Deforestation and forest degradation remain huge global environmental challenges. Over the last decades, various forest governance initiatives and institutions have evolved in global response to interlinked topics such as climate change mitigation, biodiversity conservation, indigenous rights, and trade impacts – accompanied by various levels of academic attention. Using a Delphi methodology that draws on both policy and academic insights, we assess the currently perceived state of play in global forest governance and identify possible future directions. Results indicate that state actors are seen to be key in providing supportive regulatory frameworks, yet interviewees do not believe these will be established at the global scale. Rather, respondents point to issue-specific, regional and inter-regional coalitions of the willing, involving the private sector, to innovate global forest governance. Linking forest issues with high politics may hold promise, as demonstrated by initiatives regarding illegal logging and timber trade. Confident rule-setting in support of the public good as well as responsible investments are seen as further avenues. New forest governance “hypes”, if used strategically, can provide leverage points and resources to ensure sustainability effects on the ground. At the same time, informal markets are often crucial for governance outcomes and need consideration. As such, clarifying tenure in sovereignty-sensitive ways is important, as are innovative ways for inclusive “glocal” decision-making. Lastly, new technologies, big data and citizens’ capacities are identified as potent innovation opportunities, for making global dependencies between consumption, production and deforestation visible and holding players accountable across the value chains.

Experimental information on fragment emissions is important in understanding the dynamics of nuclear collisions and in the development of transport model simulating heavy-ion collisions. The composition of complex fragments emitted in the heavy-ion collisions can be explained by statistical models, which assume that thermal equilibrium is achieved at collision energies below 100 MeV/u. Our new experimental data together with theoretical analyses for light particles from Sn+Sn collisions at 270 MeV/u, suggest that the hypothesis of thermal equilibrium breaks down for particles emitted with high transfer momentum. To inspect the system's properties in such limit, the scaling features of the yield ratios of particles from two systems, a neutron-rich system of ${}^{132}\mathrm{Sn}+{}^{124}\mathrm{Sn}$ and a nearly symmetric system of ${}^{108}\mathrm{Sn}+{}^{112}\mathrm{Sn}$, are examined in the framework of the statistical multifragmentation model and the antisymmetrized molecular dynamics model. The isoscaling from low energy particles agree with both models. However the observed breakdown of isoscaling for particles with high transverse momentum cannot be explained by the antisymmetrized molecular dynamics model.

International audience; Thymically-derived Foxp3+ regulatory T cells (Treg) critically control immunological tolerance. These cells are generated in the medulla through high affinity interactions with medullary thymic epithelial cells (mTEC) expressing the Autoimmune regulator (Aire). Recent advances have revealed that thymic Treg contain not only developing but also recirculating cells from the periphery. Although Aire is implicated in the generation of Foxp3+ Treg, its role in the biology of recirculating Treg remains elusive. Here, we show that Aire regulates the suppressive signature of recirculating Treg independently of the remodeling of the medullary 3D organization throughout life where Treg reside. Accordingly, the adoptive transfer of peripheral Foxp3+ Treg in AireKO recipients led to an impaired suppressive signature upon their entry into the thymus. Furthermore, recirculating Treg from AireKO mice failed to attenuate the severity of multiorgan autoimmunity, demonstrating that their suppressive function is altered. Using bone marrow chimeras, we reveal that mTEC-specific expression of Aire controls the suppressive signature of recirculating Treg. Finally, mature mTEC lacking Aire were inefficient in stimulating peripheral Treg both in polyclonal and antigen-specific co-culture assays. Overall, this study demonstrates that Aire confers to mTEC the ability to restimulate recirculating Treg, unravelling a novel function for this master regulator in Treg biology.

Development of Foxp3-expressing regulatory T-lymphocytes (Treg) in the thymus is controlled by signals delivered in T-cell precursors

Regulatory T lymphocytes expressing the forkhead/winged helix transcription factor Foxp3 (Treg) play a vital role in the protection of the organism from autoimmune disease and other immunopathologies. The antigen specificity of Treg plays an important role in their in vivo activity. We therefore assessed the diversity of the T-cell receptors (TCRs) for antigen expressed by Treg newly developed in the thymus of autoimmune type 1 diabetes-prone NOD mice and compared it to the control mouse strain C57BL/6. Our results demonstrate that use of the TCRα and TCRβ variable (V) and joining (J) segments, length of the complementarity determining region (CDR) 3, and the diversity of the TCRα and TCRβ chains are comparable between NOD and C57BL/6 mice. Genetic defects affecting the diversity of the TCR expressed by newly developed Treg therefore do not appear to be involved in the etiology of type 1 diabetes in the NOD mouse.

Interactions of developing T cells with Aire+ medullary thymic epithelial cells expressing high levels of MHCII molecules (mTEChi) are critical for the induction of central tolerance in the thymus. In turn, thymocytes regulate the cellularity of Aire+ mTEChi. However, it remains unknown whether thymocytes control the precursors of Aire+ mTEChi that are contained in mTEClo cells or other mTEClo subsets that have recently been delineated by single-cell transcriptomic analyses. Here, using three distinct transgenic mouse models, in which antigen presentation between mTECs and CD4+ thymocytes is perturbed, we show by high-throughput RNA-seq that self-reactive CD4+ thymocytes induce key transcriptional regulators in mTEClo and control the composition of mTEClo subsets, including Aire+ mTEChi precursors, post-Aire and tuft-like mTECs. Furthermore, these interactions upregulate the expression of tissue-restricted self-antigens, cytokines, chemokines, and adhesion molecules important for T-cell development. This gene activation program induced in mTEClo is combined with a global increase of the active H3K4me3 histone mark. Finally, we demonstrate that these self-reactive interactions between CD4+ thymocytes and mTECs critically prevent multiorgan autoimmunity. Our genome-wide study thus reveals that self-reactive CD4+ thymocytes control multiple unsuspected facets from immature stages of mTECs, which determines their heterogeneity.

Systematic examination of fusion for $^{39,41,45,47}$K + $^{28}$Si and $^{36,44}$Ar + $^{28}$Si provides insight into the impact of neutron and proton exchange on fusion for nuclei at and near the N=20 and N=28 shells. Comparison of the reduced excitation functions reveals a marked difference between the behavior of open-shell and closed-shell systems. While coupled channels calculations provide a good description for the closed-shell nuclei they significantly under-predict the fusion cross-section for open-shell nuclei. Background: Psoriasis is a chronic skin disease in which lifelong treatment is required. Recently, we faced COVID-19 pandemic where adherence to a prescribed drug could have been affected. Objectives: We conduct a study to assess this statement. Methods: The study was performed in Dermatology Department with Pharmacy database in the Hospital Universitario de La Princesa. A retrospective observational database study was performed, including all psoriatic patients with a biological therapy prescription between March 13 and May 31, 2020, with a paired control group between March 13 and May 31, 2019. Medication possession ratio (MPR) was used to determine the adherence. Results: A total of 244 patients were included in the cohort and 228 in the control group. We observed a decrease in the percentage of adherent patients of 40.0% in COVID-19 period. Overall, MPR was lower in the 2020 period than in 2019. MPRs by treatment and drug family, with the exception of etanercept, certolizumab pegol, and guselkumab, were significantly lower between the 2019 and the 2020 period. Conclusions: Adherence to a prescribed drug can be affected by different factors such as age, sex, income quintile, or stress. We have demonstrated a deep impact on adherence because of the situation created by COVID-19 pandemic. These data are in contrast with previously reported results where adherence was slightly affected, probably due to the different epidemiological situation in different countries. Further efforts are needed to be done to mitigate COVID-19 pandemic influence on the therapeutic adherence.

Background Tumor-associated myeloid cells can impede productive anti-tumor immunity. One strategy for targeting immunosuppression is myeloid reprogramming, which drives immunosuppressive myeloid cells to acquire an immunostimulatory phenotype. Triggering receptor expressed on myeloid cells-1 (TREM1) is an immunoglobulin superfamily cell surface receptor expressed on neutrophils and subsets of monocytes and tissue macrophages. TREM1 associates with DAP12 adaptor and induces proinflammatory signaling, amplifies innate immune responses, and is implicated in the development of acute and chronic inflammatory diseases. TREM1 is also enriched in tumors, specifically on tumor-associated myeloid cells. To investigate the potential of TREM1 modulation as an anti-cancer therapeutic strategy, we developed PY159, an afucosylated humanized anti-TREM1 monoclonal antibody, and characterized it in the pre-clinical assays described below. Materials and Methods An FcγR binding ELISA and a Jurkat TREM1/DAP12 NFAT-luciferase reporter cell line were used to assess PY159 binding to human FcγRs and TREM1 signaling, respectively. PY159 responses in human whole blood in vitro were evaluated by flow cytometry, transcriptional analysis of sorted leukocyte subsets, and measurement of secreted cytokines/chemokines by MSD. A Transwell system was used to evaluate PY159 effects on neutrophil chemotaxis. TREM1 expression in human tumors was validated by scRNAseq, immunohistochemistry, and flow cytometry. Anti-tumor efficacy of a surrogate anti-mouse TREM1 antibody, PY159m, was evaluated using syngeneic mouse tumor models, either as a single agent or in combination with anti-PD-1. Results PY159 afucosylation increased its binding affinity for FcγR and its ability to activate TREM1/DAP12 signaling. In human blood assays, PY159 treatment did not induce depletion of TREM1-expressing cells. Rather, it upregulated monocyte activation markers, promoted neutrophil chemotaxis, and induced proinflammatory cytokines and chemokines, which was dependent on PY159 afucosylation. In human tumors, TREM1 was detected on tumor-associated neutrophils, tumor-associated macrophages, and monocytic myeloid-derived suppressive cells. PY159 induced proinflammatory cytokines and chemokines in dissociated human tumors in vitro, demonstrating that PY159 can reprogram tumor-associated myeloid cells. A surrogate anti-mouse TREM1 antibody, PY159m, exhibited anti-tumor efficacy in several syngeneic mouse tumor models, both as single-agent and in combination with anti-PD-1. Conclusions These results show that PY159 is a TREM1 agonist that reprograms myeloid cells and unleashes anti-tumor immunity. PY159 safety and efficacy are currently being evaluated in first-in-human clinical trial (NCT04682431) involving patients resistant and refractory to standard of care therapies. A ^{13}F resonance was observed following a charge-exchange reaction between a fast ^{13}O beam and a ^{9}Be target. The resonance was found in the invariant-mass distribution of 3p+^{10}C events and probably corresponds to a 5/2^{+} excited state. The ground state was also expected to be populated, but was not resolved from the background. The observed level decays via initial proton emissions to both the ground and first 2^{+} state of ^{12}O, which subsequently undergo 2p decay. In addition, there may also be a significant proton decay branch to the second 2^{+} level in ^{12}O. The wave function associated with the observed level may be collectivized due to coupling to the continuum as is it located just above the threshold for proton decay to the 2_{2}^{+} state of ^{12}O.

A kinematically complete quasifree (p,pn) experiment in inverse kinematics was performed to study the structure of the Borromean nucleus ^{17}B, which had long been considered to have a neutron halo. By analyzing the momentum distributions and exclusive cross sections, we obtained the spectroscopic factors for 1s_{1/2} and 0d_{5/2} orbitals, and a surprisingly small percentage of 9(2)% was determined for 1s_{1/2}. Our finding of such a small 1s_{1/2} component and the halo features reported in prior experiments can be explained by the deformed relativistic Hartree-Bogoliubov theory in continuum, revealing a definite but not dominant neutron halo in ^{17}B. The present work gives the smallest s- or p-orbital component among known nuclei exhibiting halo features and implies that the dominant occupation of s or p orbitals is not a prerequisite for the occurrence of a neutron halo.

International audience; The influence of shell effects on fusion of mid-mass nuclei is explored using isotopic chains of K and Ar ions on an oxygen target. Comparison of the reduced excitation functions reveals that the fusion cross section for the open neutron-shell projectile nuclei K41 and K45 is systematically larger than for the closed neutron-shell projectiles K39 and K47. The São Paulo fusion model using matter densities from systematics fails to describe the measured excitation functions. Use of more realistic densities from a Dirac-Hartree-Bogoliubov (DHB) approach performs significantly better though it still overpredicts the closed-shell nuclei.

Many neutron star (NS) properties, such as the proton fraction within a NS, reflect the symmetry energy contributions to the Equation of State that dominate when neutron and proton densities differ strongly. To constrain these contributions at supra-saturation densities, we measure the spectra of charged pions produced by colliding rare isotope tin (Sn) beams with isotopically enriched Sn targets. Using ratios of the charged pion spectra measured at high transverse momenta, we deduce the slope of the symmetry energy to be $42 < L < 117$ MeV. This value is slightly lower but consistent with the $L$ values deduced from a recent measurement of the neutron skin thickness of $^{208}$Pb.

The formation of a dineutron in the nucleus $^{11}$Li is found to be localized to the surface region. The experiment measured the intrinsic momentum of the struck neutron in $^{11}$Li via the $(p,pn)$ knockout reaction at 246 MeV/nucleon. The correlation angle between the two neutrons is, for the first time, measured as a function of the intrinsic neutron momentum. A comparison with reaction calculations reveals the localization of the dineutron at $r\sim3.6$ fm. The results also support the density dependence of dineutron formation as deduced from Hartree-Fock-Bogoliubov calculations for nuclear matter.

Spectroscopic factors to low-lying negative-parity states in $^{11}$Be extracted from the $^{12}$B($d$,$^3$He)$^{11}$Be proton-removal reaction are interpreted within the rotational model. Earlier predictions of the $p$-wave proton removal strengths in the strong coupling limit of the Nilsson model underestimated the spectroscopic factors to the $3/2^-_1$ and $5/2^-_1$ states and suggested that deviations in the $1^+$ ground state of the odd-odd $^{12}$B due to Coriolis coupling should be further explored. In this work we use the Particle Rotor Model to take into account these effects and obtain a good description of the level scheme in $^{11}$B, with a moderate $K$-mixing of the proton Nilsson levels [110]1/2 and [101]3/2. The observation of enhanced octupole collectivity in Ba144,146 as measured in sub-barrier Coulomb excitation, consistent with static octupole deformation, has further heightened this interest. In the present work, these studies are extended to the neighboring odd-mass Ba143 to investigate the interplay between single-particle and collective octupole degrees of freedom. A new measurement of the first 92 - state lifetime is also presented. Reflection-Asymmetric Triaxial Particle Rotor Model calculations indicate that the negative-parity bands in Ba143 can be understood as a decoupled structure of νh9/2 parentage, while the positive-parity bands are built on a decoupled octupole phonon. No evidence for E3 excitation is observed in this work, but an upper limit is placed on the E3 matrix element to the lowest octupole band.

Type-I x-ray bursts can reveal the properties of an accreting neutron star system when compared with astrophysics model calculations. However, model results are sensitive to a handful of uncertain nuclear reaction rates, such as ^{22}Mg(α,p). We report the first direct measurement of ^{22}Mg(α,p), performed with the Active Target Time Projection Chamber. The corresponding astrophysical reaction rate is orders of magnitude larger than determined from a previous indirect measurement in a broad temperature range. Our new measurement suggests a less-compact neutron star in the source GS1826-24.

Background Coronary microvascular obstruction (MVO) is a noxious condition frequently occurring in patients with ST-elevation myocardial infarction (STEMI) after percutaneous coronary intervention (PCI). However, multiple mechanisms are involved in the pathogenesis of MVO and not yet fully understood. Recent studies suggested that perilipin 2 (PLIN2) may play an important role in lipid metabolism of macrophages resident in atherosclerotic plaques along with a role in enhancing oxidative stress. Purpose To study the association between PLIN2 and MVO in STEMI patients undergoing primary PCI. We also assessed the role of PLIN2 to predict future major cardiovascular events (MACEs). Methods STEMI patients undergoing primary PCI were enrolled. PLIN2 was dosed within 24 hours from admission in peripheral blood monocytes. MVO was assessed using TIMI flow grade and myocardial blush grade on coronary angiogram after PCI, and patients were stratified accordingly (MVO or noMVO). Major adverse cardiac events (defined as a composite of cardiac death, non-fatal myocardial infarction, re-admission for heart failure and target vessel revascularization) were assessed at clinical follow-up. Results Among 100 STEMI patients (mean age, 65.2±12.0 years, 81 males), 33 (33.0%) had MVO. Patients with MVO were older, had higher troponin I peak, C-reactive protein and lower left ventricular ejection fraction on admission. Patients with MVO had significantly higher levels of PLIN2 (1.03±0.28 vs. 0.90±0.16, p=0.019) compared to noMVO patients. Age [OR (95% CI) per year, 1.045 (1.005–1.087), p=0.026] and PLIN2 [OR (95% CI) per unit, 16.606 (2.027–136.030), p=0.009] were associated with MVO at univariate logistic regression analysis. However, only PLIN2 levels [OR (95% CI) per unit, 12.325 (1.446–105.039), p=0.033] were independently associated with MVO at multivariate analysis. Follow up data were available for 76 patients (76%). After a mean follow up of 182.2±126.6 days, 13 MACEs occurred. Patients with MVO had more MACEs [9 (37.5%) vs. 4 (7.7%), p Conclusions In STEMI patients undergoing primary PCI, PLIN2 was independently associated with MVO. PLIN2 was an independent predictor of MACEs at clinical follow-up. In a recent experiment, carried out at the Radioactive Isotope Beam Factory/RIKEN, the $^{25}\mathrm{F}(p,2p)^{24}\mathrm{O}$ reaction was studied at 270 MeV/A in inverse kinematics. Derived spectroscopic factors suggest that the effective core of $^{25}\mathrm{F}$ significantly differs from a free $^{24}\mathrm{O}$ nucleus. We interpret these results within the particle-rotor model and show that the experimental level scheme of $^{25}\mathrm{F}$ can be understood in the rotation-aligned coupling scheme with its $5/{2}_{1}^{+}$ ground state as the bandhead of a decoupled band. The excitation energies of the observed $1/{2}_{1}^{+}$ and $9/{2}_{1}^{+}$ states correlate strongly with the rotational energy of the effective core, seen by the odd proton, and allow us to estimate its ${2}^{+}$ energy at $\ensuremath{\approx}3.2\phantom{\rule{0.28em}{0ex}}\mathrm{MeV}$ and a moderate quadrupole deformation ${\ensuremath{\epsilon}}_{2}\ensuremath{\approx}0.15$. The measured fragmentation of the $\ensuremath{\pi}{d}_{5/2}$ single-particle strength is discussed, and some further experiments suggested.

International audience; Regulatory T lymphocytes (Treg) play a vital role in the protection of the organism against autoimmune pathology. It is therefore paradoxical that comparatively large numbers of Treg were found in the thymus of type I diabetes-prone NOD mice. The Treg population in the thymus is composed of newly developing cells and cells that had recirculated from the periphery back to the thymus. We here demonstrate that exceptionally large numbers of Treg develop in the thymus of young, but not adult, NOD mice. Once emigrated from the thymus, an unusually large proportion of these Treg is activated in the periphery, which causes a particularly abundant accumulation of recirculating Treg in the thymus. These cells then rapidly inhibit de novo development of Treg. The proportions of developing Treg thus reach levels similar to or lower than those found in most other, type 1 diabetes-resistant, inbred mouse strains. Thus, in adult NOD mice the particularly large Treg-niche is actually composed of mostly recirculating cells and only few newly developing Treg.

Twenty-one two-proton knockout (p; 3p) cross sections from neutron-rich nuclei at ~ 250 MeV/nucleon were measured. For the first time, the angular distribution of the three emitted protons were measured in coincidence with the tracker and hydrogen target MINOS, demonstrating that the (p; 3p) kinematics is consistent with two sequential proton-proton collisions within the projectile nucleus. Ratios of (p; 3p) over (p; 2p) inclusive cross sections follow the trend of other many-proton (neutron) removal reactions, further reinforcing the sequential nature of (p; 3p) in neutron-rich nuclei.

In an experiment performed at Lawrence Berkeley National Laboratory's 88-inch cyclotron, the isotope ^{244}Md was produced in the ^{209}Bi(^{40}Ar,5n) reaction. Decay properties of ^{244}Md were measured at the focal plane of the Berkeley Gas-filled Separator, and the mass number assignment of A=244 was confirmed with the apparatus for the identification of nuclide A. The isotope ^{244}Md is reported to have one, possibly two, α-decaying states with α energies of 8.66(2) and 8.31(2) MeV and half-lives of 0.4_{-0.1}^{+0.4} and ∼6 s, respectively. Additionally, first evidence of the α decay of ^{236}Bk was observed and is reported.

Here we present new information on the shape evolution of the very neutron-rich ^{92,94}Se nuclei from an isomer-decay spectroscopy experiment at the Radioactive Isotope Beam Factory at RIKEN. High-resolution germanium detectors were used to identify delayed γ rays emitted following the decay of their isomers. New transitions are reported extending the previously known level schemes. The isomeric levels are interpreted as originating from high-K quasineutron states with an oblate deformation of β∼0.25, with the high-K state in ^{94}Se being metastable and K hindered. Following this, ^{94}Se is the lowest-mass neutron-rich nucleus known to date with such a substantial K hindrance. Furthermore, it is the first observation of an oblate K isomer in a deformed nucleus. This opens up the possibility for a new region of K isomers at low Z and at oblate deformation, involving the same neutron orbitals as the prolate orbitals within the classic Z∼72 deformed hafnium region. From an interpretation of the level scheme guided by theoretical calculations, an oblate deformation is also suggested for the ^{94}Se_{60} ground-state band.

The analysis considers two different coupling schemes, namely, the strong and decoupled limits of the particle rotor model to describe the ground and isomeric states of F18, as well as the transitions to the ground-state band in F19. Our results, obtained using Nilsson amplitudes for the neutron levels 12[220], 32[211], and 52[202] calculated at a deformation ϵ2=0.32 are in agreement with the measurements and USDB shell-model calculations.

The invariant-mass distribution of $^{11}\mathrm{O}$ fragments formed in two-neutron-knockout reactions with a $^{13}\mathrm{O}$ projectile and $^{9}\mathrm{Be}$ target has been further examined. Gating on events where the $^{9}\mathrm{C}$ decay fragments produced following $2p$ emission are recoiled transversely in the projectile's frame improves the overall invariant-mass resolution. The observed peak is now shown to have contributions from at least two $^{11}\mathrm{O}$ levels, in contradiction with the suggestion of Fortune [Phys. Rev. C 99, 051302(R) (2019)]. The data, however, do not differentiate between the ground-state properties obtained by the Gamow coupled-channels calculation and the prompt $2p$-decay model of Fortune. The ground-state $2p$-decay energy is 4.25(6) MeV in a two-level fit to the data, but lower values are possible if more states contribute to the observed spectrum, as suggested in the previous analysis.

The structures of the neutron-rich Co69,71,73 isotopes were investigated via (p,2p) knockout reactions at the Radioactive Isotope Beam Factory, RIKEN. Isotopes of interest were studied using the DALI2 γ-ray detector array combined with the MINOS target and tracker system. Level schemes were reconstructed using the γ-γ coincidence technique, with tentative spin-parity assignments based on the measured inclusive and exclusive cross sections. Comparison with shell-model calculations using the Lenzi-Nowacki-Poves-Sieja LNPS and PFSDG-U interactions suggests coexistence of spherical and deformed shapes at low excitation energies in the Co69,71,73 isotopes. The distorted-wave impulse approximation (DWIA) framework was used to calculate the single-particle cross sections. These values were compared with the experimental findings.
