Background: Arterial blood pressure (ABP) is essential to provide adequate perfusion pressure and blood flow to tissues. Usually, ABP is tightly controlled through a variety of complex physiological mechanisms, and significant deviation of ABP from normal may result in end‐organ damage. Measurement of ABP is relatively straightforward and can be used to assess cardiovascular function and guide therapeutic decisions. Challenges in the interpretation of ABP may arise due to individual patient factors and may be dependent on the method of measurement. Aim of the article: This article is the first in a two‐part series and aims to review the physiology and measurement of arterial blood pressure in small animal practice. Studying behavioural lateralization in animals holds great potential for answering important questions in laterality research and clinical neuroscience. However, comparative research encounters challenges in reliability and validity, requiring new approaches and innovative designs to overcome. Although validated tests exist for some species, there is yet no standard test to compare lateralized manual behaviours between individuals, populations, and animal species. One of the main reasons is that different fine-motor abilities and postures must be considered for each species. Given that pawedness/handedness is a universal marker for behavioural lateralization across species, this article focuses on three commonly investigated species in laterality research: dogs, cats, and rats. We will present six apparatuses (two for dogs, three for cats, and one for rats) that enable an accurate assessment of paw preference. Design requirements and specifications such as zoometric fit for different body sizes and ages, reliability, robustness of the material, maintenance during and after testing, and animal welfare are extremely important when designing a new apparatus. Given that the study of behavioural lateralization yields crucial insights into animal welfare, laterality research, and clinical neuroscience, we aim to provide a solution to these challenges by presenting design requirements and innovations in methodology across species. Simple Summary: Sixteen cats were used in a model of behavioral and physiological transport stress. Cats were not accustomed to being transported. In an objective evaluation, cats wore a PetPace (PP) collar that recorded carotid pulse rate (PR) and general activity. Video cameras recorded cat behavior during the 70 min transport experience. Cats also wore a plastic collar containing either 2-methyl-2-butenal (2M2B) collars or a placebo collar. This randomized, placebo-controlled, blinded study found that cats with a 2M2B collar had a lower PR, slept more, sat less, and self-groomed more compared with cats wearing a placebo collar. Control cats hid near the back of the transport kennel, and some vomited or had excessive salivation, whereas cats with 2M2B collars did not hide, vomit, or salivate. This controlled study demonstrates behavioral and physiological benefits to transported cats from the use of 2M2B collars. Some cats experience stress when they have novel experiences, such as infrequent transport. This study was a randomized, placebo-controlled, blinded study that sought to objectively evaluate the effects of a 2M2B collar on transported cat physiology and behavior. The statistical model included effects of cat treatment (2M2B vs. control), period (70 min), sex, and interactions. Cats wearing 2M2B collars had an 8% lower PR (p < 0.01), and they slept more and did not hide at the back of the kennel. While control cats vomited or showed excess salivation, cats with 2M2B collars did not show these signs of stress. Male cats were less active during transport than females. Male cats slept more with 2M2B collars compared with male cats with a control collar, but females showed similar sleeping overall regardless of which collar they wore. Female cats increased activity during transport when they had a 2M2B collar, while male activity did not differ with control or 2M2B collars. These data support the concept that the semiochemical 2M2B can reduce stress in transported cats based on objective physiological and behavioral measures. [ABSTRACT FROM AUTHOR]

Wood vinegar, a by-product of charcoal biomass pyrolysis, has been used as a biofungicide in plant disease management because of its antimicrobial properties. However, the physiological and biochemical mechanisms through which wood vinegar alleviates biotic stress are poorly understood. In this study, pot experiments were conducted to investigate the resistance and regulation mechanism of wood vinegar prepared from different raw materials (ZM) and from a single raw material (SM) in controlling tomato (Solanum lycopersicum "Bonny Best") Fusarium wilt at different concentrations (0.3%, 0.6%, 0.9%, 1.2%, and 1.5%). The results showed that ZM and SM had significant control effects on tomato fusarium wilt under different concentrations in the same growth cycle. Under biotic stress, the two kinds of wood vinegar significantly increased the plant height, stem diameter, leaf area and yield of tomato under the concentration of 0.3%, 0.6%, 0.9% and 1.2%, and significantly reduced the content of malondialdehyde (MDA) and hydrogen peroxide (H2O2) in tomato leaves. The effect of 0.9% treatment was the most significant, ZM and SM significantly increased tomato yield by 122% and 74%, respectively, compared with CK under 0.9% treatment. However, the plant height, stem diameter and leaf area of tomato were significantly reduced under 1.5% treatment, but the content of soluble sugar, soluble protein and vitamin C in tomato fruit was the best. Compared with CK, ZM significantly increased by 14%, 193% and 67%, respectively, and SM significantly increased by 28%, 300% and 159%, respectively. Except for 0.3% treatment, both significantly increased the activities of catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD) in tomato leaves. The response intensity of two kinds of wood vinegar—physiological and biochemical—to tomato disease resistance, growth and development, showed ZM > SM. The disease index of tomato showed highly significant negative correlation with plant height, stem thickness, leaf area and antioxidant physiology CAT, and highly significant positive correlation with MDA and H2O2 content. In conclusion, ZM was more effective than SM in enhancing tomato disease resistance by promoting tomato growth and development, decreasing leaf MDA and H2O2 content, and inducing antioxidant enzyme activity in leaves at moderate concentrations. This study was aimed to reveal the efficacy of hypochlorous acid in cats with bacterial cystitis. The animal material of the study consisted of 14 cats of different races, 1-8 years of age, of different sexes, diagnosed with bacterial cystitis as a result of clinical and urine examinations and bacterial urine culture, who applied to Afyon Kocatepe University Veterinary Health Application and Research Centre Internal Diseases Clinic with complaints of lower urinary system disease and diagnosed with bacterial cystitis as a result of the examinations. After the diagnosis of bacterial cystitis, the cats were divided into 2 groups: fluid therapy + enrofloxacin + vitamin C + nitrofurantoin (group I, n = 7) and fluid therapy + enrofloxacin + hypochlorous acid (group II, n = 7). Both groups were treated for 7 days. It was observed that clinical recovery was faster according to the clinical scoring table created in the cats in group II, and bacterial growth in the urine was eliminated more quickly and effectively as a result of urine cultures. As a result, it was concluded that hypochlorous acid is effective in the clinical improvement and elimination of bacteriuria in cats with bacterial cystitis. [ABSTRACT FROM AUTHOR]

The primary objective of this study was to assess how obesity impacts the occurrence of left ventricular hypertrophy in healthy cats. The researchers evaluated the body condition score of all participating cats before conducting the study. Additionally, they performed various echocardiographic examinations, including transthoracic 2D, M-mode, and Doppler, as well as measuring systolic blood pressure. The results indicated a notable distinction in body condition scores between the non-obese and obese groups (P < .001). Moreover, significant differences were observed in terms of systolic blood pressure (SBP) and heart rate between the 2 groups (P < .001, P = .002). Echocardiographic measurements revealed statistically significant variations in left atrium and left ventricular free wall thickness between non-obese and obese cats (P < .001 and P = .003). To address obesity, the obese group underwent a lowcalorie diet, leading to improvements in echocardiographic parameters and SBP. However, these improvements did not achieve statistical significance. The study findings indicate that obesity can lead to substantial myocardial changes, resulting in left ventricular hypertrophy in cats, which has been previously reported in humans and dogs. Simple Summary: Cats are the most common pet animal worldwide. Therefore, enhancing their health and welfare is essential for owners. However, few studies have assessed the physiology of domestic cats noninvasively. In this study, we collected urine samples from domestic cats and quantified cortisol as an indicator of negative stress states, and oxytocin as a positive indicator. Furthermore, we investigated factors influencing these hormonal states, which reflect their physiological status. Our study revealed that the more frequently owners talked to and petted their cats, the higher their urinary oxytocin levels. We suggest that social interactions between cats and their owners are factors that may influence their health and welfare status. Physiological samples are beneficial in assessing the health and welfare of cats. However, most studies have been conducted in specialized environments, such as shelters or laboratories, and have not focused on cats living in domestic settings. In addition, most studies have assessed physiological stress states in cats based on cortisol, and none have quantified positive indicators, such as oxytocin. Here, we collected urine samples from 49 domestic cats and quantified urinary cortisol, oxytocin, and creatinine using ELISA. To identify factors influencing hormone levels, owners responded to questionnaires regarding their housing environment, individual cat information, and the frequency of daily interactions with their cats. Using principal component analysis, principal component scores for daily interactions were extracted. These results showed that the frequency of tactile and auditory signal-based communication by owners was positively correlated with the mean concentration of oxytocin in the urine. Additionally, this communication was more frequent in younger cats or cats that had experienced a shorter length of cohabitation with the owner. However, no factors associated with urinary cortisol concentration were identified. Our study indicates that interactions and relationships with the owner influence the physiological status of cats and suggests that oxytocin is a valuable parameter for assessing their health and welfare. The objective of this study was to determine the prevalence of obesity as a risk factor in geriatric-aged cats. By its approach, the study was quantitative, non-experimental, cross-sectional, descriptive, correlational and observational design. A total of 100 geriatric-aged cats were sampled, with a nonprobabilistic sampling type, using a directed or intentional sampling technique; the inclusion criterion was considered to be geriatric-aged cats ≥ 7 years old. A measuring tape, scales and a structured questionnaire related to risk factors such as age, breed, weight, height, environment, exercise habits, type of feeding and physiological condition were used as measuring instruments. The physical method was used to measure the Body Score Condition (BSC) of 5 points of Thatcher, based on external morphological characteristics such as: observing and palpating the difficulty to palpate the ribs, if they presented thick layer of fat, bony prominences covered by a thick layer of fat, hanging ventral bulge and widened back. There were 10% of cases with obesity and 14 cases with overweight in geriatricaged cats. The determining factors associated with obesity in geriatric-aged cats are the mixed breed, age between 7 and 10 years, larger cats and neutered cats. [ABSTRACT FROM AUTHOR]

Motor lateralization is commonly observed through preferential paw use in dogs and cats. Previous studies have uncovered sex-related differences in paw preference, hypothesizing that these differences may be related to sex hormones. The current study aimed to compare neutered and entire individuals to further investigate whether paw preference is influenced by sex hormones. Dog and cat owners were required to fill in a questionnaire with demographic information such as sex and neuter status of their pets. They then carried out two simple paw preference tasks within their homes: a "reaching for food" task and a "reaching for a toy" task. This study revealed an overall preference among the 272 dogs and 137 cats tested to use their right paw in both tasks. In cats, the degree of paw preference (i.e., regardless of the direction) was significantly influenced by an interaction between neuter status and life stage. Also in dogs, both life stage and an interaction between neuter status and life stage tended to influence the degree of paw preference. Post-hoc power analysis revealed a lack of statistical power, suggesting that future studies using a larger sample size are needed to further investigate potential effects of neuter status on paw preference. Background: Considering animals as individuals and not as species is becoming increasingly essential to animal welfare management in captive settings. Recent studies on big cat personalities and coping strategies suggest personality can help big cats cope in their surroundings. Yet a large portion of the published literature focuses on understanding either the personality or stress physiology of big cats. Our research shows how integrating an improved understanding of the personality of big cats with stress physiology may enhance welfare, especially for endangered species like African lions. By using a wild cat personality checklist, this study compared the key personality dimensions of 22 African lions with its faecal glucocorticoids and assessed factors influencing their personality and stress physiology. Results: We found two reliable personality dimensions for African lions (dominance and agreeableness) and identified key factors (sex, age and location) that may influence their personality. Further, on testing if these factors influenced the stress physiology through variations in glucocorticoid levels, there was no significant difference. However, there was a strong negative association between agreeableness and glucocorticoid levels. These results suggest that the behavioural traits loading positively and higher for agreeableness are associated with lower glucocorticoid stress levels, which may assist a lion to cope with stressors in its surroundings. Conclusions: Our findings highlight this integrated approach of linking personality and stress physiology of big cats can be beneficial for caretakers. For example, during stressful veterinary procedures or in reintroduction programs, recognizing the personality of lions can help in designing or providing them with resources that will alleviate stress. This study aimed to perform a morphometric analysis of the skull of the European shorthair cat by using computed tomographic images. Thirty-seven computed tomography (CT) studies of healthy cats' heads were used for linear measurements and index calculations of the skull and cranium. The following values were determined: skull length = 8.94 ± 0.45 cm, cranial length = 8.21 ± 0.42 cm, nasal length = 0.73 ± 0.17 cm, cranial width = 4.28 ± 0.26 cm, cranial index = 52.18 ± 3.75%, internal height of cranium = 2.88 ± 0.29 cm, external height of cranium = 3.35 ± 0.12 cm, internal length of the cranium = 5.53 ± 0.28 cm, external length of the cranium = 6.32 ± 0.28 cm, internal cranium index = 45.62 ± 4.77%, external cranium index = 53.06 ± 2.07%, internal cranium and skull index = 61.93 ± 2.38%, external cranium and skull index = 70.70 ± 1.72%, width of the foramen magnum = 1.34 ± 0.07 cm, height of the foramen magnum = 1.01 ± 0.09 cm, and foramen magnum index = 75.37 ± 5.76%. It was also found that the population was homogeneous, with the exception of nasal length (NL), and that there was a sexual dimorphism present, with males exhibiting higher dimensions. This work contributed to characterizing the morphometry of the cranium and skull of the domestic cat, a knowledge of utmost importance for the diagnosis and treatment of conditions affecting this complex anatomical region. We live in complex auditory environments, in which we are confronted with multiple competing sounds, including the cacophony of talkers in busy markets, classrooms, offices, etc. The purpose of this article is to synthesize observations from a series of experiments that focused on how spatial hearing might aid in disentangling interleaved sequences of sounds. The experiments were unified by a non-verbal task, "rhythmic masking release", which was applied to psychophysical studies in humans and cats and to cortical physiology in anesthetized cats. Human and feline listeners could segregate competing sequences of sounds from sources that were separated by as little as ∼10°. Similarly, single neurons in the cat primary auditory cortex tended to synchronize selectively to sound sequences from one of two competing sources, again with spatial resolution of ∼10°. The spatial resolution of spatial stream segregation varied widely depending on the binaural and monaural acoustical cues that were available in various experimental conditions. This is in contrast to a measure of basic sound-source localization, the minimum audible angle, which showed largely constant acuity across those conditions. The differential utilization of acoustical cues suggests that the central spatial mechanisms for stream segregation differ from those for sound localization. The highest-acuity spatial stream segregation was derived from interaural time and level differences. Brainstem processing of those cues is thought to rely heavily on normal function of a voltage-gated potassium channel, Kv3.3. A family was studied having a dominant negative mutation in the gene for that channel. Affected family members exhibited severe loss of sensitivity for interaural time and level differences, which almost certainly would degrade their ability to segregate competing sounds in real-world auditory scenes. Extracellular vesicles (EVs) contain multiple factors that regulate cell and tissue function. However, understanding of their influence on gametes, including communication with the oocyte, remains limited. In the present study, we characterized the proteome of domestic cat (Felis catus) follicular fluid EVs (ffEV). To determine the influence of follicular fluid EVs on gamete cryosurvival and the ability to undergo in vitro maturation, cat oocytes were vitrified using the Cryotop method in the presence or absence of ffEV. Vitrified oocytes were thawed with or without ffEVs, assessed for survival, in vitro cultured for 26 hours and then evaluated for viability and meiotic status. Cat ffEVs had an average size of 129.3 ± 61.7 nm (mean ± SD) and characteristic doughnut shaped circular vesicles in transmission electron microscopy. Proteomic analyses of the ffEVs identified a total of 674 protein groups out of 1,974 proteins, which were classified as being involved in regulation of oxidative phosphorylation, extracellular matrix formation, oocyte meiosis, cholesterol metabolism, glycolysis/gluconeogenesis, and MAPK, PI3K-AKT, HIPPO and calcium signaling pathways. Furthermore, several chaperone proteins associated with the responses to osmotic and thermal stresses were also identified. There were no differences in the oocyte survival among fresh and vitrified oocyte; however, the addition of ffEVs to vitrification and/or thawing media enhanced the ability of frozen-thawed oocytes to resume meiosis. BACKGROUND: Phenobarbital is one of the most commonly used drugs to treat epilepsy and other seizure disorders in dogs and cats. Hepatotoxicity following phenobarbital administration is dose-dependent. OBJECTIVES: The present study aimed to evaluate the protective action of silymarin on phenobarbital-induced hepatotoxicity in cats. METHODS: For this purpose, twenty-four healthy adult cats were randomly allotted to four equal groups. Cats in group A were given phenobarbital with dosage 16 mg/kg orally for 28 days; group B received silymarin (30 mg/kg/day for twenty-eight days) orally concurrent with phenobarbital; groups C and D were treated like group B, but silymarin was administered 3 and 48 h after administration of phenobarbital, respectively, and continued up to twenty-eight days. The serum concentrations of alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, lactate dehydrogenase, total and direct bilirubin, blood urea nitrogen and creatinine were measured before administration of phenobarbital and after 24 h, 72 h and 28 days. RESULTS: Phenobarbital elevated significantly serum concentrations of liver enzymes (in all cases), and total and direct bilirubin in two cats of group A, after 24 h (P <0.001). In groups, B and C, levels of serum enzyme activities and total and direct bilirubin remained within the normal range up to 28 days (P>0.05), while in group D, levels of serum enzyme activities (in 4 cases) were higher than the normal values (P<0.001). CONCLUSIONS: The results showed that silymarin can protect liver tissue against oxidative stress in cats with phenobarbital intoxication especially in the first 3 h post-exposure. Feline lower urinary tract disease (FLUTD) is a common problem in cats. The objectives of the study were to determine the prevalence, clinical signs, and causes of FLUTD and the risk factors for FLUTD. The medical records of 3486 cats visiting Chiang Mai University Small Animal Veterinary Teaching Hospital (VTH) between November 2016 and October 2017 were reviewed. An age-matched case-control study was performed to determine the risk factors for FLUTD by comparing 78 cats with FLUTD and 78 clinically normal age-matched cats. For each animal, potential risk data were obtained from medical records and cat owner interviews; these were analysed for associations with FLUTD. Multivariable logistic regression analysis was performed to estimate the odds ratios and to adjust for expected confounding factors. The prevalence of FLUTD in cats visiting the Chiang Mai University Veterinary Teaching Hospital was 2.2%. The most common clinical signs identified were urethral obstruction (55.1%) and haematuria (23.1%). The most common diagnoses were feline idiopathic cystitis (FIC) (57.7%) and urolithiasis (struvite) (18%). The multivariable logistic regression analysis results indicated that FLUTD was most likely to be diagnosed in castrated male cats. FIC and urolithiasis were the most common diagnoses in cats with FLUTD, and male sex and castration increased the risk of FLUTD. [ABSTRACT FROM AUTHOR]

OBJECTIVE To investigate water intake and urine measures in healthy cats provided free-choice access to a nutrient-enriched water with (NWP) or without (NW) added poultry flavoring offered at 3 different volumes in addition to tap water (TW). ANIMALS 36 domestic shorthair cats. PROCEDURES Control group cats (n = 4) received dry food with TW ad libitum throughout the study. Cats of the NW and NWP groups (n = 16/group) received the same food with TW only (period I; 7 days) followed by TW and the assigned treatment ad libitum at IX, I.5X, and 2X the volume of TW consumed in period I during periods 2 (17 days), 3 (10 days), and 4 (10 days), respectively. Liquid consumption, food intake, and total water intake (from all sources) were measured; urine collected over 48 hours in each period was measured, and urine specific gravity (USG) was determined. Data were analyzed with mixed-effects models. RESULTS TW and food calorie intake were similar among groups in period I; TW consumption by control cats did not differ during the study. Liquid consumed by drinking increased 18%, 57%, and 96% for the NWP group in periods 2, 3, and 4, respectively, with increases of 25% and 44% for the NW group in periods 3 and 4, respectively, compared with period I values for the same groups. Increased urine output and decreased USG were significantly associated with period and treatment. CONCLUSIONS AND CLINICAL RELEVANCE Increasing the volumes of NW or NWP offered to healthy cats led to increased free liquid consumption and was associated with greater urine output and dilution as measured by USG. The cat perigeniculate nucleus (PGN) is a visual sector of the thalamic reticular nucleus that consists of GABAergic neurons. It receives excitatory axon-collateral input from relay neurons of the dorsal lateral geniculate nucleus (LGN) to which it provides inhibitory input. Thus, it is usually argued that the PGN works as feedback inhibition to the LGN. At the single neuron level, however, this circuit can also provide lateral inhibition. Which inhibition dominates in the visual circuit of the thalamus has yet to be well characterized. In this study, we conducted cross-correlation analysis of single spike trains simultaneously recorded from PGN and LGN neurons in anesthetized cats. For 12 pairs of functionally connected PGN and LGN neurons with overlapped receptive fields (RF), we quantitatively compared RF properties including the spatial frequency (SF) and temporal frequency (TF) tunings of each neuron. We found the SF and TF tunings of PGN neurons and LGN neurons were similar when there was only excitatory input from the LGN neuron to the PGN neuron, but different when the PGN neuron returned inhibitory inputs back, suggesting the circuit between PGN and LGN neurons works as lateral inhibition for these properties. Cats routinely undergo handling and restraint for examinations and procedures throughout their lifetime. In recent years there has been increased awareness that some forms of restraint have the potential to negatively impact cat health and welfare, but there has been limited research to assess alternatives. Full-body restraint is known to be aversive and cause stress responses across a range of species. The current study therefore validated measures for identifying negative cat responses to handling by comparing passive restraint (n = 22; the control treatment) to full-body restraint (n = 25; negative treatment), during a physical examination. Given that cats respond differently to unfamiliar people, and handlers are often unfamiliar to cats, an Unfamiliar Person Test was used to account for differences in cat responses to an unknown handler. Cats were then restrained according to treatment, and assessed for behavioural and physiological responses during the following phases: 1) placement into restraint, 2) restraint, and 3) post-restraint. The results show the odds of struggling were 8.2 (95% CI: 1.47, 58.53; P = .009) times greater in cats being placed into full-body restraint in comparison to passively restrained cats. During the physical examination, full-body restrained cats showed more breaths per minute (P = .004), more lip licks per minute (F 1,42  = 6.18; P = .017), and were more likely to hold their ears in a back or side position during the first 15 s of handling (P < 0.0001), compared to passively restrained cats. Analysis of pupil dilation ratio (pupil diameter/iris diameter) showed an interaction between treatment and friendliness (F 1,43  = 8.39; P = .006); unfriendly cats handled using full-body restraint showed a larger pupil dilation ratio than unfriendly cats handled with passive restraint (P = .0007). Immediately after the handler released the cat from restraint, the odds of staying on the examination table were 6.1 (95% CI: 1.04, 36.10; F 1,44  = 4.23; P = .0458) times greater for cats restrained with passive restraint, in comparison to cats that underwent full-body restraint. These results provide scientific validation of negative cat responses to handling, and may be used in further research to assess the welfare effects of different handling techniques. OBJECTIVE To evaluate the effects of storage conditions and duration on cobalamin concentration in serum samples from dogs and cats. DESIGN Experiment. SAMPLE Serum samples from 9 client-owned cats and 9 client-owned dogs. PROCEDURES Serum harvested from freshly obtained blood samples was separated into 11 aliquots/animal. One aliquot (baseline sample) was routinely transported in light-protected tubes to the la boratory for cobalamin assay; each of the remaining aliquots was stored in a refrigerator (6°C; n = 5) or at room temperature (20°C) w ith exposure to daylight (5) for 24, 48, 72, 96, or 120 hours. Aliquots were subsequently wrapped in aluminum foil, frozen (-2 0 °C ), and then transported to the laboratory for measurement of cobalamin concentration, all in the same run. Percentage decrease in cobalamin concentration from baseline was analyzed by means of linear mixed modeling. RESULTS N o differences in cobalamin values were identified between cats and dogs; therefore, data for both species were analyzed together. Median baseline serum cobalamin concentration was 424 ng/L (range, 178 to 1,880 ng/L). Values for serum samples stored w ith daylight exposure at room temperature were significantly lower over time than were values for refrigerated samples. Although values for refrigerated samples did not decrease significantly from baseline values over time, values for the other storage condition did; however, the mean percentage decrease for serum samples stored at room tem perature was small (0 .14%/h; 95% confidence interval, 0.07% to 0.21 %/h). CONCLUSIONS AND CLINICAL RELEVANCE Overall, serum cobalamin concentration appeared stable for 5 days when feline and canine serum samples were refrigerated at 6°C. The effect of light and room temperature on serum cobalamin concentration, although significant, was quite small for samples stored w ith these exposures for the same 5-day period. Melanocortin 4 receptor (MC4R), which is a member of the G protein-coupled receptor (GPCR) family, mediates regulation of energy homeostasis upon the binding of α-melanocyte-stimulating hormone (α-MSH) in the central nervous system (CNS). Melanocortin 2 receptor accessory protein 2 (MRAP2) modulates the function of MC4R. We performed cDNA cloning of cat MC4R and MRAP2 and characterized their amino acid sequences, mRNA expression patterns in cat tissues, protein–protein interactions, and functions. We found high sequence homology (>88%) with other mammalian MC4R and MRAP2 encoding 332 and 206 amino acid residues, respectively. Reverse transcription-polymerase chain reaction analysis revealed that cat MC4R and MRAP2 mRNA were expressed highly in the CNS. In CHO-K1 cells transfected with cat MC4R, stimulation with α-MSH increased intracellular cyclic adenosine monophosphate (cAMP) concentration in a dose-dependent manner. Furthermore, the presence of MRAP2 enhanced the cat MC4R-mediated cAMP production. These results suggested that cat MC4R acts as a neuronal mediator in the CNS and that its function is modulated by MRAP2. In addition, our NanoBiT study showed the dynamics of their interactions in living cells; stimulation with α-MSH slightly affected the interaction between MC4R and MRAP2, and did not affect MC4R homodimerization, suggesting that they interact in the basal state and that structural change of MC4R by activation may affect the interaction between MC4R and MRAP2. Simple cells in the cat primary visual cortex usually have elongated receptive fields (RFs), and their orientation selectivity can be largely predicted by their RFs. As to the relay cells in cats’ lateral geniculate nucleus (LGN), they also have weak but significant orientation bias (OB). It is thus of interest to investigate the fine spatiotemporal receptive field (STRF) properties in LGN, compare them with the dynamics of orientation tuning, and examine the dynamic relationship between STRF and orientation sensitivity in LGN. We mapped the STRFs of the LGN neurons in cats with white noise and characterized the dynamics of the orientation tuning by flashing gratings. We found that most of the LGN neurons showed elongated RFs and that the elongation axes were consistent with the preferred orientations. STRFs and the dynamics of orientation tuning were closely correlated temporally: the elongation of RFs and OB emerged, peaked and decayed at the same pace, with unchanged elongation axis of RF and preferred orientation but consistently changing aspect ratio of RF and OB strength across time. Importantly, the above consistency between RF and orientation tuning was not influenced by the ablation of the primary visual cortex. Furthermore, biased orientation tuning emerged 20–30 ms earlier than those in the primary visual cortex. These data demonstrated that similar to the primary visual cortex, the orientation sensitivity was closely reflected by the RF properties in LGN. Rodents are intermediate hosts for many species of Sarcocystis. Little is known of Sarcocystis cymruensis that uses the Brown rat (Rattus norvegicus) as intermediate hosts and the domestic cat (Felis catus) as experimental definitive host. Here, we identified and described Sarcocystis cymruensis in naturally infected R. norvegicus from Grenada,West Indies. Rats (n = 167) were trapped in various locations in two parishes (St. George and St. David). Microscopic, thin (< 1 µm) walled, slender sarcocysts were found in 11 of 156 (7.0%) rats skeletal muscles by squash examination. A laboratory-raised cat fed naturally infected rat tissues excreted sporocysts that were infectious for interferon gamma gene knockout (KO) mice, but not to Swiss Webster outbred albino mice. All inoculated mice remained asymptomatic, and microscopic S. cymruensis-like sarcocysts were found in the muscles of KO mice euthanized on day 70, 116, and 189 post inoculation (p.i.). Sarcocysts from infected KO mice were infective for cats at day 116 but not at 70 days p.i. By transmission electron microscopy, the sarcocyst wall was "type 1a." Detailed morphological description of the cyst wall, metrocytes, and bradyzoites is given for the first time. Additionally, molecular data on S. cymruensis are presented also for the first time. Molecular characterization of sarcocysts 18S rDNA and 28S rDNA, ITS-1, and cox1 loci showed the highest similarity with S. rodentifelis and S. muris. In conclusion, the present study described the natural infection of S. cymruensis in Brown rat for the first time in a Caribbean country and provided its molecular characteristics. This study aimed to investigate the effects of the systemic administration of acepromazine, tramadol and the association of both on intraocular pressure (IOP) and pupil diameter (PD) in young healthy cats. Cats were randomly allocated into three groups (n=10/each) and intramuscular acepromazine (AG), tramadol (TG) or acepromazine combined with tramadol (ATG) were injected. PD (electronic caliper) and IOP (applanation tonometry) were assessed before (baseline) and following 15, 30, 60, and 120 minutes of treatments. It was verified that in AG, PD decreased significantly from time point 30 to 120 (P=0.002), but such reduction did not differ significantly from baseline (P=0.89). In TG, PD increased significantly from the first 15 minutes, until the last time point of evaluation (P<0.001). In ATG, PD increased significantly from time point 30 to 120 when compared to baseline (P<0.001); but significant differences from time point 30 to 120 were not seen (P=0.71). Comparisons among groups showed that PD values of TG and ATG were significantly higher than that of AG (P<0.05). IOP values, on the other hand, did not change significantly among time points and groups (P>0.05). It can be concluded that tramadol alone or in association with acepromazine produced significant mydriasis for up to 120 minutes, without changing IOP values in normal cats. Results of this study suggested that tramadol alone or in association with acepromazine caused significant mydriasis and did not change IOP values in normal cats. Therefore, it may be considered a satisfactory pre-anesthetic combination for ophthalmic surgery in cats. OBJECTIVE To determine effects of restriction feeding of a moderate-protein, high-fiber diet on loss of body weight (BW). voluntary physical activity, body composition, and fecal microbiota of overweight cats. ANIMALS 8 neutered male adult cats. PROCEDURES After BW maintenance for 4 weeks (week 0 = last week of baseline period), cats were fed to lose approximately 1.5% of BW/wk for 18 weeks. Food intake (daily). BW (twice per week), body condition score (weekly), body composition (every 4 weeks), serum biochemical analysis (weeks 0. 1.2. 4. 8, 12. and 16). physical activity (every 6 weeks), and fecal microbiota (weeks 0. 1.2.4.8. 12. and 16) were assessed. RESULTS BW. body condition score, serum triglyceride concentration, and body fat mass and percentage decreased significantly over time. Lean mass decreased significantly at weeks 12 and 16. Energy required to maintain BW was 14% less than National Research Council estimates for overweight cats and 16% more than resting energy requirement estimates. Energy required for weight loss was 11% more. 6% less, and 16% less than American Animal Hospital Association recommendations for weight loss (80% of resting energy requirement) at weeks I through 4, 5 through 8, and 9 through 18. respectively. Relauve abundance of Actinobacteria increased and Bacte-roidetes decreased with weight loss. CONCLUSIONS AND CLINICAL RELEVANCE Restricted feeding of a moderate-protein, high-fiber diet appeared to be a safe and effective means for weight loss in cats. The objective of this study was to determine the pharmacodynamics (PD) and pharmacokinetics (PK) of insulin aspart in healthy cats following intramuscular (IM) and subcutaneous (SC) injection. Eight healthy, purpose-bred cats were used in a randomized, crossover study design. Each cat had 2 isoglycemic clamps performed, one after receiving 0.25 IU/kg of insulin aspart by IM injection and one after receiving the same dose by SC injection. The two isoglycemic clamps were performed on different days, at least 48 h apart. The blood glucose, plasma endogenous insulin, and plasma insulin aspart concentrations were measured and the glucose infusion rate (GIR) was recorded during the clamp. The GIR over time was used to create a time-action curve for each clamp which was used to describe the PD of insulin aspart. Data that are normally distributed are reported as mean ± SD, while data that are not normally distributed are reported as median (25–75 percentile). When compared to the PD data that have been reported for regular insulin in healthy cats, insulin aspart had a more rapid onset (IM: 10 min [10–21.25 min], SC: 12.5 min [10–18.75 min]) and shorter duration of action (IM: 182.5 ± 34.33 min, SC: 159.38 ± 41.87 min). The onset of action ( P = 0.795), time to peak action ( P = 0.499), duration of action ( P = 0.301), and total metabolic effect ( P = 0.603) did not differ with route of administration; however, SC administration did result in a higher maximum plasma insulin aspart concentration (IM: 1,265.17 pmol/L [999.69–1,433.89 pmol/L], SC: 3,278.19 pmol/L [2,485.29–4,132.01 pmol/L], P = 0.000) and larger area under the insulin aspart vs time curve (IM: 82,662 ± 30,565 pmol/L, SC: 135,060 ± 39,026 pmol/L, P = 0.010). Insulin aspart has a rapid onset of action and short duration of effect in healthy cats when administered by IM and SC injection. Although it cannot be assumed that the PD and PK of insulin aspart will be the same in cats with diabetic ketoacidosis (DKA), our data support further investigation into the use of SC insulin aspart as an alternative to regular insulin for the treatment of DKA in cats. Degree of adiposity and dietary macronutrient composition affect incretin hormone secretion in humans and mice, but little is known about their effect in cats. In this study, 7 overweight cats were fed a maintenance diet (MD) for at least 2 wk followed by a reduced calorie diet (RCD), which was lower in fat and higher in carbohydrates and fiber. Cats were fed ad libitum initially, and then, food was restricted to achieve 1%–2% loss of body weight weekly (11 wk). When lean, cats were fed MD for 2 wk. A standardized meal test (SMT) using a third diet was performed after at least 7 d on each diet, before and after weight loss (four SMT's total). Glucose, insulin, glucagon-like peptide-1 (GLP-1), and glucose-dependent insulinotropic peptide (GIP) concentrations were measured immediately before and over 6 h after feeding the SMT. Area under the curve (AUC) was compared for GLP-1, GIP, and insulin concentrations using 2-way analysis of variance. Leaner cats had increased GIP AUC compared to obese cats ( P = 0.025). There was a trend toward increased GIP AUC on RCD compared to the MD ( P = 0.085). There was a moderate negative correlation between body fat percentage and GLP-1 AUC ( r = −0.45; P = 0.05). There was no effect of diet on GLP-1 AUC . In conclusion, degree of adiposity and dietary macronutrient content could be important in determining GIP responses not only acutely but also on a long-term basis. Background Mycophenolate mofetil (MMF), the prodrug of mycophenolic acid (MPA), is becoming increasingly popular as an alternative immunosuppressant in feline medicine. Pharmacokinetic information is not available for cats. Objective The purpose of this study was to determine whether MMF is biotransformed into the active metabolite MPA and to evaluate the disposition of MPA after a 2-hour constant rate intravenous (IV) infusion of MMF in healthy cats. Animals Healthy cats (n = 6). Methods This was a prospective pilot study. All cats were administered MMF at 20 mg/kg every 12 hours over a 2-hour constant rate infusion for 1 day. The concentrations of MPA and its derivatives in blood were determined using a validated UHPLC-UV method. Results All cats biotransformed MMF into MPA. The mean AUC0-14 h ranged from 6 to 50 h*mg/L after IV dosing of MMF. Transient large bowel diarrhea was recorded in 2 of 6 cats after medication administration. Conclusion and Clinical Importance The disposition of MPA in plasma was highly variable, which could result in high interindividual variability in the safety and efficacy of treatment with MMF in cats. Objective To compare the duration, quality of anaesthesia and analgesia, and quality of recovery of dexmedetomidine and methadone combined with either ketamine or alfaxalone. Study design Randomized, prospective clinical trial. Animals A group of 44 healthy client-owned cats presenting for ovariectomy. Methods Cats were randomly assigned to one of the two treatment groups: DAM (n= 22), which was administered intramuscularly (IM) dexmedetomidine (15 μg kg-1), methadone (0.3 μg kg-1) and alfaxalone (3 μg kg-1), and DKM (n=22), which was administered IM dexmedetomidine (15 μg kg-1), methadone (0.3 μg kg-1) and ketamine (3 μg kg-1). During anaesthesia, heart rate, respiratory rate and systolic arterial pressure were measured every 5 minutes. Cats that moved or had poor muscle relaxation were administered an additional 1 μg kg-1 of either alfaxalone (DAM) or ketamine (DKM) intravenously (IV). In cases of increased autonomic responses to surgical stimulation, fentanyl (2 μg kg-1) was administered IV. At the end of the surgery, atipamezole (75 μg kg-1) was administered IM, and the times to both sternal recumbency and active interaction were recorded. Quality of recovery was evaluated with a simple descriptive scale. The UNESP-Botucatu multidimensional composite pain scale and a visual analogue scale were used to evaluate postoperative analgesia at the return of active interaction and 1, 2 and 3 hours later. Results The additional anaesthesia and rescue fentanyl requirements were similar between groups. The quality of recovery was better in the DAM group than in the DKM group [simple descriptive scale scores: 0 (0-1) and 1 (0-3), respectively; p=0.002]. Postoperative pain scores decreased progressively over time in both groups, with no significant differences (p=0.08) between them. Conclusions and clinical relevance Both protocols provided comparable quality of anaesthesia and analgesia and were suitable for cats undergoing ovariectomy. Surveys report that 25-57% of cats are overweight or obese. The most evinced cause is neutering. Weight loss often fails; thus, new strategies are needed. Obesity has been associated with altered gut bacterial populations and increases in microbial dietary energy extraction, body weight and adiposity. This study aimed to determine whether alterations in intestinal bacteria were associated with obesity, energy restriction and neutering by characterising faecal microbiota using 16S rRNA gene sequencing in eight lean intact, eight lean neutered and eight obese neutered cats before and after 6 weeks of energy restriction. Lean neutered cats had a bacterial profile similar to obese rodents and humans, with a greater abundance (P < 0-05) of Firmicutes and lower abundance (P < 0-05) of Bacteroidetes compared with the other groups. The greater abundance of Firmicutes in lean neutered cats was due to a bloom in Peptostreptococcaceae. Obese cats had an 18 % reduction in fat mass after energy restriction (P < 0-05). Energy reduction was concurrent with significant shifts in two low-abundance bacterial genera and trends in four additional genera. The greatest change was a reduction in the Firmicutes genus, Sarcina, from 4-54 to 0-65% abundance after energy restriction. The short duration of energy restriction may explain why few bacterial changes were observed in the obese cats. Additional work is needed to understand how neutering, obesity and weight loss are related to changes in feline microbiota and how these microbial shifts affect host physiology. Obesity is regarded as one of the most significant welfare issues in companion animals. Some risk factors affecting body condition in cats have been determined, but many remain controversial. The current study aimed to investigate the risk factors for overweight and underweight in cats in metropolitan Sydney. Electronic patient records for 11 years (2005–2015) were acquired from a feline primary practice in metropolitan Sydney. The 9-point body condition score (BCS) evaluation in each visit was classified into three groups: underweight (1–3), ideal-weight (4–6) and overweight (7–9). Multinomial logistic regression was used to identify the risk factors for underweight and overweight. The risk factors tested included: breed, hair length, sex, neuter status, age at neutering, age at visit, microchip status, lifestyle, reason for the clinic visit and the number of visits by a cat in a calendar year. The BCS examiner was included as a confounder. Over the 11-year period, 4,020 cats had made 18,349 visits to the clinic, from which BCS records were available for 15,659 visits (85.3%), with 834 cats (5.3%) being underweight, 12,362 (79.0%) being ideal-weight and 2,463 (15.7%) being overweight. Although various cat breeds showed a different tendency for body condition, British Shorthair and mixed-breed cats were more likely to be overweight. Non-illness related visits and middle age (7–11 years old) were also risk factors for overweight. The risk factors for underweight included purebred cats (apart from British Shorthair and Burmese), medium-or-long hair, being intact, increase in age, not having a microchip and illness-related visits. Sex was not a significant risk factor for either overweight or underweight. The study demonstrated that the risk factors for overweight and the protective factors for underweight were not necessarily the same, highlighting the importance of investigating the risk factors for underweight and overweight separately in future research. A full 24 factorial design was applied to find the best combination of diluted reagents (HNO3 and H2O2), time, and temperature for the digestion of samples of wet feed for dogs and cats using a closed digestion block. The residual carbon concentration (RCC) was used as the response in the factorial design. All variables and their interactions significantly influenced the digestion of the feed samples, as indicated by the RCC. The conditions established for the digestion of 0.05 g (dry mass) wet feed samples were the addition of 3.0 mol/L HNO3 and 5.0% m/m H2O2 in a final volume of 10 mL, followed by heating in a closed digestion block at a temperature of 170°C for 120 min. Analyses were performed by inductively coupled plasma (ICP) optical emission spectrometry (OES). LOQs ranged from 0.2 μg/g (Mg and Sr) to 51 μg/g (P). Accuracy of the analytical method was confirmed through the analysis of the Standard Reference Materials Tomato Leaves (NIST 1573), Apple Leaves (NIST 1515), and Peach Leaves (NIST 1547). The agreement values achieved ranged from 80.2 ± 0.3% for Ba to 113.8 ± 7.1% for Zn (n = 3). Addition and recovery tests were carried out by adding the analytes to a feed sample at two concentration levels, and the recoveries were between 84 ± 6 and 114 ± 10% for macroelements (Ca, K, Mg, and P; n = 3) and between 88 ± 3 and 113 ± 7% for microelements and trace elements (B, Cu, Fe, Sr, and Zn; n = 3). The precision values achieved for the different elements, expressed as RSDs, were better than 7.3% (Zn; n = 3) except for Cu determination, that was 14.6% (n=3). The optimized analytical method was applied to 10 commercial samples of wet feed for cats and dogs, with the concentrations of Al, B, Ba, Ca, Cu, Fe, K, Mg, Mn, P, Sr, and Zn determined by ICP-OES. The main objectives of sperm cryopreservation in domestic cats are to preserve these gametes for future use, especially in valuable domestic cat breeds and to use knowledge-gained for developing sperm preservation techniques in wild felids that are threatened with extinction. To achieve acceptable quality of post-thaw sperm and results after insemination, sperm samples must be properly handled, cryopreserved and thawed by using appropriate protocols. In this paper, cryopreservation protocols of domestic cat sperm that have been reported previously are described. The subtopics include sources of sperm, freezing extenders, methods of sperm dilution, freezing storage vessels, methods of sperm cryopreservation, thawing temperature, and thawing extenders. Contents This study aimed to evaluate tissue damage of feline testicles sectioned in two different sizes (0.3 or 0.5 cm3) and submitted to different cryoprotectants (propanediol or glycerol). Testicles obtained from 12 domestic cats were sectioned in 0.3 and 0.5 cm3 sized pieces and immediately evaluated by TBARS and semi-quantitatively by histomorphology. The remaining fragments were placed in cryotubes with 1 ml Egg yolk Tris Equex STM extender containing 3% glycerol or 3% propanediol and cryopreserved by fast-freezing technique. Frozen-thawed fragments were also evaluated by TBARS and histomorphology. Statistical analysis was performed using one-way ANOVA with Student-Newmann-Keuls post hoc test, with p < .05. Fresh and cryopreserved tissues generally exhibited a similar morphology concerning detachment of cells from the basement membrane and observation of nucleoli, with a great proportion scored as 0 (no alteration). When present, alterations were slight and the morphology was considered to be good (most classified in scores 1). Pyknosis was the main anomaly observed as score 2 in 54.6% and 58.4% of 0.3-cm3 fragments cryopreserved in propanediol and glycerol, respectively (16.7% scored 2 in fresh tissue). In TBARS evaluation, 0.5-cm3 fragments cryopreserved in glycerol produced less free radical compared to the 0.3 cm3 cryopreserved in glycerol or propanediol. Our results showed that glycerol was more efficient than propanediol to cryopreserve 0.5-cm3 fragments; this might be attributed to the fact that glycerol molecular weight is larger than propanediol and so its perfusion in the testicular tissue is slower. Use of the immunosuppressant mycophenolic acid ( MPA) in cats is limited because MPA elimination depends on glucuronidation, which is deficient in cats. We evaluated formation of major (phenol glucuronide) and minor (acyl glucuronide, phenol glucoside, and acyl glucoside) MPA metabolites using liver microsomes from 16 cats, 26 dogs, and 48 humans. All MPA metabolites were formed by human liver microsomes, while dog and cat liver microsomes formed both MPA glucuronides, but only one MPA glucoside (phenol glucoside). Intrinsic clearance ( CLint) of MPA for phenol glucuronidation by cat liver microsomes was only 15-17% that of dog and human liver microsomes. However, CLint for acyl glucuronide and phenol glucoside formation in cat liver microsomes was similar to or greater than that for dog and human liver microsomes. While total MPA conjugation CLint was generally similar for cat liver microsomes compared with dog and human liver microsomes, relative contributions of each pathway varied between species with phenol glucuronidation predominating in dog and human liver microsomes and phenol glucosidation predominating in cat liver microsomes. MPA conjugation variation between cat liver microsomes was threefold for total conjugation and for phenol glucosidation, sixfold for phenol glucuronidation, and 11-fold for acyl glucuronidation. Our results indicate that total MPA conjugation is quantitatively similar between liver microsomes from cats, dogs, and humans despite large differences in the conjugation pathways that are utilized by these species. In contrast to the species studied, the corpus luteum (CL) of Iberian and Eurasian lynx physiologically persists in the ovary for more than 2 years and continues to secrete progesterone. Such persistent CL (perCL) transition into the next cycle and are present in the ovary together with the freshly formed CL (frCL) of a new ovulation. To date, the mechanisms supporting such CL persistence are not known. We analyzed the potential receptivity of feline CL to sex steroids through mRNA measurements of progesterone receptor (PGR), progesterone receptor membrane components (PGRMC) 1 and 2, estrogen receptor (ESR) 1 and ESR2, G protein–coupled estrogen receptor 1 (GPER1), and androgen receptor (AR). All receptors were present in domestic cat CL during pregnancy and the nonpregnant luteal phase, in frCL and perCL of post-mating Iberian lynx and in perCL of pre-mating Eurasian lynx. Mass spectrometry detected the presence of PGRMC1 protein in frCL and perCL of the Iberian lynx. In both domestic cat and lynx CL, PGR, PGRMC1, and ESR1 proteins were localized in luteal cells by immunohistochemistry. The mRNA levels of PGR , PGRMC1 , PGRMC2 , ESR1, and AR changed significantly throughout the domestic cat luteal phase. This may indicate involvement of these receptors in the processes of formation, maintenance, and regression of feline CL. In Iberian lynx, expression of PGRMC1 , PGRCM2 , ESR1 , GPER1, and AR was significantly higher in perCL compared with frCL, whereas ESR2 was reversed. High mRNA amounts of these receptors in perCL suggest that physiological persistence of lynx CL may be partly regulated by actions of sex steroids through their nuclear and/or membrane receptors. Today immunoassays are widely used in veterinary medicine, but lack of species specific assays often necessitates the use of assays developed for human applications. Mass spectrometry (MS) is an attractive alternative due to high specificity and versatility, allowing for species-independent analysis. Targeted MS-based quantification methods are valuable complements to large scale shotgun analysis. A method referred to as parallel reaction monitoring (PRM), implemented on Orbitrap MS, has lately been presented as an excellent alternative to more traditional selected reaction monitoring/multiple reaction monitoring (SRM/MRM) methods. The insulin-like growth factor (IGF)-system is not well described in the cat but there are indications of important differences between cats and humans. In feline medicine IGF–I is mainly analyzed for diagnosis of growth hormone disorders but also for research, while the other proteins in the IGF-system are not routinely analyzed within clinical practice. Here, a PRM method for quantification of IGF–I, IGF–II, IGF binding protein (BP) –3 and IGFBP–5 in feline serum is presented. Selective quantification was supported by the use of a newly launched internal standard named QPrEST™. Homology searches demonstrated the possibility to use this standard of human origin for quantification of the targeted feline proteins. Excellent quantitative sensitivity at the attomol/μL (pM) level and selectivity were obtained. As the presented approach is very generic we show that high resolution mass spectrometry in combination with PRM and QPrEST™ internal standards is a versatile tool for protein quantitation across multispecies. The first aim of this study was to provide a comprehensive analysis of structural and functional features of spermatozoa in semen collected from the same cat by two methods: urethral catheterization and epididymis slicing. The second aim was to assess if feline urethral (CT) and epididymal (EP) spermatozoa undergo the same changes during cryopreservation and to compare the postthaw characteristics of spermatozoa collected by the two methods. In the first phase, CT and EP semen were collected from 20 cats, and sperm motility, viability, morphology, computer-assisted sperm analysis (CASA) parameters, membrane and acrosome integrity, mitochondrial potential, lipid peroxidation, and chromatin status were assessed. In the second phase, both types of semen collected from 10 cats were cryopreserved, thawed, and the same sperm parameters were assessed as in fresh semen. Fresh CT spermatozoa (phase I) showed higher (P < 0.05) motility (subjective: median 75.0% vs. 62.5%; by CASA: mean ± SD 60.2 ± 10.7% vs. 43.1 ± 16.7%), morphology (mean ± SD, 57.5 ± 9.6% vs. 45.2 ± 15.9%), and membrane integrity (median live: 89.2% vs. 79.8%). Other parameters were not different between CT and EP spermatozoa. After cryopreservation (phase II), spermatozoa from both types of semen did not differ significantly, except for lipid peroxidation of live sperm cells (median CT: 3.5%, EP: 1.7%, P < 0.05). Urethral and EP spermatozoa showed a similar, significant drop in motility (CT to 18.6 ± 10.3% and EP to 21.6 ± 12.1%, P < 0.05), progressive motility (CT to 6.8 ± 5.9% and EP to 8.3 ± 8.8%, P < 0.05), and rapid movement (from 34.3 ± 20.6% to 8.5 ± 7.0% in CT and from 26.0 ± 14.7% to 10.1 ± 10.4% in EP, P < 0.05), whereas other motion characteristics assessed by CASA were not affected (P > 0.05). The strongest change after cryopreservation was noted in high mitochondrial potential (median CT: 1.3%, EP: 2.2%). Although cryopreservation increased acrosome damage and lipid peroxidation, the level of these changes in the population of live sperm cells remained low (median acrosome damage: CT: 3.3%, EP: 4.5%, lipid peroxidation CT: 3.5%, EP: 1.7%). Cryopreservation did not affect chromatin structure (median percent DNA fragmentation index, CT: 3.3%, EP: 2.3%). In this study, we confirmed that urethral catheterization for collection of semen allows the retrieval of spermatozoa with quality equally good as in those obtained by epididymal slicing. The applications of data on biological variation include assessment of the utility of population-based reference intervals, evaluation of the significance of change in serial results, and setting of analytical quality specifications. We investigated the biological variation of 19 biochemistry analytes and total T4, measured in serum from 7 clinically healthy domestic cats sampled once weekly for 5 weeks. Samples were frozen and analyzed in random order in the same analytical run. Results were analyzed for outliers, and the components of variance, subsequently generated by restricted maximum likelihood, were used to determine within-subject and between-subject variation (CVI and CVG, respectively), as well as analytical variation (CVA) for each analyte. Indices of individuality, reference change values, and analytical performance goals were calculated. The smallest CVI and CVG were found for calcium, chloride, and sodium, whereas the largest values were calculated for bile acids. Nine analytes (albumin, alkaline phosphatase, alanine aminotransferase, aspartate aminotransferase, cholesterol, creatinine, phosphate [phosphorus], total protein, total T4) demonstrated high individuality, indicating limited utility of population-based reference intervals. Individuality was low, and population-based reference intervals were thereby considered appropriate for 5 analytes (bile acids, calcium, fructosamine, glucose, potassium). The intermediate individuality observed for 4 analytes (creatine kinase, iron, magnesium, urea) indicated that population-based reference intervals should be used with caution. Background: Osteoarthritis (OA) is a common cause of chronic pain and dysfunction in older cats. The majority of cats with OA do not show signs of overt lameness, yet cats with orthopaedic disease are known to redistribute their body weight from the affected limb. OA can cause changes in the cat's behaviour, which is often misinterpreted as signs of aging. The aim of the present study was to investigate if cats with a previous cranial cruciate ligament (CCL) injury perform differently on the pressure mat and exhibit different behaviour compared to sound cats according to the owner's subjective assessment. Ten cats with a previous CCL injury were assessed with a pressure mat system and their owners were asked to complete an assessment questionnaire. The results were compared to those of 15 sound cats, matched to have the same weight and body condition score. Results: The front/hind limb index for peak vertical force (PVF) was significantly higher for CCL cats, and there was a decreased PVF and vertical impulse (VI) on the affected hindlimb compared to the unaffected one. The results indicate that cats with a previous CCL injury put less weight, on the affected hindlimb but for a longer time. There was a significantly higher owner assessment questionnaire score for the group of cats with CCL injury compared to sound cats. Conclusions: Cats with a previous CCL injury have a different gait pattern compared to sound cats and a different behaviour according to owner subjective assessment. It is of great importance that further studies are performed to investigate the long term effects of CCL injury as a cause of pain and physical dysfunction, and its role in the development of OA in cats. Anti-Müllerian hormone (AMH) belongs to the Transforming Growth Factor-β (TGF-β) family and produces only in ovaries by the granulosa cells of growing follicles in females. Therefore the measurement of AMH levels plays an important role for the detection of the presence or absence of t he ovaries. The aim of this study was to evaluate the individual AMH levels before and after ovariohysterectomy (OHE), to demonstrate mean AMH alterations in the absence of ovaries with blood sampling in different days after OHE, to demonstrate the age-related changes in serum AMH levels and investigate the usability of serum AMH levels as a diagnostic tool for detecting the presence of functional ovarian tissue in domestic cats. Totally 30 domestic cats were used for this study. The animals were allocated into 2 groups according to their ages; <1 year old (Group 1, n=13) and >1 year old (Group 2, n=17). Mean serum AMH concentrations of all cats at day 0 and day 3rd were found 3.15±2.25 ng/mL and 0.38±0.21 ng/mL respectively. Ten days after surgery, AMH levels of all spayed cats were below the minimum detectable concentration of the ELISA kit (<0.08 ng/mL). According to the age classification, the age of cats were correlated with AMH levels at day 0 (rs=0.293, P<0.05) and at day 3 (rs=0.410, P<0.01). It can be concluded that measurement of serum AMH concentrations is adequate for assessing the functional ovarian tissue in cats. [ABSTRACT FROM AUTHOR]

Objectives To retrospectively evaluate the complications, graft clarity, and outcomes associated with the use of commercially available porcine urinary bladder submucosa ( ACell Vet™) alone for corneal reconstruction in dogs and cats. Procedures Dogs or cats receiving an ACell Vet™ graft for corneal reconstruction due to severe ulcerative keratitis or after a keratectomy to remove a corneal sequestrum were included. All received a single layer of ACell Vet™, bandage contact lens, and temporary tarsorrhaphy. Bandage contact lens and temporary tarsorrhaphy were removed after graft vascularization or epithelialization. Topical steroids, cyclosporine, tacrolimus were started after epithelialization. Based on their last examination, outcomes were categorized into five groups based on the presence of corneal vessels, appearance of the scar, and the ability to visualize the posterior and/or the anterior segment through the grafted area. Results There were 82 eyes included in the study, with 68 eyes with sufficient follow-up time for final assessment. Scarring was minimal in 47 eyes, moderate but not enough to obscure visualization of the posterior segment in 12, and severe in nine. There were five eyes that developed phthisis bulbi, glaucoma or were enucleated and nine that were lost to follow up. Graft dehiscence occurred in 19 eyes. Twelve healed without additional surgical intervention while three required a second graft, two became phthisical, and two were enucleated. Conclusions and Clinical Relevance Corneal reconstruction with ACell Vet™ alone is a viable alternative and results in minimal scarring and complications in cats. In dogs, scarring is more pronounced than in cats and graft dehiscence rate is higher compared to conventional techniques. [ABSTRACT FROM AUTHOR]
