1. Nanocharacterisation of III-nitride semiconductors

Abstract

Scanning electron microscopy allows for the investigation of materials down to the nanometre scale. In this thesis I combine cathodoluminescence spectroscopy with complementary techniques to investigate a range of III-nitride semiconductor heterostructures. I first describe AlGaN-coreshell nanorod UV-LED structures. Here, I conducted a wideranging study investigating the optical, electrical and structural properties of these structures. The electrical measurements were enabled by a nanoprobing system for in-situ contacting within the scanning electron microscope combined with bespoke focused ion beam deposition and milling to create contacts on individual nanorods. I-V measurements and electron beam induced currents confirmed the formation of a p-n junction and successful doping within the structure. In the same microscope, I measured CL to find the emission properties and found optically active quantum wells emitting near 300nm, with some variation in energy and intensity as well as clustering. In a transmission electron microscope, energy dispersive X-ray analysis confirmed the existence of clusters occurring at a-plane nanofacets. Low temperature and time resolved measurements measure the speed of the recombination in the quantum wells, showing a reduction in the quantum confined Stark effect within these structures. Using this same combination of techniques, I also examined planar UV-LED structures. I found that screw dislocations pinned the position of hexagonal hillocks during growth. In each of the penetrated layers, higher point defect densities and three-dimensional growth enclose the threading dislocations. These point defects can compensate electrical dopants and increase parasitic recombination reducing quantum well emission intensity. I found that during growth, the emergence of multiple facets with distinct incorporation rates leads to alloy composition and doping fluctuations, resulting in hexagonal structures interconnecting and forming a network. I discovered that by changing the alloy compositions present within the LED structure, screwtype threading dislocations can either enhance recombination (non-radiative) or reduced it. Finally I discuss lateral polarity heterostructures, which contain alternating stripes of Gaand N-polar material. Electron backscattered diffraction measurements confirmed the stripes were of the intended polarity but found in addition small inclusions of undesirable material in the Ga-polar regions. With cathodoluminescence spectroscopy I investigated the properties of the two distinct domains as well as their interface. I found that the Ga-polar region displayed far greater defect luminescence, consistent with some previous studies.

Abstract

Existing barriers to efficient deep ultraviolet (UV) light-emitting diodes (LEDs) may be reduced or overcome by moving away from conventional planar growth and toward three-dimensional nanostructuring. Nanorods have the potential for enhanced doping, reduced dislocation densities, improved light extraction efficiency, and quantum wells free from the quantum-confined Stark effect. Here, we demonstrate a hybrid top-down/bottom-up approach to creating highly uniform AlGaN core-shell nanorods on sapphire repeatable on wafer scales. Our GaN-free design avoids self-absorption of the quantum well emission while preserving electrical functionality. The effective junctions formed by doping of both the n-type cores and p-type caps were studied using nanoprobing experiments, where we find low turn-on voltages, strongly rectifying behaviors and significant electron-beam-induced currents. Time-resolved cathodoluminescence measurements find short carrier liftetimes consistent with reduced polarization fields. Our results show nanostructuring to be a promising route to deep-UV-emitting LEDs, achievable using commercially compatible methods.

1. The 1970's were times of considerable change in our understanding and beliefs about the nature of 'problem drinking'. The 'Disease Concept of Alcoholism' had been shown to have limited value and no validity. The hazards of inappropriate use of the 'Alcoholism' label had been highlighted. There was no clear successor to the disease concept, although the hybrid model of 'The Alcohol Dependence Syndrome' seemed to be the nearest there was to a consensual view. But it adhered to the older notion of Inexorability, re-expressed as 'Restitution after Abstinence' , in spite of growing evidence of Reversibility. There was a need for the development of new models of alcohol problems. The Author has developed a method of examining Models of Alcohol Problems and has been able to derive the profile of the Alcohol Dependence Syndrome using this method. 2. Views about Treatment have also changed. Hospitalbased treatment for 'problem drinkers' had been shown to be of little value: minimal interventions appeared as effective. The use of: Involvement of Spouse, Therapeutic Community Milieu, Antabuse (for an Abstinence Goal) and Vigorous Follow-up differentiated more successful from less successful agencies. Assessment procedures were believed to be of great importance although the 'problem drinker' tended to be seen as an object of study and not as a participant in the process, whose views might also be of importance and value. The logic of matching the individual needs of the 'problem drinker' with what service is offered was advocated but had not been implemented systematically. Multidisciplinary Community-based Services were being suggested, but none had been established for long enough for an evaluation of efficacy to be undertaken. 3. Abstinence was no longer regarded as the major criterion of successful treatment outcome. Experimental controlled drinking programmes appeared to be promising. 4. Ho treatment system had been shown to make any impact on the prevalence of alcohol problems in a community. It was believed that gross per capita consumption of alcohol in that community was the major modifiable influence on that prevalence. By the late 1970's, no other attempt had been made to operate the major alcohol treatment resource of a whole community along the lines of the changes noted in 1 - 3 (above). 5. "The Leicestershire Community Alcohol Services" were instituted in 1978. For a population of just under one million, they provided a non-abstinence oriented, multidisciplinary community-based response. They attempted to maintain the "customers" in their own living and working environment and to reinforce their beliefs in their own essential normality and continuing responsibility for personal conduct. 6. Referral rates into the Leicestershire Services have risen greatly since their inception. "Customers" using the services had typical demographic and alcohol use characteristics of users of alcohol services in Britain generally. Mean consumption per drinking day was the alcohol equivalent for men of a bottle of spirits, for women a bottle of fortified wine (Sherry, Martini). At the time of presentation to the services, they were judged to show a high degree of insight into their drinking and other problems. At follow-up, most saw themselves as 'ex-problem drinkers', whether they continued to drink or not. 7. Using the same method as he used to derive the profile of the Alcohol Dependence Syndrome, the Author attempts to explore a new model of alcohol problems, "Determined Drinking" based predominantly on the self reports of the "customers" of the Leicestershire Community Alcohol Services. It incorporates an acceptance of Reversibility. 8. A short-term uncontrolled follow up of a sample of users of the Leicestershire Services was carried out. There was a high rate of attrition from the study, particularly of less stably housed men. But this study appeared to indicate that six months after cessation of contact, consumption of alcohol had either ceased (24.4%) or reduced to approximately half reported intake at initial assessment. Patterns of consmption had changed from all-day solitary facultative drinking to drinking in the company of family and friends in the evening. 9. Using data collected from Hospital Records, Death Certificates and Police Records, a wide range of indirect indicators of alcohol problems were compared in a natural experimental design. Three counties in the East Midlands of England, with similar demography and drinking patterns, but different styles of Alcohol Services: Derbyshire (effectively generic only), Nottinghamshire (In-patient Alcohol Treatment Unit and 'outreach') and Leicestershire (Community Alcohol Services) were compared over time. Using a mathematical modelling method of data analysis that incorporated change over time as a factor, significant differences in the time series between the three counties were found. Some baseline differences were present. But with the development of the alternative styles of alcohol services, further significant changes over time occurred. In general, compared with one or both of its neighbours, concomitant with a burgeoning usage of its Community Alcohol Services, there has been a flattening off or an actual reduction in alcohol related morbidity and criminality in Leicestershire. One possible explanation of these findings is the difference in the style of provision of Alcohol Services in the three counties. Other passible explanations, such as differences in demography, alcohol use, general medical services' or policing practices would appear unable to account for the wide spectrum of differences reported. Thus the policies and practices of the "Leicestershire Community Alcohol Services" appear to offer a timely and promising model of service provision for alcohol problems in a whole community. They now merit further examination and evaluation locally as do similar services established elsewhere, particularly in areas with very different drinking practices and prevalences of alcohol problems.

Abstract

Objective means are needed to predict and assess clinical response in patients treated for invasive aspergillosis (IA). We examined whether early changes in serum galactomannan (GM) and/or β-D-glucan (BDG) can predict clinical outcomes. Patients with proven or probable IA were prospectively enrolled, and serial GM and BDG levels and GM optical density indices (GMI) were calculated twice weekly for 6 weeks following initiation of standard-of-care antifungal therapy. Changes in these biomarkers during the first 2 and 6 weeks of treatment were analyzed for associations with clinical response and survival at weeks 6 and 12. Among 47 patients with IA, 53.2% (25/47) and 65.9% (27/41) had clinical response by weeks 6 and 12, respectively. Changes in biomarkers during the first 2 weeks were associated with clinical response at 6 weeks (GMI, P = 0.03) and 12 weeks (GM+BDG composite, P = 0.05; GM, P = 0.04; GMI, P = 0.02). Changes in biomarkers during the first 6 weeks were also associated with clinical response at 6 weeks (GM, P = 0.05; GMI, P = 0.03) and 12 weeks (BDG+GM, P = 0.02; GM, P = 0.02; GMI, P = 0.01). Overall survival rates at 6 weeks and 12 weeks were 87.2% (41/47) and 79.1% (34/43), respectively. Decreasing biomarkers in the first 2 weeks were associated with survival at 6 weeks (BDG+GM, P = 0.03; BDG, P = 0.01; GM, P = 0.03) and at 12 weeks (BDG+GM, P = 0.01; BDG, P = 0.03; GM, P = 0.01; GMI, P = 0.007). Similar correlations occurred for biomarkers measured over 6 weeks. Patients with negative baseline GMI and/or persistently negative GMI during the first 2 weeks were more likely to have CR and survival. These results suggest that changes of biomarkers may be informative to predict and/or assess response to therapy and survival in patients treated for IA.

Abstract

A two-stage revision total hip replacement (THR) remains the gold standard for treating chronic periprosthetic joint infection. The goals for the first stage are twofold: (1) remove infected tissue/implants, obtaining tissue samples for culture; (2) deliver local antibiotics and maintain hip function with a prosthesis with antibiotic-loaded acrylic cement. With extensive bone loss prefabricated spacers are not sufficient, therefore customised spacers are required. The current technique, coating cephalomedually femoral nail in cement, is complex, time consuming, functions poorly and causes wear debris from excessive friction at the femoral/acetabular articulation. We report a technique to create a custom made composite spacer using a short femoral nail, standard Exeter femoral stem (Stryker), Trident acetabular cup (Stryker) and polymethyl methacrylate (PMMA) bone cement with antibiotics. This technique restores hip biomechanics and gives a stable articulation, even in the presence of abductor deficiency, due to dual mobility bearings.

Abstract

Treating milk samples with 2,500 pulses of a 30 kV electric field per centimeter at 50 degrees C. can inactivate Mycobacterium paratuberculosis. This bacterium causes Johne's disease in animals and may be linked to Crohn's disease in humans.

Abstract

The production of R-1,2-propanediol (PD) in Escherichia coli strains having the glycerol dehydrogenase genes have been investigated. The study seeks to discover new fermentation routes for 1,2-PD, an important chemical derived from propylene, from renewable sources. It will also be demonstrated that E. coli overexpressing the methylglyoxal synthase gene produces 1,2-PD. Results reveal that expressing glycerol dehydrogenase or methylglyoxal synthase in E. coli result in the production of 0.25 g of 1,2-PD per liter.
