Abstract

The receptor for interleukin-5 (IL-5) is composed of two different subunits. The IL-5 receptor alpha (IL-5R alpha) is required for ligand-specific binding while association with the beta-chain results in increased binding affinity. Murine IL-5 (mIL-5) has similar activity on human and murine cells, whereas human IL-5 (hIL-5) has marginal activity on murine cells. We found that the combined substitution of K84 and N108 on hIL-5 by their respective murine counterpart yields a molecule which is as potent as mIL-5 for growth stimulation of a murine cell line. Since the unidirectional species specificity is due only to the interaction with the IL-5R alpha subunit, we have used chimeric IL-5R alpha molecules to define regions of hIL-5R alpha involved in species-specific hIL-5 ligand binding. We found that this property is largely determined by the NH2-terminal module of hIL-5R alpha, and detailed analysis defined D56 and to a lesser extent E58 as important for binding. Moreover, two additional residues, D55 and Y57, were identified by alanine scanning mutagenesis within the same region. Based on the observed homology between the NH2-terminal module and the membrane proximal (WSXWS-containing) module of hIL-5R alpha we located this stretch of four amino acid residues (D55, D56, Y57 and E58) in the loop region that connects the C and D beta-strands on the proposed tertiary structure of the NH2-terminal module.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract

Summary In bovine serum, the natural agglutinin for goose red blood cells was shown to be an IgM antibody which differed from immune IgM antibody in its partial inactivation during acetone-ether extraction. Agglutinins active at pH 6.2 were not readily adsorbed at pH 9, they were partially eluted from the red cells at room temperature. Both natural and immune IgM antibodies were inactivated by 64° C. for 30 min., although “7S” antibodies were not affected. Non-specific inhibitors of arbovirus haemagglutination in bovine serum had properties suggesting they were β-lipoprotein. Both low density or less) and higher density (between l-01 and 1-04) fractions caused inhibition. Kaolin adsorption of bovine serum did not remove all non-specific inhibitors. Acetone-ether extraction was the preferred technique, because it removed non-specific inhibitors more efficiently and because it caused partial inactivation of natural agglutinins without affecting immune IgM antibody.

Abstract

A semi-automatic device is described for harvesting cells or precipitates from microplates on to disposable microfilters (‘C.R.C. thimbles’). The use of this harvesting system for measuring incorporation of [ 3 H] thymidine into phytohaemagglutinin-stimulated lymphocytes is investigated. The thimbles themselves do not complicate beta counting compared with counting on glass fibre filter paper itself. An estimated counting efficiency of 25% was obtained with trichloroacetic acid washing. This could be increased to 43% by dissolving the residue off the filter with NCS Solubiliser. If a trichloroacetic acid wash is not used efficiency is lower because of increased internal absorption. At least 10 6 lymphocytes/microfilter could be harvested. The system could have wide application in immunology and cell biology.

Abstract

What seems to be an IgD antibody to insulin has been found in three of six diabetic patients investigated. The antibody has been detected by the red-blood-cell linked antigen-antiglobulin reaction. Although the IgD antibody titres in the three sera are low when compared with the IgG titres, the specificity of the reactions has been checked by careful inhibition tests.

Abstract

The oxidation of carbohydrate by periodate to produce reactive aldehyde groups which will couple to side chain amino groups of proteins has been used in three immunochemical coupling reactions. The carbohydrate of the red cell membrane was oxidised and allowed to take up BSA for passive haemagglutination. The carbohydrate of an anti-red cell antibody was oxidised and allowed to couple to BSA. Dextran was oxidised and allowed to couple to an anti-red cell antibody. These can be used for coating red cells with protein of polysaccharide respectively.
