For diagnosis, the comparative cervical test was used as a routine and confirmatory test. A total of 45,804 cattle were tested, 53 (0.1%) of which tested positive. Secondary data are essential in the evaluation of sanitary measures, allowing rapid generation of hypotheses about diseases and providing support for decision-making. Measures such as raising awareness among producers, sanitary control in the acquisition and sale of cows for reproduction, inspection of sanitary barriers, and conducting epidemiological surveys to understand the actual situation of this disease in the state of Rio Grande do Norte are crucial. Mycobacterium avium subsp. paratuberculosis (MAP) causes Johne's disease, or paratuberculosis (PTB) in ruminants, besides having zoonotic potential. It possibly changes the gut microbiome, but no conclusive data are available yet. This study aimed at investigating the influence of MAP on the faecal microbiome of cattle naturally infected with PTB. In a follow up period of 10 months, PTB status was investigated in a herd of dairy cattle with history of clinical cases. Each animal was tested for MAP infection using serum and milk ELISA for MAP anti-bodies and IS900 real-time PCR and recombinase polymerase amplification assays for MAP DNA in the faeces and milk monthly for 4 successive months, then a last one after 6 months. The faecal samples were subjected to 16S rDNA metagenomic analysis using Oxford Nanopore Sequencing Technology. The microbial content was compared between animal groups based on MAP positivity rate and production status. All animals were MAP positive by one or more tests, but two animals were consistently negative for MAP DNA in the faeces. In all animals, the phyla firmicutes and bacteroidetes were highly enriched with a small contribution of proteobacteria, and increased abundance of the families Oscillospiraceae, Planococcaceae, and Streptococcacaceae was noted. Animals with high MAP positivity rate showed comparable faecal microbial content, although MAP faecal positivity had no significant effect (p > 0.05) on the microbiome. Generally, richness and evenness indices decreased with increasing positivity rate. A significantly different microbial content was found between dry cows and heifers (p < 0.05). Particularly, Oscillospiraceae and Rikenellaceae were enriched in heifers, while Planococcaceae and Streptococcaceae were overrepresented in dry cows. Furthermore, abundance of 72 genera was significantly different between these two groups (p < 0.05). Changes in faecal microbiome composition were notably associated with increasing MAP shedding in the faeces. The present findings suggest a combined influence of the production status and MAP on the cattle faecal microbiome. Genetic, environmental, technological and financial resources are used differently in cattle herds that participate in the same breeding programme. The percentages of calves sired by sires within their own herd or from external herds vary across herds, as do the intensities of use of reproductive biotechnologies. These divergences may be related to differences in the indicators of genetic performance for economic traits. The aim of this study was to determine the factors related to herd structure and genetic resource utilization that exert the greatest influence on the genetic merit of seedstock herds within a Nellore breeding programme. The database comprised 21 factors, along with genomic‐enhanced expected progeny differences (GE‐EPDs) for growth, reproductive and carcass traits, as well as a selection index of animals from 128 herds. By combining principal component analysis and cluster analysis, we were able to group the herds. We identified statistically significant differences (p < 0.05) in the mean values of the factors, GE‐EPDs and genetic trends among the groups of herds. Differences in the percentage of sires from external herds and in sire age between the groups of herds were the factors most associated with differences in mean GE‐EPDs and genetic trends. Using young sires from other herds or lineages is an effective strategy in animal breeding. By enhancing genetic variability, this approach does not only improve the genetic quality of herds but also accelerates genetic progress in desired traits over time. Therefore, to ensure the success of this strategy, it is crucial that seedstock herds undergo a thorough selection process aimed at maximizing the genetic potential of future generations of beef cattle. Accurate livestock counts are essential for effective pastureland management. High spatial resolution remote sensing, coupled with deep learning, has shown promising results in livestock detection. However, challenges persist, particularly when the targets are small and in a heterogeneous environment, such as those in African rangelands. This study evaluated nine state-of-the-art object detection models, four variants each from YOLOv5 and YOLOv8, and Faster R-CNN, for detecting cattle in 10 cm resolution aerial RGB imagery in Kenya. The experiment involved 1039 images with 9641 labels for training from sites with varying land cover characteristics. The trained models were evaluated on 277 images and 2642 labels in the test dataset, and their performance was compared using Precision, Recall, and Average Precision (AP0.5–0.95). The results indicated that reduced spatial resolution, dense shrub cover, and shadows diminish the model's ability to distinguish cattle from the background. The YOLOv8m architecture achieved the best AP0.5–0.95 accuracy of 39.6% with Precision and Recall of 91.0% and 83.4%, respectively. Despite its superior performance, YOLOv8m had the highest counting error of −8%. By contrast, YOLOv5m with AP0.5–0.95 of 39.3% attained the most accurate cattle count with RMSE of 1.3 and R2 of 0.98 for variable cattle herd densities. These results highlight that a model with high AP0.5–0.95 detection accuracy may struggle with counting cattle accurately. Nevertheless, these findings suggest the potential to upscale aerial-imagery-trained object detection models to satellite imagery for conducting cattle censuses over large areas. In addition, accurate cattle counts will support sustainable pastureland management by ensuring stock numbers do not exceed the forage available for grazing, thereby mitigating overgrazing. Long noncoding RNAs (lncRNAs) are RNA molecules with a length greater than 200 nucleotides that do not code for functional proteins. Although, genes play a vital role in immune response against a disease, it is less known that lncRNAs also contribute through gene regulation. Bovine tuberculosis is a significant zoonotic disease caused by Mycobacterium bovis (M. bovis) in cattle. Here, we report the in-silico analysis of the publicly available transcriptomic data of calves infected with M. bovis. A total of 51,812 lncRNAs were extracted across all the samples. A total of 216 genes and 260 lncRNAs were found to be differentially expressed across all the 4 conditions—infected vs uninfected at 8- and 20-week post-infection (WPI), 8 vs 20-WPI of both infected and uninfected. Gene Ontology and Functional annotation showed that 8 DEGs were annotated with immune system GOs and 2 DEGs with REACTOME immune system pathways. Co-expression analysis of DElncRNAs with DEGs revealed the involvement of lncRNAs with the genes annotated with Immune related GOs and pathways. Overall, our study sheds light on the dynamic transcriptomic changes in response to M. bovis infection, particularly highlighting the involvement of lncRNAs with immune-related genes. The identified immune pathways and gene–lncRNA interactions offer valuable insights for further research in understanding host–pathogen interactions and potential avenues for genetic improvement strategies in cattle. Simple Summary: In recent years, the interest in virtual fencing systems for flexible animal enclosure management has increased. However, implementing such systems requires significant investment due to the need for individual collars, hindering large-scale adoption. This study examines the spatial distribution of a cow herd using GPS locations from the Nofence© system, aiming to minimize the number of collars required by identifying patterns in positions and ranks to derive a hierarchy. Contrary to expectations, no distinct pattern emerged, suggesting future studies should focus on individual interactions rather than viewing the herd as a single unit. Interest in virtual fencing has increased due to its flexibility for agriculture and rewilding. However, systems like Nofence© require large financial investments, and the need for individual collars complicates large-scale use. If cattle herds maintain cohesive groups around leading individuals, fewer collars could be used, thereby enhancing cost efficiency. This study investigates the pattern in spatial distribution in a herd of 17 Angus cows on Fanø in Denmark with GPS locations, using a Nofence© system. The aim of this paper is to determine how individuals position themselves in a herd, spatially, and identify a pattern in ranks. The method used in this study examines the distances between an individual to the rest of the herdmates using nearest neighbour and descriptive statistics. Contrary to expectations, this study did not reveal a distinct pattern in herd distribution. While some tendencies in spatial distribution patterns were observed, only a low concordance could be found ( W = 0.15 , p < 0.001 ), indicating great variability in the cattle's ranks. A cumulative curve of the ranks estimated over the entire periods, however, allowed a rough estimation of the hierarchy and allowed identification of the highest-ranked cows, making the use of a cumulative curve a possible solution to finding the high-ranked cows. This research underscores the complexity of cattle social structures and highlights the need for extended observation periods and alternative methodologies to enhance the cost-effectiveness and scalability of virtual fencing in agricultural and rewilding contexts. Emerging infectious diseases with zoonotic potential often have complex socioecological dynamics and limited ecological data, requiring integration of epidemiological modeling with surveillance. Although our understanding of SARS-CoV-2 has advanced considerably since its detection in late 2019, the factors influencing its introduction and transmission in wildlife hosts, particularly white-tailed deer (Odocoileus virginianus), remain poorly understood. We use a Susceptible-Infected-Recovered-Susceptible epidemiological model to investigate the spillover risk and transmission dynamics of SARS-CoV-2 in wild and captive white-tailed deer populations across various simulated scenarios. We found that captive scenarios pose a higher risk of SARS-CoV-2 introduction from humans into deer herds and subsequent transmission among deer, compared to wild herds. However, even in wild herds, the transmission risk is often substantial enough to sustain infections. Furthermore, we demonstrate that the strength of introduction from humans influences outbreak characteristics only to a certain extent. Transmission among deer was frequently sufficient for widespread outbreaks in deer populations, regardless of the initial level of introduction. We also explore the potential for fence line interactions between captive and wild deer to elevate outbreak metrics in wild herds that have the lowest risk of introduction and sustained transmission. Our results indicate that SARS-CoV-2 could be introduced and maintained in deer herds across a range of circumstances based on testing a range of introduction and transmission risks in various captive and wild scenarios. Our approach and findings will aid One Health strategies that mitigate persistent SARS-CoV-2 outbreaks in white-tailed deer populations and potential spillback to humans. SARS-CoV-2, the virus responsible for the COVID-19 pandemic in humans, has been detected in white-tailed deer (Odocoileus virginianus) yet we have limited understanding of its introduction and transmission within deer populations. Here, we use epidemiological models to describe SARS-CoV-2 introduction and transmission patterns within wild and captive white-tailed deer populations. We found that captive deer herds faced a higher risk of spillover of SARS-CoV-2 from humans and higher transmission rate compared to wild deer. Despite these differences in wild and captive contexts, we found that transmission in both wild and captive contexts often resulted in persisting circulation of SARS-CoV-2. This circulation was determined by deer-deer transmission in both wild and captive contexts, rather than by high rates of introduction from humans. Outbreaks in wild populations were greater when these animals could interact with captive deer populations circulating SARS-CoV-2. SARS-CoV-2 appears to circulate in both captive and wild deer across a range of circumstances. Enhancing reproductive performance is a key strategy to mitigate involuntary culling rates, thereby extending productive life (PL) and ultimately improving profitability in dairy cattle herds. A piecewise Weibull proportional hazards model was used to investigate the effect of several important reproductive traits on PL in Holstein dairy cows. Data comprised 200,747 lactation records from 82,505 cows sired by 1952 bulls across 36 dairy herds. PL was defined as the number of days from the first calving to the last milk record or censoring. The statistical model accounted for the time‐dependent fixed effects of changes in herd size, year‐season, milk production, fat and protein contents, and the time‐independent fixed effect of age at first calving. Herd‐year and sire effects were also included as random effects. Reproductive traits include calving traits such as calving ease (CE), calf size (CZ), and calf survival (CS), as well as female fertility traits such as number of inseminations per conception (NI), days from calving to first service (CFS), days from first service to conception (FSC), and days open (DO). All reproductive traits had a significant effect on PL (p < 0.001). Each reproductive trait was analysed separately. The relative risk (RR) of being culled increased as the severity of calving difficulties increased in both primiparous and multiparous cows. Cows that calved small or large calves showed a higher risk of being culled compared with those that calved medium size calves. The increased RR of culling was observed only for primiparous cows that gave birth to dead calves. In addition, cows that required more NI, a longer CFS, FSC, and DO had shorter longevity. These insights can deepen our comprehension of the factors affecting PL and provide information for refining management and breeding strategies, which could lead to increased profitability and sustainability in Iranian dairy farming. Introduction: Bovine cattle raising is the most important line of the economy of Caquetá department and El Doncello municipality, according to the figures given by the Departmental Committee of Cattlemen there is a herd of 2,175,065 cattle heads in the region, many of the rural families depend on said economic activity to generate their income; To do this, the life analysis methodology examines the assets that communities have which are called community capital, and the approach they use to meet their fundamental human needs. Objective: To assess sustainable means and the application of community capital in cattle rancher families in the municipality of El Doncello, Caquetá (Colombia). Materials and methods: For the collection of information the producer families of the municipality of El Doncello were assessed; based on the sources of the Departmental Committee of Cattlemen, for this purpose families were surveyed and participatory workshops were carried out using the livelihood methodology framing the community capitals (human, natural, financial, physical and social) using 36 variables for the assessment of them, and the fundamental human needs (basic of the person, the environment and of action) that grouped 15 analysis variables; Subsequently, the interpretation and analysis of the data was made through a principal component analysis-PCA relating the typologies and each of the variables assessed in this study; all the data were run in the InfoStat Program. Results: It was found that 939 families base their economy on livestock production in the municipality of El Doncello, the human capital of these families was distributed in 72 men representing 15.55 %, 81 women (17.49 %), 143 boys (30.89 %) and 167 girls (36.07 %), finding three types of families: Typology 1: families with <50 bovines on their farms; Typology 2: between 51 and 100 bovines, and Typology 3: >101 bovines, where financial capital is the one that represents the greatest contribution given its characteristics of income generation and well-being for families. The principal component analysis-ACP carried out for social capital divided according to the first principal component-PC1 with a contribution of 62.3 % to Family Typology 1 (families with <50 head of cattle) to the positive end of said component strongly associated with the variables greater participation in projects and organizations to which they belong, with the particularity of having a greater number of members at home; from Typology 3 to the negative end of said component associated with the variables greater number of shared spaces in which they participate and greater level of family participation. In the same way, for natural capital, a separation was observed between Family Typology 1, associated with the variables greater number of water sources, mainly due to the fact that a large part of these producers were in the mountain range zone, greater area in stubble, greater number of spaces used and more activities for consumption, such as plantations of plantain, cocoa, cassava and other subsistence crops and Family Typology 2 strongly associated with the variables greater number of trees in pastures and greater percentage of natural products. One of the most important capitals within the producers is the financial one, for which the following was observed: the first principal component-PC1, with a contribution of 43.7%, separates Typology 1 from the positive end of said component associated with the variable income from daily wages and income from subsidies, these families are the ones that generate the least amount of assets and through which they diversify their income that are not only dependent on livestock production as such, but also on other types of economic activities, of the Typology 2 to the negative extreme associated with the variables income from transfers, credits, income from formal employment and income from the sale of processed products, such as cheese. The second main component-PC2, with a contribution of 24.4 %, groups Family Typology 3 strongly associated with the highest income from the sale of space-use products, These families, by owning a greater number of cattle heads, base a large part of their economy in said activity, but they diversify it with other types of income such as rubber crops, cocoa, pineapple, plantain, cassava, among others. Finally, the fundamental human needs-FHN can be defined as very similar for the three types of families studied, with slight adjustments in the basic needs for Typology 1, where food, health, shelter and security are classified as bad and regular. Although the situation does not vary much for Typology 2, but it does look better for Typology 3 in which satisfaction is good, except for security, everyone has the same perception, this is mainly due to the presence of certain illegal armed groups in the region. Regarding the needs of the person, the environment and action, there are no variations between the different types. Conclusions: Finally, it was possible to observe a high cultural tie that cattle ranchers have for this economic activity that is evidenced in human capital, where we see the number of people who directly depend on livestock and its derivatives. Also, in terms of financial capital, it can be observed that there is a great diversification of the income of families that not only depend on livestock, but also on income from formal employment, daily wages, loans and sales of other crops such as plantain, cassava, rubber and cocoa; In terms of social and natural capital, a good sustainable appropriation of environmental care and the participation of rural communities are observed. Finally, a low contribution to the fundamental human needs of the producing communities is denoted, this mainly due to the social and economic conditions of the region; In this sense, this study has repercussions as a first diagnosis in the management of the livelihoods of rural cattle-raising communities in the department of Caquetá. Understanding how disease moves through wildlife communities is essential to managing outbreaks of zoonotic diseases across the globe. Bovine tuberculosis is a disease caused by the bacterium Mycobacterium bovis that can threaten domestic and wildlife species. The mechanism by which Mycobacterium bovis is spread between species is still poorly understood. Previous reviews are limited in the breadth of species considered and are primarily concerned with transmission from wildlife to domestic species.We conducted a review and analysis of Mycobacterium bovis prevalence rates in European wildlife species to identify species of concern for the transmission of bovine tuberculosis in a wildlife community. We subsequently conducted a narrative review of these species assessing the risk of Mycobacterium bovis transmission in a wildlife community based on available literature.We calculated weighted mean disease prevalence rates to be highest in fallow deer (Dama dama, 20%), Eurasian badgers (Meles meles, 11%), wild boar (Sus scrofa, 9%) and red foxes (Vulpes vulpes, 4%). We considered these species to be of particular concern for the transmission of Mycobacterium bovis and selected them as the focus of our narrative review and risk assessment. Our risk assessment considered disease pathology, spatiotemporal activity patterns and animal behaviour as factors affecting the likelihood of Mycobacterium bovis transmission between wildlife species.We found that prior research has principally focused on a few individual species, but that Mycobacterium bovis transmission through a wildlife community is likely more complex. We determined that disease transmission between multiple species may compound the severity of an outbreak of bovine tuberculosis. Broad, multi‐species sampling campaigns and standardised Mycobacterium bovis testing protocols should be implemented in future studies. The article provides an overview of IBR testing in Irish beef herds as part of the National Beef Welfare Scheme (NBWS) 2023. The study found that IBR remains endemic in the Irish beef cattle sector, but at a lower prevalence than previously reported. The herd-level prevalence was 48.8%, and the animal-level prevalence was 11.4%. The study suggests that targeted interventions in herds with a within-herd prevalence below 20% could lead to containment and potentially facilitate the eradication of IBR on a broader scale. Local comparisons of effects, responses and mitigations to the Covid-19 pandemic are of vital importance in building a sustainable tourism. This is particularly the case for conservancies in Africa which is largely dependent on international tourism. Qualitative interviews were carried out in the Kenya Maasai Mara Wildlife Conservancies Association (MMWCA)with landowners, lodge managers and staff, tourism operators, community organisations and NGOs between January and May 2021. The MMWCA is an important case study as conservancies pay lease payments to more than 14,528 landowners through tourism revenues. The results show how partner conservancies took different paths in securing payments of leases and salaries by rotating staff, attracting international funding and by targeting domestic tourism. Meanwhile, landowners experimented with alternative economic activities such as cattle herding and diary production. The study shows the strength of MMWCA as a stakeholder partnership to proactively design measures including renegotiation of lease-payments, in soliciting external funding and in re-distributing funding. The positive role of domestic tourism is also stressed. The pandemic brought to the forefront discussions on equity and benefit sharing and on the sustainability of the model itself. Recommendations are given to strengthen possibilities for alternative incomes sources and for a diversification of strategies of the MMWCA partners, including the need to stimulate domestic tourism as a parallel source of income. Bovine tuberculosis (bTB) continues to have significant economic and veterinary health impacts on cattle herds where the disease remains endemic. The continual tailoring of policies to address such maintenance requires an in-depth analysis of national data, underpinning new control strategies. In Ireland, when outbreaks occur, ancillary testing of herd mates deemed to be at the highest risk of exposure to reactors is undertaken using the interferon gamma (GIF) test. This highest risk cohort was hypothesised to be of a higher future risk despite this ancillary testing. We used a dataset from Ireland to model bovine test failure to the comparative tuberculin skin test using a survival analysis (observations: 39,248). Our primary exposure of interest was whether an animal that tested negative had a GIF test after the disclosure of infection within a herd during a bTB breakdown. There was evidence that animals with a negative GIF test during a breakdown had an increased risk of failing a test relative to other animals from the same herds without this exposure. The time to failure was 48.8% (95%CI: 38.3–57.5%) shorter for the exposed group relative to the unexposed group during a two-year follow-up period (2019–2022; time ratio: 0.51; 95%CI: 0.43–0.62; p < 0.001). The results from this study suggest that animals who were GIF-tested, having been deemed to have a higher risk of exposure, subsequently had shorter time-to-test failure periods. The absolute numbers of failure are small (only 2.5% of animals go on to fail during 2-year follow-up). Importantly, however, a high proportion of these high-risk herds included in the dataset failed at least one test at the follow-up (21/54 herds), impacting breakdown duration or recurrence. The frequent zoonotic disease known as "bovine tuberculosis" is brought on by the Mycobacterium bovis bacteria, which can infect both people and animals. The aim of this review article is to provide an explanation of the etiology, history, epidemiology, pathogenesis, clinical symptoms, diagnosis, transmission, risk factors, public health importance, economic impact, treatment, and control of bovine tuberculosis. Primarily, bovine tuberculosis affects cattle, but other animals may also be affected. Bovine tuberculosis is present throughout the world, with the exception of Antarctica. Cattle that contract bovine tuberculosis might suffer from a persistent, crippling illness. In the early stages of the disease, there are no symptoms. The tuberculin test is the primary method for detecting bovine tuberculosis in cows. Depending on its localized site in the infected animal, M. bovis can be found in respiratory secretions, milk, urine, feces, vaginal secretions, semen, feces, and exudates from lesions (such as lymph node drainage and some skin lesions). This illness generally lowers cattle productivity and could have a negative financial impact on the livestock business, particularly the dairy industry. The most effective first-line anti-tuberculosis chemotherapy consists of isoniazid, ethambutol, rifampin, and streptomycin. Second-line drugs used against bovine tuberculosis include ethionamide, capreomycin, thioacetazone, and cycloserine. To successfully control and eradicate bovine tuberculosis, developed nations have implemented routine testing and culling of infected animals under national mandatory programs. The aim of this work was to analyze the R. microplus (Canestrini, 1888) infestation in two bovine herds with different degrees of natural resistance (i.e., Hereford and Braford) to ticks subjected to an identical chemical treatment scheme to ticks at the same farm, to demonstrate the impact on tick control of the incorporation of a more resistant bovine breed. Two groups of ten Hereford and Braford cows each were subjected to eleven chemical treatments between August 2022 and October 2023 (four fluazuron, two fipronil 1%, one ivermectin 3.15% and four immersion in a dipping vat with a combination of cypermethrin 10% and ethion 40%). Tick population was shown to be susceptible to ivermectin, fluazuron and the mix cypermethrin 10%-ethion 40% and resistant to fipronil according to in vitro tests. Tick infestation was significantly greater in the Hereford cows than in the Braford cows. Tick infestation in both Hereford and Braford breeds was similar when treatment with functional drugs was applied, but when a block of the treatments was done with drugs with decreased functionality due to resistance (i.e. fipronil), treatment failure was manifested more strongly in the most susceptible breed. The incorporation of cattle breeds with moderate or high resistance to R. microplus is instrumental to optimize the efficacy and sustainability of chemical control of ticks in a scenario where resistance to one or more chemical groups is almost ubiquitous, because it favors the biological control of this parasite. In November 2021, an investigation was conducted into an outbreak of abortion, stillbirth, and the birth of calves with congenital abnormalities (arthrogryposis and hydranencephaly) at a dairy farm in Dasht-e-Mughan city, Ardabil province. A total of 70 cows experienced these issues. To determine the cause of the outbreak, post-mortem brain tissue samples were collected from two calves affected by hydranencephaly, which occurred shortly after their birth. Polymerase chain reaction (PCR) testing was conducted for multiple viruses, including bovine viral diarrhea (BVD), border disease, Akabane, Schmallenberg, and bluetongue viruses (BTVs). The samples were positive only for Akabane virus. Serum samples were collected from a group of 60 cattle, consisting of 45 adult cows and 15 younger calves aged between 8 to 10 months. These samples were analyzed to detect the presence of antibodies against the Akabane and Schmallenberg viruses. Both of these viruses are known to be responsible for causing abortion, stillbirth, and congenital abnormalities in calves. Among 45 cows that tested by competitive enzyme-linked immunosorbent assay (cELISA), 26.66% and 33.33% exhibited antibodies against Akabane and Schmallenberg viruses, respectively. Notably, 20.00% of cows co-exhibited antibodies for both viruses. Despite PCR evidence implicating Akabane virus as the principal etiology of clinical signs observed in the affected herd, the high coseropositivity to Schmallenberg virus, warrants a thorough investigation into potential viral interactions. Further research is required to determine the source of the virus and their transmission routes. This study aimed to enhance our understanding of the agreement between two sampling methods for the detection of bovine respiratory disease (BRD) pathogens in calves using high-throughput real-time qPCR (ht-RT-qPCR). In total, 233 paired nasal swab (NS) and non-endoscopic bronchoalveolar lavage (nBAL) samples were collected from 152 calves from 12 Danish cattle herds. In 202 of the observations, the calves were examined using a standardized clinical protocol. Samples were tested for three viruses (bovine respiratory syncytial virus, bovine corona virus, and influenza D virus) and six bacteria (Histophilus somni, Mannheimia haemolytica, Mycoplasma bovis, Mycoplasma species, Pasteurella multocida, and Truepurella pyogenes). The results showed age-related differences in disease and pathogen occurrence, with the highest detection rates in calves aged 35 days or older. Poor to moderate agreement was found between the NS and nBAL results. The presence of Mannheimia haemolytica in both NS and nBAL in younger calves and in nBAL in older calves was associated with clinical BRD. There was a potential link between BRD and influenza D virus in older calves, although it was only found in one herd in a small sample size. Overall, NS was a relatively poor predictor of pathogens in the lower respiratory tract. A recent article in the New England Journal of Medicine discusses the detection of avian influenza A(H5N1) virus in wastewater in ten cities. The highly pathogenic virus was detected in samples from March 4 to July 15, 2024, after not being detected in previous samples. The article suggests that wastewater monitoring can be a useful tool for surveillance and detection of viral adaptations. Background: Peste des Petits Ruminants (PPR) is a world organization for animal health (WOAH) notifiable and economically important transboundary, highly communicable viral disease of small ruminants. PPR virus (PPRV) belongs to the genus Morbillivirus of the family Paramyxoviridae. Aim: The present cross-sectional epidemiological investigation was accomplished to estimate the apparent prevalence and identify the risk factors linked with peste des petits ruminants (PPR) in the previously neglected northern border regions of Pakistan. Method: A total of 1300 samples (serum = 328; swabs = 972) from 150 flocks/herds were compiled from sheep (n = 324), goats (n = 328), cattle (n = 324), and buffaloes (n = 324) during 2020–2021 and tested using ELISA for detection of viral antibody in sera or antigen in swabs. Results: An overall apparent prevalence of 38.7% (504 samples) and an estimated true prevalence (calculated by the Rogan and Gladen estimator) of 41.0% (95% CI, 38.0–44 were recorded in the target regions. The highest apparent prevalence of 53.4% (85 samples) and the true prevalence of 57.0%, 95% Confidence Interval (CI) were documented in the Gilgit district and the lowest apparent prevalence of 53 (25.1%) and the true prevalence of 26.0%, 95% Confidence Interval (CI), 19.0–33.0) was reported in the Swat district. A questionnaire was designed to collect data about associated risk factors that were put into a univariable logistic regression to decrease the non-essential assumed risk dynamics with a P-value of 0.25. ArcGIS, 10.8.1 was used to design hotspot maps and MedCalc's online statistical software was used to calculate Odds Ratio (OR). Some of the risk factors significantly different (P < 0.05) in the multivariable logistic regression were flock/herd size, farming methods, nomadic animal movement, and outbreaks of PPR. The odds of large-sized flocks/herds were 1.7 (OR = 1.79; 95% Confidence Interval (CI) = 0.034–91.80%) times more likely to be positive than small-sized. The odds of transhumance and nomadic systems were 1.1 (OR = 1.15; 95% Confidence Interval (CI) = 0.022–58.64%) and 1.0 (OR = 1.02; 95% Confidence Interval (CI) = 0.020–51.97%) times more associated to be positive than sedentary and mixed farming systems, respectively. The odds of nomadic animal movement in the area was 0.7 (OR = 0.57; 95% Confidence Interval (CI) = 0.014–38.06%) times more associated to be positive than in areas where no nomadic movement was observed. In addition, the odds of an outbreak of PPR in the area were 1.0 (OR = 1.00; 95% Confidence Interval (CI) = 0.018–46.73%) times more associated to be positive than in areas where no outbreak of PPR was observed. Conclusions: It was concluded that many northern regions considered endemic for PPR, large and small ruminants are kept and reared together making numerous chances for virus transmission dynamic, so a big threats of disease spread exist in the region. Zoonotic diseases represent a significant societal challenge in terms of their health and economic impacts. One Health approaches to managing zoonotic diseases are becoming more prevalent, but require novel thinking, tools and cross-disciplinary collaboration. Bovine tuberculosis (bTB) is one example of a costly One Health challenge with a complex epidemiology involving humans, domestic animals, wildlife and environmental factors, which require sophisticated collaborative approaches. We undertook a scoping review of multi-host bTB epidemiology to identify trends in species publication focus, methodologies, and One Health approaches. We aimed to identify knowledge gaps where novel research could provide insights to inform control policy, for bTB and other zoonoses. The review included 532 articles. We found different levels of research attention across episystems, with a significant proportion of the literature focusing on the badger-cattle-TB episystem, with far less attention given to tropical multi-host episystems. We found a limited number of studies focusing on management solutions and their efficacy, with very few studies looking at modelling exit strategies. Only a small number of studies looked at the effect of human disturbances on the spread of bTB involving wildlife hosts. Most of the studies we reviewed focused on the effect of badger vaccination and culling on bTB dynamics with few looking at how roads, human perturbations and habitat change may affect wildlife movement and disease spread. Finally, we observed a lack of studies considering the effect of weather variables on bTB spread, which is particularly relevant when studying zoonoses under climate change scenarios. Significant technological and methodological advances have been applied to bTB episystems, providing explicit insights into its spread and maintenance across populations. We identified a prominent bias towards certain species and locations. Generating more high-quality empirical data on wildlife host distribution and abundance, high-resolution individual behaviours and greater use of mathematical models and simulations are key areas for future research. Integrating data sources across disciplines, and a "virtuous cycle" of well-designed empirical data collection linked with mathematical and simulation modelling could provide additional gains for policy-makers and managers, enabling optimised bTB management with broader insights for other zoonoses. Contagious bovine pleuropneumonia (CBPP) is an infectious and contagious bacterial respiratory disease that affects cattle with significant economic losses to the African animal industry. The use of ELISA kits based on monoclonal antibodies (mAbs) will aid in quick and precise diagnosis of CBPP, contributing to disease control and prevention in cattle. Thus, this research aims to develop and evaluate monoclonal antibodies against CBPP (T1/44) antigen for use in ELISA kits for CBPP diagnosis. Hybridoma technology was used to develop monoclonal antibodies that recognize and bind to the CBPP (T1/44) antigen. The antibody-secreting hybridomas were produced after immunizing mice with purified CBPP antigens. The hybridomas were screened for high sensitivity, specificity, and liking to the antigen. The selected mAbs were assessed for sensitivity and specificity against CBPP antigen using different immunoassays, dot-blot, ELISA, and mouse mAb isotyping. The monoclonal antibodies were profoundly specific, with a higher hindrance to CBPP antigen (<0.50 OD) while lacking cross-reactivity to other antigens. The monoclonal antibodies could distinguish CBPP antigen at low concentrations, showing their high sensitivity (>80% PI). The isotyped mAbs of intrigued appeared to have a place in the IgG class. These identified monoclonal antibodies can be utilized to develop an ELISA kit for CBPP diagnosis, which would give a fast, precise, and cost-effective strategy for screening and checking CBPP in cattle herds. Context: The energy availability for cow–calf systems is related to climate disturbances, and energy use is influenced by the age of the cows. Both factors determine the productivity and resilience of herds after climate disruption. Aims: Identify the time needed for the herds composed of cows of a varying age to achieve resilience after energy restriction in the pre- and postpartum. Methods: The resilience of systems was compared by deterministic dynamic simulation and herds were considered initially stable but different in (1) energy availability for all herd in the first year of production, namely at 50% (low, L), 75% (medium, M) of 100% (high, H) of the requirements according to the NRC (2000 , 2016) , with these energy levels being restricted to 60 days before and 60 days after the birth, and (2) the maximum age of the cull cow (lifetime, LT), namely 4 (LT4L, LT4M, LT4H), 6 (LT6L, LT6M, LT6H) of 11 (LT11L, LT11M, LT11H) years. From the second year, availability returned to meet the energy requirements of all animals. The availability of energy for the herd was simulated in natural and cultivated pasture. The productivity was determined as the relation between the kilograms of animals sold and the area used for production over a productive cycle. The system model was considered resilient when 95% of the standard productivity was reached (prior to energy restriction). Key results: The time required to return to stability was 2 years in the LT6 and LT11, whereas LT4 required 3 years. Furthermore, LT6 had a higher productivity than did the other herds. The older the culled cow was, the greater was the change in the composition of the product commercialised. Conclusions: Cows with an intermediate culling age allowed a sustainable intensification of the system, because they showed better productivity than did older cull cows. Furthermore, after climate disturbance, they presented more resilience than did younger cull cows. The level of energy restriction did not directly influence the time to resilience within each evaluated discard-age group. Implications: Herds that culled females before reaching adult age are less resilient after disturbances, such as changes in climate or changes of any nature that interfere with reproductive rates. The sufficient length of the cow's lifetime in the beef cattle herd allows to mitigate problems in the cow–calf systems generated by the reduction in food availability. The intermediate age (6 years) of culling the cows allowed better resilience of the herd, after restriction (50% or 75%) of energy during 60 days pre- and 60 days postpartum for the cows. Background and Aim: Bovine viral diarrhea (BVD) is endemic in North Africa and the Mediterranean Basin with high socioeconomic impacts. However, there are no data on this disease in Libya. One of the aims of this study was to provide data on BVD in Libya, to fill in the gap in the region and to investigate the level of seroprevalence of BVD virus (BVDV) in Libya and associated risk factors. Material and Methods: A total of 1599 serum samples were collected from cattle herds belonging to seven Libyan regions. All sera were assayed using a screening enzyme-linked immunosorbent assay for the detection of antibodies against BVDV. Results: The overall seroprevalence of BVDV was estimated to be 48.6% (95% confidence interval, 46.08%-50.98%). A seroprevalence rate of 36.8% was detected in cattle aged <1 year, 41.0% in cattle aged between 1 and 2 years, and 49.7% in cattle aged >2 years. Statistically significant differences (p = 0.001) were observed between age groups. BVDV seroprevalence was significantly associated with geographical region (p = 0.033). Conclusion: To the best of our knowledge, this is the first study on BVD in Libya, and the results suggest that BVD is endemic in Libya. Simple Summary: Beef cattle farming is an extremely important economic activity in Brazil, mainly in the northern region. Bovine hoof disorders impair the beef production chain; however, there are few studies on the subject, especially on the incidence of Bovine Dewclaw Dermatitis (BDCD). Thus, this is a pioneering investigative study to highlight the main factors predisposing animals to BDCD in beef cattle herds in the Amazon Biome. This study shows that risk factors related to climate, pasture conditions, and the physical structure of bovine management facilities are directly linked to BDCD onset in herds, affecting the beef production sector. Bovine Dewclaw Dermatitis (BDCD) is a hoof disease characterized by inflammation of the second and fifth accessory digits and the skin in this region. This pathology is poorly described in the literature; however, it has recently been observed in beef cattle in the Amazon Biome, Brazil. The objective of this study was to perform a clinical diagnosis and identify the risk factors associated with BDCD onset in cattle in the studied biome. Samples were collected from eight farms with extensive breeding systems located in Xinguara, Rondon do Pará, Curionópolis, and Ipixuna do Pará in the state of Pará, Brazil. A total of 706 Nellore and Nellore crossbred with taurine bovine of both sexes were evaluated, with males aged between 2 and 4 years and a mean weight of 650 kg, and females aged between 2 and 11 years and a mean weight of 400 kg. Distal extremities were inspected during cattle management, and in cases of dewclaw lesions, a specific examination was carried out after proper restraint. Cattle were diagnosed with BDCD on all farms analyzed. Of the 706 cattle inspected, 49 (6.94%) showed BDCD, of which 19 (38.77%) were Nellore and 30 (61.22%) were crossbred. This was the first study to determine BDCD's occurrence in extensive farming systems in the Amazon region, also showing that pastures with large amounts of stumps and stones, the physical structure of pens, and trauma and injury incidence during animal management are the most important predisposing factors for the onset of BDCD. It commonly affects lactating and recently weaned calves, but dairy cows can also be impacted resulting in significant losses to the milk production chain. This research aimed to investigate the serological status of dairy cows for the four main bovine respiratory viruses. This study took place in the central-eastern mesoregion of the state of Paraná, which has the highest milk productivity in Brazil. A total of 497 serum samples were assessed from unvaccinated dairy cows from 39 herds. This study aimed to determine the presence of antibodies to bovine alphaherpesvirus 1 (BoAHV1), bovine viral diarrhea virus (BVDV), bovine parainfluenza virus 3 (BPIV3), and bovine respiratory syncytial virus (BRSV) using virus neutralization tests. The frequencies of BoAHV1, BVDV, BPIV3, and BRSV seropositive cows and herds were 71.4, 56.3, 96.8, and 63.4%, and 79.5, 82.0, 100, and 84.6%, respectively. Rates of more than 50% of cows and herds seropositive for the 4 viruses indicate that these respiratory pathogens are endemic in dairy herds in the region evaluated. In this region for BRD prevention, we recommend implementing cow vaccination programs that provide passive immunity in calves and active immunity in cows. This study aims to determine the serological profile of high-yielding dairy cows for four main viruses (bovine alphaherpesvirus 1 (BoAHV1), bovine viral diarrhea virus (BVDV), bovine parainfluenza virus 3 (BPIV3), and bovine respiratory syncytial virus (BRSV)) related to bovine respiratory disease (BRD) in cattle herds worldwide. In this survey, 497 blood serum samples were collected from non-vaccinated dairy cows without clinical respiratory signs in 39 herds in the central-eastern mesoregion of Paraná State, South Brazil. The presence of neutralizing antibodies was determined by virus neutralization (VN) tests. VN antibodies against BoAHV1, BVDV, BPIV3, and BRSV were detected in 355 (71.4%), 280 (56.3%), 481 (96.8%), and 315 (63.4%) serum samples, respectively. The frequencies of seropositive herds for BoAHV1, BVDV, BPIV3, and BRSV were 79.5 (n = 31), 82.0 (n = 32), 100 (n = 39), and 84.6% (n = 33), respectively. The frequencies of seropositive cows varied according to the type of herd management and the number of cows in the herd. The detection of VN antibodies in unvaccinated dairy cattle herds demonstrated the endemic circulation of the four viruses in the herds evaluated. Background: Digital dermatitis (DD) is a contagious hoof infection affecting cattle worldwide. The disease causes lameness and a reduction in animal welfare, which ultimately leads to major decreases in milk production in dairy cattle. The disease is most likely of polymicrobial origin with Treponema phagedenis and other Treponema spp. playing a key role; however, the etiology is not fully understood. Diagnosis of the disease is based on visual assessment of the feet by trained hoof-trimmers and veterinarians, as a more reliable diagnostic method is lacking. The aim of this study was to evaluate the use of an enzyme-linked immunosorbent assay (ELISA) on bulk tank milk samples testing for the presence of T. phagedenis antibodies as a proxy to assess herd prevalence of DD in Swedish dairy cattle herds. Results: Bulk tank milk samples were collected in 2013 from 612 dairy herds spread across Sweden. A nationwide DD apparent prevalence of 11.9% (8.1–14.4% CI95%) was found, with the highest proportion of test-positive herds in the South Swedish regions (31.3%; 19.9–42.4% CI95%). Conclusions: This study reveals an underestimation of DD prevalence based on test results compared to hoof trimming data, highlighting the critical need for a reliable and accurate diagnostic method. Such a method is essential for disease monitoring and the development of effective control strategies. The novelty of ELISA-based diagnostic methods for DD, coupled with the disease's polymicrobial origin, suggests an avenue for improvement. Developing an expanded ELISA, incorporating antigens from various bacterial species implicated in the disease, could enhance diagnostic accuracy. The significance of this study is underscored by the extensive analysis of a substantial sample size (612). Despite its importance, few studies have analysed the disease's behaviour in Colombia, and none have developed maps using geographic information systems (GIS) to characterise it; as such, we developed this study to describe the temporal–spatial distribution of bovine tuberculosis in Colombia over a period of 19 years. Methods: A retrospective cross-sectional descriptive study, based on reports by the Colombian Agricultural Institute (ICA), surveillance of tuberculosis on cattle farms in Colombia from 2001 to 2019 was carried out. The data were converted into databases using Microsoft Access 365®, and multiple epidemiological maps were generated with the QGIS® version 3.36 software coupled to shape files of all the country's departments. Results: During the study period, 5273 bovine tuberculosis cases were identified in multiple different departments of Colombia (with a mean of 278 cases/year). Regarding its temporal distribution, the number of cases varied from a maximum of 903 cases (17.12% of the total) in 2015 to a minimum of 0 between 2001 and 2004 and between 2017 and 2019 (between 2005 and 2016, the minimum was 46 cases, 0.87%). Conclusions: GIS are essential for understanding the temporospatial behaviour of zoonotic diseases in Colombia, as is the case for bovine tuberculosis, with its potential implications for the Human and One Health approaches. Background: Bovine Genital Campylobacteriosis (BGC), a worldwide distributed venereal disease caused by Campylobacter fetus subsp. venerealis (Cfv), has a relevant negative economic impact in cattle herds. The control of BGC is hampered by the inexistence of globally available effective vaccines. The present in silico study aimed to develop a multi-epitope vaccine candidate against Cfv through reverse vaccinology. Results: The analysis of Cfv strain NCTC 10354 proteome allowed the identification of 9 proteins suitable for vaccine development. From these, an outer membrane protein, OmpA, and a flagellar protein, FliK, were selected for prediction of B-cell and T-cell epitopes. The top-ranked epitopes conservancy was assessed in 31 Cfv strains. The selected epitopes were integrated to form a multi-epitope fragment of 241 amino acids, which included 2 epitopes from OmpA and 13 epitopes from FliK linked by GPGPG linkers and connected to the cholera toxin subunit B by an EAAAK linker. The vaccine candidate was predicted to be antigenic, non-toxic, non-allergenic, and soluble upon overexpression. The protein structure was predicted and optimized, and the sequence was successfully cloned in silico into a plasmid vector. Additionally, immunological simulations demonstrated the vaccine candidate's ability to stimulate an immune response. Conclusions: This study developed a novel vaccine candidate suitable for further in vitro and in vivo experimental validation, which may become a useful tool for the control of BGC. Social interactions are fundamental properties of gregarious species, helping to establish dominance hierarchies and maintain social bonds within groups. Cattle, Bos taurus , are social ungulates which engage in affiliative and agonistic relationships with other individuals. Although there are approximately 1.5 billion cattle on the planet, the opportunity to research cattle behaviour in free-ranging groups is rare, as there are few feral populations worldwide. Cattle engage in positive social behaviours such as allogrooming, where one individual licks the body of another. The relationship between affiliative behaviours and other individual characteristics (such as sex and dominance status) are frequently studied in other gregarious species, but are largely undetermined in cattle. To investigate the relationships between sex, dominance status and allogrooming, we observed a mixed-sex feral cattle herd in Hong Kong, recording dominance interactions and allogrooming events. The hierarchy was linear and stable, consisting of 22 adult males and 33 adult females. We found that dominant females received more allogrooming than subordinate females, but subordinate females did not perform it more. Males performed allogrooming more towards females than other males, but females groomed both sexes equally. Sex affected dominance position, with males obtaining higher status than females, but not all females were subordinate to males. These preferential allogrooming patterns improve our knowledge of sex-specific interactions and help us to understand the dynamics of agonistic and affiliative behaviours in multimale, multifemale ungulate groups. This research examined the positivity ratio of Crimean–Congo haemorrhagic fever (CCHF) antibodies in cattle and sheep within Namibia's Omaheke region after a human disease outbreak in the same geographical area. A total of 200 samples (100 cattle and 100 sheep) were randomly collected from animals brought to two regional auction sites, and then tested using the ID Screen® CCHF Double Antigen Multi-Species Enzyme-Linked Immunosorbent Assay kit. Of the cattle samples, 36% tested positive, while 22% of the sheep samples were seropositive. The cattle had a significantly higher positivity ratio than sheep at the individual animal level (p = 0.0291). At the herd level, 62.5% of cattle herds and 45.5% of sheep flocks had at least one positive animal, but this difference was statistically insignificant (p = 0.2475). The fourteen cattle farms with at least one seropositive animal were dispersed across the Omaheke region. In contrast, the ten sheep farms with seropositive cases were predominantly situated in the southern half of the region. The study concluded that the CCHF is endemic in the Omaheke region and likely in most of Namibia, underscoring the importance of continued surveillance and preventive measures to mitigate the impact of CCHFV on animal health and potential spillover into human populations. Paratuberculosis, also known as Johne's disease, is a chronic wasting disease caused by Mycobacterium avium subs paratuberculosis (MAP) in ruminants. Paratuberculosis causes a significant reduction of milk production in the affected dairy sheep or goats and increase the cost of diagnosis, treatment, and culling of the infected animals. Paratuberculosis is currently recognised as a disease of major economic significance in cattle, sheep, goats, and wild ruminants globally. Recent reports also suggest that paratuberculosis affects wild ruminants and farmed deer. Despite the widespread occurrence of MAP, there are variations in the seroprevalence and global distribution patterns of disease among small ruminants and deer herds due to the influence of interacting epidemiological variables in different places. This systematic review aims to provide insights on the current global seroprevalence status and distribution pattern of paratuberculosis among small ruminants and deer herds. The review compiled, analyzed, and narratively synthesized 36 eligible research articles published between January 1, 2010, and January 31, 2024, from the SCOPUS and PubMed databases based on the 22-point Strengthening the Reporting of Observational Studies in Epidemiology (STROBE) checklist. The average global seroprevalence of paratuberculosis in sheep was 14.02% (0.7-66.8), with the highest rate in Canada (66.8%) and the lowest in Austria (0.7%). Comparatively, the average global seroprevalence in goats was 18.44% (0.3-83), with the highest rate in Canada (83%) and the lowest in the West Indies (0.3%). The average global prevalence of paratuberculosis in deer was 14.76% (3.7-30.2), with the highest rate in Spain (30.2%) and the lowest rate in the Czech Republic (3.7%). This review revealed that Canada is a hot spot for both caprine and ovine paratuberculosis and there was a higher global seroprevalence rates in goats than sheep and deer. The lack of data on the seroepidemiology of paratuberculosis among small ruminant stock in Southeast Asia and other regions is a gap in our current knowledge of its distribution. Therefore, seroprevalence surveys of paratuberculosis among the small ruminant and deer livestock are required to furnish information for planning suitable interventions in these areas. Simple Summary: Paratuberculosis impacts animal welfare and the economic performance of dairy herds by causing reduced milk yield and carcass weight and premature culling. Due to the ability of the infectious agent Mycobacterium avium subsp. paratuberculosis (MAP) to survive in the environment for a long time, the limited diagnostic accuracy of tests, and the long incubation period, successful elimination of MAP from a cattle herd is doubted. This study describes measures applied to control the disease in a 450-head dairy herd and, as the result of these measures, the progress of prevalence reduction to a level where the infectious agent was not detectable anymore in any individual sample and from the liquid manure of the herd. MAP shedders were detected by the bacteriological cultivation of individual faecal samples from each cow in annual intervals and MAP shedders were removed from the herd in a timely manner. Calves were kept outside the barn of adult cows and hygienic measures to minimize their MAP exposure were established. The results demonstrate that the elimination of MAP might be possible within 10 years. Voluntary control programs are effective in controlling paratuberculosis in closed herds by providing adequate diagnostic, logistic, and financial support for farmers. This longitudinal case study provides an in-detail report of the process towards the elimination of Mycobacterium avium subsp. paratuberculosis (MAP) from a closed 450-head commercial dairy herd. In parallel, two diagnostic approaches were applied to all cows in annual intervals during 2012–2022: detection of MAP in individual faecal samples by bacteriological cultivation on solid medium and detection of MAP-specific antibodies by ELISA. For each annual sampling, the kappa coefficients for test agreement and the survival rates of MAP-positive and MAP-negative cows were calculated. Applying a multivariable linear regression model revealed a significantly lower fat-corrected 305-day milk yield for MAP-positive cows. The true prevalence of MAP shedders reduced from 24.2% in 2012 to 0.4% in 2019 and during 2020–2022, no MAP shedder was identified. Test agreement was generally low and bacteriological cultivation showed positive results earlier than the ELISA. In the first years of control, the survival of MAP shedders was longer than in the final stage. In conclusion, the elimination of MAP from a dairy herd might be feasible within a decade. Simple Summary: Johne's disease, caused by Mycobacterium avium subsp. paratuberculosis (MAP), is endemic in cattle herds in the UK and throughout the world. It is a wasting disease with a protracted incubation period, during which clinical signs are not apparent, and is responsible for considerable economic losses. Infection can be transmitted silently from animal to animal, and MAP can find its way into the food chain through the products of infected animals. Ante-mortem diagnosis of the disease is normally achieved through detection of the organism (by faecal culture or PCR) or serological tests, but agreement as to a positive diagnosis is often poor and the certainty of identifying all positive animals is low, particularly in early-stage infections. Mycobacterial cells contain high-molecular-weight lipids, mycolic acids (MAs), which are recognised by antibodies in active infections. Here, we show that a simple assay using different classes of single synthetic MAs can detect antibodies in serum from cattle with MAP infections and distinguish uninfected animals from animals positive both to faecal culture and a commercial serodiagnostic assay. It does so without interference from vaccination either for MAP (a Gudair vaccination) or for bovine TB (BCG vaccination). When the assay is optimised to distinguish negative samples from those positive only by faecal PCR, it identifies additional positives that are not seen using a commercial serodiagnostic assay; this suggests that, with further development, it may provide a method for detecting infections before the commercial serodiagnostic assay can. The problem: Ante-mortem diagnosis of Johne's disease, caused by Mycobacterium avium subsp. paratuberculosis (MAP), is normally achieved through faecal culture, PCR, or serological tests, but agreement as to which samples are positive for Johne's disease is often poor and sensitivities are low, particularly in early-stage infections. The potential solution: Mycobacterial cells contain very complex characteristic mixtures of mycolic acid derivatives that elicit antibodies during infection; this has been used to detect infections in humans. Here, we explore its application in providing an assay differentiating infected from vaccinated animals (DIVA assay) for Johne's disease in cattle. Method: Antibody responses to different classes of mycolic acid derivatives were measured using ELISA for serum from cattle positive for MAP by both faecal PCR and commercial serum ELISA, or just by PCR, and from animals from herds with no history of Johne's disease, bovine tuberculosis reactors, BCG-vaccinated, BCG-vaccinated and M. bovis-infected, and Gudair-vaccinated animals. Results: The best-performing antigens, ZAM295 and ST123—the latter a molecule present in the cells of MAP but not of Mycobacterium bovis—achieved a sensitivity of 75% and 62.5%, respectively, for serum from animals positive by both faecal PCR and a commercial MAP serum ELISA, at a specificity of 94% compared to 80 no-history negatives. Combining the results of separate assays with two antigens (ST123 and JRRR121) increased the sensitivity/specificity to 75/97.5%. At the same cut-offs, animals vaccinated with Gudair or BCG vaccines and bTB reactors showed a similar specificity. The specificity in BCG-vaccinated but M. bovis-infected animals dropped to 85%. Combining the results of two antigens gave a sensitivity/specificity of 37.5/97.5% for the full set of 80 PCR-positive samples, detecting 30 positives compared 16 for IDEXX. Conclusion: Serum ELISA using synthetic lipids distinguishes effectively between MAP-negative cattle samples and those positive by both PCR and a commercial MAP serodiagnostic, without interference by Gudair or BCG vaccination. Background: Contagious bovine pleuropneumonia [CBPP] is a transboundary animal disease of cattle caused by Mycoplasma mycoides subsp. mycoides [Mmm]. CBPP causes severe economic losses to livestock producers in sub-Saharan Africa mainly due to high mortality, morbidity, reduction in productivity as well as livestock trade restrictions. This study aimed at determining seroprevalence of Mmm in cattle from Karamoja region, north-eastern Uganda; data that are required to design and implement risk based CBPP control program. Methods: We randomly collected blood samples from 2,300 cattle spread across Karamoja region. Serum was extracted and screened for antibodies against Mycoplasma mycoides subsp. mycoides [Mmm] using the competitive enzyme linked immunosorbent assay [cELISA]. Results: A quarter [25.4%; 95% CI: 23.7–27.3] of the screened cattle [n = 2,300] were sero-positive for Mmm. Amudat and Kaabong districts recorded the lowest [12.3%] and highest [30.7%] Mmm seroprevalence respectively. Increasing age, overnight stay in cattle kraals and location [certain districts, villages, herds and sub counties] of the cattle herds, the factors that promote animal commingling, were the most significant risk factors of seroconversion with Mmm. Conclusion: Results from this study indicated a higher seroprevalence of Mmm in Karamoja region cattle herds. This could be due to the increased frequency of CBPP outbreaks in recent years. To be effective, CBPP vaccination programs should target high risk herds along the international borders and other hotspot areas [e.g., parishes or sub counties] where cattle commingling is high. Introduction: Thailand experienced a nationwide outbreak of lumpy skin disease (LSD) in 2021, highlighting the need for effective prevention and control strategies. This study aimed to identify herd-level risk factors associated with LSD outbreaks in beef cattle herds across different regions of Thailand. Methods: A case--control study was conducted in upper northeastern, northeastern, and central regions, where face-to-face interviews were conducted with farmers using a semi-structured questionnaire. Univariable and multivariable mixed effect logistic regression analyses were employed to determine the factors associated with LSD outbreaks. A total of 489 beef herds, including 161 LSD outbreak herds and 328 non-LSD herds, were investigated. Results and discussion: Results showed that 66% of farmers have operated beef herds for more than five years. There were very few animal movements during the outbreak period. None of the cattle had been vaccinated with LSD vaccines. Insects that have the potential to act as vectors for LSD were observed in all herds. Thirty-four percent of farmers have implemented insect control measures. The final mixed effect logistic regression model identified herds operating for more than five years (odds ratio [OR]: 1.62, 95% confidence interval [CI]: 1.04--2.53) and the absence of insect control management on the herd (OR: 2.05, 95% CI: 1.29--3.25) to be associated with LSD outbreaks. The implementation of insectvector control measures in areas at risk of LSD, especially for herds without vaccination against the disease, should be emphasized. This study provides the first report on risk factors for LSD outbreaks in naïve cattle herds in Thailand and offers useful information for the development of LSD prevention and control programs within the country's context. Although several brands of tuberculin purified protein derivatives (PPDs) are available for diagnosing bovine tuberculosis (bTB), comparative studies to determine their diagnostic accuracy are infrequent. In Ecuador we compared two different PPD brands for bTB diagnosis using skin testing and measuring skin thickness increase. Additionally, we evaluated four PPD brands, including those used for skin testing, in the Bovine Tuberculosis Interferon Gamma Test (IFN-γ test) measuring IFN-γ induction in whole blood. The study included 17 naturally tuberculosis-infected PPD and IFN-γ test positive bovines. Both the field and laboratory results showed significant differences in classifying the 17 bovines as bTB positive or negative. We hypothesize that several factors, such as the genetic background of the cows, sensitization to environmental mycobacteria, M. bovis strains involved in the bTB infection, and the manufacturing procedures of the PPDs, could have influenced the immune reaction toward the different tuberculin PPD brands. Our study emphasizes the necessity for comparative studies aimed at determining the diagnostic accuracy of PPD brands for bTB diagnosis as well as the development of standardized methods for PPD production and potency determination. Herding cattle across the landscape requires three species – horse, cattle, and human – to move together in a goal-orientated, albeit human-centered, activity. In this multispecies activity, they must synchronize through embodied communication and develop a shared understanding of moving conjointly. Together, all three species are socialized, or "zoocialized," to learn to engage in a shared community of communication where they develop a sense of "timing" and "feel" of the others to enable their directed movement together. While not denying or downplaying the power and pain integral to cattle ranching, we explore and interpret the interspecies, multispecies communication, collaboration, and choreography. Based on a multispecies ethnographic methodology, we draw on experiences from cattle herding in the USA, Canada, and Sweden. What emerges are intricate relations of agency, shaped by meanings of species, where human and nonhuman social actors learn to meet and construct a vibrant multispecies community of communication. Optimising antimicrobial usage (AMU) in livestock is pivotal to counteract the emergence of antimicrobial resistance. We analysed AMU in more than 1000 cattle herds over 11 years (2008–2018) in the Aosta Valley (Italy), a region where 80% of farms house less than 50 cattle. Dairy cows accounted for over 95% of AMU. AMU was estimated using the defined daily dose animal for Italy (DDDAit) per biomass for the whole herd and a treatment incidence 100 (TI100) for cows. Average annual herd-level AMU was low, with 3.6 DDDAit/biomass (range: 3.2–4.0) and 1.2 TI100 in cows (range: 1.1–1.3). Third and fourth generation cephalosporins, which are critical for human medicine, represented almost 10% of usage, and intramammary antimicrobials accounted for over 60%. We detected significant downward temporal trends in total AMU, as well as a positive relationship with herd size. The magnitude of such effects was small, leaving scant room for further reduction. However, the frequent use of critical antimicrobials and intramammary products should be addressed, following the principles of prudent AMU. Our findings highlight the importance of monitoring AMU even in low-production, smallholding contexts where a low usage is expected, to identify any deficiencies and implement interventions for further AMU optimisation. Bovine babesiosis has substantial economic implications in the cattle industry, emphasizing the need for a thorough understanding of the genetic diversity of the causative apicomplexan pathogen. Although babesiosis has been extensively studied globally, the genetic diversity of Babesia species in Malaysian and Nigerian cattle remains unreported. This study aims to bridge this gap by detecting and characterizing Babesia species in selected cattle herds. Our investigation explores the genetic diversity of Babesia species in cattle from Selangor, Malaysia, and Ribah, Nigeria. Blood samples revealed a 32.9% infection rate via PCR analysis. Further genetic analysis detected variations in Malaysian Babesia bigemina isolates but genetic similarity among Nigerian isolates. Conversely, all Babesia bovis isolates displayed genetic homogeneity. Persistent tuberculosis (TB) in cattle populations in England has been associated with an exchange of infection with badgers (Meles meles). A badger control policy (BCP) commenced in 2013. Its aim was to decrease TB incidence in cattle by reducing the badger population available to provide a wildlife reservoir for bovine TB. Monitoring data from 52 BCP intervention areas 200–1600 km2 in size, starting over several years, were used to estimate the change in TB incidence rate in cattle herds, which was associated with time since the start of the BCP in each area. A difference in differences analysis addressed the non-random selection and starting sequence of the areas. The herd incidence rate of TB reduced by 56% (95% Confidence Interval 41–69%) up to the fourth year of BCP interventions, with the largest drops in the second and third years. There was insufficient evidence to judge whether the incidence rate reduced further beyond 4 years. These estimates are the most precise for the timing of declines in cattle TB associated with interventions primarily targeting badgers. They are within the range of previous estimates from England and Ireland. This analysis indicates the importance of reducing transmission from badgers to reduce the incidence of TB in cattle, noting that vaccination of badgers, fertility control and on farm biosecurity may also achieve this effect. Background: Bovine tuberculosis (bTB) is a chronic disease that results from infection with any member of the Mycobacterium tuberculosis complex. Infected animals are typically diagnosed with tuberculin-based intradermal skin tests according to World Organization of Animal Health which are presently in use. However, tuberculin is not suitable for use in BCG-vaccinated animals due to a high rate of false-positive reactions. Peptide-based defined skin test (DST) antigens have been identified using antigens (ESAT-6, CFP-10 and Rv3615c) which are absent from BCG, but their performance in buffaloes remains unknown. To assess the comparative performance of DST with the tuberculin-based single intradermal test (SIT) and the single intradermal comparative cervical test (SICCT), we screened 543 female buffaloes from 49 organized dairy farms in two districts of Haryana state in India. Results: We found that 37 (7%), 4 (1%) and 18 (3%) buffaloes were reactors with the SIT, SICCT and DST tests, respectively. Of the 37 SIT reactors, four were positive with SICCT and 12 were positive with the DST. The results show that none of the animals tested positive with all three tests, and 6 DST positive animals were SIT negative. Together, a total of 43 animals were reactors with SIT, DST, or both, and the two assays showed moderate agreement (Cohen's Kappa 0.41; 95% Confidence Interval (CI): 0.23, 0.59). In contrast, only slight agreement (Cohen's Kappa 0.18; 95% CI: 0.02, 0.34) was observed between SIT and SICCT. Using a Bayesian latent class model, we estimated test specificities of 96.5% (95% CI, 92–99%), 99.7% (95% CI: 98–100%) and 99.0% (95% CI: 97–100%) for SIT, SICCT and DST, respectively, but considerably lower sensitivities of 58% (95% CI: 35–87%), 9% (95% CI: 3–21%), and 34% (95% CI: 18–55%) albeit with broad and overlapping credible intervals. Conclusion: Taken together, our investigation suggests that DST has a test specificity comparable with SICCT, and sensitivity intermediate between SIT and SICCT for the identification of buffaloes suspected of tuberculosis. Our study highlights an urgent need for future well-powered trials with detailed necropsy, with immunological and microbiological profiling of reactor and non-reactor animals to better define the underlying factors for the large observed discrepancies in assay performance, particularly between SIT and SICCT. Background and Aim: Bovine anaplasmosis (BA) is one of the most important diseases of ruminants worldwide, causing significant economic losses in the livestock industry due to the high morbidity and mortality in susceptible cattle herds. Anaplasma marginale is the main causative agent of BA occurring worldwide in tropical and subtropical regions. This study aimed to investigate the first molecular detection and genetic diversity of A. marginale in dairy cattle in Khon Kaen Province, Thailand. Materials and Methods: Blood samples were collected from 385 lactating cows from 40 dairy farms in five districts of Khon Kaen, regardless of age and health status. To detect A. marginale, all DNA preparations were used for molecular diagnosis using a single polymerase chain reaction with the msp4 gene target. A phylogenetic tree was constructed from the msp4 gene sequences using molecular genetic characterization. Genetic diversity was calculated as haplotype diversity, haplotype number, number of nucleotide differences, nucleotide diversity, and average number of nucleotide differences. Results: The overall prevalence of A. marginale was 12.72% (49/385). The highest prevalence (17.19%) was found in Ubolratana district, followed by Muang, Kranuan, Khao Suan Kwang, and Nam Phong districts (14.94%, 14.74%, 13.79%, and 3.70%, respectively). Phylogenetic analysis showed that A. marginale was closely related to isolates from Australia (98.96%), China (99.68%), Spain (99.74%), and the USA (99.63%). Conclusion: The molecular prevalence of BA in dairy cattle is the first to be observed in this area, and the genetic variability with separated clusters shown in the msp4 gene of A. marginale revealed species variation in dairy cattle. This significant genetic diversity contributes to the understanding of the diversity of A. marginale and will be important for the control and prevention of A. marginale in dairy cattle. This paper considers the role of social research and human behaviour in attempts to eradicate bTB. Future attempts to eradicate bTB are likely to involve an increasing range of sophisticated technologies. However, the acceptance and use of these technologies is likely to depend on a range of behavioural incentives. The use of appropriate behavioural nudges may facilitate bTB eradication, but the paper contends that of more value are socio-cultural approaches to understanding behaviour. Specifically, the concepts of the 'good farmer' and 'real vets' are discussed to show how bTB eradication is dependent on social identities. This study investigated the impact of parity, period of calving, and season of calving on various production and reproduction traits in Hardhenu cattle. Third‐parity cows outperformed first and second‐parity cows, exhibiting significantly higher total milk yield (TMY), 305 days milk yield (305d MY), peak yield (PY), and artificial insemination per conception (AIPC) (p <.01). The Lactation length (LL) was inversely related to parity, with first‐parity cows having extended lactations (p <.05). The period of calving had a substantial influence on production traits. The season of calving was critical, affecting production traits differently across parities. First‐parity cows had the highest TMY and 305d MY during winter and summer calving, with summer calving associated with shorter LL (p <.05). Second‐parity cows showed higher TMY and 305d MY during summer calving, while third‐parity cows had higher TMY in winter (p <.01). PY followed similar patterns to TMY, with LL affected by the season of calving. Additionally, the study explored interactions between parity, period of calving, and season of calving on reproduction traits. Third‐parity cows had higher AIPC compared to first and second‐parity cows (p <.01). Reproduction traits were also significantly affected by the period of calving, with cows calving in the 2017–2021 period displaying longer service period (SP), calving interval (CI), dry period (DP), and AIPC (p <.01). These findings highlight the complex interplay of biological, environmental, and management factors in shaping the productivity and reproductive efficiency of Hardhenu cattle, emphasizing the need for multifaceted considerations to optimize herd performance. Bovine tuberculosis and paratuberculosis are endemic in many areas worldwide. This work aims to study cytokines production and gene expression profiles of bovine macrophages infected with Mycobacterium bovis and Mycobacterium paratuberculosis subsp. avium (MAP) strains to identify potential diagnostic biomarkers. Bovine bone marrow stem cells were differentiated into macrophages and subsequently infected in vitro with different spoligotypes of M. bovis and MAP field strains (as single infections and coinfections), using different multiplicity of infection. Supernatant and cell pellets were collected 24 h, 48 h, and one week post-infection. Preliminarily, gene expression on cell pellets of IL-1β, IL-2, INFγ, IL-6, IL-10, IL-12, and TNFα was assessed by qRT-PCR one week p.i. Subsequently, IL-1β and IL-6 were measured by ELISA and qRT-PCR to investigated their production retrospectively 24 h and 48 h p.i. A variability in macrophages response related to the concentration of mycobacteria, the coinfection with MAP, and M. bovis spoligotypes was identified. An early and constant IL-6 increase was observed in the M. bovis infection. A lower increase in IL-1β was also detected at the highest concentration of the two M. bovis spoligotypes one week post-infection. IL-6 and IL-1 β production was reduced and differently expressed in the MAP infection. IL-6 appeared to be the earliest cytokines produced by bovine macrophages infected with M. bovis.
