The oxidation of 5-hydroxymethylfurfural (HMF) to 2,5-diformylfuran (DFF) using oxygen (1 atm) with a TEMPO and CuCl catalyst system is investigated using a range of imidazolium-based ionic liquids (ILs) and various bases at different HMF substrate loadings (10-50%). This represents the first example of HMF to DFF conversion in ionic liquid media under homogeneous catalysis conditions, revealing dramatic differences in performance between the ILs. In the non-coordinating, hydrophobic ionic liquid, [bmim][NTf2], 90% DFF yield is obtained at 5 mol% catalyst loading after 6 hours at 80 °C at a very high 40% HMF loading. Increasing the temperature to 100 °C leads to a lower yield, attributed to loss of volatile TEMPO from the reaction medium. A system using TEMPO and pyridine immobilized within the ionic liquid [bmim][NTf2] results in selective conversion of HMF to high purity DFF. It also allows the DFF formed to be isolated by sublimation in 81% yield before a further cycle is performed. Subsequent catalyst deactivation is probed by X-ray photoelectron spectroscopy (XPS). Synthesis from fructose in a two-step process achieves a 55% isolated DFF yield. This approach overcomes significant drawbacks previously reported for this transformation, such as solvent toxicity, separation and purification problems as well as the need for high oxygen pressures. Further oxidation of HMF with this system leads to a 62% yield of 5-formyl-2-furancarboxylic acid (FFCA). The separation of this compound can be achieved by sublimation of DFF followed by solvent extraction.

Homogeneous Co/Mn/Br catalyzed aerobic oxidation of benzyl alcohol in acetic acid to benzaldehyde was performed in polytetrafluoroethylene microreactors operated under slug flow at temperatures up to 150°C and pressures up to 5 bar. Depending on the bubble velocity and length, a wetted or dewetted slug flow was observed, characterized typically by a complete or partially wetting liquid film around the bubble body. The latter flow suffered from a limited interfacial area for mass transfer. Experiments at temperatures up to ca. 90°C were under kinetic control given no product yield difference under wetted and dewetted slug flows and were used to establish a simplified kinetic expression (first order in benzyl alcohol and zero order in oxygen). This allows to develop a mass transfer model combined with an instantaneous reaction regime that well described the experimental results at higher temperatures where mass transfer was limiting in the dewetted slug flow.

Catalytic glycerol conversion by means of either photon or thermal energy is of great importance and can be realized by metal supported on TiO2 systems. Although various procedures have been employed to synthesize efficient metal/TiO2 catalysts, the promotional mechanisms for both reactions are still unclear due to the lack of well-defined systems. Here, we have deposited gold nanoparticles on a series of highly crystalline anatase TiO2 substrates with different crystallite sizes (7, 12, 16, 28 nm) by both direct precipitation and sol-immobilization methods to examine the effect of metal deposition methods and TiO2 sizes on both photo- and thermal catalytic glycerol reforming. For photocatalytic H2 evolution from glycerol, optimum performance was observed for the Au supported on 12 nm TiO2 for both deposition methods. For thermal catalytic glycerol oxidation, all catalysts show a similar selectivity to glycerate (>70%) regardless of the TiO2 size and metal deposition method; however, the metal deposition method significantly influences the catalytic activity. In situ UV-vis spectrometry reveals that the optimized photocatalytic performance originates from enhanced charge transfer kinetics and a more negative Fermi level for proton reduction, whereas electrochemical analysis reveals that the promoted glycerol oxidation is caused by the enhanced oxygen reduction half-reaction.

The newly designed electrically neutral complexes [Ru(acac)(2)(Q)] (1-3) involving redox-active polycyclic aromatic hydrocarbon (PAH) derived quinonoids (Q): Q(1)((O,O)) (1) Q(2)((O,NH)) (2) Q(2)((NH,NH)) (3) (acac = acet- ylacetonate) were prepared from the metal precursor [Ru-II(acac)(2)(CH3CN)(2)] and preformed pyrene-4,5-dione (Q(1)) and partially deprotonated pyrene-4,5-diamine (H(4)Q(2)), respectively. The structural characterization of 1-3 established their molecular identities including intermolecular pi-pi stacking interactions between the extended pi-system of pyrene in the adjacent molecules and the hydrogen bonded 1D-polymeric form of 3. The redox sensitive C-O and C-N bond distances of Q in 1, 2 and 3 revealed the dominating ground state electronic forms of [(acac)(2)Ru-III-Q(1)((O,O))(center dot-)] (S = 0), [(acac)(2)Ru-III-Q(2)((O,NH)center dot-)] (S = 0) and [(acac)(2)Ru(II)Q(2)((NH,NH)o)] (S = 0), respectively, where strong antiferromagnetic coupling between Ru-III(t(2g)(5)) and Q(center dot-) resulted in S = 0 state in 1 or 2. Complexes 1-3 exhibited reversible single oxidation and reduction within the potential window of +/- 1.5 V versus SCE in CH3CN, which progressively shifted to the negative potential on moving from 1 to 2 to 3, primarily due to the difference in electronegativity between O and N donors of Q. The collective consideration of experimental (EPR, electronic spectra) and theoretical (DFT, TD-DFT) results of 1(n)-3(n) (n = +1, 0, -1) revealed (i) extensive mixing of metal-ligand orbitals due to the inherent covalency factor and (ii) Q(center dot-) and Ru-II based oxidations of 1/2 and 3, respectively, led to the {Ru-III-Q(o)} electronic form at the metal-ligand interface of the oxidized state (1(-).3(-)), while the reduced state (1(-)-3(-)) could best be described by the resonating form of {Ru-II-Q(center dot-)} {Ru-III-Q(2-)}.

Molecular oxygen is without doubt the greenest oxidant for redox reactions, yet aerobic oxidation is one of the most challenging to perform with good chemoselectivity, particularly on an industrial scale. This collaborative review (between teams of chemists and chemical engineers) describes the current scientific and operational hurdles that prevent the utilisation of aerobic oxidation reactions for the production of speciality chemicals and active pharmaceutical ingredients (APIs). The safety aspects of these reactions are discussed, followed by an overview of (continuous flow) reactors suitable for aerobic oxidation reactions that can be applied on scale. Some examples of how these reactions are currently performed in the industrial laboratory (in batch and in flow) are presented, with particular focus on the scale-up strategy. In recent years, several research groups have focused their attention on the development of synthetic and functional models of biological systems based on copper. Here we report the synthesis strategy, characterization, and catalytic activity for new copper-based porphyrins (CuP2 and CuP3) isolated by metallation of the corresponding free base derivatives, H2P2 and H2P3. The solid CuP2S was obtained from the reaction of H2P2 with copper(II) acetate (10 equiv.). These complexes have been used in homogeneous (CuP2 and CuP3) and heterogeneous (CuP2S) oxidative catalysis of 3,5-di-tert-butylcatechol and catechol, in the presence of air, with or without hydrogen peroxide. The obtained results show that the new copper porphyrins CuP2 and CuP3 and the CuP2S material are able to efficiently mimic the activity of catechol oxidase, the latter being easily reused and maintaining its activity for more than three catalytic cycles.

Prediction of oxidative drug metabolism at the early stages of drug discovery and development requires fast and accurate analytical techniques to mimic the in vivo oxidation reactions by cytochrome P450s (CYP). Direct electrochemical oxidation combined with mass spectrometry, although limited to the oxidation reactions initiated by charge transfer, has shown promise in the mimicry of certain CYP-mediated metabolic reactions. The electrochemical approach may further be utilized in an automated manner in microfluidics devices facilitating fast screening of oxidative drug metabolism. A wide range of in vivo oxidation reactions, particularly those initiated by hydrogen atom transfer, can be imitated through the electrochemically-assisted Fenton reaction. This reaction is based on O-O bond activation in hydrogen peroxide and oxidation by hydroxyl radicals, wherein electrochemistry is used for the reduction of molecular oxygen to hydrogen peroxide, as well as the reduction of Fe(3+) to Fe(2+). Metalloporphyrins, as surrogates for the prosthetic group in CYP, utilizing metallo-oxo reactive species, can also be used in combination with electrochemistry. Electrochemical reduction of metalloporphyrins in solution or immobilized on the electrode surface activates molecular oxygen in a manner analogous to the catalytical cycle of CYP and different metalloporphyrins can mimic selective oxidation reactions. Chemoselective, stereoselective, and regioselective oxidation reactions may be mimicked using electrodes that have been modified with immobilized enzymes, especially CYP itself. This review summarizes the recent attempts in utilizing electrochemistry as a versatile analytical and preparative technique in the mimicry of oxidative drug metabolism by CYP.

A variety of aromatic, aliphatic and conjugated alcohols were transformed into the corresponding carboxylic acids and ketones with aq 70% t-BuOOH in the presence of catalytic amounts of bismuth(III) oxide. This method possesses a wide range of capabilities, does not involve cumbersome work-up, exhibits chemoselectivity and proceeds under ambient conditions. We suggest that the reactivity of Au nanocatalysts can be greatly increased by doping the oxide substrate on which they are placed with an electron donor. To demonstrate this, we perform density functional theory calculations on a model system consisting of a 20-atom gold cluster placed on a MgO substrate doped with Al atoms. We show that not only does such substrate doping switch the morphology of the nanoparticles from the three-dimensional tetrahedral form to the two-dimensional planar form, but it also significantly lowers the barrier for oxygen dissociation by an amount proportional to the dopant concentration. At a doping level of 2.78%, the dissociation barrier is reduced by more than half, which corresponds to a speeding up of the oxygen dissociation rate by five orders of magnitude at room temperature. This arises from a lowering in energy of the s and p states of Au. The d states are also lowered in energy, however, this by itself would have tended to reduce reactivity. We propose that a suitable measure of the reactivity of Au nanoparticles is the difference in energy of sp and d states.

The syntheses and spectroscopic characterization of two 1,2,4-triazole-based oxovanadium(V) complexes are reported: 1(-)[VO(2)L1](-) and 2 [(VOL2)(2)(OMe)(2)] (where H(2)L1 = 3-(2'-hydroxyphenyl)-5-(pyridin-2"-yl)-H-1-1,2,4-triazole, H3L2 = bis-3,5-(2'-hydroxyphenyl)-1H-1,2,4-triazole). The ligand environment (N,N,O vs O,N,O) is found to have a profound influence on the properties and reactivity of the complexes formed. The presence of the triazolato ligand allows for pH tuning of the spectroscopic and electrochemical properties, as well as the interaction and stability of the complexes in the presence of hydrogen peroxide. The vanadium(IV) oxidation states were generated electrochemically and characterized by UV-vis and EPR spectroscopies, For 2, under acidic conditions, rapid exchange of the methoxide ligands with solvent [in particular, in the vanadium(IV) redox state] was observed.

In this paper the oxidation of milled wood lignin (MWL), catalysed by three enzymes, i.e. laccase, tyrosinase and horseradish peroxidase (HRP) was studied. The oxidation was followed by measuring the consumption of O2 during laccase and tyrosinase treatment and of H2O2 during HRP treatment. Both laccase and HRP were found to oxidise lignin effectively, whereas the effect of tyrosinase was negligible. The changes in MWL molecular-weight distributions caused in the reactions were analysed by gel permeation chromatography. Both laccase and HRP treatments were found to polymerise MWL. Peroxidase treatment was found to decrease the amount of phenolic hydroxyls in MWL, whereas no such effect could be detected in the laccase-treated sample. Both laccase and HRP treatments were, however, found to increase the amount of conjugated structures in MWL. The formation of phenoxy radicals during the treatments was studied by electron paramagnetic resonance spectroscopy. Phenoxy radicals were detected in both laccase and HRP-treated samples. The amount of the formed phenoxy radicals was found to be essentially constant during the detected time (i.e. 20–120 min after the addition of enzyme).

The reaction of the multidentate ligand N-(2-(aryldiazenyl)phenyl)picolinamide, HL (3a, 3b and 3c) [HL is ArN = NC6H4NHC(O)Py, where Ar is C6H5 for HL1 (3a), p-MeC6H4 for HL2 (3b) or p-ClC6H4 for HL3 (3c), and Py stands for pyridyl], with Cu(CH3COO)(2)center dot H2O and Na2PdCl4 separately afforded the complexes [(L)(2)Cu] (4a, 4b and 4c) and [(L)PdCl] (5a, 5b and 5c) respectively, where the deprotonated ligand L- binds copper(II) and palladium(II) in a tridentate (N,N,N) fashion. X-ray structures of [(L-3)(2)Cu] (4c) and [(L-1)PdCl] (5a) were determined to confirm the molecular structures. The newly synthesized complex 4a exhibits catalytic activity toward the oxidation of alcohols to the corresponding carbonyl compounds and oxidation of organic thioethers to sulfoxide and sulfone, whereas the complex 5a is an active catalyst for Suzuki and Heck reactions. The role of manganese (Mn) oxides in catalyzing the abiotic oxidation of polynuclear aromatic compounds (PNAs) has been investigated by monitoring the oxidation of dihydrodiol and dione PNAs and corresponding reduction of Mn in an aqueous environment. 1,4-, 1,3-, and 2,3-naphthalenediol, 1,4-naphthoquinone, and l,4-dihydroxy-9,10-anthracenedione (Quinizarin) were oxidized in an oxic, aqueous environment, where each of these organic compounds is soluble in water, except Quinizarin. A mathematical model is presented, which describes the redox reactions, and suggests that electron transfer/organic release from the oxide surface is rate limiting with the reaction apparently proceeding through a free-radical formation. Results indicate significant oxidation of the organics, that the oxidation of the diols is kinetically more favorable than that of the dione, and that lower molecular weight PNAs containing diol and dione functional groups are susceptible to abiotic-catalyzed oxidation, thereby suggesting that progressively more complex PNA molecules may also be susceptible to oxidation.

Enveloped animal viruses, such as Semliki Forest virus (SFV), utilize a membrane fusion strategy to deposit their genome into the cytosol of the host cell. SFV enters cells through receptor-mediated endocytosis, fusion of the viral envelope occurring subsequently from within acidic endosomes. Fusion of SFV has been demonstrated before to be strictly dependent on the presence of cholesterol in the target membrane. Here, utilizing a variety of membrane fusion assays, including an on-line fluorescence assay involving pyrene-labeled virus, we demonstrate that low-pa-induced fusion of SFV with cholesterol-containing liposomal model membranes requires the presence of sphingomyelin or other sphingolipids in the target membrane. The minimal molecular characteristics essential for supporting SFV fusion are encompassed by a ceramide. The action of the sphingolipids is confined to the actual fusion event, cholesterol being necessary and sufficient for low-pH-dependent binding of the virus to target membranes. Complex formation of the sphingolipids with cholesterol is unlikely to be important for the induction of SFV-liposome fusion, as sphingolipids that do not interact appreciably with cholesterol, such as galactosylceramide, effectively support the process. The remarkably low levels of sphingomyelin required for half-maximal fusion (1-2 mole%) suggest that sphingolipids do not play a structural role in the SFV fusion process, but rather act as a cofactor, possibly activating the viral fusion protein in a specific manner.

Wilson disease (WD) is a rare hereditary condition that is caused by a genetic defect in the copper-transporting ATPase ATP7B that results in hepatic copper accumulation and lethal liver failure. The present study focuses on the structural mitochondrial alterations that precede clinical symptoms in the livers of rats lacking Atp7b, an animal model for WD. Liver mitochondria from these Atp7b–/– rats contained enlarged cristae and widened intermembrane spaces, which coincided with a massive mitochondrial accumulation of copper. These changes, however, preceded detectable deficits in oxidative phosphorylation and biochemical signs of oxidative damage, suggesting that the ultrastructural modifications were not the result of oxidative stress imposed by copper- dependent Fenton chemistry. In a cell-free system containing a reducing dithiol agent, isolated mitochondria exposed to copper underwent modifications that were closely related to those observed in vivo. In this cell-free system, copper induced thiol modifications of three abundant mitochondrial membrane proteins, and this correlated with reversible intramitochondrial membrane crosslinking, which was also observed in liver mitochondria from Atp7b–/– rats. In vivo, copper-chelating agents reversed mitochondrial accumulation of copper, as well as signs of intra-mitochondrial membrane crosslinking, thereby preserving the functional and structural integrity of mitochondria. Together, these findings suggest that the mitochondrion constitutes a pivotal target of copper in WD.

Various aromatic, aliphatic and conjugated aldehydes were transformed to the corresponding carboxylic acids with 70% t-BuOOH solution (water) in the presence of catalytic amounts (10 mol%) of Mohr's salt. This method possesses functional group compatibility, does not involve cumbersome work-up, exhibits chemoselectivity since other functional groups remain intact and proceeds under mild conditions. Standard coagulation assays were performed with control and oxidized fibrinogen (Fg), using prothrombin time (PT; 12.5 ± 0.4 vs 25 ± 0.8 seconds, P < .001) and activated partial thromboplastin time (aPTT; 33 ± 2.5 vs 63 ± 4.7 seconds, P < .001). Fibrin clot (MA), clot formation initiation (r), and rate of clot lysis (LY30) were measured, a reflection exposure of Fg to Fe3+/ ascorbate oxidative system by thrombelastograph (TEG) analysis (0, 6, 12, 24, and 48 hours, 6.2 ± 1.3 vs 5.5 ± 1.2, 4.3 ± 1.0 [P < .01], 3.9 ± 1.6, 3.2 ± 0.8, [P < .001]). Maximum amplitude level was found to be lower than control (69.1 ± 7.2 vs 67.9 ± 12.4, 64.0 ± 11.4, 60.2 ± 21.2, 42.2 ± 15.2, P < .001). The lysis rate was changed according to oxidation time between Fg exposed to Fe3+/ascorbate and control exposed to Fe 3+/ascorbate for the same treatment time (1.9 ± 0.71 vs 7 ± 0.5, 1.6 ± 0.1, 1.2 ± 0.5, 0.9 ± 1.3, P < .001). We revealed dysregulation of hemostatic system with contribution of oxidized Fg, which was in direct proportion to the intensity of Fg oxidation.

A new proteomics technique for analyzing 3-nitrotyrosine-containing peptides is presented here. This technique is based on the combined fractional diagonal chromatography peptide isolation procedures by which specific classes of peptides are isolated following a series of identical reverse-phase HPLC separation steps. Here dithionite is used to reduce 3-nitrotyrosine to 3-aminotyrosine peptides, which thereby become more hydrophilic. Our combined fractional diagonal chromatography technique was first applied to characterize tyrosine nitration in tetranitromethane-modified BSA and further led to a high quality list of 335 tyrosine nitration sites in 267 proteins in a peroxynitrite-treated lysate of human Jurkat cells. The oxidation of each alcohol by permanganate ion is found to be first order with respect to each of the reactants resulting in an overall second order rate expression.

Semliki Forest virus (SFV) utilizes a membrane fusion strategy to introduce its genome into the host cell. After binding to cell-surface receptors, virus particles are internalized through receptor-mediated endocytosis and directed to the endosomal cell compartment. Subsequently, triggered by the acid pH in the lumen of the endosomes, the viral envelope fuses with the endosomal membrane. As a result of this fusion reaction the viral RNA gains access to the cell cytosol. Low-pH-induced fusion of SFV, in model systems as well as in cells, has been demonstrated previously to be strictly dependent on the presence of cholesterol in the target membrane. In this paper, we show that fusion of SFV with cholesterol-containing liposomes depends on sphingomyelin (SM) or other sphingolipids in the target membrane, ceramide representing the sphingolipid minimally required for mediating the process. The action of the sphingolipid is confined to the actual fusion event, cholesterol being necessary and sufficient for low-pH-dependent binding of the virus to target membranes. The 9-hydroxyl group on the sphingosine backbone plays a key role in the SFV fusion reaction, since 3-deoxy-sphingomyelin does not support the process. This, and the remarkably low levels of sphingolipid required for half-maximal fusion (1-2 mol%), suggest that the sphingolipid does not play a structural role in SFV fusion, but rather acts as a cofactor, possibly through activation of the viral fusion protein. Domain formation between cholesterol and sphingolipid, although it may facilitate SFV fusion, is unlikely to play a crucial role in the process.

This study investigates the effects that manganese(IV) dioxide particles have on 2,3-naphthalenediol at varying pH levels (i.e., initial pH of 4.58, 5.85, and 8.75) and under different organic concentration conditions (4×10−3, 4×10−4, and 4×10−5 M), and assesses the importance of Mn oxides on abiotic catalysis of the multiple-ringed aromatic compound. Proton concentration affected the rates of reductive dissolution; as the pH values increased, the rate of reductive dissolution decreased, as predicted by theory. Also, as the concentration of naphthalenediol increased, the rate of reductive dissolution increased, although not proportionally; thus indicating that a majority of the active sites had been occupied. In addition, the results tend to confirm that electron transfer/organic release from the oxide surface is the rate-limiting step. This study demonstrates that in an oxic environment and in the presence of 2,3-naphthalenediol, MnO2 particles undergo reductive dissolution; in the process, naphthalenediol is oxidized. An oxidation by-product of reductive dissolution is an insoluble polymerized organic. The organic by-product was deep-brown in appearance, very similar to that of "humified" material. Using infrared spectroscopy, energy-dispersive x-ray analysis, and a microelemental analysis, the "humified" products appeared to be comprised mainly of constituents originating from naphthalenediol.

Enveloped animal viruses, such as Semliki Forest virus (SFV), utilize a membrane fusion strategy to deposit their genome into the cytosol of the host cell. SFV enters cells through receptor-mediated endocytosis, fusion of the viral envelope occurring subsequently from within acidic endosomes. Fusion of SFV has been demonstrated before to be strictly dependent on the presence of cholesterol in the target membrane. Here, utilizing a variety of membrane fusion assays, including an on-line fluorescence assay involving pyrene-labeled virus, we demonstrate that low-pa-induced fusion of SFV with cholesterol-containing liposomal model membranes requires the presence of sphingomyelin or other sphingolipids in the target membrane. The minimal molecular characteristics essential for supporting SFV fusion are encompassed by a ceramide. The action of the sphingolipids is confined to the actual fusion event, cholesterol being necessary and sufficient for low-pH-dependent binding of the virus to target membranes. Complex formation of the sphingolipids with cholesterol is unlikely to be important for the induction of SFV-liposome fusion, as sphingolipids that do not interact appreciably with cholesterol, such as galactosylceramide, effectively support the process.
