The neural circuits responsible for behavior remain largely unknown. Previous efforts have reconstructed the complete circuits of small animals, with hundreds of neurons, and selected circuits for larger animals. Here we (the FlyEM project at Janelia and collaborators at Google) summarize new methods and present the complete circuitry of a large fraction of the brain of a much more complex animal, the fruit fly Drosophila melanogaster. Improved methods include new procedures to prepare, image, align, segment, find synapses, and proofread such large data sets; new methods that define cell types based on connectivity in addition to morphology; and new methods to simplify access to a large and evolving data set. From the resulting data we derive a better definition of computational compartments and their connections; an exhaustive atlas of cell examples and types, many of them novel; detailed circuits for most of the central brain; and exploration of the statistics and structure of different brain compartments, and the brain as a whole. We make the data public, with a web site and resources specifically designed to make it easy to explore, for all levels of expertise from the expert to the merely curious. The public availability of these data, and the simplified means to access it, dramatically reduces the effort needed to answer typical circuit questions, such as the identity of upstream and downstream neural partners, the circuitry of brain regions, and to link the neurons defined by our analysis with genetic reagents that can be used to study their functions.Note: In the next few weeks, we will release a series of papers with more involved discussions. One paper will detail the hemibrain reconstruction with more extensive analysis and interpretation made possible by this dense connectome. Another paper will explore the central complex, a brain region involved in navigation, motor control, and sleep. A final paper will present insights from the mushroom body, a center of multimodal associative learning in the fly brain.

Focused Ion Beam Scanning Electron Microscopy (FIB-SEM) generates 3D datasets optimally suited for segmentation of cell ultrastructure and automated connectome tracing but is limited to small fields of view and is therefore incompatible with the new generation of ultrafast multibeam SEMs. In contrast, section-based techniques are multibeam-compatible but are limited in z-resolution making automatic segmentation of cellular ultrastructure difficult. Here we demonstrate a novel 3D electron microscopy technique, Gas Cluster Ion Beam SEM (GCIB-SEM), in which top-down, wide-area ion milling is performed on a series of thick sections, acquiring < 10 nm isotropic datasets of each which are then stitched together to span the full sectioned volume. Based on our results, incorporating GCIB-SEM into existing single beam and multibeam SEM workflows should be straightforward and should dramatically increase reliability while simultaneously improving z-resolution by a factor of 3 or more.

Background and aims Diabetes contributes to atherosclerosis partially through induction of oxidative stress. Both vitamin D receptor (VDR) and retinoid X receptor (RXR) agonists exhibit anti-atherogenic effects. Methods We explored the effects of combination treatment with VDR and RXR agonists (represented by calcitriol and bexarotene, respectively) on atherosclerosis progression and the mechanisms involved, using a diabetes model of mice. The animals were intragastrically fed calcitriol (200 ng/kg, twice-a-week), bexarotene (10 mg/kg, once-daily) either alone or in combination for 12 weeks. Results VDR and RXR agonists delayed atherosclerosis progression independent of serum lipid and glucose levels, and significantly reduced the protein expression of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase subunit gp91phox and nuclear factor-kappa B (NF-κB) subunit p65, as well as plasma biomarkers of oxidative stress and inflammation. Combination therapy alleviated atherosclerosis and inhibited indexes of oxidative stress and inflammation to a greater extent than either monotherapy. In the in vitro study, naturally occurring VDR ligand 1α,25-dihydroxyvitamin D 3 (1,25[OH] 2 D 3 ) and RXR ligand 9-cis retinoic acid (9-cis-RA), both significantly inhibited high-glucose-induced endothelial cell apoptosis. Co-administration of VDR and RXR ligands produced synergistic protection against endothelial apoptosis by antagonizing the protein kinase C /NADPH oxidase/reactive oxygen species pathway. The inhibitory effects of 9-cis-RA on oxidative stress was attenuated when VDR was downregulated by VDR siRNA; however, downregulation of RXR by RXR siRNA imposed no influence on the effects of 1,25(OH) 2 D 3 . Conclusions Combination treatment with VDR and RXR agonists synergistically alleviated diabetic atherosclerosis through inhibition of oxidative stress, and the preventive effects of RXR agonist may partially depend on VDR activation.

Mn-passivated nanocrystalline ZnO:S thin films were fabricated by thermally oxidizing Mn-doped ZnS (ZnS:Mn) films prepared by electron beam evaporation. Mn was introduced to passivate the surface defects of ZnO and to improve the optical properties. X-ray diffraction (XRD) and photoluminescence (PL) spectra at 81.9 K indicated the S content in ZnO thin film gradually decreased with increasing annealing temperature. The fitted result of the temperature-dependent PL spectra in the range from 81.9 to 302.2 K showed that S dopant could broaden the optical band gap energy of ZnO. Room temperature PL spectra confirmed that the ultraviolet peak shifted to lower energy with the decrease of S content in the thin film because of the Burstein–Moss effect.

AlN nanocrystals were prepared in organic solvent at atmospheric pressure and low temperature by the Schlenk technique. Both hexagonal and cubic AlN nanocrystals were obtained. The hexagonal nano-AlN powder possessed a wurtzite structure with a =3.124 A, c =5.024 A, the average grain size was about 2 nm. The lattice constant of the cubic nano-AlN was a =9.171 A, the average grain size was about 4 nm. The structural and optical properties of the obtained AlN were analyzed. The emission related to deep-level defects was investigated by using temperature-dependent photoluminescence.

In this paper, a neural network based uniformity controller is developed for the linear chemical-mechanical planarization (CMP) process. The control law utilizes the metrology measurements of the wafer uniformity profile and tunes the pressures of different air-bearing zones on Lam linear CMP polishers. A feedforward neural network is used to self-learn the CMP process model and a direct inverse control with neural network is utilized to regulate the process to the target. Simulation and experimental results are presented to illustrate the control system performance. Compared with the results by using statistical surface response methods (SRM), the proposed control system can give more accurate uniformity profiles and more flexibility.

Non-linear equations of transverse motion for a generally laminated, truncated, conical shell are derived by use of the dynamic virtual work principle. The effects of transverse shear deformation, rotatory inertia and geometric initial imperfection are taken into account. A solution for non-linear free vibrations of a symmetrically laminated, cross-ply, geometrically perfect, thick, conical shell with its two ends both clamped and both simply supported is expressed in the form of generalized double Fourier series with time-dependent coefficients. The Galerkin procedure furnishes a system of equations for time functions which are solved by the method of harmonic balance. Non-linear vibrations of circular cylindrical shells and annular plates are treated as special cases. Numerical results in non-linear vibrations of symmetric cross-ply conical laminates are presented graphically for different parameters. Present results are also compared with available data for special cases.

A Fourier-Bessel series solution is formulated for the non-linear vibration of an unsymmetrically laminated moderately thick shallow spherical shell with its edge elastically restrained against rotation. The effects of transverse shear deformation and rotatory inertia are included in the analysis. The equations of motion are reduced to a set of non-linear ordinary differential equations for the time-dependent coefficients in the series by making use of the Galerkin method. These equations are solved by the method of harmonic balance for undamped free vibrations. The corresponding postbuckling problem is treated as a special case. Numerical results in non-linear free vibration, buckling and postbuckling are presented for various ratios of base radius to thickness and shell rise to thickness, number of layers, material properties and boundary conditions. The present results are compared with available data.
